Availability of seeds and provision of “safe sites” for seedling recruitment are essential for successful restoration of seminatural grassland communities. Inability to provide species‐specific conditions for seedling recruitment appears to be a major factor limiting establishment of fen‐meadow species on restoration sites. This contention was tested in the field for both germination and establishment conditions for a selection of fen‐meadow species. A Cirsio‐Molinietum fen meadow and an agriculturally semi‐improved species‐poor grass dominated rush pasture were used. Seeds of Carex ovalis, Cirsium dissectum, Molinia caerulea, Succisa pratensis, and Holcus lanatus were sown onto treatments comprising either irrigation or no irrigation, presence or absence of existing vegetation canopy, and presence or absence of soil disturbance. Germination of all except H. lanatus was higher in the fen meadow than in the rush pasture. The fen‐meadow site was less susceptible to drought, provided more light to the seed environment, and showed a stronger day–night variation in relative humidity compared with the rush pasture. All the fen‐meadow species responded strongly to the experimental treatments, whereas H. lanatus showed only a small response. Soil disturbance was the major factor that increased germination. Removal of the vegetation canopy improved germination only in S. pratensis. Conditions affecting survival of seedlings were different from those affecting seed germination. Seedling survival was greater on the fen‐meadow site than on the rush pasture. Canopy presence was the major factor that reduced seedling survival. Few seedlings survived in the presence of the rush pasture canopy. Irrigation and soil disturbance were of minor importance for seedling survival on both sites. Safe sites for seed germination and seedling establishment of fen‐meadow species existed on the fen meadow even without soil disturbance and gap creation. Safe sites for seedling recruitment were not present in the rush pasture. The need for species‐specific definition of safe site characteristics at the two stages of seedling recruitment (i.e., for seed germination and for seedling survival) was demonstrated. The implications of these findings for restoration of seminatural grasslands are discussed. 相似文献

We determined the effects of zeatin (ZEA), isopentenyl adenine (2iP), kinetin (KIN), benzyladenine (BA), and thidiazuron (TDZ)
on seed germination, elongation of seedling shoots and roots, frequency of regeneration, and the number of regenerants per
seedling in Lotus corniculatus L. Sterilized seeds were cultured in vitro on Murashige and Skoog (1962) medium containing 3% sucrose, 0.7% agar, and various cytokinins (0, 0.08, 0.22, 0.35, 0.80, 2.20, and 3.50 μM). After 30
days, seedlings were transferred to cytokinin-free medium for another 60 days. All cytokinins stimulated the rate and percentage
of seed germination at least twofold in optimum concentrations; TDZ and ZEA were the most active, followed closely by BA,
whereas KIN and 2iP stimulated germination in higher concentrations only. Elongation of shoots and roots was strongly inhibited
at the lowest TDZ and BA concentrations, whereas ZEA, KIN, and 2iP exerted moderate, dose-dependent inhibition. The frequency
of regenerant-producing seeds was highest on ZEA and BA, whereas the greatest number of regenerants per seedling was found
on TDZ. It is concluded that the culture of seeds on cytokinin-containing media, followed by transfer to cytokinin-free medium,
is a suitable procedure for rapid production of a large number of uniform regenerants. The presumed role of particular cytokinins

Endogenous cytokinins were quantified and identified in germinating achenes and developing seedlings of Tagetes minuta L. incubated at 25 °C over a 144 h period. The process of germination (radicle emergence) was completed 38 h after commencement
of imbibition. Subsequent growth was considered to cover seedling establishment. Eighteen isoprenoid cytokinins, belonging
to the zeatin (9), dihyrozeatin (5) and isopentenyladenine (4) groups and one aromatic cytokinin, benzyladenine, were identified.
The total isoprenoid cytokinin concentration increased upon imbibition, reached a peak by 48 h and subsequently decreased
with seedling development. The individual cytokinin groups and the respective derivatives within each group did, however,
not follow such a consistent trend. During the course of the experiment, the ribotides and ribosides were present in the highest
concentrations, reaching a peak at 48 h and decreasing thereafter. The free bases and O-glucoside remained at low levels throughout
the experiment. Isopentenyladenine-9-glucoside increased dramatically in the developing seedlings and after 144 h was the
predominate cytokinin. Benzyladenine was the only aromatic cytokinin detected throughout the experiment. It was present in
high concentrations in the dry achenes and declined rapidly upon imbibition. 相似文献

