Ion Effects on Calcium Accumulation by Cardiac Sarcoplasmic Reticulum   总被引:6,自引:3,他引:3  
The effects of monovalent cations on the active calcium-accumulating ability of cardiac sarcoplasmic reticulum were assessed. Grana prepared in an ion-free system accumulated calcium when ATP and Mg++ were present. Sodium ion and to a lesser extent lithium but not K+ reduced the amount of calcium taken up. The reduction of calcium binding by Na+ is not due to inhibition of uptake but to a rapid release of the radiocalcium bound. The amount of calcium released by sodium does not appear to be enough to explain contraction on the basis of sodium influx into muscle, but may be significant in the regulation of tension.  相似文献

Abstract: Somatostatin (SS) is a neuropeptide that is distributed in various regions of the CNS, where it may act as a neurotransmitter or neuromodulator. SS produces multiple effects in the CNS through interactions with membrane receptors. In particular, SS inhibits various secretory responses in different cell types. In the present study, we have investigated the effects of exogenous application of SS on the intracellular free Ca2+ concentration ([Ca2+]i) in PC12 cells, a rat pheochromocytoma cell line. SS did reduce the magnitude of the secondary, maintained Ca2+ influx brought about by K+ depolarization. Similar effects were obtained with the application of SS analogues, such as d -Trp8-SS, d -Trp8- d -Cys14-SS, CGP-23996, and SMS-201995. In addition, treatment with cyclo-SS, a SS antagonist, did not alter [Ca2+]i. Experiments with selective blockers of different voltage-dependent Ca2+ channels, such as methoxyverapamil (D600) and Ω-conotoxin GVIA, demonstrated that the effects of SS on [Ca2+]i were mediated by voltage-dependent Ca2+ channels of the L type. Control experiments with a membrane potential indicator, i.e., the fluorescent dye bisoxonol, excluded that SS influenced the level of the membrane potential. SS effects on PC12 cells suggest the possibility that this neuropeptide plays a role in the modulation of cell functional activity by altering Ca2+ influx.  相似文献

Calcium release and binding produced by alterations in pH were investigated in isolated sarcoplasmic reticulum (SR) from skeletal muscle. When the pH was abruptly increased from 6.46 to 7.82, after calcium loading for 30 sec, 80–90 nanomoles (nmole) of calcium/mg protein were released. When the pH was abruptly decreased from 7.56 to 6.46, after calcium loading for 30 sec, 25–30 nmole of calcium/mg protein were rebound. The calcium release process was shown to be a function of pH change: 57 nmole of calcium were released per 1 pH unit change per mg protein. The amount of adenosine triphosphate (ATP) bound to the SR was not altered by the pH changes. The release phenomenon was not due to alteration of ATP concentration by the increased pH. Native actomyosin was combined with SR in order to study the effectiveness of calcium release from the SR by pH change in inducing super-precipitation of actomyosin. It was found that SR, in an amount high enough to inhibit superprecipitation at pH 6.5, did not prevent the process when the pH was suddenly increased to 7.3, indicating that the affinity of SR for calcium depends specifically on pH. These data suggest the possible participation of hydrogen ion concentration in excitation-contraction coupling.  相似文献

利用扫描电镜、透射电镜方法,观察氮离子束注入对芦荟细胞壁结构的影响。扫描电镜观察结果显示,在离子注入剂量达1×1016N+/cm2时,叶片表面发现了离子束刻蚀的小洞。透射电镜半薄切片观察也发现离子束轰击后,芦荟叶片表皮的横切面都有不同程度被打烂的情况,初步认为是离子束轰击引起的刻蚀效应,这与扫描电镜的观察结果相一致。研究结果可为离子束介导转基因技术的研究提供参考。  相似文献

为了进一步研究细胞外钙离子浓度变化对甲壳动物感光细胞超微结构的影响,应用透射电子显微镜显示了日本沼虾感光细胞,在暗适应时高钙离子浓度中温育的感光器细胞的感杆束直径下降,微绒毛排列零乱;多囊体、板膜体数量增加;色素颗粒散布在细胞质中,呈现出光适应的结构特征。而温育在低钙离子溶液和生理溶液中的感光器细胞结构相同,呈现出暗适应的结构特征。另外,细胞器中储存的钙离子也受细胞外钙离子浓度的影响,在高钙离子溶液中温育后细胞器储存的钙离子量增加,膜下储泡囊、多囊体、线粒体、色素颗粒等细胞器中的焦锑酸钙结晶颗粒比温育在低钙溶液中的细胞明显增多。结果显示,细胞外钙离子浓度变化引起细胞内钙离子浓度变化,从而影响感光器细胞的结构而影响其生理功能。  相似文献

