小麦背景中黑麦1R染色体的遗传变异   总被引:8,自引:0,他引:8  
运用细胞遗传学方法鉴定了来源于中国春×M27(1R/1D代换系)的花粉植株和F2单株染色体组成。发现7个花粉植株中出现7种染色体组成变异类型,每株呈现一类变异;而27个F2单株中,存在11种染色体组成变异类型,变异频率仅为37.0%,低于花粉植株。花粉植株群体中,观察到一个能稳定向后代传递的小麦/1R小片段易位,但F2群体中未检测到小麦/黑麦易位。表明常规染色体工程结合花药培养是有计划、有目的实现异源染色体小片段向小麦转移的简便、高效、快速途径。  相似文献

浓缩狂犬病疫苗接种后不良反应与血清抗体水平分析   总被引:7,自引:1,他引:6  
报导了≥2.5IU浓缩狂苗人体接种后的不良反应其反应率为1.31%与以往未浓缩常规苗的反应率为0.73%比较两者具有显著性差异(X2=6.94,P<0.01),浓缩苗以注射1~3针时的不良反应多,临床表现以局部极痛、红肿、瘙痒、急性荨麻疹和过敏性休克等。但其血清抗体阳转率7天时为40.0%以上,14天时为100%,而常规苗仅是40.0%,抗体滴度浓缩苗第7天时达到有效保护水平(中和试验)。显示了其抗体产生早,水平高特点,关于如何减少浓缩狂苗不良反应问题,作者提出了几点建议  相似文献

研究以名贵月季品种“落霞”、“墨红”、“和平”、“金背大红”等为试材,以MS基本培养基附加不同浓度的6-BA、NAA,以诱导腋芽增殖为微繁途径。结果表明:无菌外植体的建立培养基为MS+6-BAO.5-2+NAAO.05-0.1+Vc100;芽增殖培养基为MS十BA1+NAA0.05,在此培养基上繁殖系数为5.5;生根培养基为MS+IAA1+IBA1+NAA0.1-肌醇,生根率在95%左右。生根苗移栽成活率在85%左右。同时对繁殖工艺流程、成本、商品苗产率等问题进行了讨论,给出在繁殖系数为F、生根数为b时,有效繁殖系数f=F—b,试管苗实际增殖倍数Nn=(F—b)(n-1)·F、生根苗数R1-n=f-1;当生根率为r1,移栽成活率为r2,则商品苗产率为C=r1·r2·Rn=r1·r2·b(f-1)/(f-1)  相似文献

用0.1%秋水仙碱溶液处理甜菊实生苗生长点,可诱变产生甜菊多倍体(四倍体)植株,用8次点滴处理,诱变率可达31.25%。染色体数鉴定表明:四倍体甜菊染色体数是2n=44,而二倍体甜菊染色体数是2n=22.形态学和解剖学的观察表明,四倍体比二倍体甜菊植株的茎矮壮,叶片增大,长度增长2.1倍,宽度扩大2.3倍,叶片加厚1.7倍,叶色更浓绿,叶片气孔数减少,气孔变大。叶片糖苷含量测定表明:四倍体的叶片含量为15.7%,而二倍体的叶片含量为10.8%,前者比二倍体叶片含量提高4.9%。  相似文献

本文对上海浦东东海海滩上[31°1'N]芦苇植被自堤至海水边各地段的生物生产力、生长特性及土壤状况作了测定。其平均生物生产力为2.074±0.554kgm ̄(-2)a ̄(-1),其中地上部和地下部生物量分别为1.433±0.382和0.641±0.566kgm ̄(-2)a ̄(-1)。观察了自芦苇出土起,大气CO_2浓度加倍对芦苇生长的影响。结果表明,处理44d后植株叶面积和比叶重相应提高,生物生产力比对照增加53.7%,其光合产物主要分配在地上部分。试验结束时,CO_2加浓处理的叶片叶绿素含量和在正常空气中的光合速率与对照相似。  相似文献

