The Nonhuman Primate Pregnancy Test may be useful for diagnosis of pregnancy in squirrel monkeys between 40 and 60 days in pregnancy. However, single determinations have an inherent 10% risk of false negative responses caused by low chorionic gonadotropin levels; thus, initial negative test responses should be followed within one week by an independent confirmatory test. Preliminary results with this hemagglutination inhibition test compare favorably with bioassay and may be useful, in conjunction with conventional uterine palpation, for diagnosis of pregnancy in squirrel monkeys.  相似文献

Levels of estradiol and progesterone in peripheral serum of seven cynomolgus monkeys (Macaca fascicularis) were measured from about the 30th day of pregnancy until parturition. Although the pattern of each steroid in circulation differed somewhat from the respective patterns in rhesus monkeys (Macaca mulatta), there were basic similarities. On the day of delivery, the corpus luteum was excised and in vitro incubation of dispersed luteal cells was performed. Isolated luteal cells produced progesterone under control conditions and responded to the addition of HCG with enhanced steroidogenesis. Accordingly, "rejuvenation" of the corpus luteum may occur during advanced gestation in cynomolgus monkeys. These findings, along with establishing the efficacy of the Subhuman Primate Pregnancy Test kit to diagnose pregnancy in this macaque, extend previous evidence for utility of cynomolgus monkeys as a primate model for study of steroid hormones in pregnancy.  相似文献

The Nonhuman Primate Pregnancy Test (NPPT) kit and the Gravindex Slide Test (GST) were compared for the diagnosis of pregnancy in an orangutan. The NPPT was found to consistently detect chorionic gonadotropin earlier and longer than the GST. During approximately the last trimester of pregnancy, both tests registered negative results. It was concluded that the NPPT is a simple, effective pregnancy test for breeding age orangutans.  相似文献

The performance of The Subhuman Primate Pregnancy Test Kit was evaluated for routine detection of early (days 19-21) pregnancy in the rhesus monkey. Out of 123 confirmed matings, 19 resulted in pregnancy. In the pregnant animals the kit had an accuracy of 73.7%. In the nonpregnant females the accuracy was higher, 88.5%. False positives were encountered in ovariectomized females as well as adult intact males.  相似文献

The Nonhuman Primate Pregnancy Test (NHPPT) kit was evaluated for diagnosis of pregnancy in baboons. Unreadable controls rendered 33% of all tests inconclusive. Refrigeration and dilution did little to improve the number of inconclusive tests. Positive-and false-negative results from readable tests on days 20 to 27 of confirmed pregnancy were 75% and 25%, respectively. The NHPPT did not always provide an accurate diagnosis of pregnancy and often proved unreadable.  相似文献

Abstract: The reproduction performance of captive owl monkeys, a breed used extensively in biomedical research, was observed at the Battelle Primate Facility (BPF). The colony grew through captive breeding, imports from the Peruvian Primatological Project, and others to a peak size of 730. It included seven karyotypes of Aotus sp. Results showed that owl monkeys can breed successfully in a laboratory in numbers sufficient to sustain modest research programs. Reproductive success increases when pairs are compatible, of the same karyotype, and stabilized; however, mated pairs of different karyotype are also productive. Under conditions of controlled lighting and heating, owl monkeys at BPF showed no birth peak nor birth season.  相似文献

Administration of human chorionic gonadotropin to pregnant bonnet monkeys(Macaca radiata) at 55–60 days and 130–140 days of pregnancy resulted in a significant increase in serum progesterone levels. This effect could be observed even in lutectomized monkeys. However, no significant change in the serum estrogen level was noticed. These results suggest that although no chorionic gonadotropin is detectable in the serum after 35 days of pregnancy, the foetoplacental steroidogenic system is still responsive to exogenous gonadotropic stimulation.  相似文献

