The effects of light and temperature on flowering and pollentube growth were studied in watermelon [Citrullus lanatus(Thunb.)Matsum. and Nakai, cv. Early Yates] plants grown in controlledenvironment cabinets. All female flowers were pollinated inone group of plants; none was pollinated in the other group. Temperature increase from 25 °C to 35 °C with daylengthof 14 h and light intensity of 32 klx caused increase in flowernumber per plant, proportion of male flowers, ovary length anddiameter, ovule number per ovary, rate of pollen tube growthand percentage of penetrated ovules at 24 hand 48 h after pollination.Very few flowers were produced at 40 °C, but there was ahigh proportion of male flowers. Increase in daylength from14 h to 24 h at 25 °C with light intensity of 32 klx alsoincreased number of flowers per plant, ovary length and diameterand number of ovules per ovary but sex expression and rate ofpollen tube growth were unaffected. Reduction in daylength from14 h to 8 hat 25 °C and light intensity of 32 klx and reductionin light intensity from 32 klx to 8 klx at 25 °C and 14h daylength both produced an increase in the percentage of immatureovules. The presence of fruit on the vine resulted in fewerflowers per plant and in reduced ovary legnth and diameter underall conditions tested. The results are discussed in relation to the fruiting responseof the plant.  相似文献

Watermelon [Citrullus lanatus (Thunb.) Matsum. and Nakai] plug seedlings were stored at 15°C in the light at a photosynthetic photon flux density of 15 µmol·m−2·s−1 or in darkness for 6 days, to evaluate their chloroplast ultrastructure, and associated photosynthetic characteristics. Storage in the dark caused swelling, disordered granal arrangement, and starch grain disappearance in the chloroplasts. In contrast, the chloroplasts stored in the light were relatively normal. As a result, the light-stored seedlings had a significantly higher chlorophyll content, Fv/Fm, and Pn than did dark-stored seedlings. Regardless of whether the seedlings were stored in light or darkness, the Gs and Ls of the seedlings significantly decreased, while the Ci obviously increased when the Pn decreased after 6 days of storage. This result suggests that the decreased Pn is not solely a stomatal effect, as the effects on the chloroplasts contributed to this photosynthetic inhibition. Six days after transplanting, seedlings that were stored in the light or darkness for 2 or 4 days showed complete recovery of chloroplast ultrastructure, chlorophyll content, Fv/Fm, Gs and Pn. When the storage period increased to 6 days, the dark-stored seedlings had a significantly lower Fv/Fm and Pn than the light-stored and control seedlings 6 days after transplanting, which was mainly ascribed to incomplete recovery of chloroplast ultrastructure. Furthermore, the light-stored seedlings exhibited a significantly higher shoot dry weight during storage and a higher percentage dry weight increase after transplanting than the dark-stored seedlings. These effects were enhanced by prolonged storage (4 to 6 days). This study demonstrated that dim light during storage is beneficial for maintaining chloroplast ultrastructure as well as photosynthetic efficiency in watermelon seedlings, thus contributing to the rapid recovery of post-storage photosynthetic performance, which ensures the transplant quality of the seedlings after removal from storage.  相似文献

Prerooted cuttings of grape-vine (Vitis vinifera L., cultivarMuscat Gordo Blanco, syn. Muscat of Alexandria) were grown ingrowth cabinets for 13 weeks after bud-burst. Environmentalconditions were light intensities of 3600 foot candles (f.c.),18oo f.c., and 9oo f.c. in all combinations with temperaturesof 30 ?C day/25 ?C night, 25 ?C/20 ?C, and 20 ?C/15 ?C. Daylengthwith both fluorescent and incandescent lighting was 16 h with8-h nights. Measurements of main and lateral shoot lengths,leaf numbers, and leaf areas were made at weekly intervals for9 weeks. At 13 weeks the plants were harvested and dry weightsof roots, parent canes, stems, and leaves measured. Dry weights increased with increasing light intensity and weregreaert at 25 ?C than at 20 ?C or 30 ?C. The new shoot: rootratio remained at approximately 7.o under all conditions, butincreasingly more dry weight was in leaves and correspondinglyless in stems with increased temperature. Stem lengths and leafarea values were in most cases greatest at 25 ?C followed inturn by 30 ?C and 20 ?C values. Light intensity had no cleareffect. Apical dominance increased with increasing temperature.Internode length was stable under all conditions. The erraticgrowth of lateral shoots was one factor making the results difficultto interpret.  相似文献

