Fifty-two isolates of Fusarium species were obtained from soybean seeds from various parts of Korea and identified as Fusarium oxysporum, F. moniliforme, F. semitectum, F. solani, F. graminearum, or F. lateritium. These isolates were grown on autoclaved wheat grains and examined for toxicity in a rat-feeding test. Nine cultures were toxic to rats. One of these, a culture of Fusarium sp. strain KCTC 16677, produced apicidin, an antiprotozoal agent that caused toxic effects in rats (including body weight loss; hemorrhage in the stomach, intestines, and bladder; and finally death) when rats were fed diets supplemented with 0.05 and 0.1% apicidin. The toxin was toxic to brine shrimp (the 50% lethal concentration was 40 μg/ml) and was weakly cytotoxic to human and mouse tumor cell lines.  相似文献

Each of 12 cultures ofFusarium, comprising four species, isolated from moldy soybeans suspected of being involved in illness of wild geese, were grown separately in autoclaved moist rice, in autoclaved moist soybeans, and in surface sterilized-disinfected soybeans, assayed for various mycotoxins, and fed to rats. Four additional cultures that produced known toxins on rice were also grown on soybeans as controls. All isolates, except one ofF moniliforme, grown in rice resulted in weight loss of rats, and that one resulted in weight gain; 12 of the isolates caused death. One isolate ofF poae grown in soybeans caused death when consumed by rats, but none of the other 15 resulted in weight loss or overt injury. Much larger amounts of zearalenone, deoxynivalenol (DON), T-2 toxin, neosolaniol, T-2 tetraol, wortmannin, and moniliformin were produced by the cultures on rice than on soybeans, but more HT-2 toxin was produced by one isolate ofF poae grown on soybeans than when grown on rice. Soybeans appear to be a poor substrate for elaboration of most of the toxins produced by the isolates tested.  相似文献

Surface sterilised stelar tissues from onion roots exhibiting typical pink root symptoms were plated on water agar and incubated under cool white fluorescent light (CWFL) for 12 h day-1 at 20°C. After 4–5 days, the plates were examined with the 10? objective of a compound microscope and tissues producing conidia of Fusarium spp. were discarded. Sub-cultures of isolates suspected to be Pyrenochaeta terrestris, grown from Fusarium-negative tissues, were transferred to chloroamphenicol-amended (500 ppm) corn-meal (CCMA) agar plates and incubated at 24°C under CWFL for a further 10–14 days. Cultures with setose pycnidia were identified as P. terrestris. Plates were again scanned for conidia of Fusarium spp., and cultures negative for Fusarium spp., but not producing pycnidia, were suspected to be P. terrestris. These were compared with known isolates on CCMA. On this medium growth of P. terrestris was slow, producing appressed, pinkish colonies which were circular with a smooth margin. All isolates of P. terrestris isolated by the procedure caused pink root of onion when tested, whereas none of the Fusarium spp. isolated produced the disease. However, a mixed inoculums of P. terrestris and Fusariurn solani produced typical pink root symptoms.  相似文献

Studies have shown that nutrient requirement of suckling kits is not satisfied, but they can be fed a double quantity of milk (double nursing) resulting in improved BW and weight gain. The aim of our trials was to give additional solid feed during the early suckling period (3 to 15 days of age) when rabbit kits drink exclusively milk. Two experiments were conducted with animals from Pannon Rabbit Breeding Program. In experiment 1 (n=77 does, 734 kits) the does received commercial feed (C) or C pellet supplemented with 0.2 g powdered thyme/kg (CT). Within both dietary groups of the does three groups of litters were formed: no additional solid creep feeding (N); soya bean-based pellet (S); S pellet with 1% added powdered thyme (ST). In group S and ST, cylinder-shaped solid pellets were made. At the beginning (3 days of age) two pieces of pellets were placed daily into the nestbox after nursing. Later on it was increased to six pellets till 15 days of age. The kits consumed the additional solid feed (S and ST), however, it did not affect the BW, weight gain or survival. In experiment 2 (n=30 does, 240 kits) all does consumed commercial feed. The additional feed for kits was based on commercial piglet feed. Three groups were formed: the litters in control group were fed no additional solid feed (K), kits were fed additionally with pellets (8 mm of diameter) based on piglet feed powder, pellet adhesive and water (PI), and extra glycerin powder was added to the mixture of piglet feed powder and water (PG). The experiment lasted from the age of 3 days till 21 days. At the beginning six pellets were placed on the nest material. Later on the amount was gradually increased to 24 pellets till age of 15 days. The kits consumed the pellets. The BW of PI group differed from group PG at age of 5, 9, 12 and 21 days by +7.3%, +6.5%, +5.9%, +4.8%, respectively (P<0.05) and from group K at age of 12 days by +5.9% (P< 0.05). The differences were more expressed at age of 16 and 19 days in favour of group PI (from K by +7.1%, +6.9% and from PG by +5.9%, +5 8%, respectively, P<0.01) and at 21 days of age (from K by +6.2%, P<0.01). To find appropriate composition of creep feed for kits further studies are needed.  相似文献

