We have studied the effects of curare on responses resulting from iontophoretic application of several putative neurotransmitters onto Aplysia neurons. These neurons have specific receptors for acetylcholine (ACh), dopamine, octopamine, phenylethanolamine, histamine, gamma-aminobutyric acid (GABA), aspartic acid, and glutamic acid. Each of these substances may on different specific neurons elicit at least three types of response, caused by a fast depolarizing Na+, a fast hyperpolarizing Cl-, or a slow hyperpolarizing K+ conductance increase. All responses resulting from either Na+ or Cl- conductance increases, irrespective of which putative transmitter activated the response, were sensitive to curare. Most were totally blocked by less than or equal to 10-4 M curare. GABA responses were less sensitive and were often only depressed by 10-3 M curare. K+ conductance responses, irrespective of the transmitter, were not curare sensitive. These results are consistent with a model of receptor organization in which one neurotransmitter receptor may be associated with any of at least three ionophores, mediating conductance increase responses to Na+, Cl-, and K+, respectively. In Aplysia nervous tissue, curare appears not to be a specific antagonist for the nicotinic ACh receptor, but rather to be a specific blocking agent for a class of receptor-activated Na+ and Cl- responses.  相似文献

The influence of external calcium variation (0.7–28 mM) on acetylcholine (ACh) induced conductance changes was examined under voltage clamp conditions in snail neurons in which ACh elicitis hyperpolarizing responses and in which the main current-carrying ion species is Cl?. Raising external calcium to 28 mM as well as lowering external calcium to 1.75 mM decreased the ACh-induced condutance change without altering the reversal potential for ACh-induced currents. Lowering external calcium to 0.7 mM increased the ACh-induced conductance change and shifted the reversal potential to less negative values. The results at 28 mM calcium can be best explained on the assumption that excess calcium interferes with the interaction between ACh and the receptor. The results at 1.75 mM calcium can be satisfactorily explained in terms of the concept that fixed membrane charges play a role in regulating Cl? permeation through transmitter sensitive membranes. Evidence was also obtained that in snail neurons receptor inactivation increases when external calcium is reduced.  相似文献

γ-Aminobutyric acid (GABA), a known inhibitory neurotransmitter in mammals, can elicit two different types of excitatory response in the nervous system of the marine mollusc, Aplysia. These responses are depolarizing when GABA is applied ionophoretically, and result from either an increase in membrane conductance to Na+ or a decrease in conductance to K+. In addition, GABA on other neurons causes an inhibitory response similar to that commonly found in other preparations. Although not all neurons have GABA receptors, identified single cells consistently have the same type of response. These observations suggest the possibility that GABA may function in at least some preparations as an excitatory neurotransmitter in addition to its documented inhibitory function.  相似文献

Isolated olfactory receptor neurons from the squid Lolliguncula brevis respond to betaine, a repellent odorant, with hyperpolarizing receptor potentials. Using perforated-patch techniques, we determined that the hyperpolarizing conductance was selective for Cl and could be reversibly blocked by the Cl channel blockers 4-acetamido-4′-isothio-cyanatistilbene-2,2′disulfonic acid and niflumic acid. Gramicidin-patch recordings revealed that [Cl]i in squid olfactory receptor neurons is normally very low compared to vertebrate olfactory receptor neurons, and that activating a Cl conductance would hyperpolarize the cell in vivo. The lack of dependence on internal or external K+ or Na+ ruled out the possibility that the Cl conductance was generated by a cation-dependent cotransporter or pump. Common G-protein-dependent signalling pathways, including phospholipase C, arachidonic acid, and cyclic nucleotides, do not appear to be involved. Ca2+ imaging experiments showed that betaine did not affect [Ca2+]i, suggesting that the Cl current is not Ca2+ dependent. Abstract Radioisotope equilibration techniques have been used to determine the intracellular concentration of K+, Na+ and Cl?, together with the unidirectional ion fluxes across the plasmalemma of Porphyra purpurea. Influx and efflux of 42K+, 24Na+ and 36C1? are biphasic, the rapid, initial uptake and loss of tracer from individual thalli being attributable to desorption from extracellular regions. Cellular fluxes are slower and monophasic, cells discriminating in favour of K+ and Cl? and against Na+. A comparison between the equilibrium potential of individual ion species and the measured membrane potential demonstrates that there is an active component of K+ and Cl? influx and Na+ efflux. ‘Active’ uptake and ‘passive’ loss of K+ and Cl? are reduced when plants are kept in darkness, suggesting that a fraction of the transport of K+ and Cl? may be due to ‘exchange diffusion’ (K+/K+ and Cl?/Cl?antiport).  相似文献

