Allelopathic effects of fresh tissue and dry powder of a nonsexual and a sexual strain of the macroalga Ulva pertusa on the growth of the microalgae Heterosigma akashiwo and Alexandium tamarense were evaluated using long-term coexistence culture systems in which several concentrations of macroalga fresh tissue and dry powder were used. The effects of macroalga culture medium filtrate on the two HAB algae were also investigated. Moreover, isolation co-culture systems were built to confirm the existence of allelochemicals and preclude the growth inhibition by direct contact. Short-term algicidal effect assays of macroalgae on H. akashiwo were carried out to measure the rate of algal cell lysis. The results of the coexistence assays showed that the growth of H. akashiwo and A. tamarense was strongly inhibited by fresh tissue and by dry powder of both strains of U. pertusa. The allelochemicals were lethal to H. akashiwo at relatively higher concentrations. The macroalga culture medium filtrate exhibited no apparent growth inhibitory effect on the two HAB algae under initial or semicontinuous filtrate addition, which suggested that continuous release of small quantities of rapidly degradable allelochemicals from the fresh tissue of both strains of U. pertusa was essential to effectively inhibit the growth of H. akashiwo and A. tamarense.  相似文献

Global environmental changes, including ocean acidification, have been identified as a major threat to scleractinian corals. General predictions are that ocean acidification will be detrimental to reef growth and that 40 to more than 80 per cent of present-day reefs will decline during the next 50 years. Cold-water corals (CWCs) are thought to be strongly affected by changes in ocean acidification owing to their distribution in deep and/or cold waters, which naturally exhibit a CaCO(3) saturation state lower than in shallow/warm waters. Calcification was measured in three species of Mediterranean cold-water scleractinian corals (Lophelia pertusa, Madrepora oculata and Desmophyllum dianthus) on-board research vessels and soon after collection. Incubations were performed in ambient sea water. The species M. oculata was additionally incubated in sea water reduced or enriched in CO(2). At ambient conditions, calcification rates ranged between -0.01 and 0.23% d(-1). Calcification rates of M. oculata under variable partial pressure of CO(2) (pCO(2)) were the same for ambient and elevated pCO(2) (404 and 867 μatm) with 0.06 ± 0.06% d(-1), while calcification was 0.12 ± 0.06% d(-1) when pCO(2) was reduced to its pre-industrial level (285 μatm). This suggests that present-day CWC calcification in the Mediterranean Sea has already drastically declined (by 50%) as a consequence of anthropogenic-induced ocean acidification.  相似文献

The cosmopolitan presence of Ulva spp. is partly due to its great reproductive ability, but relatively little information is available for the radiation conditions triggering reproduction. In the present study, we investigated the effect of photon irradiance, photoperiod, and spectral qualities of light on growth and reproduction of Ulva pertusa.During 8-day culture of discs cut from marginal parts of the thallus of U. pertusa, the size of the thallus discs was greatest at 10 μmol m−2 s−1, while saturation of reproduction occurred at 30 μmol m−2 s−1. The minimum photon irradiance allowing growth and reproduction was 5 and 10 μmol m−2 s−1, respectively. Rapid increases in the size and subsequent initiation of sporulation were observed in samples transferred to saturating irradiance from 5 μmol m−2 s−1 or darkness for 9 days. When exposed to different photoperiods (8:16-, 12:12-, 16:8-h LD and continuous light regimes) combined with different photon irradiances (10 and 100 μmol m−2 s−1), U. pertusa thallus showed that the thallus size attained at the low irradiance was similar in daylengths longer than 12 h per day, while the surface area increased in parallel with increasing light duration at the high irradiance. The degree of sporulation at 10 μmol m−2 s−1 varied, ranging from no sporulation in 8:16-h LD to 80% in 16:8-h LD and continuous light. On the other hand, there was no remarkable difference in the degree of sporulation between the photoperiods except for slightly smaller percentage sporulation in 8:16-h LD at 100 μmol m−2 s−1.At 5 μmol m−2 s−1, the growth of U. pertusa was markedly lower in green than in blue or red light, but there was no sporulation in any spectral quality. The degree of sporulation at 20 μmol m−2 s−1 was almost twice as much in blue or red as in green light.The size of plants irradiated with 1.0 W m−2 of UV-B for 20-40 min increased by 18-21% relative to control, whereas higher UV irradiance caused inhibition of growth. There was a significantly lower incidence of sporulation in UV-B-irradiated plants with the degree of reduction being greater in those exposed to higher UV doses. The total biologically effective UV-B dose for 50% inhibition of sporulation was 0.085 Doseeff kJ m−2. The time required to achieve 50% inhibition would be longer than 13 h at depths below 1 m in Ahnin coastal waters. The vertical attenuation coefficient of PAR (λ=400-700 nm) and UV-B (λ=300-320 nm) in April 1998 at Ahnin on the eastern coast of Korea was 0.21 m−1 for KPAR and 0.54 m−1 for KUV-B. A large reduction of light quantity and rapid disappearance of blue wavelength occurred by shading from overlying thalli.Percentage inhibition of sporulation was only 14-18% at 150-200 μmol m−2 s−1 compared with 70% at 10 μmol m−2 s−1, when plants were incubated under different irradiances of PAR immediately after UV-B exposures.These different photoadaptive strategies for sporulation may in part account for the great ecological success of U. pertusa.  相似文献

