Summary In Coprinus radiatus, a cross homozygous for nic-2 mutation yields two types of nic-strains. The first one (nic-2F) retains the character of the original mutant, reverting at meiosis with a frequency of 20 to 30%. All crosses with the second type (nic-2 fa) show a sharply reduced reversion rate.The analysis of the progeny of crosses involving a nic-2 fa strain show the appearance of a third type of strains (nic-2 fn). The reversion of nic-2 fn strain is low, but this character is not imposed to the partner in a cross with a nic-2F strain.  相似文献

Chlorate-resistant mutants ofS. typhimurium LT2 and LT7 and ofS. abony have been isolated, which are deficient in the biosynthesis of nicotinic acid and thiamin and in the fermentation of inositol. These mutants could be divided into 5 groups. The most likely gene order isnicB-chlG-thiB-inlB. This segment is transferred early in conjugation experiments with Hfr H2 and Hfr B2 as donors. In time-of-entry experiments with Hfr B2 as donor the segment entered about 3 minutes afterpur C. Consequently this segment maps in the 79- to 82-minutes region of the genetic map. From recombinant analysis of nic+ recombinants obtained in a four-point cross between Hfr B2 and ahis carBpur C del (nic chl G) acceptor the incorporation frequency of the transferred donor fragment was calculated to be about 0.41. The number of crossing-over events per minute length of the chromosome was about the same as in similar crosses betweenE. coli Hfr and F. However, between thenic and thepur C markers it was much higher; it may therefore be inferred that there is a higher probability for a crossing-over event in the regions adjacent to the region that is deleted in the recipient.In crosses betweenS. abony Hfr H2 del (nic thi inl chl) and F strains no recombinants were observed which have obtained the deletion from the donor. Nearly all auxotrophic or nic+ recombinants obtained in a cross between Hfr B2 and a F del (nicBthiBchlGinlB) strain have inherited all markers of the donor, which are present in the deletion of the recipient.  相似文献

A chlorophyll b-less mutant of Chlamydomonas reinhardtii (Pg 27) was isolated after UV irradiation of the wild type cells. This photosynthetically competent mutant totally lacks chlorophyll b and the CP2 chlorophyll-protein complex. However, SDS-PAGE, proteolytic digestions and immunodetections demonstrated that the 24–25 Kd apoproteins of the lacking CP2 complex are still present in thylakoids of the Pg27 mutant. It is concluded that this CP2-less mutant is affected in the biosynthesis pathway of chlorophyll b.This CP2-less mutant was crossed with a CP1-less mutant (Fl5) Fluorescence emission spectra and fluorescence inductions in the presence of DCMU were analysed in the resulting (cp 2 , cp 1 + ), (cp 2 + , cp 1 ), (cp 2 + , cp 1 + ), cp 2 , cp 1 )tetratype. Differences in PS 2 optical cross section and in the relative amplitude or localisation of fluorescence emission peaks fit well with a quadripartite model where PS1 and PS2 would each correspond to a reaction centre core complex (CP1 and CP2 respectively) associated to a light harvesting antenna (LHC1 and LHC2 respectively). To investigate the role of ammonium-assimilating enzyme in heterocyst differentiation, pattern formation and nitrogen fixation, MSX-resistant and GS-impaired mutants of Anabaena 7120 were isolated using transposon (Tn5-1063) mutagenesis. Mutant Gs1 and Gs2 (impaired in GS activity) exhibited a similar rate of nitrogenase activity compared to that of the wild type under dinitrogen aerobic conditions in the presence and absence of MSX. Filaments of Gs1 and Gs2 produced heterocysts with an evenly spaced pattern in N2-grown conditions, while addition of MSX altered the interheterocyst spacing pattern in wild type as well as in mutant strains. The wild type showed complete repression of heterocyst development and nitrogen fixation in the presence of NO3 or NH4 +, whereas the mutants Gs1 and Gs2 formed heterocysts and fixed nitrogen in the presence of NO3 and NH4 +. Addition of MSX caused complete inhibition of glutamine synthetase activity in wild type but Gs1 and Gs2 remained unaffected. These results suggest that glutamine but not ammonium is directly involved in regulation of heterocyst differentiation, interheterocyst spacing pattern and nitrogen fixation in Anabaena.  相似文献

