Germination responses of species from the native plant communities of southwestern Western Australia can be related to syndromes of life history, fire response, and seed storage, and also to factors related to environmental stress. The Mediterranean-type climate of the region with periodic drought and recurrent fires affects the production of viable seeds in plants of limited stature and rooting depth. Fire response ephemerals and species cued to flower by fire tend to produce viable, readily germinable seeds, but there are instances where seed production is aborted in these predominantly herbaceous life forms. Clonal, rhizomatous species often produce mainly inviable seeds. Production of viable seeds in woody species of these highly diverse communities may also be restricted by limitations to cross pollination. Obligate post-fire seeding species tend to produce a greater proportion of viable seeds than species which are capable of resprouting following fire. Serotinous species, whether post-fire re-seeders or post-fire resprouting species, produce mainly viable seeds, which germinate readily once freed from protective fruits. Species of the legume families and a few others of the soil seed bank produce innately dormant seeds which can be germinated following heat shock treatments which simulate the effects of fire. Heat shock in these species appears mainly as a mechanism to crack the hard seed coats, but the effect of heat to denature seed coat inhibitors has not been eliminated. Western Australian species do not seem to break dormancy when exposed to leachates from burned wood as has been observed in comparable habitats in California and South Africa, but further research is advised. Germination in many native southwestern Australian species is cued by temperatures that correspond to the winter rainfall period. There are also indications that an after-ripening period of warm, dry storage increases percentage of germinable seeds. Stimulation of germination by hormones is almost unresearched in Western Australia, but germination percentages have been increased in a small number of species of horticultural potential. Stimulation of germination by soil nutrient concentrations is almost unresearched in Western Australia, except for the inhibitory effect of excess sodium chloride levels inEucalyptus andMelaleuca. These species only germinate when osmotic effects are reduced to lower levels as would occur when winter rains dilute soil salts. Application of research on seed germination has already enhanced the establishment of seedlings in the restoration of mine sites and is becoming important in aspects of the breeding and selection of native plants for the cut flower, bedding plant and essential oil industries.

Facing the current biodiversity crisis, the value of ex situ conservation has been increasingly acknowledged in international treaties and legislations. Seed banks are a good way of conserving biodiversity, providing that seeds are of high quality and at maximum viability. However, despite the number of established ex situ facilities, there is little information on seed viability in botanic garden seed banks. This paper analyses the status of the seed collection of the National Botanic Garden of Belgium by determining the germination capacity and viability of seeds that have been stored for 1–26 years. It aims at: (1) ensuring that existing storage conditions provide effective ex situ conservation of threatened species; (2) providing viability data on threatened species; (3) planning future collection and storage efforts for seeds of West European species of conservation value. Results from this study showed that the germination and viability percentages of the 250 tested species reached on average 59 and 79% respectively. Some families typically performed better than others. Within a species, consistent results were not always obtained. Over a quarter of accessions exhibited some degree of dormancy. Considering the current lack of knowledge in seed germination and dormancy of many rare and threatened species, we believe that the quality of a seed collection should be estimated by its viability and not by its germination percentage. This study calls for further research in order to better understand the biology of a range of threatened native species.

Seed viability and germination are key factors in the success of restoration efforts, especially when stored seeds are used. However, the effect of seed storage on germination of most of the native Arabian species is not well documented. We investigated the effect of storage time and role of the seed mucilage in regulating germination, dormancy, salinity tolerance and consequential survival strategy of F. aegyptia in an unpredictable arid desert setting. Effect of light and temperature during germination was studied under two photoperiods and two thermoperiods using intact and de-mucilaged seeds. Presence of mucilage and thermoperiod did not affect the germination. However, seed collection year and photoperiod had a highly significant effect on the germination. Increasing salinity levels decreased the germination of F. aegyptia but ungerminated seeds were able to germinate when salinity stress was alleviated. Seed storage at room temperature enhances the germination percentage, indicating that F. aegyptia seeds have physiological dormancy and it can be alleviated by after-ripening at dry storage. In addition, F. aegyptia seeds show ability to germinate at lower salinity concentration and remain viable even at higher saline conditions, indicating their adaptability to cope with such harsh environmental conditions.