将西瓜野生种质PI296341抗枯萎病生理小种1的抗性基因连锁的RAPD标记OPP01.700进行克隆、测序,Southern杂交证明此标记为1个单拷贝,并转化为SCAR标记,简化了SCAR扩增产物的检测技术。上述技术在抗病转育后代造反中得到了很好的应用,初步建立了西瓜抗枯萎病育种分子标记辅助选择技术系统。  相似文献

分子标记在作物育种、资源遗传多样性分析、遗传图谱构建等方面有着广泛的应用,常见的分子标记方法主要有RFLP、RAPD、SSR、AFLP、SCAR、CAPS、SNP等.迄今为止,国内外学者已经利用分子标记技术对西瓜和甜瓜的20多个质量性状进行了标记和定位.本文主要对西瓜和甜瓜质量性状的分子标记和定位研究进展进行了综述,发现RAPD标记使用最多,而更具优势的SSR标记使用较少;对抗病虫等育种相关的基因研究较多,且多数标记遗传距离偏大,达不到图位克隆的要求,而对西瓜和甜瓜遗传基础相关的基因研究较少.  相似文献

Volatile flavor components of watermelon fruit (Citrullus vulgaris) were obtained by distilling juice under reduced pressure and extracting the resulting distillate with Freon-11. The volatiles were separated into acidic and neutral fractions and then analyzed by gas chromatography and gas chromatography-mass spectrometry.

Fifty-two compounds were found for the first time as flavor components of watermelon. Among them, the following compounds have not been previously reported as naturally occurring flavor components: 4-oxononanal and 2-hydroxy-, 2-pentyloxy-, 2-hexyloxy-, 2-octyloxy-, 2-nonyloxy-, 2-[(Z)-3-noneyloxy]-, 2-[(Z)-2-nonenyloxy]-, 2-[(Z,Z)-3,6-nonadienyloxy]-, 2-benzyl-oxy-, and 2-phenetyloxy-5-pentyltetrahydrofurans.  相似文献

Changes in the dry weight (dry wt.), total and neutral lipids,and fatty acid composition were determined in the cotyledonsand axis of Citrullus lanatus cv. Sugar baby seedlings duringtheir first 12 d growth in the dark, at 25 ? 1 ?C. The major stored reserves were mobilized between days 3 and8. The lipid concentration after day 4 decreased rapidly inthe cotyledons. On the first day the lipid concentration inthe axis was higher than in the cotyledons but decreased rapidlyand from day 5 up to the end of the experiments it remainedat very low values. In the cotyledons, the change in the fattyacid composition was relatively small compared to the significantchange observed in the axis. The neutral lipids of the cotyledonsdecreased sharply between days 2 and 6 while their fatty acidcomposition suffered no significant change. The axis, comparedto the cotyledons, contained a small but concentrated amountof neutral lipids, which decreased up to day 6. Thereafter,an increase was observed. The fatty acid composition of theaxis neutral lipids changed significantly, linoleic and oleicacid decreased while palmitic and linolenic acid increased. 为探究环境因子对针叶蕨藻(Caulerpa sertularioides)生长的影响,对不同盐度、温度和光照强度下针叶蕨藻的生长和叶绿素荧光参数进行了研究。结果表明:藻体日特定生长率(SGR)、最大光量子产量(Fv/Fm)、实际光合效率(Yield)、电子传递速率(ETR)和光化学淬灭(qP)随盐度升高呈先上升后下降的变化趋势,非光化学淬灭(qN)则呈相反的变化趋势,藻体光合活性和固碳效率在盐度27.5‰时达到最高,且与25‰和30‰盐度的差异显著(P<0.05,n=3)。藻体SGR、Fv/Fm、Yield、ETR和qP随温度升高而下降,qN则相反,藻体光合活性和固碳效率在26℃下达到最高,且与28℃和30℃的差异显著(P<0.05,n=3)。藻体的SGR、Fv/Fm、Yield、ETR和qP随光照强度升高呈先上升后下降的变化趋势,qN则相反,且在18.75 μmol/(m2·s)弱光照下出现轻微光抑制,藻体生长、光合活性及固碳效率在光照强度25.00 μmol/(m2·s)时达到最高,但与18.75和31.25 μmol/(m2·s)的差异不显著(P>0.05,n=3)。因此,针叶蕨藻在27.5‰盐度、26℃和25.00 μmol/(m2·s)光照强度下生长最快且光合作用能力最高。  相似文献

