UV absorption by aflatoxins was monitored in GY agar medium by UV photography. In the UV photographs, aflatoxin-producing molds were identified as gray or black colonies, whereas aflatoxin-nonproducing molds appeared as white colonies. By cellophane transplantation experiments and silica gel thin-layer chromatography, the products absorbing UV light substantially were found to be mainly aflatoxins B1 and G1 excreted from the mold mycelium into the agar medium. UV absorption did not occur when the agar medium contained aflatoxin-noninducible carbon sources instead of glucose. Various inhibitors of aflatoxin production, such as dichlorovos and dimethyl sulfoxide, also decreased the intensity of UV absorption. These results indicate that this technique can be used as a simple, safe, and rapid method of screening aflatoxin-producing molds.  相似文献

Aflatoxin concentrations in agar media were estimated with a direct technique that quantifies the fluorescence of agar containing aflatoxins. Tubes containing 5 ml of an agar medium inoculated with spores of aflatoxin-producing Aspergillus isolates were incubated for 3 days at 30 degrees C and set in a carriage specifically designed to carry culture tubes in a scanning densitometer. Fluorescence (450 nm and above) was elicited in the agar by UV light (365 nm) and photometrically measured. Agar fluorescence directly correlated (r2 = 0.89 +/- 0.05, P less than 0.001) with the concentration of aflatoxin within the range 0 to 18.7 micrograms/g. The lowest aflatoxin concentration detected was 50 ng/g. The technique successfully differentiated the aflatoxigenic potentials of Aspergillus isolates.  相似文献

Thirty seven strains of the Aspergillus flavus group isolated from animal mixed feeds have been screened for their ability to produce aflatoxins in yeast extract and sucrose (YES), aflatoxin producing ability (APA), and coconut agar medium (CAM) media. The concentration and detection of the aflatoxins by different methods is compared. Five known aflatoxin-positive and one aflatoxin-negative strains have been used as controls. Only 5 out of the 37 strains (13.5%) were aflatoxin-producers in YES medium. Of these five strains and the five known aflatoxin-positive strains, only three showed blue fluorescence in APA medium and four in CAM medium. Generally, the aflatoxin concentration in CAM medium was higher than in YES and APA media. Using the agar-plug method and by direct spotting of the YES broth on TLC plates, some aflatoxin-producing strains were not detected.  相似文献

The crystalline compound produced in large quantity in liquid medium by Aspergillus parasiticus UNBF A12, a high aflatoxin-producing strain isolated from the air in the Federal District of Brazil, was identified as kojic acid. The effect of pH on the production of crystalline kojic acid and aflatoxins by the strain was studied. Aflatoxin-producing ability medium was tested for its ability to distinguish aflatoxin-positive from aflatoxin-negative strains of Aspergillus flavus in naturally occurring populations from corn at harvest. All of the aflatoxin-positive strains and some of the aflatoxin-negative strains produced aflatoxins when cultured on cracked corn. Although the data indicate that aflatoxin-producing ability medium is not entirely reliable in distinguishing potential aflatoxin-producing strains of A. flavus from nontoxigenic strains, it is significant that the medium did not yield false-positives.  相似文献