Salt‐affected farmlands are increasingly burdened by chlorides, carbonates, and sulfates of sodium, calcium, and magnesium. Intriguingly, the underlying physiological processes are studied almost always under NaCl stress. Two faba bean cultivars were subjected to low‐ and high‐salt treatments of NaCl, Na2SO4, and KCl. Assimilation rate and leaf water vapor conductance were reduced to approximately 25–30% without biomass reduction after 7 days salt stress, but this did not cause severe carbon shortage. The equimolar treatments of Na+, K+, and Cl? showed comparable accumulation patterns in leaves and roots, except for SO42? which did not accumulate. To gain a detailed understanding of the effects caused by the tested ion combinations, we performed nontargeted gas chromatography–mass spectrometry‐based metabolite profiling. Metabolic responses to various salts were in part highly linearly correlated, but only a few metabolite responses were common to all salts and in both cultivars. At high salt concentrations, only myo‐inositol, allantoin, and glycerophosphoglycerol were highly significantly increased in roots under all tested conditions. We discovered several metabolic responses that were preferentially associated with the presence of Na+, K+, or Cl?. For example, increases of leaf proline and decreases of leaf fumaric acid and malic acid were apparently associated with Cl? accumulation.  相似文献

Summary 1. The effects of bath-applied recombinant human interleukin-1 (rhIL-1) and interleukin-2 (rhIL-2) on the calcitonin (CT)-induced outward current recorded from identified neurons (R9–R12) ofAplysia kurodai were investigated with conventional voltage-clamp and pressure ejection techniques.2. Micropressure ejection of CT onto the soma of the neuron induced a slow outward current [I o(CT); 4–6 nA in amplitude, 30–40 sec in duration] associated with a decrease in input membrane conductance.3.I o(CT) was increased by hyperpolarization.4. The extrapolated reversal potential was +10 mV. Additionally,I o(CT) was sensitive to changes in (Na+)o but not to changes in (K+)o, (Ca2+)o, and (Cl)o.5. Micropressure-ejected forskolin produced a slow outward current similar to that induced by CT.6. Bath-applied rhIL-1 and rhIL-2 (10–40 U/ml) reduced the CT-induced current in identifiedAplysia neurons without affecting the resting membrane conductance or the holding current.7. The inhibitory effects of both cytokines on the current were completely reversible. Heat-inactivated rhIL-1 and rhIL-2 were without effect.8. These results suggest that the immunomodulators, IL-1 and IL-2, can modulate the CT-induced outward current associated with a decrease in Na+ conductance in the nervous system ofAplysia. Summary This paper reports experiments designed to assess the relations between net salt absorption and transcellular routes for ion conductance in single mouse medullary thick ascending limbs of Henle microperfusedin vitro. The experimental data indicate that ADH significantly increased the transepithelial electrical conductance, and that this conductance increase could be rationalized in terms of transcellular conductance changes. A minimal estimate (G c min ) of the transcellular conductance, estimated from Ba++ blockade of apical membrane K+ channels, indicated thatG c min was approximately 30–40% of the measured transepithelial conductance. In apical membranes, K+ was the major conductive species; and ADH increased the magnitude of a Ba++-sensitive K+ conductance under conditions where net Cl absorption was nearly abolished. In basolateral membranes, ADH increased the magnitude of a Cl conductance; this ADH-dependent increase in basal Cl conductance depended on a simultaneous hormone-dependent increase in the rate of net Cl absorption. Cl removal from luminal solutions had no detectable effect onG e , and net Cl absorption was reduced at luminal K+ concentrations less than 5mm; thus apical Cl entry may have been a Na+,K+,2Cl cotransport process having a negligible conductance. The net rate of K+ secretion was approximately 10% of the net rate of Cl absorption, while the chemical rate of net Cl absorption was virtually equal to the equivalent short-circuit current. Thus net Cl absorption was rheogenic; and approximately half of net Na+ absorption could be rationalized in terms of dissipative flux through the paracellular pathway. These findings, coupled with the observation that K+ was the principal conductive species in apical plasma membranes, support the view that the majority of K+ efflux from cell to lumen through the Ba++-sensitive apical K+ conductance pathway was recycled into cells by Na+,K+,2Cl cotransport.  相似文献

