【目的】研究黑腹果蝇Drosophila melanogaster对高盐的产卵避性,并通过高盐下后代生存率和发育历期的变化来揭示这一现象的生物学意义。【方法】应用产卵双选择装置,检测黑腹果蝇和雅库巴果蝇D. yakuba雌成虫分别对含有0. 25和1 mol/L NaCl的培养基的产卵选择性;利用产卵装置,检测黑腹果蝇对1 mol/L NaCl的位置效应;通过强迫产卵实验检测钠盐胁迫下黑腹果蝇的产卵能力;通过毛细管摄食法和观察口器延伸,检测钠盐胁迫下黑腹果蝇的摄食行为。利用黑暗条件和突变体Orco2分别探究视觉和嗅觉对高盐的产卵选择性。通过外科手术法摘除黑腹果蝇前足部分味觉感受器,并使用咸味突变体Ionotropic Receptor 76b (IR76b),研究介导该行为的感觉系统。在含有不同浓度NaCl (0,0. 25,0. 50,0. 75和1 mol/L)的培养基上培养黑腹果蝇,探究NaCl对黑腹果蝇后代的发育历期和存活率的影响。【结果】雌性黑腹果蝇产卵对高盐产生避性反应,对0. 25和1 mol/L NaCl的产卵偏嗜指数分别为-0. 45和-0. 59;雅库巴果蝇对高盐的反应与黑腹果蝇相同,对0. 25和1 mol/L NaCl的产卵偏嗜指数分别为-0. 78和-0. 99;并且两种果蝇均对0. 25和1 mol/L KCl和CaCl2产生产卵避性反应。黑腹果蝇避开含0. 25和1 mol/L NaCl培养基,位置指数分别为-0. 74和-0. 88。高盐不影响黑腹果蝇的生育能力,其对0,0. 25和1 mol/L NaCl的产卵指数分别为5. 59,6. 63和5. 41。黑腹果蝇对0. 25和1 mol/L NaCl摄食产生明显排斥反应,摄食指数分别为-0. 37和-0. 41,且随浓度增大食欲越低。同时在黑暗条件下,黑腹果蝇对0. 25和1 mol/L NaCl仍产生了显著的避性反应,产卵偏嗜指数分别为-0. 49和-0. 59,摘除前足的黑腹果蝇对0. 25 mol/L NaCl的产卵偏嗜指数为-0. 05,IR76b突变体对高盐的产卵避性反应消失,对0. 25和1 mol/L NaCl的产卵偏嗜指数分别为0. 46和0. 39。0. 25 mol/L NaCl明显延长了后代成蛹时间0. 40 d,后代幼虫存活率降低了16. 5%,而在1 mol/L NaCl上后代卵完全不能形成蛹和成虫。【结论】黑腹果蝇雌成虫通过咸味感觉器官感知高盐而产生产卵避性反应,以促进后代的生长发育和提高后代的幼虫存活率。  相似文献

【目的】研究黑腹果蝇Drosophila melanogaster对短小乳杆菌Lactobacillus brevis有氧与无氧代谢产物的产卵选择,并解析这些代谢产物影响果蝇产卵行为的机制和生物学意义。【方法】应用产卵双选择装置,检测成年雌果蝇产卵的选择行为;利用黑暗条件(暗盒)、嗅觉突变体和摘除前足味觉感受器的方式探究介导该行为的感觉系统;通过在不同浓度乳酸(0, 70, 280 mmol/L)的培养基上培养黑腹果蝇,探究乳酸对其后代的发育历期和存活率的影响;利用免疫荧光染色法检测乳酸对果蝇肠道上皮细胞增殖情况的影响。【结果】短小乳杆菌无氧代谢产物引起成年雌性黑腹果蝇产卵避性,产卵指数为-0.47;短小乳杆菌有氧代谢产物乳酸含量为0.4 mmol/L,无氧代谢产物乳酸含量为126.8 mmol/L;雌性果蝇对乳酸同样产生产卵避性反应,且产卵避性随乳酸浓度的增高而增强。切除前足的果蝇对乳酸的产卵避性显著降低。与对照相比, 70 mmol/L的乳酸使幼虫的成蛹时间和成虫羽化时间分别延长了1.42 d和1.17 d,存活率降低了36.0%。乳酸明显破坏肠道上皮的完整性,促进肠道上皮细胞的增殖。【结论】短小乳杆菌无氧代谢产物引起成年果蝇产卵避性。果蝇主要通过味觉感知短小乳杆菌无氧代谢产物乳酸,这引起果蝇产卵避性,从而提高后代的生长发育和幼虫的存活率。  相似文献

