Molecular markers [random amplified polymorphic DNA (RAPD) and restriction fragment length polymorphism (RFLP)] were used to determine the frequency of DNA polymorphism in grain sorghum (Sorghum bicolor (L.) Moench). Twenty-nine oligonucleotide primers were employed for RAPDs, generating a total of 262 DNA fragments, of which 145 were polymorphic in at least one pairwise comparison between 36 genotypes. Individual primers differed significantly in their ability to detect genetic polymorphism in the species. The overall frequency of polymorphisms was low with a mean frequency of 0.117 polymorphisms per RAPD band being obtained from all pairwise comparisons between genotypes, with maximum and minimum values of 0.212 and 0.039, respectively. Results from phenetic analysis of bandsharing data were consistent with current sub-specific groupings of the species, with clusters of Durra, Zerazera, Caud-Nig, Caud-Kaura and Caffrorum being discernible. The results also indicated that individuals of a similar taxonomic grouping but different geographic origin may be genetically less identical than previously considered. Similar frequencies of polymorphism to that obtained with RAPDs were obtained with RFLPs. Results from these experiments indicated that a high level of genetic uniformity exists within S. bicolor.  相似文献

The sequence of pollen development from the tetrad stage to the mature tricellular grain was studied in freshly harvested anthers of Sorghum bicolor. This pattern of development was then compared with that occurring during panicle pretreatment and subsequent anther incubation in vitro. It was found that during pretreatment at 7° C mitoses of the vegetative cell were induced in up to 30% of the pollen. During anther incubation procallus development was highly polarised with contributions from both the generative and vegetative cells. After pretreatment at 14 or 20° C the generative cell became detached from the pollen wall and it was not possible to determine whether subsequent development involved only the vegetative cell or both the vegetative and generative cells.Although retarded pollen grains were observed both in vivo and in vitro, and were occasionally seen to divide in culture, they did not appear to be the source of the procalluses produced.  相似文献

The effect of various abiotic stresses on germination rate, growth and soluble sugar content in Sorghum bicolor (L.) Moench cv. CSH 6 seed embryos and endosperm during early germination was investigated. Under stress conditions germination, water potential and tissue water content decreased markedly. Subsequently, this reduction resulted in marked decreases in fresh weight both in embryos and endosperm. Conversely, a substantial increase in dry weight was observed. Furthermore, a considerable increase in the sugar contents in both embryo and endosperm was detected. The fructose level was always higher than glucose and sucrose in response to various stresses. However, as compared to the control the level of glucose and sucrose was higher in embryos and endosperm after stress treatments. Based upon these results a possible physiological role of sugars in the germination of sorghum seeds is discussed.  相似文献

Summary Restriction endonuclease patterns of chloroplast DNA (cpDNA) were consistently distinguishable between fertile and male-sterile cytoplasms of sorghum [Sorghum bicolor (L.) Moench], whereas no differences in restriction patterns of cpDNA among male-sterile (A1) lines, including six isocytoplasmic strains, were revealed in this study. Summary Foreign pollen tubes in the stigma of Zea mays can be prevented from reaching the ovary cavity by the unusual length of the pollen tube pathway. A simple and rapid procedure is described for overcoming this difficulty by pollinating the basal parts of the stigmas without removing the ensheathing bracts (husks). The method maintains high humidity in the vicinity of the ovaries, and by conserving photosynthetic tissues probably also ensures a more normal O2 /CO2 balance in the neighbourhood of the stigmas than do bagging procedures. It is shown that Sorghum pollen tubes readily reach the ovary after pollination by the method. Their presence induces some of the characteristic post-pollination effects caused by Zea pollen tubes, but they frequently also stimulate premature enlargement of the nucellus and lysis of nucellar cells. Although Sorghum tubes have been traced across the inner ovary wall, they have not been seen to enter the micropyle, and hybrid embryos have not yet been obtained.  相似文献

