An account of gonadal morphology and reproductive traits of the Amazonian arapaimatid, Arapaima gigas (Schinz 1822), is given. The only functional ovary (left) was foliaceous in shape having the ventral surface without an ovarian capsule. The absence of the ovarian capsule ventral portion, absence of oviduct and funnel‐shaped coelomic cavity are features that imply that oocytes are freed in the coelomic cavity before being shed to the aquatic environment through a genital papilla. The only functional testis (left) was cord‐like with a maximum diameter of 1–1.5 cm and was connected to the genital papilla through a spermatic duct. Size at sexual maturity was 145–154 cm and 115–124 cm, total length class, for females and males, respectively.  相似文献

Most nests of Arapaima gigas in floodplains of the Amazon were built at the margins of the forests surrounding temporary or permanent lakes and their connecting channels. They were found under forested levees in locations that were shallow and sandy. From 2000 to 2005, the adult population increased six-fold but nest density increased only two-fold.  相似文献

This study evaluated the influence of diets supplemented with 500, 800, 1200 mg kg-1 of vitamin C (ascorbic acid or AA) and vitamin E (alpha-tocopherol or alpha-T) on the physiological responses of pirarucu fed for 2 months. Weight and mortality were not affected by dietary vitamin type or their concentrations. Significant increase (p<0.05) on the red blood cells count was obtained on treatments with 800 and 1200 mg AA kg-1 and on the hemoglobin concentration on treatment with 500 mg alpha-T kg-1 relatively to control. Mean corpuscular volume presented a significant decrease (p<0.05) on treatment with 800 and 1200 mg AA kg-1 when compared to control. Mean corpuscular hemoglobin concentration was significantly high (p<0.05) on treatment with 500 mg alpha-T kg-1. Only in vitamin C treatments, we noticed a significant increase (p<0.05) in the number of leucocytes relative to control. All fish in the vitamin-supplemented treatments, except 500 mg AA kg-1, had high total protein values compared to control. Fish treated with 800 or 1200 mg alpha-T kg-1 also showed increases in plasma glucose concentrations. Our results suggest that 800 and 1200 mg AA kg-1 are probably the most suitable concentrations for pirarucu diets, although high vitamin E diets are not necessary for quantitative leucocyte increases for this species.  相似文献

The sexual differentiation of Salaria (= Blennius ) pavo is described from the stage of hatching to a body length of 35 mm. At hatching, the primordial germ cells (PGCs) can be recognized clearly. At a standard body length of 5 mm, they begin to protrude into the peritoneal cavity and at 14 mm they transform to oogonia. At 17 mm length, the first oocytes can be observed. In males at a standard length of 16–17 mm, the first signs of a differentiation into a testis can be recognized. Shortly after the differentiation of the male sex, the division of the male gonad into a testis and a testicular gland can be seen. The fine structural characteristics of the PGCs and of differentiation stages are presented.  相似文献

