Bean arcelin     
Summary SDS-PAGE of seed proteins from the seeds of a nondomesticated bean of Mexican origin (Phaseolus vulgaris L., PI 325690) revealed the presence of a novel 38 kd protein which appeared to be neither an altered phaseolin nor a lectin fraction. The protein was named arcelin, after Arcelia, the town in the state of Guerrero near which PI 325690 had been collected. The pure line, UW 325, was derived by self fertilization of the plant from a single arcelin-containing seed of PI 325690. Despite a low percentage seed phaseolin (14.6%), seed phenotype, seed germination, plant growth, pollen fertility, and percentage seed protein of UW 325 were normal. Analyses of F2 and F3 seeds from a single F1 plant of the cross SanilacXPI 325690-3 revealed that arcelin expression was inherited as a single gene and that presence was dominant to absence of arcelin. The mean percentage phaseolin in the seeds of homozygous dominant Arc/Arc F3 families (14.0%) was significantly lower than that of the homozygous recessive arc/arc seeds (44.7%). The distribution of percentage phaseolin values for seeds within segregating families was bimodal and nonoverlapping. Without exception, seeds containing arcelin (Arc+phenotype) contained a lower percentage phaseolin than seeds lacking arcelin (Arc-phenotype). Although arcelin presence was associated with low percentage phaseolin, the Arc/Arc and Arc/arc genotypes were similar for seed weight and percentage total seed protein.  相似文献

Selection and random genetic drift are the two main forces affecting allele frequencies in common bean breeding programs. Therefore, knowledge on allele frequency changes attributable to these forces is of fundamental importance for breeders. The changes in frequencies of alleles of biochemical markers were examined in F2 to F7 populations derived from crosses between cultivated Mesoamerican and Andean common bean accessions (Phaseolus vulgaris L.). Biochemical markers included the seed proteins phaseolin, lectin and other seed polypeptides, and six isozymes. The Schaffer’s test detected a high significant linear trend of the 63% of the polymorphic loci studied, meaning that directional selection was acting on those loci. Associations between seed size traits, phaseolin seed-storage protein and isozyme markers were detected based on the comparisons of the progeny genotypic means. In the interracial populations the intermediate form PhaH/T, b6, and Rbcs 98 alleles had a positive effect on seed size. In the inter-gene pool populations, a higher transmission of Mesoamerican alleles in all loci was showed, although the Andean alleles PhaT, Skdh 100 , Rbcs 98 , and Diap 100 showed positive effects on seed weight. Our results suggest that phaseolin and other seed proteins markers are linked to loci affecting seed size. These markers have good potential for improving the results of the selection and should be considered as a strategy for germplasm enhancement and to avoid the reduced performance of the inter-gene pool populations.  相似文献

An intron-less phaseolin gene [15] was used to express phaseolin polypeptides in transgenic tobacco plants. The corresponding amounts of phaseolin immunoreactive polypeptides and mRNA were similar to those found in plants transformed with a bean genomic DNA sequence that encodes an identical -phaseolin subunit. These results justified the use of the intron-less gene for engineering of the phaseolin protein by oligonucleotide-directed mutagenesis. Each and both of the two Asn residues that serve as glycan acceptors in wild-type phaseolin were modified to prevent N-linked glycosylation. Wild-type (wti–) and mutant phaseolin glycoforms (dgly 1, dgly 2 and dgly 1,2) were localized to the protein body matrix by immunogold microscopy. Although quantitative slot-blot hybridization analysis showed similar levels of phaseolin mRNA in transgenic seed derived from all constructs, seed from the dgly 1 and dgly 2 mutations contained only 41% and 73% of that expressed from the wild-type control; even less (23%) was present in seed of plants transformed with the phaseolin dgly 1,2 gene. Additionally, the profile of 25–29 kDa processed peptides was different for each of the glycoforms, indicating that processing of the full-length phaseolin polypeptides was modified. Thus, although targeting of phaseolin to the protein body was not eliminated by removal of the glycan side-chains, decreased accumulation and stability of the full-length phaseolin protein in transgenic tobacco seed were evident.Abbreviations bp base pair(s) - DAF days after flowering - GUS -glucuronidase - kb kilobase - kDa kilodalton  相似文献