【目的】研究东方醋杆菌(Acetobacter orientalis)和短小乳杆菌(Lactobacillus brevis)通过氧气浓度调控黑腹果蝇(Drosophila melanogaster)产卵的偏嗜性,并研究其对后代存活率和发育历期的生物学意义。【方法】利用双相选择装置解析果蝇产卵行为。利用分光光度仪和pH计测量细菌的OD值与pH值。通过改变果蝇的视觉、味觉和嗅觉感官以检测介导果蝇产卵行为的感觉系统。用后代的发育历期与存活率评价其生物学意义。【结果】果蝇产卵对东方醋杆菌具有趋避性,产卵指数为–0.76,对短小乳杆菌具有趋向性,产卵指数为0.5,并且二者接种比例决定了果蝇的产卵选择。氧气作为一个调节因子,可改变果蝇对东方醋杆菌和短小乳杆菌的产卵偏嗜性。氧气是东方醋杆菌生长必需的条件,有氧培养的OD值为1.3,而无氧培养的OD值为0.4。氧气是短小乳杆菌生长非必需的条件,有氧和无氧培养的OD值均为2.2。破坏嗅觉可严重地影响果蝇对细菌的产卵偏嗜性。东方醋杆菌和短小乳杆菌均促进了果蝇的生长,分别提高存活率1倍和1.5倍,分别缩短果蝇发育历期1 d和2 d。【结论】东方醋杆菌和短小...  相似文献

对动物体内单个细胞的谱系进行分析有助于追踪其在发育过程中的作用,但是体内各种组织都是由很多形态、结构、功能各不相同的细胞构成的复杂系统,这种复杂性严重阻碍了对单个细胞的研究。嵌合克隆技术(Mosaic technique)和标记技术(Labeling technique)的出现为这一研究提供了强有力的手段。文章介绍了近几年来黑腹果蝇(Drosophila melanogaster)研究中常用的7种嵌合克隆标记方法,包括FRT介导的有丝分裂重组(FRT-mediated mitotic recombination)、MARCM(Mosaic analysis with a repressible cell marker)、TSG(Twin spotgenerator)、Twin-spot MARCM、Q-MARCM(Q system-based MARCM)、Coupled MARCM和G-TRACE(Gal4technique for real-time and clonal expression)技术,详述了这些技术的原理及应用,并对不同技术进行了对比。运用这些技术研究者可以从单细胞水平进行遗传学标记和操作,特别是在神经系统等复杂系统中追踪单个细胞的发育过程。果蝇中的这些技术也将为其他模式生物追踪细胞谱系提供参考。  相似文献

性别的形成包括两个过程,即性别决定和性别分化。果蝇的性别控制研究包括性别决定、性别分化、性别鉴定、性别诱导和性别控制5个方面。性别决定是在两种不同发育途径之间的选择,它提供了一个研究基因调控的模式系统。果蝇的性别决定问题已经研究得相当详细[1]。性别分化是使胚胎向着雌性或雄性发育的过程,决定了性别表型。果蝇的性别分化也取得了不少研究成果。近年来,许多重要的性别调控基因已被克隆和鉴定。随着果蝇基因组全序列测定的完成,果蝇的性别控制研究将会更为深入而完善。本文对与黑腹果蝇性别决定和性别分化相关的一些问题进行综述。  相似文献

