Water movement from intracristal spaces in isolated liver mitochondria   总被引:1,自引:0,他引:1  
When analyzing mitochondria isolated in a sucrose medium that had been embedded for thin sectioning according to one low denaturation embedding technique, large intracristal spaces were present in close to 90% of the mitochondria. The two crista membranes were closely apposed in only 40% of all cristae. When the mitochondria were transferred to an incubation medium, the percentage of mitochondria with intracristal spaces was reduced to 40%. About 90% of all cristae were lacking any space separating the two crista membranes. The presence of inorganic phosphate in the medium was required for the closing of the intracristal spaces. The percentage of cristae lacking an intracristal space remained the same after addition of substrate for respiration (state 4) and of ADP (state 3). Inhibition or uncoupling of respiration led to an increase in the percentage of intracristal spaces, showing that oxidative phosphorylation is required to maintain the crista membranes closely apposed. The appearance and disappearance of the intracristal spaces was an indication of water movements across the crista membranes. The mean volume of the mitochondria increased 33% when they were transferred from the sucrose medium to the incubation medium, showing that the removal of water from the cristae was not caused by a passive osmotic effect. Addition of substrate made the volume decrease by 28%. After further addition of ADP, the volume decreased another 23%. No change in volume was associated with inhibition or uncoupling of respiration. The observations revealed that water can move into or out of the cristae independently of water movement out from the entire mitochondrion. Therefore, the water moving out from or into the cristae is translocated across the cristae membrane. The observations are interpreted to reveal the presence of a mechanism that actively prevents water from accumulating in the crista membrane. This mechanism allows for a low water activity to be maintained within the membrane. The variations in the frequency of intracristal spaces occurred without any simultaneous changes in the width of the space appearing between the two surface membranes after isolation of the mitochondria. The observations, therefore, do not agree with the concept that there is an outer compartment that communicates freely with intracristal spaces.  相似文献

The structure and volume of isolated mitochondria embedded for electron microscopy during different respiratory states were analyzed in thin sections. Three different embedding methods were compared; osmium tetroxide fixation/acetone dehydration, glutaraldehyde fixation/acetone dehydration, and glutaraldehyde fixation-osmium tetroxide postfixation/acetone dehydration. Analysis of fresh mitochondria, isolated in a sucrose medium, revealed the presence of a homogeneous population with respect to structure when any of the three methods were applied. After fixation with osmium alone, or in combination with glutaraldehyde, nearly 100% of the mitochondria were in a "condensed" conformation. Mitochondria fixed with glutaraldehyde alone resulted in a population of mitochondria that had large spaces separating the two membranes of the cristae which corresponds to the condensed conformation as observed after osmium fixation. Transfer of the mitochondria to the incubation medium led to the appearance of two classes of mitochondria with respect to size. One class had a volume close to that observed when suspended in sucrose, and another class was present that was 30-45% larger. In osmium fixed or in double-fixed preparations, these small and large classes corresponded to "condensed" and "orthodox" forms of mitochondria respectively. When glutaraldehyde was used alone as the fixative, the two size classes were also present. However, the mitochondria were homogeneous with respect to structure. In these mitochondria, the width of the space that separated the cristae membranes had become reduced when compared to mitochondria suspended in sucrose. The two size classes were also present in samples of mitochondria prepared during both states 3 and 4. State 4 conditions did not lead to any significant increase of the number of condensed mitochondria. In state 3 preparations, 65-70% of the population were condensed. The condensed and orthodox forms could be related to normal and swollen forms of mitochondria. Conditions that led to a swelling also led to an increase in the number of orthodox mitochondria in osmium-fixed material. The different appearance of the mitochondria is explained by the different conditions for fixation of the mitochondria that exist when nonswollen and swollen mitochondria are fixed. This difference is particularly crucial in the case of osmium tetroxide due to the unique way this fixative, among generally used fixatives, denatures proteins.  相似文献

The effect of propylthiouracil-induced neonatal hypothyroidism on some aspects of the biogenesis of free (non-synaptosomal) mitochondria in the cerebral hemispheres and in the cerebellum of developing rat has been studied. The results obtained show that in hypothyroid rats mitochondrial DNA synthesis is delayed, mitochondrial RNA synthesis is not affected and cytochrome aa3 content of mitochondria is lower than in controls. Furthermore ultrathin sections of 14-and 21-day old hypothyroid rat cerebella show mitochondria with an altered ultrastructural organization and large intracristal spaces.  相似文献