One culture ofF avenaceum, 4 cultures ofF oxysporum, and 11 cultures of Fsambucinum were isolated from soil samples of pasture in New Zealand in 1987. All cultures, when grown on rice media and fed to rats caused a weight loss in rats as well as toxic signs including hemorrhaging and congestion, uterine enlargement, and hematuria. 6 out of 16 cultures caused death in rat feeding tests.F oxysporum #1 killed rats (feeding test) within 5-12hrs. 10 cultures produced zearalenone (19 to 8,849 ppm), 8 cultures produced nivalenol (32 to 117 ppm), 1 culture,F sambucinum #8, produced wortmannin (40 ppm), and 5 cultures produced moniliformin (19 to 9,000ppm). We report for the first time the co-occurrence of zearalenone, nivalenol, and moniliformin produced byF sambucinum #3 in culture.F avenaceum #1 andF oxysporum cultures (nos 1, 2, and 3) produced moniliformin alone.F oxysporum #4 produced zearalenone alone as well.F sambucinum #5 caused erythema in the small intestine of rats and 100% mortality and did not produce any known toxin(s). Nivalenol when administered to the stomach of rats orally at levels 10, 20, and 40mg/kg body weight caused inflammation in the intestines, coma, and death. The mycotoxins T-2 toxin, HT-2 toxin, T-2 tetraol, diacetoxyscirpenol (DAS), monoacetoxyscirpenol (MAS), deoxynivalenol (DON), 3-acetyl-and 15-acetyldeoxynivalenol, depoxynivalenol, fusarenon-X, alpha-and beta-zearalenone, and fusarochromanone (TDP-1) were not detected in the extracts of these cultures.  相似文献

1,3-Propanediol (PAD) was fed to rats for 15 weeks, and its effects on hepatic and testicular DNA were studied. The control rats were fed a casein-based diet that contained 10% tocopherol-stripped corn oil with 30 IU of d,l-α-tocopherol acetate/kg; the experimental rats were fed the same diet with 500 ppm of PAD. Homogenates prepared from the livers of each group of rats converted 1,3-propanediol to malondialdehyde (MDA) with equal efficacy, but homogenates of testes did not catalyze this conversion. After 10–15 weeks of feeding the diets, the hepatic DNA of the rats fed PAD had less template activity, more bound tryptophan and more DNA-protein and interstrand DNA cross-links than that of the control rats. As measured by template activity and bound tryptophan, testicular DNA of the experimental rats was not different from that of the control rats; however, there was slightly more cross-linking in the testicular DNA of experimental rats than in that of control rats. Testes of the experimental rats contained more lipid-soluble fluorophores than did those of the control rats. The results are consistent with the conclusion that PAD was converted to MDA in vivo and that MDA is the reactive species that caused the observed biological damage.  相似文献