Here we report the first study of clearance rate and respiration rate of a deep living bivalve, Acesta excavata (J.C. Fabricius, 1779) (Mollusca: Limidae). We found that A. excavata had extreme values both for clearance and respiration rates compared to other bivalves. It has the second largest clearance rate ever reported, 13.36 l h− 1 g− 1, and the second lowest value of respiration rate, 0.12 ml O2 h− 1 g− 1. The gill area of 7063 mm2 g− 1 is one of the largest found in bivalves so far. We suggest that these values indicate a physiological adaptation to the low and irregular food supply in the deep sea rather that a specific adaptation to depth.  相似文献

Offshore banks and seamounts sustain diverse megafaunal communities, including framework reefs formed by cold-water corals. Few studies have quantified environmental effects on the alpha or beta diversity of these communities. We adopted an interdisciplinary approach that used historical geophysical data to identify topographic highs on Hatton Bank, which were surveyed visually. The resulting photographic data were used to examine relationships between megafaunal communities and macrohabitat, the latter defined into six categories (mud, sand, cobbles, coral rubble, coral framework, rock). The survey stations revealed considerable small-scale variability in macrohabitat from exposed Late Palaeocene lava flows to quiescent muddy habitats and coral-built carbonate mounds. The first reported evidence for coral carbonate mound development in UK waters is presented, which was most pronounced near present-day or former sites of topographic change, suggesting that local current acceleration favoured coral framework growth and mound initiation. Alpha diversity varied significantly across macrohabitats, but not between rock and coral rubble, or between smaller grain sized categories of cobbles, sand and mud. Community composition differed between most macrohabitats, and variation in beta diversity across Hatton Bank was largely explained by fine-scale substratum. Certain megafauna were clearly associated with particular macrohabitats, with stylasterid corals notably associated with cobble and rock habitats and coral habitats characterized by a diverse community of suspension-feeders. The visual surveys also produced novel images of deep-water megafauna including a new photographic record of the gorgonian coral Paragorgia arborea, a species not previously reported from Rockall Plateau. Further interdisciplinary studies are needed to interpret beta diversity across these and other environmental gradients on Hatton Bank. It is clear that efforts are also needed to improve our understanding of the genetic connectivity and biogeography of vulnerable deep-water ecosystems and to develop predictive models of their occurrence that can help inform future conservation measures. Mechanisms governing the behavior of coral planulae are not well understood, particularly those manifesting themselves between the time when the larvae are released and when they settle. Larvae from the hermatypic coral Porites astreoides Lamarck were exposed to different levels of hydrostatic pressure—103.4, 206.9, 310.3, 413.8, and 517.1 kPa (including ambient pressure). Data were collected at stops of the above pressures for 15 min each, respectively. This was done in both an increasing sequence and a decreasing one. When exposed to increases in pressure from 103.4 kPa, larvae swam upward (negative barotaxis) in a spiraling motion. Upon exposure to decreasing pressure from 517.1 kPa, larvae moved downward (positive barotaxis), but the magnitude of the vertical movement was much less than in the case of increasing pressure. This suggests that these larvae are more sensitive to increased pressure than decreasing pressure. High variance was also observed in the responses of these larvae at both the intra- and inter-colony levels. Thus, this behavioral trait is variable within the population. The trait may be genetically based, and thus may be susceptible to alteration by natural selection, although this remains to be demonstrated. Cold-water corals, such as Lophelia pertusa, are key habitat-forming organisms found throughout the world''s oceans to 3000 m deep. The complex three-dimensional framework made by these vulnerable marine ecosystems support high biodiversity and commercially important species. Given their importance, a key question is how both the living and the dead framework will fare under projected climate change. Here, we demonstrate that over 12 months L. pertusa can physiologically acclimate to increased CO2, showing sustained net calcification. However, their new skeletal structure changes and exhibits decreased crystallographic and molecular-scale bonding organization. Although physiological acclimatization was evident, we also demonstrate that there is a negative correlation between increasing CO2 levels and breaking strength of exposed framework (approx. 20–30% weaker after 12 months), meaning the exposed bases of reefs will be less effective ‘load-bearers’, and will become more susceptible to bioerosion and mechanical damage by 2100.  相似文献