The Nile tilapia is one of the most important fish species for aquaculture worldwide. Understanding their reproductive biology is essential for improving their aquaculture methods. The morphological and quantitative dynamics of ovarian recrudescence of Oreochromis niloticus was studied for 21 days postspawning. To accomplish this, breeding females were kept in controlled conditions and ovarian samples were collected weekly for histological, ultrastructural and morphometric analyses. Ovarian follicle morphology revealed an intense synthesis activity of the follicular cells, which actively contributed to formation of the zona radiata and oocyte development following spawning. Recently spawned ovaries contained follicles at all developmental stages, but they were predominantly early primary growth (~42%) and full‐grown follicles (~20%). Remnants of spawning, postovulatory follicle complexes represented approximately 5% of the former ovarian follicles immediately after spawning, and less than 1% after 7 days. Atretic follicles accounted for approximately 2% of the follicles studied during the period. The stock of primary growth follicles was stable during ovarian recrudescence, indicating their availability for continuous recruitment. Only the frequency of full‐grown follicles significantly increased in the ovaries during recrudescence, representing approximately 35% of the follicles 21 days postspawning. The diameters of all follicles were significantly different between the periods analyzed. The ovaries' morphological characteristics, the maintenance of young follicles stocks and the gradual and significant increase in the proportion and diameter of full‐grown follicles showed a rapid ovarian recovery and follicular growth of O. niloticus, in 21 days at 29.5°C, necessary for the next spawning. Rad54 is an important factor in the homologous recombination pathway of DNA double-strand break repair. However, Rad54 knockout (KO) mice do not exhibit overt phenotypes at adulthood, even when exposed to radiation. In this study, we show that in Rad54 KO mouse the germline is actually altered. Compared with the wild-type (WT) animals, these mice have less premeiotic germ cells. This germ cell loss is found as early as in E11.5 embryos, suggesting an early failure during mutant primordial germ cells development. Both testicular and ovarian KO germ cells exhibited high radiation sensitivity leading to a long-term gametogenesis defect at adulthood. The KO female germline was particularly affected displaying decreased litter size or sterility. Spermatogenesis recovery after irradiation was slower and incomplete in Rad54 KO mice compared with that of WT mice, suggesting that loss of germ stem cell precursors is not fully compensated along the successive rounds of spermatogenesis. Finally, spermatogenesis recovery after postnatal irradiation is in part regulated by glial-cell-line-derived neurotrophic factor (GDNF) in KO but not in irradiated WT mice, suggesting that Sertoli cell GDNF production is stimulated upon substantial germ cell loss only. Our findings suggest that Rad54 has a key function in maintaining genomic integrity of the developing germ cells.  相似文献

A study on the reproductive status of Belonogaster juncea juncea individuals was conducted in Cameroon on 21 pre- and 15 post-emergence colonies. We compared the females’ size and ovarian development, and verified if they were inseminated or not. Dominant females were inseminated, had well developed ovaries, and were the largest individuals in pre-emergence colonies; in post-emergence colonies their size did not differ significantly from that of non-dominant females. Most subordinate females of both pre- and post-emergence colonies had thread-like ovaries, but a few had well developed ovaries. In pre-emergence colonies 13 subordinates had thread-like ovaries and several yellow bodies indicating that they had already laid eggs. We deduced that subordinates with well developed ovaries joined the foundations shortly before our study, while those with several yellow bodies had arrived some time before and their ovaries had regressed. At the end of the post-emergence phase appear non-dominant females with well developed ovaries. They are the future foundresses. Some females can disperse just after emergence and join a new foundation without mating. Nevertheless, most co-foundresses were inseminated when they joined the foundations, and so had the potential to reproduce, negating the hypothesis of swarm founding in B. j. juncea. The gene germ cell-less (gcl) plays an important role in the early differentiation of germ cells in Drosophila. We isolated the gcl homolog of the model teleost medaka (Oryzias latipes) using degenerated primers and an ovary cDNA bank. The predicted amino acid sequence of medaka gcl showed 92, 68 and 31% overall identity to mouse, human and Drosophila gcl respectively. RT-PCR revealed stronger expression in the ovary and weaker expression in testis, brain, heart, liver and muscle tissue. Expression in early embryos indicates the presence of maternal mRNA. By in situ hybridisation (ISH), gcl could not be detected in embryos. In contrast to vasa, ISH revealed expression of gcl in the ovary but not in the testis. Mol. Reprod. This study describes the structural and ultrastructural characteristics of gonadal sex differentiation and expression of Vasa, a germline marker, in different developmental stages of embryos and newborn fry of the barred splitfin Chapalichthys encaustus, a viviparous freshwater teleost endemic to Mexico. In stage 2 embryos, the gonadal crest was established; gonadal primordia were located on the coelomic epithelium, formed by scarce germ and somatic cells. At stage 3, the undifferentiated gonad appeared suspended from the mesentery of the developing swimbladder and contained a larger number of germ and somatic cells. At stages 4 and 5, the gonads had groups of meiotic and non-meiotic germ cells surrounded by somatic cells; meiosis was evident from the presence of synaptonemal complexes. These stages constituted a transition towards differentiation. At stage 6 and at birth, the gonad was morphologically differentiated into an ovary or a testis. Ovarian differentiation was revealed by the presence of follicles containing meiotic oocytes, and testicular differentiation by the development of testicular lobules containing spermatogonia in mitotic arrest, surrounded by Sertoli cells. Nuage, electron-dense material associated with mitochondria, was observed in germ cells at all gonadal stages. The Vasa protein was detected in all of the previously described stages within the germ-cell cytoplasm. This is the first report on morphological characteristics and expression of the Vasa gene during sexual differentiation in viviparous species of the Goodeidae family. Chapalichthys encaustus may serve as a model to study processes of sexual differentiation in viviparous fishes and teleosts.  相似文献

