In order to investigate the aerobic granules cultured under alternating aerobic and anoxic conditions, a sequencing batch reactor (SBR) was operated without the presence of a carrier material. Nitrification and denitrification occurred alternately in the SBR operation, with an increased nitrification efficiency of up to 97% and a high chemical oxygen demand (COD) removal efficiency of up to 95%. It was observed that physical characteristics of granule play an important role in the performance of the SBR process. Light microscopy was used to observe the time dependent development of the granules in the SBR. Based on the microscopic observations, some floc-like sludges remained in the form of a mixture with granules for 30 days of operation. Even though various granule sizes had been formed in the reactor after 50 days, the granule sizes were primarily from 1 +/- 0.35 to 1.3 +/- 0.45 mm, rarely exceeding 2 mm. The granules were analyzed by a combination of microelectrodes and fluorescent in situ hybridization (FISH), which provides more detailed information on what happens inside the granules. Based on their results, ammonia oxidizing bacteria (AOB) existed primarily in the upper and middle layers of the granule. Assuming a first-order reaction for nitrification, most of the nitrification is likely to occur from the surface to 300 microm into the granular thickness.  相似文献

Comparison of the characteristics of anammox granules of different sizes   总被引:1,自引:0,他引:1  
The physical properties and performance of anaerobic ammonium oxidation (anammox) granules of different sizes (0.5 ~ 1.0 mm, 1.0 ~ 1.5 mm, and above 1.5 mm) have been investigated. The values of the settling velocity increased with increasing size of the granules. There was no significant difference in metal contents among granules of different sizes, in which calcium, magnesium and iron were the predominant ions. Different start-up periods showed that larger granules were better able to resist adverse impacts and their activities could quickly be recovered. The specific activities of granules of different sizes were 0.55, 0.62, and 0.52 g N/ (g/VSS/day), respectively, which implied the activity of 1.0 ~ 1.5 mm granules was the highest. Larger anammox granules were better able to resist temperature shock and nitrogenous shock loading. However, larger granules were also shown to contain bigger gas tunnels and interior hollows, which decrease the stability of anammox granules. With the comprehensive consideration of bacteria activity, granule stability and shock resistance capacity, the properties of granules within the size range of 1.0 ~ 1.5 mm were found superior to others in this study.  相似文献

AIMS: To obtain biomass and porosity profiles for aerobically grown granules of different diameters and to determine a suitable range of granule diameters for application in wastewater treatment. METHODS AND RESULTS: Microbial granules were cultivated in an aerobic granulated sludge reactor with model wastewaters containing acetate, or ethanol plus acetate, or glucose as the main carbon source. Granules were formed by retaining microbial aggregates using a settling time of 2 min. Sampled granules had diameters ranging from 0.45 to 3 mm. Microbial biomass in the granules was detected with the nucleic acid stain SYTO 9 and confocal laser scanning microscopy. The thickness of the microbial biomass layer was proportional to the granule diameter, and had a maximum value of 0.8 mm. The thickness of the microbial biomass layer correlated with the penetration depth of 0.1 microm fluorescent beads into the granule. CONCLUSIONS: The microbial biomass and porosity studies suggest that aerobically grown microbial granules should have diameters less than a critical diameter of 0.5 mm, if deployed for wastewater treatment applications. This critical diameter is based on the assumption that whole granules should have a porous biomass-filled matrix. SIGNIFICANCE AND IMPACT OF THE STUDY: This work could contribute to the development of aerobic granulation technology for effective biological wastewater treatment.  相似文献

