Environmental prenatal stress (EPS) has effects on fetuses that are long-lasting, altering their hormone levels, brain morphology and behavior when they reach maturity. In previous research, we demonstrated that EPS affects the expression of induced maternal behavior (MB), the neuroendocrine system, and morphology of the sexually dimorphic accessory olfactory bulb (AOB) involved in reproductive behavior patterns. The bed nucleus of the accessory olfactory tract (BAOT) is another vomeronasal (VN) structure that plays an inhibitory role in rats in the expression of induced maternal behavior in female and male virgins. In the present study, we have ascertained whether the behavioral, neuroendocrine, and neuromorphological alterations of the AOB found after EPS also appear in the BAOT. After applying EPS to pregnant rats during the late gestational period, in their female offspring at maturity we tested induced maternal behavior, BAOT morphology and plasma levels of testosterone (T), estradiol (E2), progesterone (P), adrenocorticotropic hormone (ACTH) and corticosterone (Cpd B). EPS: a) affected the induction of MB, showed a male-like pattern of care for pups, b) elevated plasma levels of Cpd B and reduced E2 in comparison with the controls, and c) significantly increased the number of BAOT neurons compared to the control females and comparable to the control male group. These findings provide further evidence that stress applied to pregnant rats produces long-lasting behavioral, endocrine and neuroanatomical alterations in the female offspring that are evident when they become mature.  相似文献

In mice, tactile stimulation of the nipples appears to be critical for the onset of postpartum maternal aggression. Surgical removal of the nipples (thelectomy) blocks aggression if performed prior to parturition. In rats, indirect evidence suggests a similar role for nipple stimulation in maternal aggression. Two experiments were undertaken to determine whether thelectomy prior to mating reduces pregnancy-induced and/or postpartum aggression in this species. In the first, thelectomized and sham-thelectomized females were subjected to home cage tests (pups, if any, present) with unfamiliar male intruders on Gestation Days 18 and 21 and Lactation Days 3 and 5. Additional groups of thelectomized females were tested one time only on either Lactation Day 5 or 12. Thelectomized and control females were equally aggressive; postpartum, nearly all females in both groups attacked. Experiment 2 used females that were hysterectomized-ovariectomized (HO) on Gestation Day 16. Such females are not aggressive prior to initiating maternal behavior, but become highly aggressive (over 80% attacking) after commencing maternal care. Females again were thelectomized or sham-thelectomized prior to mating. On Day 16 HO was performed, and 48 hr later continuous exposure to pups was begun. After the females had displayed maternal behavior for 1.5-2 days, intruder tests were conducted. All females attacked at least once, with no differences between treatment groups. Thus thelectomy does not reduce maternal aggression in the rat. This finding, however, does not preclude a role for tactile ventral stimulation in mediating maternal aggression.  相似文献

The present study was designed to investigate the role of the medial preoptic area (MPOA) and the bed nucleus of the stria terminalis (BNST) in the onset and maintenance of maternal behavior in sheep. In a first experiment, the MPOA or BNST were transiently inactivated during the first 2 h postpartum in primiparous ewes with the use of the anaesthetic lidocaine. MPOA inactivation greatly impaired the display of maternal behavior whereas inactivation of BNST or of adjacent sites (septum or diagonal band of Broca) or infusion of cerebrospinal fluid did not. In a separation/reunion lamb test (S/R) performed at 2 h postpartum, ewes with MPOA inactivation exhibited little reaction after separation of their lambs and did not show any motivation to reunite with them. Ewes with BNST inactivation showed intermediate performances in the S/R test. Moreover, in control ewes that were maternal for the first 2 h postpartum, MPOA or BNST inactivation performed in the following 12 h induced deficits in the S/R test, indicating that the MPOA and to a lesser extent BNST are also involved in the maintenance of maternal behavior. A second experiment showed that, in multiparous ewes, MPOA inactivation at parturition induced less deficit in the display of maternal behavior and in the S/R test than in primiparous mothers. These findings indicate that the MPOA and, to some extent, the BNST are functionally involved in the initiation and in the maintenance of maternal behavior in sheep, but this involvement is influenced by maternal experience.  相似文献