低温大曲是清香型白酒酿造的核心因素之一,为酿造过程提供了物系、酶系和菌系。酵母菌是白酒发酵过程中最重要的功能微生物类群,研究大曲中的酵母菌种类和含量具有重要意义,对大曲质量评价也具有重要参考价值。但用常规酵母菌分离技术和培养基从大曲中分离酵母菌时易受优势丝状真菌(霉菌)的干扰,霉菌常常很快长满培养皿,将酵母菌覆盖,难以对酵母菌进行定量计数、观察和分离纯化。本研究根据酒醅发酵过程中随乙酸和乳酸含量的升高,霉菌含量急剧降低而酵母菌含量逐渐升高的现象,用常规培养基YPD为基础培养基,测试了添加不同量的乙酸、乳酸和丙酸(后者为常用食品防腐和防霉剂),以及不同比例的乙酸乳酸组合物,对低温大曲中霉菌的抑制效果和对酵母菌生长的影响进行探究,发现在灭菌后的YPD中添加3.3mL/L乙酸、2.0mL/L丙酸或在乙酸乳酸1:3的情况下添加2.0mL/L乙酸,30℃培养3-5d之内可有效抑制低温大曲中的霉菌,实现对酵母菌的有效分离、计数和纯化培养,而单加乳酸对霉菌,特别是黄曲霉的抑制效果差。随后测试了以前我们从清香型白酒大曲和酒醅发酵过程中分离的13属18种酵母菌在这些培养基上的生长情况,发现这些酵母菌中的绝大多数,尤其是大曲和酒醅中的优势酵母菌种,均可以在这些加酸培养基上良好生长。综合考虑培养基的成本、配制的简便性及分离效果等因素,本研究推荐将灭菌后添加3.3mL/L乙酸的YPD固体培养基作为低温大曲酵母菌分离和定量计数的优化培养基。  相似文献

A general medium named malt-yeast extract-sucrose agar (MYSA) containing oxgall was designed. The medium was intended for the enumeration and isolation of molds and yeasts in routine examinations of animal feed stuffs. In this study MYSA was tested as a general medium for mycological examination of silage. The medium was compared with dichloran-rose bengal medium (DRBC) in an examination of more than 500 specimens of big bale grass silage. Selected characteristics of known fungal species commonly isolated from feeds were examined after growth on MYSA and DRBC and on malt extract agar, used as a noninhibitory control medium. MYSA suppressed bacterial growth, without affecting the growth of fungi common in feeds. The fungi growing on MYSA were easily recognized, and the medium seemed to slow radial growth of fungal colonies, which permitted, easy counting. The number of species found was higher on MYSA than on DRBC. When we compared MYSA with DRBC for mycological examination of grass silage samples, MYSA was found to be the medium of choice.  相似文献

A new reliable, fast, and simple method for the detection of aflatoxigenic Aspergillus strains, consisting of the addition of a cyclodextrin (a methylated β-cyclodextrin derivative) to common media used for testing mycotoxin production ability, was developed. We propose the use of this compound as an additive for fungal culture media to enhance the natural fluorescence of aflatoxins. The production of aflatoxins coincided with the presence of a bright blue or blue-green fluorescent area surrounding colonies when observed under long-wavelength (365-nm) UV light after 3 days of incubation at 28°C. The presence of aflatoxins was confirmed by extracting the medium with chloroform and examining the extracts by high-pressure liquid chromatography with fluorescence detection.  相似文献

A new distinctive strain of Aspergillus nomius that produces the potent mycotoxins, aflatoxins, is described from pistachio, pecan, and fig orchards in California. Similar to the typical strain of A. nomius (as represented by the ex-type), the O strain produced both B and G aflatoxins but not cyclopiazonic acid, had similar conidial ornamentation, and grew poorly at 42°C. Furthermore, previous published DNA sequence supports that the new strain is very closely related to the ex-type of A. nomius. However, the O strain differs from the ex-type in several morphological characters. The ex-type was initially described as producing “indeterminate sclerotia” that appear as large (up to 3 mm long) elongated sclerotia on surfaces of media. The O strain produces only small spherical sclerotia (mean diameter <0.3 mm) submerged in the medium. In addition, the O strain has predominantly uniseriate conidial heads, whereas the typical strain of A. nomius has predominantly biseriate heads. The O strain colony color on both Czapek solution agar and Czapek yeast extract agar was more yellowish than the ex-type of A. nomius and other common aflatoxin-producing fungi. Isolates of the O strain reported here from several orchards represent the first report of A. nomius in California.  相似文献

All of 33 samples of dried shrimps, shrimp paste, peanut butter and raw groundnut kernels were contaminated with fungi. Aspergillus and Penicillium spp. were the predominant types in dried shrimps and raw groundnut kernels but no Aspergillus spp. were present in peanut butter or shrimp paste samples. Among 81 Aspergillus isolates obtained from dried shrimps and raw groundnut kernels, 10 were A. flavus/A. parasiticus, of which five were potential aflatoxin-producing A. flavus strains. No aflatoxins were detected in the food samples although some were visibly mouldy and some had high mould counts. The occurrence of aflatoxin-producing strains of A. flavus in dried shrimps and raw groundnut kernels warrants further investigation of these foods and their products as potentially significant sources of aflatoxins.  相似文献