低强度He-Ne激光对红细胞变形性的影响已得到广泛的认可和应用,但其具体调节机制尚不明确,通过研究低强度He-Ne激光对红细胞胞浆内钙离子浓度的影响,探讨其对红细胞变形性影响的机制。采用A23187处理过的红细胞,分别用5 mW和9 mW激光照射后,观察红细胞胞浆内钙离子浓度变化。结果显示:低强度He-Ne激光照射后的红细胞与无照射组的红细胞相比,红细胞胞浆内钙离子浓度显著降低,但在本实验中钙离子浓度的降低与激光照射剂量无显著相关。由此得出结论:降低红细胞胞浆内钙离子浓度可能是低强度He-Ne激光调节红细胞变形性的重要机制。  相似文献

Studies were conducted on the protective action of Ca against growth inhibition and death of pollen from Crinum asiaticum and Catharanthus roseus. Ca ions not only promoted pollen germination and pollen tube growth, but also were antagonistic to pollen injuries induced by various chemical and physical treatments. Most of the chemicals used seemed to act as osmotic inhibitors. Among the chemical agents tested, DNP inhibited pollen germination more strikingly with Ca than without it, whereas pollen tube growth was enhanced. Another metabolic inhibitor, low temperature, showed a similar effect on pollen germination. Insignificant or no protective action of Ca was observed when IAA, TIBA and coumarin, were applied. Large populations of pollen when grown in cultural media witbout Ca showed a partial protective action. The protective action of Ca in pollen growth against various inhibitors required other cations such as Mg and K ions. The promoting of pollen growth by Ca also required tbese otber cations. The protective action of Ca is considered to be based on its binding in tbe pollen cell walls, particularly in the pectic regions. This gives rise to a decreased permeability and increased structural rigidity against tbe chemical and physical inhibitors.  相似文献

目的:研究胞外不同浓度的镁离子对SD成年大鼠心肌细胞钙瞬变的影响。方法:采用激光共聚焦显微镜系统同步配合阈上电刺激探测心肌细胞钙瞬变。结果:胞外低浓度的镁离子(0 mmol /L,0.5 mmol /L; 正常镁离子浓度为1 mmol/L)可以升高钙瞬变的峰值(P〈0.05),但不影响钙瞬变的持续时间(以钙瞬变的半高宽表示)(P〉0.05);胞外高浓度的镁离子(2.5 mmol /L,5 mmol /L,10 mmol /L)均可抑制钙瞬变的峰值(P〈0.05);其中5 mmol/L和10 mmol/L的胞外镁还能延长钙瞬变的持续时间(P〈0.05)。结论:正常情况下镁对心肌细胞钙瞬变有抑制作用。  相似文献

The effects of taurine on ATP-dependent calcium ion uptake and protein phosphorylation of rat retinal membrane preparations were investigated. Taurine (20 mM) stimulates ATP-dependent calcium ion uptake by twofold in crude retinal homogenates. In contrast, it inhibits the phosphorylation of specific membrane proteins as shown by acrylamide gel electrophoresis and autoradiography. The close structural analogue of taurine, 2-aminoethylhydrogen sulfate, demonstrates similar effects in both systems, i.e., stimulation of ATP-dependent calcium ion uptake and inhibition of protein phosphorylation, whereas isethionic acid and guanidinoethanesulfonate have no effect on either system. A P1 subcellular fraction of the retinal membrane preparation that contains photoreceptor cell synaptosomes has a higher specific activity for the uptake of calcium ions. Phosphorylation of specific proteins in the P1 fraction is also inhibited by the addition of 20 mM taurine. Taurine has no effect on retinal ATPase activities or on phosphatase activity, thus suggesting that it directly affects a kinase system.  相似文献

The walls of normal root hairs of white mustard, corn and tomatoseedlings developed in moist air following seed treatment with45CaCl2, were tested by radioautography for incorporated calcium.Radioactivity was absent or faint over the growing tips butbecame concentrated immediately behind this point. It extendedback along the walls of the hairs, increasing in concentrationtowards the base. Since the hairs were subjected to plasmolysisand prolonged extraction with hot water prior to making theradioautographs, the distribution of radioactivity indicatedthe presence of bound 45Ca. These findings are compatible withthe view that gradual calcification is essential for the normalgrowth and form of the hair. Radioautographs of mustard root hairs produced in an aqueoussolution of 45CaCl2 showed conspicuous radioactivity throughoutthe whole length of the hairs but particularly over the tips.Excessive wall calcification at the tips of these hairs couldbe the cause of their stunted growth.  相似文献