The physiological behaviour of Pseudomonas fluorescens strain R2fN was compared to that of transconjugants [R2fN(RP4)], and two aggregation phenotypes were identified (Agr and Agr+). Agr+ phenotype is characterized by the appearance of macroscopic aggregates when cells are growing in liquid media. Transconjugants exhibited Agr+ phenotype whereas wild type strain represented Agr. Evidence is presented to support correlation between Agr+ phenotype acquisition and the presence of the broad-host range plasmid RP4 in strain R2fN. In addition, according to bacterial adherence to hydrocarbon test the transconjugant cells appeared to be very hydrophilic whereas wild type R2fN cells were hydrophobic.  相似文献

Two types of nitrosoguanidine-induced rifampicin-resistant mutants ofNostoc muscorum were isolated and characterized. Compared with the wild type, the strainrif-1 (rif r het + nif + blu) revealed high growth rate, heterocyst frequency (10%–12%), nitrogenase activity, phycocyanin pigment, photosynthetic O2 evolution, and higher activities of phosphoribulokinase and Fd-NADP+-oxidoreductase. The heterocyst spacing pattern in the mutant was altered and did not respond to 7-azatryptophan, -2-thienylalanine, rifampicin, andl-methionine-dl-sulfoximine treatment. The second type of mutantrif r -2 (rif r het + nif ) did not show nitrogenase activity and was unable to grow on molecular nitrogen even under microaerobic conditions, although it produced heterocysts (6%–7%) under these conditions of incubation. The pattern of macromolecular synthesis, particularly of RNA and protein, the rate of acetylene reduction, and photosynthetic O2 evolution were not affected in the mutant strains with the treatment of drug. The characteristics of the mutants reflected the possibility of pleiotropic mutation and also suggested that therif marker is most likely associated in close genetic proximity with regulatory gene(s) ofhet andnif system.  相似文献

Time-correlated single photon counting was used to study energy trapping and detrapping kinetics at 295 K in Rhodobacter sphaeroides chromatophore membranes containing mutant reaction centers. The mutant reaction centers were expressed in a background strain of Rb. sphaeroides which contained only B880 antenna complexes and no B800-850 antenna complexes. The excited state decay times in the isolated reaction centers from these strains were previously shown to vary by roughly 15-fold, from 3.4 to 52 ps, due to differences in the charge separation rates in the different mutants (Allen and Williams (1995) J Bioenerg Biomembr 27: 275–283). In this study, measurements were also performed on wild type Rhodospirillum rubrum and Rb. sphaeroides B880 antenna-only mutant chromatophores for comparison. The emission kinetics in membranes containing mutant reaction centers was complex. The experimental data were analyzed in terms of a kinetic model that involved fast excitation migration between antenna complexes followed by reversible energy transfer to the reaction center and charge separation. Three emission time constants were identified by fitting the data to a sum of exponential decay components. They were assigned to trapping/quenching of antenna excitations by the reaction center, recombination of the P+H charge-separated state of the reaction center reforming an emitting state, and emission from uncoupled antenna pigment-protein complexes. The first varied from 60 to 160 ps, depending on the reaction center mutation; the second was 200–300 ps, and the third was about 700 ps. The observed weak linear dependence of the trapping time on the primary charge separation time, together with the known sub-picosecond exciton migration time within the antenna, supports the concept that it is energy transfer from the antenna to the reaction center, rather than charge separation, that limits the overall energy trapping time in wild type chromatophores. A barley mutant RPr84/90 has been isolated by selecting for plants which grow poorly in natural air, but normally in air enriched to 0.8% CO2. After 5 minutes of photosynthesis in air containing14CO2 this mutant incorporated 26% of the14C carbon into phosphoglycollate, a compound not normally labelled in wild type (cv. Maris Mink) leaves.The activity of phosphoglycollate phosphatase (EC 3.1.1.18) was 1.2 nkat mg–1 protein at 30°C in RPr 84/90 compared to 19.2 nkat mg–1 protein in the wild-type leaves. Phosphoglycollate phosphatase activity was not detected after protein separation by electrophoresis of leaf extracts from the mutant on polyacrylamide gels; on linear 5% acrylamide gels three bands with enzyme activity were separated from extracts of wild type plants. Gradient gel electrophoresis followed by activity staining showed two bands in Maris Mink tracks of MW 86,000 and 96,000, but no bands in 84/90. This is the first report of isozymes of phosphoglycollate phosphatase in barley which were absent in the mutant extracts. Our results confirm an earlier report of isozymes of this phosphatase in Phaseolus vulgaris [18].The photosynthetic rate of RPr 84/90 in 1% O2, 350 l CO2 l–1 was 9–12 mg CO2 dm–2 h–1 at 20°C, whereas the wild-type rate was 27–29 mg CO2 dm–2 h–1 at 20°C. In 21% O2, 350 l CO2 l–1 the rate was 2–3 mg CO2 dm–2 h–1 in the mutant and 20 mg CO2 dm–2 h–1 in the wild type.Genetic analysis has shown that the mutation segregates as a single recessive nuclear gene.  相似文献

