A method of measuring CO2gas exchange (caused, for example, by microalgal photosynthesis on emersed tidal mudflats) using open flow IR gas analyzers is described. The analyzers are integrated in a conventional portable photosynthesis system (LI-6400, LI-COR, Nebraska, USA), which allows manipulation and automatic recording of environmental parameters at the field site. Special bottomless measuring chambers are placed directly on the surface sediment. Measurements are performed under natural light conditions and ambient CO2concentrations, as well as under different CO2concentrations in air, and various PAR radiation levels produced by a LED light source built into one of the measurement chambers. First results from tidal channel banks in a north Brazilian mangrove system at Bragança (Pará, Brazil) under controlled conditions show a marked response of CO2assimilation to CO2concentration and to irradiance. Photosynthesis at 100molmol–1CO2in air in one sample of a well-developed algal mat was saturated at 309mol photons m–2s–1, but increased with increasing ambient CO2concentrations (350 and 1000mol mol–1CO2) in the measuring chamber. Net CO2assimilation was 0.8mol CO2m–2s–1at 100mol mol–1CO2, 5.9mol CO2m–2s–1at 350mol mol–1CO2and 9.8mol CO2m–2s–1at 1000mol mol–1CO2. Compensation irradiance decreased and apparent photon yield increased with ambient CO2concentration. Measurements under natural conditions resulted in a quick response of CO2exchange rates when light conditions changed. We recommend the measuring system for rapid estimations of benthic primary production and as a valuable field research tool in connection with certain ecophysiological aspects under changing environmental conditions.  相似文献

Summary The objectives of this study included directin vivo measurements of circulating blood gases, pH, heart rate, and blood pressure during voluntary dives of unrestrained Nile monitor lizards. A Radiometer flow-through cuvette was employed for continuous recording of arterial PO2, PCO2 and pH. Hematological properties revealed no particular adaptations for diving. Mean values were: hematocrit = 24%; hemoglobin concentration = 7.1 g %; oxygen capacity = 9.3 vol %; red cell dimensions = 22×12 ; red cell count = 0.67 million/l. The respiratory properties of the blood, studiedin vitro andin vivo, show distinct adaptations to habitual diving. Oxygen affinity of blood is low (P50 = 42.4 mm Hg at pH 7.45, 25 °) and the dissociation curve is markedly sigmoid (n = 3.1). These features, coupled with a Bohr factor ( logP 50/pH) of –0.48, ensure increased utilization of oxygen while maintaining relatively high tissue PO2. Arterial pH decreases during diving from about 7.5 to 7.1 due to combined respiratory and metabolic acidosis. High plasma bicarbonate (30 mM/l at PCO2 = 25 mm Hg) and a buffering capacity of H C3 O/ pH = 18.9 mM/l increase the tolerance to this acidosis and prolong diving time. Thein vivo oxygen dissociation curve shows a 90 % depletion of arterial oxygen content during typical dives. Diving elicited a rapidly developing bradycardia with maximum of 85 % reduction in heart rate. The temperature sensitivity of HbO2 binding was very low (H = –3kcal). This would minimize the HbO2 affinity increase accompanying the decrease in body temperature likely to occur in lizards going from sun basking to submergence in water.Supported by a grant from the Danish Natural Science Research Council.  相似文献

We have previously reported (Kim et al., J. Dairy Res., 1999, in press) three novel angiotensin-I-converting enzyme (ACE) inhibitory peptides, Tyr-Pro-Glu-Arg, Tyr-Tyr-Pro-Gln-Ile-Met-Gln-Tyr and Asn-Asn-Val-Met-Leu-Gln-Trp, isolated from the tryptic digest of recombinant human s1-casein. A series of C-terminal di- and tripeptides from these three peptides have now been synthesized and their ACE inhibitory activities have been measured. Among the peptides tested, the tripeptide Leu-Gln-Trp had the highest ACE inhibitory activity (IC50=3.8 mol l–1).  相似文献

Summary The enzymatic transformation of desacetyl-lanatoside A (DLA) to its secondary glycoside, digitoxin, in solutions of -and -cyclodextrins is effected using of -glucosidase from barley. Due to the interaction of cyclodextrins (CyDs) with desacetyl-lanatoside A, an increase in solubility of the latter of 24.5 and 230 times was observed for -cyclodextrin and -cyclodextrin, respectively. Kinetic studies of the enzymatic transformation gave for -glucosidase the values KM=3.3×10–4 mol. dm–3 and Vmax=0.557 mol mg–1 min–1 when the substrate was the deacetyl-lanatoside A complex with -cyclodextrin, while in the case of the complex with -cyclodextrin these values were KM=5.45×10–4 mol dm–3 and Vmax=0.896 mol mg–1 min–1.  相似文献