Solutions of aflatoxins B1, B2, G1 and G2 and solid films of aflatoxin B1 were irradiated with UV light of wavelength corresponding to the absorption band of aflatoxins at approximately 365 nm (approximately 27 500 cm-1). The reaction was followed spectrophotometrically. Simultaneously the mutagenicity of samples of aflatoxins was determined using Salmonella typhimurium/mammalian microsome test. UV-irradiation caused a modification in all aflatoxins studied (in aqueous and nonaqueous solutions as well as in a solid film); the final photoproducts lost completely the mutagenic activity.  相似文献

AIMS: To find a supplemental ingredient that can be added to routinely used growth media to increase conidial production and decrease aflatoxin biosynthesis in small sclerotial (S strain) isolates of Aspergillus flavus. METHODS AND RESULTS: Molasses was added to three commonly used culture media: coconut agar (CAM), potato dextrose agar (PDA), and vegetable juice agar (V8) and production of conidia, sclerotia, and aflatoxins by A. flavus isolate CA43 was determined. The effect of nitrogen sources in molasses medium (MM) on production of conidia, sclerotia and aflatoxins was examined. Water activity and medium pH were also measured. Conidia harvested from agar plates were counted using a haemocytometer. Sclerotia were weighed after drying at 45 degrees C for 5 days. Aflatoxins B(1) and B(2) were quantified by high-performance liquid chromatography. Addition of molasses to the media did not change water activity or the pH significantly. Supplementing CAM and PDA with molasses increased conidial production and decreased aflatoxins. Two-fold increased yield of conidia was found on MM, which, like V8, did not support aflatoxin production. Adding ammonium to MM significantly increased the production of sclerotia and aflatoxins, but slightly decreased conidial production. Adding urea to MM significantly increased the production of conidia, sclerotia and aflatoxins. CONCLUSIONS: Molasses stimulated conidial production and inhibited aflatoxin production. Its effect on sclerotial production was medium-dependent. Water activity and medium pH were not related to changes in conidial, sclerotial or aflatoxin production. Medium containing molasses alone or molasses plus V8 juice were ideal for conidial production by S strain A. flavus. SIGNIFICANCE AND IMPACT OF THE STUDY: Insight into molecular events associated with the utilization of molasses may help to elucidate the mechanism(s) that decreases aflatoxin biosynthesis. Targeting genetic parameters in S strain A. flavus isolates may reduce aflatoxin contamination of crops by reducing the survival and toxigenicity of these strains.  相似文献