The value of intra- and interracial populations in common bean (Phaseolus vulgaris L.) needs to be determined in order to create useful genetic variation for maximizing gains from selection, broadening the genetic base of commercial cultivars, and making efficient use of available resources. Five large-seeded parents of race Nueva Granada (N), two small-seeded race Mesoamerica (M), and one medium-seeded race Durango (D) were hybridized to produce one intraracial (N x N) and three interracial (two N x M and one N x D) populations. Seventy-nine F2-derived F6 lines randomly taken from each population along with their parents were evaluated for agronomic traits and markers at Palmira and Popayán, Colombia, in 1990 and 1991. Variation for agronomic traits and for morphological, protein, and isozyme markers was larger in interracial populations than in the intraracial population. Mean seed yield of all lines as well as yield of the highest yielding line from two interracial populations were significantly higher than that of the intraracial population. The highest ( 0.80±0.15) heritability was recorded for 100-seed weight. Values for seed yield varied from 0.19±0.17 to 0.50±0.16. Gains from selection (at 20% selection pressure) for seed yield ranged from 3.9% to 11.4%. Seed yield was positively associated with biomass yield, pods/m2, and days to maturity, but harvest index showed negative correlations with these traits and a positive value with 100-seed weight. Polymorphism was recorded for phaseolin, lectins, protein Group-1 and protein Group-2 fractions, and six isozyme loci. Lines with indeterminate growth habit had significantly (P < 0.01) higher seed yield than lines with determinate growth habit in a Redkloud x MAM 4 population. Also, 23 other associations of markers with agronomic traits other than seed yield were recorded. Of these associations, lines with T phaseolin, the Diap1 2 allele, and lilac flower color tended to possess greater seed weight.  相似文献

 Arcelin seed proteins of common bean (Phaseolus vulgaris L.) confer resistance to bruchid pests, and in vitro results suggested that greater resistance could be achieved by increasing the concentration of arcelin. We created backcross lines having arcelin alleles (SMARC lines), or alleles for the related protein phytohemagglutinin (PHA) (SMPHA lines), and a null allele for phaseolin to determine if seeds lacking phaseolin would contain increased quantities of arcelin or PHA proteins. To test the affects of genetically removing phaseolin, SMARC and SMPHA lines were derived as pairs of phaseolin-containing and phaseolin-null lines. Parental, SMARC, and SMPHA lines were grown in a replicated greenhouse trial and measured for days-to-flower, days-to-maturity, seed weight, and for quantities of phaseolin, arcelin dimer, PHA, and total proteins. There were no differences between pairs of phaseolin and phaseolin-null lines for days-to-flower, seed weight or total protein, and inconsistent differences for days-to-maturity. Arcelin concentrations were significantly increased in two of four pairs of SMARC lines, and PHA concentration was significantly greater in four of five pairs of SMPHA lines. Phaseolin, the major seed storage protein of Phaseolus vulgaris L., is degraded in the cotyledons in the first 7–10 d following seed germination. We assayed cotyledon extracts for protease activity by using [3H]phaseolin as a substrate and then fractionated the digestion mixtures by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in order to identify the cleavage products. The cotyledons of 4-d-old seedlings contain an endopeptidase which cleaves the polypeptides of [3H]phaseolin (apparent molecular weights=51 000, 48 000, 46 000 and 43 000) into three discrete clusters of proteolytic fragments (M rs=27 000, 25 000 and 23 000). Endopeptidase activity is not detected in the cotyledons until the protein content of these organs starts to decline, shortly after the first day of seedling growth. Endopeptidase activity increases to a maximum level in the cotyledons of 5-d-old seedlings and then declines to a minimum value by day 10. The enzyme was purified 335-fold by ammonium-sulfate precipitation, organomercurial-agarose chromatography, gel filtration and ion-exchange chromatography. The endopeptidase constitutes 0.3% of the protein content in the cotyledons of 4-d-old seedlings. It is a cysteine protease with a single polypeptide chain (M r=30 000). Optimum hydrolysis of [3H]phaseolin occurs at pH 5. The enzyme is irreversibly inactivated at pH values above 7 and at temperatures above 45° C. The endopeptidase attacks only a limited number of peptide bonds in [3H]phaseolin, without causing any appreciable change in the native molecular weight of the storage protein. The endopeptidase is also able to hydrolyze the bean-seed lectin, phytohemagglutinin. The gene SFB encodes an F-box protein that has appropriate S-haplotype-specific variation to be the pollen determinant in the S-RNase-based gametophytic self-incompatibility (GSI) reaction in Prunus (Rosaceae). To further characterize Prunus SFB, we cloned and sequenced four additional alleles from sweet cherry (P. avium), SFB 1 , SFB 2 , SFB 4 , and SFB 5 . These four alleles showed haplotype-specific sequence diversity similar to the other nine SFB alleles that have been cloned. In an amino acid alignment of Prunus SFBs, including the four newly cloned alleles, 121 out of the 384 sites were conserved and an additional 65 sites had only conservative replacements. Amino acid identity among the SFBs ranged from 66.0% to 82.5%. Based on normed variability indices (NVI), 34 of the non-conserved sites were considered to be highly variable. Most of the variable sites were located at the C-terminal region. A window-averaged plot of NVI indicated that there were two variable and two hypervariable regions. These variable and hypervariable regions appeared to be hydrophilic or at least not strongly hydrophobic, which suggests that these regions may be exposed on the surface and function in the allele specificity of the GSI reaction. Suppression of phaseolin and lectin accumulation in common bean resulted in higher concentrations of bean seed polypeptides with apparent molecular weights of 54 kDa and from 70 to 84 kDa on SDS-polyacrylamide gel electrophoresis. Polypeptides of 54 and 56 kDa segregated as products of different alleles. Genes for the 54/56 kDa bands and phaseolin were estimated to be 26.2±3.7 map units apart. The 54 kDa band phenotype manifested by SDS-PAGE consisted of from one to three polypeptides of 54 kDa MW on 2D gels, and the 56 kDa phenotype consisted of one polypeptide of 56 kDa plus two minor polypeptides of 54-54.5 kDa molecular weight. The pKI of these polypeptides was approximately 5.25. The methionine content of the 54 kDa polypeptides of the cultivar Great Northern Star was 1.6±0.1 g/100 g protein, which was not statistically different from the value (1.5±0.1%) obtained for phaseolin isolated by the same procedure. F2 seeds deficient for phaseolin and lectin contained as much total N per g as wild-type seeds and were not shrunken, but contained 50% more free amino acids. Studies with the induced lysogens of λS +R+, λS-R+, λS+R- and λS-R- phages have shown that while theS gene product is essential for the action of intracellularR gene product to release the periplasmic alkaline phosphatase in the presence of EDTA, the latter gene product can bring about this effect while acting onEscherichia coli cells from outside, in the absence of functionalS gene product; chloroform, could help the intracellularR gene product in effecting bacterial lysis in the absence ofS gene product. These result support the premise that theS gene product facilitates theR gene product in crossing the cytoplasmic membrane into the periplasmic space such that the latter can act on the peptidoglycan layer of the host cell thus causing both the release of alkaline phosphatase and cell lysis. Summary In a study of partial self-compatibility in Brassica oleracea, flower number, seeded siliqua and seed production were recorded on self and cross-pollinated inflorescences of 32 progenies obtained by inter-crossing and selfing 8 plants homozygous for the incompatibility alleles S2, S5, S15 and S45.Progeny differences for both self-and out-cross seed production could be largely attributed to G.C.A. effects which were essentially uncorrelated. For cross-pollinated inflorescences heterosis was also important. Significant differences were found for selfed seed set and its two components, the proportion of flowers producing seeded siliquae and the numbers of seed per seeded siliqua, between parents with the same S-allele which could not be attributed to S-genotype alone. No evidence of increased self-compatibility in particular S-allele heterozygotes (mutual weakening) could be found.Outcross seed production depended primarily on the numbers of seeds set per seeded siliqua while self seed production was largely determined by the proportion of flowers which produced seeded siliqua. It is suggested that this is a key character for the production of inbred lines with reduced partial self-compatibility.  相似文献

