The production of extracellular enzymes by the thermophilic fungus Thermomyces lanuginosus was studied in chemostat cultures at a dilution rate of 0.08 h–1 in relation to variation in the ammonium concentration in the feed medium. Under steady state conditions, three growth regimes were recognised and the production of several extracellular enzymes from T. lanuginosus was recorded under different nutrient limitations ranging from nitrogen limitation to carbon/energy limitation. The range and the production of carbohydrate hydrolysing enzymes and lipase increased from Regime I (NH4Cl 600 mg l–1) to Regime III (NH4CI 1200 mg l–1), whereas production of protease was highest in Regime II (600 mg l–1 < NH4Cl <1200 mg l–1).  相似文献

The kinetics of the release of chitinolytic activity (endochitinase EC 3.2.1.14, \-N-acetyglucosaminidase EC 3.2.1.30) by a yeast cell wall lytic Arthrobacter species was studied. The organism was cultivated on yeast cell wall, mycelium of Trichoderma reesei, colloidal chitin, N-acetylglucosamine, glucosamine and mixtures with acetate. With the exception of yeast cell wall, these substrates were used as the sole source of carbon and nitrogen. The growth on colloidal chitin (0.5%) proceeded at a maximum specific growth rate (umax) of 0.23 h–1 and yielded 2700 mU1–1 chitinase. Yeast cell wall and mycelium of T. reesei supported more rapid growth (max = 0.30 h–1 and 0.25 h–1 respectively) but yielded reduced chitinase activity (565 mUl–1 and 700 mUl–1). The growth rate on glucosamine (max = 0.24 h–1) was reduced when this was mixed with acetate (max = 0.12 h–1), whereas the enzyme yield was increased from 720 mUl–1 to 960 mUl–1. The same effect on growth rate was observed with glucose and equimolar mixtures of glucose and acetate, indicating a strong impact of the organic acid on carbohydrate transport or metabolism. The growth of adapted cells on N-acetylglucosamine was comparable to that observed on an equimolar mixture of glucosamine and acetate, indicating that N-acetylglucosamine is rapidly hydrolysed by adapted cells.  相似文献

Degradation of polyaromatic hydrocarbons by Burkholderia cepacia 2A-12   总被引:1,自引:1,他引:0  
A new strain of bacterium degrading polyaromatic hydrocarbons (PAHs), Burkholderia cepacia 2A-12, was isolated from oil-contaminated soil. Of three PAHs, the isolated strain could utilize naphthalene (Nap) and phenanthrene (Phe) as a sole carbon source but not pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the strain was enhanced by the addition of other organic materials such as YE, peptone, glucose, and sucrose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g YE l–1, an optimum concentration, was supplemented into the basal salt medium (BSM) with 215 mg Phe l–1, the specific growth rate (0.30 h–1) and Phe-degrading rate (29.6 mol l–1 h–1) were enhanced approximately ten and three times more than those obtained in the BSM with 215 mg Phe l–1, respectively. Both cell growth and PAH degradation rates were increased with increasing Phe and Pyr concentrations, and B. cepacia 2A-12 had a tolerance against Phe and Pyr toxicity at the high concentration of 730–760 mg l–1. Through kinetic analysis, the maximum specific growth rate ( max) and PAH degrading rate ( max) for Phe were obtained as 0.39 h–1 and 300 mol l–1 h–1, respectively. Also, max and max for Pyr were 0.27 h–1 and 52 mol l–1 h–1, respectively. B. cepacia 2A-12 could simultaneously degrade crude oil as well as PAHs, indicating that this bacterium is very useful for the removal of oils and PAHs contaminants.  相似文献