The present study investigated hormonal mediation of maternal behavior and accumbal dopamine (DA) responses to pup-stimuli, as measured in microdialysis samples collected from the nucleus accumbens shell of female rats in non-homecage environment. In Experiment 1, samples were collected before and after continuous homecage pup experience from either intact postpartum or cycling females. In Experiment 2, samples were collected before and after responding maternally in homecage from ovariectomized females given either parturient-like hormone or sham treatments. After baseline sample collection in the dialysis chamber, pup and food stimuli were individually presented to females. Upon sampling completion, all animals were placed back into their homecage with donor pups for several days, and then the sample collection procedure was repeated. Prior to stimulus presentation, postpartum and hormone-treated females had decreased basal DA release compared to their controls. In response to pup stimuli, only postpartum and hormone-treated females had increased DA release compared to basal release (both sampling days). In response to food stimuli, all females had increased DA responses from basal; although there were group differences on the initial day of sampling. Findings suggest that hormones associated with inducing maternal behavior in the postpartum rat play a significant role in modifying accumbal dopaminergic responses on first exposure to pup stimuli in the rat. However, the postpartum experience provides further modifications to this brain region to promote DA responses to pup stimuli.  相似文献

An association between pregnancy levels of estrogen and progesterone and maternal behavior has been demonstrated in several taxonomic orders of nonprimate and primate mammals, but has not so far been investigated in the gorilla. In this study we investigated whether prepartum titers of urinary estrone conjugates (E1C) or pregnanediol-3alpha-glucuronide (PdG) were related to postpartum maternal behavior in eight multiparous Western lowland gorilla females (Gorilla gorilla gorilla) housed in four zoological gardens. Urine samples were collected from each study animal for 14 days prepartum and 14 days postpartum, and measures of maternal responsiveness were scored during the first 15 days postpartum. Urine samples were assayed with radioimmunoassay for E1C and PdG. Results for the peripartum profiles of urinary E1C, as well as postpartum profiles of PdG, agree with previous findings for the gorilla, while results for late-pregnancy profiles of urinary PdG were inconclusive in confirming a prepartum increase or decrease. Neither prepartum levels of E1C or PdG, nor the E1C/PdG ratio were found to be related to measures of postpartum maternal behavior. This lack of association between late-pregnancy E1C titers per se and postpartum maternal behavior is contrary to findings in nonprimate and other primate species.  相似文献

The role of prolactin in maintaining the maternal behavior of the rat was investigated. Ergocornine, a drug that interferes with the release of prolactin from the anterior pituitary, was injected daily into postpartum females for a period of 21 days. Those receiving ergocornine did not differ significantly from control females on any measure of maternal behavior, the control females having received either ergocornine plus prolactin or simply the vehicle. Since both lactation and the decidual cell response to uterine traumatization were inhibited after ergocornine, and since each were at least partially reversed by exogenous prolactin, it was evident that ergocornine interfered successfully with the postpartum release of prolactin. That at the same time maternal behavior was unaffected, supports the conclusion that prolactin is not essential in maintaining the maternal responsivity of the postpartum female rat.  相似文献

Plants accumulate high levels of Gamma amino butyric acid (GABA) in response to different environmental stresses and GABA metabolism has different functions such as osmotic and pH regulation, bypass of tricarboxylic acid cycle, and C:N balance. The cytoplasmic male sterile (CMS) II mutant of Nicotiana sylvestris has a deletion in the mitochondrial gene nad7 which encodes the NAD7 subunit of complex I which causes increased leaf respiration, impaired photosynthesis, slower growth and increased amino acid levels. In this study we aimed to elucidate the role of GABA and GABA metabolism in different genotypes of the same plant system under salt stress (100mM NaCl) in short (24h) and long (7, 14 and 21 days) terms. We have investigated the differences in leaf fresh and dry weights, relative water content, photosynthetic efficiency (F(v)/F(m)), glutamate dehydrogenase (GDH, EC 1.4.1.4) and glutamate decarboxylase (GAD, EC 4.1.1.15) enzyme activities, GABA content and GAD gene expression profiles. GDH activity showed variations in CMSII and wild type (WT) plants in the first 24h. GAD gene expression profiles were in good agreement with the GAD enzyme activity levels in CMSII and WT plants after 24h. In long-term salinity, GAD activities increased in WT but, decreased in CMSII. GABA accumulation in WT and CMSII plants in short and long term was induced by salt stress. Variations in GDH and GAD activities in relation to GABA levels were discussed and GABA metabolism has been proposed to be involved in better performance of CMSII plants under long term salinity.  相似文献