 Arcelin seed proteins of common bean (Phaseolus vulgaris L.) are toxic to one of the most damaging pests of bean seeds, Zabrotes subfasciatus (Boheman), but they appear to have little effect on another important bean pest, Acanthoscelides obtectus (Say), when introduced into standard cultivars by backcrossing. With the goal of increasing arcelin concentration to improve resistance, we modified seed-protein composition by introducing a null allele for the major seed protein, phaseolin, into lines (SMARC1, 2 and 4) or three phytohemagglutinin types (SMPHA lines). These lines were tested for resistance to both insects by measuring percentage insect emergence (%E) and days-to-adult emergence (DAE). For SMARC lines, arcelin type was the most important factor in resistance levels, with SMARC1 lines being most resistant, SMARC2 lines intermediate, and SMARC4 lines the least resistant to both bruchids. Additionally, the absence of phaseolin was a significant factor in the resistance of SMARC lines to A. obtectus. SMARC1 lines without phaseolin had half the percentage insect emergence of lines with phaseolin. Summary The expressed activity in pollen and stigma was determined for both S alleles of sixteen S-alíele heterozygous genotypes and for one of the two S alleles of two additional heterozygotes. Activities were measured using pollen tube penetration and seed set data from reciprocal crosses between each S-allele heterozygote and its two corresponding S-allele homozygotes.In pollen the S-allele activities ranged from zero to 100% inhibition of pollen tube penetration and seed set, and in the stigma they ranged from 8 to 100% inhibition. Of the sixty-eight S-allele activities measured, thirty-three (48%) were 90 to 100% inhibition, nine (13%) were 80 to 89% inhibition and one to five were within each ten-unit range below 80% inhibition.In an S-allele heterozygote, each subset of two S alleles had an activity for each allele in both pollen and stigma which was highly repeatable among duplicate pollinations within and among successive years. Each subset of two S alleles had a specific S-allele interaction in the pollen, and the same or another specific interaction in the stigma. In pairings with six other S alleles, allele S 2 had four calculated levels of activity in pollen that ranged from 88 to 94%, and five levels in the stigmas between 15 and 94%. When paired in a heterozygote, alleles S 3 and S 5 had activities ranging between 42 and 59%, representing mutual weakening of S-allele activity. Also, heterozygote S 15 S 3 had pollen activities, respectively, of 25 and 6%, i.e. mutual weakening in the pollen.These results indicate that in heterozygous combination with a series of other S alleles, each S-allele may have activity in pollen and also in stigma that potentially is between zero and 100% inhibition. They further indicate that the defined sexual-organ X S-allele-interaction Types I, II, III and IV are extremes; all intermediate variations including complete weakening of both alleles are possible. Recessiveness is weakening of the activity of but one of the two S alleles. Summary Allelic complexity is a key feature of self-incompatibility (S) loci in gametophytic plants. We describe in this report the allelic diversity and gene structure of the S locus in Solanum tuberosum revealed by the isolation and characterization of genomic and cDNA clones encoding S-associated major pistil proteins from three alleles (S 1, S r1, S 2). Genomic clones encoding the S1 and S2 proteins provide evidence for a simple gene structure: Two exons are separated by a small intron of 113 (S 1) and 117 by (S 2). Protein sequences deduced from cDNA clones encoding S1 and Sr1 proteins show 95% homology. 15 of the 25 residues that differ between these S 1and S r1alleles are clustered in a short hypervariable protein segment (amino acid positions 44–68), which corresponds in the genomic clones to DNA sequences flanking the single intron. In contrast, these alleles are only 66% homologous to the S 2allele, with the residues that differ between the alleles being scattered throughout the sequence. DNA crosshybridization experiments identify a minimum of three classes of potato S alleles: one class contains the alleles S 1, S r1and S 3, the second class S 2and an allele of the cultivar Roxy, and the third class contains at present only S 4. It is proposed that these classes reflect the origin of the S alleles from a few ancestral S sequence types.  相似文献