The effect of melengestrol acetate (MGA) on the organ weight and the mammary lobulo-alveolar development in rats was studied. 33 adult female rats were divided into 4 groups: 1) 5 mcg MGA/gm feed; 2) a normal diet; 3) ovariectomized and fed 5 mcg MGA/gm feed; and 4) ovairectomized and fed a normal diet for 30 days when the rats were sacrificed. In the second experiment, 21 primiparous female rats were ovariectomized, and 10 days later 1 group was injected with 2 mcg estradiol for 10 days, while the 2nd group was injected with 50 mcg MGA/day, and the 3rd group with estradiol plus MGA in the above doses. The animals were sacrificed after 10 days of treatment. MGA decreased anterior pituitary, ovary, uterus, and adrenal weight, but enhanced (p less than .01) mammary lobulo-alveolar development in intact rats. No effect on mammary development in ovariectomized rats was noticed whether the drug was given orally or by injection; however uterine and adrenal weights were reduced. MGA plus estradiol caused significant (p less than .01) mammary growth in ovariectomized rats as compared with that in rats given MGA or estradiol alone. Uterine weight was increased slightly after supplementation with estradiol, but adrenal weight did not show improvement. It is suggested that MGA is without any estrogenic activity and therefore requires the presence of ovaries or estrogen to exhibit development of mammary growth.  相似文献

A field trial was conducted for 2 years in an Arredondo fine sand containing a tillage pan at 15-20 cm deep to determine the influence of subsoiling on the distribution of corn roots and plant-parasitic nematodes. Soil samples were taken at various depths and row positions at 30, 60, and 90 days after planting in field corn subsoiled under the row with two chisels and in non-subsoiled corn. At 30 and 60 days, in-row nematode population densities to 60 cm deep were not affected by subsoiling compared with population densities in nonsubsoiled plots. After 90 days, subsoiling had not affected total root length or root weight at the 20 depth-row position sampling combinations, but population densities of Meloidogyne incognita and Criconemella spp. had increased in subsoiled corn. Numbers of Pratylenchus zeae were not affected. Subsoiling generally resulted in a change in distribution of corn roots and nematodes in the soil profile but caused little total increase in either roots or numbers of nematodes. Fusarium spp. are plant pathogens producing fumonisins and trichothecenes that both affect human and animal health. In the present study, 40 fungal strains were isolated and species identified from 35 shrimp feed samples and from 61 agricultural raw materials. F. verticillioides was the predominant species (85 %) mostly found in corn and soybean meal, while no Fusarium contamination was detected in shrimp feed. Levels of 10 % of F. oxysporum were isolated from peanut and 5 % of F. equiseti contamination in corn and peanut. To determine the ability of toxin production, enzyme-linked immunosorbent assay, polymerase chain reaction, and ultra-pressure liquid chromatography-tandem mass spectrometry were performed. All but four of the fumonisin-producing strains contained the FUM1 gene. No Fusarium synthesized T-2 toxin nor contained the Tri5 gene. This survey brings more data on mycotoxin contamination in the food chain of animal feed production, and leads to the awareness of the use of contaminated raw materials in shrimp farming.  相似文献

Thirty-six kangaroo rats were captured in the Mojave Desert near Las Vegas. Eight rats, group A, were kept at room temperature, 25 ± 2 °C. Ten, Group B, were kept at 13–15 °C and fed ad libitum. Eighteen, group C, were kept at 13–15 °C and fed only 2 g of dry oats daily. Rate of O2 consumption, VO2, food intake, rectal temperature, Tre., body weight and composition were measured in all groups. Group B showed a significant increase in VO2, and food intake and no change in Tre body weight as compared to group A. After 2 days on the limited food intake group C began to lose weight, and Tre then began to fluctuate ranging between 14 and 36 °C; the animals exhibited hibernation when the Tre was low. Eight of the 18 rats of group C reached Tre values of 14 °C or below; only one of these survived. The lowest Tre in the other 10 was 15 °C; all survived. Chemical analysis of the homogenized rats showed a significant decrease in body fat in group C to average 1.0% contrasted with 7.8% in group B and 3.7% in group A. The VO2 ranged from 1.9 ml/g hr at a Tre of 36.5 to 0.2 ml/g hr at a Tre of 15 °C. In conclusion D. merriami utilizes hibernation as an effective adaptive mechanism to carry out their essential body functions when on a limited food intake. It seems that maintaining a level of more than 1.5% of body fat is essential for successful arousal from hibernation.  相似文献

