Type I collagen in tetrapods is usually a heterotrimeric molecule composed of two 1 and one 2 chains. In some teleosts, a third chain has been identified by chromatography, suggesting that type I collagen should also exist as an 1(I)2(I)3(I) heterotrimer. We prepared, from zebrafish, three distinct cDNAs identified to be those of the collagen 1(I), 2(I), and 3(I) chains. In this study on the evolution of fibrillar collagen chains and their relationships, an exhaustive phylogenetic analysis, using vertebrate fibrillar collagen sequences, showed that each chain constitutes a monophyletic cluster. Results obtained with the newly isolated sequences of the zebrafish showed that the 3(I) chain is phylogenetically close to the 1(I) chain and support the hypothesis that the 3(I) chain arose from a duplication of the 1(I) gene. The duplication might occur during the duplication of the actinopterygian genome, soon after the divergence of actinopterygians and sarcopterygians, a hypothesis supported by the demonstration of a syntenic evolution between a set of fibrillar collagen genes and Hox clusters in mammals. An evolutionary scenario is proposed in which phylogenetic relationships of the chains of fibrillar collagens of vertebrates could be related to Hox cluster history. Jacalin andM. pomifera agglutinin are T-antigen specific lectins with 44 structures that show far greater microheterogeneity than plant lectins from other families, due to multiple genetic isoforms and post-translational processing. Electrospray mass spectrometry and combined liquid chromatography-electrospray mass spectrometry were used to characterize the various forms. For both lectins, the mass data were consistent with previous protein sequencing of the major -chain species of 133 residues and three -chain species of 20 or 21 residues. In addition, for jacalin the mass of one minor -chain species was consistent with a second of the four reported gene sequences. However, the glycopeptide -chain form and one -chain form did not match any of the genes, suggesting a fifth gene remains to be found. ForM. pomifera agglutinin, three more -chain forms were found, but all six could arise from only two genes, with additional post-translational proteolysis and post-translational substitution with an unidentified component of 106 Da creating the set of six forms. Using immunoelectron microscopy and isoform-specific antibodies against Na,K-ATPase to study changes in Na,K-ATPase in rat erythroblastic cells during maturation, we unexpectedly observed numerous antigenic sites against the 3-isoform in the cytoplasmic phase. There was an increase in the number of 3-isoforms after denucleation of the erythroblast. The increase was transient. As the reticulocyte matured into a red blood cell, the number of 3-isoforms was reduced drastically. This 3-isoform was distributed in a reticular pattern resembling the double layers of endoplasmic reticulum. Western blot analysis confirms the presence of the 3-isoform in these cells. X-ray microanalysis of the erythroid series of cells in the bone marrow shows that sodium concentration in the young reticulocyte is higher than that in the nucleated erythroblast. The reason for the transient increase in this pump p rotein is not clear. It is possible that the increase in sodium concentration in the reticulocyte plays a role in the increase in pump protein synthesis.  相似文献

Zusammenfassung Die vorliegende Arbeit ist die 2. einer Reihe, deren Ziel die vergleichend-physiologische Untersuchung der Dynamik zentralnervöser Selbststeuerungssysteme sein soll. Die bei Anpassungsvorgängen auftretenden Änderungen des motorischen Verhaltens der Lebewesen werden als Schwingungsvorgänge im mathematischen Sinne aufgefaßt. Die Güte der Anpassung wird definiert als die zu der Einschwingungskurve gehörige Fläche.Ebenso wie die in der I. Mitteilung untersuchten Axolotl (Amblystoma) zeigen auch die hier benutzten Tische (Carassius carassius) periodisch gedämpften Einschwingungstyp. Dieser scheint bei niederen Wirbeltieren häufig zu sein (verbunden mit relativ großer Fläche, d. h. geringer Güte), während bei höheren Wirbeltieren, besonders beim Menschen, aperiodische Einschwingung (mit nahezu minimaler Fläche, also optimaler Güte) vorherrscht.Die Versuche bezogen sich hier nicht (wie bei der I. Mitteilung) auf die Anpassungsleistung von Einzeltieren, sondern auf die Verhaltensänderung eines Kollektivs. Da es sich bei diesen Fischen um ausgesprochene Schwarm-Tiere handelt, erschien dieser Weg als der biologisch richtigere.  相似文献

