采用PCR技术从rec-M13mp18中扩增出120 bp的大鼠肝tRNAIle合成基因片段,经限制性内切酶BstNⅠ酶切后作为模板,利用T7 RNA聚合酶在体外无细胞体系转录由T7启动子带动的大鼠肝tRNAIle基因,生成不含修饰碱基的tRNAIle,并对体外转录反应条件进行了优化,回收的tRNA产量可达DNA模板量的40倍.  相似文献

Summary DNA synthesis in vitro using intact duplex T7 DNA as template is dependent on a novel group of three phage T7-induced proteins: DNA-priming protein (activity which complements a cell extract lacking the T7 gene 4-protein), T7 DNA polymerase (gene 5-protein plus host factor), and T7 DNA-binding protein. The reaction requires, in addition to the four deoxyribonucleoside triphosphates, all four ribonucleoside triphosphates and is inhibited by low concentrations of actinomycin D. Evidence is presented that the priming protein serves as a novel RNA polymerase to form a priming segment which is subsequently extended by T7 DNA polymerase. T7 RNA polymerase (gene 1-protein) can only partially substitute for the DNA-priming protein. At 30°C, deoxyribonucleotide incorporation proceeds for more than 2 hours and the amount of newly synthesized DNA can exceed the amount of template DNA by 10-fold. The products of synthesis are not covalently attached to the template and sediment as short (12S) DNA chains in alkaline sucrose gradients. Sealing of these fragments into DNA of higher molecular weight requires the presence of E. coli DNA polymerase I and T7 ligase. Examination of the products in the electron microscope reveals many large, forked molecules and a few eye-shaped structures resembling the early replicative intermediates normally observed in vivo.  相似文献

Coding sequences for a hammerhead ribozyme designed to cleave lexA mRNA in a targeted manner was cloned under phage T7 promoter and expressed in E. coli strain BL-21 (DE3) expressing T7 RNA polymerase under the control of IPTG-inducible lac UV-5 promoter. Ribozyme expression in vivo was demonstrated by RNase protection assay. Also, total RNA extracted from these transformed cells following induction by IPTG, displays site-specific cleavage of labeled lexA RNA in an In vitro reaction. The result demonstrates the active ribozyme in extracts of cell transformed with a recombinant cassette and goes beyond the earlier demonstration of the stability of In vitro synthesized ribozyme in cell extracts. The observed rise in lexA mRNA rules out any role for protease activity or resulting fragments of lexA protein in de-repression of RNA. (Mol Cell Biochem 271: 197–203, 2005)  相似文献

Background  

The DNA-dependent RNA polymerase from T7 bacteriophage (T7 RNAP) has been extensively characterized, and like other phage RNA polymerases it is highly specific for its promoter. A combined in vitro / in vivo selection method has been developed for the evolution of T7 RNA polymerases with altered promoter specificities. Large (103 – 106) polymerase libraries were made and cloned downstream of variant promoters. Those polymerase variants that can recognize variant promoters self-amplify both themselves and their attendent mRNAs in vivo. Following RT / PCR amplification in vitro, the most numerous polymerase genes are preferentially cloned and carried into subsequent rounds of selection.  相似文献

Stimulation of T7 DNA polymerase by a new phage-coded protein   总被引:10,自引:0,他引:10  
Summary A bacteriophage-induced DNA-binding protein was purified from T7 infected E. coli. The protein has a molecular weight of about 25000, as judged by SDS-polyacrylamide gel electrophoresis. The purified protein binds to single-stranded but not to native T7 DNA. Like the T4 gene-32 protein and the 22000-dalton unwinding protein of E. coli, the T7 25000 protein lowers the melting temperature of poly d(A-T). Using partially single-stranded T7 DNA as template-primer, the protein stimulates in vitro DNA synthesis by T7 DNA polymerase about five-fold. It was also found that the DNA-unwinding protein of E. coli stimulates T7 DNA polymerase to approximately the same extent. However, neither of the unwinding proteins stimulate DNA polymerase I of E. coli.  相似文献