In this study of embryo development in Phaseolus vulgaris L., we found that immature embryonic axes placed in culture show a growth lag before germinating. The length of this lag phase varies according to axis age at excision, but is not affected by transfer to fresh medium, alteration of sucrose concentration between 0.5 and 2%, or whether the culture medium is liquid or agar-solidified. The lag phase was shortened by both actinomycin D and cordycepin treatment, and by treatment with 10-5 to 10-6 M benzyladenine. The effect of abscisic acid (ABA) varied with concentration: below a certain level, it had no effect on the lag phase, but above that level it inhibited, germination. This threshold concentration was 10-7 M for 20-d-old axes but increased to 10-5 M by the time the axes were 32 to 34 d old. To determine whether the axes were continuing their embryonic development during the lag phase, we tested them for desiccation-tolerance and for synthesis of phaseolin, a seed storage protein which is specific for embryos of P. vulgaris. The ability to germinate after rapid desiccation was acquired by axes at 26 d past anthesis; when axes younger than this were placed in culture, they developed desiccation-tolerance during the lag phase of growth, indicating that they were continuing embryonic maturation. Phaseolin was present in isolated axes, although at lower levels than in cotyledons. It accumulated during axis development in parallel with total protein, staying at about 1% of total protein content. When isolated immature axes were pulsed with 3H-or 14C-amino acids, they incorporated label into phaseolin, shown by precipitation with anti-phaseolin antibody. Isolated axes from mature seeds, however, did not synthesize detectable amounts of phaseolin. Immature axes cultured in vitro for a period of one to several days continued synthesizing phaseolin until the day prior to visible germination. Treatment of cultured axes with ABA increased the amount of precursor amino acids incorporated into protein, but had a small or no effect on the relative proportion of phaseolin synthesized. We conclude that P. vulgaris axes in culture continue to develop embryonically for a period of time which seems to be under intrinisc control by the axis. This contrasts with precocious germanation, a pattern of embryo behavior seen in many other species. When such embryos are excised from seeds while immature and placed in culture, they switch promptly from embryo development into germination. If ABA or water stress is responsible for preventing precocious germination, it may be that a high level of ABA is maintained or synthesized internally by embryonic axes of Phaseolus, while in other embryos the maternal environment supplies ABA and/or causes water stress.Abbreviations ABA Abscisic acid - BA benzyladenine  相似文献

Cross-compatibility relationships in almond are controlled by a gametophytically expressed incompatibility system partly mediated by stylar RNases, of which 29 have been reported. To resolve possible synonyms and to provide data for phylogenetic analysis, 21 almond S-RNase alleles were cloned and sequenced from SP (signal peptide region) or C1 (first conserved region) to C5, except for the S 29 allele, which could be cloned only from SP to C1. Nineteen sequences (S 4 , S 6 , S 11 S 22 , S 25 S 29 ) were potentially new whereas S 10 and S 24 had previously been published but with different labels. The sequences for S 16 and S 17 were identical to that for S 1 , published previously; likewise, S 15 was identical to S 5 . In addition, S 4 and S 20 were identical, as were S 13 and S 19 . A revised version of the standard table of almond incompatibility genotypes is presented. Several alleles had AT or GA tandem repeats in their introns. Sequences of the 23 distinct newly cloned or already published alleles were aligned. Sliding windows analysis of Ka/Ks identified regions where positive selection may operate; in contrast to the Maloideae, most of the region from the beginning of C3 to the beginning of RC4 appeared not to be under positive selection. Phylogenetic analysis indicated four pairs of alleles had ‘bootstrap’ support > 80%: S 5 /S 10 , S 4 /S 8, S 11 /S 24 , and S 3 /S 6 . Various motifs up to 19 residues long occurred in at least two alleles, and their distributions were consistent with intragenic recombination, as were separate phylogenetic analyses of the 5′ and 3′ sections. Using immunochemical methods the authors investigated the evolutionary taxonomic distribution of the reserve seed protein “phaseolin” in cultivars ofPhaseolus vulgaris, in a series of species ofPhaseolus, and in representatives of some additional genera ofViciaceae. “Phaseolin” is typical of the seed ofPhaseolus vulgaris L.: it was detected in all 658 investigated cultivars — and also in species related toPhaseolus vulgaris L.(Ph. vulgaris L. ssp.aborigineus Burk.,Ph. polyanthus Green,Ph. dumosus Macf.,Ph. coccineus L., and in an undescribed species from the group ofPh. vulgaris L. -Ph. coccineus L.). A protein immunochemically somewhat similar to ?phaseolin“ occurs inPh. acufifolius A. Gray. In all other taxa“phaseolin” is absent.  相似文献