The absorbance change of the weak base dye probe, Acridine orange, was used to monitor alterations of pH gradients across renal brush border membrane vesicles. The presence of Na+/H+ or Li+/H+ exchange was demonstrated by diluting Na2SO4 or Li2SO4 loaded vesicles into Na+- or Li+-free solutions, which caused dye uptake. About 20% of the uptake was abolished by lipid permeable cations such as valinomycin-K+ or tetraphenylphosphonium, indicating perhaps the presence of a finite Na+ conductance smaller than electroneutral Na+/H+ exchange. The protonophore tetrachlorosalicylanilide raised the rate of dye uptake under these conditions, hence the presence of an Na+ conductance greater than the H+ conductance was suggested. K+ gradients also induced changes of pH, at about 10% of the Na+ or Li+ rate. Partial inhibition (21%) was seen with 0.1 mM amiloride indicating that K+ was a low affinity substrate for the Na+/H+ exchange. Acceleration both by tetrachlorosalicylanilide (2-fold) and valinomycin (4-fold) suggested the presence of 2 classes of vesicles, those with high and those with low K+ conductance. The larger magnitude of the valinomycin dependent signal suggested that 75% of the vesicles had a low K+ conductance. Inward Cl? gradients also induced acidification, partially inhibited by the presence of tetraphenylphosphonium, and accelerated by tetrachlorosalicylanilide. Thus both a Cl? conductance greater than the H+ conductance and a Cl?/OH? exchange were present. The rate of Na+/H+ exchange was amiloride sensitive with a pH optimum of 6.5 and an apparent Km for Na+ or Li+ of about 10 mM and an EA of 14.3 kcal per mol. A 61-fold Na2SO4 gradient resulted in a pH gradient of 1.64 units which increased to 1.8 with gramicidin. An equivalent NaCl gradient gave a much lower ΔpH even in the presence of gramicidin showing that the H+ and Cl? pathways could alter the effects of the Na+/H+ exchange.  相似文献

Illumination of an Aplysia giant neuron evokes a membrane hyperpolarization which is associated with a membrane conductance increase of 15%. The light response is best elicited at 490 nM: the neuron also has an absorption peak at this wavelength. At the resting potential (-50 to -60 mV) illumination evokes an outward current in a voltage-clamped cell. This current reverses sign very close to EK calculated from direct measurements of internal and external K+ activity. Increases in external K+ concentration shift the reversal potential of the light-evoked response by the same amount as the change in EK. Decreases in external Na+ or Cl- do not affect the response. Therefore, the response is attributed to an increase in K+ conductance. Pressure injection of Ca2+ into this neuron also hyperpolarizes the cell membrane. This effect is also due largely to an increase in K+ conductance. The light response after Ca2+ injection does not appear to be altered. Pressure injection of EGTA abolished or greatly reduced the light response. The effect was reversible. We suggest that light acts upon a single pigment in this neuron, releasing Ca2+ which in turn increases K+ conductance, thereby hyperpolarizing the neuronal membrane.  相似文献