Translin associated factor X (TRAX) is a binding partner of TB-RBP/Translin. A cDNA encoding the 260 C-terminal amino acids of KIF2Abeta was isolated from mouse testis cDNAs in a yeast two-hybrid library screen for specific TRAX-interacting proteins. KIF2Abeta was expressed predominantly in the mouse testis and enriched in germ cells. The interaction of full-length KIF2Abeta or its C-terminus with TRAX was verified using in vitro synthesized fusion proteins. Deletion mapping of the TRAX-binding region of KIF2Abeta indicated that amino acids 514-659 were necessary and sufficient for the interaction in vivo. Confocal microscopy studies using GFP-fusion proteins demonstrated that KIF2Abeta colocalizes with TRAX in a perinuclear location. KIF2Abeta does not interact with TB-RBP, suggesting that either TRAX can function as an adaptor molecule for motor proteins and TB-RBP, or that this interaction reveals an undescribed role for TRAX in germ cells. The interaction with KIF2Abeta suggests a role for TRAX in microtubule-based functions during spermatogenesis.  相似文献

In the early juveniles of Ciona intestinalis, primordial germ cells arise on the degenerated mass of the resorbed tadpole tail, and assemble to form a discrete gonad rudiment. The present study elucidated the morphological sequences during differentiation of the gonad rudiment into the testis and ovary. In 11- to 12-day juveniles, the gonad rudiment, an elongate sac, divided into the testicular and ovarian rudiments. The testicular rudiment separated as a round vesicle from the thickened wall of the elongate sac. The original sac, after separation of the round vesicle, developed into the ovary. In the testicular rudiment, germ cells formed a continuous central mass without association of somatic cells, while in the ovarian rudiment, each germ cell was associated with somatic cells within the epithelium composing the wall of the rudiment. In 13- to 15-day juveniles the testicular rudiment changed into branched tubes ending in club-shaped follicles. Cells characterized by many flattened cisternae of rough endoplasmic reticulum (distal cells) constituted the distal wall of each follicle. Spermatogenic cells were freely present in the follicular lumen, but the largest spermatogonia were in contact with the distal cells. Both in the testicular and ovarian rudiments, germ cells entered meiosis in 18-day juveniles. A novel body (periesophageal body) was found just beneath the ventral margin of the esophageal opening. It comprised irregular follicles made up of one cell type whose cytoplasm, filled with round vesicles and Golgi complexes, was suggestive of an endocrine function. Fragments derived from the periesophageal body were present around the developing ovary.  相似文献

