An increased rigidity of erythrocyte membranes in four-week old dystrophic chickens compared to closely related normal controls has been suggested using electron spin resonance. These findings suggest that similar to the case of human Duchenne muscular dystrophy, chicken muscular dystrophy may be associated with a generalized membrane defect.  相似文献

Myofibrillar proteins synthesized in vitro by normal and dystrophic chicken muscle polysomes were purified and analyzed by SDS gel electrophoresis. No substantial difference in the synthesis of myofibrillar proteins could be detected. These observations suggest that the loss of muscle mass that is observed in muscular dystrophy is not related to a translational defect in the dystrophic polysomes.  相似文献

Myogenesis is the developmental program that generates and regenerates skeletal muscle. This process is impaired in patients afflicted with myotonic dystrophy type 1 (DM1). Muscle development is disrupted in infants born with congenital DM1, and recent evidence suggests that defective regeneration may contribute to muscle weakness and wasting in affected adults. DM1 represents the first example of a human disease that is caused, at least in part, by pathogenic mRNA. Cell culture models have been used to demonstrate that mutant DM1 mRNA takes on a gain-of-function and inhibits myoblast differentiation. Although the molecular mechanism(s) by which this mutant mRNA disrupts myogenesis is not fully understood, recent findings suggest that anomalous RNA-protein interactions have downstream consequences that compromise key myogenic factors. In this review, we revisit morphological studies that revealed the nature of myogenic abnormalities seen in patients, describe cell culture systems that have been used to investigate this phenotype and discuss recent discoveries that for the first time have identified myogenic events that are disrupted in DM1.  相似文献

Partially purified elongation factor 1 preparations from calf brain, sheep brain, calf liver, and rabbit reticulocytes have been compared in their ability to interact with GTP and Phe-tRNA. A nitrocellulose filter assay has been used to study these interactions, and with all the EF1 preparations studied, evidence has been obtained for the formation of a Phe-tRNA·-EF1·-GTP complex. The ternary complex reacts with calf brain ribosomes in the presence of poly(U) resulting in a rapid hydrolysis of GTP and the binding of Phe-tRNA to the ribosome. Indirect evidence indicates that EF1·GDP is a product of this reaction. In the absence of poly(U) the intact complex reacts with the ribosomes without hydrolysis of GTP. The stability of the ternary complex was different with the various EF1 preparations, but the most stable complexes were prepared with calf brain EF1. Sephadex chromatography of the ternary complex shows that it contains a low molecular-weight species of the enzyme.  相似文献

Pyruvate kinase and creatine phosphokinase activities in breast muscle extracts and in serum, and protein content of the muscle extracts were determined during the first eight weeks of development of control and dystrophic chickens. In the dystrophic chicken serum enzyme levels were significantly greater than, and muscle protein content and enzyme activities on a gram wet weight basis were significantly less than control values, by the second week after hatching and thereafter. For both muscle and serum the relative differences between control and dystrophic groups was greater for pyruvate kinase than crearine phosphokinase. On a specific activity basis only pyruvate kinase levels in dystrophic muscle were significantly less than control values in 2–8-week-old chickens.  相似文献

The activity of motoneurons supplying the brachial biceps muscle was examined in eight control subjects and 26 patients affected by Duchenne muscular dystrophy. The patients were subdivided into two groups: one whose motor units (MU) fired with normal rates (N group) and the other whose MU firing rates were higher as compared to controls (I group). Firing rates of motoneurons of patients from group I increased more rapidly with increasing force level. The relationship between the standard deviation of interspike intervals and their mean value, sigma(Tm), was shifted towards the shorter intervals and lower standard deviations in both groups of patients. The numerical values describing these changes correlated with the severity of disease. The MU recruitment was comparable for control subjects and for patients. Experimental results as well as computer simulations indicate that the break-point of the function sigma(Tm) is correlated with motoneuronal properties, and in particular with the afterhyperpolarization (AHP) duration. In muscular dystrophy this break-point corresponds to the shorter interspike intervals. Therefore, we propose that the motoneurons in muscular dystrophy are altered either in response to the muscle degeneration or as a result of the disease itself.  相似文献

Dystroglycan is a central component of dystrophin-glycoprotein complex that links extracellular matrix and cytoskeleton in skeletal muscle. Although dystrophic chicken is well established as an animal model of human muscular dystrophy, the pathomechanism leading to muscular degeneration remains unknown. We show here that glycosylation and laminin-binding activity of alpha-dystroglycan (alpha-DG) are defective in dystrophic chicken. Extensive glycan structural analysis reveals that Galbeta1-3GalNAc and GalNAc residues are increased while Siaalpha2-3Gal structure is reduced in alpha-DG of dystrophic chicken. These results implicate aberrant glycosylation of alpha-DG in the pathogenesis of muscular degeneration in this model animal of muscular dystrophy.  相似文献