Summary The course of the CO2 evolution rates of soil samples has been followed continuously in the absence and in the presence of various organic compounds. After an incubation period of 300 hours at 13 and 20°C the CO2 evolution from pasture soil (containing 1.76% soil organic carbon) amounted to 0.13 and 0.44g CO2–C.g soil–1.h–1, respectively. For arable soil (containing 1.20% soil organic carbon) the rates amounted to 0.04 and 0.09 g CO2–C.g soil–1.h–1, respectively.At 20°C larger amounts of the organic substrates added to the soil supplied with 20 g NH4NO3–N.g soil–1 were lost as CO2 than at 13°C, indicating a higher efficiency of the growth of microorganisms at lower temperatures. In the absence of NH4NO3 the respiration rates were initially higher than in its presence, suggesting that a part of the soil microflora is inhibited by low concentrations of NH4NO3. The amounts of carbon lost were low for phenolcarboxylic acids with OH groups in the ortho position. The replacement of one of these groups by a methoxyl group resulted in a larger amount of the C lost as CO2. The replacement of the COOH group by a C=C–COOH group had a decreasing effect on the decomposition of the phenolic acids tested. The decomposition of vanillic acid,p-hydroxybenzoic acid, and of the benzoic acids with OH groups in the meta position was as complete as that of glucose, amino acids or casein. The decomposition of bacterial cells to CO2 was considerably less than that of glucose.No evidence could be obtained that the low percentage of substrate converted to CO2 at the time of maximal respiration rate was due to the decreasing diffusion rate of substrate to the microbial colonies in the soil during the consumption of substrate.  相似文献

Eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid productivities from chemostat cultures of an isolate of Isochrysis galbana have been studied. The productivities reached in the interval of dilution rates between 0.0295 h–1 and 0.0355 h–1 were 1.5mg·1–1·h–1 for lipids, 300 g·1–1·h–1 for EPA and 130g1·1–1·h–1 for DHA. Furthermore, light attenuation by mutual shading, and agitation speed influences on growth and fatty acid composition were analysed. A model relating steady-state dilution rates to internal average light intensity has been proposed, the parameter values of which obtained by non-linear regression were: maximum specific growth rate (max)=0.0426 h–1; the affinity of cells to light (Ik) = 10.92 W·m–2; the exponent (n) = 5.13; regression coefficient (r 2)=0.9999. Physical and chemical conditions, particulate matter and N-uptake were characterized at two sampling sites at the eastern German coast of the Baltic Sea (Pomeranian Bay) over the annual period of 1997 (February–November). The inshore sampling sites (5 m water depth) differed with respect to the potential influences of river run-off and salt water exchange (mean values of salinity: 7.05 and 8.72 PSU), respectively. The mean org-Cdiss/org-Cpart-ratios (4.9 and 12.6) fell in the same order of magnitude (1.0–12.6) as values of neighboring inshore waters, and increasing values reflect an enhancement of the trophic level. Beside differences of nitrogen concentrations (dissolved inorganic nitrogen: 1.8–23.8 and 0.9–9.9 mol l–1), particulate nitrogen (4.30–41.01 and 2.69–9.08 mol l–1) and absolute uptake of N-nutrients (mean sum of NH4 +, urea, NO3 uptake rates: 0.141 and 0.087 mol l–1 h–1), specific uptake of 15N-labelled nutrients (NH4 +, urea, NO3 ) as well as the relationships between the measured variables characterize distinguishable inshore systems. The high variability at the shallow sampling sites prevents, however a simple resolution of the seasonal courses. Light dose could be identified as a potential key in order to describe long-term variations of N-uptake at the station with higher organic matter concentration (station KW), but phytoplankton development is better reflected in the seasonal course of N-uptake at the other station. Specific nitrogen uptake rates (NH4 +: 0.0009–0.0353 h–1, urea: 0.0001–0.0137 h–1, NO3 : 0.000004–0.0009 h–1) and relative nitrogen preferences indicate extraordinary importance of reduced nitrogenous nutrients (NH4 +, urea) at both stations throughout the year.  相似文献