【背景】共生菌群可以影响宿主的生理、代谢以及神经行为,黑腹果蝇是研究宿主与共生菌作用机制的优秀遗传模型。【目的】分离和鉴定黑腹果蝇体内的共生菌,并研究其对果蝇生长和发育的影响。【方法】利用YG固体培养基分离果蝇肠道菌;革兰氏染色、生化鉴定和16S rRNA基因比对鉴定菌种;体内定殖和世代传递实验验证细菌与果蝇的共生关系;建立无菌和悉菌模型,通过发育历期和生长速率来验证其促生长作用;RT-PCR检测促生长分子基因表达;免疫荧光染色检测果蝇肠道细胞的增殖;葡萄糖氧化酶法检测葡萄糖浓度。【结果】分离到一株能够明显促进果蝇生长发育的细菌,经鉴定为霍氏肠杆菌(Enterobacter hormaechei),该菌可以稳定定殖于黑腹果蝇肠道以及培养基内,而且可以世代垂直传递。该菌可明显地促进果蝇的生长,并通过加速促胸腺激素的分泌来发挥促生长作用,同时可以促进果蝇肠道细胞的增殖。霍氏肠杆菌相较于无菌果蝇可以降低体内葡萄糖水平。【结论】霍氏肠杆菌是黑腹果蝇的有益共生菌,实验证实其可以促进果蝇的生长和发育。  相似文献

果蝇是近几年发现危害樱桃果实的一类重要害虫,在国内外樱桃产区均有发生。天水地区危害甜樱桃的果蝇有3个种,分别是黑腹果蝇Drosophila melanogaster Meigen、铃木氏果蝇Drosophila suzukii(Matsumura)和海德氏果蝇Drosophila hydei(Sturtevant),黑腹果蝇为优势种。作者记述黑腹果蝇对甜樱桃果实的危害情况、寄主范围及其生活史、生活习性、发育历期与温度的关系等,调查发现蚂蚁是樱桃果蝇的天敌之一。  相似文献

目的 分离和鉴定黑腹果蝇肠道共生微生物,并研究其促进果蝇身体发育的功能。方法 利用Hungate滚管技术,分离厌氧细菌;运用定植实验证明其为果蝇肠道共生菌;用悉菌实验来检测细菌对果蝇发育的影响。结果 本研究从野外捕获的果蝇体内分离到蜂房哈夫尼菌(Hafnia alvei),而且证实它能够在果蝇肠道内有效地定植并且能在培养基中稳定持续地存在,说明蜂房哈夫尼菌是果蝇肠道共生菌。此外,蜂房哈夫尼菌能显著地缩短无菌果蝇的发育周期及提高生长速率。结论 证明了蜂房哈夫尼菌是果蝇肠道的共生菌,并且其可以有效地促进果蝇的生长发育。  相似文献

【目的】黑腹果蝇Drosophila melanogaster肠道中栖生着众多微生物,通过分离和研究其内共生菌,以研究肠道菌群的多态性和作用。【方法】利用Hungate滚管技术从黑腹果蝇成虫肠道分离厌氧细菌;通过记录果蝇的发育历期和生长速率,检测该细菌对果蝇发育和生长的影响。【结果】首次从黑腹果蝇肠道内分离到一株产气荚膜梭菌Clostridium perfringens。该菌能够有效地定植到果蝇肠道内,是果蝇肠道共生菌。产气荚膜梭菌显著地缩短无菌果蝇的发育历期,将无菌果蝇成蛹天数由20 d缩短到8.1 d,羽化天数由30 d缩短到12.7 d。该菌还可以提高果蝇生长速率。【结论】本研究揭示了产气荚膜梭菌是果蝇的内共生菌,可以通过提高生长速率而有效地促进果蝇的生长和发育。  相似文献

Oviposition preference for ureasupplemented food was assayed by simultaneous choice trials on five pairs of closely related laboratory populations of Drosophila melanogaster.Each pair of populations had been derived from a separate ancestral population about 85 generations prior to this study. One population in each pair had been subjected to selection for larval tolerance to the toxic effects of urea; the other population served as a control. Considerable variation in oviposition preference was seen both within and among populations, with four of the ten populations showing a significant mean preference for ureasupplemented food. The degree of specificity shown by individual females was surprisingly high, leading to a bimodal distribution of oviposition preference in some populations. Overall, selection for larval tolerance to urea did not significantly affect oviposition preference. However, the data indicated that pairwise comparisons between randomly selected populations from the two larval selection regimes would lead to a range of possible outcomes, suggesting, in several cases, that selection for larval urea tolerance had led to significant differentiation of adult oviposition preference for urea in one or the other direction. The results, therefore, highlight the importance of population level replication and caution against the practice, common in ecological studies, of assaying oviposition preference in two populations that utilize different hosts in nature, and then drawing broad evolutionary inferences from the results.  相似文献