An attempt has been made in this paper to review our present understanding of luteal function during the periimplantation period and in particular hormonal requirement for implantation and maintenance of early pregnancy in the non-human primate.In a fertile cycle thecorpus luteum is apparently rescued from luteolysis by chorionic gonadotropin secreted by the implanted blastocyst, In the bonnet monkey the serum progesterone titers during the luteal phase of a fertile cycle seems higher compared to that of nonmated cycling monkeys. This suggested that thecorpus luteum is receiving some stimulatory signal from the blastocyst even prior to implantation. The recent demonstration that human blastocyst in culture secretes into the medium human chorionic gonadotropin essentially support the above assumption. However, attempts to extend the luteal phase of cycling unmated monkeys with exogenous human chorionic gonadotropin injection has hitherto not met with complete success suggesting that there could be other than chorionic gonadotropin, additional luteal stimulatory factors the unimplanted blastocyst is secreting.Corpus luteum is the principle source of both progesterone and estrogen produced during the periimplantation period and dysruption of luteal function, brought about by either lutectomy or ovariectomy or luteinizing hormone antiserum treatment, followed by progesterone supplementation leads to maintenance of pregnancy. This has lead to questioning the need for estrogen in the maintenance of early pregnancy. Recent work using Zuclomiphene, an antiestrogen during days 5–11 of cycle in rhesus monkeys mated between day 9–14, has however, suggested that estrogen may be required for implantation. Further work is needed to arrive at an unequivocal decision regarding the need for estrogen in maintenance of early pregnancy in the primate.  相似文献

We have established the interval over which urinary chorionic gonadotropin can be detected by a radioimmunoassay during pregnancy in the golden lion tamarin. Preliminary findings indicate the potential value of this radioimmunoassay system for (1) diagnosis of pregnancy at about four weeks after fertilization; (2) estimation of the expected time of delivery; and (3) identification of individual monkeys having an apparent high risk of spontaneous abortion.  相似文献

Hormonal profiles during postpartum estrus, time of conception, and pregnancy were determined in urine samples from six cotton-top tamarins (Saguinus oedipus oedipus). Noninvasive collection techniques permitted daily sampling throughout lactation and pregnancy. Urinary estrone (E1), estradiol (E2), and both bioactive and immunoreactive luteinizing hormone/chorionic gonadotropin (LH/CG) measures revealed an interval of 19 ± 2.07 (S E M) days between parturition and the postpartum ovulatory LH peak. An increase in both E1 and E2 was seen prior to the LH peak; however, E1 and E2 continued to increase to their highest concentrations after the LH peak. Since postpartum ovulations resulted in pregnancy, neither postpartum estrus nor conception was suppressed by lactation. The length of gestation (measured from the LH peak to parturition) was 183.7 ± 1.14 (S E M) days, which is at least 30 days longer than that previously reported for other callitrichid species. Both E1 and E2 reached their maximum levels during midpregnancy but showed a rapid decline at parturition. Gestational levels of CG were first detectable approximately 20 days after the LH peak and continued to be elevated for approximately 80 days. The Sub-Human Primate Tube Test (SHPTT) for pregnancy did not detect the LH Peak and was less sensitive than other methods in detecting CG. Two RIA methods and a bioassay technique could not distinguish between LH and CG. We concluded that monitoring both estrogen and LH concentration was needed to determine when ovulation occurs in the cotton-top tamarin, since peak values of estrogen are seen after the ovulatory LH peak. Also, these tamarins were pregnant the majority of the time, indicating an unusually high fertility rate in contrast to most noncallitrichid primate species.  相似文献

Serum levels of estradiol (E2), progesterone (P), prolactin (Prl), monkey chorionic gonadotropin (mCG), dehydroepiandrosterone sulfate (DHEAS), and cortisol (C) in pregnant cynomolgus monkeys are described. mCG, E2, and Prl patterns resembled those of pregnant rhesus monkeys. P patterns differed from data from the literature. DHEAS patterns did not follow E2 patterns. C levels did not change during pregnancy.  相似文献

Serum testosterone concentrations have been determined during the menstrual cycle and early pregnancy in the bonnet monkey, Macace rediata. During the cycle, there is an increase around the time of ovulation and a secondary peak in the late luteal phase. In pregnancy, there is a distinct peak around 23-25 days, a period which corresponds to the peak of chorionic gonadotropin reported by Atkinson et al. (1975) in Rhesus monkeys. Administration of exogenous hCG causes a significant rise in the serum testosterone level in cycling monkeys.  相似文献

Background  This case report describes the treatment of three male owl monkeys ( Aotus nancymaae ) diagnosed with chronic dry eye with a topical cyclosporine product, Restasis®, approved for use in humans. These owl monkeys had ocular disease resulting from procedures performed at a biotechnology company. They were moved to the Center for Neotropical Primate Research and Resources at University of South Alabama to be incorporated into the breeding colony.
Materials and methods  Schirmer tear testing was performed initially and during the course of treatment to monitor efficacy of twice daily administered Restasis®. The goals of treatment were to reduce pain and/or distress and if possible to quantitatively increase tear production.
Results and discussion  All animals had improvements in conjunctival inflammation and had an increase in tear production.  相似文献

