When A. pullulans is grown on a glucose medium with a limiting nitrogen source and low buffer capacity, the yeast-like cells that are originally present undergo a transition to chlamydospores. The initial pH must be around 6 for the transition to take place under optimal conditions. On the above-mentioned medium pH decreases to values below 2 in the first two days; if this decrease is prevented, either by buffering the medium or by repeatedly adjusting the pH to its original value no chlamydospores form.  相似文献

Summary Polysaccharide formation by Aureobasidium pullulans was affected by the nitrogen source in the medium, and its yield fell when excess ammonium ions were present, even under conditions which otherwise supported its synthesis. Experiments carried out with pH control gave results which question some of the earlier published data, so it is possible to obtain high polysaccharide yields at low pH (2.5), and mycelial growth at high pH (6.5) with certain nitrogen sources. Growth and production of zeaxanthin by Flavobacterium sp were studied using different carbon and nitrogen sources in a chemically defined medium. The best growth was supported by sucrose, but glucose yielded similar carotenoid concentrations. Both asparagine and glutamine stimulated growth and pigment formation. Carotenoid production and glucose consumption increased as a function of asparagine concentration. In the presence of asparagine, high glucose concentrations decreased pigment production without affecting biomass formation. In the absence of glucose, asparagine could not support growth and zeaxanthin production. When compared to the effect of 55 mM glucose, 10 mM oxaloacetate increased growth and carotenoid production. Pyruvate and other intermediates of the citric acid cycle showed a similar stimulatory effect. The intermediates of glycolysis: glucose 6-phosphate and fructose 1,6-diphosphate did not support growth. These results suggest that Flavobacterium sp utilizes asparagine primarily as a nitrogen source for growth and production of zeaxanthin. Summary Glucoamylase production kinetics was greatly affected by medium composition. While maltose and cassava flour induced glucoamylase synthesis, fructose clearly repressed it, reaching a maximum enzyne activity value (Am) of only 6% of those obtained with the former carbon sources. Among the nitrogen sources the best result was obtained with urea, reaching Am values of about 40% higher than those obtained with ammonium sulphate.  相似文献

During the last century, the global biogeochemical cycles of carbon (C) and nitrogen (N) have been drastically altered by human activities. A century of land‐clearing and biomass burning, followed by fossil fuel combustion have increased the concentration of atmospheric CO2 by approximately 20%, and since the mid‐1900s, the use of agricultural fertilizers has been the primary driver of an approximate 90% increase in bioavailable N. Geochemical records obtained through stable isotope analysis of terrestrial and marine biota effectively illustrate rising anthropogenic C inputs. However, there are fewer records of anthropogenic N, despite the enormous magnitude of change and the known negative effects of N on ecosystem health. We used stable isotope values from independent octocorals (gorgonians) sampled across the Western Atlantic over the last 143 years to document human perturbations of the marine C and N pools. Here, we demonstrate that in sea plumes δ13C values and in both sea plumes and sea fans δ15N values declined significantly from 1862 to 2005. Sea plume δ 13C values were negatively correlated with increasing atmospheric CO2 concentrations and corroborate known rates of change resulting from global fossil fuel combustion, known as the Suess effect. We suggest that widespread input of agricultural fertilizers to near‐shore coastal waters is the dominant driver for the decreasing δ 15N trend, though multiple anthropogenic sources are likely affecting this trend. Given the interest in using δ 15N as an indicator for N pollution in aquatic systems, we highlight the risk of underestimating contributions of pollutants as a result of source mixing as demonstrated by a simple isotope‐mixing model. We conclude that signals of major human‐induced perturbations of the C and N pools are detectable in specimens collected over wide geographic scales, and that archived materials are invaluable for establishing baselines against which we can assess environmental change.  相似文献

Cytochromes ofAureobasidium pullulans have been identified and partially characterized using low-temperature and carbon-monoxide-difference spectroscopy. The presence ofa-,b-, andc-type cytochromes is demonstrated, as are other unidentified redox components. During exponential growth in batch culture, cytochrome levels showed complex changes. Changes in respiration rates and in the levels of cytochromea+a 3 closely paralleled cellular growth: both increased exponentially until stationary phase, when no further increase occurred. Theb- andc-type cytochromes showed biphasic increases, initially doubling every, generation time and then increasing more slowly during the stationary phase. Sensitivity of respiration to 100M potassium cyanide gradually decreased during exponential growth, falling from virtually 100% inhibition after about 20 h growth to 30% inhibition in the stationary phase. The results suggest that in stationary-phase cultures, an alternative cyanide-insensitive but salicylhydroxamic-acid-sensitive terminal oxidase also operates.  相似文献