Self-incompatibility in the Solanaceae is mediated by S-RNase alleles expressed in the style, which confer specificity for pollen recognition. Nicotiana alata has been successfully used as an experimental model to elucidate cellular and molecular aspects of S-RNase-based self-incompatibility in Solanaceae. However, S-RNase alleles of this species have not been surveyed from natural populations and consequently the S-haplotype diversity is poorly known. Here the molecular and functional characterization of seven S-RNase candidate sequences, identified from a natural population of N. alata, are reported. Six of these candidates, S 5 , S 27 , S 70 , S 75 , S 107 , and S 210 , showed plant-specific amplification in the natural population and style-specific expression, which increased gradually during bud maturation, consistent with the reported S-RNase expression. In contrast, the S 63 ribonuclease was present in all plants examined and was ubiquitously expressed in different organs and bud developmental stages. Genetic segregation analysis demonstrated that S 27 , S 70 , S 75 , S 107 , and S 210 alleles were fully functional novel S-RNases, while S 5 and S 63 resulted to be non-S-RNases, although with a clearly distinct pattern of expression. These results reveal the importance of performing functional analysis in studies of S-RNase allelic diversity. Comparative phylogenetic analysis of six species of Solanaceae showed that N. alata S-RNases were included in eight transgeneric S-lineages. Phylogenetic pattern obtained from the inclusion of the novel S-RNase alleles confirms that N. alata represents a broad sample of the allelic variation at the S-locus of the Solanaceae.  相似文献

氧化铈纳米颗粒(CeO2NPS),因具有较强的自由基清除能力和抗氧化酶特性,已被证明可提高植物的耐盐性,但其对辣椒种子引发作用和机制尚不明确。为揭示CeO2NPS种子引发处理辣椒对盐胁迫下的萌发及幼苗生长的影响,以辣椒品种(Capsicum annuum)茂蔬360为试验材料,设置了7个CeO2NPS浓度(0、0.05、0.1、0.2、0.3、0.4、0.5 mmol·L-1),以未引发处理组为对照,研究不同浓度CeO2NPS种子引发处理后对盐胁迫下辣椒种子萌发、幼苗生物量和生理生化指标的影响。结果表明:(1)0.5 mmol·L-1 CeO2NPS种子引发处理后的种子,其可溶性蛋白质、脯氨酸含量和过氧化氢酶(CAT)活性、抗坏血酸(AsA)含量和AsA/DHA比值显著提高,超氧阴离子(O2-)含量显著降低; 盐胁迫下,该处理种子的发芽率、发芽势、发芽指数、活力指数最大。(2)0.4 mmol·L-1 CeO2NPS种子引发处理的幼苗在盐胁迫下的鲜重、干重和根长最大,幼苗的可溶性蛋白质、AsA含量和AsA/DHA比值均显著提高。综上认为,CeO2NPS引发处理不仅可通过降低种子水势、促进贮藏物质代谢和提高抗氧化能力提高种子在盐胁迫下的发芽率,还可在苗期通过增强蛋白合成和抗坏血酸-谷胱甘肽循环(AsA-GSH)促进盐胁迫下幼苗的生长。  相似文献

Developing cotyledons of Phaseolus vulgaris L. were labeled for 30 min with [3H] amino acids, homogenized, and the proteins fractionated on sodium dodecylsulfate (SDS) polyacrylamide gels. Fluorographs of these gels showed that the polypeptides of phaseolin, the major reserve protein of P. vulgaris, were synthesized as precursors which could be distinguished from the polypeptides of mature phaseolin by their slightly lower mobility. When extracts of cotyledons labeled for 45 min with [3H] amino acids were fractionated on isopynic sucrose gradients, radioactive phaseolin banded at the same density (1.14 g cm-3) as the endoplasmic reticulum (ER)-marker enzyme NADH-cytochrome c reductase. Fractionation in the presence of 3 mM MgCl2 indicated that the newly-synthesized phaseolin was associated with the rough ER. Pulse-chase experiments showed that phaseolin was transiently associated with the ER, and later accumulated in the protein bodies. Treatment of isolated ER with proteinase K showed that phaseolin polypeptides were degraded only if Triton X-100 was present, indicating that phaseolin was membrane-protected, probably enclosed within the vesicles. ER-associated phaseolin associated to an 18S form at pH 4.5 in the presence of 0.3 M NaCl and 100 mM sodium acetate. The polypeptides of ER-associated phaseolin had a slightly lower mobility on SDS-gels than polypeptides of protein body phaseolin. ER-associated phaseolin had a carbohydrate content of 6.8%, while protein body-derived phaseolin had a carbohydrate content of 6.2%. When cotyledons were labeled simultaneously with [14C] amino acids and [3H] glucosamine or with [14C] amino acids and [3H] mannose, the [3H]/[14C] ratio of ER-derived phaseolin was similar to that of protein body derived phaseolin, indicating that the faster mobility on SDS-gels was not due to the detachment of carbohydrate. Experiments in which the carbohydrate side chains were removed with endoglycosidase H, and the resulting polypeptides subjected to electrophoresis in SDS-gels showed that the differential mobility of the glycopolypeptides of phaseolin resided in their polypeptide chains.  相似文献