离体培养半夏Pinellia ternata 茎尖获得小植株。用其叶柄作外植体在MS添加2,4-D 和KT的培养基上诱导出愈伤组织,继代培养中挑选出一种表面呈颗粒状、易分散、生长快的愈伤组织。在MS添加KT0.5 m g/L, NAA 0.2 m g/L的培养基上,叶柄可直接分化形成小植株,愈伤组织通过形成小块茎的途径产生完整植株。块茎顶端芽分化过程中,先形成的叶原基或幼叶总是覆盖着后面的叶原基而出现一种依次叠套的特殊结构。液体浅层培养对试管苗的增殖速率比固体培养快1 倍。叶柄和愈伤组织的小植株分化率均在70% 以上,移栽成活率为100% 。试管苗的块茎产量(鲜重)比对照(用小块茎繁殖)的净块茎产量(鲜重)高103% 。试管苗块茎的总生物碱含量为0.344% ,野生和人工栽培半夏块茎的总生物碱含量分别为0.264% 和0.203% 。  相似文献

以草木樨状黄芪无菌苗茎切段为材料,在含1-2mg/L2,4-D和0-0.5mg/L6BA的MS培养基上培养获得大量愈伤组织,愈伤组织诱导率在95%以上,愈伤组织在附加0.2mg/LKT,1mg/L6BA,0或0.5mg/LNAA,500mg/LCH 和200mg/L YE的MS培养基上诱导丛生芽,并进而发育成苗。苗的分化频率为100%。分化苗或其茎的切段在不国源植物激素的1/2MS培养基上可出现根的分化,分化频率达90%以上,再生植株经炼苗后移栽成活率达80%以上。  相似文献

本文报道茄属果树可乐茄(SolanumquitoenseLam.)叶肉原生质体的分离、培养及植株再生。幼嫩叶片原生质体经酶游离、纯化后,以1×104个/ml密度培养于稍加改良K8p(附加2,4-D0.5mgL(-1)、NAA1.0mgL(-1)和BA0.5mgL(-1))的培养基中,三天后开始分裂,一周分裂3—4次。一个月形成小细胞团,植板率为0.1—0.2%,小细胞团转培养于MS+2,4-D0.5mgL(-1)上增殖后进行分化。原生质体来源愈伤组织在IAA(0.1—1.0mgL(-1))与BA或ZT组合的培养基中能诱导器官发生,芽分化率最高可达42.9%;但IAA、BA、ZT三者一起使用未见任何器官分化。小芽在MS+IAA0.2mgL(-1)中生根成植株。可乐茄叶肉原生质体的植株再生,可应用于育种和茄属植物遗传工程研究。  相似文献

取荷兰鸢尾(IrisxiphiumL.var.hybridum)鳞茎片不同部位外植体块,接种于附加不同激素配比的基本培养基上,其中取自鳞茎片基部外植体块2mm×2mm×2mm,培养基为MS+BA1.0mg/L+NAA0.2mg/L的不定芽诱导率为最高(70%);最理想的增殖培养基为MS+BA2.0mg/L+NAA0.2mg/L。不定芽直径4~5mm,培养基为MS+BA0.2mg/L+NAA0.5mg/L有利于不定根的发生,诱导生根率达833%。试管苗不经练苗可直接出瓶,移栽于泥炭∶田土∶河沙=1∶1∶1(V/V)的基质中。  相似文献

