Seven common wombats (Vombatus ursinus) were exposed and two of these were re-exposed to Sarcoptes scabiei var. wombati (Acari: Sarcoptidae). For wombats exposed for the first time, five exposed to 5,000 mites on their shoulder developed moderate to severe parakeratotic mange after 11 wk compared with two given 1,000 mites that developed mild clinical signs of mange after 11 wk. For re-exposed wombats, one of two given 5,000 mites developed mild parakeratotic mange and the other developed severe parakeratotic mange. Initial signs of mange were erythema followed by parakeratosis, alopecia, excoriation and fissuring of parakeratotic crust and skin. Erythema usually became apparent within 14 days after exposure (DAE) or within 24 hrs of re-exposure. Parakeratosis was visible 14-21 DAE and alopecia first occurred 35-77 DAE. Clinical signs increased in severity over time and lesions spread slowly from the site of exposure. Mangy wombats scratched excessively, lost weight, and exhibited a significant neutrophilia compared with control wombats. Treatment of mange with three injections of ivermectin, 300 micrograms/kg, 10 days apart led to complete resolution of clinical signs. However mites were not entirely eliminated until wombats received a second regime of treatment.  相似文献

The cellular response in the dermis of common wombats (Vombatus ursinus) with sarcoptic mange exhibited some typical aspects of an immune response to Sarcoptes scabiei. There was an induction phase for wombats experimentally infected with S. scabiei represented by absence of a dermal inflammatory infiltrate for at least 12 days after infection. T lymphocytes, plasma cells, mast cells, and neutrophils then entered the dermis, consistent with a type IV (delayed) hypersensitivity response. In free-living wombats with severe parakeratotic sarcoptic mange eosinophils were also present in the dermis suggesting that a type I (immediate) hypersensitivity response may develop after a type IV hypersensitivity response. Absence of plasma cells and B lymphocytes in free-living wombats with severe parakeratotic sarcoptic mange compared with their presence in wombats experimentally infected with S. scabiei suggested that some immune tolerance may develop with severe infections. A large proportion of cells in the dermal response were not identified but were possibly cells of connective tissue. The thickness of the epidermis increased within 4 days in response to S. scabiei infection. Some antibodies raised against human leucocyte antigens CD3, CD5, HLA-DP, DQ, DR, and CD79b cross-reacted with leucocyte antigens of common wombats and were used to identify cell types in inflammatory infiltrates using immunohistochemistry.  相似文献

The life cycle of Sarcoptes scabiei var. canis was systematically investigated in vivo. The life cycle of females and males consisted of an egg, larva, protonymph, and a tritonymph that gave rise to an adult. Development from egg to adult required 10.06-13.16 days for the male and 9.93-13.03 days for the female. Egg incubation times were greater than 50.1 to less than 52.97 hr. Larval duration was between 3.22 and 4.20 days. The durations of protonymphal stages that were destined to become females and males were greater than 2.40 to less than 3.40 days and greater than 2.33 to less than 3.33 days, respectively. Tritonymphs destined to become females and males molted in greater than 2.22 to less than 3.22 days and greater than 2.42 to less than 3.42 days, respectively. During development, all life stages frequently left their burrows and wandered on the skin surface.  相似文献

Sarcoptes mange is an important disease that affects the health, mobility and longevity of bare-nosed wombats (Vombatus ursinus) in Australia. We measured the activity patterns of wombats infected with varying levels of mange in an agricultural riparian environment using motion-sensing cameras. Over a 10 month period we observed a greater increase in nocturnal activity of mange-free wombats than those with mange on six or more body sections. The percentage of mange-infected wombats was greater during the day, but in the 6 h after midnight, wombats were detected with fewer mange-infected segments. Air temperature at the time of wombat detections was generally higher for individuals with mange than those without mange. Our results show that diurnal activity of wombats is likely to increase with higher levels of infection by sarcoptes mange. Increased diurnal activity is likely to present serious problems for the persistence of sustainable populations of wild wombats.  相似文献

