TwoPyxidanthera morphs of questionable taxonomic rank are described in the eastern United States. We analyzed leaf samples from ninePyxidanthera populations (four of each morph and one with intermediate morphology) for 13 allozyme loci. Our results do not support differentiation of the two morphs at the species level. Mean genetic identity among populations was high ( = 0.97), and typical of that found for conspecific populations. The proportion of total genetic diversity found among populations was low (GST = 0.079). Several low frequency alleles were confined to each of the morphs, being found in some of the populations of each morph, but not all.  相似文献

Factors enabling and limiting self-fertilization were examined in populations of Epipactis palustris, E. helleborine and E. atrorubens located in NE Poland. In these species we found self-compatibility within the same flower (even 100% of fruit set from induced autogamy) and the same inflorescence (reaching 90–100% of fruit set from induced geitonogamy). Facultative autogamy was found only in E. palustris (even 82.5% of fruit set in a bagged inflorescence), due to underdevelopment of the clinandrium and projection of pollinia over the rostellum. Allogamy was predominant in E. helleborine (maximum to 2.8% of fruits set spontaneously) and E. atrorubens (maximum to 3.4%), attributable to a well-developed rostellum and clinandrium and to the position of pollinia. Autonomous selfing takes place during flower wilting, when the viscidium becomes evanescent and the pollinia are fragile. Selfing is enabled mostly by pollinator behaviour, promoting both geitonogamy and autogamy, and influencing mixed-mating.  相似文献

Genetic divergence between population samples ofOrchis laxiflora and ofO. palustris from various European locations was studied by electrophoretic analysis of 25 enzyme loci. An average genetic distance of DNei = 1.24 was found between the two taxa, with 12 out of 25 loci showing alternative alleles (diagnostic loci). Genetic heterogeneity was observed within bothO. laxiflora andO. palustris, when northern and southeastern populations were compared, being lower in the former taxon (D = 0.06), than in the latter (D = 0.16). Karyologically, 2n = 36 was found for bothO. laxiflora andO. palustris. O. laxiflora andO. palustris produce hybrids, described asO. ×intermedia. Genotype analysis of several sympatric samples showed the presence of hybrid zones, including F1 hybrids and, in low proportions, recombinant classes, putatively assigned to Fn and backcrosses, as well as a few introgressed individuals of both taxa. These data indicate that hybrids are only partially fertile, with a very limited mixing up of the two parental gene pools; this is also shown by the lack of significant lowering of genetic distances when sympatric and allopatric heterospecific samples are compared. Accordingly,O. laxiflora andO. palustris form a syngameon; nevertheless they can be considered as good taxonomic species, with virtually distinct gene pools, which evolve independently. The genetic variability inO. laxiflora andO. palustris is remarkably low ( e = 0.05 and e = 0.02, respectively). In particular, nearly complete absence of polymorphic loci was found inO. palustris from northcentral Europe. Two hypotheses are considered to explain the low genetic variability of this endangered species.  相似文献

Fourteen enzyme systems were analysed in leaf parenchyma of nine native and introduced populations of teak. These enzyme systems were encoded by 20 putative loci of which 18 were polymorphic. Populations showed a general lack of heterozygosity (average FIS = 0.11). On average over the 18 polymorphic loci, the genetic differentiation among provenances varied according to the estimator: 0.09 for GST, 0.12 for FST and 0.19 for . The cluster analysis showed two main gene pools, the first consisted of the Indian provenances and the second of African, Indonesian and Thai provenances. Genetic distances among populations of the same group were similar, and lower than the genetic distances between populations from different groups. The factorial analysis on genotypes of seedlings also showed the same geographic differentiation into two major groups. The possible natural distribution of teak in Java is discussed.  相似文献