Influence of environmental factors on origin of colchicine-induced true-breeding diploid mutants in sorghum. Amer. Jour. Bot. 46(2) : 119-125. Illus. 1959.—Factors involved in the induction of true-breeding diploid mutants following colchicine treatment of sorghum seedlings (line ‘Experimental 3‘) were studied by growing untreated and treated seedlings in aseptic cultures under controlled conditions and examining mature plants and progenies for genetic changes. Factors found to increase rate of early growth and survival of treated seedlings are: use of agar media rather than quartz sand plus liquid media, presence of major minerals in the medium, and presence of up to 2% sucrose in the medium. The beneficial effect of sucrose is contingent upon the presence of major minerals. IAA from 0.01 p.p.m. to 0.1 p.p.m. increased rate of both root and top growth of treated seedlings during early development. Increased osmotic values of the medium increased early growth rate and survival of treated seedlings up to a concentration equivalent to 0.123 molar sucrose while early growth of control seedlings fell off with every increase in osmotic value. This shift in optimum osmotic value of the medium would seem to indicate an increase in the osmotic concentration of the tissues of colchicine-treated seedlings. Progeny tests of some of the treated plants from the culture experiments demonstrated that, out of 137, none were true-breeding diploid mutants. There were instances of polyploidy and minor changes, but the 2 treated plants which were the only obvious mutants gave progenies which segregated for many characters. Improved growing conditions for treated seedlings decreased the colchicine effect of induction of true-breeding diploid mutants as well as the colchicine effects of retarded seedling growth and decreased survival.  相似文献

1981-1990年对冷箭竹(Bashaniafangenia)开花、结实、种子萌发、幼苗更新复壮等生殖特性进行了观察。冷箭竹开花周期为45-55年,开花持续期约10年,1983年盛花期测定茎秆上着生花序的长度平均为170.8cm,每1茎秆上的小花数为1280.3枚,每朵小花从开放至萎蔫的持续期约6天,花药持续开裂时间为0.77天,花粉生活力达96.7%,柱头承受花粉时间为2.3天,雌蕊子房发育为果实平均为3586粒/m ̄2,未受粉的瘪粒为13994粒/m ̄2。种子生活力为27.6%,种子发芽率为15%-20%,当年生幼苗成活率为76%-93%。  相似文献

Given that seeds fertilized by slowly growing pollen are of low quality genetically, we theoretically reanalyzed the hypothesis that plants selectively abort ovules fertilized later to enhance the mean quality of resulting seeds. We assumed that both superior and inferior pollen exist, the superior pollen growing faster to fertilize ovules, resulting in seeds of higher quality than those of ovules fertilized by inferior pollen. We developed two models to determine the conditions under which selective abortion is favored. In the first model, ovules in one flower are fertilized by pollen grains that arrive at different times, with each visit bringing both fast- and slow-growing pollen. In the second model, ovules in two flowers are fertilized by all pollen grains that arrive at the same time. In the first model, we found that selective abortion based on the order of fertilization is never advantageous irrespective of the duration of the time lag between the two visits. Rather, random abortion is possibly favored. In the second model, although selective abortion based on the order of fertilization can be advantageous, the parameter region favoring it is rather restricted. This is because overproduction can be advantageous only if the quantity of the superior pollen is not limited in one flower but is limited in the other flower. In addition, the degree of overproduction was very low, implying that the merit of overproduction (increase in the number of superior seeds) is low compared to the cost of overproducing ovules. These results suggest that selective abortion of ovules based on the order of fertilization is not as advantageous as previously considered.  相似文献

In the present paper, an experiment was conducted to study the effects of soil moisture content on dry nursery seedling quality in Guangzhou in 1995. Through comparing the difference of dry nursery seedlings and wet nursery seedlings, we found a close relationship between soil moisture content and seedling growth. The seedling emergence of dry nursery seedling was more even, tidy and faster, and the survival rate was higher than that of wet nursery seedling. Dry nursery seedlings had small plant stature, slow leaf stretching speed and low individual seedling dry weight, but had high dry/fresh weight ratio. This was abeneficial factor for seedlings to recover from transplanting shock more quickly. As com-pared with the wet nursery seedlings, dry nursery seedlings had poor rooting ability,but had more vigorous white roots and fewer rust roots. It was the possibly important reasonfor dry nursery seedlings to form strong“explosive force”.  相似文献