Forty-one fungal isolates (one isolate per species) representing common plant pathogens and food crop contaminants were grown on sterile, polished rice and assayed for mutagenic activity in the Salmonella typhimurium-microsome system. Initially, single doses of aqueous and chloroform extracts of the moldy rice were assayed against the TA100 tester strain by incorporating extracts into the growth medium and by applying small quantities on disks placed on the agar surface. Suspected activity was examined further by analysis of several doses in the plate incorporation assay. Extracts of two aflatoxin-producing isolates (Aspergillus flavus and A. parasiticus) showed pronounced mutagenic activity, as did extracts of five other isolates (A. heterothallicus, A. nidulans, A. terricola, Alternaria tenuis, and Fusarium moniliforme) which did not contain detectable aflatoxins. Seven additional isolates (Botrytis cineria, Ceratocystis fimbriata, Cladosporium herbarum, Fusarium solani f. sp. pisi, Penicillium oxalicum, Thermomyces lanuginosus, and Verticilium albo-atrum) revealed activity which was possibly mutagenic; i.e., mutagenic responses were not observed in both the disk and incorporation assays, and clear dose-related activity was not observed in the incorporation assay. The potential of sodium bicarbonate to inhibit growth of and aflatoxin synthesis by Aspergillus parasiticus was examined in Czapek's agar (CA), a medium in which fluorescence under UV light indicates aflatoxin production. Incorporation of sodium bicarbonate (SB) into CA at 0.011, 0.022, and 0.033 mol% reduced cell viability 63-, 10(3)-, and greater than 10(7)-fold, respectively. Colonies resulting from surviving cells did not fluoresce under UV light, but thin-layer chromatography analysis of culture extracts detected aflatoxins. Potassium bicarbonate (KB) at 0.011 and 0.022 mol% produced inhibitory effects similar to those of SB, but NaCl and silica had no effect. After 7 days, control cultures had the normal aflatoxin distribution (B1 greater than G1 greater than B2 greater than G2), but this distribution shifted to B2 greater than B1 approximately equal to G2 greater than G1 during prolonged incubation. Cultures supplemented with SB and KB contained mostly aflatoxins B1 and G1 after 28 days. Both SB and KB raised the pH of CA to 7.5 to 8.5 at the time of growth. Culture growth on CA adjusted to pH 7.5 to 8.5 with NaOH was not inhibited but exhibited reduced fluorescence and elevated levels of aflatoxins B1 and G1. Thus, while bicarbonate inhibition of growth could not be attributed to pH elevation, the lack of culture fluorescence on CA-SB and CA-KB and the altered aflatoxin distribution were caused by the ability of SB and KB to elevate pH.  相似文献

Human foreskin cell cultures in scheduled DNA synthesis (S phase) of the cell cycle were exposed to UV irradiation at a dose of 10 J · m?2 in the presence of insulin. These treated cell populations, when selectively passaged in a high amino acid supplemented complete growth medium (CM) after 20 Dulbecco's phosphate buffered saline (pH 6.8) (PDL), were able to be grown in soft agar. These treated cell populations were also grown in 1% serum supplemented growth medium and at 41°C in 10% serum supplemented growth medium. Cell populations 4–5 PDL after treatment exhibited altered colony morphology and altered lectin agglutination profiles but would not grow in soft agar. These events appeared to be associated with the early stages in the expression phase of the transformed phenotype. After 20 PDL, we observed that these cells would grow in soft agar at a frequency of 20 colonies/105 cells seeded in soft agar. The cell populations derived from these colonies, when propagated and injected into the nude mice, formed myxofibromas at the injection sites rather than the type of tumor (fibrosarcoma) previously described for chemical carcinogen-induced neoplasms.  相似文献

The goal of this study was to develop a simple plating medium to allow large-scale screening of water samples for the presence of Helicobacter pylori. Five conventional plating media (brain heart infusion, brucella agar, Columbia blood agar base, campylobacter agar kit Skirrow, and HPSPA medium), each containing a commercial antibiotic supplement, were initially evaluated. Eight strains selected as common waterborne organisms (Acinetobacter, Aeromonas, Bacillus, Escherichia coli, Enterobacter, Enterococcus, Helicobacter pylori, and Pseudomonas strains) were individually plated onto each of these media. Three organisms (Acinetobacter, E. coli, and H. pylori) were able to grow on all five media. This growth was unacceptable since Helicobacter grows very slowly and competing organisms must be inhibited for up to 7 days. Therefore, a more selective medium (HP agar) containing a novel mixture of growth supplements plus amphotericin B and polymyxin B was developed. This medium also included a phenol red color indicator for urease production. Aliquots of nonsterile well water that contained native flora (Flavobacterium, Serratia, Citrobacter, Pasteurella, Ochrobactrum, Rahnella, and unidentified molds) and were further adulterated with the eight strains listed above (10(6) CFU of each strain per 100 ml) were spiked with H. pylori and were plated. In spite of the heavy mixed microbial load, only H. pylori colonies grew during 7 days of incubation at 37 degrees C. The color indicator system allowed presumptive identification of H. pylori colonies sooner (12 to 20 h) than the conventional media tested allowed. The HP formulation developed in this study provides a medium with superior selectivity for H. pylori from mixed microbial populations in water and reduces the time required to complete the assay.  相似文献