Transport of 86Rb+/K+, 22Na+, 36Cl?, and [3H]indole acetic acid (IAA) has been studied on suspension-cultured cells of the parsley, Petroselinum crispum (Mill) Nym. By compartmental analysis two intracellular compartments of K+, Na+, and Cl? have been identified and ascribed to the cytoplasm and vacuole; half-times of exchange were around 200 s and 5 h, respectively. According to the Ussing-Teorell flux equation, active transport is required for the influx into the cytoplasm at the plasmalemma (K+, Cl?) and the tonoplast (K+, Na+, Cl?). The plasmalemma permeability pattern, PK:PNa:PCl=1.00:0.24:0.38, features an increased chloride permeability compared with cells from higher plant tissues. IAA uptake showed an exponential timecourse, was half-maximal after 10 min, and a linear function of the IAA concentration from 10?9 to 10?5 M. IAA and 2,4-dichlorophenoxy acetic acid reduce the apparent influx of K+, Na+, Cl? during the initial 30 min after addition and subsequently accelerate both in- and efflux of these ions. We discuss that auxins could affect the ion fluxes in a complex way, e.g. by protonophorous activity and by control of the hypothetical proton pump.  相似文献

In the dispersed acinar cells of the submucosal nasal gland in the guinea pig, intracellular Na+ concentration ([Na+]i) was measured with a microfluorimetric imaging method and the cytosolic indicator dye, sodium-binding benzofuran isophthalate, under HCO3?-free conditions. In the unstimulated condition, the [Na+]i was averaged to 12.8 ± 5.2 mM. Addition of 100 μM ouabain or removal of external K+ caused an increase in [Na+]i. Replacement of external Cl? with NO3? or addition of 0.5 mM furosemide reversibly decreased the [Na+]i. The recovery process from the reduced [Na+]i was inhibited by removal of either K+ or Cl? in the bath solution. These findings indicate the presence of a continuous influx of Na+ coupled with K+ and Cl? movement. Application of acetylcholine (ACh, 1 μM) caused an increase in [Na+]i by about 15–20 mM, which was completely inhibited by addition of 10 μM atropine. Increased cytosolic Na+ induced by ACh was extruded by the Na+-K+ pump. Removal of external Cl? and addition of 50 μM dimethylamiloride inhibited ACh-induced increase in [Na+]i by about 66% and 19%, respectively. In both unstimulated and stimulated state, Na+-K+ pump, Na-K-Cl cotransport, and Na+-H+ exchange play a critical role in maintaining intracellular electrolyte environment and in controlling a continuous secretion of nasal fluids. © 1995 Wiley-Liss, Inc.  相似文献

We have previously reported on the biochemical properties of a Na+,K+,2Cl?-cotransport in HeLa cells and here we deal with aspects of its physiological regulation. Na+,K+,2Cl?-cotransport in HeLa cells was studied by 86Rb+ influx and 86Rb+/22Na+ efflux measurements. The effects of rat atrial natriuretic peptide (ANP), isoproterenol, and amino acids on 86Rb+ flux, mediated by the bumet-anide-sensitive Na+, K+, 2Cl?-cotransport system and the ouabain-sensitive Na+/K+-pump, were investigated. ANP reduced bumetanide-sensitive 86Rb+ influx under isotonic as well as under hypertonic conditions. Similar decrease of bumetanide-sensitive 86Rb+ influx was observed in the presence of 8-bromo-cGMP, while neither isoproterenol as a β-receptor agonist nor 8-bromo-cAMP-could alter bumetanide-sensitive 86Rb+ influx. Furthermore, efflux of 86Rb+ and 22Na+ was greatly reduced in the presence of bumetanide and ANP. Together with our recent findings, showing functionally active, high affinity receptors for ANP on HeLa cells (Kort and Koch, Biochim. Biophys. Res. Commun. 168:148–154, 1990), this study indicates that ANP participates in the regulation of the Na+, K+, 2Cl?-cotransport system in HeLa cells. Further measurements revealed that amino acids as present in the growth medium (Joklik's minimal essential medium) and the amino acid derivative α-methyl-aminoisobutyric acid (metAlB, 1 and 5 mM, respectively) also reduced Na+, K+, 2Cl?-cotransport-mediated 86Rb+ uptake and diminished the stimulatory effect of hypertonicity on the cotransporter. In addition, the Na+/K+-pump was markedly stimulated in the presence of amino acids, while neither ANP and 8-Br-cGMP nor isoproterenol and 8-Br-cAMP had a significant effect on the activity of the Na+/K+-pump.  相似文献