Seed banking following internationally agreed standards is an important way for preserving collections of wild plant species ex situ; but this method is not suitable for desiccation sensitive species. Lauraceae comprehends some of the dominant species in the evergreen broadleaved forest in the south of China and contains many species both of ecological and economical importance. However, study on seed biology such as germination and desiccation tolerance of this family is scarce. Seeds of 9 species from 5 genera of this family were collected and their dormancy status and germination requirement were studied; also their desiccation tolerance were determined using a modified 100 seed test. The results showed that seeds of Cinnamomum camphora probably have intermediate physiological dormancy; seeds of Actinodaphne forrestii, Actinodaphne obovata, Cinnamomum migao, Lindera metcalfiana var. dictyophylla, Lindera communis and Neolitsea polycarpa are non deep physiological dormant; Seeds of Cinnamomum burmannii and Phoebe glaucophylla may have no or negligible dormancy. All 9 species lost seed viability after desiccated to 286%-716% moisture content while still retained considerable viability with moisture content ranged from 1732% to 4487% after moist storage; thus seeds of the 9 species are all desiccation sensitive and can not be stored at the conventional seed bank conditions.

Seed predators that severely affect seed germination rates are well known for many plant species. Here, we hypothesised that due to differences in resource allocation within fruits, seed predation can negatively affect non-predated seeds in infested fruits when predation occurs during fruit maturation (a ‘top-down’ effect). We addressed this question using a system of bruchid beetles on Mimosa trees and we also investigated whether seed quality (nitrogen concentration) affects beetle body mass, which would have implications for adult fitness (‘bottom-up’ effect). To assess spatial variation, bottom-up and top-down effects were investigated in two plant populations. Nitrogen concentration was significantly higher in seeds from non-infested fruits than from infested fruits. This supports the hypothesis that resource allocation may differ between seeds from infested and non-infested fruits. Germination experiments showed that seeds from non-infested fruits germinated better than non-predated seeds from infested fruits. It was also confirmed that seed quality affected bruchid body mass. There was also evidence that more resources were taken from well-developed seeds. These results showed that seed predation can damage non-predated seeds.

Effective seed storage after sourcing (harvesting or purchasing) is critical to restoration practitioners and native seed producers, as it is key to maintaining seed viability. Inadequate seed storage can lead to a waste of both natural and economic resources when seeds of poor quality are sown. When working with native species with unknown storage behavior, general assumptions can be made based on studies on related species, and standard practices may be applied with caution; however, an investigation should be conducted to understand if specific storage requirements are needed and for how long seeds can be stored before they lose significant viability. In this paper of the Special Issue Standards for Native Seeds in Ecological Restoration, we provide an overview of the key concepts in seed storage and the steps to take for effective storage of native seeds for restoration use.

Seed quality and seedling establishment are the most important factors affecting successful crop development. They depend on the genetic background and are acquired during seed maturation and therefor, affected by the maternal environment under which the seeds develop. There is little knowledge about the genetic and environmental factors that affect seed quality and seedling establishment. The aim of this study is to identify the loci and possible molecular mechanisms involved in acquisition of seed quality and how these are controlled by adverse maternal conditions. For this, we used a tomato recombinant inbred line (RIL) population consisting of 100 lines which were grown under two different nutritional environmental conditions, high phosphate and low nitrate. Most of the seed germination traits such as maximum germination percentage (Gmax), germination rate (t50) and uniformity (U8416) showed ample variation between genotypes and under different germination conditions. This phenotypic variation leads to identification of quantitative trait loci (QTLs) which were dependent on genetic factors, but also on the interaction with the maternal environment (QTL × E). Further studies of these QTLs may ultimately help to predict the effect of different maternal environmental conditions on seed quality and seedling establishment which will be very useful to improve the production of high-performance seeds.

Ex situ conservation in seed banks is a potential complementary conservation strategy for native plant species. It is well established that ex situ seed banking of native wild plants prolongs seed viability and thereby preserves genetic and species diversity for future use.We evaluated ex situ storage potential of eight halophytic species from deserts in the United Arab Emirates(UAE)by studying seed germination.Specifically,we examined the germinability of freshly collected seeds and seeds stored for three years in a seed bank.We also examined the effect of light conditions on fresh and stored seed germination.Fresh seeds of seven of the eight species tested had a higher germination rates under 12/12 h light/dark fluctuations than did those exposed to total darkness.Storage reduced light sensitivity in Halocnemum strobilaceum,Suaeda aegyptiaca,Salsola drummondii and Salsola imbricata, but increased the requirement for light in Arthrocnemum macrostachyum. In Anabasis setifera, storage decreased germination percentage when there was a 12-hour light/dark fluctuation, but increased germination rate when exposed to the dark treatment.Storage significantly reduced germination in both the light/dark and dark treatments in Suaeda vermiculata and S.aegyptiaca.Germination speed also responded differently to storage;whereas Timson's index significantly increased in A.macrostachyum and H.strobilaceum,it significantly decreased for S.drummondii,S.aegyptiaca and S.vermiculata.Germination of these species at a range of temperatures requires further testing;additionally,we strongly suggest that these laboratory findings be complemented by field studies.