Production of hydrogen peroxide has been found in Ulva rigida (Chlorophyta). The formation of H2O2 was light dependent with a production of 1.2 mol·g FW–1·h–1 in sea water (pH 8.2) at an irradiance of 700 mol photons m–2·s–1. The excretion was also pH dependent: in pH 6.5 the production was not detectable (< 5 nmol·g FW–1·h–1) but at pH 9.0 the production was 5.0 mol·g FW–1·h–1. The production of H2O2 was totally inhibited by 3-(3,4-dichlorophenyl)-1,1 dimethylurea (DCMU). The ability of U. rigida growing in tanks (7501) under a natural light regime to excrete H2O2 was checked and found to be seven times higher at 08.00 hours than other times of the day. The H2O2 concentration in the cultivation tank (density: 2 g FW·l–1) reached the highest value (3 M) at 11.00 hours. Photosynthesis was not influenced by H2O2 formation. The H2O2 is suggested to come from the Mehler reaction (pseudocyclic photophosphorylation). With an oxygen evolution of 120 mmol·g FW–1·h–1 at pH 8.2 and 90 mmol·g FW–1·h–1 at pH 9.0, 0.5% and 2.7% of the electrons were used for extracellular H2O2 production. Summary From acetylene reduction assays over a 10-month period starting in April 1979, nodule activities averaged 18.78 (se 4.67) moles C2H4 g nodule dw–1 h–1 forAlnus rubra and 59.95 (se 12.14) moles C2H4 g nodule dw–1 h–1 forCytisus scorparius. Plant rates were 1.91 (se. 47) moles C2H4 plant–1 h–1 forA. rubra and 0.55 (se. 17) moles C2H4 plant–1 h–1 forC. Scoparius. Plant activity and total leaf N were strongly correlated with the dw of other plant parts, but nodule activity and percent leaf N were not. Plant and nodule activities were not associated with temperature, moisture stress, precipitation events or percent light for either species over the growing season nor for 54A. rubra sampled in mid-season 1979 on one replication. After 5 to 6 growing seasons, 14A. rubra on the same site ranged from 30 to 332 cm in height and showed strong correlation between nodule dw, leaf dw, plant size and total leaf N. Results from this study and others indicate logistic equations may be modified to predict the effect of adding a N2 fixing plant to a population of non N2 fixing trees.  相似文献