The dynamics of coupled biological oscillators can be modeled by averaging the effects of coupling over each oscillatory cycle so that the coupling depends on the phase difference between the two oscillators and not on their specific states. Average phase difference theory claims that mode locking phenomena can be predicted by the average effects of the coupling influences. As a starting point for both empirical and theoretical investigations, Rand et al. (1988) have proposed d/dt= — K sin ), with phase-locked solutions =arcsin( /K), where is the difference between the uncoupled frequencies and K is the coupling strength. Phase-locking was evaluated in three experiments using an interlimb coordination paradigm in which a person oscillates hand-held pendulums. was controlled through length differences in the left and right pendulums. The coupled frequency c was varied by a metronome, and scaled to the eigenfrequency v of the coupled system K was assumed to vary inversely with c. The results indicate that: (1) and K contribute multiplicatively to (2) =0 or = regardless of K when =0; (3) 0 or regardless of when K is large (relative to ); (4) results (1) to (3) hold identically for both in phase and antiphase coordination. The results also indicate that the relevant frequency is c/v rather than c. Discussion high-lighted the significance of confirming =arcsin(/K) for more general treatments of phase-locking, such as circle map dynamics, and for the 11 phase-entrainment which characterizes biological movement systems.  相似文献

The study, addressed to understand whether or not human platelets possess a unique thiol-oxidase whose activity could be modulated by signalling pathway initiated upon the activation of Receptor-Ck revealed the existence of disulphide-dependent oxidation within these cells and this phenomenon was regulated by Receptor-Ck-dependent generation of second messengers especially phosphatidic acid (PA); cAMP and cGMP. Purification of this activity revealed the existence of 47 kDa protein having thiol-oxidase activity. Keeping in view these results we propose that the existence of this novel 47 kDa Thiol-oxidase within human platelets may provide a crucial switch for the regulation of Receptor-Ck-dependent mevalonate pathway in human platelets.  相似文献

DNA molecules from lysates of microsporocytes at different stages of meiosis of Lilium longiflorum were examined in the electron microscope. In all of the preparations, only linear (non-circular) molecules were found. They had a mean length of between 59 and 75 , and a range of lengths which varied from 1 to 115 , to 1 to 210 . From the data obtained, there was no indication of a change in molecular form or size of DNA molecules associated with the process of meiosis.  相似文献

Summary Information regarding the relative levels of salt tolerance between cultivars of Kentucky bluegrass (Poa pratensis L.) is lacking. The objectives of this study were to 1) develop a simple, quick and sensitive method of screening turfgrass species for NaCl tolerance and 2) to compare the relative salt tolerance of five cultivars of Kentucky bluegrass (Ram I, Adelphi, Baron, Bensun, and Nassau) to other known salt tolerant turfgrass species such as alkalaigrass (Puccinellia distans (L.) Parl. cv. Fults) and two cultivars of red fescue (Festuca rubra L. Dawson, and Checker).Alkalaigrass and both cultivars of red fescue retained a high level of salt tolerance compared to the Kentucky bluegrass cultivars. Significant variability in salt tolerance was apparent among the Kentucky bluegrass cultivars with Adelphi and Ram I exhibiting the best overall tolerance.  相似文献