Phenylalanine ammonia-lyase (PAL), the first enzyme in the phenylpropanoid pathway, plays a critical role in plant growth, development, and adaptation. PAL enzymes are encoded by a gene family in plants. Here, we report a genome-wide search for PAL genes in watermelon. A total of 12 PAL genes, designated ClPAL1-12, are identified . Nine are arranged in tandem in two duplication blocks located on chromosomes 4 and 7, and the other three ClPAL genes are distributed as single copies on chromosomes 2, 3, and 8. Both the cDNA and protein sequences of ClPALs share an overall high identity with each other. A phylogenetic analysis places 11 of the ClPALs into a separate cucurbit subclade, whereas ClPAL2, which belongs to neither monocots nor dicots, may serve as an ancestral PAL in plants. In the cucurbit subclade, seven ClPALs form homologous pairs with their counterparts from cucumber. Expression profiling reveals that 11 of the ClPAL genes are expressed and show preferential expression in the stems and male and female flowers. Six of the 12 ClPALs are moderately or strongly expressed in the fruits, particularly in the pulp, suggesting the potential roles of PAL in the development of fruit color and flavor. A promoter motif analysis of the ClPAL genes implies redundant but distinctive cis-regulatory structures for stress responsiveness. Finally, duplication events during the evolution and expansion of the ClPAL gene family are discussed, and the relationships between the ClPAL genes and their cucumber orthologs are estimated.  相似文献

During germination of Citrullus lanatus, pyrophosphate-dependent phosphofructokinase (PFP) activity is induced. The peak of PFP activity coincides with the maximum gluconeogenic flux and high fructose-2,6-bisphosphate (Fru-2,6-P2) concentrations. Determination of cytosolic fructose-1,6 bisphosphatase (FBPase) activity in crude extracts is unreliable because of the high PFP activity. The FBPase activity, after correction for the contaminating PFP, is only one-third of the PFP activity. Purified cytosolic FBPase is inhibited by Fru-2,6-P2. The low cytosolic FBPase activity and high Fru-2,6-P2 most probably result in inadequate in vivo activity to catalyze the observed gluconeogenic flux. The total PFP activity is sufficient to catalyze the required carbon flux.  相似文献

以NC89烟草品种为材料,用白纱布遮荫研究了100%、80%、50%和20%全光照处理对烤烟生长发育、光合特性及化学成分的影响.结果表明:(1)随光照强度的减弱,植株茎杆变细,叶片的长宽比增加,叶片数减少,出叶速度变慢,干物质重逐渐减少,并以80%的全光照条件下的株高、叶面积最大.(2)净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)、叶绿素a/b值均随光照减弱而逐渐下降,胞间CO2浓度(Ci)、叶绿素含量逐渐升高;叶绿素荧光参数Fm、ФPSⅡ、Fv/Fm、QP值逐渐增加.(3)烟碱、淀粉、还原糖、水溶性总糖、总酚含量随光照减弱而降低,而总氮和钾含量升高.综合分析各项测定指标发现,在大于或等于80%全光照(光照强度为770~1 125μmol?m-2?s-1)条件下,烤烟的生长指标变化不大,品质指标符合不同卷烟工业需要,适合优质烟叶的生产.  相似文献