Summary Spermidine has a differential influence on T3 and T7 DNA-directed enzyme synthesis in vitro on the translational level. T7 early RNAs were synthesized in vitro by transcribing T7 DNA with Escherichia coli RNA polymerase and treating the resulting precursor molecule with ribonuelease III. Oligonucleotide fragments from the 5′ and 3′ termini of several of the cleaved species were then selectively isolated. Structural analysis revealed sequences identical to the corresponding in vivo RNAs. Thus, the T7 early RNAs found in phage-infected cells appear to be the direct products of RNAase III cleavage of a large precursor molecule. We conclude further that RNAase III action on this particular natural substrate is a sequence-specific event.  相似文献

Summary Enzyme synthesisin vitro directed by T7 heteroduplex DNA is observed when the H strand [the strand assuming the higher density in combination with poly(G,U)], carries the wild allele. This is true for enzyme synthesis mediated byE. coli RNA polymerase or by T7 phage RNA polymerase. The full-length gene for Marburg virus (MV) nucleoprotein (NP) was cloned in prokaryotic pQE32 under the control of the T5 promoter and in eukaryotic pTM1 under the control of the T7 RNA polymerase promoter. Recombinant NP was synthesized in Escherichia coliand in human kidney cell line 293 cotransfected with recombinant vaccinia virus vTF7-3 expressing T7 RNA polymerase. On evidence of electron microscopy with immune detection, recombinant NP formed tubules of two types in E. coliand of a single type in cell line 293. ELISA and immunoblotting with polyclonal and monoclonal antibodies revealed common antigenic determinants in recombinant NP and natural MV NP.  相似文献

Analysis of the nucleotide sequence at the 5′-triphosphate termini of RNA chains synthesized by T7 RNA polymerase from T7 DNA template indicates that nearly all RNA chains synthesized in this polymerase reaction contain the sequence, pppGpGp. In addition, studies carried out on T7 DNA-dependent 32PPi exchange into ribonucleoside triphosphates suggest that immediately following the guanine residues at the 5′-end of RNA formed in the T7 RNA polymerase reaction, there is one or more adenine residues. These results indicate a high degree of specificity of initiation of RNA synthesis by T7 RNA polymerase.  相似文献

SYNOPSIS. DNA-dependent RNA polymerases have been solubilized from homogenates of Crithidia fasciculata using gentle extraction procedures. RNA polymerase I and II are separated on DEAE cellulose at 0.07M (NH4)2SO4 and 0.13M (NH4)2SO4 respectively. RNA polymerase II is inhibited 80% by α-amanitin (25 μg/ml). Both RNA polymerases require DNA as a template, ribonucleoside triphosphates and Mn2+. The synthesis of RNA as a product is inhibited by DNase. RNase, pronase and actinomycin D. Purified kinetoplast and nuclear DNA can serve as templates for the RNA polymerases. Denatured DNA templates are preferred. The synthesis of RNA continues for at least an hour and is inhibited by trypanocidal drugs including suramin. antrycide, acriflavine, ethidium bromide and berenil. Complementary RNA synthesized in vitro from C. fasciculata kinetoplast DNA hybridizes with C. fasciculata kinetoplast DNA but not with C. fasciculata nuclear DNA or Blastocrithidia culicis kinetoplast DNA, Escherichia coli, T4 or calf thymus DNAs. The complementary RNA synthesized in vitro from C.fasciculata kinetoplast DNA sediments at 4–5S.  相似文献

A DNA-membrane complex isolated from Escherichia coli infected with bacteriophage T7 contains newly synthesized T7 DNA and the T7 DNA polymerase (gene 5 product). The DNA present in the complex appears to exist as a concatemer which contains single-strand breaks and possibly internal single-stranded regions (gaps). The complex is capable of synthesizing T7 DNA by using endogenous template, and part of the DNA is made by a semiconservative mechanism. A portion of the in vitro synthesized DNA sediments in alkaline sucrose as 10-11S material. This DNA is converted to a larger-molecular-weight material after treatment with T4 polynucleotide ligase and E. coli DNA polymerase I.  相似文献

为了建立一种适于实验室乃至常规定量检测mRNA表达的、可快速构建cRNA标准曲线的方法,设计带有T7启动子序列和PolyT序列的引物对目的基因和内参照进行PCR,克隆入载体作为体外合成cRNA的模板,快速构建cRNA标准.结果表明:该曲线的线性范围至少达6个数量级,相关系数为0.99.该法快速、简便,适用于所有靶基因.  相似文献