The kinetics of the light-driven Cl? uptake pump of Synechococcus R-2 (PCC 7942) were investigated. The kinetics of Cl? uptake were measured in BG-11 medium (pHo, 7·5; [K+]o, 0·35 mol m?3; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 mol m?3) or modified media based on the above. Net36Cl? fluxes (?Cl?o,i) followed Michaelis-Menten kinetics and were stimulated by Na+ [18 mol m?3 Na+ BG-11 ?Cl?max= 3·29±0·60 (49) nmol m?2 s?1 versus Na+-free BG-11 ?Cl?max= 1·02±0·13 (54) nmol m?2 s?1] but the Km was not significantly different in the presence or absence of Na+ at pHo 10; the Km was lower, but not affected by the presence or absence of Na+ [Km = 22·3±3·54 (20) mmol m?3]. Na+ is a non-competitive activator of net ?Cl?o,i. High [K+]o (18 mol m?3) did not stimulate net ?Cl?o,i or change the Km in Na+-free medium. High [K+]o (18 mol m?3) added to Na+ BG-11 medium decreased net ?Cl?o,i [18 mol m?3K+ BG-11; ?Cl?max= 2·50±0·32 (20) nmol m?2 s?1 versus BG-11 medium; ?Cl?max= 3·35±0·56 (20) nmol m?2 s?1] but did not affect the Km 55·8±8·100 (40) mmol m?3]. Na+-stimulation of net ?Cl?o,i followed Michaelis-Menten kinetics up to 2–5 mol m?3 [Na+]o but higher concentrations were inhibitory. The Km for Na+-stimulation of net ?Cl?o,i [K1/2(Na+)] was different at 47 mmol m?3 [Cl?]o (K1/2[Na+] = 123±27 (37) mmol m?3]. Li+ was only about one-third as effective as Na+ in stimulating Cl? uptake but the activation constant was similar [K1/2(Li+) = 88±46 (16) mmol m?3]. Br? was a competitive inhibitor of Cl? uptake. The inhibition constant (Ki) was not significantly different in the presence and absence of Na+. The overall Ki was 297±23 (45) mmol m?3. The discrimination ratio of Cl? over Br? (δCl?/δBr?) was 6·38±0·92 (df = 147). Synechococcus has a single Na+-stimulated Cl? pump because the Km of the Cl? transporter and its discrimination between Cl? and Br? are not significantly different in the presence and absence of Na+. The effect of changing [K+], [Na+] and [Cl?] in nutrient solution was studied in bullfrog antrum with and without HCO3? in nutrient. In 25 mM HCO3? (95% O2/5% CO2) and in zero HCO3? (100% O2), nutrient pH was maintained at 7.3. Changing from 4 to 40 mM K+ or from 81 to 8.1 mM Cl? gave a decrease 10 min later in transmucosal PD (nutrient became more negative) — a normal response. These responses were less in zero than in 25 mM HCO3?. A decrease from 102 to 8 mM Na+ decreased PD (anomalous response of electrogenic NaCl symport). This effect was attenuated or eliminated in zero HCO3?. In contrast, change from 4 to 40 mM K+ gave initial anomalous PD response and change from 102 to 8 mM Na+, initial normal PD response with either zero or 25 mM HCO3?. Both responses were associated with (Na+ + K+)-ATPase pump and were greater in zero than in 25 mM HCO3?. Initial PD increases in zero HCO3? are explained as due to increase in the resistance of passive conductance and/or NaCl symport pathways. Thus, removal of HCO3? modifies conductance pathways of nutrient membrane.  相似文献

