Summary Intermediate filaments of epithelial cells generally consist of specific combinations of keratins. However, cultured epithelial cells from certain tissues and some epithelial tumors have been shown also to express vimentin. In the present study, the expression of vimentin by epithelial cells in healing corneal wounds (partial thickness penetrating wounds) and in tissue culture was analyzed. Both immunohistochemical and immunotransblot analyses indicated that although vimentin was not detected in the normal rabbit corneal epithelium in vivo, cultured rabbit corneal epithelial cells co-express keratins and vimentin. At 1 day post-wounding, vimentin was not detectable in the epithelial cells that had covered the denuded stroma. However, at 2 days post-wounding, the epithelium at the base of the epithelial plug immunoreacted with both anti-vimentin and antikeratin monoclonal antibodies. Immunotransblot analyses of the extracts of the epithelial plugs confirmed the presence of vimentin (Mr=58k). The 58k band was not detected in the extract of normal rabbit corneal epithelium. At day/5, vimentin was no longer detectable in the epithelium. This study demonstrated that corneal epithelial cells transiently co-express vimentin and keratins in vivo during wound healing and in tissue culture. The time-course of the transient expression of vimentin suggests that the vimentin expression in the epithelial cells during healing is not linked to cell proliferation or to the centripetal migration of the epithelium during early stages (first 24 h) of healing, but may be linked to cell-matrix interactions or the migration of basal cells in the upward direction at the following stage of healing.  相似文献

Summary This study aims to describe the regulation of vimentin and cytokeratin expression during differentiation of primary mesenchymal cells in the 7 day old rabbit embryo; unusual intermediate filament protein expression patterns have already been found in this species at later embryonic stages. Double-labelling indirect immunofluorescence assays with a panel of monoclonal intermediate filament antibodies are performed on frozen sections and compared with aldehyde-fixed plastic-embedded tissues. The histological part of the study, serving as a basis for the topographical orientation in the immunostained frozen sections, emphasises many similarities between the primitive streak embryos of the rabbit and the chick. The immunohistochemical analysis reveals cytokeratin expression to varying degrees in all germ layers. Vimentin expression, always in combination with cytokeratin expression, is found in a few cells of the ectoderm, endoderm and lateral mesoderm, but not in the primary mesenchymal cells of either the primitive node or the primitive streak. The results are discussed in relation to recent experimental findings on differentiation and morphogenetic processes in the primitive streak embryo. While these complex expression patterns make it seem unlikely that intermediate filament protein subtypes are expressed independently of cellular function during development, no indication can be found for a relation between vimentin expression and the morphogenetic changes thought to be important during mesoderm formation.Supported by the Deutsche Forschungsgemeinschaft (Wa 359-9) and by the Netherlands Cancer Foundation Offprint requests to: C. During a long-term study on reproduction of the European rabbit (Oryctolagus cuniculus L.) that was carried out on a population living under semi-natural conditions, occasionally single pups per litter were discovered being stuck in remnants of the foetal membranes. Here we describe this so far unknown phenomenon, quantify the occurrence, and investigate the consequences for the individual growth rate and for survival. Furthermore, we tested if maternal social rank and age were correlated with the occurrence of this phenomenon. Pups comprising the described adherence of the foetal membranes had lower growth rates than their unaffected litter siblings, and usually died during the nestling period or short after emergence above ground. Our findings point out that there was an individual predisposition of the mother for having affected litters. However, an impact of maternal social rank or age on the occurrence of this feature in their offspring was not supported by our data. Nevertheless, a pathological background also has to be considered as a possible cause.  相似文献

The relative importance of predation risk and food quality on spatial grazing pressure and activity patterns was tested in a central-place foraging herbivore: the European rabbit. Rabbits grazed less with increasing distance from their burrows into adjacent grassland, thereby creating a gradient of increasing vegetation height and plant biomass and decreasing plant nutrient concentration. When nitrogen concentration was experimentally increased by 150% through fertilizing and mowing, rabbits visited these plots four times more frequently than the untreated control plots. Addition of predator scent (mink pellets) did not result in different patch use by rabbits. The combined addition of fertilizer and mink pellets had the same effect as addition of fertilizer alone. However, the mink pellets changed the temporal activity pattern of rabbits as measured with infrared detectors. Rabbits were predominantly nocturnal but shifted their activities to the day when mink pellets were added, resulting in equal activities during night and day. We conclude that rabbits are sensitive to perceived predation risk, but that this does not influence their spatial grazing pressure. A selection for the highest food quality on the other hand can explain the observed natural rabbit grazing gradient. Food quality was highest close to the burrows, therefore rabbits selecting for high quality food should forage most intensely close to the burrows and only move further away for higher quality items or when the vegetation close to their burrows is depleted. Through intensive grazing close to the burrows rabbits facilitated for themselves either through stimulating fresh protein rich re-growth or the return of nutrients through faeces or both. This is in contrast with central-place foraging theory where intense feeding close to the burrow is assumed to lead to reduced food resources.  相似文献