Sarcoptic mange has been identified as the most significant infectious disease affecting the common wombat (Vombatus ursinus). Biochemistry and haematology values for wild common wombats have not been published previously. A 35% of the wombat population were exhibiting clinical signs of the disease. Significant changes in blood haematology and biochemistry parameters were observed in affected animals. A seasonal variation in numbers of animals affected was demonstrated. Females appeared to be more commonly affected than males although the reason for this is not forthcoming.  相似文献

Brimer et al. (Vet. Parasitol. 51: 123-135, 1993 and 59: 249-255, 1995) developed a migration assay for acaricidal effect of acetylcholinesterase inhibitors and macrocyclic lactones utilising Sarcoptes scabiei var. suis mites. In contrast to many others, this assay is fully quantitative but quite time-consuming. The aim of the present investigation was to modify this assay to become faster and simpler. As a result accurate determinations can now be obtained within 6h, as opposed to 24h. Furthermore it was demonstrated that also Otodectes cynotis mites can be used with only minor modifications of the procedures. The cholinesterase inhibitor diazinon and the formamide amitraz were used as acaricides. Thus, the mite migration assay now has been proven useful for acaricidal compounds belonging to three chemical groups with different modes of action, namely organophosphorous cholinesterase inhibitors, macrocyclic lactones acting on the glutamanergic/GABAegic motoneurons, and formamide inhibitors of the octopamine systems of arthropods.  相似文献

  • 1 Disease epizootics can significantly influence host population dynamics and the structure and functioning of ecological communities. Sarcoptic mange Sarcoptes scabiei has dramatically reduced red fox populations Vulpes vulpes in several countries, including Britain, although impacts on demographic processes are poorly understood. We review the literature on the impact of mange on red fox populations, assess its current distribution in Britain through a questionnaire survey and present new data on resultant demographic changes in foxes in Bristol, UK.
  • 2 A mange epizootic in Sweden spread across the entire country in < 10 years resulting in a decline in fox density of up to 95%; density remained lowered for 15–20 years. In Spain, mange has been enzootic for > 75 years and is widely distributed; mange presence was negatively correlated with habitat quality.
  • 3 Localized outbreaks have occurred sporadically in Britain during the last 100 years. The most recent large‐scale outbreak arose in the 1990s, although mange has been present in south London and surrounding environs since the 1940s. The questionnaire survey indicated that mange was broadly distributed across Britain, but areas of perceived high prevalence (> 50% affected) were mainly in central and southern England. Habitat type did not significantly affect the presence/absence of mange or perceived prevalence rates. Subjective assessments suggested that populations take 15–20 years to recover.
  • 4 Mange appeared in Bristol's foxes in 1994. During the epizootic phase (1994–95), mange spread through the city at a rate of 0.6–0.9 km/month, with a rise in infection in domestic dogs Canis familiaris c. 1–2 months later. Juvenile and adult fox mortality increased and the proportion of females that reproduced declined but litter size was unaffected. Population density declined by > 95%.
  • 5 In the enzootic phase (1996–present), mange was the most significant mortality factor. Juvenile mortality was significantly higher than in the pre‐mange period, and the number of juveniles classified as dispersers declined. Mange infection reduced the reproductive potential of males and females: females with advanced mange did not breed; severely infected males failed to undergo spermatogenesis. In 2004, Bristol fox population density was only 15% of that in 1994.

Density-dependent processes may have multiple effects on populations, which among other things include the regulation of population abundance and of the relative distribution of life-cycle stages within populations. The epiphytic habitat is often characterized as highly ephemeral and therefore epiphytic orchid populations may never achieve density-dependent regulation. In this study, we investigated the potential for density-dependent regulation in epiphytic and lithophytic orchids by examining the association between seedlings, juvenile and adult life-history stages in the Caribbean endemic orchid,Lepanthes rupestris in a cross-sectional study of 179 populations surveyed in the Luquillo National Forest along a riparian area where it is locally abundant. Under density-dependent regulation we expected a negative association between the ratio of seedling/adults and juveniles/adults and total population density. Population density was in the range of 140 individuals per m2, however patch sizes were small and mostly limited to less than 0.5 m2 with a maximum of 3 m2. We found no evidence of reduction of the ratio of seedlings or juveniles to adults as population size increased in either tree or boulder populations suggesting negative density dependence for population regulation inL. rupestris is either rare or occurs at even higher densities than those measured here. Moreover, we found positive (although weak) relationship between the ratio of seedlings and juveniles to adults and population size, suggesting that facilitation may be occurring.  相似文献