It has previously been suggested that inhibition of the proofreading 3-5 exonuclease activity of DNA polymerase may play an important role in generation of UV-induced mutations inEscherichia coli. Our previous work showing that overproduction of , the proofreading subunit of DNA polymerase III, counteracts the SOS mutagenic response ofE. coli seemed to be consistent with this hypothesis. To explore further the nature of the antimutagenic effect of we constructed plasmid pMK17, which encodes only two of the three highly conserved segments of — Exol and ExoII; the third segment, ExoIII, which is essential for 3–5 exonuclease activity, is deleted. We show that at 40°C, over-production of the truncated e subunit significantly delays production of M13 phage, suggesting that the protein retains its capacity to bind to DNA. On the other hand, the presence of pMK17 in atrpE65 strain growing at 40°C causes a 10-fold decrease in the frequency of UV-induced Trp+ mutations. This antimutagenic effect of the truncated s is effectively relieved by excess UmuD,C proteins. We also show that the presence of plasmid pIP21, which contains thednaQ49 allele encoding an subunit that is defective in proofreading activity, almost completely prevents generation of UV-induced mutations in thetrpE65 strain. We propose that the DNA binding ability of free , rather than its 3–5 exonuclease activity, affects processing of premutagenic UV-induced lesions, possibly by interfering with the interaction between the UmuC-UmuD-RecA complex and Pol III holoenzyme. This interaction is probably a necessary condition for translesion synthesis.  相似文献

The effects of biogenic amines, glucagon, and insulin on the cAMP-dependent protein kinase A (PKA) activity have been studied in the muscle tissue of the freshwater bivalve mollusc Anodonta cygnea. It was shown that serotonin, glucagon, and insulin both in vivo and in vitro stimulated PKA activity, whereas isoproterenol inhibited it. The stimulating effect of serotonin and inhibiting effect of isoproterenol was blocked by serotoninergic (cyproheptadine) and adrenergic (propranolol) inhibitors, which confirms specificity of the effect of biogenic amines on the PKA activity. Taking into account participation of adenylyl cyclase system in action of the above hormones, the revealed hormonal effects on the PKA activity produce metabolic effects via the following chain reaction. In the case of serotonin and glucagon: receptor Gs-protein AC cAMP PKA phosphorylation of glycogen synthase (GS) and glucose-6-phosphate dehydrogenase (G6PDH) and inhibition of their activity; in the case of isoproterenol: -adrenoreceptor Gi-protein AC inhibition decreasing PKA inhibition of phosphorylase and stimulation of GSI and G6PDH. A participation is suggested of the insulin-stimulated AC signaling system in the mechanism of the mitogenic insulin effect mediated, as shown in this work, via the PKA activation, but not of the metabolic effect of insulin.  相似文献

The amounts of a 160-kDa amylase and a 140-kDa -amylase (A. Burgess-Cassler and S. H. Imam, Curr. Microbiol. 23:207–213, 1991) secreted into culture medium by the starchutilizingLactobacillus amylovorus were enhanced by the use of cyclodextrin (CD) as the carbon source. The levels of total extracellular -amylase obtained with glucose as the carbon source could be boosted severalfold by use of CD. The best enhancer was -CD, and the rank order of best to least effective was -CD>-CD=-CD>glucose.Another amylase, a 65-kDa -amylase, which degraded para-nitrophenyl-(1,4)-d-glucopyranoside, was also detected in this study. The most effective enhancer in this case was -CD, and the rank order was -CD>-CD>-CD glucose. Despite its ability to degradep-nitrophenylated glucose, this enzyme did not convert maltose to glucose. It showed a cleared zone on starch zymograms and did degrade short maltodextrins to maltose. Neither this new -amylase nor the 140-kDa -amylase exhibited any detectable ring-decyclizing (cyclodextrinase) activity against -or -CD.Other extracellular amylases (not characterized here) appeared to be similarly enhanced by CDs. Although the precise mechanism by which this effect is accomplished remains undefined, CDs can be useful inducing agents, boosting the expression and/or secretion of otherwise low-level enzymes, either as additives to growth media or as sole carbon source.  相似文献

Summary We have identified a common restriction fragment length polymorphism of the fibrinogen gene with the enzyme TaqI. This polymorphism is probably due to a single base change that creates or destroys a TaqI recognition site about 1000 basepairs from the 3 end of the fibrinogen géne. The frequency of the rare allele in 83 unrelated healthy individuals is 0.33. We have used in situ hybridisation of the fibrinogen cDNA to localise the gene on chromosome 4q29–31. We have confirmed this regional localisation by restriction fragment detection in a human x Chinese hamster somatic cell hybrid which contains a translocated human chromosome 4 with a breakpoint at 4q26. Summary Darwin's phrase place in natural economy, andSpencer's term correspondence can be regarded as first attempts to express the organism-environment relationships. The same concept has more recently been approached from the point of view of (1) life-form, (2) external activities, and (3) habitat. Though all these points are interlocking, they have been stressed differently in the writings of American and European ecologists. It is proposed that the term niche would be most useful and rational if applied to the total of relationships between a living organism (population, species) and its complete environment, both biotic and abiotic.  相似文献