Using the soybean callus bioassay it has been shown that dormant lettuce seeds ( Lactuca sativa L. cv. Grand Rapids) contain large amounts of water soluble cytokinins and small amounts of butanol soluble ones. When the seeds are irradiated with red light, or imbibed with 5 mg/1 gibberellic acid in the dark, the total cytokinin content of the seeds decreases, the level of water soluble cytokinins decreases, and the level of the butanol soluble cytokinins increases. Far-red light does not reverse this effect completely although cytokinin activity in the butanol extracts decreases following such irradiation. It is proposed that the interconversion of cytokinins initiated by red light, or gibberellic acid in the dark, is one of the primary events leading to radicle elongation in light-sensitive lettuce seed. 相似文献

Experiments with Grand Rapids lettuce seeds ( Lactuca sativa L.) maintained in darkness or irradiated with red light have shown that the inhibition of germination induced by low concentrations of ABA (2, 4, 6 μ M) could be overcome by gibberellins (GA 3 or GA 4). The same results were obtained, although to a lesser extent, under the influence of two out of the four cytokinins tested (K and BAP) for seeds maintained in darkness. To suppress the block induced by higher concentrations of ABA (for example 8 μ M), it was necessary to apply a cytokinin (K, BAP, Z or 2iP) and a gibberellin (GA 4 or GA 3) simultaneously, or a cytokinin following a red light treatment. Experiments conducted in darkness in which ABA (8 μ M) was applied together with a cytokinin (BAP) and a gibberellin (GA 4) showed that the gibberellin and the cytokinin played similar roles towards each other and towards ABA. 相似文献

The primary site of photosynthetic carbon dioxide fixation wasdetermined in the leaves of two monocot species by means ofmicroautoradiographic techniques. We have used antibodies directed against the 16.5 kilodalton protein L3, the most abundant integral protein of maize ( Zea mays L. cv Mo 17) lipid bodies, to follow the fate of this protein in scutellar parenchyma cells of maize during germination and subsequent seedling growth. Using gel electrophoresis and immunoblotting as well as immunocytochemical electron microscopy, we found that the amount of L3 decreases gradually during the first 3 to 4 days of seedling growth and more rapidly over the course of the next several days. Immunogold localization of the protein on thin sections indicated that L3 is found exclusively in the surface phospholipid monolayer of lipid bodies. The density of L3 in the surface layer of individual lipid bodies does not change during seedling growth; therefore, the decrease in the amount of L3 can be attributed to a decrease in the number of lipid bodies rather than to selective removal of protein components from the surface of all lipid bodies. Thus, L3 is apparently degraded at the same time as the matrix lipid of each lipid body. Unlike lipase, L3 does not appear to be transferred to other cellular compartments such as vacuoles during late stages of seedling growth. 相似文献

Five cereals and two related grasses were tested for adventitious shoot production from tissue cultures using methods concordant with those reported to be successful for cereals. The five cereals I wheat ( Triticum aestivum L.), oats ( Avena sativa L.), and maize ( Zea mays L.) Pioneer hybrid 3369A, the Bolivian race Pororo and the Equadorian race Chococenõl were all found to proliferate in culture through an aberrant root-like mechanism of growth which had the external appearance of callus. Two related species, teosinte ( Zea mexicana Reeves and Mangelsdorf) and tripsacum ( Tripsacum dactyloides L.), were less successful in culture, but grew in the same way. Oats, and probably Pororo and Chococeño, initiated presumptive shoot meristems directly from root vascular tissues within this root-like growth. Hybrid maize and wheat initiated no shoot meristems and produced only roots. The occasional shoot production observed in wheat was discounted as simple carryover of existing shoot apices from the primary embryo cultures. This study suggests that the incidence of shoot regeneration in cultures of these cereals may be related more directly to adventitious bud formation on roots than to any controlled de novo organogenesis from undifferentiated callus. 相似文献

