Isolated root cultures which could be maintained over severalmonths by serial subculture were established from Brassica oleraceavar. italica cv. Green Comet F1. A modified White's medium wasfound to be the best of several salt compositions tested. Theeffects on isolated root growth of the following were also examined;nutritional components, culture vessel type and closure, pHof the medium and auxin type and concentration. Using a mediumdevised for Green Comet, root cultures were established fromsix other B. oleracea, B. napus and B. campestris cultivars. It was possible to regenerate shoots from segments of culturedroots by incubation on agar-solidified media containing cytokininand auxin. The effects on regeneration of various auxins andcytokinins were investigated; the combination of Picloram withKN gave the highest frequency of shoot formation. It was demonstratedthat isolated roots retained their regenerative ability overa period of 5 months in culture. Thylakoid vesicles were purified from spinach (Spinacea oleracea L. ) leaves by sucrose density gradient centrifugation and incorporated into planar lipid bilayers by stirring. At least one type of voltage-dependent K+ single-channel currents was found. Its conductance (between +60 mV and –60 mV ) was about 55 pS in symmetrical (cis: trans) 250 mmol/L KC1. This channel was sensitive to TEA (tetraethylammonium chloride) and the permeability ratio (PK+/PCl-) was about 14. 9. The selectivity of 55 pS channel determined from both reversal potentials under bi-ionic conditions or from conductance measurements in symmetrical solutions, was in the seguence of K+〉Na+〉Li+ 〉NH4+〉 Cs+. This potassium channel could act as involved in charge-balancing during light-driven proton uptake by thylakoid.  相似文献

Metal and metalloid resistances in plant species and genotypes/accessions are becoming increasingly better understood at the molecular and physiological level. Much of the recent focus into metal resistances has been on hyperaccumulators as these are excellent systems to study resistances due to their very abnormal metal(loid) physiology and because of their biotechnological potential. Advances into the mechanistic basis of metal(loid) resistances have been made through the investigation of metal(loid) transporters, the construction of mutants with altered metal(loid) transport and metabolism, a better understanding of the genetic basis of resistance and hyperaccumulation and investigations into the role of metal(loid) ion chelators. 以羽衣甘蓝10个品种的游离小孢子培养,研究其胚状体及其再生植株诱导方法的结果表明,琼脂糖和活性炭对诱导胚状体发生及发育有促进作用;改良MS培养基中添加0.01%的活性炭可促进植株再生;确定1/2MS NAA 0.1 mg·L-1是优化生根的培养基;小孢子再生植株成活率可达74.6%.  相似文献

Ca2+ and Mg2+ content of cellulose fibres is of relevance for a wide range of applications e.g. textile processing, pulp/paper, food. Sorption of Ca2+ and Mg2+ ions were found on lyocell type regenerated cellulose fibres. Higher affinity was found for Ca2+ ions compared to Mg2+ ions. At pH 9, fibre saturation was observed at a calcium binding capacity of 18–20 mmol/kg. A carboxylic group content of 18 mmol COOH per kg fibre material was determined based on the Methylene Blue absorption. This indicates a 1:1 molar stoichiometry between the carboxylic groups present in the fibres and the bound Ca2+ ions. Thus it is proposed that the salt in fibre shows the general composition (Cell-O? Ca2+ X?), X? being an anion bound in the salt to achieve charge neutrality.The sorption of Ca2+ also can be demonstrated by complex formation with 1,2-dihydroxy-9,10-anthraquinone (alizarin) which forms a red-violet Ca2+-complex. Colour fixation thus can be used as an indicator for the Ca2+-ions bound in the fibre.  相似文献