The potential acaricidal properties of an Ailanthus altissima bark extract were assessed against two common species of animal ectoparasitic mites, Psoroptes cuniculi and Sarcoptes scabiei var. cuniculi, in vitro. A. altissima bark extract was obtained by ethanol thermal circumfluence and tested at four concentrations (1.0, 0.5, 0.25 and 0.125 g/ml) on mites collected from rabbits. Compared to the fenvalerate treatment group, the A. altissima bark exhibited significant acaricidal properties for both mite species treated. The extract of concentrations of 1.0, 0.5 and 0.25 g/ml killed all tested S. scabiei within 7 h, however, only 1.0 and 0.5 g/ml of extract killed all treated P. cuniculi. The median lethal time (LT50) values at 1, 0.5 and 0.25 g/ml were 0.60, 0.78, 1.48 h for S. scabiei and 0.74, 1.29, 3.33 h for P. cuniculi. The median lethal concentration (LC50) for P. cuniculi was approximately 1.6 times that for S. scabiei var. cuniculi at 4 h. The extract showed stronger toxicity against S. scabiei than against P. cuniculi. Mortality rates increased with increasing concentration of extract administered and with increasing time post-treatment, indicating that the acaricidal activity of A. altissima bark extract is both time-dependent and dose-dependent. This is the first report on acaricidal activity of A. altissima against P. cuniculi and S. scabiei var. cuniculi. It indicates that A. altissima contain potential acaricidal compounds. Our study is the first step to develop potentially novel compounds from A. altissima for the effective control of mites in livestock.  相似文献

Three red foxes (Vulpes vulpes) were experimentally infected with Sarcoptes scabiei isolated from a naturally infected wild red fox. A fourth red fox served as a control. The first signs of sarcoptic mange became evident on the 31st day post infection (dpi). The signs gradually increased thereafter and between dpi 49 and 77 characteristic lesions of hyperkeratosis developed. Two of the infected foxes developed severe sarcoptic mange, and one of these animals died on dpi 121. The third fox developed a chronic hyperkeratotic lesion on its back, at the site where the mites had been applied. On dpi 127 the surviving foxes were treated systemically with ivermectin, and within 4 weeks the skin lesions had healed except on the pinnae of one animal. Antibodies to S. scabiei var. vulpes were demonstrated in the infected foxes by an ELISA with which seroconversion was seen around 4 weeks post infection (wpi). Western blot analysis of sequential sera of the infected animals demonstrated antibody activity consistently after the 2nd wpi. All life-stages ofSarcoptes scabiei var.canis survive in the hosts' environment for several days to several weeks depending on r.h. and temperature. Survival of larvae was comparable to males; survival of nymphs was comparable to females. Females and nymphs generally survived longer than larvae and males.Low temperature (10–15°) and high r.h. prolonged survival of all life stages. At 10–15°C, females and nymphs survived 1–3 weeks at 97% r.h., 1–2 weeks at 75% r.h. and 5–8 days at 45% r.h. At 20–25°C, survival was significantly reduced but all life-stages survived at least 2 days at 25% r.h. and 5–6 days at 75–100% r.h. Long survival off the host coupled with host-seeking behavior of these mites make it likely that environmental contamination is a source of scabies in domestic and wild mammals, and in humans.  相似文献