Summary A comparison of soluble protein, esterase, GDH and ADH isoenzyme patterns in seeds of different steriles, maintainers and restorer lines exhibited similarities as well as differences. Soluble protein patterns from sterile and maintainer lines differed both qualitatively and quantitatively. Based on the esterase patterns, male steriles with different cytoplasms could be separated into three groups (i) Ck 60A and B; Nagpur A and B, (ii) M 35-1A and 1 B, M 31-2A and 2B, (iii) G1A and B, VZM2A and 2B. Each group could further be differentiated on the basis of minor differences in esterase isoenzyme patterns within each group. Forty-five sorghum germplasm growing in the Eastern Highlands of Ethiopia were evaluated for 10 qualitative traits. Phenotypic frequencies between the accessions from each of the nine Aanaas and Alemaya University, grouped in 10 localities were tabulated. Phenotypic diversity index, H′, was analysed and the result indicated the between localities component of diversity to be relatively smaller than the variation in H′ among characters within localities. The value of H′ for all sample germplasm ranged from 0.36 to 0.95 with a mean of 0.71. The results showed that there is a wide morpho-agronomical diversity among the sample germplasm studied. This information can be used for the conservation of these germplasm resources and future improvement work of the sorghum crop.  相似文献

Four seed lots of sorghum cultivar IS 34213 were obtained which had been harvested early or at full maturity and then dried either swiftly in the sun or slowly in the shade. The earlier harvested seeds were lighter, smaller and of lower density. Earlier harvested seeds had pericarps which permitted faster water uptake and greater leakage of electrolytes. Water uptake and electrolyte leakage were positively correlated. The method of seed drying had no effect on water uptake or leakage. Greater leakage was associated with lower final germination. The results are interpreted in relation to germination testing.  相似文献

The objective of this work was to study the effect of two concentrations (10 and 100 mg kg−1) of phenanthrene, a ubiquitous polycyclic aromatic hydrocarbon (PAH), on root exudation of the remediating plant Sorghum bicolor (L.) Moench under controlled conditions in a pot experiment. It was found that the phenanthrene concentration of 10 mg kg−1 did not cause significant effects on plant survival and growth but had little stimulating effect on carbohydrate exudation. The contamination with phenanthrene at 100 mg kg−1 inhibited accumulation of plant shoot and root biomass, decreasing the carboxylic acid, carbohydrate, and amino acid amounts released by sorghum root into the rhizosphere. However, root exudation per unit of root surface was not changed significantly with increasing phenanthrene concentration. There were no differences in qualitative composition of root exudates under the influence of PAH were found. The observed alterations in the ratio between the main root-exuded components are assumed to manifest adaptive alterations occurring in the plant as a response to pollutant stress. The activity of three oxidoreductases (oxidase, peroxidase, and tyrosinase) released by sorghum roots was clearly progressive to the increasing phenanthrene concentration in the substrate. Under the influence of phenanthrene, the population of phenanthrene-degrading microorganisms in sorghum root zone increased, and their share in the total number of culturable heterotrophs increased as well. The main promotional factor was the pollutant; however, the stimulating effect of the plant root exudates was also involved. The increased pollutant-degrading microbial population and activity of the extracellular root enzymes are presumed to be important for the rhizodegradation of PAH.  相似文献

Summary Amino acid composition of proteins from anthers of milo and Indian origin male steriles were determined. Comparison of amino acid between A and B lines showed lower contents of histidine, threonine, glutamic acid, glycine, leucine and phenylalanine and higher contents of alanine, serine, proline and tyrosine in line A compared to line B. Alanine content in anthers of A lines was more than two fold higher than that in the anthers from B lines. Marked differences in amino acid composition of anthers of A and B lines are suggestive of their involvement in male sterility. Cytoplasmic male steriles of Indian origin M35-1A and M31-2A showed greater similarity but differed from milo, VZM2A and B.  相似文献