Triploidy is a viable condition in teleosts. However, in many salmonids, the triploid condition in the female results in sterility as gametogenesis appears to be disrupted. Although the underlying mechanisms regulating the gonadal development of teleosts have not been clearly elucidated, the reversal of phenotypic sex by the administration of the appropriate exogenous steroid during early development supports the argument that gonadal steroids play a pivotal role in sexual differentiation and subsequent gonad development in these fish. To determine whether the failure of normal ovarian development in triploid female rainbow trout (Oncorhynchus mykiss) is due to an absence or reduction of endogenous sex steroids, ovarian morphology was compared between diploid and triploid juvenile rainbow trout treated with exogenous estradiol-17β (E2). The ovaries of both untreated and E2 treated diploid fish, at 145 days post-fertilization, contained synchronously developing oocytes in the perinucleolar stage, whereas ovaries from untreated and estradiol-treated triploid fish of the same age were considerably smaller and devoid of developing oocytes. No differences in the ovaries of triploid untreated fish and triploid fish treated with E2 were observed. It is reported that exposure to exogenous E2 during the period of gonadal differentiation is not sufficient to induce oocyte development in triploid rainbow trout. © 1996 Wiley-Liss, Inc.  相似文献

The germ cell lineage is first recognized as a population of mitotically proliferating primordial germ cells that migrate toward the gonadal ridge. Shortly after arriving at the gonadal ridge, the germ cells begin to initiate a commitment to gamete production in the developing gonad. The mechanisms controlling this transition are poorly understood. We recently reported that a mouse germ cell nuclear antigen 1 (GCNA1) is initially detected in both male and female germ cells as they reach the gonad at 11.5 days postcoitum (dpc). GCNA1 is continually expressed in germ cells through all stages of gametogenesis until the diplotene/dictyate stage of meiosis I. Since GCNA1 expression commences soon after primordial germ cells arrive at the gonadal ridge, we wanted to determine whether the gonadal environment was essential for induction of GCNA1 expression. By examining GCNA1 expression in germ cells that migrate ectopically into the adrenal gland, we determined that both the gonadal and adrenal gland environments allow GCNA1 expression. We also examined GCNA1 expression in Ftz-F1 null mice, which are born lacking gonads and adrenal glands. During embryonic development in the Ftz-F1 null mice, the gonad and most germ cells undergo apoptotic degeneration at about 12.5 dpc. While most of the germ cells undergo apoptosis without expressing GCNA1, a few surviving germs cells, especially outside the involuting gonad clearly express GCNA1. Thus, although the Ftz-F1 gene is essential for gonadal and adrenal development, induction of GCNA1 expression in germ cells does not require Ftz-F1 gene products. The finding that germ cell GCNA1 expression is not restricted to the gonadal environment and is not dependent on the Ftz-F1 gene products suggests that GCNA1 expression may be initiated in the germ cell lineage by autonomous means. The present study describes the morphology and ultrastructural features of postnatal follicular development in the volcano mouse ( Neotomodon alstoni alstoni ), an endemic Mexican rodent. By the first week of age, germ cells were organized in clusters within the ovigerous cords, and only 51.8% of them were associated with somatic cells. At the ultrastructural level, pairing chromosomes and cellular junctions between germ and pregranulosa cells, such as desmosomes, were observed. At this time, the zona pellucida could not be detected in the formed follicles. From 15 to 28 days postpartum, growing follicles were located at the medulla and inner cortex of the ovary, but most were atretic. The first preovulatory follicles were seen at 40 days. Likewise, corpora lutea were observed at this stage of development, which shows that the volcano mouse is a spontaneous ovulator. The follicular development of the volcano mouse shows strong similarities with that of the golden hamster, particularly during the first week. The morphological changes observed during postnatal follicular development of the volcano mouse follow the same general histological pattern as reported for other mammals, although the timing of these events is species-specific.  相似文献

To evaluate the possible role of germ cells on sex differentiation of the gonads in vertebrates, the teleost fish, medaka ( Oryzias latipes ), was used to generate a gonad without germ cells. The germ cell-deficient medaka reveals multiple effects of germ cells on the process of sex differentiation. The previously isolated mutant medaka, hotei , with the excessive number of germ cells may support the contention that the proliferation of germ cells is related to feminization of the gonad. Futhermore, we show that two modes of proliferation for either maintenance of germ cells or commitment to gametogenesis are important components of the sex differentiation of medaka developing gonads. An intimate cross talk between germ cells and gonadal somatic cells during the sex differentiation will be discussed.  相似文献