Endoxylanases from the thermophilic fungus, Thermomyces lanuginosus ATCC 44008 (cellulase free wild and mutant strains), were purified to homogeneity by anion-exchange and molecular-sieve chromatographic methods. The purified enzymes were monomers with molecular masses of 22 kDa (wild type) and 24 kDa (mutant), estimated by SDS-PAGE and gel filtration. As glycoproteins, the purified enzymes had 0.74% (wild type) and 11.8% (mutant) carbohydrate contents, and pI values of 5.8 and 6, respectively. The optimal pH and temperature values of wild type xylanase were determined to be pH 7 and 60 °C, whereas pH 6.7 and 70 °C, were optimal for the purified mutant enzyme (K m and V max values of 3.7 mg ml–1 and 670 mol min–1 xylose compared to the kinetic values of the purified wild type xylanase –5.1 mg ml–1 and 385 mol min–1 xylose). Inhibition studies suggested the possible involvement of histidine, tryptophan residues and carboxylic groups in the binding or catalysis.  相似文献

Summary Two auxotrophic mutants ofRhizobium trifolii which are deficient in nodulating ability have been isolated. Both mutants (strain RS 164 His and strain RS213 Leu) appear to synthesize abnormal extracellular polysaccharides as compared with the wild type strain RS 55. Summary The membrane currents of wild typeParamecium tetraurelia and the behavioral mutantteaA were analyzed under voltage clamp. TheteaA mutant was shown to have a greatly increased outward current which was blocked completely by the combined use of internally delivered Cs+ and external TEA+. This, along with previous work (Satow, Y., Kung, C., 1976,J. Exp. Biol. 65:51–63) identified this as a K+ current. It was further found to be a calcium-activated K+ current since this increased outward K+ current cannot be elicited when the internal calcium is buffered with injected EGTA. The mutationpwB, which blocks the inward calcium current, also blocks this increased outward K+ current inteaA. This shows that this mutant current is activated by calcium through the normal depolarization-sensitive calcium channel. While tail current decay kinetic analysis showed that the apparent inactivation rates for this calcium-dependent K+ current are the same for mutant and wild type, theteaA current activates extremely rapidly. It is fully activated within 2 msec. This early activation of such a large outward current causes a characteristic reduction in the amplitude of the action potential of theteaA mutant. TheteaA mutation had no effect on any of the other electrophysiological parameters examined. The phenotype of theteaA mutant is therefore a general decrease in responsiveness to depolarizing stimuli because of a rapidly activating calcium-dependent K+ current which prematurely repolarizes the action potential.  相似文献