Presence of anaerobic bacteroides in aerobically grown microbial granules   总被引:6,自引:0,他引:6  
Microbial granules were grown in a column-type sequential aerobic sludge blanket reactor inoculated with activated sludge flocs taken from a wastewater treatment plant and containing a medium with glucose as the main carbon source. The reactor selected for granules that could settle rapidly by employing a short settling time of 2 min. Matured granules with diameters between 2 and 3 mm were examined for anaerobic bacteria as their presence can signal the onset of diffusion limitation problems that can potentially diminish granule stability due to the bacterial production of fermentation gases and organic acids under anaerobic conditions. To detect the anaerobes in the granules, clones were constructed from 16S rRNA PCR amplicons. Two sequence types associated with a strict anaerobe Bacteroides spp. were identified from these clones. Fluorescence in situ hybridization (FISH) followed by confocal laser scanning microscopy (CLSM) demonstrated that cells of Bacteroides spp. were concentrated at a depth of approximately 800 mm below the surface of the granule. Cell enumeration using flow cytometry showed that the percentage of labeled cells of Bacteroides spp. compared to total bacterial cells in the granules was 0.56%. This is the first study to use a suite of culture-independent techniques to report the presence of a defined species of anaerobic bacteria in aerobically grown microbial granules.  相似文献

Size-effect on the physical characteristics of the aerobic granule in a SBR   总被引:12,自引:0,他引:12  
Owing to a fast growth rate, aerobic granules display a wide range of sizes, approximately 0.3-5.0 mm in diameter. As the diameter increases, the aerobic granule undergoes serial morphological and physical changes that could cause problems to the reactor operation, a phenomenon which, however, has not been fully studied hitherto. In this study, aerobic granules from a sequencing batch reactor were mechanically separated into various size-categories in order to investigate their physical properties, including sludge volumetric index (SVI), settling velocity (sv), specific surface hydrophobicity, granule strength, total solids, percentage volatile solids and other structural properties. Also, the live and dead biomass distribution was examined under a confocal laser scanning microscope after treatment with nucleic acid viability stains. Regardless of size, the biomass (both live and dead) was densest in the outer layer of the granule, which was about 600+/-50 microm thick. The live cells appeared only in the peripheral zone, while dead biomass spread into the inner zone. The biomass distribution pattern justified the changing physical properties of the granules as they grew bigger. As size increased, the sv, granule total density and biomass density increased but not in parallel with the size increment, while the granule strength, specific surface hydrophobicity and SVI decreased. Nonetheless, beyond a threshold size (4.0 mm diameter), the granules presented peculiar values in those properties, deviating from the initial trends. This was due to both inner and outer structural changes. The physical properties associated significantly with the size factor, for which the correlation coefficients were above 0.67. In view of biological viability and physical properties, the operational size-range suggested for optimal performance and economically effective aerobic SBR granular sludge is a diameter of 1.0-3.0 mm.  相似文献

Aerobic granulation with brewery wastewater in a sequencing batch reactor   总被引:5,自引:0,他引:5  
Aerobic granular sludge was cultivated in a sequencing batch reactor fed with brewery wastewater. After nine-week operation, stable granules with sizes of 2-7 mm were obtained. With the granulation, the SVI value decreased from 87.5 to 32 mL/g. The granular sludge had an excellent settling ability with the settling velocity over 91 m/h. Aerobic granular sludge exhibited good performance in the organics and nitrogen removal from brewery wastewater. After granulation, high and stable removal efficiencies of 88.7% COD(t), 88.9% NH(4)(+)-N were achieved at the volumetric exchange ratio of 50% and cycle duration of 6h. The average COD(t) and COD(s) of the effluent were 212 and 134 mg/L, respectively, and the average effluent ammonium concentration was less than 14.4 mg/L. Nitrogen was removed due to nitrification and simultaneous denitrification in the inner core of granules.  相似文献