Spiny rats from Venezuela show an extensive karyotypic diversification (2n=24 to 2n=62) and little morphological differentiation. This study reports genetic distance, heterozygosity and polymorphism based upon 22 loci in semispecies and allospecies of the Proechimys guairae superspecies from N Central Venezuela, as compared with Proechimys urichi, a member of the Proechimys trinitatis superspecies from eastern Venezuela. Four chromosome forms of the P. guairae complex are included, each characterized by karyotypes of 2n=46 (Fundamental Number=72), 2n=48 (FN=72), 2n=50 (FN=72) and 2n=62 (FN=74). Proechimys urichi has a distinetive karyotype of 2n=62 (FN=88). The overall mean value of Nei's genetic identity index for all pair-wise comparisons is I=0.942±0.011. Mean identity within the P. guairae complex is =0.969±0.033. Mean identity between P. urichi and members of that complex is =0.889±0.011. Within the P. guairae complex, increased genetic divergence is correlated with higher karyotypic divergence. Heterozygosity varies from H=0.059 to H=0.153, with a mean value of H0.059. The mean percent of polymorphic loci is P=18.2±3.9 after the 0,95% polymorphism criterion, and P=20,5±5.2 after the 0.99% criterion. These results are compared with similar data from fossorial and non-fossorial rodents. Spiny rats are non-fossorial, forest-dwelling rodents which have undergone a speciation process with little genetic divergence and extensive chromosome rearrangements.  相似文献

Since high levels of genetic diversity may ensure long-term survival of a plant species, it is essential to preserve the genetic diversity of the species. Tipularia japonica and Epipactis papillosa are rare terrestrial orchids in southern Korea with fewer than 50 mature individuals in a population and southern Japan and considered to be threatened (endangered or vulunerable). To obtain knowledge of how the genetic variation of these species is partitioned within and among populations in Korea, I used enzyme electrophoresis to examine the genetic diversity of each eight known populations of the two species from South Korea. Twenty-three (E. papillosa) and 24 putative loci (T. japonica) resolved from 15 enzyme systems revealed no variation either within or among populations of each species (0.0% of the percentage of polymorphic loci, %P). Previous studies, in contrast, showed that their more widely distributed disjunct congeners T. discolor and E. helleborine harbored high allozyme-based genetic diversity within populations in eastern United States (%P = 75%) and in Denmark (%P = 73.6%), respectively. In theory, small population size leads to allelic fixation at many loci over generations within a population, resulting in population genetic divergence or differentiation. In this regard, the complete lack of genetic differences between conspecific populations of T. japonica and E. papillosa cannot be explained by genetic drift. Instead, the present allozyme data suggest that recent origin from the same genetically depauperate ancestral or source population could result in this observation. The current status of T. japonica and E. papillosa (rarity and lack of genetic variation) significantly threatens the long-term survival of the species in Korea.  相似文献

Random amplified polymorphic DNA (RAPD) markers were used to determine the levels and pattern of molecular variation in four populations of Elymus trachycaulus, and to estimate genetic similarity among different populations of E. trachycaulus from British Columbia and the Northwest Territories and one population of Elymus alaskanus from the Northwest Territories. Based on 124 RAPD bands (loci), mean percent polymorphic loci for E. trachycaulus (PP) was 67.4% (a range 41.2% to 86.3%), and mean gene diversity (He) for E. trachycaulus species was 0.23 (range 0.18 to 0.27). The total genetic diversity was 0.32. Differentiation among populations was 31% (FST = 0.31) with most of the genetic variation found within populations (69%). The genetic structure of ten natural populations of Arabidopsis thaliana (L.) Heynh. at eight isozyme loci was studied. The populations were located in the northern part of the species range, 200 km from the north to the south along the Onega Lake coast in Karelia. Considerable genetic diversity (P 99% = 43.7, H obs = 0.003) was revealed that is not typical of populations of self-pollinating plant species. A direct correlation between the proportion of polymorphic loci and geographical latitude was shown (r = 0.68; P < 0.05). It is suggested that a high polymorphism level in Karelian Arabidopsis thaliana (L.) populations increasing from the south to the north is due to extreme environmental conditions in the northern part of the species range. The distribution of genetic diversity within and between populations is typical of self-pollinating species: the larger part of the total diversity resides among populations (G ST = 0.583).  相似文献