The neurotransmitters mediating the synaptic interactions in the pyloric system of the stomatogastric ganglion of a stomatopod, Squilla oratoria, were examined. Putative transmitters were applied iontophoretically to the pyloric cells. Glutamate and GABA produced inhibitory responses in all motoneurons but acetylcholine did not. These inhibitory responses were due to increases in conductance to either K+ or Cl or both, and blocked by picrotoxin. The inhibitory postsynaptic potentials evoked by the constrictor and dilator neurons were different in their time courses, reversal potentials, ion selectivities, and picrotoxin sensitivities. Glutamate is a transmitter candidate for inhibitory synapses made among the pyloric cells as well as for their neuromuscular junctions. In some cells, glutamate and acetylcholine evoked excitatory responses which were blocked by joro spider toxin and by tubocurare, respectively. They mediated the extrinsic inputs to modulate the pyloric rhythm. The transmitter, glutamate, is conserved in the ganglion neurons between stomatopods and decapods during evolution. Use of two transmitters, glutamate and acetylcholine, may have evolved in decapods, while the ionic mechanism is preserved in both orders. The neuromodulators, acetylcholine and -aminobutyric acid, are conserved between both orders. Alfalfa (Medicago sativa L.) is a moderately salt-tolerant plant. This study was conducted to evaluate responses of two contrasting alfalfa genotypes (OMA-84-salt sensitive and OMA-285-salt-tolerant) to components (Na+, and/or Cl?) of salt stress. Alfalfa genotypes were exposed to Na+???salts (without chloride), Cl????salts (without sodium), and NaCl (sodium chloride) stresses with two concentrations (30 and 150 mM). The treatments, involving macronutrients, with the same osmotic potentials, were taken as control. Salt stress, irrespective of type and intensity, caused a significant reduction in plant biomass, physiological (net photosynthetic rate, photosystem II efficiency, chlorophyll fluorescence, water use efficiency, maximum yield of primary photochemistry, and electron transport rate), and shoot mineral (calcium, magnesium, phosphorus, and potassium) contents compared to control; however, this reduction was in the order of NaCl (150 mM)?>?Na+ (150 mM)?>?Cl? (150 mM). The alfalfa genotype OMA-285 sustained growth under both types of salt stresses than the genotype OMA-84 due to less accumulation of Na+ and Cl? ions, maintenance of higher K+/Na+ ratio, and better photosynthetic activities. In conclusion, salt stress caused a significant reduction in alfalfa growth, this reduction was more under NaCl stress and the effect was mainly additive. The alfalfa genotype OMA-285 sustained growth under salt stresses than the genotype OMA-84 due to ionic homeostasis. However, the tested genotypes were more sensitive to Na+ toxicity than the Cl? toxicity, and the contrasting genotypes differed in tissue tolerance of high Na+ and Cl?. Further research is needed to evaluate tissue tolerance in a diverse and large group of alfalfa genotypes to elucidate the general salt tolerance mechanism in alfalfa.