A thermophilic upflow anaerobic sludge blanket (UASB) reactor degrading acetate was started by applying published methods (W. M. Wiegant and A. W. A. de Man, Biotechnol. Bioeng. 28:718-77, 1986) for production of granules dominated by Methanothrix spp. The reactor was inoculated with thermophilic digested sludge. No granules were observed during the first 7 months of start-up of the UASB reactor. However, after the concentrations of potassium, phosphate, ammonium, and magnesium in the medium were gradually increased, granules developed, indicating that there was a critical concentration of one or more of the ions required for production of granules from the starting material. After several years of stable operation, the effect of removing 60% of the granular sludge was investigated. Immunologic qualitative and quantitative studies showed that removal of the granular sludge resulted in an increase in the number of the predominant methanogens, antigenically related to Methanosarcina thermophila TM-1 and Methanosarcina mazeii S-6, and Methanobacterium thermoautotrophicum delta H and GC1. These changes were accompanied by modifications of the microanatomy of the granules, as demonstrated histochemically and immunohistochemically. The results indicated that different catabolic pathways dominated in different regions of the granules, i.e., acetate oxidation in the middle of the granules, where there is a low acetate concentration, and an aceticlastic reaction in the outer surfaces, with a high acetate concentration. The results also showed that removal of granules from a UASB reactor which has been under steady-state operation for a long period can improve the reactor's performance via formation of denser and larger granules with improved microbial activities.  相似文献

AIMS: The aim of this study is to evaluate the utility of aerobically grown microbial granules for the biological treatment of phenol-containing wastewater. METHODS AND RESULTS: A column-type sequential aerobic sludge blanket reactor was inoculated with activated sludge and fed with phenol as the sole carbon source, at a rate of 1.5 g phenol l-1 d-1. Aerobically grown microbial granules first appeared on day 9 of reactor operation and quickly grew to displace the seed flocs as the dominant form of biomass in the reactor. These granules were compact and regular in appearance, and consisted of bacterial rods and cocci and fungi embedded in an extracellular polymeric matrix. The granules had a mean size of 0.52 mm, a sludge volume index of 40 ml g-1 and a specific oxygen utilization rate of 110 mg oxygen g VSS-1 h-1 (VSS stands for volatile suspended solids). Specific phenol degradation rates increased with phenol concentration from 0 to 500 mg phenol l-1, peaked at 1.4 g phenol g VSS-1 d-1, and declined with further increases in phenol concentration as substrate inhibition effects became important. CONCLUSIONS: Aerobically grown microbial granules were successfully cultivated in a reactor maintained at a loading rate of 1.5 g phenol l-1 d-1. The granules exhibited a high tolerance towards phenol. Significant rates of phenol degradation were attained at phenol concentrations as high as 2 g l-1. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study to demonstrate the ability of aerobically grown microbial granules to degrade phenol. These granules appear to represent an excellent immobilization strategy for microorganisms to biologically remove phenol and other toxic chemicals in high-strength industrial wastewaters.  相似文献

A mathematical model was developed to describe the anaerobic ammonium oxidation (ANAMMOX) process in a granular upflow anaerobic sludge blanket (UASB) reactor. ANAMMOX granules were cultivated in the UASB reactor by seeding aerobic granules. The granule‐based reactor had a great N‐loading resistant capacity. The model simulation results on the 1‐year reactor performance matched the experimental data well. The yield coefficient for the growth and the decay rate coefficient of the ANAMMOX granules were estimated to be 0.164 g COD g?1 N and 0.00016 h?1, respectively. With this model, the effects of process parameters on the reactor performance were evaluated. Results showed that the optimum granule diameter for the maximum N‐removal should be between 1.0 and 1.3 mm and that the optimum N loading rate should be 0.8 kg N m?3 d?1. In addition, the substrate micro‐profiles in the ANAMMOX granules were measured with a microelectrode to explore the diffusion dynamics within the granules, and the measured profiles matched the predicted results well. Mathematical models are useful tools to optimize the performance of granular sludge reactors. In these models, typically a uniform granule size is assumed for the whole reactor, even though in reality the granules follow a size distribution and the granule size as such affects the process performance. This study assesses the effect of the granule size distribution on the performance of a granular sludge reactor in which autotrophic nitrogen removal is realized through one-stage partial nitritation-anammox. A comparison is made between different approaches to deal with particle size distributions in one-dimensional biofilm models, from the use of a single characteristic diameter to applying a multiple compartment model. The results show a clear impact on the conversion efficiency of the way in which particle size distribution is modeled, resulting from the effect of the granule size on the competition between nitrite oxidizing and anammox bacteria and from the interaction between granules of different sizes in terms of the exchange of solutes. Whereas the use of a uniform granule size is sufficient in case only the overall reactor behavior needs to be assessed, taking into account the detailed granule size distribution is required to study the solute exchange between particles of different sizes. For the latter purpose, the application of the widespread software package Aquasim is limited and the development of dedicated software applications is required.  相似文献

