In the absence of cations, the release of proteins from pollen tubes ofNicotiana tabacum in culture is greatly dependent on boron concentration and inversely related to growth stimulation. The minimum of proteins in the medium occurs at 100 mg 1? of boric acid, which is the optimum concentration for growth. The shift of boron level to this optimum further increases the proportion of proteins bound to the insoluble pollen tube fraction; on the other hand the amount of soluble proteins is not affected inside pollen tubes, but greatly decreased in the medium. The loss of proteins into the medium is considerably reduced by oalcium and also at the optimal boron concentration and in the presence of K+ and Mg2+ ions. The rate of tube elongation, however, is slightly decreased and the duration of pollen growth activity is not extended. The release of proteins is not affected by potassium and is slightly reduced by magnesium. The possible mechanism of calcium and boron action on protein release is discussed in relation to the exocytotic function of secretory vesicles and it is suggested that boron supports the incorporation of proteins transported to the growing tip into the pollen tube wall structures.  相似文献

Illus. 1963.—A pollen population effect occurs whenever pollen grains are grown in vitro. Small pollen populations germinate and grow poorly if at all, under conditions which support excellent growth of large pollen populations. The pollen population effect is overcome completely by a growth factor obtained in water extracts of many plant tissues. This factor is shown to be the calcium ion, and its action confirmed in 86 species representing 39 plant families. Other ions (K+, Mg++, Na+) serve in supporting roles to the uptake or binding of calcium. The high requirement of calcium (300–5000 ppm, as Ca (NO3)2·4H2O, for optimum growth) and low calcium content of most pollen may conspire to give calcium a governing role in the growth of pollen tubes both in vitro and in situ. It is suspected that ramifications of this role extend to the self-incompatibilities of plants and to the curious types of arrested tube growth distinguishing, for example, the orchids. A culture medium which proved its merit in a wide variety of pollen growth studies included, in distilled water, 10% sucrose, 100 ppm H3BO3, 300 ppm Ca (NO3)2·4H2O, 200 ppm MgSO4·7H2O and 100 ppm KNO3.  相似文献

Summary The hydrodynamics of mature pollen rehydration in Nicotiana tabacum was used to study reversible inhibition of pollen germination in vitro. Tobacco pollen was incubated for various times in media containing calcium, potassium and magnesium salts, boric acid, and exhibiting different osmotic pressures as a function of sucrose concentration. Total inhibition of germination with complete viability preservation was achieved for 56 h by keeping the grains in medium with 80% sucrose, since typical percentages of germination and pollen tube lengths were recovered after this treatment. These effects were considered as consequences of natural osmoregulation of rehydration/germination in mature pollen. The possibility of applying these findings to the incubation of pollen with Agrobacterium tumefaciens to develop a pollen transformation method is discussed.  相似文献

The addition of boric acid or calcium, or both boric acid and calcium to the warm water in which the styles were treated led to a further enhancement of pollen tube growth in otherwise incompatible styles. It was established that developing pollen tubes ofOenothera organesis obtain sugar from the stylar tissue through which they grow. The rate of this utilization of endogenous sugar was stimulated by the addition of boric acid. It was further established that with increased pollen tube development there was reduction in callose plugs as well as in content of endogenous stylar sugar.  相似文献

Plant‐derived smoke stimulates seed germination in numerous plant species. Smoke also has a positive stimulatory effect on pollen germination and pollen tube growth. The range of plant families affected my smoke still needs to be established since the initial study was restricted to only three species from the Amaryllidaceae. The effects of smoke‐water (SW) and the smoke‐derived compounds, karrikinolide (KAR1) and trimethylbutenolide (TMB) on pollen growth characteristics were evaluated in seven different plant families. Smoke‐water (1:1000 and 1:2000 v:v) combined with either Brewbaker and Kwack's (BWK) medium or sucrose and boric acid (SB) medium significantly improved pollen germination and pollen tube growth in Aloe maculata All., Kniphofia uvaria Oken, Lachenalia aloides (L.f.) Engl. var. aloides and Tulbaghia simmleri P. Beauv. Karrikinolide (10?6 and 10?7 m ) treatment significantly improved pollen tube growth in A. maculata, K. uvaria, L. aloides and Nematanthus crassifolius (Schott) Wiehle compared to the controls. BWK or SB medium containing TMB (10?3 m ) produced significantly longer pollen tubes in A. maculata, K. uvaria and N. crassifolius. These results indicate that plant‐derived smoke and the smoke‐isolated compounds may stimulate pollen growth in a wide range of plant species.  相似文献