The proton translocation stoichiometry (H+/ATP ratio) was investigated in membrane vesicles from a Synechocystis 6803 mutant in which the serine at position 37 in the hydrophilic loop of the c-subunit from the wild type was replaced by a negatively charged glutamic acid residue (strain plc37). At this position the c-subunit of chloroplasts and the cyanobacterium Synechococcus 6716 already contains glutamic acid. H+/ATP ratios were determined with active ATP synthase in thermodynamic equilibrium between phosphate potential (G p ) and the proton gradient ( H +) induced by acid–base transition. The mutant displayed a significantly higher H+/ATP ratio than the control strain (wild type with kanamycin resistance) at pH 8 (4.3 vs. 3.3); the higher ratio also being observed in chloroplasts and Synechococcus 6716. Furthermore, the pH dependence of the H+/ATP of strain plc37 resembles that of Synechococcus 6716. When the pH was increased from 7.6 to 8.4, the H+/ATP of the mutant increased from 4.2 to 4.6 whereas in the control strain the ratio decreased from 3.8 to 2.8. Differences in H+/ATP between the mutant and the control strain were confirmed by measuring the light-induced phosphorylation efficiency (P/2e), which changed as expected, i.e., the P/2e ratio in the mutant was significantly less than that in the wild type. The need for more H+ ions used per ATP in the mutant was also reflected by the significantly lower growth rate of the mutant strain. The results are discussed against the background of the present structural and functional models of proton translocation coupled to catalytic activity of the ATP synthase.  相似文献

The parental strain (A+T+) of Saccharomyces cerevisiae and mutants, deficient in catalase T (A+T), catalase A (AT+) or both catalases (AT), grew on ethanol and oleic acid with comparable doubling times. Specific activities of catalase were low in glucose- and ethanol-grown cells. In the two oleic acid-grown A+-strains (A+T+ and A+T) high catalase activities were found; catalase activity invariably remained low in the AT+ strain and was never detected in the AT strain. The levels of -oxidation enzymes in oleic acid-grown cells of the parental and all mutant strains were not significantly different. However, cytochrome C peroxidase activity had increased 8-fold in oleic acid grown A strains (AT+ and AT) compared to parental strain cells. The degree of peroxisomal proliferation was comparable among the different strains. Catalase A was shown to be located in peroxisomes. Catalase T is most probably cytosolic in nature and/or present in the periplasmic space.  相似文献

The last two amino acids of the nascent peptide at the ribosomal P-site influence the efficiency of termination readthrough at the stop codon UGA (Mottagui-Tabar et al (1994) EMBO J 13, 249–257; Björnsson et al (1996) EMBO J 15, 1696–1704). Here we analyze this effect on readthrough by wild type or a UGA suppressor form (Su9) of tRNATrp by varying the codons at positions −1 and −2 at the 5′ side of UGA. Strains with wild-type or mutant (ArBr) forms of elongation factor Tu (EF-Tu) were analyzed (Vijgenboom et al (1985) EMBO J 4, 1049–1052). The effect on readthrough by changing these −1 and −2 codons is different on the two forms of tRNATrp and is also dependent on the structure of EF-Tu. Readthrough by the tRNATrp-derived suppressor, but not wild-type tRNATrp, is sensitive to the van der Waals volume of the last amino acid in the nascent peptide. Together with mutant EF-Tu, both forms of tRNATrp are sensitive. The data suggest that the C-terminal amino acid in the nascent peptide is in a functional interaction with the EF-Tu ternary complex. This interaction is changed by mutation in tRNATrp at position 24 or in EF-Tu at position 375. No indication of a changed interaction between the mutant EF-Tu and the penultimate amino acid could be found. Mutant forms of RF2 (Mikuni et al (1991) Biochimie 73, 1509–1516) and ribosomal proteins S4 and S12 (Fáxen et al (1988) J Bacteriol 170, 3756–3760) were found not to be altered in sensitivity to the last two amino acids in the nascent peptide.  相似文献