The genetic control of adult-plant blackleg [Leptosphaeria maculans (Desm.) Ces. et De Not.] resistance in rapeseed (Brassica napus L.) was studied in the F2 and first-backcross populations of the cross Maluka (blackleg-resistant) x Niklas (highly susceptible). A L. maculans isolate possessing high levels of host specificity (MB2) was used in all inoculations. Resistance/susceptibility was evaluated using three separate measures of crown-canker size, i.e. the percentage of crown girdled (%G), external lesion length (E) and internal lesion area (%II). Disease severity scores for the F2 and first-backcross populations based on E and %II gave discontinuous distributions, indicating major-gene control for these measures of resistance; but those for %G were continuous, indicating quantitative genetic control for this measure. Chi-square tests performed on the (poorly-defined) resistance classes, based on E, in the F2 and first-backcross populations indicated the likelihood for resistance being governed by a single, incompletely dominant major gene. Although the distributions of the F2 and first-backcross populations, based on%II, were clearly discontinuous, the observed segregation ratios for resistance and susceptibility did not fit any of the numerous Mendelian ratios which were considered. Differences in inheritance of resistance according to the assessment method and blackleg isolate used, were discussed.  相似文献

Summary Collagen and fibronectin synthesis by trisomic and triploid fibroblasts derived from human spontaneous abortuses was studied. It was demonstrated that the level of fibronectin and collagen production in fibroblasts with trisomy 7, trisomy 9, and triploidy was reduced as compared with diploid cells. A correlation between this observation and an increased rate of intracellular 14C-procollagen degradation was also established for the anomalous strains. No difference in hydroxylation of 14C-proline residues in 1() and 2() collagen chains and no fluctuation in the collagen type (): type ratio was found in the strains with the abnormal karyotypes. It was concluded that differentiation of the abnormal fibroblasts was impaired. The data also favour the hypothesis that the deficiency of the fibroblasts in producing proteins may account for a variety of anatomic abnormalities of embryos.  相似文献

Epipactis helleborine (L.) Crantz (Orchidaceae, Neottieae) can spread by sexual or vegetative propagation. The choice of strategy likely depends on the environmental conditions. The rhizome is the organ of vegetative reproduction; hence, it is crucial to understand its development. Unfortunately, it is hardly possible to investigate rhizome morphology directly, since E. helleborine is a protected species in most European countries. The goal of our investigation was to infer the growth patterns of underground parts of an orchid population from long-term annual observations of its aboveground shoots. We implemented the Minimum Spanning Tree method to determine a likely set of underground connections between shoots and to simulate the annual growth of new rhizomes. Furthermore, we modelled the spatial distribution of shoots with a density kernel estimator to compare the density gradients with the direction of growth of the rhizomes. Observed shoot numbers fluctuated between 72 and 183 from year to year. Our results suggest that (1) vegetative reproduction prevails in the studied population, (2) the population consists of about a dozen clones with a diameter of up to 6 m, (3) rhizomes produce up to five new shoots at one branch end per year, (4) rhizomes develop in the direction of decreasing population density, and (5) nodes of rhizomes may produce new offshoots after up to 7 years of dormancy.  相似文献

The purpose of his research was to describethe organization of genetic variation in thefederally endangered plant taxon Eriogonumovalifolium var. vineum using allozymes. Such information can help prioritize sites andmanagement choices for capturing andmaintaining genetic variation and can reducethe number of populations necessary to committo conservation, thus reducing costs andconflicts with competing land uses. Information on genetic diversity patterns alsoprovides insight into evolutionary anddemographic history of a taxon which canprovide means to assess future risk of erosionof diversity. Similar to other Eriogonumovalifolium varieties, E. ovalifoliumvar. vineum is diverse atboth the taxon (proportion of polymorphic loci[P] = 0.55 and alleles per locus [A] =5.45 [SD = 2.5]) and population (P = 0.56 [SD =0.11] and A = 2.68 [SD = 0.35]) levels. Gene diversity (H e) withinpopulations averaged 0.19 (SD = 0.03). Wefound some evidence for limited clonalreproduction within populations. Populationswere moderately differentiated from one another(S = 0.14) and showed moderatedeviations from Hardy-Weinberg equilibriumwithin populations (f = 0.14 and F =0.19). Mean Nei's genetic distance (D)among all pairs of populations was 0.02. Populations with high levels of multiplemeasures of genetic diversity, high levels ofdifferentiation, and low levels of apparentinbreeding are suggested as conservationpriorities.  相似文献