The physical structure and activity of aerobic granules, and the succession of bacterial community within aerobic granules under constant operational conditions and shock loading were investigated in one sequencing batch reactor over ten months. While the maturation phase of the granulation process began on day 30, the structure of microbial community changed markedly until after three months of reactor operation under constant conditions with a loading rate of 1.5 g phenol L−1 day−1. A shock loading of 6.0 g phenol L−1 day−1 from days 182–192 led to divergence of bacterial community, an inhibition of the biomass activity, and a decrease in phenol removal rate in the reactor. However, phenol was still completely removed under this disturbance. After the shock loading, the mean sizes of aerobic granules increased, and the activity of the microbial population within the granules decreased, although there appeared highly resilient for the dominant bacterial community of aerobic granules which mainly included β-Proteobacteria. Correlation analysis suggested that biomass concentration and biomass loading were significantly related to the community composition of aerobic granules during the whole operational period. The development of a relatively stable bacterial community in aerobic granules implied that those distinct dominant microbes in aerobic granules were favorably selected and proliferated under the operational conditions.  相似文献

This paper deals with the characteristics of anaerobic microbial granules grown in an UASB reactor treating catechol bearing synthetic wastewater (SWW). The specific methanogenic activity of the sludge showed an increase in trend with an increase in the organic loading rate and the catechol concentration in the SWW. The settling velocity of individual granules in the size range of 0.5-2.5mm was found to be in the range of 30-75mh(-1). The ash content in the sludge was 11.7% with a sludge volume index of 18-20mlg(-1). The inorganic elemental distribution within the granules showed a decrease except that for phosphorous and cobalt, which increased by approximately 12% and 18%, respectively, after the treatment of SWW. Scanning electron microscopy (SEM) coupled with electron disperse X-ray analysis showed an increase in the sulphur content by approximately 300% after the treatment of SWW. Surface mineral composition of the granules determined by XRD analysis indicated the existence of vuagnatite (CaAlSiO(4)(OH)). SEM observation of the granules showed the predominance of Methanosaeta and Methanobacterium type of species on the surface along with a variety of other species.  相似文献

树木细根具有高度空间异质性,确定合理的细根取样策略是林木细根研究的前提。通过在福建省三明米槠天然常绿阔叶林内随机钻取96个土芯,分析细根生物量和形态特征的空间变异特征,并估计各指标所需的取样数量。结果表明:(1)随着径级增加,细根各指标变异系数增大,相应的取样数量增加;(2)随着土壤深度增加,单位面积细根生物量变异程度和相应的取样数量均增加。在置信水平为95%、精度为80%的条件下,直径为0-1 mm和1-2 mm的细根,分别采集16和42个样品可以满足测定单位面积细根生物量,采集17和31个样品可以满足测定单位面积细根长度,采集25和33个样品可以满足测定单位面积细根表面积。Shapiro-Wilk检验表明,除表层土壤0-1 mm细根单位面积生物量符合正态分布外,其余细根生物量和形态指标数据均不符合正态分布。研究结果为亚热带常绿阔叶林细根的合理取样提供了科学依据。  相似文献