One hundred zebra fish per tank were maintained for 112 days at 24°C or 28°C in glass aquaria and fed a diet of flaked food made without cellulose (13.45 kJ g?1, metabolizable energy, Type A) or with cellulose (8.71 kJ g?1, metabolizable energy, Type B). Each experimental condition was repeated in triplicate (12 tanks). The weight of food given daily to the fish was based on daily records of survivors (from which mortality rates were calculated) and wet wt of fish (measured every 14 days) in each tank. All fish were fed with the same weight of food per day and the quantity of energy in the food in excess of standard metabolism (as a proportion of SM) was approximately 0–5 for fish maintained at 28°C and fed food B, 1–0 for fish maintained at 24°C and fed food B, 1–5 for fish maintained at 28°C and fed food A, and 2.2 for fish maintained at 24°C and fed food A. Non-ionized ammonia, nitrite and nitrate nitrogen in the tanks did not reach toxic levels although there was an increase in total ammonium nitrogen in one tank and a subsequent heavy mortality. It was assumed that this was caused by the build up of pathogenic bacteria. Apart from this tank, mortality was highest in tanks at 28°C with fish fed food A and second highest in tanks at 24°C with fish fed on the same diet. Growth was measured in units of length, wet and dry weights, carbon and energy. There was a good correlation (P < 0.001) between carbon (mgC mg?1) and calorific (J mg?1) values and a conversion factor of 46.2 J (mgC)?1 was derived. Fish maintained at 24°C and fed food A had the highest rates of growth both in weight and in energy value per unit weight. Fish fed the same diet but kept at 28°C had the lowest growth rates. Both these groups of fish had the highest coefficients of variation in wet weights which increased during the experiment, indicating an increase in interaction within the tanks. There was agreement between the energy value of fish sampled for growth and a condition factor based on the length-weight relationships of fish remaining in the tanks. A correlation (P < 0.05) was found between instantaneous mortality and growth rates for fish between tanks when those maintained at 28°C and fed on food A were ignored.  相似文献

Of 53 isolates of Chaetomium globosum Kunze isolated from corn, manufactured feed, and fiberboard, grown on autoclaved corn, and fed to rats, 25 were lethal within 4 to 6 days. Thirty-six single ascospore cultures of one isolate were uniformly lethal when similarly grown and fed to rats. Incubation of the fungus on corn for 6 to 8 weeks resulted in greater toxicity than shorter incubation times. Premortem symptoms indicated damage to the central nervous system of the affected animals, and postmortem lesions included hemoglobinuria, hemorrhagic enteritis, and subdural hemorrhaging. Corn invaded by C. globosum and toxic to rats had no detectable effects on swine when fed as their sole ration for 6 weeks, and appeared to be consumed with relish. The toxic substance was best obtained by defatting the fungus-invaded corn with petroleum ether (bp, 60 to 70 C), extraction with acetone, and further purification by elution from a silica gel column with acetone and 5% acetone in chloroform.  相似文献

The objective of the present study was to investigate the effect of feeding two transgenic corn lines containing the mCry1Ac gene from Bacillus thuringiensis strain (BT-799) and the maroACC gene from Agrobacterium tumefaciens strain (CC-2), respectively, on growth, egg quality and organ health indicators. Expression of the mCry1Ac gene confers resistance to Pyrausta nubilalis and the maroACC gene confers tolerance to herbicides. Healthy hens (n=96 placed in cages; 3 hens/cage) were randomly assigned to one of four corn–soybean meal dietary treatments (8 cages/treatment) formulated with the following corn: non-transgenic near-isoline control corn (control), BT-799 corn, CC-2 corn and commercially available non-transgenic reference corn (reference). The experiment was divided into three 4-week phases (week 1 to 4, week 5 to 8 and week 9 to 12), during which hens were fed mash diets. Performance (BW, feed intake and egg production) and egg quality were determined. Following slaughter at the end of 12 weeks of feeding (n=8/treatment), carcass yield and organ weights (heart, liver, spleen, lung, kidneys, stomach and ovary) were recorded; organs and intestines were sampled for histological analysis. Analysis of serum biochemistry parameters to assess the liver and kidney function were performed. No differences in BW, egg production and production efficiency were observed between hens consuming the control diet and hens consuming the BT-799 or CC-2 diet. Haugh unit measures and egg component weights were similar between the control and test groups. Carcass yield was not affected by the diet treatment. Similar organosomatic indices and serum parameters did not indicate the characteristics of organ dysfunction. All observed values of the BT-799 and CC-2 groups were within the calculated tolerance intervals. This research indicates that the performance, egg quality, organ health and carcass yield of laying hens fed diets containing the BT-799 or CC-2 corn line were similar to that of laying hens fed diets formulated with the non-transgenic near-isoline corn with comparable genetic backgrounds.  相似文献