Zusammenfassung Im Buchenwald des Naturschutzgebietes Serrahn (Mecklenburg) achtete Verf. seit 1949 auf den Zwergschnäpper. 1956 und 1957 brüteten dort über 12 Paare.Ankunft frühestens am 9. Mai. Als Erste erscheinen vorwiegend ältere (rotkehlige) . Den ersten folgen sehr bald die ersten .Balzflug, Zeigen der Nisthöhle, Copula sowie Variationen des Neststandes werden beschrieben.Fast stets baute nur das , in einem Falle 3 Tage lang. Während der Bauzeit setzt das seine Singflüge eifrig fort. Gepaarte verstummen, sobald ihr begonnen hat, fest auf dem Gelege zu brüten. Nur Junggesellen singen den ganzen Sommer über.An einem günstig gelegenen Nest wurde das Verhalten des Paares vom 4.(?) Bruttag ab bis zum Ausfliegen der Jungen (im Alter von 13 Tagen) aus einem dicht davor angebrachten Schirm genau beobachtet und aufgezeichnet. Nach dem Verlassen des Nestes suchen die Jungen unter Führung ihrer Eltern die nächste Dickung auf. Sobald sie selbständig geworden sind, beginnt die Jugendmauser, 3 Wochen danach die Wanderung ins Winterquartier. Die letzten Zwergschnäpper verschwanden meist vor Mitte September.Angaben über Gesang, Bedeutung der Rufe, Nahrung.Attrappen-Versuche am Nest ergaben, daß die Eltern genau zwischen dem (gefährlichen) Sperber und dem (ungefährlichen) Kuckuck zu unterscheiden wußten und ihnen gegenüber unterschiedliche Alarmlaute brachten. Many genes have been located in wheat chromosomes, yet little is known about the location of genes for resistance to Ustilago tritici, which causes loose smut. Crosses were made between the loose smut susceptible alien substitution lines Cadet 6Ag(6A) and Rescue 6Ag(6A) (lines in which Agropyron chromosome 6 is substituted by wheat chromosome 6A) and four cultivars resistant to U. tritici race T19: Cadet, Kota, Thatcher and TD18. The segregating progeny were tested for reaction to race T19 and for the level of binding with a monoclonal antibody specific to a chromosome 6A-coded seed protein. The antibody, which does not bind to seed protein extracts in the absence of the 6A chromosome, was used as a chromosome marker. An association was established between resistance to race T19 and the presence of chromosome 6A for each of the cultivars tested, indicating that resistance to race T19 resides in chromosome 6A. Ustilago tritici race T19 resistance in Cadet appears to be located in the short arm of chromosome 6A, based on the evaluation of the Cadet 6A long ditelosomic stock, which was susceptible, and the Cadet 6A-short: 6-Agropyron- short alien translocation stock, which was resistant.  相似文献