鱼类早期资源调查中不同网具采集效率的研究   总被引:4,自引:0,他引:4  
用网目为0.500和0.776mm的筛绢分别制成长度为1.5、2.0和2.5m的弶网、圆锥网和纺锤形网,采用网箱式、滤过式集苗桶和非滤过式集苗桶3种集苗器,于1998年5-6月在长江九江江段进行了采集仔鱼效果的比较研究。结果显示,弶网采集的数量最少,纺锤形网采集的数量最多。不同长度网具采集的数量没有显著差异,但长度为1.5m的圆锥网所采集仔鱼的规格明显小于长度为2.0和2.5m圆锥网所采集的。用网口面积分别为0.38m~2和0.20m~2的圆锥网组与不同类型集苗器进行比较,所采集仔鱼的数量和规格均没有差异。不同网目网具所采集仔鱼的数量没有显著差异,但网目为0.500mm圆锥网所采集仔鱼的平均规格显著小于网目为0.776mm圆锥网的。综合比较,以网目为0.500mm的纺锤形网的采样误差较小。  相似文献

在长江河口九段沙盐沼湿地的一条潮沟中,我们比较了两种不同网目尺寸(4mm和8mm)的插网(fyke net)及日夜潮对游泳动物群落采样效率的影响。共捕获鱼类16种5476尾、虾2种4982只。采用方差分析、柯尔莫诺夫-斯米尔诺夫非参数检验法和无度量多维标定分析对两种网目插网的捕获效率,样品的体长分布,群落结构及日夜潮间的差异进行了分析。结果表明,网目尺寸对样品中游泳动物的物种数和鱼类的多度没有影响,但是小网目的插网对虾类有较高的捕获率。两种网目尺寸的插网均在夜潮中捕获较多的鱼类个体,但虾类在日潮捕获较多。在日潮中,网目尺寸对捕获物中五种优势游泳动物的体长分布无显著影响。在夜间,小网目的插网对斑尾复缎虎和脊尾白虾的较大个体具有较高的捕获率,但对较大个体鲅的捕获率不如大网目的插网。样品中游泳动物的群落组成在日夜潮间存在明显差异,大网目捕获物的样本间差异比小网目小。  相似文献

The strictly anaerobic, pentachlorophenol (PCP) degrading bacterium DCB-2 was immobilized in an Upflow Anaerobic Sludge Blanket (UASB) reactor containing sterile granules. PCP and lactate were fed to the reactor and the concentration of chlorophenols in the effluent were monitored for 641 days. PCP was found to be degraded and transformed into 3.4.5-trichlorophenol in the reactor where DCB-2 was introduced into the granular sludge. PCP was still transformed to 3.4.5-trichlorophenol when the hydraulic retention time was decreased to six hours which was much lower than the generation time of DCB-2 insuring no free living cells in the reactor. This indicated that DCB-2 was immobilized in the granular layer. A control reactor that contained only sterile granules did not dechlorinate PCP indicating that the performance in the inoculated reactor was only due to the introduced bacteria. Immobilization of DCB-2 in the granules was further demonstrated by adding an antibody raised against DCB-2 to sliced granules. Bacteria thus visualized formed a net structure inside the granules. No DCB-2 bacteria could be found in granules from the control reactor. When lactate was omitted from the influent, the reactor still dechlorinated PCP in accordance with our findings that lactate was not used by DCB-2. This suggested that the reducing equivalents for reductive dechlorination were derived from the granules themselves. The reactor performance was 120 mol·l reactor-1·day-1, comparable to the best described performance of a UASB-reactor and to aerobic reactors. Our study demonstrates that granules can be constructed which possess specific abilities such as a dechlorinating activity and at the same time be high performing. This result have implications for eco-engineering of granules for anaerobic treatment of contaminated waters.  相似文献