A post-fire morel collected from Populus simonii stands in Mt. Qingling was identified as Morchella crassipes Mes-20 by using nuclear ribosomal DNA internal transcribed spacer phylogeny. It was inoculated into sweet corn to observe colonized roots in purified culture and in greenhouse experiments. The elongation and maturation zones of sweet corn were remarkably colonized at the cortex intercellular and intracellular cells, vessel cells, and around the Casparian strip, forming ectendomycorrhiza-like structures. Colonization was also observed in the zone of cell division proximal to the root cap. Greenhouse assays with sweet corn showed that this morel stimulated the development of the root system and significantly increased the dry root biomass. M. crassipes also significantly reduced the incidence of Fusarium verticillioides in the kernels of mature ears when inoculated into young ears before Fusarium inoculation and prevented Fusarium infection in corn ears compared with that of the control in the greenhouse. When grown under axenic conditions, M. crassipes produced the phytohormones abscisic acid, indole-3-acetic acid, and salicylic acid. The benefits to plants elicited by M. crassipes may result from these phytohormones which may improve the drought resistance, biomass growth and resistance to Fusarium.  相似文献

Eighteen Fusarium crookwellense isolates from the continents of Australia, Europe, and North America were compared for their ability to produce mycotoxins on corn at 25 °C after 2 weeks. Extracts from corn fermented with each Fusarium isolate were analyzed by thin-layer chromatography (TLC) and gas chromatography/mass spectroscopy (GS/MS) for mycotoxins. Toxins detected were zearalenone (13 isolates), fusarin C (11 isolates), nivalenol (4 isolates), and diacetoxyscirpenol (2 isolates). Zearalenone and fusarin C were produced by isolates from each continent, while nivalenol was detected in the Fusarium isolates originating from Australia and one isolate from the United States.The mention of firm names or trade products does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned  相似文献

Crompton D. W. T., Singhvi A., Nesheim M. C. and Walters D. E. 1981. Competition for dietary fructose between Moniliformis (Acanthocephala) and growing rats. International journal for Parasitology11: 457–461. The food intake, weight gain and blood sugar of young rats, fed on diets containing growth-limiting amounts of fructose and infected with Moniliformis dubius (Acanthocephala) for 6 weeks, were measured and compared with values obtained from similar uninfected rats which had been treated in the same manner. The growth of the rats was closely related to the fructose content of the diet. However, on average, the infected rats gained less weight than their uninfected counterparts. Significant differences between the values for blood sugar from infected and uninfected rats were not detected when the diet contained 2% fructose. The blood sugar of infected rats fed on a diet containing 4% fructose was found to be significantly less than that of similar uninfected rats. Information was also obtained about the numbers, sex, dry weight and location of the Moniliformis in the small intestines of their hosts. Male and female Moniliformis from rats fed on the 4% fructose diet were found to be heavier than those from rats fed on the 2% fructose diet. The results can be interpreted to suggest that the reduction in the growth rate of the infected rats is due to competition for fructose between Moniliformis and the rat.  相似文献

Reproduction of the corn cyst nematode (Heterodera zeae) and its effect on growth of corn (Zea mays) was studied in plant growth chambers at 24, 27, 30, 33, and 36 C. Reproduction of H. zeae increased directly with increase in temperature from 24 to 36 C. Fifteen-cm-d pots of corn seedlings inoculated with a mixture of 5,000 eggs + J2 and maintained for 8 weeks in growth chambers contained an average of 7,042 cysts + females at 36 C, but only 350 cysts + females at 24 C. Fresh weights of plants without nematodes were highest at 27 C and lowest at 36 C. Nematodes suppressed plant fresh weight by an average of 30% at 27 C and by 27% at 33 C but did not suppress plant weight at 36 C. Heterodera zeae has the highest reported temperature optimum for reproduction of any cyst nematode.  相似文献