Gonadotropins are routinely administered to produce multiple oocytes for clinical in vitro fertilization (IVF) treatment, laboratory research, and livestock industries. Studies in mice have shown gonadotropin stimulation using equine chorionic gonadotropin (eCG) affects the endometrium, implantation, and fetal development. Evidence from clinical studies also indicates that stimulation with recombinant human follicle-stimulating hormone (rhFSH) may be detrimental to the endometrium and implantation rates. We investigated the effect of rhFSH in mice on maternal plasma hormone concentrations and uterine gene and protein expression and the effect of a stimulated maternal environment on pregnancy. Adult females were stimulated with rhFSH or eCG, followed by human chorionic gonadotropin (hCG). On day 4 of pseudopregnancy, mice either had embryos transferred to the uterus or were killed, and blood and uterine samples were collected. Pregnancy outcomes were examined on day 15. Gonadotropin stimulation increased plasma progesterone concentrations on day 4 compared with controls, whereas estradiol concentrations were unaffected. Stimulation also reduced uterine leukemia inhibitory factor (Lif) mRNA, but the expression of estrogen and progesterone receptors (Esr1 and Pgr), homeobox gene Hoxa10, and Vegf mRNA were unchanged. Furthermore, distribution of uterine PGR protein expression was altered by stimulation, but LIF protein was unchanged. Stimulated embryo transfer recipients had lower pregnancy rates than controls, and fetuses from the rhFSH group had reduced weight, length, and maturity. These results demonstrate that gonadotropin stimulation with rhFSH or eCG alters the preimplantation maternal environment, which results in reduced pregnancy rates and fetal development in the mouse.  相似文献

The radioreceptorassay developed to determine serum luteinizing hormone level in the cynomolgus monkey was evaluated for its usefulness in early pregnancy diagnosis by the detection of serum chorionic gonadotropin (CG). Blood samples were collected at weekly intervals from the 1st to the 5th week after conception to determine changes in circulating levels of CG. In the pregnancy cases, serum CG levels increased to above 50 μg/ml in almost all animals. By the determination of CG 3 weeks after conception, 86% of all pregnant cases exhibited a positive response. Cases that were negative 3 weeks after conception were followed by a repeated test in the next week. According to this test schedule, 95% of the pregnant cases were detected by 4 weeks after conception, and 5% were undetected as negative responses because their CG levels were low.  相似文献

A practical, noninstrumented enzyme-linked immunosorbant assay (NELISA) for the measurement of urinary monkey chorionic gonadotropin (mCG) has been developed for the detection of early pregnancy in macaque monkeys for use in both the laboratory and the field. Five rhesus monkeys (Macaca mulatta) and six crab-eating monkeys (Macaca fascicularis) were tested for the presence of mCG in urine on gestational days (GDs) 12 to 35. The mCG NELISA detected pregnancy as early as GD 14, with an average earliest detection at GD 16.5 +/- 1.4 (n = 11). Out of 90 tests, 27 false-negative and zero false-positive tests were obtained, for an accuracy of 70.0%. Without the aid of a spectrophotometer, the presence of mCG in pregnant monkey samples was indicated by a dark green color change. Nonpregnant monkey urine samples, on the other hand, exhibited no color change. These findings suggest that the simple, economical, and reliable urinary mCG NELISA may be useful for diagnosing early pregnancy in these and related species. Because capture and restraint are unnecessary for collecting urine samples, the mCG NELISA has widespread potential for confined and free-ranging animals.  相似文献