Random amplified polymorphic DNA(RAPD) was employed to detect a molecular marker linked to Fusarium wilt resistant gene in the wild watermelon ( Citrullus lanatus (Thunb.) Mansfeld var. citroides ) germplasm P1296341. The resistance to race 1 Fusarium wilt of PI296341 was controlled by one dominant gene. A RAPD marker OPPO1/700 was proved to be linked to the resistant gene. The genetic distance is 3.0 cM (centimorgan). This work has provided a solid basis for molecular marker-assisted selection (MAS) for disease resistance, and made location and cloning of disease resistant genes possible.  相似文献

以瓠瓜品种‘福州芋瓠’为试材,在南方塑料大棚内密闭式光照植物培养架中,以荧光灯作为共同对照(CK),采用发光二极管(LED)调制光谱能量分布,设置50、100、120μmol·m~(-2)·s~(-1) 3个水平(表示为L50、L100、L120),红、蓝配比设置3∶7和7∶3等2个水平(表示为RB3∶7、RB7∶3),包括对照组共7个处理组合,各处理每天照光12h(8:00~20:00),研究不同光强和红蓝光配比对瓠瓜幼苗生长及其生理生化特性的影响。结果表明:(1)在处理7~14d时,瓠瓜幼苗叶片的叶绿素a、叶绿素b、叶绿素总量及其类胡萝卜素含量均随着处理天数的增加而增加,但处理后期,各色素指标有所下降。(2)光强一致时,红光比重大的处理组瓠瓜幼苗叶片可溶性糖含量较蓝光比重大的处理组增大更为显著;蓝光比重大的处理组的瓠瓜幼苗叶片可溶性蛋白质含量较红光比重大的处理组增大更为显著。(3)在光质红蓝配比分别RB3∶7、RB7∶3条件下,生长指标(株高、茎粗、叶面积、下胚轴长、壮苗指数和生物量)及若干生理生化特性(光合色素、可溶性糖、可溶性蛋白质和丙二醛含量)均随着光强的增大而显著提高,而MDA含量则显著降低。研究表明,瓠瓜幼苗在光强为120μmol·m~(-2)·s~(-1)下生长较好,且红蓝光配比为7∶3时更有利于瓠瓜幼苗的生长。  相似文献

The effects of light intensity and temperature on the changesin growth and development induced by sodium 2 4-dichlorophenoxyacetatehave been studied in multifactorial experiments. Random samplesfrom a clonal population of Lemna minor were subjected to threeintensities (700, 275, and 180 foot-candles) and three temperatures(30°, 25°, and 20° C.), while varying concentrationsof the growth regulator were added to a phosphate-buffered culturesolution (pH 5:1 There were up to five sampling occasions whenthe dry weights and, for some purposes, the total frond areaswere determined. The replication was at least twofold, and thelargest experiment involved 720 dry-weight determinations. At each combination of light and temperature, sublethal concentrationsof the growth regulator induced cumulative reductions in therelative growth rate and the order of the depression increasedprogressively with concentration-3, 9, 27, and 45 p.p.m. Becauseof the cumulative effects and the great differences in the growthrates of the controls under the nine combinations of light andtemperature, the statistical analysis of the data was involved.Employing two different approaches it was concluded that therelative depression in the growth rate increased with temperaturewhile the growth of plants receiving the highest intensity wasslightly more inhibited. At a concentration (27 p.p.m.) which reduced both the relativegrowth and frond area, over all combinations of light intensityand temperature the net assimilation rate during the periodso–2 and 2–4 days was depressed by 2 and 12 per cent.respectively. There was some indication that the depressionwas larger at the highest temperature in the second period. At greatly increased concentrations the fronds exhibited chioroticsymptoms, and the concentrations required to produce a standarddegree of chiorosis at the end of 3 days were determined undertwelve combinations of light intensity and temperature. It wasfound that varying the intensity during the time of treatmentwas without effect, but that there was a highly significantinfluence of the light level prior to treatment: the lower theintensity the smaller was the concentration subsequently requiredto induce chlorosis. Likewise, with a rise in temperature themagnitude of the equi-effective concentration was reduced. These interactions are discussed in relation to changes in theratio of frond area to frond weight, the mechanism of uptake,and possible physiological effects at cell level.  相似文献