In the wild type strain (stock no. 1227) of Thermoactinomyces vulgaris, as reported earlier [Sinha and Singh (1980) Biochem. J. 190, 457–460], all phosphatase isoenzymes (three alkaline — AlpI, AlpII and AlpIII, and one acidic — Acp) are present. However, the auxotrophic mutants, the strains 1286 (thi ), 1279 (nic , ura ) and 1278 (thi , ura ) exhibited two alkaline phosphatase isoenzymes (AlpII and AlpIII), but AlpI was lacking. In the strain 1261 (nic , thi ), only AlpIII was expressed, and AlpI and AlpII isoenzymes were missing. The results suggest that the strains, which require either thiamine (1286 and 1278) or nicotinamide (1279) for their growth, were AlpI mutants; and the strain (1261), which requires both thiamine and nicotinamide for its growth, was AlpI /AlpII double mutant. There was no direct correlation between uracil auxotrophy and the expression of phosphatases. The uniform expression of AlpIII and Acp in all the strains, irrespective of their nutrient requirements, suggest that these constitutive phosphatases are species-specific. The specific activities of the thermophilic acid and alkaline phosphatases were maximum in the wild type strain (1227) of T. vulgaris. The next in phosphatase activity was the strain 1279 (an AlpI mutant), followed by their decrease, in order, in the strains 1286 and 1278 (which were also AlpI mutants); while least activity of these enzymes was observed in the obligate thermophile strain 1261 (AlpI /AlpII double mutant).  相似文献

5-2 is a mutant of Arabidopsis thaliana which is partially resistant to fusicoccin in vivo. We have analysed fusicoccin binding and the activity and amount of H+-ATPase in plasma membrane isolated from mature leaves of the wild type and of mutant 5-2. Fusicoccin binding was similar in plasma membrane from the two genotypes, while H+-ATPase activity was markedly (c. 50%) lower in plasma membrane from mutant 5-2 than in that from the wild type. The H+-ATPase of mutant 5-2 was activated by fusicoccin as much as that of the wild type. In plasma membrane from mutant 5-2, the amount of immunodetectable H+-ATPase, quantified by densitometry of Western blots, was about half that in the wild type. These results indicate that the major defect of mutant 5-2 detectable at the plasma membrane level is a reduction in the amount of H+-ATPase.  相似文献

Summary The short-trichome-forming, non-heterocystous and non-nitrogen-fixing (het nif) mutant of the nitrogen-fixing blue-green alga Nostoc muscorum was isolated by N-methyl-N-nitro-N-nitrosoguanidine (NTG)-mutagenesis after penicillin enrichment technique and characterized. The mutant did not grow and fix nitrogen in combined-nitrogen-free medium while in nitrate-containing medium it grew well (K=0.112/day, G=64.27 h), although its growth was comparatively poor than the parent alga (K=0.128/day; G=56.14 h). The mutant was stable and both the het and nif characters reverted to wild type (het + nif+) with the reversion frequency of 2.62×10-7.The het nif mutant tolerated 0.5 g/ml of streptomycin sulphate on the agar medium and its streptomycin resistant mutant capable of growing in presence of 10g/ml of streptomycin was isolated spontaneously with a frequency of 1.45×10-8. These streptomycin resistant isolates (het nif strR) resisted 100 g/ml of streptomycin sulphate on the agar medium and 200 g/ml in liquid medium. Spontaneous virus-resistant mutant of het nif strR was isolated with a mutation frequency of 4.02×10-4.The data of genetic recombination experiments suggested that there is transfer of both het and nif genes to het nif strain with the frequency of 2×10-6 to 2×10-5 simultaneously. There was increase in recombination frequency with increasing the incubation period. From a pool of Medicago truncatula mutants—obtained by gamma-irradiation or ethyl methanesulfonate mutagenesis—impaired in symbiosis with the N-fixing bacterium Sinorhizobium meliloti, new mutants are described and genetically analysed, and for already reported mutants, complementary data are given on their phenotypic and genetic analysis. Phenotypic data relate to nodulation and mycorrhizal phenotypes. Among the five new mutants, three were classified as [Nod+ Fix Myc+] and the mutations were ascribed to two loci, Mtsym20 (TRV43, TRV54) and Mtsym21 (TRV49). For the two other new mutants, one was classified as [Nod–/+ Myc+] with a mutation ascribed to gene Mtsym15 (TRV48), and the other as [Nod Myc-/+] with a mutation ascribed to gene Mtsym16 (TRV58). Genetic analysis of three previously described mutants has shown that [Nod–/+ Myc+] TR74 mutant can be ascribed to gene Mtsym14, and that [Nod–/+ Myc–/+] TR89 and TRV9 mutants are ascribed to gene Mtsym2 (dmi2). Using a detailed analysis of mycorrhizal phenotype, we have observed a delayed typical arbuscular mycorrhizal formation on some mutants that present thick lens-shaped appressoria. This phenotype was called [Myc–/+] and mutants TR25, TR26, TR89, TRV9, P1 and Y6 were reclassified as [Myc–/+]. Mutant P1 was reclassified as [Nod–/+] because of a late nodulation observed on roots of this mutant.  相似文献