Abstract

In the present study, we used 12 genotypes of sorghum originated from different countries (five sweet, four grain and three forage). These different genotypes and types of sorghum were evaluated for the agro-morphological traits that are associated with the estimated sugar and bioethanol yield to estimate their phenotypic diversity. Analysis of variance showed significant differences between different types of sorghum for all the evaluated traits. Sweet sorghum genotypes, however, showed better performance with respect to all studied traits than the other genotypes. A positive significant correlation was observed between plant height, leaf number, leaf area, biomass yield, cane and bagasse yields, and the predicted bioethanol yield. Both, cluster and principal component analysis were performed to group the genotypes according to their agro-morphological and molecular similarity coefficients. For analytical approaches, the Iranian grain and forage genotypes clustered separately from the other genotypes. The clustering patterns obtained from the molecular dominant markers had higher discriminatory power than using morphological characters to separate sweet genotypes from the forage and grain sorghum ones. The results clearly indicated that sweet sorghum can be grown in Germany and maintains its superiority in biomass production and sugar yield over grain and forage sorghum types.  相似文献

Flooding evoked a differential response in the activities of enzymes of fermentation pathway in leaves and roots of flood sensitive (S-308) and flood-tolerant (SSG-59-3) cultivars of sorghum. Activities of alcohol dehydrogenase (ADH), lactate dehydrogenase (LDH) and alanine aminotransferase (AlaAT) enhanced in roots of SSG-59-3 during 72 h of flooding. In contrast, a transient increase in the activities was discerned in roots of S-308 up to 24 h flooding followed by a decline in activities of these enzymes. In leaves of SSG-59-3, the activities of ADH and LDH increased to about three fold during flooding stress as compared to that in the non-flooded control plants. Though elevation in activities of these enzymes was observed in leaves of S-308 up to 48 h of flooding, the magnitude of enhancement was much lower than that in SSG-59-3. Alanine aminotranferase activity depressed in leaves of both the cultivars but the level of decline was more pronounced in sensitive cultivar S-308 as compare to tolerant SSG-59-3. The amount of alcohol, lactic acid and alanine were higher in both roots and leaves of SSG-59-3 than that in S-308 during flooding stress. It is thus apparent that roots and leaves of flood tolerant variety tends to attain greater capacity to perform reactions of various fermentation pathways to sustain production of ATP under flooded conditions.  相似文献

Abstract

The scope of biocontrol agents in the control of sorghum grain molds was investigated both in vitro as well in vivo. Their utility in enhancing the germination and seedling vigor of mold-infected seeds were assessed. The biocontrol agents, Trichoderma viride, T. harzianum, T. hamatum, T. koeningii and Pseudomonas fluorescens (isolates collected from the state of Tamil Nadu, India) were efficient in checking the growth of mold pathogens when grown in Potato Dextrose Agar (PDA) as dual cultures. The bacterial bioagent P. fluorescens was most efficient in checking the growth of all the test pathogens viz., Fusarium moniliforme, Curvularia lunata and Alternaria alternata (66.8, 68.5 and 64.9% inhibition of colony growth respectively) followed by other bioagents for their antagonistic effect. The in vivo effectiveness of the bioagents were enhanced when combined with other components like host-resistance, cultural and chemical control strategies, paving the way for a possible Integrated Disease Management. The percent incidence and severity of molds were lowest (19.44 and 13.5% respectively) with the treatment T6 (seeds harvested at physiological maturity + propiconazole @ 0.02% + P. fluorescens @ 1 × 108 cfu ml?1) that recorded maximum 100 seed weight (2.905 g), followed by 2.871 g in T2 (seeds harvested at physiological maturity + P. fluorescens). The effect of bioagents in enhancing the germination and seedling vigor was most promising. Of all the treatments, P. fluorescens resulted in maximum germination (88%) and seedling vigor (2635.60 vigor index), followed by treatment with T. viride (86.6% and 2478.47 respectively).  相似文献