Bags of Pinus strobus wood chips with moisture contents of 38, 92, 164, and 217% (oven dry weight) were inoculated with Bursaphelenchus xylophilus and incubated at 30 C in order to determine the effect of wood moisture on nematode population development. Nematodes were extracted after 2, 4, 8, and 12 weeks. Population levels were greatest in wood chips with a moisture content of 38% and decreased successively with each higher moisture content. In chips with the three lower moisture contents, populations peaked at 2 weeks, but at 217% moisture, they peaked at 8 weeks. By 12 weeks, nematode populations had declined in wood chips with 92 and 164% moisture contents. The fungi most frequently isolated from the wood chips were Alternaria, Fusarium, Gliocladium, Graphium, Penicillium, Trichoderma, and Mucorales.  相似文献

The primary objective of this study was to examine the effects of different inclusion rates of dietary triticale replacing corn grain in starter rations of dairy calves on feed intake, average daily gain (ADG), feed efficiency, skeletal growth, fecal score, and selected blood and ruminal parameters. In all, 30 4-day-old Holstein calves (15 female and 15 male) were blocked by gender and birth weight, and then randomly allotted to three treatment groups (n=10 calves/treatment) and received either a corn-based diet without triticale (T0), or a diet containing 16% (T16) or 32% (T32) triticale (25% and 50% corn replaced by triticale, respectively) for 10 weeks. Calves were weaned on day 43 of study and remained on the study until day 70. During post-weaning period, calves fed T32 had the lowest starter intake (1.78 kg/day; P=0.02). In contrast, ADG was not affected in pre-weaning and overall periods, but calves fed T32 had a lower ADG (P=0.04) as compared with calves fed T0 in post-weaning period. No dietary effect was detected for feed efficiency. Dietary treatment did not affect heart girth and body length; however, height at wither and hip at weaning increased in calves fed triticale compared with T0. No detectable effects were observed in serum glucose and β-hydroxybutyrate. No difference was detected in blood urea nitrogen on day 35 either but on day 50 and day 70 the greatest concentration was recorded for calves fed T16. Compared with the control, ruminal ammonia concentration was increased for calves fed T32 (4.34 v. 7.50 mmol/l) and T16 (4.01 v. 8.12 mmol/l) on day 35 and day 50. No difference was detected in ruminal pH on day 35 or day 50; however, calves fed T32 had the lowest pH (6.11) at 70 days. No significant effect was detected in days scoured, respiratory score and general appearance. Under our experimental conditions, it appears that triticale at 16% dry matter (i.e. replacing corn grain up to 25%) in the starter diet does not have adverse effects on the performance and intake of calves. Therefore, substituting corn partially with triticale in calf starter diets may prove beneficial in places where corn is less abundant or its price is prohibitive. The low number of calves per treatment however may have limited the statistical power to detect significant differences among treatments, possibly affecting the results, which should be interpreted with caution.  相似文献

The embryogenic development of Rolylenchulus borealis, at 24-26 C, was completed on corn, in 12-15 days, and the life-cycle of the nematode from egg to egg required 35-40 days at 20-25 C. Juveniles remained in the soil as preinfective stages for 17-19 days before becoming adults. Only immature vermiform and swollen egg-laying females were found attached to corn roots. Eggs were laid in a gelatinous matrix on the root surface; the number of eggs per egg mass was 45 ± 28 on corn roots. Bean, green pea, potato, sorghum, and sweet potato were also found to be hosts of R. borealis. The nematode established a permanent feeding site on corn root in an endodermal cell that became hypertrophied. Pericyclic cells close to the feeding site showed granular cytoplasm and nuclei with hypertrophied nucleoli. A cell wall ingrowth was also noted around the area of stylet penetration into the endodermal cell.  相似文献