The genetic diversity, subdivision, and differentiation of nine populations of Norway spruce (Picea abies (L.) Karst.) in Ukrainian Carpathians were studied using electrophoretic analysis of variability of enzyme systems in 346 trees aged from 80 to 150 years. Based on electrophoretic fractionation of enzymes extracted from seed endosperms in vertical slabs of 7.5% polyacrylamide gel, 20 loci of nine enzyme systems (ADH, ACP, DIA, GDH, GOT, MDH, LAP, FDH, SOD) were identified, and 71 allele variant were revealed. Each tree was heterozygous on average in 15.8% of its genes. The populations were characterized by low subdivision (F ST = 0.017) and differentiation (D N = 0.005). The main contribution to heterogeneity of population genetic structure was made by loci Dia-3, Lap-1, and Sod-3. Clustering and multivariate analysis revealed no observed trends in geographical or altitudinal position of the populations.  相似文献

Summary 3 families with hereditary angioedema in 9 members are described. The serum concentrations of complement factors C3, C4, and C-inhibitor (CINH) were determined in all available family members. C inhibiting activity was characteristically low or absent in sera from patients. CINH and C4 concentrations were also low, whereas C3 was present in normal concentrations in patients and in unaffected family members. In all 3 families fatal attacks of laryngeal edema had occured. 4 individuals, 2 children and 2 adults, had low concentrations of CINH and C4 but had never experienced symptoms of angioedema.
Zusammenfassung 3 Familien mit hereditärem Angioödem bei insgesamt 9 Mitgliedern werden beschrieben. Die Serumkonzentrationen der Komplementfaktoren C3, C4 und des C-Inhibitors (CINH) wurden bei allen erreichbaren Familienmitgliedern bestimmt. Die C-Hemmaktivität und die Konzentrationen von CINH und C4 waren in den Patientenseren niedrig. Der C3-Serumspiegel war jedoch bei Patienten und gesunden Familienmitgliedern normal. In allen 3 Familien sind tödlich verlaufende Anfälle von Larynxödem vorgekommen. 4 Familienmitglieder, 2 Kinder und 2 Erwachsene, waren trotz niedriger Serumkonzentrationen von C4 und CINH asymptomatisch.

This report describes an integrative analysis of seed-oil-content quantitative trait loci (QTL) in Brassica napus , using a high-density genetic map to align QTL among different populations.

Abstract

Rapeseed (Brassica napus) is an important source of edible oil and sustainable energy. Given the challenge involved in using only a few genes to substantially increase the oil content of rapeseed without affecting the fatty acid composition, exploitation of a greater number of genetic loci that regulate the oil content variation among rapeseed germplasm is of fundamental importance. In this study, we investigated variation in the seed-oil content among two related genetic populations of Brassica napus, the TN double-haploid population and its derivative reconstructed-F2 population. Each population was grown in multiple experiments under different environmental conditions. Mapping of quantitative trait loci (QTL) identified 41 QTL in the TN populations. Furthermore, of the 20 pairs of epistatic interaction loci detected, approximately one-third were located within the QTL intervals. The use of common markers on different genetic maps and the TN genetic map as a reference enabled us to project QTL from an additional three genetic populations onto the TN genetic map. In summary, we used the TN genetic map of the B. napus genome to identify 46 distinct QTL regions that control seed-oil content on 16 of the 19 linkage groups of B. napus. Of these, 18 were each detected in multiple populations. The present results are of value for ongoing efforts to breed rapeseed with high oil content, and alignment of the QTL makes an important contribution to the development of an integrative system for genetic studies of rapeseed.  相似文献