The ultrastructure of septa and septum-associated septal pore caps are important taxonomic markers in the Agaricomycotina (Basidiomycota, Fungi). The septal pore caps covering the typical basidiomycetous dolipore septum are divided into three main phenotypically recognized morphotypes: vesicular-tubular (including the vesicular, sacculate, tubular, ampulliform, and globular morphotypes), imperforate, and perforate. Until recently, the septal pore cap-type reflected the higher-order relationships within the Agaricomycotina. However, the new classification of Fungi resulted in many changes including revision of existing and addition of new orders. Therefore, the septal pore cap ultrastructure of more than 325 species as reported in literature was related to this new classification. In addition, the septal pore cap ultrastructures of Rickenella fibula and Cantharellus formosus were examined by transmission electron microscopy. Both fungi have dolipore septa associated with perforate septal pore caps. These results combined with data from the literature show that the septal pore cap-type within orders of the Agaricomycotina is generally monomorphic, except for the Cantharellales and Hymenochaetales.It appears from the fungal phylogeny combined with the septal pore cap ultrastructure that the vesicular-tubular and the imperforate type both may have arisen from endoplasmic reticulum. Torrubiella is a genus of arthropod-pathogenic fungi that primarily attacks spiders and scale insects. Based on the morphology of the perithecia, asci, and ascospores, it is classified in Clavicipitaceae s. lat. (Hypocreales), and is considered a close relative of Cordyceps s. 1., which was recently reclassified into three families (Clavicipitaceae s. str., Cordycipitaceae, Ophiocordycipitaceae) and four genera (Cordyceps s. str, Elaphocordyceps, Metacordyceps, and Ophiocordyceps). Torrubiella is distinguished morphologically from Cordyceps s. lat. mainly by the production of superficial perithecia and the absence of a well-developed stipitate stroma. To test and refine evolutionary hypotheses regarding the placement of Torrubiella and its relationship to Cordyceps s. lat., a multi-gene phylogeny was constructed by conducting ML and Bayesian analyses. The monophyly of Torrubiella was rejected by these analyses with species of the genus present in Clavicipitaceae, Cordycipitaceae, and Ophiocordycipitaceae, and often intermixed among species of Cordyceps s. lat. The morphological characters traditionally used to define the genus are, therefore, not phylogenetically informative, with the stipitate stromata being gained and/or lost several times among clavicipitaceous fungi. Two new genera (Conoideocrella, Orbiocrella) are proposed to accommodate two separate lineages of torrubielloid fungi in the Clavicipitaceae s. str. In addition, one species is reclassified in Cordyceps s. str. and three are reclassified in Ophiocordyceps. The phylogenetic importance of anamorphic genera, host affiliation, and stipitate stromata is discussed.  相似文献