Despite their pest status in numerous areas throughout the World, the populations of European rabbits (Oryctolagus cuniculus) have strongly decreased in South Western Europe since the mid-20th century. Such a decrease constitutes a major threat on top predators and calls for a better understanding of its mechanisms to provide suitable management responses. Infectious diseases have been invoked as the main responsible factors, but they cannot by themselves explain the magnitude of this decrease. Habitat fragmentation may indeed act as a synergetic factor, and habitat use studies are needed to better understand the impact of fragmentation on rabbit population dynamics. We investigated the variability of home range size with respects to age, sex and season in three wild populations of rabbits using telemetry. Home ranges were smaller in the highest density populations (7333 and 6878 vs. 20,492 m2) suggesting differences in habitat quality between the populations. In addition, home range sizes were larger during the reproductive season for both sexes, and adults tended to have smaller home ranges than juveniles. Clearly, the home range sizes reported here were smaller than those previously reported in rabbits.  相似文献

In altricial mammals and birds, the presence of a large number of litter or brood mates often affects the development of individual offspring by reducing the share of resources provided by the parents. However, sibling presence can also be favourable, conferring thermoregulatory benefits when ambient temperatures are low. Consequently, shifts in the relation between costs and benefits of sibling presence can be expected as a function of the thermal environment. In a study of a European rabbit population (Oryctolagus cuniculus) living in a field enclosure, we investigated the effects of litter size and soil temperature on pup growth over 7 years. Temperatures inside the subterranean nests were positively correlated with soil temperature and with litter size. Soil temperature varied strongly across the breeding season, ranging from 3 to 21°C. Under warmer soil temperature conditions (10–15°C and >15°C), pup growth decreased with increasing litter size, where litters of two pups (smallest litter size considered) showed the highest growth rates. In contrast, under colder soil temperature conditions (<10°C), the highest growth rates were found in litters of three pups. We also asked if such temperature-dependent differences in the optimal pup growth rates might be explained by differences in maternal characteristics, which might affect lactational performance. We assessed maternal performance using females’ postpartum body mass and social rank. However, we did not find consistent differences in maternal characteristics between females giving birth to different-sized litters during different soil temperature conditions, which would have provided an alternative explanation for the observed differences in litter size-dependent pup growth. We conclude that under colder soil temperature conditions, the thermal benefits of a greater number of littermates outweigh the negative consequences of competition for milk, leading to an environment-dependent shift in the optimal litter size for individual growth in this species.  相似文献

Herbivores living in seasonal environments commonly have to deal with reduced food availability and quality during the winter season. The present study investigated how European rabbits Oryctolagus cuniculus living in a grassland habitat adapt their feeding behaviour to cope with this situation. Both adult and subadult animals markedly increased their feeding rates throughout the course of the winter, with the rates in subadults being generally higher. Nevertheless, both age classes showed a significant decline in body mass throughout the winter averaging 22.3% of the autumn mass in subadult and 9.7% in adult animals. In late winter, an increase in the faecal sand content was found, indicating that the animals were feeding on ground-level plant parts. Measurements of nitrogen content of (1) grass leaves and stems and (2) ground-level sprouts and roots demonstrated a decrease in the food quality of grass leaves and stems from early to late winter. In contrast, the nitrogen content of roots and ground-level sprouts remained stable. It can be speculated that the animals increasingly switched to ground-level plant parts as alternative food in the course of the winter in order to satisfy their needs for a sufficient source of nitrogen.  相似文献

Summary Fertilization and early embryo and endosperm development were examined in Phaseolus vulgaris x P. acutifolius, P. vulgaris x P. lunatus crosses and their reciprocals. The number and length of pollen tubes were not different between selfings and interspecific crosses. Fertilization was completed in all matings and the time of fertilization was maternally dependent which may reflect the degree of maturation of embryo sacs at pollination. A large difference between reciprocal crosses was found in the time of endosperm and embryo division in relation to the time of fertilization. When P. vulgaris was the female parent and P. acutifolius the male parent, endosperm division occurred at the same time as in P. vulgaris upon selfing, while in P. vulgaris x P. lunatus crosses the time of endosperm division was intermediate as compared with the two parents. The time lapse between fertilization and endosperm and embryo division in P. acutifolius x P. vulgaris crosses was longer than in either parent upon selfing. In P. lunatus x P. vulgaris crosses, endosperm division occurred in only 7–12% of the ovules at 72 hours after pollination. Embryo development in these ovules was limited to the four cell stage although the endosperm was at the free nuclei stage. Summary The fate of the germinal vesicle-derived protein, nucleoplasmin, was followed in embryos and tadpoles of Xenopus using monoclonal antibodies and indirect immunofluorescent staining. Nucleoplasmin was found in all nuclei up to feeding tadpole stages. Thereafter its level decreased in all nuclei. It was not detected in nuclei of advanced tadpoles or of adults. Contrasting with another protein, N1, that was previously monitored in the nuclei of dividing gonia of both sexes, nucleoplasmin was only detected in the nuclei of ovarian oocytes starting at diplotene. Traces of nucleoplasmin have also been found in a rapidly-dividing fibroblastic cell-line by immunohistology and protein blotting.  相似文献