An ultrastructural study of life cycle stages of Pneumocystis carinii in infected rat lungs in situ was undertaken utilizing 8 different modes of fixation. Three of the fixatives employed gave good fixation of cysts and intracystic bodies, but for the trophic forms fixation was only fair. Both the trophic forms and intracystic bodies have nuclear pores. The mitochondria of the organism have cristae that appear lamellar. One of the fixation modes revealed a thin, electron-dense layer on the outer surface of the cell wall, a "fuzzy coat" that had not been described previously. This material appears to mediate tight adhesion of trophic forms with other trophic forms, cysts, and with pneumocytes of the lung alveolus.  相似文献

Sarcoptes scabiei (Acari: Sarcoptidae) causes a common contagious skin disease that affects many mammals. Here, the complete mitochondrial genome of a mite, S. scabiei var. nyctereutis, from Japanese wild raccoon dogs was analyzed. The 13,837 bp circular genome contained 13 protein-coding genes, two rRNA genes, and 22 tRNA genes. For the first time, two tRNAs (alanine and tyrosine), that were thought to be absent in scabies mites from other animals, were predicted to have short, non-cloverleaf structures by in silico annotation and detected by RT-PCR, sequencing, and northern analysis. The mitochondrial genome structure of S. scabiei is similar to that of Psoroptes cuniculi and Dermatophagoides farinae. While small and unusual tRNA genes seem to be common among acariform mites, further experimental evidence for their presence is needed. Furthermore, through an analysis of the cox1 gene, we have provided new evidence to confirm the transmission of this mite between different animal hosts.  相似文献

Hatched second stage juveniles of Globodera rostochiensis were found to be more sensitive than either unhatched juveniles or adult males to low concentrations of oxamyl (EC50 value for activity 0.5 μg oxamyl ml-1). Development of juveniles in roots was significantly inhibited by 2.0 μg oxamyl ml-1 probably due to impairment of normal feeding activity. The behavioural events necessary for orientation of juveniles towards roots for invasion and orientation of males towards females for fertilisation were found to be impaired by 0.5 and 1.0 μg oxamyl ml-1 respectively. A comparison with previous work on Meloidogyne incognita showed G. rostochiensis juveniles to be more sensitive to oxamyl.  相似文献

Genome ploidy in different stages of the Giardia lamblia life cycle   总被引:2,自引:0,他引:2  
The early diverging eukaryotic parasite Giardia lamblia is unusual in that it contains two apparently identical nuclei in the vegetative trophozoite stage. We have determined the nuclear and cellular genome ploidy of G. lamblia cells during all stages of the life cycle. During vegetative growth, the nuclei cycle between a diploid (2N) and tetraploid (4N) genome content and the cell, consequently, cycles between 4N and 8N. Stationary phase trophozoites arrest in the G2 phase with a ploidy of 8N (two nuclei, each with a 4N ploidy). On its way to cyst formation, a G1 trophozoite goes through two successive rounds of chromosome replication without an intervening cell division event. Fully differentiated cysts contain four nuclei, each with a ploidy of 4N, resulting in a cyst ploidy of 16N. The newly excysted cell, for which we suggest the term 'excyzoite', contains four nuclei (cellular ploidy 16N). In a reversal of the events occurring during encystation, the excyzoite divides twice to form four trophozoites containing two diploid nuclei each. The formation of multiple cells from a single cyst is likely to be one of the main reasons for the low infectious doses of G. lamblia .  相似文献

The petroleum ether extract of neem oil and its four fractions separated by column chromatography was diluted at different concentrations with liquid paraffin. The acaricidal bioassay was conducted using a dipping method. The results indicated that the median lethal concentration (LC50) of the petroleum ether extract (at the concentration of 500.0ml/l) was 70.9ml/l, 24h after treatment. At concentrations of 500.0, 250.0, 125.0, 62.5 and 31.2ml/l, the median lethal times (LT50) of the petroleum ether extract were 8.7, 8.8, 10.8, 11.5 and 13.1h, respectively. Thin-layer chromatography (TLC) showed that the petroleum ether extract of neem oil separated into four fractions (F1-F4). Acaricidal activity of 68.3% and 100.0% in the F2 and F4 was confirmed. These results suggest that petroleum ether extracts of neem oil and its four fractions possess useful acaricidal activity in vitro.  相似文献