The formation of granules grown on glucose in an upflow anaerobic sludge blanket (UASB) reactor was investigated. Total granular sludge concentration retained in the UASB reactor was 34.5 g MLSS/l (30.0 g MLVSS/l) during 240 d operation on glucose minimum medium with the supplementation of 1.07 g NaHCO3 per 1 g glucose. This realized a high-rate methanogenic fermentation of glucose of 17.6 g COD/l-reactor-d at 3.4 d−1 of space velocity. The granules formed were relatively small, ranging mainly from 0.4 to 0.5 mm, had a relatively low cell density of 0.0542–0.0560 g MLVSS/ml, and had low specific gravity (0.97–1.19) due to very low ash content (11–13%). Electron microscopic analysis showed that Methanothrix spp. appeared dominant over the granules. The specific metabolic activities of bacterial trophic groups were the highest for H2 followed by glucose, acetate, and propionate.  相似文献

This study attempted to address a fundamental question of whether metabolic behaviors of aerobic granules are different from their counterparts, such as activated sludge and biofilms. A series of respirometric experiments were carried out using mature aerobic granules with mean sizes of 0.75–3.4 mm. Results suggested that metabolism of aerobic granules comprised three consecutive phases: (i) conversion of external dissolved organic carbon to a poly-β-hydroxybutyrate-like substance; (ii) growth of aerobic granules on the stored poly-β-hydroxybutyrate-like substance derived from phase I, and (iii) subsequent endogenous metabolism of aerobic granules. The stoichiometric analysis revealed that the conversion yields of external dissolved organic carbon to the poly-β-hydroxybutyrate-like substance, the growth yields of biomass on storage, and the overall growth yields of biomass on external dissolved organic carbon were not significantly correlated to the sizes of aerobic granules, i.e., the metabolism of aerobic granules would be size independent. The conversion coefficients and growth rates of aerobic granules were found to be comparable with those reported in the activated sludge and biofilms cultures, indicating that there would not be significant difference in the metabolisms of aerobic granules over activated sludge and biofilms. This information will be useful for modeling and designing aerobic granular sludge processes.  相似文献

The objective of this study was to define minimum size of bioparticles that could be classified as granules, to offer all advantages of granular sludge. Based on the theory of sedimentation, the minimum diameter bioparticles, which should be considered as granules was found out for specific gravity of sludge ranging between 1.01 and 1.05. For example, for specific gravity of 1.035 the minimum diameter of granules required for better sludge retention was 0.34 mm. The diameter based on this theory was evaluated by carrying out settling column analysis of a granular sludge obtained from lab-scale UASB reactor and verified with microscopic observation. To find out the effect of granules size on the nature of biodegradability, specific methanogenic activity (SMA) was carried out. It was observed that SMA increased with size of bioparticles tested in the range of 0.27-3.03 mm. The change in VSS/SS ratio and specific gravity was observed with size of granules. Consideration of variation in specific gravity with size of granules increased the degree of validation of sedimentation theory for the calculation of granules diameter.  相似文献

Hydrogen-producing granules with an excellent settling ability were cultivated in an upflow anaerobic sludge blanket reactor treating a sucrose-rich synthetic wastewater. The physicochemical characteristics of granules were evaluated in this study. The mature granules had a diameter ranging from 1.0 to 3.5 mm and an average density of 1.036 +/- 0.005 g/mL, whereas they had good settling ability and a high settling velocity of 32-75 m/h. The low ratio of proteins/carbohydrates for the extracellular polymeric substances (EPS) in the granules suggests that carbohydrates rather than proteins, might play a more important role in the formation of the H(2)-producing granules. The contact angle of the mature granules, 54 +/- 2 degrees , was larger than that of the seed sludge (38 +/- 2 degrees ), indicating that the microbial cells in the H(2)-producing granules had higher hydrophobicity. The granules had fractal nature with a fractal dimension of 1.78. Their porosities were in the range of 0-0.70, and increased with increasing granule size. The ratios between the observed and predicted settling velocities by Stokes' law were in a range of 1.00-1.50, and the fluid collection efficiency of the granules ranged from 0 to 0.19, indicating that their permeabilities were lower and that there was little advective flow through their interior. Experimental results also suggest that molecular diffusion appeared to play an important role in the mass transfer through the H(2)-producing granules.  相似文献
