The bacteria of genera Pseudomonas oxidizing maleic, fumaric, italic acids and their anhydrides in sewage of industry of maleic anhydride have been isolated from active sludge. During the autoselection a strain of P. stutzeri 832 capable to oxidize 12.5 - 25.0 g/l of maleic acid on 90 - 70% correspondingly in 3 days has been isolated. Destructive activity of isolated strain was retained after 5-year storage in lyophylized state and under vaseline oil.  相似文献

好氧甲烷氧化菌是一群以甲烷为碳源和能源的细菌。好氧甲烷氧化菌在自然环境中分布广泛,人类已从土壤、淡水和海洋沉积、泥炭沼泽、热泉、海水和南极环境分离到甲烷氧化菌的纯培养。好氧甲烷氧化菌可分为14个属,包括研究较为深入的隶属于变形菌门Alpha和Gamma纲的细菌,以及属于疣微菌门的极端嗜热嗜酸甲烷氧化菌。最近,好氧甲烷氧化菌还被发现存在于苔藓类植物(尤其是泥炭苔藓)共生体中,兼性营养好氧甲烷氧化菌也被发现。本文通过对好氧甲烷氧化菌的分类、生理生化特征、分子生物学检测方法以及微生物生态学中的研究成果的总结与分析,以及对甲烷氧化菌研究所面临的问题进行讨论,以期为今后进一步开展好氧甲烷氧化菌及其在碳循环中的作用研究提供参考。  相似文献

Methanol oxidation was studied in several RuMP and serine type methylotrophic bacteria. On the basis of the distribution of the dissimilatory enzymes and the electrophoretic mobility of the methanol dehydrogenases, the methanol and methane oxidizers of the RuMP type belong to two different taxonomic groups. The pink pigmented facultative serine type methylotrophs represent another taxon.  相似文献

The efficacy of disrupting cells of methane oxidizing bacteria M. trichosporium strain 20 by air decompression was measured. The efficacy increased with a rise of the gas pressure, density of the bacterial suspension and the ratio of the air and suspension volumes. The relationship between the efficacy of disruption of the microorganisms and the rate of pressure reduction showed a distinct maximum. The efficacy of the bacterial disruption did not change with an increase of the time of saturation of the bacterial suspension by air from 30 sec. to 60 min.  相似文献

Recently discovered microorganisms affiliated to the bacterial phylum NC10, named "Candidatus Methylomirabilis oxyfera", perform nitrite-dependent anaerobic methane oxidation. These microorganisms could be important players in a novel way of anaerobic wastewater treatment where ammonium and residual dissolved methane might be removed at the expense of nitrate or nitrite. To find suitable inocula for reactor startup, ten selected wastewater treatment plants (WWTPs) located in The Netherlands were screened for the endogenous presence of M. oxyfera using molecular diagnostic methods. We could identify NC10 bacteria with 98% similarity to M. oxyfera in nine out of ten WWTPs tested. Sludge from one selected WWTP was used to start a new enrichment culture of NC10 bacteria. This enrichment was monitored using specific pmoA primers and M. oxyfera cells were visualized with fluorescence oligonucleotide probes. After 112 days, the enrichment consumed up to 0.4 mM NO(2)(-) per day. The results of this study show that appropriate sources of biomass, enrichment strategies, and diagnostic tools existed to start and monitor pilot scale tests for the implementation of nitrite-dependent methane oxidation in wastewater treatment at ambient temperature.  相似文献

好氧甲烷氧化菌能以甲烷作为碳源和能源,对全球甲烷消除的贡献率高达10%–20%,还能有效地合成有价值的甲烷来源生物产品。文中介绍了好氧甲烷氧化菌的甲烷氧化代谢机理,总结了好氧甲烷氧化菌在填埋场甲烷减排、煤矿通风气治理、合成生物产品、油气藏勘探等领域的实际应用功效和研究热点,即污染物去除和产品合成效率的影响因素。基于对甲烷氧化菌规模化培养方法的研究,本文认为加强培养过程中代谢产物对甲烷氧化菌活性和种群结构影响的研究,以及开发经济高效的替代培养基和培养技术的研究将有利于好氧甲烷氧化菌生物技术的应用推广。  相似文献