通过观察镁离子对紫外线辐射损伤后细胞活性与光产物产量的影响,以明确镁离子对UVB辐射细胞损伤的防护作用.在实验中,选择成人成纤维细胞(HDF-a)和角质形成细胞(HaCaT)进行细胞培养,筛选小剂量UVB辐射进行诱导性损伤,观察不同浓度镁离子(0.1 mmol/L,0.2 mmol/L,0.4 mmol/L,0.8 mmol/L,1.2 mmol/L)对辐射损伤的细胞活性、基因组断裂情况及光产物产量的影响.研究结果显示,在UVB辐射后,两个细胞系细胞活性显著下降,镁离子培养液孵育的细胞活性均比UVB辐射损伤对照组显著升高,而镁离子孵育后细胞光产物嘧啶二聚体(CPDs)量明显减少.实验研究证实,镁离子对UVB辐射损伤的培养细胞具有防护作用,为UV辐射所致的DNA损伤的修复提供理论基础.  相似文献

Nutrient resorption from senescing tissues increases plant nutrient-use efficiency, and may be an adaptation to nutrient limitation. In some tree species, retranslocation of nutrients from sapwood during heartwood formation is a comparable process. We measured Ca, Mg and K concentrations in Atlantic white cedar (Chamaecyparis thyoides) stemwood samples taken from two swamps in the northeastern United States and compared them to soil mineral nutrient availability at each site. We found that Ca, Mg and K concentrations were 60–700% higher in sapwood than in the immediately adjacent heartwood, indicating retranslocation of these nutrients from senescing sapwood. Sapwood nutrient concentrations were similar between the two sites. However, nutrient concentrations in the heartwood differed significantly between the sites, as did the relative degree of Ca and Mg retranslocation from senescing sapwood. We found these differences between sites to be inversely related to significant differences in exchangeable Ca, Mg and K as well as Al concentrations in the soil. These findings suggest that the degree of nutrient retranslocation from senescing sapwood may be influenced by soil nutrient availability. Salt tolerant spinach (Spinacia oleracea) and salt sensitive pea (Pisum sativum) plants were exposed to mild salinity under identical growth conditions. In order to compare the ability of the two species for extra- and intracellular solute compartmentation in leaves, various solutes were determined in intercellular washing fluids and in aqueously isolated intact chloroplasts. In pea plants exposed to 100 millimolar NaCl for 14 days, apoplastic salt concentrations in leaflets increased continuously with time up to 204 (Cl) and 87 millimolar (Na+), whereas the two ions reached a steady concentration of only 13 and 7 millimolar, respectively, in spinach leaves. In isolated intact chloroplasts from both species, sodium concentrations were not much different, but chloride concentrations were significantly higher in pea than in spinach. Together with data from whole leaf extracts, these measurements permitted an estimation of apoplastic, cytoplasmic, and vacuolar solute concentrations. Sodium and chloride concentration gradients across the tonoplast were rather similar in both species, but spinach was able to maintain much steeper sodium gradients across the plasmamembrane compared with peas. Between day 12 and day 17, concentrations of other inorganic ions in the pea leaf apoplast increased abruptly, indicating the onset of cell disintegration. It is concluded that the differential salt sensitivity of pea and spinach cannot be traced back to a single plant performance. Major differences appear to be the inability of pea to control salt accumulation in the shoot, to maintain steep ion gradients across the leaf cell plasmalemma, and to synthesize compatible solutes. Perhaps less important is a lower selectivity of pea for K+/Na+ and NO3/Cl uptake by roots.  相似文献

The aim of the study was to measure with microbeam PIXE elements such as Na, K, Mg, and Ca in cardiac tissue after various treatments in vivo, which affect the cardiomyocyte integrity. It was assumed that local deviations from normal electrolyte levels indicate the degree of cardiac damage. The first step in this feasibility study was comparison of severely damaged cardiac tissues with controls. Severe cardiac damage was introduced by the so-called Ca paradox. Experiments were performed with isolated rat hearts, perfused retrogradely with an oxygenated crystalloid buffer. Results indicated that severe cardiac damage was accompanied with almost complete disappearance of the normal intracellular electrolyte composition as a result of the loss of membrane integrity. Identifications of smaller and more locally present ischemic damages on basis of altered electrolyte levels appeared to be feasible. However, the prerequisite was that the mobility of electrolytes be kept under control during tissue sampling and sample preparation, when physiological mechanisms stop to maintain gradients.  相似文献