Community of arbuscular mycorrhizal (AM) fungi in a coastal vegetation on Okinawa island in Japan was examined. A sampling plot was established in a colony of Ipomoea pes-caprae (Convolvulaceae) on the beach in Tamagusuku, Okinawa Pref, in which eight root samples of I. pes-caprae and three root samples each of Vigna marina (Leguminosae) and Paspalum distichum (Poaceae) were collected. Partial 18S rDNA of AM fungi was amplified from the root samples by polymerase chain reaction (PCR) with primers NS31 and AM1. Restriction fragment length polymorphism analysis with HinfI and RsaI for cloned PCR products revealed that two types of Glomus sp., type A and type B, were dominant in the colony. Among them, the fungi of type A were especially dominant near the edge of the colony facing the sea. A phylogenetic analysis showed that the AM fungi of type B are closely related to Glomus intraradices and those of type A are nearly related to type B. From the sequence data, it was also found that type A was further divided into two types, type A1 and A2. One representative strain each of the three types, type A1, A2, and B, propagated from single spore each, was examined for the growth of sorghum (Sorghum bicolor) at three different salinity levels, 0, 100, and 200 mM NaCl. At the non-salt-treated condition, the type B fungus was the most effective on shoot growth enhancement of the host plant, whereas at the salt-treated conditions, the type A2 fungus was the most effective. An efficient suppression of Na + translocation into the shoot by the examined AM fungi was found. These results suggested that the AM fungi dominant near the sea are adapted to salt-stressed environment to alleviate the salt stress of host plants.  相似文献

The studies were carried out at Radzików, Central Poland in the years 1982–1991, using a progeny of an Indian population. It was proved that Catharanthus roseus may be grown in Poland under field conditions as an annual crop, using plants previously raised in glass or plastic houses. However, the culture of C. roseus in the greenhouse resulted in a higher yield of herb and particularly in a better setting of fruits and seeds than under field conditions. The yield of seeds obtained under cover was several times higher than in the field. Growing of plants under cover may be justified, as greenhouses usually are not utilised in the summertime. Consequent mass selection conducted during successive seasons has lead to obtaining the lines better adapted to the conditions of a cool climate and hence more productive. Improvement of generative traits, resulting from selection, was more pronounced than improvement of the vegetative indices. The ‘yellow silique’ has been recognised as an optimal stage for harvest, as at this stage siliques could be easily separated from the stems, whereas the risk of seed shedding was still minimal. Germinability of seeds obtained from siliques harvested at this stage was high and uniform.  相似文献

Poor emergence of maize seedlings, due to high soil temperatures, is a major limitation of crop potential in the lowland tropics. Ability to germinate at high temperature (>c. 37° C) is related to the temperature sensitivity of the embryo, and there is considerable genotypic variation for this character.Respiration and mitochondrial phosphorylation proceed normally in seeds imbibing at 41° C, and ATP levels are adequate for germination. However, the specific activities of several important enzymes are lower, and the rate of protein synthesis is severely reduced compared with seeds imbibing at 28° C. The depression of the rate of protein synthesis in the embryos of several tropical hybrids imbibing at high temperature correlated with their known temperature sensitivity. Some effects of methyl jasmonate (Me-Ja) on sunflower (Helianthus annuus L.) seed germination and seedling development are described and compared with those of ABA. Both growth regulators have very similar action. They inhibit germination, but high concentrations of O2 in the atmosphere suppress this inhibitory action. Depending on the concentration, Me-Ja inhibits root and hypcotyl growth, however the root is more sensitive to Me-Ja than to ABA. Me-Ja also strongly reduces oxygen uptake during germination and inhibits chlorophyll biosynthesis in isolated cotyledons.  相似文献

The N-substituted phthalimide AC 94377 (1-(3-chlorophthalimido)-cyclohexanecarboxamide) was equally effective as a mixture of the gibberellins A4 and A7 (GA4/7) in breaking dormancy and stimulating germination of celery seeds when either was used in combination with ethephon or daminozide as a seed soak. Whereas seedlings emerging from GA4/7-treated seeds became etiolated in comparison with those from untreated seeds, those from AC 94377-treated seeds showed normal development. Preharvest sprays of gibberellic acid (GA3) increased the height of mature plants in comparison with untreated controls by about 16 per cent whereas AC 94377 was ineffective. The yield from GA3-treated plots was about 10 per cent greater than that from AC 94377-treated plots.  相似文献