The effects of a 60 min exposure to photosynthetic photon flux densities ranging from 300 to 2200 mol m–2s–1 on the photosynthetic light response curve and on PS II heterogeneity as reflected in chlorophyll a fluorescence were investigated using the unicellular green alga Chlamydomonas reinhardtii. It was established that exposure to high light acts at three different regulatory or inhibitory levels; 1) regulation occurs from 300 to 780 mol m–2s–1 where total amount of PS II centers and the shape of the light response curve is not significantly changed, 2) a first photoinhibitory range above 780 up to 1600 mol m–2s–1 where a progressive inhibition of the quantum yield and the rate of bending (convexity) of the light response curve can be related to the loss of QB-reducing centers and 3) a second photoinhibitory range above 1600 mol m–2s–1 where the rate of light saturated photosynthesis also decreases and convexity reaches zero. Summary The function of the caecal bulb, and its adaptation to chronic high- or low-Na+ intake, was investigated by in vivo perfusion of anaesthetised birds. Effects of acute aldosterone injection (125 g·kg–1 body mass) were also measured.Evidence was found for primary active net absorption of Na+, inducing parallel Na-linked absorption of water and Cl and secretion of K+. Around 20–35% of total Cl absorption and K+ secretion were independent of Na+ fluxes, and these components appear to be driven by passive processes with apparent conductances of 6.3×10–3 (G Cl) and 1.1×10–3 (G K) S·cm–2.Acetate (40mM) stimulated Na+ fluxes (8.5–9.9 Eq·cm–2·h–1) and Na-linked water fluxes (27–44 l·cm–2·h–1). Summary The kinetics ofBordetella pertussis growth was studied in a glutamate-limited continuous culture. Growth kinetics corresponded to Monod's model. The saturation constant and maximum specific growth rate were estimated as well as the energetic parameters, theoretical yield of cells and maintenance coefficient. Release of pertussis toxin (PT) and lipopolysaccharide (LPS) were growth-associated. In addition, they showed a linear relationship between them. A fermentation medium based on millet (Pennisetum typhoides) flour hydrolysate and a four-phase feeding strategy for fed-batch production of baker's yeast,Saccharomyces cerevisiae, are presented. Millet flour was prepared by dry-milling and sieving of whole grain. A 25% (w/v) flour mash was liquefied with a thermostable 1,4--d-glucanohydrolase (EC 3.2.1.1) in the presence of 100 ppm Ca2+, at 80°C, pH 6.1–6.3, for 1 h. The liquefied mash was saccharified with 1,4--d-glucan glucohydrolase (EC 3.2.1.3) at 55°C, pH 5.5, for 2 h. An average of 75% of the flour was hydrolysed and about 82% of the hydrolysate was glucose. The feeding profile, which was based on a model with desired specific growth rate range of 0.18–0.23 h–1, biomass yield coefficient of 0.5 g g–1 and feed substrate concentration of 200 g L–1, was implemented manually using the millet flour hydrolysate in test experiments and glucose feed in control experiments. The fermentation off-gas was analyzed on-line by mass spectrometry for the calculation of carbon dioxide production rate, oxygen up-take rate and the respiratory quotient. Off-line determination of biomass, ethanol and glucose were done, respectively, by dry weight, gas chromatography and spectrophotometry. Cell mass concentrations of 49.9–51.9 g L–1 were achieved in all experiments within 27 h of which the last 15 h were in the fedbatch mode. The average biomass yields for the millet flour and glucose media were 0.48 and 0.49 g g–1, respectively. No significant differences were observed between the dough-leavening activities of the products of the test and the control media and a commercial preparation of instant active dry yeast. Surficial sediment and sedimenting material were sampled during spring and summer 1991 in Lake Erken. Sediment was analyzed for redox potential, P concentrations and bacterial biomass. Sedimentation and chlorophyll a concentrations of sedimenting matter were determined. Additionally, different phosphorus forms in surficial sediment were quantified using sequential fractionation. The resulting dataset was used to study the effects of sedimentation events following phytoplankton blooms and benthic bacterial biomass on the size of the various phosphorus pools in the sediment.Sedimentation of spring diatoms caused a rapid increase in the NH4Cl- and NaOH-extractable P (NH4Cl–P and NaOH–rP) in the sediment. During sedimentation, NaOH–rP and NH4Cl–P increased within 3 days from 422 ± 17 g g–1 DW to 537 ± 8.0 g g–1 DW and from 113 ± 13 g g–1 DW to 186 ± 26 g g–1 DW, respectively. The NaOH–nrP (non-reactive P) fraction made up about 17% of Tot-P in sediment samples, whereas NaOH–rP and HCl–P made up 25% each. All P forms showed considerable seasonal variation. Significant relationships were found between bacterial biomass and the NaOH–nrP and NH4Cl–P fractions in the sediment, respectively. Also regressions of NaOH–nrP and NH4Cl–P versus the chlorophyll a concentration of sedimenting matter were highly significant. These regressions lend support to the conjecture that NaOH–nrP is a conservative measure of bacterial poly-P.  相似文献