Numerous studies have demonstrated that Sprague-Dawley rats exhibit a wide range of locomotor reactivity when placed in a novel environment. High Responder (HR) rats show exaggerated locomotor response to novelty, enhanced neuroendocrine stress reactivity, decreased anxiety-like behavior, and propensity to self-administer psychostimulants, compared to the less active Low Responder (LR) animals. Few studies have explored the early environmental factors which may underlie the HR-LR differences in emotional reactivity. Considering the enormous impact of maternal care on rodent neurodevelopment, we sought to examine maternal behavior in HR-LR dams to determine whether they exhibit differences which could contribute to their offspring's differential temperaments. Females, like males, can be classified as HR versus LR, showing marked differences in novelty-induced locomotor activity and anxiety-like behavior. HR-LR mothers behaved differently with their pups during the first two postpartum weeks. LR dams spent greater time licking and nursing their pups compared to HR dams, with the most prominent differences occurring during the second postpartum week. By contrast, when non-lactating HR-LR females were presented with orphaned pups, the pattern of maternal response was reversed. HR females were more responsive and showed greater maternal care of the novel pups compared to LR females, which were probably inhibited due to fear of the unfamiliar pups. This underscores the critical interplay between the female's emotional phenotype, her hormonal status and her familiarity with the pup as key factors in determining maternal behavior. Future work should explore neural and hormonal mechanisms which drive these HR-LR differences in maternal behavior and their impact on the development of the offspring.  相似文献

GABA is the principal neurotransmitter of the mammalian circadian system, and its activity is subject to diurnal and circadian variations, with maximal values in hypothalamic turnover, content and binding during the night. In this study we have examined rhythms in the proconvulsant effect of inhibition of glutamate decarboxylase (GAD) in hamsters (Mesocricetus auratus) as well as the anticonvulsant effect of androsterone, a neurosteroid that positively modulates the GABA(A) receptor. Administration of 10-60 mg/Kg of 3-mercaptopropionic acid (3-MPA, a GAD inhibitor) induced convulsions that were analyzed by an ad-hoc severity scale, with a lower sensitivity threshold at 24:00 h. Moreover, the latency for first and maximal convulsive response times was significantly lower at night. A similar temporal profile (maximal effect at midnight) was found for picrotoxin-induced seizures. Androsterone (40 mg/Kg) completely inhibited 3-MPA-induced tonic/clonic seizures at 12:00 h, while it had a partial inhibitory effect at 24:00 h. These results support the importance of temporal regulation of GABAergic modulation in the central nervous system.  相似文献

Female rats with maternal experience display a shorter onset of maternal responsiveness compared to those with no prior experience. This phenomenon called ‘maternal memory’ is critically dependent on the nucleus accumbens (NA) shell. We hypothesized that activation of OT receptors in the NA shell facilitates maternal memory. In Experiment 1, postpartum female rats given 1 hour of maternal experience were infused following the experience with either a high or low dose of an OT antagonist into the NA shell and tested for maternal behavior after a 10-day pup isolation period. Females receiving a high dose of the antagonist showed a significantly longer latency to exhibit full maternal behavior after the pup isolation period compared to females that received vehicle or a high dose of antagonist in a control region. In Experiment 2, postpartum female rats were infused with either a high or low dose of OT into the NA shell after a 15-minute maternal experience and tested for maternal behavior after a 10-day pup isolation period. There were no significant differences between the females infused with OT and females treated with a vehicle infused into the NA shell or with OT infused into the control region. One possible reason for a lack of facilitation is a floor effect, since females in the control groups displayed a rapid maternal response after the pup isolation period. These findings suggest that OT receptors, likely in combination with other neurotransmitters, in the NA shell play a role in the consolidation of maternal memory.  相似文献