Spinach (Spinacia oleracea L. cv. Clermont) leaves grown in open-top chambers and exposed to three different concentrations of ozone were measured for gas exchange, chlorophyll a fluorescence, antioxidant systems, and lipid peroxidation at the end of growing season. High O3 concentration reduced Fv/Fm, indicating that the efficiency in the energy conversion of photosystem 2 (PS2) was altered. The rate of non-cyclic electron transport rate and the capacity to reduce the quinone pool were also affected. The development of non-photochemical quenching was not high enough to decrease the photon excess in the PS2. The limitation of photosynthetic activity was probably correlated with stomata closure and with an increase in intercellular CO2 concentration. Under oxidative stress, superoxide dismutase (SOD) activity was stimulated in parallel with lipid peroxidation. We did not find any differences in the ascorbate (AsA) pool and ascorbate peroxidase (APX) or glutathione reductase (GR) activities between air qualities. Small, but similar responses were observed in spinach leaves exposed to ambient ozone concentration.  相似文献

为获得芸薹属白菜Brassica campestris与青花菜Brassica oleracea var. botrytis的种间体细胞杂交体,以青花菜和白菜的子叶与下胚轴为材料,分离制备原生质体,用40%聚乙二醇 (Polyethylene glycol,PEG) 进行原生质体融合。融合细胞在以0.3 mol/L 蔗糖、0.3 mol/L葡萄糖为渗透稳定剂,附加0.2 mg/L 2,4-D+0.5 mg/L 6-苄氨基嘌呤 (6-BA) +0.1 mg/L 1-萘乙酸 (NAA) +0.1 mg/L激动素 (Kinetin,Kin) 的改良K8p培养基中液体浅层培养。将包埋于0.1%琼脂糖的8~10个细胞期的细胞在添加0.3 mol/L蔗糖和2 mg/L 6-BA+2 mg/L玉米素 (Zeatin,ZEA) +1 mg/L NAA+0.5 mg/L Kin的Kao培养基中诱导愈伤组织。愈伤组织转到MS+5 mg/L ZEA+2 mg/L IAA诱导不定芽。将长1~2 cm的不定芽转到1/2 MS+0.2 mg/L NAA诱导生根。将生根的植株转移到花盆,并对其杂种性质进行形态学、细胞学和分子生物学鉴定。结果表明,融合细胞培养2~7 d后发生第1次分裂,培养35 d后植板率为0.66%,不定芽再生率达3.7%。形态学观察显示,绝大多数再生植株的叶面积较大,株型和叶型为两种杂交亲本的中间型。染色体计数结果显示,再生植株染色体数目为2n=38。流式细胞仪测定DNA含量显示,再生植株DNA含量是亲本之和。随机扩增多态性DNA (Random amplified polymorphic DNA,RAPD) 和基因组原位杂交 (Genomic in situ hybridization,GISH) 分析结果证明再生植株具有双亲基因组。体细胞杂种花粉育性比较低,杂交、回交后其育性逐渐获得恢复。  相似文献