Summary The involvement of exogenous calcium ions in the regulation of pollen tube formation has been investigated in Haemanthus albiflos L. and Oenothera biennis L. by following the changes that occur in pollen germination, tube growth, and 45+Ca2+ uptake and distribution upon application of Verapamil (an inhibitor of calcium channels), lanthanum (a Ca2+ substitute), and ruthenium red (believed to raise the intracellular calcium level). It was found that exogenous Ca2+ takes part in the formation of the calcium gradient present in germinating pollen grains and growing pollen tubes. Ca2+ ions enter the cells through calcium channels. Raising or reducing 45Ca2+ uptake causes disturbances in the germination of the pollen grains and in the growth of the pollen tubes.  相似文献

The ability of pine pollen to grow in vitro is discussed in relation to its in situ. Optimal conditions of growth in vitro were investigated. All the experiments were made with pollen of Pinus mugo Turra. A satisfactory medium is described as that which at pH 5.2 and at 29 °C can lead to a tube growth bigger than in vivo. The initial growth (0–30) hours) in terms of tube growth and respiration is described in detail. It is shown that a correlation exists between tube growth and respiration. A method for estimating the metabolism and growth of the pollen tube, based on the uptake of 32p-labelled phosphate, is described. Addition of known stimulators of plant growth and extension does not stimulate tube growth. Temperature studies show that under 20°C germination does not take place.  相似文献

Isolated frog rod outer segments (ROS) with a leaky plasma membrane showed a bicarbonate-dependent, ATP-activated45Ca accumulation. This calcium uptake requires magnesium and is specific for ATP; Other nucleotides, ITP, GTP, UTP and the non-hydrolysable analogue of ATP --methylene ATP did not substitute for ATP.45Ca accumulation was inhibited by mersalyl, ethylmaleimide, ruthenium red, oligomycin and dicyclohexylcarbodiimide and was unaffected by ouabain. Addition of taurine to the incubation medium enhanced45Ca uptake in a concentration-dependent manner; increases of more than 100% being produced by 25 mM taurine. The taurine-induced stimulation of45Ca uptake was also sensitive to the tested inhibitors. The effect of taurine was only exerted on the bicarbonate-dependent, ATP-activated45Ca uptake. Calcium accumulation observed in the absence of ATP or in a tris-buffered medium was unaffected by taurine. Other amino acids, glycine, GABA, -alanine, glutamic acid and the taurine analogue guanidinoethyl-sulfonate did not stimulate45Ca uptake. These results suggest that taurine is affecting a Mg-ATPase activity responsible for calcium accumulation in frog ROS.  相似文献

Summary In Brewbaker and Kwack's medium (BK) only 16% of the pollen grains germinated, and these produced pollen tubes having a maximum length of 25 m. With a solution based on Monnier's medium 47% germination and 160-mlong pollen tubes were observed. Calcium was shown to be essential for germination; the optimal concentration was 880 mg/l calcium chloride. The optimal concentrations of magnesium sulphate and boric acid were 360 and 50 mg/l, respectively. Germination at pH 4.0 but also pH 8.0 and the presence of vitamins B1 and B6 (1 mg/l each) were stimulatory. Polyethylene glycol (PEG) was superior to sucrose as an osmoticum and germination and tube length were significantly improved using PEG 4000 at a concentration of 120 g/l (0.03 M). Equimolar concentrations of PEG 400 and PEG 600 gave inferior results. Combining PEG with sucrose in the medium did not improve germination or increase tube length.  相似文献

Summary When the pollen tubes of Pisum sativum (initial line) and of its mutants are grown on a standard medium containing only sucrose, boric acid and agar-agar, no difference in maximum length was observed. But, while pollen tubes of the initial line took nine hours to reach this length, pollen tubes of the mutants needed only six hours. Growth seems to be faster in pollen tubes of the mutants than in those of the initial line.Further investigations examined the influence of twenty-one amino acids on pollen tube growth. With the initial line, these substances can be classified into three groups: those that promote pollen tube growth; those which have no influence upon its growth; and those which reduce its growth. The amino acids of each group are characterized by special structural properties. Those amino acids which accelerate pollen tube growth of the initial line show variable effects on the pollen tubes of the mutants. In some cases the same behaviour of pollen tubes can be observed whether amino acids are added or not, in others the addition of amino acids has a positive effect on pollen tube growth, though less than on pollen tubes of the initial line, and in a single case the addition of an amino acid is followed by a negative effect on growth.  相似文献