An embedding technique has been developed to overcome difficulties that confront light and electron microscopists working with so-called “hard-to-embed” plant tissue. The method was originally described for freeze-dried material. It uses a modified Quickfit Rotaflo Valve and low heat to generate high pressure to aid in the infiltration and embedding of tissue with propylene oxide and plastic. The technique is not too cumbersome and requires 6 days from the dehydration step to the end of the polymerization process. Thick sections (1-2 μm) obtained from material prepared by this method stain readily with toluidine blue, and thin sections for the electron microscope stain satisfactorily following standard treatment with uranyl acetate and lead citrate. The thin sections are stable under the beam of the electron microscope. Results indicate that the quality of tissue preservation with this high pressure embedding technique is as good as that observed using standard embedding methods for electron microscopy.  相似文献

Electron microscopy shows that intact mitochondria can be isolated from neck-muscle stored at 144h post-mortem at 4°. Isolated mitochondria, all in the condensed configuration, have clearly defined outer and inner membranes, outer compartments and intracristal spaces; a larger proportion of swollen ones was isolated from the 144h than from the 120 h post-mortem tissue.Mitochondria from 96 h tissue still retained the following % of the initial values for the ADP/O ratio, respiratory control index (RCI) and state 3 respiratory rate observed for 0–5h tissue: malate+pyruvate, 100, 72 and 53; succinate, 80, 30 and 74; ascorbate+ tetramethyl-p-phenylencdiamine (TMPD), 92, 88 and 72.Both the succinate and ascorbate-TMPD oxidase systems appear to have a critical storage time of about 70 h, whereas the malate+pyruvate system has one of about 96 h. Asharp decline of the ADP/O ratio, RCI and the state 3 respiratory rate occurred after this time, but these three parameters were better preserved in the ascorbate-TMPD oxidase system.The oxidation of the citric acid cycle intermediates in the neck-muscle mitochondria thus shows a higher sensitivity to post-mortem ageing with respect to cytochrome oxidase activity. This is probably due to post-mortem muscle acidification.  相似文献

Ordered arrays of structured material were visualized in the intracristal space of isolated beef heart mitochondria in two ways. Under standard conditions of fixation, structured material in the intracristal space appeared as paracrystalline arrays nestled between two apposing membranes. When mitochondria were preincubated with phosphotungstic acid (PTA) prior to fixation, the structures in the mitochondrial intracristal space took on an open lattice structure. Such structures, either paracrystalline or lattice, could not be demonstrated in the mitochondrial matrix space under these conditions.Pretreatment with PTA prior to fixation increased greatly the frequency with which structured material was observed within the mitochondrial intracristal space. Visualization of the PTA-induced lattice structures appeared to be pH dependent, being most clearly seen between pH 7·0 and 7·5. Above pH 7·5, lattice structures could not be seen, whereas at pH values below 7·0, the observed structures in the intracristal space no longer retained an organized lattice structure but became amorphous. Increasing the concentration of PTA from 0·1% to 3·5% or the incubation time from 5 sec to 1 h did not significantly alter the frequency of observation of lattice structures, as long as the mitochondrial preincubation with PTA was carried out between pH 7·0 and 7·5.  相似文献

Summary Adult mice were treated daily with HCG (human chorionic gonadotropin). The most notable changes were found 24 hours after the second injection of 100 i.u. Nearly all lipid droplets had disappeared. Mitochondria showed a constriction linking the two portions of the organelle with widened intracristal spaces. Doughnut-shaped, bifurcated, and elongated mitochondria occurred commonly; their intracristal spaces were consistently widened. The Golgi complex was well developed. Dense bodies and phagolysosomes seemed to disappear. The results indicate that the mitochondrion is the first and most responsive organelle affected by HCG treatment. A hypothesis is advanced that the mitochondria of the Leydig cell provide the energy required for the lipolytic effect of HCG.  相似文献

Sweetpotato mitochondria, that showed respiratory control, were studied with respect to ultrastructure. If fixed in media containing sucrose at 0.4 M, the cristae were dilated and the matrix was highly condensed. A more orthodox ultrastructural form was observed when the mitochondria were fixed in a medium containing sucrose at 0.25 M, i.e., the matrix was more expanded, the cristae were less dilated, and peripherally, the inner membrane element lay adjacent to the outer membrane element. These results are discussed in terms of a sucrose-accessible space (space between outer and inner membrane elements including intracristal space), and a space relatively inaccessible to sucrose (matrix). Ultrastructural shifts were not observed with change in metabolic steady state of the mitochondria. High resolution electron micrographs showed that the ultrastructure of sweetpotato mitochondria is very similar to that of animal mitochondria.