Haloxylon salicornicum and Salsola vermiculata (Amaranthaceae) are two perennial shrubs producing fruits with differently coloured perianths (yellow vs pink) on separate individuals. To test the impact of different storage periods (four, eight and twelve months) and temperature conditions (freezing, room temperature and natural field temperatures) on the seed germination of the two seed categories, germination tests were carried out on both species. For both species, collected from wild plants in the Arabian desert (Qatar), seed germination was tested under two photoperiods (light 12/12; dark 24 hours) at 20/30°C for winged (intact) and de‐winged seeds. For each species, all the considered factors (perianth colour, wing presence, photoperiod, storage period and storage conditions) had significant effects on seed germination (p < 0.001), while the interactions among factors varied in their level of significance depending on the species. In both species, yellow seeds germinated considerably better than the pink ones, the presence of wings slowed the germination, and both storage period and temperature conditions highly influenced their germination ability. Our results indicate that seeds from different perianth colours have different germination strategies to delay their germination and distribute the species through time by escaping germination time by imposing some kind of after‐ripening or seed dormancy. The variation in storage time and storage conditions might help in extending their germination period, where one set of seeds germinate immediately, and the other set of seeds becomes dormant and thus contributes to the formation of a soil seed bank for the long‐term recruitment of seedlings. This study highlights an ecological advantageous strategy for these two species growing in the unpredictable desert habitat.

Germination of freshly collected seeds of three sympatric herbaceous species native to fire‐prone environments in south‐western Australia was significantly improved through the application of novel combinations of dry heat, gibberellic acid, smoke water and dry afterripening. For fresh seeds, combinations of dry heat, gibberellic acid and/or smoke water resulted in >80% germination in Austrostipa elegantissima (Poaceae) and Stylidium affine (Stylidaceae) seeds and >60% germination in Conostylis candicans (Haemodoraceae) seeds, compared with <10% germination of control seeds. For fresh seeds, two broad germination patterns were observed in response to smoke water: nil – low germination for both control and smoke water‐treated seeds (A. elegantissima and S. affine); and a significant smoke response (35%) compared with control seeds (1%) (C. candicans). During afterripening, high germination for A. elegantissima seeds was achieved following 3 months storage of seeds at equilibrium relative humidities of 23–75%, but seeds stored at 5–13% equilibrium relative humidities took 6–36 months to achieve similar levels of germination. Germination of C. candicans seeds also increased after 3 months storage, to >60% at each equilibrium relative humidity and further increases over time were slight. For S. affine seeds >60% germination was achieved only after 36 months storage at 50% equilibrium relative humidity. Seeds from all three species were smoke‐responsive at some point, but the interaction/effects of afterripening on the smoke response varied significantly between species. This study highlights an apparent effect of seed dormancy status on response to smoke and a surprisingly high level of ecological variation in pre‐germination requirements (cues) for these co‐occurring species that may relate to variation(s) in microsite selection forces operating on the soil seed bank of the different species.

The main storage compound in lupin seeds is protein, whose content can reach up to 45–50 % of dry matter. However, seeds of some lupin species can also contain quite a large amount of storage lipid. The range of lipid content in lupin seeds is from about 6 to about 20 % of dry matter. Storage lipid in developing seeds is synthesized mainly from sugars delivered by mother plants. During seed germination, one of the main end-products of storage lipid breakdown is also sugars. Thus, the sugar level in tissues is considered an important regulatory agent, during both lipid accumulation and lipid breakdown. Generally, in developing legume seeds, there is a strong negative relation between accumulation of storage protein and storage lipid. Results obtained in developing lupin cotyledons cultured in vitro pointed to the possibility of a positive relation between protein and lipid accumulation. Such a positive effect could be caused by nitrate. During lupin seed germination and seedling development, the utilization of storage lipid is enhanced under sugar deficiency conditions in tissues and is controlled at the gene expression level. However, under sugar starvation conditions, autophagy is significantly enhanced, and it can cause disturbances in storage lipid breakdown. The hypothesis of pexophagy, i.e., autophagic degradation of peroxisomes under sugar starvation conditions during lupin seed germination, has been taken into consideration. The flow of lipid-derived carbon skeletons to amino acids was discovered in germinating lupin seeds, and this process is clearly more intense in sucrose-fed embryo axes. At least four alternative or mutually complementary pathways of carbon flow from storage lipid to amino acids in germinating lupin seeds are postulated. The different strategies of storage compound breakdown during lupin seed germination are also discussed.