Behavioral and physiological responses to hypoxia were examined in three sympatric species of sharks: bonnethead shark Sphyrna tiburo, blacknose shark, Carcharhinus acronotus, and Florida smoothhound shark, Mustelus norrisi, using closed system respirometry. Sharks were exposed to normoxic and three levels of hypoxic conditions. Under normoxic conditions (5.5–6.4mg l–1), shark routine swimming speed averaged 25.5 and 31.0cm s–1 for obligate ram-ventilating S. tiburo and C. acronotus respectively, and 25.0cm s–1 for buccal-ventilating M. norrisi. Routine oxygen consumption averaged about 234.6 mg O2kg–1h–1 for S. tiburo, 437.2mg O2kg–1h–1 for C. acronotus, and 161.4mg O2 kg–1 h–1 for M. norrisi. For ram-ventilating sharks, mouth gape averaged 1.0cm whereas M. norrisi gillbeats averaged 56.0 beats min–1. Swimming speeds, mouth gape, and oxygen consumption rate of S. tiburo and C. acronotus increased to a maximum of 37–39cm s–1, 2.5–3.0cm and 496 and 599mg O2 kg–1 h–1 under hypoxic conditions (2.5–3.4mg l–1), respectively. M. norrisi decreased swimming speeds to 16cm s–1 and oxygen consumption rate remained similar. Results support the hypothesis that obligate ram-ventilating sharks respond to hypoxia by increasing swimming speed and mouth gape while buccal-ventilating smoothhound sharks reduce activity.  相似文献

Connective tissue of the freshwater pulmonateLymnaea stagnalis was shown to contain fucosyltransferase activity capable of transferring fucose from GDP-Fuc in 1–2 linkage to terminal Gal of type 3 (Gal1–3GalNAc) acceptors, and in 1–3 linkage to GlcNAc of type 2 (Gal1–4GlcNAc) acceptors. The 1–2 fucosyltransferase was active with Gal1–3GalNAc1-OCH2CH=CH2 (K m=12 mM,V max=1.3 mU ml–1) and Gal1–3GalNAc (K m=20 mM,V max=2.1 mU ml–1), whereas the 1–3 fucosyltransferase was active with Gal1–4GlcNAc (K m=23 mM,V max=1.1 mU ml–1). The products formed from Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4GlcNAc were purified by high performance liquid chromatography, and identified by 500 MHz1H-NMR spectroscopy and methylation analysis to be Fuc1–2Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4(Fuc1–3)GlcNAc, respectively. The Zn(II) binding by partial peptides of human protamine HP2: HP21–15; HP21–25, HP226–40, HP237–47, and HP243–57 was studied by circular dichroism (CD). Precipitation of a 20mer DNA by these partial peptides and the effects of Zn(II) thereon were investigated using polyacrylamide gel electrophoresis (GE). The results of this study suggest that reduced HP2 (thiol groups intact) can bind Zn(II) at various parts of the molecule. In the absence of DNA, the primary Zn(II) binding site in reduced HP2 is located in the 37–47 sequence (involving Cys37, His39, His43, and Cys47), while in the presence of DNA, the strongest Zn(II) binding is provided by sequences 12–22 (by His12, Cys13, His19, and His22) and 43–57 (His43, Cys47, Cys53, and His57). In its oxidized form, HP2 can bind zinc through His residues of the 7–22 sequence. Zn(II) markedly enhances DNA binding by all partial peptides. These findings suggest that Zn(II) ions may be a regulatory factor for sperm chromatin condensation processes.  相似文献

Diurnal variation in hydrological variables and dissolved inorganic nutrients such as PO inf4 sup3– -P, N O inf2 sup– -N, NO inf3 sup– -N and NH inf4 sup+ -N were studied in three interconnected biotopes including freshwater, marine and mangrove brackish water of the Kakinada coastal zone, Andhra Pradesh. Samples were collected at intervals of 3 hours, for a period of 24 hours. In the marine environment salinity varied from 26 to 32 whereas in the mangrove waters it fluctuated from 12 to 20 and in both biotopes salinity showed bimodal type of oscillation. Dissolved oxygen content was high in the mangrove waters during day time but decreased rapidly during the night hours. In the marine environment POf4 p3–-P concentration varied from 0.345 to 1.195 g at l–1, NO inf3 sup– -N from 1.03 to 6.62 g at l–1 and NO inf2 sup– -N from 0.086 to 0.506 g at l–1. The highest and the lowest concentrations of PO inf4 sup3– -P, NO inf3 sup– -N, NO inf2 sup– -N recorded in the mangrove waters were 0.790 and 0.325 g at l–1, 7.10 and 1.60 g at l–1 and 0.278 and 0.060 g at l–1, respectively. The concentration of PO inf4 sup3– -P, NO inf3 sup– -N and NO inf2 sup– -N were high in the freshwater canal, the maximum and minimum values being 1.110 and 0.730 g at l–1, 26.40 and 9.98 g at l–1 and 0.520 and 0.252 g at l–1 respectively. The concentration of ammonia was relatively high in the mangrove water. Gross and net primary production in the mangrove water was 4 times higher than in the marine biotope. There was no export of dissolved nutrients from the mangrove environment to the adjacent marine waters.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