Purity and homogeneity of mitochondrial fractions were followed both by phase-contrast and electron microscopy. Preparations from sweetpotato, using older methods, were relatively homogeneous with respect to particle type and size, whereas avocado preparations contained a high proportion of chloroplasts and cellular debris. A method of purification involving sucrose-density-gradient centrifugation was developed. Purified mitochondria exhibited respiratory control and appeared similar to unpurified mitochondria under the electron microscope.

Liver mitochondria were exposed in vitro at 30°C to microwave radiation (2.45 GHz) during the following states of respiraton: resting, state 1; substrate dependent, state 2; ADP stimulated, state 3; and ADP depleted, state 4. At 10 or 100 mW/g, with succinate as substrate, no effect of exposure was observed on states 1–4 or the respiratory control index (state 3/state 4) of either tightly or loosely coupled mitochondria. When glutamate was used as substrate, no effects were observed at 10 mW/g. However, in the loosely coupled mitochondria the 100 mW/g exposure produced an increase in states 2 and 4 and a decrease in the respiratory control index. The results suggest that the function of loosely coupled mitochondria can be affected at high power levels of microwave radiation.  相似文献

Approximately 40% of the mitochondria in the sphincter muscle of the crayfish vas deferens have prismatic-type cristae. In cross section, the angular cristae have either triangular or rhomboid profiles which are surrounded by a hexagonal array of electron-dense dots. In longitudinal section, these mitochondria exhibit both thick and thin parallel lines, which represent cristae and filaments, respectively. It is postulated that the matrix of the prismatic-type mitochondria is packed with rodlets composed of an electron-dense core and a less dense shell. Close packing of these rodlets results in the regular hexagonal dot array. Deletion of fascicles of 3 or 4 rodlets results in spaces with triangular or rectangular cross sections. Lining of these spaces with membranes results in cristae with triangular or rhomboid cross sections.  相似文献

Summary Autoradiography was found to be a useful method for studying the function of mitochondria. It might provide means to investigate the mitochondrial ATP-ase activity. Applying this method, different problems of metal salt-techniques, firstly the aspecific reactions, can be eliminated. The incubation solution contained 3H-ATP at 100 c activity. The autoradiographic precipitate seems to appear on the membrane of the mitochondria and in the intracristal space. Atractylate treatment inhibits completely the permeation of 3H-ATP. So-called arreactive mitochondria with retained structure lacking of reaction-precipitate were found besides the mitochondria revealing pronounced activity. This might refer to the manifestation of a functional state. Experiments with P32 ATP may contributs to the more exact determination of original place of the enzyme.  相似文献

Studies with liver mitochondria from rats which starved for 48 hours showed the rate of ADP-stimulated respiration to be 20% lower than in the presence of an uncoupler. This effect was eliminated by preincubation of mitochondria with carnitine. Mitochondria from fed rats were characterized by a considerable decrease of states 3 and 4 respiration. In this case carnitine produced no effect. Preincubation of mitochondria from the liver of fed rats with alpha-ketoglutarate resulted in a substantial increase of the states 3 and 4 respiratory rates. There proved to exist at least two types of regulation of adenine nucleotide transport through the inner mitochondrial membrane depending on the metabolic state of the organism, i.e. by inhibition of adenine-nucleotide translocase by cytoplasmic acyl-CoAs and by control of intramitochondrial adenine nucleotide pool.  相似文献