山玉兰(Magnolia delavayi)是木兰科木兰属的常绿乔木或大型灌木,是重要的园林造景、庭院绿化素材,也是重要的育种资源。山玉兰花粉的研究对其杂交育种的成败具有重要影响,但目前尚未见其花粉活力的相关报道。该研究以新鲜的山玉兰花粉为对象,采用悬滴培养法分析了温度、光照以及培养液的蔗糖和硼酸浓度对山玉兰花粉萌发的影响。结果表明:(1)山玉兰花粉萌发时,最适宜的温度为27℃。(2)光暗条件下,山玉兰花粉以浓度为5%的蔗糖培养效果最佳,其萌发率在16%以上;而硼酸浓度则以0.001%的培养效果最佳。(3)蔗糖与硼酸共同作用可有效促进花粉萌发和花粉管生长。其中,在光照条件下,以5.0%蔗糖+0.001%硼酸为最适宜的培养液,花粉萌发率达41.27%,花粉管长达281.49μm;而在黑暗条件下,则以5.0%蔗糖+0.01%硼酸为最适宜的培养液,花粉萌发率达45.71%,花粉管长达254.00μm。该研究结果为进一步开展人工辅助授粉、发掘山玉兰的种质资源工作奠定了基础。  相似文献

The metabolism of purine- and pyrimidine nucleotides in pine pollen (Pinus mugo) grown in suspension cultures have been examined. In the ungerminated dehydrated pollen, the presence of ATP has been demonstrated. Incubation of the pollen in a germination medium leads to an exhaustion of the ATP pool, which is restored with the onset of oxygen uptake. By labelling pollen cultures with 32P-orthophosphate, it has been possible to quantitate the nucleotide components of the pollen, and thereby to measure changes in the nucleotide pattern at various growth stages. The most marked changes occur during the initial phase of tube growth when a large increase in the ribonucleoside triphosphate and the sugar nucleotide pools is observed. The contents of ATP and UDP-glucose are further increased if starch synthesis is initiated by the addition of sucrose to the culture medium. In order to determine whether nucleotides in pine pollen are synthesized from de novo pathways or via reutilization pathways, from breakdown products of nucleic acids, pollen was incubated with 14C-labelled precursors of both the de novo and the reutilization pathways. Incorporation experiments established de novo synthesis of ATP and GTP from glycine, and de novo synthesis of CTP and UTP from orotic acid. The operation of pathways for the utilization of exogenous nucleosides was also demonstrated. While uridine, cytidine and adenosine are incorporated into nucleoside triphosphate to a great extent, only minor incorporation of inosine and guanosine is observed. These reutilization pathways might be of importance for the synthesis of nucleotides during tube growth in situ. Addition of inhibitors of glycolysis and oxidative phosphorylation drastically reduces the level of ribonucleoside triphosphates, indicating a rapid turnover of the nucleotide pool.  相似文献