Immunohistochemical staining for S-100 protein and the intermediate filaments keratin and vimentin, was made in 41 salivary adenomas. In pleomorphic adenomas, great heterogeneity in the staining, as well as multiple and co-expressions of these proteins were found in the outer tumor cells of tubulo-ductal structures and modified myoepithelial cells, but not in the luminal tumor cells. All the outer tumor cells stained for S-100 protein, 97% for K8.12 keratin and 85% for vimentin. Of these cells, 29% showed multiple expression of K8.12 keratin, vimentin, and S-100 protein, and 17% showed co-expression of K8.12 and S-100 protein. Modified and neoplastic myoepithelial cells showed similar expressions of these proteins to those of outer tumor cells; myoepithelioma cells displayed the most complicated pattern, being positive for KL1, PKK1, and K8.12 keratins, vimentin and S-100 protein. In luminal tumor cells there was a heterogeneous expression of KL1 and PKK1 in 82%, and of KL1, PKK1, and K8.12 in only 14.7%. Based on the immunohistochemical findings obtained with different monoclonal antibodies in pleomorphic salivary adenomas, outer tumor cells may be derived from ductal basal cells and luminal tumor cells from intercalated duct cells.  相似文献

Summary The occurrence of cytokeratins, vimentin, and desmin in the dome epithelia and adjacent non-dome epithelia in four locations of gut-associated lymphoid tissues (GALT) of adult and newborn rabbits (Peyer's patches, sacculus rotundus, caecal lymphoid patches and appendix) was studied with monoclonal antibodies, using the indirect immunoperoxidase technique. In all locations investigated in adult animals, antibodies specific for vimentin labelled (1) M-cells, which engulf intraepithelial lymphocytes, (2) columnar epithelial cells at the base of the domes lacking an apparent contact with lymphocytes (immature M-cells), and (3) flat cells, which lie in the lamina propria under the dome epithelium, and which line the basal lamina with thin cytoplasmic processes. In newborn rabbits, columnar epithelial cells resembling the immature M-cells of adults were selectively stained with vimentin antibodies. In M-cells, the strongest immunoreactivity was present in the perinuclear region and close to the pocket membrane, whereas the most apical and most basal parts of the cytoplasm showed no vimentin-immunoreactivity. Enterocytes in the dome epithelium and in the non-dome epithelium were vimentin-negative. M-cells and enterocytes bound antibodies against cytokeratin peptides 18 and 19 in adults and newborn animals. Compared with enterocytes, M-cells showed less intense staining for cytokeratins. Dome epithelia and no-dome epithelia did not contain desmin-immunoreactive cells. The results suggest that vimentin is a sensitive marker for M-cells in rabbit GALT.  相似文献

Knowledge about the factors determining habitat use is especially interesting for herbivores living under seasonal climates as they have to deal with food shortage during the drought season. In this context, different-aged individuals are expected to respond differently to seasonal variations because nutritional requirements and predation risk can vary with age. We investigated adult and juvenile European rabbit (Oryctolagus cuniculus) habitat use in a Mediterranean ecosystem of central Spain, during spring, summer and winter. Relationships between adult and juvenile rabbit pellet abundances and 11 environmental variables related to food availability and refuge density were analysed by means of multiple regression, and evaluated using information theory to identify the set of models best supported by the data. Density of warren entrances was the more constant predictor of habitat use for juvenile rabbits in all the seasons. Herbaceous vegetation volume had a negative influence and was the strongest predictor for adult rabbit habitat use in spring and winter. In summer, green vegetation cover became the strongest positive habitat use predictor. These results suggest that adults prefer to forage in low volume swards ensuring a wide sensory range for the detection of approaching predators. However, the arrival of summer and its associated food depletion forces them to shift toward more open productive areas where green vegetation persists, but at the expense of higher predation risk. Seasonal variation induces minor changes in juvenile habitat use due to their strong dependence on warrens. Thus, our results show that rabbit habitat use is influenced by animal age and seasonal variations in resources.  相似文献