Summary The term centrophobism is introduced to describe a newly discovered modification of search behavior in the walking fruitfly,Drosophila melanogaster: the avoidance of the center of an arena after diethylether narcosis. Evidence for the effect is obtained by comparison of the tracks of etherized and non-etherized flies under the influence of olfactory attractant around the center of the arena (Fig. 3). The tracks can be distinguished by their mean radial distance from the central district of the arena. Centrophobia denotes the relative difference of the distances of etherized flies and non-etherized controls (Fig. 4).Etherized flies avoid the center of the arena in spite of the attraction of olfactory, thermal or visual cues. The avoidance is significant even in the absence of conspicuous sensory cues for the discrimination of center and surround. The centrophobia obtained in the arena can be used to estimate the efficacy of attractants in the non-etherized control flies (Figs. 6, 7).The lowest possible dose of ether sufficient to elicit narcosis is sufficient to induce centrophobia. None of the other prevalent insect anaesthetics, CO2, N2 and cold, substitutes ether in the present experiments (Figs. 8, 9).Centrophobia arises immediately after ether narcosis. Once induced the effect lasts apparently undiminished for the life time of the flies (Fig. 9).Centrophobia has been found in either sex of the 9 strains tested so far (Fig. 5). Four strains including mutants deficient in wing formation (vestigial) or learning (dunce) show either temporal decline or partial suppression of centrophobia. The anomalous properties are actually due to enhanced spontaneous centrophobism in the non-etherized control groups of these strains (Fig. 10).  相似文献

Summary Drosophila melanogaster is able to perform osmotropotaxis under open-loop conditions. With an optimal stimulus the average turning tendency to the side of higher concentration corresponds to a circular track with radiusr=0.8 cm. The response amplitude does not decrease within 1 or 2 h. Unilaterally antennectomized flies in an homogeneous odor field show a permanent turning tendency towards their intact side.The smallest concentration ratio to elicit osmotropotaxis in normal flies ranges between 610 and 910 at high and between 210 and 510 at an about 50 times lower odor intensity. No negative tropotaxis (i.e. turning to the side of lower concentration) is observed, even with strong repellents.  相似文献

Previous studies have shown that the esterase 6 (EST6) enzyme ofD. melanogaster is mainly produced in the sperm ejaculatory duct of the adult male and comparisons of wild-type males with laboratory null mutants have suggested that the enzyme plays a role in reproductive fitness. In this study we have compared 18 field-derived lines each isoallelic forEst6 for differences in five components of male reproductive fitness. No consistent fitness differences were found among lines differing in respect of the two major allozyme classes EST6-F and EST6-S, despite other evidence that these two classes are not selectively equivalent in the field. However, differences in reproductive fitness were found among lines differing in the minor mobility variants that segregate within EST6-F and EST6-S. A failure to distinguish among these minor forms may explain the discrepancies in previous studies on the effects of the major EST6 allozymes on reproductive fitness. The most significant associations we have found between EST6 and reproductive fitness were due to variation in EST6 activity levels. Male EST6 activity levels were found to be positively correlated with their time to first mating, negatively correlated with the numbers of eggs laid and progeny produced by their mates, and negatively correlated with the frequency with which their mates remate. We conclude that some EST6 variants differ in components of male reproductive fitness operative in laboratory cultures. However, the evidence for fitness differences is stronger for variants affecting the amount, rather than the structure of the enzyme, and the direction of the differences varies between some of the fitness components tested.  相似文献

A temperature sensitive lethal allele of thewingless locus ofDrosophila melanogaster together with previously studied lethal and viable alleles in this locus, has been used to study some properties of this locus. These studies show the existence of two lethal phases for thewingless lesion; one during embryogenesis and another during pupation. By growing embryos with temperature sensitivewingless lesion at the permissive temperature and letting the larvae develop at non-permissive temperature, a large-scale cell death and subsequent regeneration were seen to occur in the mutant wing discs. This cell death followed by regeneration alters the normal developmental potential of the wing disc. Disc transplantation experiments show that these discs are incapable of differentiating into wing blade structures.  相似文献

Summary Using a fully automatized procedure and a training scheme symmetrical with respect to the visual stimuli as well as to the aversive stimulus,Drosophila could be conditioned to blue and yellow lights. With constant test conditions and variation of the light intensities during the training procedure, it could be shown that the flies respond primarily to the colour of the light.We are grateful to Dr. W.