The ovulatory cycle and pregnancy of the endangered Grevy’s zebra (Equus grevyi) was characterized by analyzing excretion patterns of fecal estradiol and progesterone. Pregnancy was further described by assay of urinary equine chorionic gonadotropin. Courtship and mating behavior were similar to that seen in the domestic horse, but cycle length was longer, ranging from 28 to 35 days. Steroid hormone profiles during pregnancy were likewise similar to the horse, but gestation length was longer (391 and 406 days) in the two mature mares. A young mare was first seen to mate when 1 year 3 months of age, at which time she conceived but then aborted at 101 days’ gestation. Conception occurred after one and possibly two more abortions. A healthy foal was born after a gestation period of 425 days, when the mare was 3 years 4 months old, the youngest age at first foaling to be reported for this species. Equine chorionic gonadotropin (eCG) was first detected between 35 and 40 days’ gestation, as in the horse, returning to baseline at about 195 days. However, in one young mare, eCG was also detectable around the time of ovulation. The eCG antibody may have cross‐reacted with luteinizing hormone, which would have been reaching peak concentrations at about that time. The general agreement of the endocrine results with observed occurrence of mating behavior and of parturition demonstrates that assay of excreted hormones can be used to document puberty, monitor ovarian cycles, and diagnose and monitor pregnancy in Grevy’s zebra mares. A radioreceptorassay (RRA) for macaque luteinizing hormone (LH)/chorionic gonadotropin (CG) was adapted from the clinical RRA for human LH/CG, Biocept-G?, for the purposes of detection of pregnancy prior to day 20 of gestation and for estimation of the time of ovulation in macaques. The 90-min assay procedure was simple, accurate, and reliable. Seventy-five rhesus monkeys (Macaca mulatta) and 20 crab-eating monkeys (Macaca fascicularis) were tested for the presence of CG in the serum on estimated days 17–20 of pregnancy. Of a total of 160 tests, four false negative and 0 false positive tests were obtained, for an accuracy of 97.5%. The preovulatory LH peak was detected in 19 rhesus monkeys by semiquantitative RRA of LH/CG. Ovulation was confirmed in these 19 animals by the presence of a fresh corpus luteum at laparotomy 2–10 days after ovulation, collection of an embryo, pregnancy, or subsequent cycle history. The short, simple assay procedure and the low inter-and intraassay coefficients of variation (7.3 and 3.7%, respectively) allow use of this assay in an economical, predictive, as well as retrospective, capacity for estimation of the time of ovulation in rhesus monkeys. The sensitivity, reliability, species nonspecificity, simplicity, and rapidity of performance of this RRA for LH/CG are features which add up to a useful new management tool for breeding macaques for research purposes.  相似文献

Frequent urine samples from three habituated free-ranging adult female mountain gorillas were collected during a four-week period in order to characterize, by analysis of urinary hormones, their ovarian cycle, and to diagnose pregnancy. A conceptive cycle and two cycles that did not result in full term pregnancy were monitored. Behavioral and physiological observations of mating and labial swellings were correlated to urinary hormone results. Mating occurred on days with elevated estrogens. Labial swelling was maximum near or at peak estrogen levels. Pregnancy was diagnosed at the field site by measurement of chorionic gonadotropin using commercially available human pregnancy detection kits. In one pregnancy, gestation length was 254 days. The pregnanediol glucuronide concentrations of the mountain gorilla did not change during the nonconceptive cycle. During the conceptive cycle, the concentrations did not increase until the approximate time of implantation. This finding suggests marked differences in steroid production or metabolism between the mountain gorilla and the captive lowland gorilla.  相似文献

The effect of super-ovulatory dose of pregnant mare serum gonadotropin and human chorionic gonadotropin on ovulation, advancement of ovulation, subsequent embryo development and implantation were studied in the hamster. Groups of hamsters received pregnant mare serum gonadotropin injection on day 1 of the estrous cycle followed by human chorionic gonadotropin injection either at 56 or 76 h later, pregnant mare serum gonadotropin alone on day 1 or human chorionic gonadotropin alone on day 3.The combination therapy (pregnant mare serum gonadotropin and human chorionic gonadotropin) resulted in super-ovulation (an average of 40 mature ova/animal) while human chorionic gonadotropin alone yielded an average of 10 mature ova/animal. Ovulation was advanced by 24 h by giving human chorionic gonadotropin at 56 h instead of 76 h after pregnant mare serum gonadotropin. Subsequent embryo development and implantation occurring under different hormonal regimens were studied. The ova obtained by giving human chorionic gonadotropin injection at 56 h were poorly fertilizablein vivo and hence the pregnancy rate was low (6 %). These ova however, were fertilizablein vitro, suggesting that the low fertilization rate and developmental failure may be due to inhibition of sperm capacitation/transport because of premature human chorionic gonadotropin administration. In the group receiving human chorionic gonadotropin alone on day 3 there was fertilization and cleavage, but no implantation occurred due to failure of functional corpora lutea. However, administration of progesterone and estrone from day 2 of gestation resulted in 80% implantation and sustenance of pregnancy. On the other hand, the pregnant mare serum gonadotropin and human chorionic gonadotropin combination therapy resulted in super-pregnancy. The number of fetuses present at term was higher in the group receiving pregnant mare serum gonadotropin alone than in the group receiving the combination therapy. Embryo resorption however was higher (37%) in the latter group compared with the former (9.5%). However, preimplantation embryos were found to be viable as evidenced by fluorescein diacetate staining.  相似文献