To better understand the roles that hormones and experience play in infant survival and maternal behavior in a biparental primate species, we analyzed urinary estrone (E(1)C) and pregnanediol glucuronide (PdG) from 24 socially housed titi monkey (Callicebus cupreus) females over 54 pregnancies (N = 1,430 samples). Pregnancies were categorized according to whether the infant survived (N = 35) or not (N = 19), and by maternal parity (primiparous: N = 9; multiparous: N = 45). Mothers of infants that survived had a significantly greater drop in PdG from the third trimester to the first week postpartum than mothers of infants that did not survive. Multiparous mothers had a greater increase in PdG from the first to the third trimester as well as greater increases in the E(1)C:PdG ratio from the first to the third trimester and from the third trimester to the first week postpartum. There were positive relationships between third trimester PdG and maternal carrying and nursing during the first week postpartum, and between maternal age and carrying during the infant's first month of life. There was a negative correlation between maternal age and PdG during the third trimester. These results suggest that elevated progesterone during late pregnancy followed by progesterone withdrawal immediately following parturition is associated with greater probability of infant survivorship and maternal behavior in this species, and that older females engage in more postpartum maternal care.  相似文献

We previously reported that bilobalide, a constituent of Ginkgo biloba L. leaves, protected mice against convulsions induced by 4-O-methylpyridoxine (MPN). To elucidate the mechanism of the anticonvulsant activity of bilobalide, this study examined the effect of bilobalide on MPN-induced changes in the levels of gamma-aminobutyric acid (GABA) and glutamate, and in the activity of glutamic acid decarboxylase (GAD) in the hippocampus, cerebral cortex and striatum of the mouse. GABA levels and GAD activity in the hippocampus and cerebral cortex were significantly enhanced by bilobalide treatment (30 mg/kg, p.o., for 4 days) alone. MPN significantly decreased GABA levels and GAD activity in the three brain regions tested compared with those in the control. Pretreatment with bilobalide effectively suppressed the MPN-induced reduction in GABA levels and GAD activity in the hippocampus and cerebral cortex. On the other hand, there were no significant differences in the glutamate levels in the three regions despite various treatments. These results suggested that bilobalide prevents MPN-induced reduction in GABA levels through potentiation by bilobalide of GAD activity, and this effect of bilobalide contributes to its anticonvulsant effect against MPN-induced convulsions.  相似文献

A study was conducted in subtropical northern Mexico (26 degrees N) to determine whether the presence of estrous females can improve the response of seasonally anovulatory goats to the introduction of bucks in the group. The induction of estrous activity was studied in three groups of anovulatory lactating goats during seasonal anestrus. These females were of the Mexican Creole breed. In the control group (sexually inactive (SI), n = 20), two control (SI) bucks exposed to normal seasonal daylength variations were used. In the second group (SI + E, n = 20 + 3), two control males were also used, but in addition, three females of the group were in estrus at the time of male introduction. In the third group (sexually active, SA + E, n = 19 + 4), anovulatory females were exposed to two bucks made sexually active by exposure to 2.5 months of long days (16L:8D) followed by two subcutaneous 18 mg melatonin implants, and four estrous females were also present when introducing the bucks. In all groups, males were introduced on 15 March and estrous detection was conducted twice daily for 15 days. The sexual activity of the bucks was observed from 08:00 to 10:00 h during the first five days of exposure to females. More females displayed estrous behavior in the first 15 days following the introduction of the males in the SA + E group (18/19) as compared with the SI or SI + E groups (2/20 and 0/20, respectively; P < 0.001). No difference was observed between the two latter groups. Thirteen females of SA + E group showed a second estrus between days 6 and 11 (short estrous cycle duration: 5.4 +/- 0.4 days). By contrast, in the SI group none showed a second estrus. The sexual behavior of the males in the SA + E group was greater as compared with that of the males in SI and SI + E groups (over 80% of the total sexual activity recorded in the three groups; P < 0.001). By contrast, no differences were found between SI and SI + E males. These results indicate that the presence of estrous females alone at the time of buck introduction is not sufficient to induce an adequate stimulation of seasonally inactive males. The use of sexually active bucks is necessary to induce reproductive activity in anovulatory females, whereas preparation of the bucks with long days followed by melatonin implants allows them to gain such a capacity.  相似文献