Complete annual cycles of N2O and CH4 flux in forest soils at a beech and at a spruce site at the Höglwald Forest were followed in 1997 by use of fully automatic measuring systems. In order to test if on a microsite scale differences in the magnitude of trace gas exchange between e.g. areas in direct vicinity of stems and areas in the interstem region at both sites exist, tree chambers and gradient chambers were installed in addition to the already existing interstem chambers at our sites. N2O fluxes were in a range of –4.6–473.3 g N2O-N m–2 h–1 at the beech site and in a range of –3.7–167.2 g N2O-N m–2 h–1 at the spruce site, respectively. Highest N2O emissions were observed during and at the end of a prolonged frost period, thereby further supporting previous findings that frost periods are of crucial importance for controlling annual N2O losses from temperate forests. Fluxes of CH4 were in a range of +10.4––194.0 g CH4 m–2 h–1 at the beech site and in a range of –4.4––83.5 g CH4 m–2 h–1 at the spruce site. In general, both N2O-fluxes as well as CH4-fluxes were higher at the beech site. On a microsite scale, N2O and CH4 fluxes at the beech site were highest within the stem area (annual mean: 49.6±3.3 g N2O-N m–2 h–1; –77.2±3.1 g CH4 m–2 h–1), and significantly lower within interstem areas (18.5±1.4 g N2O-N m–2 h–1; –60.2±1.8 g CH4 m–2 h–1). Significantly higher values of total N, C and pH in the organic layer, as well as increased soil moisture, especially in spring, in the stem areas, are likely to contribute to the higher N2O fluxes within the stem area of the beech. Also for the spruce site, such differences in trace gas fluxes could be demonstrated to exist (mean annual N2O emission within (a) stem areas: 9.7±0.9 g N2O-N m–2 h–1 and (b) interstem areas: 6.2±0.6 g N2O-N m–2 h–1; mean annual CH4 uptake within (a) stem areas: –26.1±0.6 g CH4 m–2 h–1 and (b) interstem areas: –38.4±0.8 g CH4 m–2 h–1), though they were not as pronounced as at the beech site.  相似文献

Summary The kinetics of -mannanase (EC 3.2.1.77) and -mannosidase (EC 3.2.1.24) formation by a yeast cell wall lytic Arthrobacter species were studied. Growth () on yeast mannan was multiphasic and caused by mannose (=0.29 h–1) liberated by enzyme action from mannan. Early enzyme formation was soon repressed by mannose and depressed by its restricted availability during late exponential and stationary growth. Synthesis of -mannosidase occurred predominantly at the late stage of substrate utilization. Fructose was detected as an equally potent inducer for -mannanase formation as yeast mannan, being a simple and cheap substrate for large-scale cultivation. Growth on fructose was reduced (=0.20 h–1), enzyme synthesis being growth associated; nevertheless, comparable -mannanase levels [180 (U) units l–1] were formed. -Mannosidase activity was only detectable in small amounts. Continuous culture experiments gave values for maximal productivity of mannanase of 18 U h–1 g–1 and enzyme yield per biomass (Y EA/X) of 100 U g–1. Moreover, the substrate saturation constant (Monod constant) and maintenance coefficient were estimated for fructose as 115 mg l–1 and 4 mg h–1 g–1, respectively.  相似文献

Nitrification and denitrification rates were estimated simultaneously in soil-floodwater columns of a Crowley silt loam (Typic Albaqualfs) rice soil by an15N isotopic dilution technique. Labeled NO 3 was added to the floodwater of soil-water columns, half were treated with urea fertilizer. The (NO 3 +NO 2 )–N and (NO 3 +NO 2 )–N concentrations in the floodwater were measured over time and production and reduction rates for NO 3 calculated. Nitrate reduction in the urea amended columns averaged 515 mol N m–2h–1 and nitrification averaged 395 mol N m–2h–1 over the 35–153 d incubation. The nitrification rate for 4–19 d sampling period (1,560 mol N m–2h–1) in the urea amended columns was almost 9 times greater than the reduction rate (175 mol N m–2h–1) over the same period. Without the addition of urea the NO 3 production rate averaged 32 mol N m–2h–1 and reduction 101 mol N m–2h–1.  相似文献