In vitro pollen germination and tube length studies are valuable in elucidating mechanisms (germination capacity and rate, tube growth rate) possibly associated with genetic differences in male transmission. On each of two collection dates, the percentage germination and tube length of the binucleate pollen grains from five diverse sesame (Sesamum indicum L.) genotypes were determined at eight times (30, 60, 90, 120, 150, 180, 240, 300 min) after inoculation on a semisolid medium containing 10% (100 g l-1) sucrose (C12H22O11), 0.4% (4 g l-1) purified agar (Fisher Lot 914409), 0.1% (1 g l-1) calcium nitrate [Ca(NO3)2 ⋅ 4H2O] and 0.01% (100 mg l-1) boric acid (H3BO3). Before heating, the pH of the medium was adjusted to 7.0 with a 0.1 N potassium hydroxide (KOH) solution. Over the five genotypes, 5% germination was found 30 min after inoculation and a maximum of 37% germination 120 min after inoculation with no significant changes thereafter. As indicated by the highly significant genotype×time after inoculation interaction, the genotypes differed in the time at which germination was initiated and maximum germination attained. Over all five genotypes, the tube length was 91 μm 30 min after inoculation, reaching a maximum of 1000 μm 300 min after inoculation. As shown by the highly significant genotype×time after inoculation interaction, the genotypes differed in the time at which tube length was observed and the maximum tube length was attained. Little or no relationship between percent germination and tube length was observed among the genotypes. For both percent germination and tube length, the statistical significance of collection date and its interactions with genotype and time after inoculation indicated that environment in the form of collection date was also an influencing factor. These results indicated that genetic differences among genotypes were present for in vitro germination capacity, germination rate and tube growth rate and that these factors singly or in combination could alter male transmission of genetic elements. Summary. An acid phosphatase (acPAse) activity was released during germination and tube growth of pollen of Lilium longiflorum Thunb. By inhibiting components of the secretory pathway, the export of the acPase activity was affected and tube growth stopped. Brefeldin A (1 μM) and cytochalasin D (1 μM), which block the production and transport of secretory vesicles, respectively, inhibited the acPase secretion. The Ca2+ channel blocker gadolinium (100 μM Gd3+) also inhibited acPase secretion and tube growth, whereas 3 mM caffeine, another Ca2+ uptake inhibitor, stimulated the acPase release, while tube growth was inhibited. The Yariv reagent (β-D-glucosyl)3 Yariv phenylglycoside stopped tube growth by binding to arabinogalactan proteins of the tube tip cell wall but did not affect acPase secretion. A strong correlation between tube growth and acPase release was detected. The secreted acPase activity had a pH optimum at pH 5.5, a K M of 0.4 mM for p-nitrophenyl phosphate, and was inhibited by zinc, molybdate, phosphate, and fluoride ions, but not by tartrate. In electrophoresis gels the main acPase activity was detected at 32 kDa. The conspicuous correlation between activity of the secretory pathway and acPase secretion during tube elongation strongly indicates an important role of the acPase during pollen tube growth and the secreted acPase activity may serve as a useful marker enzyme assay for secretory activity in pollen tubes Received July 25, 2001 Accepted January 15, 2002  相似文献

The stimulation of the plasma membrane (PM) H+-ATPase by boric acid was studied on a microsomal fraction (MF) obtained from ungerminated, boron-dependent pollen grains of Lilium longiflorum Thunb. which usually need boron for germination and tube growth. ATP hydrolysis and H+ transport activity increased by 14 and 18%, respectively, after addition of 2-4 mM boric acid. The optimum of boron stimulation was at pH 6.5-8.5 for ATP hydrolysis and at pH 6.5-7.5 for H+ transport. No boron stimulation was detected when vanadate was added to the MF, whereas an increase of 10-20% in ATP hydrolysis and H+ transport was still measured in the presence of inhibitors specific for V -type ATPase (nitrate and bafilomycin) and F-type ATPase (azide), respectively. A vanadate-sensitive increase in ATP hydrolysis activity was also observed in partially permeabilized vesicles (0.001%[w/v] Triton X-100) suggesting a direct interaction between borate and the PM H+-ATPase rather than a weak acid-induced stimulation. Additionally, we measured the effect of boron on membrane voltage (Vm) of ungerminated pollen grains and observed small hyperpolarizations in 48% of all experiments. Exposing pollen grains to a more acidic pH of 4 caused a depolarization, followed in some experiments by a repolarization (21%). In the presence of 2 mM boron such hyperpolarizations, perhaps caused by an enhanced activity of the H+-ATPase, were measured in 58% of all tested pollen grains. The effects of boron on Vm may be reduced by additional stimulation of a K+ inward current of opposite direction to the H+-ATPase. All experiments indicate that boron stimulates an electrogenic transport system in the plasma membrane which is sensitive to vanadate and has a pH optimum around 7, i.e. the plasma membrane H+-ATPase. A boron-increased PM H+-ATPase activity in turn may stimulate germination and growth of pollen tubes.  相似文献