Great variability involving 36 quantitative and 31 qualitative characters, as well as the habitats occupied has been shown in 18 populations of Elymus caninus from Poland, based on the detailed population and statistical analyses of morphological characters. The variability of these characters proves that the intraspecific units which have been distinguished on the basis of these characters do not have systematic significance. The two morphotypes of E. caninus (pauciflorum and caninus) described in this study will provide the basis for further genetic studies aimed at establishing their significance for considerations in the systematics of the species.It is the first paper from the series: Biodiversity of wild Triticeae (Poaceae) in Poland.  相似文献

Isozyme analysis was used to assess the origin of the DanishEpipactis renzii, a local endemic of open coastal dunes. It seems to have evolved recently in several local populations of the more or less allogamousE. helleborine subsp.neerlandica. The obligately autogamousE. renzii is restricted to the easternmost part of the Danish range ofE. helleborine subsp.neerlandica, indicating that transition to obligate autogamy has been a selective advantage in that area. This may be explained by water stress, caused by a higher evapotranspiration (cf. the mean temperature of July which increases towards the east) — obligate autogamy shortens the time span from anthesis to fruit set due to a rapid pollination process. We propose thatE. renzii should only be recognized as a variety ofE. helleborine subsp.neerlandica.  相似文献

The genetic diversity and population structure of 25 natural populations of Thymus capitatus, from five ecological areas, were analysed using eight isozymes. For all populations, 11 polymorphic loci were detected, and several of them showed rare alleles. A high genetic diversity within populations (Ap = 1.82; P = 62.88%; Ho = 0.116) and an excess of heterozygosity were observed. A high level of inbreeding within populations was observed (FIS = 0.471). High differentiation and low gene flow (Nm = 0.821) were detected among populations (FST = 0.219). The genetic variation within and among ecological groups varied according to the bioclimate. Population structuration depends on geographic distance between sites rather than on bioclimate (Mantel’s test; r = 0.251; P = 0.004). Nei’s genetic distances (D) values calculated between pairs of populations were globally low with a mean of 0.047. The unweighted pairwise groups method using arithmetic average dendrogram showed fourth sub‐clusters. Population groupings occur with evident relationship to geographic location. The substantial differentiation and the high genetic similarities between populations indicate that populations have been recently isolated as a result of anthropic pressure. In situ conservation must first focus on populations with a high level of genetic diversity and rare alleles. Ex situ preservation should be elaborated by collecting seeds within populations that showed a high level of genetic diversity in each ecological group.  相似文献

The terrestrial orchid Epipactis helleborine is a morphologically variable species with a wide geographical distribution. It is found throughout Europe and continues eastwards to Siberia, China and Japan. It is usually pollinated by social wasps and displays the morphological characteristics of an outcrossing species. In warm, temperate areas of Japan, E. helleborine often appears in alpine or subalpine regions, and has never been found in low‐altitude forests, except for coastal pine forests. The coastal population of E. helleborine is often classified at the variety level, as E. helleborine var. sayekiana, and the inland populations are known as E. helleborine var. papillosa. It is possible that E. helleborine var. sayekiana possesses a distinctive selfing strategy, as its autonomous self‐pollination has evolved in dry habitats, such as coastal dunes. The present study investigated the pollination biology of E. helleborine var. sayekiana and var. papillosa to detect differences in their reproductive systems. Unexpectedly, both E. helleborine var. papillosa and E. helleborine var. sayekiana were found to possess a self‐pollination strategy and were therefore rarely visited by insects. Self‐pollination occurs at the end of the flowering season and probably acts to ensure pollination even if insect‐mediated pollination fails. Moreover, there are no floral differences between E. helleborine var. papillosa and E. helleborine var. sayekiana. These observations suggest that it is not necessary to distinguish between these two varieties. Furthermore, E. helleborine var. papillosa was pollinated by hoverflies, suggesting that E. helleborine var. papillosa is probably biologically distinct from its mother species, E. helleborine. © 2013 The Linnean Society of London, Botanical Journal of the Linnean Society, 2013, 173 , 733–743.  相似文献