Field studies to examine the in situ assimilation and production of ammonium (NH4 +) by bacterial assemblages were conducted in the northern Gerlache Strait region of the Antarctic Peninsula. Short term incubations of surface waters containing 15N-NH4 + as a tracer showed the bacterial population taking up 0.041–0.128 g-atoms Nl–1d–1, which was 8–25% of total NH4 + uptake rates. The large bacterial uptake of NH4 + occurred even at low bacterial abundance during a rich phytoplankton bloom. Estimates of bacterial production using 3H-leucine and -adenine were l.0gCl–1 d–1 before the bloom and 16.2 g Cl–1 d–1 at the bloom peak. After converting bacterial carbon production to an estimate of nitrogen demand, NH4 + was found to supply 35–60% of bacterial nitrogen requirements. Bacterial nitrogen demand was also supported by dissolved organic nitrogen, generally in the form of amino acids. It was estimated, however, that 20–50% of the total amino acids taken up were mineralized to NH4 +. Bacterial production of NH4 + was occurring simultaneously to its uptake and contributed 27–55% of total regenerated NH4 + in surface waters. Using a variety of 15N-labelled amino acids it was found that the bacteria metabolized each amino acid differently. With their large mineralization of amino acids and their relatively low sinking rates, bacteria appear to be responsible for a large portion of organic matter recycling in the upper surface waters of the coastal Antarctic ecosystem.  相似文献

Feeding rates of Daphnia pulex fed a range of levels of the alga Chlamydomonas reinhardi of 15 °C are strongly density-dependent. At lower densities, Daphnia (30 1–1) fed at higher rates than crowded (270 1–1) Daphnia which manifest a relatively depressed saturation feeding response. At 30 individuals/liter, Daphnia consumed 8.5 – 15.7 × 104 cells d–1h–1 (on a volume basis, 12.1 – 22.2 × 106 m3), at 270 L–1 3.7 – 3.9 × 104 (5.2 – 5.5 = 106 m3 cells d–1h–1 when feeding on algae at 80 000 cells ml–1 (11.3 × 106 m3 ml–1). The feeding rate data best fit an Ivlev feeding function. An autoallelopath might be causing the repression. Water preconditioned with crowded Daphnia completely repressed feeding in uncrowded Daphnia after six hours.  相似文献

Summary The maximum specific growth rate (max) of Hansenula polymorpha MH30 on xylose as the sole source of carbon and energy is 0.175 h–1, on methanol 0.21 h–1, on glycerol 0.27 h–1 and on glucose 0.61 h–1. On mixtures of xylose plus methanol, xylose plus glycerol, xylose plus glucose and glycerol plus glucose H. polymorpha MH30 grows faster: 0.36 h–1, 0.37 h–1, 0.47 h–1 and 0.52 h–1, respectively. Attempts have been made to explain these somewhat surprising results, especially the fact that the growth rates on xylose plus methanol and xylose plus glycerol exceed the specific growth rates of these on even the faster partner in the mixture. This paper provides an approach for optimizing the cell density (Xc) and dilution rate (D) in a chemostat for a Pichia pastoris continuous fermentation for the extracellular production of a recombinant protein, interferon (INF-). The objective was to maximize the volumetric productivity (Q, mg INF- l–1 h–1), which was accomplished using response surface methodology (RSM) to model the response of Q as a function of Xc and D within the ranges 150 Xc 450 g cells (wet weight) l–1 and 0.1 mD0.9 m (m=0.0678 h–1, the maximum specific growth rate obtained from a fed-batch phase controlled with a methanol sensor). The methanol and medium feed rates that resulted in the desired Xc and D were determined based on the mass balance. From the RSM model, the optimal Xc and D were 328.9 g l–1 and 0.0333 h–1 for a maximum Q of 2.73 mg l–1 h–1. The model of specific production rate (, mg INF- g–1 cells h–1) was also established and showed the optimal Xc=287.7 g l–1 and D=0.0361 h–1 for the maximum (predicted to be 8.92×10–3 mg–1 g–1 h–1). The methanol specific consumption rate (, g methanol g–1 cells h–1) was calculated and shown to be independent of the cell density. The relationship between and (specific growth rate) was the same as that discovered from fed-batch fermentations of the same strain. The approach developed in this study is expected to be applicable to the optimization of continuous fermentations by other microorganisms.  相似文献