The relationship between pollen and progeny performance has been a subject of many studies but the evidence for pollen-tube growth rate as an indicator of progeny fitness is equivocal. We used an anemophilous tree, Betula pendula, to examine the relationship between pollen-tube growth rate and seed and seedling performance. We crossed nine maternal plants with pollen from six pollen donors in a clonal B. pendula seed orchard, measured the pollen-tube growth rates for every cross, and analyzed the performance of the resulting seeds and seedlings. The only significant positive correlation was found between pollen-tube growth rate and seed mass when we controlled for seed number per inflorescence. Using seed mass as a covariate, we found that only maternal parent had a significant effect on the number of seeds per inflorescence, the percentage of germinable and embryonic seeds, and early seedling growth. Both maternal and paternal parents had significant effects on seedling height after 85 d of growth. These results are in concordance with the general view that maternal effects are usually most apparent in seed characters and during early plant growth. This study does not provide strong evidence for the theory of pollen-tube growth rate as an indicator of progeny quality.  相似文献

2001年在郑州黄河沿岸对外来植物小花山桃草(Gaura parviflora)的生物学特性、地上生物量动态进行了研究。结果表明:①小花山桃草各构件的变化特征为:茎高、叶数和花序数在6月25日都达到生长高峰,随后茎高、花序数继续增长,叶数迅速降低。②小花山桃草每株花序数为10.9枝。每个花序产134.23朵小花,产126.75粒种子,结实率为94.43%,不育率为5.57%。③小花山桃草单株地上生物量、叶生物量、茎生物量在6月25日达到最大,花序生物量在7月20日达到最大。在生长初期,小花山桃草占样方内所有植物生物量的78%,7月20日达100%。④小花山桃草入侵可能与植株高大、幼苗期短、生长快以及具有较强的传播和繁殖能力有关。  相似文献

The roots of rain forest plants are frequently colonized by arbuscular mycorrhizal fungi (AMF) that can promote plant growth in the nutrient poor soils characteristic of these forests. However, recent studies suggest that both the occurrence of AMF on rain forest plants and the dependence of rain forest plants on AMF can be highly variable. We examined the occurrence and levels of AMF colonization of some common seedling species in a tropical and a subtropical rain forest site in Queensland, Australia. We also used a long-term database to compare the growth and mortality rates of seedling species that rarely formed AMF with those that regularly formed AMF. In both forests, more than one-third of the seedling species rarely formed AMF associations, while 40% of species consistently formed AMF in the tropical site compared to 27% in the subtropical site. Consistent patterns of AMF occurrence were observed among plant families at the two sites. Variation among seedling species in AMF occurrence or colonization was not associated with differences in seed mass among species, variation in seedling size and putative age within a species, or lack of AMF inoculum in the soil. Comparisons of four seedling species growing both in the shaded understory and in small canopy gaps revealed an increase in AMF colonization in two of the four species in gaps, suggesting that light limitation partially explains the low occurrence of AMF. Seedling survival was significantly positively associated with seed biomass but not with AMF colonization. Furthermore, seedling species that regularly formed AMF and those that did not had similar rates of growth and survival, suggesting that mycorrhizal and nonmycorrhizal strategies were equivalent in these forests. Furthermore, the high numbers of seedlings that lacked AMF and the overall low rate of seedling growth (the average seedling required 6 years to double its height) suggest that most seedlings did not receive significant indirect benefits from AMF through connection to canopy trees via a common mycorrhizal network.  相似文献

用不同浓度秋水仙素处理野生南荻×芒(Miscanthus lutarioriparia×Miscanthus sinensis)远缘杂交后代以诱导产生多倍体,并对变异株进行形态学和细胞学鉴定,以期获得稳定的四倍体植株并分析其生理特性。结果表明:(1)采用秋水仙素加入培养基处理法和秋水仙素溶液浸泡处理法都可获得一定频率的多倍体植株;胚性愈伤组织以0.2%秋水仙素浸泡处理48h的诱变效果较好,四倍体诱导率达8.7%;芽在0.05%秋水仙素培养基中处理15d较好,四倍体诱导率达10.6%;生根苗在0.1%秋水仙素培养基中处理10d较好,四倍体诱导率达11.1%。(2)经体细胞染色体计数,加倍植株染色体数为2n=4x=76,对照植株的染色体数目为2n=2x=38。(3)生长2年的多倍体植株形态、叶片大小、茎粗、茎壁厚、节间等性状表现出巨大性和超亲优势。  相似文献