Successful restoration of sedge meadow wetlands is limited by lack of information regarding reintroduction of sedge (Carex) propagules. While restoration from seed is common for prairie restorations, little is known about the germination characteristics of many wetland plants, including sedges. We present the results of a 2.5-year study on seed germination and viability for five species of Carex common to sedge meadow and prairie pothole wetlands in temperate North America. Seed storage and germination conditions were investigated to determine the optimum combination for maintaining seed viability and stimulating germination rates over time. Seeds were germinated under seven different temperature and three moisture regimes after storage for 4, 10, and 14 months under one of four different storage regimes (dry-warm, dry-cold, moist-cold, and wet-cold). The efficacy of short-term wet-cold stratification to stimulate germination of 2.5-year-old seed after long-term dry storage was also investigated. Carex stricta, Carex comosa, and Carex lacustris showed the greatest germination response after wet-cold or moist-cold storage, while Carex lasiocarpa and Carex rostrata showed similar rates of germination after either wet-cold or dry-warm storage. Wet-cold long-term storage was associated with a high level of viability in all five species after 2.5 years. Viability and germination rates were reduced in Carex stricta, Carex comosa, and Carex lasiocarpa after long-term dry-cold storage. Germination rates of seeds stored dry for 2.5 years are not improved by short-term wet-cold treatment in any species tested. Carex seeds should be stored under wet-cold conditions to maintain seed viability over time, thus increasing the likelihood of seeding success for sedge meadow restoration.

Prunus necrotic ringspot rvirus (PNRSV) was able to invade the immature apricot seed including the embryo. The amount of virus was very high inside the embryo compared with that present in the cotyledons. PNRSV infection produced an oxidative stress in apricot seeds as indicated by the increase in lipid peroxidation, measured as thiobarbituric acid-reactive substances. This lipid peroxidation increase was parallelled with an imbalance in the seed antioxidant enzymes. A significant decrease in the ascorbate–GSH cycle enzymes as well as in peroxidase (POX) activity took place in infected seeds, suggesting a low capability to eliminate H2O2. No changes in superoxide dismutase (SOD) or catalase activity were observed. A significant decrease in polyphenoloxidase (PPO) activity was also observed. Native PAGE revealed the presence of three different SOD activity bands in apricot seeds: a Mn-containing SOD and two CuZn-containing SODs. Only an isozyme with catalase, glutathione reductase (GR) or PPO activity was detected in both healthy and infected apricot seeds. Regarding POX staining, three bands with POX activity were detected in native gels in both healthy and infected seeds. The gel results emphasise that the drop detected in POX, GR and PPO activities in PNRSV-infected apricot seeds by kinetic analyses was also evident from the results obtained by native PAGE. The oxidative stress and the imbalance in the antioxidant systems from PNRSV-infected apricot seeds resemble the hypersensitive response observed in some virus–host interactions. This defence mechanism would inactivate PNRSV during seed formation and/or the storage period or even during seed germination. Those results can explain the decrease in seed germination and the low transmission of PNRSV by seeds in apricot trees.

The global push to achieve ecosystem restoration targets has resulted in an increased demand for native seeds that current production systems are not able to fulfill. In many countries, seeds used in ecological restoration are often sourced from natural populations. Though providing seed that is reflective of the genetic diversity of a species, wild harvesting often cannot meet the demands for large‐scale restoration and may also result in depletion of native seed resources through over harvesting. To improve seed production and decrease seed costs, seed production systems have been established in several countries to generate native seeds based on agricultural or horticultural production methods or by managing natural populations. However, there is a need to expand these production systems which have a primary focus on herbaceous species to also include slower maturing shrub and tree seed. Here we propose that to reduce the threat of overharvest on the viability of natural populations, seed collection from natural populations should be replaced or supplemented by seed production systems. This overview of seed production systems demonstrates how to maximize production and minimize unintended selection bias so that native seed batches maintain genetic diversity and adaptability to underpin the success of ecological restoration programs.