The effect of carbon sources, glucose and sucrose, and nitrogen sources such as ammonia, glutamate andl-citrulline on the activities of glutathione metabolic enzymes has been studied. Yeast and mycelial cells were used to identify changes in activity levels of glutathione reductase (GSSGR), glutathione transferase (GST), glutathione peroxidase (GPX) and -glutamyl transpeptidase (GGT). Enzyme activities from cells grown in sucrose media were lower than in glucose media regardless of the enzyme tested, morphological form, or the growth interval. In all enzymes except GST, activity was higher in yeast form than in mycelia, regardless of nitrogen source, with lower activity from 24 to 72 h than at 96 h. In citrulline media, yeast form showed the maximum GST, GGT, and GPX activity. In ammonia-amended media, mycelia showed maximum activity in GGT, whereas in glutamate media, mycelia showed the maximum activity in GST. Also, the type of nitrogen source had no effect on GPX activity in the mycelial form. Finally, changing the nitrogen source showed no significant effect on GSSGR activity, either in the yeast or mycelial form.  相似文献

The effects of different carbon and nitrogen sources on the growth, nitrogenase activity, and carbon metabolism of Gluconacetobacter diazotrophicus were investigated. The amino acids asparagine, aspartic acid, and glutamic acid affected microbial growth and nitrogenase activity. Several enzymatic activities involved in the tricarboxylic acid cycle were affected by the carbon source used. In addition, glucose and gluconate significantly increased the oxygen consumption (respiration rate) of whole cells of G. diazotrophicus grown under aerobic conditions. Enzymes responsible for direct oxidation of glucose and gluconate were especially active in cells grown with sucrose and gluconate. The presence of amino acids in the apoplastic and symplastic sap of sugarcane stems suggests that these compounds might be of importance in the regulation of growth and nitrogenase activity during the symbiotic association. The information obtained from the plant-bacterium association together with the results of other biochemical studies could contribute to the development of biotechnological applications of G. diazotrophicus.  相似文献

Effects of different pH and carbon sources on pullulan production, UDP-glucose level and pullulan-related synthases activity inAureobasidium pullulans Y68 were examined. It was found that more pullulan was produced when the yeast strain was grown in the medium with initial pH 7.0 than when it was grown in the same medium with constant pH 6.0. The results also show that higher pullulan yield was obtained when the cells were grown in the medium containing glucose than when they were cultivated in the medium supplementing other carbon sources. Our results demonstrate that the more pullulan was synthesized, the less UDP-glucose was left in the cells ofA. pullulans Y68. However, it was observed that more pullulan was synthesized; the cells had higher pullulan-related synthase activity. Therefore, high pullulan yield was related to low UDP-glucose level and high pullulan-related synthases activity inAureobasidium pullulans Y68.  相似文献

研究了8种有机碳源和4种氮源对三角褐指藻(Phaeodactylum tricornutum)生长的影响.结果表明,三角褐指藻具有兼养生长的能力,碳浓度为5mmol/L和50mmol/L时,葡萄糖、果糖、乙酸钠和甘油对其生长有明显的促进作用,乳酸钠和乙醇抑制藻细胞生长,半乳糖和柠檬酸钠对其生长随有机碳浓度而异.甘油、葡萄糖和乙酸钠的适宜浓度范围分别为5 -800mmol/L、100 -400mmol/L和50 -300mmol/L.培养液中分别加入50mmol/L甘油、400mmol/L葡萄糖和150mmol/L乙酸钠,培养第14天,三角褐指藻的最大生物量分别为对照的1.45倍、1.25倍和1.11倍.甘油兼养生长的最优氮源是尿素,适宜浓度范围为0.88-8.8mmol/L.当尿素浓度为4.4mmol/L时,最高生物量可达1.31g/L.  相似文献

Carbon and nitrogen influence the lipid accumulation and arachidonic acid production by Mortierella alpina CCF 185 cultivated at 28°C for 7 days on a rotary shaker. The fungus grown with glucose, starch or dextrin as the carbon source produced satisfactory amounts of lipid and arachidonic acid. The maximum arachidonic acid values were obtained on media with yeast extract or NaNO3 as the nitrogen source. The C/N ratio of 40:1 resulted in the highest yields of lipid and arachidonic acid with different amounts of NaNO3 concentration.  相似文献