The changes occurring in the axis of the developing seedlingof Theobroma cacao L. were observed over a 96-h period. It wasfound that during the first 24-h period, greatest change hadoccurred in the hypocotyl; the epicotyl and the root meristemhad apparently remained dormant. In the hypocotyl, vascularizationwhich had begun in the mature embryo continued: starch grainshad disappeared from the cells of the cortex and pith, the latterof which were seen to be binucleate. During the second 24-hperiod, greatest change was seen to have taken place in theroot meristem, resulting in the production of a root which consistedof a central pith, a poly-arch stele, and cortical tissue. Betweenthe root and the hypocotyl an apparently undifferentiated regionwas observed. This region was seen to have differentiated duringthe third 24-h period, leading to the production of a completering of xylem, external to which were groups of phloem, theformer being separated from the latter by parenchymatous tissue.This region is interpreted as being transitional between rootand shoot. During the fourth 24-h period, the adventitious andlateral root primordia initiated earlier, were seen to havedifferentiated to the extent that the disposition of their tissueswas evident. 相似文献

The objective of this study was to evaluate the ability of the phytohormone S-abscisic acid (S-ABA) to protect maize seedlings grown under drought stress and to measure their increased drought tolerance. The maize hybrids ‘Zhengdan 958’ (ZD958; drought tolerant) and ‘Xundan 20’ (XD20; drought sensitive) were treated with nutrient solutions of different concentrations (1, 2, 4, 8, and 10 mg/kg) of S-ABA under polyethylene glycol (PEG, 15% w/v, MW 6000) simulated drought stress. Optimal concentrations of S-ABA were designed to be sprayed onto the leaves of seedlings, and their effect on endogenous ABA, malondialdehyde (MDA), osmotic substances, antioxidant enzyme activities, and Asr1 gene expression in seedlings were studied. Results indicated that, under drought stress, S-ABA treatment significantly improved maize seed germination rate (GR), germination energy (GE), and seedling biomass (p < 0.05). After spraying 4 mg/kg S-ABA onto leaves, the endogenous hormone ABA, osmotic substances, antioxidant enzyme activities, and expressive quantity of the Asr1 gene were extended and MDA content dropped significantly (p < 0.05). Moreover, ZD 958 endogenous ABA content, osmotic substances content, antioxidant enzyme activity and Asr1 gene expressive quantity were higher than that of XD 20 (p < 0.05). In conclusion, S-ABA treatment increased the content of endogenous ABA, induced an increase in antioxidant enzyme activity and Asr1 gene expression level, reduced the oxidative damage caused by drought to maize leaves, and improved the adaptability of maize seedlings to withstand drought stress. The promoting effect of S-ABA on the drought-tolerant variety ZD 958 was more obvious (p < 0.05). These results serve as a reference for the use of S-ABA in mitigating drought stress in maize. 相似文献

The morphology of flowering of several cultivars of Dioscorearotundata, Poir is described. Unisexuality and dioecism occurcommonly although complete flowers have been observed occasionally.The pollen grains are sticky and cannot be dispersed by wind;small insects are probably the main agents of pollination. Pollengrain viability appears to be low. Fruit and seed set are alsovery low and usually not more than five viable seeds are producedon one plant. Production of hybrid plants is feasible if filledseeds are selected and germinated following the end of seeddormancy. Epidermal strips and leaf fragments of Commelina and leaf fragmentsof maize were incubated on solutions containing naturally-occurringor synthetic cytokinins and/or ABA. The effects of these treatmentson stomatal behaviour were assessed. Cytokinins alone did notpromote stomatal opening in either species but concentrationsof both zeatin and kinetin from 10 3 to 10 1 molm 3 caused some reversal of ABA-stimulated closure ofmaize stomata. The reversal of the ABA effect increased withincreasing cytokinin concentration. Cytokinins had no effecton ABA-stimulated closure of Commelina stomata. When appliedalone, at high concentration (10 1 mol m 3), to Commelina epidermis or leaf pieces both zeatin and kinetin restrictedstomatal opening.