为评价多氯联苯对人体发育的影响,本研究以大鼠为模型,研究多氯联苯(PCB)对母性行为的影响。将50只同期怀孕的Long-Evans大鼠随机分为5组,在怀孕第7-18天,每天给大鼠肌注四氯联苯或芝麻油。1组、2组,按每kg体重分别注射1mg、20mg2,2′,4,4′-四氯联苯(PCB47);3组、4组,按每kg体重分别注射0.25mg、1mg3,3′,4,4′-四氯联苯(PCB77);5组(对照组)注射芝麻油。产仔后的第0、3、6、12、15天分别观察母鼠的筑窝、舔仔、自发叼仔和诱发叼仔、蹲伏和在窝内的时间。结果表明:对照组母鼠在30min的观察中,其窝内时间与幼仔出生后天娄和负相关,而接触PCB的母鼠的窝内时间与幼仔出生天数无相关关系。接触PCB47的母鼠的筑巢、蹲伏、舔仔、自发叼仔行为在幼仔出生后的第0天和6天与对照组有显著差异,而接触PCB77的母鼠仅在第15天与对照组大鼠有区别。在诱导叼仔的实验中,接触PCB77的母鼠的叼仔时间在幼仔出生后的第12天增加。  相似文献

Ethanolamine O-sulphate (400 mg/kg, i.p.) was administered to rat pups at 9 days of age and on alternate days up to 17 days of age. At 18 days of age, gamma-aminobutyric acid (GABA) concentration was increased (three- to fourfold), glutamic acid decarboxylase (GAD) activity reduced to 55% of control, and the number of GABAA and GABAB binding sites increased in the cerebral cortex. This is the same pattern of change as seen previously with oral administration of ethanolamine O-sulphate to the adult rat but the changes occur more rapidly in the developing rat. A lower dose of ethanolamine O-sulphate (100 mg/kg, i.p.), administered according to the same schedule, caused a twofold increase in cortical GABA at 18 days of age whereas GAD activity and GABAA binding were not significantly altered.  相似文献

Testosterone and oestradiol can modulate GABA synthesis in sexually regressed goldfish. Here we investigated their effects on the mRNA expression of two isoforms of the GABA synthesizing enzyme glutamate decarboxylase (GAD(65) and GAD(67), EC 4.1.1.15). Full-length GAD clones were isolated from a goldfish cDNA library and sequenced. Goldfish GAD(65) encodes a polypeptide of 583 amino acid residues, which is 77% identical to human GAD(65). Goldfish GAD(67) encodes a polypeptide of 587 amino acid residues and is 82% identical to human GAD(67). Goldfish GAD(65) and GAD(67) are 63% identical. Sexually regressed male and female goldfish were implanted with solid silastic pellets containing testosterone, oestradiol or no steroid. Semiquantitative PCR analysis showed that oestradiol significantly increased GAD(65) mRNA expression in female hypothalamus and telencephalon, while testosterone resulted in a significant increase only in telencephalon. GAD(67) mRNA levels were not affected by steroids in females. In contrast, both steroids induced significant decreases of GAD(65) and GAD(67) mRNA levels in male hypothalamus, but had no effect on GAD mRNA expression in male telencephalon. Our results indicate that modulation of GAD mRNA expression is a possible mechanism for steroid action on GABA synthesis, which may have opposite effects in males and females.  相似文献

As evidence accumulates regarding the influence of hormones and stress-related conditions on maternal behavior, it becomes critical to better understand the relationship between physiological stress and the ability to cope with infants. Eight Japanese macaque females were observed 3 hr per week during the first 12 weeks after parturition; fecal samples were collected twice a week from each mother, starting 4 weeks before parturition and ending 4 weeks after parturition. Time spent in contact, maternal responsiveness, latency of response, and maternal rejection were measured and correlated with peripartum excreted cortisol and estradiol metabolite levels. Two indices of peripartum hormonal status were also tested against behavior: the postpartum stress index, and the postpartum cortisol/prepartum estradiol ratio (F/E). Postpartum cortisol levels showed a positive correlation with maternal rejection. The cortisol/estradiol ratio was positively correlated with rejection and latency of response, and negatively correlated with maternal responsiveness. Prepartum cortisol levels and the postpartum stress index did not correlate with any aspect of maternal behavior. Our findings suggest that hypothalamic-pituitary-adrenal (HPA) axis activity per se is not enough to predict the quality of interaction between mother and infant. Only when cortisol is high relative to estradiol could it be symptomatic of a possible negative feedback response involving stress, adrenal activity, and the ability of mothers to cope with the additional problems imposed by newborns.  相似文献