We surveyed the genetic diversity of the expressed major histocompatibility complex class II DQA locus in natural populations of European brown hares, Lepus europaeus, from Austria and Belgium (267 individuals in total). Based on cDNA sequences, we designed hare-specific primers to amplify the highly variable second exon of the DQA gene. Using cloning–sequencing methodology and capillary electrophoresis single-strand conformation polymorphism, we found ten alleles of the DQA exon 2 locus across these two European regions, of which eight are described for the first time. To search for signals of selection and recombination in the evolution of the DQA gene within the leporids, we augmented our sample with orthologous DQA alleles from the European rabbit, Oryctolagus cuniculus, in order to carry out a species level, species pairwise comparison. We found evidence of recombination in the history of the DQA sequences in leporids with some recombinant alleles bridging the species divide. In both species, selection on peptide binding site codons can be detected, though stronger for the rabbit. This result suggests that there may be a differential selection pressure in the deeper evolutionary history of these two species due to differences in several demographic and ecological traits likely subjecting them to differential selection by parasites. Finally, evolutionary relationships show a widespread and statistically significant intermingling of alleles from the two species. The many macroparasites shared between hares and rabbits may explain this pattern of trans-species polymorphism. The present study consisted of two experiments that evaluated experimental infections of Haemaphysalis leporispalustris ticks by a Brazilian strain of Rickettsia rickettsii, and their effect on tick biology. In experiment I, ticks were exposed to R. rickettsii during the larval, nymphal or adult stages by feeding on rabbits (Oryctolagus cuniculus) needle-inoculated with R. rickettsii, and thereafter reared on uninfected rabbits for the entire next tick generation. Regardless of the tick stage that acquired the infection, all subsequent tick stages were shown to be infected by PCR (infection rates varying from 1.3 to 41.7%), and were able to transmit R. rickettsii to uninfected rabbits, as demonstrated by rabbit seroconversion, guinea pig inoculation with rabbit blood, and PCR on rabbit blood. In Experiment II, ticks were exposed to R. rickettsii during the larval stage by feeding on rabbits co-infested with R. rickettsii-infected adult ticks, and thereafter reared on uninfected rabbits until the next generation of larvae. Again, all subsequent tick stages were shown to be infected by PCR (infection rates varying from 3.0 to 40.0%), and were able to transmit R. rickettsii to uninfected rabbits. Thus, it was demonstrated that larvae, nymphs, and adults of H. leporispalustris were able to acquire and maintain the R. rickettsii infection by transstadial and transovarial transmissions within the tick population, with active transmission of the bacterium to susceptible rabbits by all parasitic stages. Analyses of biological parameters of uninfected and R. rickettsii-infected tick lineages were performed in order to evaluate possible deleterious effects of R. rickettsii to the infected tick lineages. Surprisingly, all but one of the four R. rickettsii-experimental groups of the present study showed overall better biological performance than their sibling uninfected control ticks. Results of the present study showed that H. leporispalustris could support infection by a high virulent strain of R. rickettsii for at least two generations, in which infected tick lineages tended to have better performance than uninfected ticks. Our results support a possible role of H. leporispalustris in the enzootic maintenance of R. rickettsii in Latin America, as previously suggested by earlier works.  相似文献

The distribution of the intermediate filament (IF) proteins desmin, keratin, and vimentin was studied immunohistochemically in bovine ovaries. Special attention was paid to granulosa cells to examine possible marked changes of IF distribution in relation to folliculogenesis during ovarian development. Therefore, ovaries were used from fetuses from 3 months of gestation onward, calves, heifers, and cows. In all ovaries, desmin immunoreactivity was restricted to smooth muscle cells in blood vessel walls. Keratin appeared a characteristic of the ovarian surface epithelium. Co-localization of keratin and vimentin was observed in the epithelium of rete ovarii tubules in fetuses and calves, and in cortical cord epithelium and pregranulosa cells of primordial follicles in fetuses at 3–7 months of gestation. Vimentin was demonstrated in endothelium and in fibroblasts. In addition, vimentin immunoreactivity was present in granulosa cells of primary, secondary, and antral follicles. In antral follicles, these granulosa cells mainly had an elongated appearance and either contained an oblong or a round nucleus. Those with an oblong nucleus were characteristic for atretic antral follicles. In nonatretic follicles, numerous vimentin immunore-active, elongated granulosa cells with a round nucleus were observed, especially in the peripheral granulosa layer and in small (<3 mm in diameter) antral follicles. Additionally, in antral follicles, protrusions of vimentin-positive corona radiata cells were observed, that penetrated the zona pellucida to contact the oocyte. The data show that the distribution of vimentin containing IFs is associated with various aspects of granulosa cell activity, as mitosis, atresia, and intercellular transport. © 1995 Wiley-Liss, Inc.  相似文献