The rate of PSI mediated cyclic electron transport was studied in wild type and mutant cells of Synechocystis sp. PCC 6803 deficient in NDH-1 (M55) or succinate dehydrogenase (SDH) that are responsible for the dark reduction of the plastoquinone pool. Kinetics of P700 photooxidation and P700+ dark reduction in the presence of 5·10−5 M 3-(3,4-dichlorophenyl)-1,1-dimethylurea have been registered as light induced absorbance changes at 810 nm resulting from illumination of cells with 730-nm actinic light for 1 sec. It is shown that in the absence of dehydrogenases the rate of dark reduction of P700+ in both mutants did not decrease but even increased in NDH-1-less mutant cells as compared with the rate in wild type cells. Dibromothymoquinone drastically reduced the rate of P700+ dark reduction both in wild type and in mutant cells. Thus, the cyclic electron transfer from ferredoxin through the plastoquinone pool to P700+, which is independent from dehydrogenases, takes place in all the types of cells. Preillumination of cells of wild type and both mutants for 30 min or anaerobic conditions resulted in delay of P700 photooxidation and acceleration of P700+ dark reduction, while the level of photosynthesis and respiration terminal acceptors (NAD(P)+ and oxygen) decreased. It appears that the rate of P700 photooxidation and P700+ dark reduction in cyclic electron transport in Synechocystis wild type and mutant cells is determined by the level of NADP+ and oxygen in stroma. A possible approach to evaluation of the levels of these acceptors in vivo is proposed, based on kinetic curve parameters of P700 photoconversions induced by 730-nm light with 1-sec duration.  相似文献

Summary To learn more about the ways in which genes silenced by insertion mutations can be reactivated, we have undertaken a systematic investigation of Gal+ revertants of the polar mutant galOP-306::IS1 in Escherichia coli K12. The selective conditions used excluded reversion to wild type by precise excision of IS1. In this system (which resisded on a multi-copy plasmid) reversion to the Gal+ phenotype occurred with a frequency of about 10-7 per cell and per generation. Analysis of the revertants revealed that — with the single exception of the previously published chromosomal mutant sis1 — alterations in the structure of IS1 lead to reactivation of gal operon expression. These events fall into four classes: (I) insertion of IS2 at position 327 in IS1, insertion of IS2 at position 687 in IS1, (III) insertion of a hitherto undetected mobile element, IS150, at position 387, (IV) a 16-bp deletion encompassing IS1 coordinates 553–568. Of some 200 independent reversion events studied, all but one were of types I–III i.e. they involved the intervention of a second mobile element.  相似文献