The following stereological parameters of mitochondria were calculated in rat germ cells during the spermatogenesis: volume density of matrix, outer compartment, outer membrane and inner membrane, surface density of outer membrane and inner membrane. They were the basis to calculate the partition coefficient of matrix and partition coefficient of outer compartment. The matrix volume demonstrated a decreasing in mitochondria of germ cells during spermatogenesis. The relative volume of outer compartment was calculated with the intracristal spaces and revealed increasing from spermatogonia to spermatids. The partition coefficient for the matrix significantly decreased. Our observations suggest that transformation of mitochondrial configuration during spermatogenesis and spermiogenesis is the expression of intensive metabolic processes and activity of membrane transport in germ cells.  相似文献

Exposure of exponentially growing L1210 cells in vitro to 5-10 micrograms/ml of rhodamine 123 (R123) for 16-48 hr inhibits cell proliferation and induces cell arrest in the G1A phase of the cell cycle. The cells remain viable during the arrest and resume growth after removal of R123; extended exposure to R123 is cytotoxic. Exposure to R123 results in morphological alterations in mitochondria of all cells observed; specifically, mitochondria of R123-treated cells are characterized by a distention of the intracristal spaces and a significant increase in the number of matrix granules. Gross morphological changes of mitochondria include formation of extended organelles and the appearance of doughnut-shaped structures.  相似文献

The formation of paracrystalline inclusions in mitochondria following postmortem aging in situ is further investigated. Inclusions are found to localize either in the mitochondrial intracristal space or between the inner and outer membrane. The greater majority of the inclusions are of the intracristal type. pH is not an important factor in the formation of the inclusions, since lowering the pH of the mitochondrial suspensions does not increase the number of mitochondria with inclusions. It is concluded that changes associated with aging during the prolonged preservation of the mitochondria both in situ and in in vitro experiments are probably the principal cause for the formation of the mitochondrial inclusions.  相似文献

A method which gives good quality 1-2 μm thick sections of undecaldfied cancellous and thin cortical bones for light miuoscopy is described. Formalin fixed material is dehydrated in graded acetones and embedded in a modiEed formula of Spurr's low viscosity embedding medium. After a 16 hour polymerisation period at 60 C, sections are cut at 1-2 μm thickness on a Porter-Blum JB4A rotary microtome Using glass knives. Sections are attached to clean glass slides with heat, the resin degraded in bromine vapour and removed in acetone. This allows comparative ease of staining. The technique is rapid, does not interfere with tetracycline fluorescence and the same specimens can be used to prepare thick sections for microradiography.  相似文献

The subcellular localization of Factor VIII/von Willebrand protein (VIII R:Ag) is studied with monoclonal antibody and gold immunocytochemical technique. Monoclonal antibody against purified VIII R:Ag is brightly fluorescent on megakaryocytes and platelets. In E.M., gold immunolabeling is performed on thin cell sections of human megakaryocytes and platelets. Different embedding materials are used to preserve the antigenicity : Epon embedded megakaryocytes show a high concentration of VIII R:Ag in alpha-granules using 4F9 monoclonal antibody. In comparison, lowicryl K4M embedded material does not improve the same specificity, only a few platelets granules were stained. This subcellular localization, in full agreement with biochemical results appears visualized for the first time in E.M.  相似文献

Structural changes of mitochondria in correlation to their energetic state have been observed as matrix expansion and condensation. In this communication we describe a morphological correlation in freeze-fractured mitochondrial membranes which is also dependent on the metabolic state of the organelle: the frequency by which the fracture plane following the inner or outer boundary membrane deviates by jumping from one membrane to the other is higher in phosphorylating mitochondria when compared to freshly isolated or energized mitochondria. These deflections of the fracture plane occur mostly in minimal, short steps showing close apposition of the two boundary membranes. We therefore conclude that the observed change in morphological appearance is produced by a change in interactions between the inner and outer membranes correlated to the different functional states of the inner membrane.  相似文献

The use of 2-hydroxyethyl-methacrylate (GMA) as embedding medium for histological investigations in phytopathology A new plastic embedding technique is described for subsequent thin sectioning of plant tissues. In comparison to the paraffinmethod the GMA polymerization system is less time consuming. The excellent preservation of well-fixed tissue is fully asserted, as the embedding medium is not removed from the sections. In lightmicroscopic studies convincing results were obtained with different staining procedures; specific evidence for polysaccharides, pectine and nucleic acids was carried out with thin sections of 2-8 μm thickness, also by fluorescence microscopy. The GMA-embedding technique seems to be of value for various histological investigations in phytopathology.  相似文献