DNA polymorphism patterns linked to the A-globin gene were analyzed in healthy Japanese using four different restriction endonucleases. The chromosomes with the A-globin gene were mapped through an evaluation of the presence of seven different restriction sites (HincII 5 to ; HindIII in G and A; HincII in, and 3 to, 1; AvaII in ; Bam-HI 3 to ). Among 36 chromosomes analyzed, 20 chromosomes had a haplotype of [+–––––+]. Among 55 individuals examined, 7 possessed a homozygous haplotye of [+–––––+]. All Japanese with the AT-globin gene had a subhaplotype of [–++–+] 5 to the -globin gene. Their major haplotypes were [–++–+–+] and [–++–++–]. It was expected that the presence of the AT-globin gene in Japanese may be deduced from subhaplotypes 5 to the -globin gene.  相似文献

Ministik Lake (longitude 113°01; latitude 53°21), a well-mixed, shallow (mean depth 1.83 m), eutrophic lake is characterized by eutrophic chlorococcalean and cyanophycean phytoplankton associations, and little change in standing crop size with increasing depth. Standing crops and primary productivity are low during the winter but pronounced spring and late summer/early autumn maxima occur. Mean yearly areal standing crop (B) and primary productivity (A) were 199.2 mg m–2 chlorophyll a and 319.5 mg C hr–1 m–2 respectively. Annual productivity was estimated at 1399,6 g C m–2yr–1. The mean increase in the extinction coefficient () per unit increase in standing crop (B) was 0.03 In units m–1. High non-algal light attenuation (q) occurred averaging 46% which prevented the ratio B from attaining more than 74.2% of the theoretical maximum except once when selfshading occurred. Insignificant relationships existed between B (mg m–3 chlorophyll a) and Amax (mg C hr–1 m–3), A and B and A and B. Close correlations existed between A and Amax/ and A and I0 (W m–2). The depth of the euphotic zone (Zeu) varied between 0.6 and 2.0 m; the average relationship between zeu and was zeu = 3.78/te, and the mean standing crop in the euphotic zone represented 58.3% of the theoritical maximum. The high q values made the model of Talling (1957) inapplicable to this lake. The Q10 value for the lake was 1.20. max (mg C chlorophyll a –1 hr–1) was closely related to both temperature and irradiance, and depressed by high pH values.Growth of the cyanophycean algae was correlated with temperature, chlorophycean algae with phosphate-phosphorus and temperature and the diatoms with dissolved silica and phosphate-phosphorus, but only in the case of Chaetoceros elmorii did any nutrient appear limiting. Indirect evidence that free CO2 limited photosynthetic rates is provided by the max:pH relationship.  相似文献

The eddy correlation technique was employed to measure net ecosystem carbon dioxide (CO2) (NEE) and water vapor exchange (LE) over a C3/C4 co-occurring wet temperate Miscanthus-type grassland in the Kanto plain of Japan in the 1999 growing season. The maximal mean canopy height and maximal leaf area index were 1.0m and 5.5, respectively. The daily maximal LE was approximately 540Wm–2. The maximum value of daily accumulative LE was 16.3MJday–1. Daily variation of the decoupling factor () suggests that in the morning LE decoupled with the atmosphere, and the available energy was the major driving force for LE, whereas in the afternoon LE coupled strongly with the atmosphere, and the atmospheric evaporative demand played a critical role in LE. The decline in (from 0.8 to 0.5) with the growing season demonstrates that LE decoupled from the atmosphere in the later growth season. The peak NEE value was 57.4µmolCO2m–2s–1 (the positive value signifies the canopy carbon gain was from the air). The maximal daily integrated NEE was 1.06molCO2m–2day–1 observed during the peak growth stage. A rectangular hyperbolic model was used to describe the relation between daytime NEE and incident photosynthetic photon flux density (PPFD). The net ecosystem CO2 was not light-saturated up to a PPFD level of 2000µmol m–2s–1. The initial slope estimated with the NEE–PPFD response model was approximately 0.042molCO2mol–1photon on average. The canopy light compensation point ranged from 210 to 430µmolm–2s–1 with an average of approximately 310µmolm–2s–1. Both the initial slope and the canopy light compensation point decreased as the canopy senesced. The switch in dominance from C3 to C4 plants played an important role in the canopy fluxes.  相似文献