The data presented here describe ratio-imaging of in intracellular free calcium (Ca2+i) during the self-incompatibility (SI) response in pollen. Use of the ratiometric indicator, fura-2 dextran, in pollen tubes of Papaver rhoeas has provided new, detailed information about the spatial-temporal alterations in Ca2+i, and has permitted calibration of alterations in the concentration of intracellular free calcium ([Ca2+]i) in the SI response. Ratio images demonstrate that, like other pollen tubes, normally growing P. rhoeas pollen tubes exhibit a tip-focused gradient of Ca2+bfi, with levels reaching 1–2 μM at the extreme apex of the pollen tube. Non-growing pollen tubes did not exhibit this tip-focused gradient. Basal levels of Ca2+i in the shank of the pollen tube were fairly consistent and had a mean value of 210 nM, with low-level fluctuations +/? 50 nM observed. Challenge with incompatible S proteins resulted in S-specific, rapid and dramatic alterations in [Ca2+]i within a few seconds of challenge. Increases in [Ca2+]i were visualized in the subapical/shank regions of the pollen tube and alterations in [Ca2+]i in this region subsequently increased for several minutes, reaching> 1.5 μM. At the pollen tube tip, a diminution of the tip-focused gradient was observed, which following some fluctuation, was reduced to basal levels within ~1 min. Our data suggest that some of these alterations in [Ca2+]i might be interpreted as a calcium wave, as the changes are not global. Although the increases in [Ca2+]i in the subapical/shank region are very rapid, because tip [Ca2+]i oscillates during normal growth, it is difficult to ascertain whether the increases in the shank of the pollen tube precede the decreases in [Ca2+]i at the pollen tube tip.  相似文献

The calcium inhibitors A23187, EGTA and La3+ inhibit pollen grain germination and growth of pollen tubes of Lilium davidii var. unicolor at different concentrations. Treatment with 10−4 or 10−5 M ionophores A23187 reduced germination rate and resulted in distortion of pollen tube. Addition of 2 or 10 mM of the chelator EGTA disturbed the direction of pollen tube growth and extended the diameter of pollen tube as observed by light and confocal microscopy. The Ca2+-channel blocker lanthanum chloride (La3+) restrained germination or markedly caused transformation of pollen tube. Furthermore, all treatments led to disappearance of any calcium gradient. Calcium distribution in pollen grain and pollen tube was altered as shown by confocal microscopy for each treatment. This indicates that the inhibitors influence pollen development by affecting the calcium gradient which may play a critical role in germination and tube growth. Fourier transform infrared (FTIR) spectra indicated slight increases in contents of amide I and a substantial decrease in the content of aliphatic esters and saturated esters in treated pollen tubes compared with normal pollen tubes. The FTIR analysis confirmed that EGTA and La3+ weakened the accumulation of ester in pollen tubes, which may be associated with an increased content of amide I.  相似文献

A simple, reliable medium for pollen germination of Cajanus cajan was developed by modifying Brewbaker and Kwack (BK) medium. Past attempts of C. cajan pollen germination in artificial media were not successful. A medium containing polyethylene glycol 4000 (PEG) showed more than 90% germination for C. cajan var. Pusa 33 only when the young buds (36 h before anthesis) were kept in pollen germination medium (PGM) for 36 h before pollen extraction. Supplementation of PGM with epsilon-amino caproic acid (EACA), an amino acid, showed improved pollen germination in Pusa 33 and also helped to avoid preconditioning of young buds before pollen extraction. It was also observed that there is a genotypic difference in the level of EACA required for in vitro pollen germination. Thus a complete medium for C. cajan genotypes consists of 37.5% sucrose+ 15% PEG 4000+250 mg l(-1) boric acid+300 mg l(-1) calcium nitrate+100 mg l(-1) potassium nitrate+ 200 mg l(-1) magnesium sulphate+1% agar+EACA (0, 100, 250, 500, 750 or 1000 mg l(-1)).  相似文献

Petunia (Petunia hybrida Vilm. cv. ‘Snowstorm') plants were grown in saline solution (NaCl, MgCl2, and/or CaCl2) of 0, 1, 2, and 3 bars osmotic pressures. Pollen viability was tested by tetrazolium chloride staining and by germination (by the hanging drop method, using 15 % sucrose and 0.01 % boric acid as the nutrient medium, at 27 ± 1 C). Pollen viability decreased with increased salinity. Pollen from plants grown in single salt solutions of NaCl, MgCl2, and CaCl2 (each at 0, 1, 2, or 3 bars osmotic pressure) was germinated in base culture medium. Pollen viability decreased more with NaCl than with MgCl2 or CaCl2. In vitro studies of the effects of three salts, viz., NaCl, MgCl2, and CaCl2, on pollen germination and tube growth showed that NaCl inhibited germination and pollen tube growth more than did MgCl2 or CaCl2. MgCl2 was least injurious, and even promoted tube growth at 0.5 and 0.75 bars osmotic pressure. Adding low concentrations of MgCl2 reduced the toxic effect of NaCl and increased the percentage of germination. CaCl2 reduced the effect of NaCl less than did MgCl2. We conclude that specific ion effects were more important than osmotic pressure.  相似文献