Recent studies in primates have suggested that pre- and peripartum sex steroid hormones may be important determinants of maternal behavior and motivation, since higher levels of prepartum estrogen are associated with maternal competency and infant survivorship. To test the cross-species generality of this finding, we monitored excreted profiles of estradiol (E(2)), progesterone metabolites (pregnanediol glucuronide; PdG), and E(2):PdG ratios throughout pregnancy in black tufted-ear marmosets (Callithrix kuhlii). Additionally, we wanted to determine the extent to which variability in maternal carrying effort was related to hormonal factors and relative levels of maternal experience. For six females, hormonal profiles were determined by enzyme immunoassay for two pregnancies, one in which infants survived at least 2 weeks postpartum and one in which infants did not survive. Our within-subjects analyses revealed significant differences in mean prepartum E(2) levels for females in the different infant survival conditions. In contrast to previous findings, however, females exhibited significantly higher prepartum E(2) levels when their infants did not survive a minimum of 2 weeks postpartum, relative to when their infants did survive. Maternal carrying effort was also negatively and significantly correlated with prepartum E(2) levels. Unlike previous reports in callitrichid primates, then, our data suggest that high concentrations of prepartum E(2) in callitrichid primates are not necessarily associated with competent maternal behavior and may instead be associated with poor infant survivorship and inadequate maternal care. Further, our results appear to be convergent with research focusing on human mothers and may represent a common underlying mechanism linking prepartum estrogen and postpartum affect and behavior in some primates.  相似文献

Studies on the maternal transfer of photoperiodic information in mammals indicate that the daily photoperiod perceived by the mother during the gestation-lactation period is communicated to the fetus either through the placenta or via the milk. However, the impact of photoperiodic exposures during gestation and lactation on the maternal pineal and reproductive physiology has not been reported for any tropical rodent. The exposure of pregnant female Indian palm squirrels (Funambulus pennanti) to constant light (24 h light:0 h dark), constant dark (0 h light:24 h dark), long daylength (14 h light:10 h dark) or short daylength (10 h light:14 h dark) during early gestation (< 30 days) resulted in the resorption of pregnancy, while during late gestation (> 30 days), it did not interfere with the maintenance of pregnancy. Alterations in photoperiodic condition during late gestation and lactation altered the postpartum recovery process. Pineal gland activity, as assessed by pineal mass, protein content and plasma melatonin, was lowest during the breeding phase, but increased gradually after parturition until the next breeding phase. During gestation and lactation, constant light, long daylength and short daylength conditions were less effective, while constant dark condition had a profound effect in depressing pineal gland activity, which subsequently advanced postpartum recovery. Hence, lactating females under constant darkness prepare themselves for next mating much earlier than females under natural daylength (12 h light:12 h dark) conditions. Therefore, photoperiodic information, mediated via the pineal gland, may be important for maintaining gestation physiology as well as postpartum recovery in female rodents.  相似文献

Early influences such as maternal stress affect the developmental outcome of the offspring. We created an animal model of postpartum depression/stress based on giving high levels of corticosterone (CORT) to the rat dam, which resulted in behavioral and neural changes in the offspring. This study investigated whether highly elevated levels of maternal CORT during pregnancy or the postpartum result in higher levels of CORT in the stomach milk, serum, and brain of offspring. Dams received daily injections of CORT (40 mg/kg) or oil (control) either during pregnancy (gestational days 10–20) or the postpartum (Days 2–21). Pups that were exposed to high gestational maternal CORT had higher CORT levels in serum, but not in stomach milk or brain, on postnatal day (PND) 1. However, on PND7, pups that were exposed to high postpartum maternal CORT had higher CORT levels in stomach milk and brain, but not in serum. Conversely on PND18, pups that were exposed to high postpartum maternal CORT had higher CORT levels in serum, but not in brain (prefrontal cortex, hypothalamus, or hippocampus). Moreover, 24 h after weaning, there were no significant differences in serum CORT levels between the groups. Thus, CORT given to the dam during pregnancy or the postpartum results in elevated levels of CORT in the offspring, but in an age‐ and tissue‐dependent manner.