The contribution of the presence of solvent to the conformations adopted by disaccharide fragments within the repeat unit of gellan have been studied by molecular modelling techniques. Initial conformational energy searches, using a dielectric continuum to represent the solvent, provided starting geometries for a series of molecular dynamics simulations. The solution behaviour from these simulations was subsequently compared to fibre diffraction data of the potassium gellan salt. The present calculations indicate considerable flexibility of the glycosidic linkages, and this is discussed in relation to its effect on gel formation. One of the fragments was solvated with explicit water molecules. These calculations showed the same conformational behaviour as those simulations conducted in implicit solvent.Finally, a series of molecular dynamics (MD) simulations were performed to study the calcium binding to gellan. 用Fura-2显微荧光测钙技术,研究了用外源性一氧化氮(NO)供体S-亚硝基谷胱甘肽(GSNO)诱导的,人脐静脉内皮细胞系ECV-304细胞胞内游离钙离子浓度([Ca2+i )升高以及硒的抑制效应.结果表明,GSNO作用于ECV-304细胞,短时间内即可导致其胞内游离钙离子浓度升高.胞外液换为无钙液或向胞外液中加入CdCl2(1 mmol/L)对GSNO引起的[Ca2+i升高无影响.提示,GSNO刺激主要引起胞内钙库释放.而且,一氧化氮清除剂血红蛋白(Hb)对这一过程有抑制作用,说明GSNO引起的胞内钙库释放由NO介导.经亚硒酸钠(1 μmol/L)处理的细胞,其NO引起的[Ca2+i升高幅度明显被抑制,说明NO的这种作用可能与细胞的氧化还原状态有关.  相似文献

5-((R)-1-Hydroxyethyl)-furo[2,3-c]pyridine ((R)-FPH) is a useful chiral building block in the synthesis of pharmaceuticals. An NADH-dependent alcohol dehydrogenase (AFPDH) isolated from Candida maris catalyzed the reduction of 5-acetylfuro[2,3-c]pyridine (AFP) to (R)-FPH with 100% enantiomeric excess. The gene encoding AFPDH was cloned and sequenced. The AFPDH gene comprises 762 bp and encodes a polypeptide of 27,230 Da. The deduced amino acid sequence showed a high degree of similarity to those of other members of the short-chain alcohol dehydrogenase superfamily. The AFPDH gene was overexpressed in Escherichia coli under the control of the lac promoter. One L of the cultured broth of an E. coli transformant coexpressing AFPDH and the glucose dehydrogenase (GDH) gene reduced 250 g of AFP to (R)-FPH in an organic solvent two-phase system. Under coupling with NADH regeneration using 2-propanol, 1 L of the cultured broth of an E. coli transformant expressing the AFPDH gene reduced 150 g of AFP to (R)-FPH. The optical purity of the (R)-FPH formed was 100% enantiomeric excess under both reaction conditions.  相似文献

The effect of egg yolk phosvitin on the permeation and absorption of calcium was investigated in vitro in relation to calcium gluconate concentration. Obtained results indicate that phosvitin significantly reduces the intestinal calcium absorption from 1 and 10 mM of calcium gluconate solution. It is associated with the formation of the complex of Ca (II) ions with phosvitin. The process of calcium permeation increases under phosvitin influence when calcium gluconate concentrations rise up to 10 mM. At a higher concentration of calcium gluconate (20 mM), no effect of phosvitin was seen on permeation of calcium ions.  相似文献

In order to clarify the interaction of calcium ion with casein, the volume change associated with the interaction was measured by dilatometric procedures. When CaCl2 was added to the casein solutions at neutral pH, a volume increase occurred and reached a constant saturated value of about 700 ml per 106 g protein with increasing CaCl2 concentrations for whole-, αs- and β-casein solutions, but there was no volume change for κ-casein solution. On the other hand, the binding of calcium ion to the casein fractions was determined by a gel filtration procedure at pH 6.0 to 9.0. The number of Ca2+ ions bound to the caseins increased with the CaCl2 concentration and pH value, and the relative order of binding capacities for the caseins was: αs-casein > whole-casein > β-casein > κ-casein.

It was found that the volume changes obtained by the dilatometry were smaller than the calculated volume increases based on the assumption that these are caused by the binding of Ca2+ ion to the caseins. Therefore it is necessary to introduce another factor which reduces the volume increase due to the Ca2+ ion binding in order to reasonably explain the measured volume changes. At present it is presumed that there occurs the unfolding of peptide chain of casein molecule on Ca2+ ion binding, which has been known to decrease the volume of the protein solution.  相似文献