Planktonic microbial interactions in the central basin of Lake Baikal were examined on a summer day in 1999. The subsurface maxima of bacterial abundance and chlorophyll concentration were recorded at the same depth, whereas the vertical distribution of heterotrophic nanoflagellates was the inverse of those of bacteria and picophytoplankton. Release of extracellular organic car-bon (EOC) from phytoplankton was estimated by the NaH14CO3 method as 2.4µgCl–1day–1. Bacterial production (4.3µgCl–1day–1), estimated in a bottle incubation experiment using size-fractionated water samples, exceeded the EOC released. Thus, other supplying sources of organic matter are needed for the bacterial production. Grazing (2.6µgCl–1day–1) was also estimated in the experiment and accounted for 60% of the bacterial production. This is the first report on the microbial food web in the central basin of Lake Baikal.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Enteropeptidase (enterokinase, EC 3.4.21.9) hydrolyzes peptide bonds formed by carboxyl groups of Lys or Arg residue if less than four negatively charged amino acid residues are in positions P 2P 5 of its substrate. We determined the kinetic parameters of three substrates of this type: human angiotensin II (AT) (DR VYIHPF) and the Hb(2–8) (LTAEEK A) and Hb(1–9) (MLTAEEK AA) peptides of the cattle hemoglobin -chain. The K m values for all the substrates (10–3 M) were one order of magnitude higher than those of the typical synthetic substrates of enteropeptidase or chimeric proteins with the –DDDDK– full-size linker (K m 10–4 M). The k cat values for AT and Hb(2–8) were also close and low (30 min–1). The general hydrolysis efficiency of such substrates is no more than 1% of the corresponding value for the typical peptide and protein substrates of the enteropeptidase. However, the elongation of Hb(2–8) peptide by one amino acid residue from both its N- and C-termini results in a dramatic increase in the catalytic efficiency of the hydrolysis: the k cat value for Hb(1–9) is 1510 min–1, which means that it is hydrolyzed only three times less effective than the chimeric protein with the full-size linker.  相似文献

In order to maintain axenic seedstock cultures axenically of thecommercially important red seaweed, Porphyra yezoensis, aprocedure was developed for axenic isolation and culture of conchocelis andmonospores. For axenic isolation of the conchocelis, contaminated microalgaewere most effectively removed by filtering contaminated samples through a100-m mesh after sonication. Removal of bacteria and otheralgaewas accomplished using a mixture of 5 agents (0.02% chitosan, 100 gml–1 GeO2, 10 gml–1 ampicillin, 40 gml–1 kanamycin and 200 gml–1 streptomycin). Axenic single colonies wereisolatedfrom a semi-solid medium prepared from 1% transfer gel. After collectingmonospores from the 40–50% density layer on a percoll-gradient, removalofbacteria and fungi from the monospores was accomplished using a mixture of 5antibiotics (3.5 g ml–1 nystatin, 2 mgml–1 ampicillin, 400 gml–1 kanamycin, 50 gml–1 neomycin and 800 gml–1 streptomycin). The production of erythritol and the erythritol yield from glucose by Torula sp. were improved, in increasing order, by supplementing with 10 mg MnSO44H2O l–1, 2 mg CuSO45H2O l–1, and both 10 mg MnSO44H2O l–1 and 2 mg CuSO45H2O l–1. Mn2+ decreased the intracellular concentration of erythritol, whereas Cu2+ increased the activity of erythrose reductase in cells. These results suggest that Mn2+ altered the permeability of cells, whereas Cu2+ increased the activity of erythrose reductase in cells.  相似文献

Whilst reliable estimates of chiasma frequencies can usually not be obtained, the probability (b) of a chromosome arm to be bound by at least one chiasma can often be determined. In the absence of interference this probability equals (1–e –2), where 2 is the average chiasma frequency of the chromosome arm and the average crossover frequency or map length. In the presence of interference is shown to retain its genetic meaning as an additive metric that may describe the chromosome arm or other distinctive chromosome segment in terms of genetic recombination. It is a form of potential map length, comparable to, but numerically different from the regular map length. It is termed provisionally crossing-over potential.A chromosome with armsm andn with crossing-over potentials and will form ring bivalents with a frequency (1–e –2).(1–e –2); open bivalents with a frequency (1–e –2).e –2+(1–e –2).e –2; univalent pairs with a frequencye –2.e –2. Estimates of these frequencies yield equations from which and may be solved.