In this study, we apply Fry's classification of environmental factors to demonstrate the limiting effects of oxygen and its interaction with temperature on the growth of juvenile P. lethostigma. We also evaluated the properties of two metabolic indices, marginal metabolic scope (MMS) and limiting oxygen concentration (LOC), as indicators of metabolic scope. We found that oxygen limitation has its greatest impact near the optimum temperature for growth of the species. At 29 °C a reduction from 6.00 mg/L to 4.00 mg/L caused a 50% reduction in growth rate while at 27 °C the reduction had no significant effect on growth rate. The results are particularly relevant because these temperatures and oxygen concentrations are commonly observed in nursery areas during summer months. At all temperatures fish from the lowest oxygen treatment (1.75 mg/L) had negative growth rates. Comparisons between daily oscillating oxygen treatments and constant treatments failed to demonstrate significant effects. At temperatures past the optimum, growth rates between the 6.00 mg/L and 4.00 mg/L treatments were not statistically different. LOC was significantly affected by temperature, oxygen, and their interaction. Estimates were positively correlated with oxygen treatment (R2 > 0.71) and negatively correlated with temperature at moderate and low oxygen concentrations (R2 > − 0.84). MMS was significantly affected by temperature and oxygen and was significantly correlated with oxygen treatment (R2 > − 0.91), but correlations with temperature were not as clear. In conclusion, oxygen and temperature interactions have significant effects on metabolic scope and growth rates of fish, well above the accepted hypoxia threshold of 2.00 mg/L and MMS has proved a useful estimator of the metabolic scope of the organism within an environment.  相似文献

Species of Lutjanus Bloch are highly valued fish in some fisheries of the world and some species are cultured in the Southeast Asia especially in South China. Wild larvae are still the major source of mariculture in South China because artificial breeding techniques for most Lutjanus species have not yet been available. The Nansha coral reefs (also called Spratly Archipelago) water area, which is located in the South China Sea, is the main habitat and spawn area for Lutjanus in China. Larval identification of Lutjanus is important for relative ecological studies and mariculture, but larvae of many closely related species, such as those of the genus Lutjanus, are different to be distinguished morphologically. In the present study, a PCR-based fingerprinting technique called amplified fragment length polymorphism AFLP was used in the characterization and identification of 11 Lutjanus species captured in Nansha coral reefs. Optimal AFLP patterns were obtained with primer combination of E+AGC/M+CAA selective nucleotides. There were in total 132 AFLP loci in all specimens, and AFLP markers of each species varied from 44 to 69, but only 7 markers were fixed in all specimens. Meanwhile, high levels of intraspecific homogeneity were observed. All 11 species of Lutjanus were successfully identified by the comparative analyses of AFLP patterns. Moreover, neighbour-joining and UPGMA analyses of AFLP data were compared with current morphological taxonomic systems.  相似文献

The respiratory responses to increasing temperature and progressive hypoxia were examined relative to temperature acclimation in the nonindigenous, brown mussel, Perna perna (Mytilidae) from the Gulf of Mexico. When oxygen uptake rate (V?O2) was recorded at near full air O2 saturation, rate-temperature curves for Texas specimens of P. perna were sigmoidal, V?O2 generally increasing with increasing temperature but becoming suppressed as temperatures approached 10 and 30 °C, corresponding closely to this species' incipient thermal limits. At each tested temperature, V?O2 did not differ among individuals acclimated to 15, 20, or 25 °C. Lack of thermal acclimation was also reflected in acclimatory Q10 values>1.0 (range=1.34-2.14) recorded across acclimation groups at test temperatures equivalent to acclimation temperature. Low acute respiratory Q10 values in all acclimation groups across 15-20 °C indicated a limited capacity for thermal regulation of V?O2 within this temperature range. The ability of P. perna to regulate O2 uptake with progressive hypoxia was temperature-dependent, increasing from poor O2 regulation at 10 °C to good regulation at 30 °C. The O2 regulatory ability of P. perna and other open-water mytilids in declining O2 concentrations does not greatly differ from that of estuarine heterodont bivalves, suggesting that it is not a major factor preventing open-water species, such as P. perna, from invading estuarine environments. However, P. perna's inability to regulate O2 uptake at temperatures>25 °C combined with its relatively low upper thermal limit of 30 °C will likely prevent it from establishing permanent estuarine populations on Gulf of Mexico shores.  相似文献