Mycotoxins are natural, secondary metabolites produced by fungi on agricultural commodities in the field and during storage under a wide range of climatic conditions. The ochratoxin A (OTA) is a nephrotoxic mycotoxin produced by several species of Aspergillus and Penicillium. In this study, the influence of carbon and nitrogen sources on ochratoxigenic Penicillium species was assessed. The ochratoxigenic Penicillium species were isolated from poultry feed samples of Andhra Pradesh, India. The isolated ochratoxigenic Penicillium species were identified, screened and characterised as OTA producers by high performance thin layer chromatography (HPTLC) and confirmed by high performance liquid chromatography (HPLC). This experiment was carried out using Czapak yeast Autolysate (CYA) medium with different carbon (C) and nitrogen (N) sources at pH 6.5 and incubated at 25 ± 2°C under dark condition. Maximum OTA production was recorded in the presence of D-glucose followed by D-galactose and D-lactose as carbon sources. Similarly, the maximum amount of OTA production was observed in thiourea followed by potassium nitrate as nitrogen source. However, OTA production, final pH of the medium, and mycelial yield and OTA production of both the species of Penicillium varied with C and N present in the medium. The kinetics of the both species of Penicillium was observed for 30 days at an interval of three days. The maximum amount of OTA was detected by 12 and 15 days incubation periods for Penicillium nordicum and Penicillium verrucosum, respectively.  相似文献

Microorganisms can produce lipases with different biochemical characteristics making necessary the screening of new lipase-producing strains for different industrial applications. In this study, 90 microbial strains were screened as potential lipase producers using a sensitive agar plate method with a suitable medium supplemented with Tween 20 and also a liquid culture supplemented with olive oil. The highest cell growth and lipase production for Candida viswanathii were observed in triolein and oleic acid when used as the only pure carbon source. Renewable low-cost triacylglycerols supported the best cell growth, and olive oil was found to be the best inducer for lipase production (19.50 g/L and 58.50 U). The selected conditions for enzyme production were found with yeast extract as nitrogen source and 1.5 % (w/v) olive oil (85.70 U) that resulted in a good cell growth yield (YX/S?=?1.234 g/g) and lipase productivity (1.204 U/h) after 72 h of shake-flask cultivation. C. viswanathii lipase presented high hydrolytic activity on esters bonds of triacylglycerols of long-chain, and this strain can be considered an important candidate for future applications in chemical industries.  相似文献

Three isolates ofAspergillus niger produced polygalacturonase (PG) and pectin methyl galacturonase (PMG) in the presence of organic and inorganic nitrogen sources. Complete inhibition of PG PMG cellulase (Cx) and proteinase synthesis was found in the presence of cystine in all isolates. Maximum biomass was found in sodium nitrate whereas no isolate could grow in the presence of cystine. A correlation between biomass and enzyme production could be obtained when sodium nitrate and cystine were added to the medium separately. All isolates produced pectic cellulolytic and proteolytic enzymes in the presence of various native carbon sources. Sodium polypectate was found to be the best carbon source for the production of PG and PMG; pectin inhibited completely the production of PG and PMG. Maximum cellulase production was brought about by cotton in all three isolates. Maximum proteinase production was observed with gelatin which served as poor substrate for fungal growth. Sucrose supported maximum fungal growth in comparison with all other native carbon sources. The increased production of pectolytic cellulolytic and proteolytic enzymes in the presence of sodium polypectate reflected a stimulation rather than an induction of synthesis of these enzymes.  相似文献

The effect of the nitrogen source and the C/N ratio of the growth medium on the biosynthesis, composition, and structure of the exopolysaccharides (EPSs) of Aurebasidium pullulans (de Bary) Arnaud var. aubasidani Yurlova var. nov. and A. pullulans var. pullulans was studied. A. pullulans var. pullulans and A. pullulans var. aubasidani strains synthesized the maximum amounts of EPSs in the presence of, respectively, a reduced nitrogen source ((NH4)2SO4) and an oxidized nitrogen source (NaNO3) in the medium. The data presented confirm the validity of using the chemical composition and structure of the major cetavlon-precipitated fraction of A. pullulans EPSs for the characterization of intraspecies taxa.  相似文献

本研究旨在阐明出芽短梗霉在不同氮源培养基中形态和胞外多糖的积累及化学成分变化。采用摇瓶法培养出芽短梗霉。三种培养基的氮源分别为硝酸钠(培养基1,M1)、硫酸氨、酵母膏(培养基2,M2)和硫酸氨、蛋白胨和酵母膏(培养基3,M3)。M1培养基中,菌丝体和单细胞的生物量积累均比M2、M3低,但胞外多糖的产量则等于甚至略超过M2和M3。在指数生长的前期,白色菌丝体和酵母状细胞状态占优势。指数生长的后期,以厚垣孢子、肿大细胞和黑色菌丝体占优势。胞外多糖都能为茁霉多糖酶水解为麦芽糖和麦芽三糖,说明这些多糖的化学组成都具有(1→4,1→6)-α结构的茁霉多糖。但M1中产生的茁霉多糖结构单元为麦芽糖和麦芽三糖,且二者比例相当。M2中茁霉多糖的麦芽糖结构单元明显减少,而M3中144h后麦芽糖结构单元完全消失。这似乎表明氧化性的氮源和低溶解氧水平可能是造成茁霉多糖结构单元同时具有麦芽糖和麦芽三糖的原因。  相似文献