The stimulation of H+ extrusion by hyper-osmotic stress (0.2–0.3 M mannitol) in cultured cells of Arabidopsis thaliana (L.) Heynh. was shown to be associated with an inhibition of Cl? efflux, whereas hypo-osmotic stress, inhibiting H+ extrusion, early and strongly stimulated Cl? efflux. In this paper, we investigate the contribution of other factors [K+ transport and transmembrane electric potential difference (Em)] to the hyper-osmotic-induced activation of the plasma membrane (PM) H+-ATPase. The effects of mannitol (MA) on K+ transport and on Em were compared with those of fusicoccin (FC) since the modes of action of osmotica and of the toxin in stimulating H+-ATPase activity seem to differ at least in some steps. The changes in H+ extrusion induced by hyper- or hypo-osmotic stress were opposite and could be reversed by the application of the respective opposite stress. The effect of MA on H+ extrusion was dependent on the presence of K+ (or Rb+) similarly to that of FC, while Na+ and Li+, which also stimulated the FC effect, were ineffective on that of MA. The MA effect was independent of the anions (Cl?, SO42?, NO3?) accompanying K+. K+ net uptake and K+ influx were stimulated by both MA and FC. Tetraethylammonium (TEA+) and Cs+ inhibited both MA- and FC-induced H+ extrusion, suggesting the involvement of K+ channels. MA (0.2 M) induced a strong hyperpolarization of Em both in the absence and in the presence of K+. The hyperpolarizing effect of MA was also found when the cells were already hyperpolarized by FC, and was rapidly reversed by removing the osmoticum from the medium. In the presence of the lipophilic cation tributylbenzylammonium (TBBA+), MA was no longer able to stimulate H+ extrusion, while FC still stimulated it. In cells pretreated with TBBA+, which strongly depolarized Em, the subsequent addition of FC repolarized it, while the hyperpolarizing effect of MA was lacking. On the contrary, in cells pretreated with Erythrosine B (EB), Em was strongly depolarized and the following addition of FC did not hyperpolarize it, while the hyperpolarizing effect of MA was still observed. These results suggest that the mechanism of MA in activating H+ extrusion and K+ uptake is different from that of FC. The rise in net K+ uptake seems to be driven by the activation of some hyperpolarizing system that does not seem to depend on a direct activation of PM H+-ATPase, but rather on the inhibition of Cl? efflux induced by hyper-osmotic stress.  相似文献

Salinity tolerance in wild (Glendale) and hatchery (Quinsam) pink salmon Oncorhynchus gorbuscha (average mass 0·2 g) was assessed by measuring whole body [Na+] and [Cl?] after 24 or 72 h exposures to fresh water (FW) and 33, 66 or 100% sea water (SW). Gill Na+, K+‐ATPase activity was measured following exposure to FW and 100% SW and increased significantly in both populations after a 24 h exposure to 100% SW. Whole body [Na+] and whole body [Cl?] increased significantly in both populations after 24 h in 33, 66 and 100% SW, where whole body [Cl?] differed significantly between Quinsam and Glendale populations. Extending the seawater exposure to 72 h resulted in no further increases in whole body [Na+] and whole body [Cl?] at any salinity, but there was more variability among the responses of the two populations. Per cent whole body water (c. 81%) was maintained in all groups of fish regardless of salinity exposure or population, indicating that the increase in whole body ion levels may have been related to maintaining water balance as no mortality was observed in this study. Thus, both wild and hatchery juvenile O. gorbuscha tolerated abrupt salinity changes, which triggered an increase in gill Na+, K+‐ATPase within 24 h. These results are discussed in terms of the preparedness of emerging O. gorbuscha for the marine phase of their life cycle.  相似文献

ACh-induced depolarization (D response) in D cells markedly decreases as the external Na+ is reduced. However, when Na+ is completely replaced with Mg++, the D response remains unchanged. When Na+ is replaced with Tris(hydroxymethyl)aminomethane, the D response completely disappears, except for a slight decrease in membrane resistance. ACh-induced hyperpolarization (H response) in H cells is markedly depressed as the external Cl- is reduced. Frequently, the reversal of the H response; i.e., depolarization, is observed during perfusion with Cl--free media. In cells which show both D and H responses superimposed, it was possible to separate these responses from each other by perfusing the cells with either Na+-free or Cl--free Ringer's solution. High [K+]0 often caused a marked hyperpolarization in either D or H cells. This is due to the primary effect of high [K+]0 on the presynaptic inhibitory fibers. The removal of this inhibitory afferent interference by applying Nembutal readily disclosed the predicted K+ depolarization. In perfusates containing normal [Na+]0, the effects of Ca++ and Mg++ on the activities of postsynaptic membrane were minimal, supporting the current theory that the effects of these ions on the synaptic transmission are mainly presynaptic. The possible mechanism of the hyperpolarization produced by simultaneous perfusion with both high [K+]0 and ACh in certain H cells is explained quantitatively under the assumption that ACh induces exclusively an increase in Cl- permeability of the H membrane.  相似文献