Metabolic responses of sand fiddler crab, Uca pugilator, populations in northwest Florida are greatly influenced by seasonal temperature fluctuations. Crabs acclimated at 20 °C and immediately transferred to either 14 or 26 °C produced an acute metabolic response with respective temperature quotient (Q10) values of 3.46 and 3.91. Crabs acclimated at 10 and 20 °C exhibited a Q10 of 2.62 indicating a partial compensation response. A brumation (reverse) response (Q10 value of 20.11) was observed for acclimated crabs between 5 and 10 °C. Brumation is advantageous during winter when food supplies are scarce and crabs must survive extensive periods of inactivity.  相似文献

‘Resilience’, the capacity of the coral symbiosis with dinoflagellate algal symbionts (‘zooxanthellae’) to recover after bleaching, is a little-studied but crucial aspect of coral responses to bleaching stressors. This study investigated the response of the zooxanthella population in the coral Porites cylindrica after bleaching either naturally on a shallow subtidal reef or experimentally in response to elevated temperature and darkness. Coral resilience was influenced by the nature and duration of the stressor. Corals strongly bleached by natural stressors were less resilient than those that had been partially bleached; and a similar recovery profile was obtained for corals experimentally bleached by exposure to elevated temperature, in which recovery was slower for corals thermally-stressed 96 h than for 72 h. The opposite trend was evident for corals exposed to darkness, indicating that the bleaching trigger had a strong impact on coral resilience. When P. cylindrica recently recovered from bleaching was subjected to a repetition of bleaching stressors, it did not display acclimation, i.e. experience-mediated acquisition of resistance to bleaching stressors. The zooxanthella populations in all corals tested throughout the experiments were typed by PCR-RFLP as clade C, indicating that coral responses were not accompanied by any substantial change in zooxanthella composition at the cladal level.  相似文献

In the light of the poor culturability of Acidobacteria and Verrucomicrobia species, group-specific real-time (qPCR) systems were developed based on the 16S rRNA gene sequences from culturable representatives of both groups. The number of DNA targets from three different groups, i.e. Holophagae (Acidobacteria group 8) and Luteolibacter/Prosthecobacter and unclassified Verrucomicrobiaceae subdivision 1, was determined in DNA extracts from different leek (Allium porrum) rhizosphere soil compartments and from bulk soil with the aim to determine the distribution of the three bacterial groups in the plant-soil ecosystem. The specificity of the designed primers was evaluated in three steps. First, in silico tests were performed which demonstrated that all designed primers 100% matched with database sequences of their respective groups, whereas lower matches with other non-target bacterial groups were found. Second, PCR amplification with the different primer sets was performed on genomic DNA extracts from target and from non-target bacteria. This test demonstrated specificity of the designed primers for the target groups, as single amplicons of expected sizes were found only for the target bacteria. Third, the qPCR systems were tested for specific amplifications from soil DNA extracts and 48 amplicons from each primer system were sequenced. All sequences were > 97% similar to database sequences of the respective target groups. Estimated cell numbers based on Holophagae-, Luteolibacter/Prosthecobacter- and unclassified Verrucomicrobiaceae subdivision 1-specific qPCRs from leek rhizosphere compartments and bulk soils demonstrated higher preference for one or both rhizosphere compartments above bulk soil for all three bacterial groups.  相似文献

Ambrosia beetles (Coleoptera: Scolytinae) are associated with strictly entomochoric and mutualistic fungi. We studied the mycobiota associated with Scolytodes unipunctatus, ambrosia beetles that infest Cecropia trees in Central America. Isolates were characterized using morphology and rDNA sequences (ITS region, LSU, and SSU rDNA). Four species are described here: Raffaelea scolytodis sp. nov. (Ophiostomatales), Gondwanamyces scolytodis sp. nov., Custingophora cecropiae sp. nov., and Graphium sp. (Microascales). The genus Custingophora is emended to include Knoxdaviesia anamorphs of Gondwanamyces based on uniformity of DNA sequences and phenotype.  相似文献