Ever since the early decades of this century, there have emerged a number of competing schools of ecology that have attempted to weave the concepts underlying natural resource management and natural-historical traditions into a formal theoretical framework. It was widely believed that the discovery of the fundamental mechanisms underlying ecological phenomena would allow ecologists to articulate mathematically rigorous statements whose validity was not predicated on contingent factors. The formulation of such statements would elevate ecology to the standing of a rigorous scientific discipline on a par with physics. However, there was no agreement as to the fundamental units of ecology. Systems ecologists sought to identify the fundamental organization that tied the physical and biological components of ecosystems into an irreducible unit: the ecosystem was their fundamental unit. Population ecologists sought, instead, to identify the biological mechanisms regulating the abundance and distribution of plant and animal species: to these ecologists, the individual organism was the fundamental unit of ecology, and the physical environment was nothing more than a stage upon which the play of individuals in perennial competition took place. As Joel Hagen has pointed out, the two schools were thus dividied by fundamentally different and irreconcilable assumptions about the nature of ecosystems.Notwithstanding these divisive efforts to elevate the image of ecology, the discipline remained in the shadows of American academia until the mid-1960s, when systems ecologists succeeded in projecting ecology onto the national scene. They did so by seeking closer involvement with practical problems: they argued before Congress that their approach to the theoretical problems of ecology was uniquely suited to the solution of the impending environmental crisis. With the establishment of the International Biological Program, they succeeded in attracting unprecedented levels of funding for systems ecology research. Theoretical population ecologists, on the other hand, found themselves consigned to the outer regions of this new institutional landscape. The systems ecologists' successful capture of the limelight and the purse brought the divisions between them and population ecologists into sharper relief — hence the hardening of the division of ecology observed by Hagen.45  相似文献

Summary Growth of the fungus Penicillium charlesii G. Smith on glucose, minimal salts medium results in the appearance of -d-mannopyranosidase activity capable of hydrolyzing p-nitrophenyl--d-mannopyranoside. No activity is found until depletion of the carbon source, after which the enzyme activity rapidly increases in the mycelium. Prolonged incubation of the culture results in the appearance of small amounts of -mannosidase activity in the growth medium. The initial release of a mannose-containing polysaccharide into the medium precedes the appearance of -mannosidase by several days.  相似文献

Carboxyl groups of HbS are readily activated by water-soluble carbodiimide atpH 6.0 and room temperature. These o-acylurea intermediates (activated carboxyl) are accessible for nucleophilic attack by amines. With glycine ethyl ester, the amidation is very selective for the -carboxyl of Glu-43() and more than 65% of the glycine ethyl ester incorporated is on this carboxyl group. In contrast, glucosamine derivatizes the -carboxyl group of Glu-22() as well as that of Glu-43() to nearly the same degree. However, the total amidation of HbS by glucosamine is lower than that with glycine ethyl ester. The differential selectivity of the two amines is apparently related to the differences in the microenvironment of the -carboxyl groups of Glu-22() and Glu-43(), which either facilitates or refracts the aminolysis of the activated carboxyl with the two amines to different degrees. The carboxyl groups of isolated -chain exhibit a higher reactivity for amidation with glycine ethyl ester than does the tetramer. The carboxyl groups of Glu-22() and Glu-43() and that of Asp-47() are all activated by carbodiimide suggesting that the higherpKa of these carboxyl groups (facilitating the activation) is a property of tertiary interaction of the polypeptide chain. The interaction of the -chain with -chain, i.e., generation of the quaternary interactions, reduces overall reactivity of the carboxyl groups of the protein. The higher selectivity of hemoglobin S for amidation at Glu-43() with glycine ethyl ester compared with that of isolated -chain appears to be primarily a consequence of decreased amidation at sites other than at Glu-43().  相似文献

Diatoms are valuable indicators of palaeosedimentary environments in coastal wetlands. In this paper, the utilization of diatoms in coastal reconstructions will be demonstrated in a case study, the Poortvliet boring (southwest Netherlands). Diatoms supply valuable information about palaeosalinity gradients and palaeotide levels in addition to the existing geological data set, obtained from sediment cores.The ecological data of taxa are usually gathered from diatom studies in recent coastal environments. The study of the Poortvliet boring shows that information can also be obtained from the past (the past as key to the past) by comparing the trends of the relative abundances of species of which the ecology is well-known to those of which the ecology is unknown. It is argued both on recent and fossil data that Cymatosira belgica and Rhaphoneis minutissima have a tychoplanktonic life form.  相似文献

Data are presented to evaluate further the abundance/biomass comparison method for the detection of pollution disturbance in marine benthic communities. The results suggest that this technique cannot be used in estuarine intertidal communities. The grossly polluted configurations appeared under unpolluted conditions, while undisturbed patterns were found under polluted conditions.