Numbers of heterotrophic, methane oxidizing and sulfate reducing bacteria were counted in Lake Vechten. A dynamic distribution pattern was found in the stratified lake. A maximum of heterotrophs (numbers of 109 bact./l) occured in the deepest part of the lake in spring and in the metalimnion during summer-stratification. These bacteria use nearly all available oxygen in the hypolimnion. It was found that the concentration of available organic material and the oxygen tension caused the numbers of heterotrophs in the metalimnion to be high.The maximal numbers of methane oxidizers (numbers of 5.105 bact./l) were found at a depth of maximal methane concentration: the de-oxygenated hypolimnion. Preliminary evidence indicated that these organisms were facultative methane oxidizers and must be regarded as micro-aerophyllics. By oxidizing methane they removed the residual oxygen under the metalimnion.The sulfate reducing bacteria could be observed in the hypolimnion only. Decreased SO inf4 sup–2 concentration and increased numbers, of bacterai were found in the bottom water. An association between the methane oxidizers and the sulfate reducers could be deduced. It was assumed that favourable redox requirements for obligate anaerobic sulfate reducers were the results of the activities of the methane oxidizing bacteria.The dynamic distribution equilibrium of the investigated groups of bacteria was disturbed by the autumn turn-over. The heterotrophic and methane oxidizing bacteria decreased in number at that period and were equally distributed, no sulfate reducers could be detected in the free water of Lake Vechten.  相似文献

【背景】反硝化厌氧甲烷氧化(Denitrifying anaerobic methane oxidation,DAMO)是以硝酸盐或亚硝酸盐为电子受体以甲烷为电子供体的厌氧氧化过程,对认识全球碳氮循环、削减温室气体排放和开发废水脱氮新技术等方面具有重要意义。【目的】认识以硝酸盐和亚硝酸盐为电子受体的DAMO微生物富集过程和结果的差异性。【方法】在序批式反应器(Sequencing batch reaetor,SBR)内接种混合物,分别以硝酸盐和亚硝酸盐为电子受体连续培养800 d,定期检测反应器基质浓度变化、计算转化速率;利用16S rRNA基因系统发育分析研究功能微生物的多样性,利用实时荧光定量PCR技术定量测定功能微生物。【结果】以亚硝酸盐为电子受体的1、3号反应器富集到了DAMO细菌,未检测到DAMO古菌;以硝酸盐为电子受体的2号反应器富集到了DAMO细菌和古菌的混合物;3个反应器的脱氮速率经过初始低速期、快速提升期,最终达到稳定,但2号快速提升期开始时间比1、3号晚了80 d左右,达到稳定的时间更长,稳定最大速率为1、3号的44.7%、40.3%。【结论】硝酸盐和亚硝酸盐对富集产物有决定性影响;以硝酸盐为电子受体富集得到的DAMO古菌和细菌协同体系可以长期稳定共存,DAMO古菌可能是协同体系中脱氮速率的限制性因素。  相似文献

Methane oxidation coupled to denitrification is mediated by 'Candidatus Methylomirabilis oxyfera', which belongs to the candidate phylum NC10. The distribution of putative denitrifying methane-oxidizing bacteria related to "M. oxyfera" was investigated in a freshwater lake, Lake Biwa, Japan. In the surface layer of the sediment from a profundal site, a phylotype closely related to "M. oxyfera" was most frequently detected among NC10 bacteria in PCR analysis of the 16S rRNA gene. In the sediment, sequences related to "M. oxyfera" were also detected in a pmoA gene library. The presence of NC10 bacteria was also confirmed by catalyzed reporter deposition fluorescence in situ hybridization (CARD-FISH). Denaturing gradient gel electrophoresis (DGGE) and quantitative real-time PCR indicated that the abundance of the "M. oxyfera"-related phylotype was higher in the upper layers of the profundal sediment. The horizontal distribution of the putative methanotrophs in lake sediment was also analyzed by DGGE, which revealed that their occurrence was restricted to deep water areas. These results agreed with those in a previous study of another freshwater lake, and suggested that the upper layer of the profundal sediments is the main habitat for denitrifying methanotrophs.  相似文献