Summary Rice and peanut plants were grown in nutrient solution with varying concentrations of phosphorus, potassium, calcium, and magnesium. Growth response and concentration of the elements in the plant tops were recorded and from these critical and adequate values for P, K, Ca, and Mg were defined. These were for P at 25 days of growth of rice plants 0.70–0.80 and 0.80–0.86; at 50 days of growth 0.18–0.26 and 0.26–0.40; and at 75 days 0.26–0.36 and 0.36–0.48 per cent of dry matter respectively. For K they were 3.75–4.25 and 4.25–4.35 per cent at 25 days of growth; 3.7–4.0 and 4.0–4.62 per cent at 50 days of growth; and 3.5–3.62 and 3.62–3.99 per cent at 75 days of growth resp. At 100 days of growth the values for Ca and Mg were established as 0.36–0.45, 0.45–0.65; and 0.12–0.17, 0.17–0.30 per cent respectively. Summary The effects of A1 on the growth and mineral composition of different cultivars of triticale (X Triticosecale, Wittmack), wheat (Triticum aestivum L.) and rye (Secale cereale L.) growing in 1/5 strength Steinberg solutions containing 0 or 6 ppm A1 were evaluated after 32 days. Aluminum increased the concentrations of P and K in the roots and K in the tops of most of the cultivars tested. A1 tolerant triticale retained a lower concentration of Mg in the roots and tops than the A1 sensitive triticale, when subjected to A1 stress. In addition, A1 treatments resulted in smaller increases in root P for the A1 tolerant triticale than for the A1 sensitive cultivars.The concentration of root Ca and P of the A1 tolerant wheat cultivars were significantly below that of the more sensitive plants. Aluminum tolerance in rye appeared to be associated with lower Ca and higher Mg concentrations in the tops. The accumulation of P and A1 in the roots was characteristic of sensitivity in triticale, wheat and rye.  相似文献

芸薹属(Brassica)植物(甘蓝型油菜,白菜,甘蓝)是重要的经济作物,它们之间的关系可用禹氏三角表示.CAMTA是在进化过程中高度保守的一类转录因子家族,该家族成员在芸薹属植物抗逆境胁迫过程中具有重要作用.为了探讨芸薹属CAMTA3基因家族的功能,本研究利用生物信息学手段,鉴定了芸薹属(白菜,甘蓝和甘蓝型油菜)CAMTA3基因家族的8个基因,并从蛋白质的理化性质、家族进化与基因结构和上游启动子等方面进行了分析研究.分析表明,芸薹属CAMTA3基因家族8个成员在进化过程中具有较高的保守性,其亲缘关系与禹氏三角理论相符,且各成员在植物响应抗胁迫过程中占重要地位.研究结果为芸薹属CAMTA3基因家族的功能研究提供了一定的信息基础.  相似文献

Nutrient imbalances of declining sugar maple (Acer saccharum Marsh.) stands in southeastern Quebec have been associated with high exchangeable Mg levels in soils relative to soil K and Ca. A greenhouse experiment was set up to test the hypothesis that the equilibrium between soil exchangeable K, Ca, and Mg ions influences the growth and nutrient status of sugar maple seedlings. Also tested was whether endomycorrhization can alter nutrient acquisition under various soil exchangeable basic cations ratios. Treatments consisted of seven ratios of soil exchangeable K, Ca, and Mg making up a total base saturation of 58%, and a soil inoculation treatment with the endomycorrhizal fungus Glomus versiforme (control and inoculated), in a complete factorial design. Sugar maple seedlings were grown for 3 months in the treated soils. Plant shoot elongation rate, dry biomass and nutrient concentrations in foliage were influenced by the various ratios of soil cations. The predicted plant biomass and foliar K concentration were highest at a soil Ca saturation of 38%, a soil K saturation of 12%, and a soil Mg saturation of 8%. Potassium concentration in foliage was dependent on the level of Ca and Mg saturation in the soil when soil K saturation was close to 12%. Foliar Ca and Mg levels were more dependent on their corresponding levels in soil than foliar K. Colonization by G. versiforme did not influence seedling growth and macronutrient uptake. The results confirm that growth and nutrition of sugar maple are negatively affected by imbalances in exchangeable basic cations in soils.  相似文献

研究了以西兰花叶为原料提取叶绿素制备叶绿素铜钠盐的工艺条件。通过正交试验对西兰花叶叶绿素提取工艺进行了优化,结果表明西兰花叶叶绿素提取的最佳条件为:乙醇浓度95%;料液比1∶6;萃取温度为70℃;萃取时间2 h;叶绿素铜钠盐制备条件为:5%NaOH,65℃皂化30 min;1%硫酸调pH 2.0,65℃酸化30 min;15%硫酸铜,65℃铜代反应60 min,成盐NaOH调pH 12,65℃1 h。  相似文献
