根据南极假丝酵母脂肪酶B (CALB)的基因序列将CALB基因进行TA克隆、酶切鉴定及测序后,亚克隆至大肠杆菌-乳酸乳球菌穿梭表达栽体pMG36e-Nisl中,构建重组表达栽体pMG36e-Nisl-CALB.设计特异性引物P3和P4,对重组质粒pMG36e-NisI-CALB进行红霉素抗性基因的敲除,以构建食品级表达载体pMG36N-CALB,后再将两种重组质粒分别电转化入乳酸乳球菌MG1363,以Nisin为选择压力,考察CALB在MG1363中的表达情况.结果显示,成功构建了表达载体pMG36e-NisI-CALB及pMG36N-CALB,两株重组菌在含有20 IU Nisir/mL的培养基中均生长情况良好,遗传性能稳定,且经水解圈鉴定,CALB能够进行活性表达.进一步研究发现,CALB基因整合到乳酸乳球菌MG1363染色体中.  相似文献

The vancomycin-resistant enterococci GV1, GV2 and GV3, which were isolated from droppings from broiler farms in Japan have been characterized as VanA-type VRE, which express high-level vancomycin resistance (256 or 512 microg ml(-1), MIC) and low-level teicoplanin resistance (1 or 2 microg ml(-1), MIC). The vancomycin resistances were encoded on plasmids. The vancomycin resistance conjugative plasmid pMG2 was isolated from the GV2 strain. The VanA determinant of pMG2 showed the same genetic organization as that of the VanA genes encoded on the representative transposon Tn1546, which comprises vanRSHAXYZ. The nucleotide sequences of all the genes, except the gene related to the vanS gene on Tn1546, were completely identical to the genes encoded on Tn1546. Three amino acid substitutions in the N-terminal region of the deduced VanS were detected in the nucleotide sequence of vanS encoded on pMG2. There were also three amino acid substitutions in the vanS gene of the GV1 and GV3 strains in the same positions as in the vanS gene of pMG2. Vancomycin induced the increased teicoplanin resistance in these strains.  相似文献

Drug resistance and the transferability of resistance were examined in 218 Enterococcus faecium clinical isolates obtained from in-patients of a Japanese university hospital between 1990 and 1999. One hundred and sixty one isolates (73.9%) were drug-resistant and 127 (58.2%) isolates were resistant to two or more drugs. Vancomycin resistant E. faecium (VRE) was not isolated. The transferability of drug-resistance to an E. faecium strain was examined by broth or filter mating. Six (12.5%) of the 48 gentamicin resistance traits, and fifty (50%) of the 101 erythromycin resistance traits were transferred by filter mating. The gentamicin resistance traits of five isolates and the erythromycin resistance traits of four isolates were transferred to the recipient strains by both broth mating and filter mating at a frequency of about 10(-6) and 10(-5) per donor cell, respectively. The five gentamicin resistant strains were shown to harbor pMG1-like plasmids on the basis of their Southern hybridization with pMG1 (65.1 kbp, Gm(r)), which transfers efficiently between enterococci by broth mating. Each of the four erythromycin resistant transconjugants obtained by broth mating harbored a large conjugative plasmid (more than 100 kbp). The plasmids showed no homology with well-characterized enterococcal conjugative plasmids such as pAD1, pPD1, pAM(beta)1, pIP501 and pMG1 by Southern hybridization. Of the erythromycin resistance traits that transferred only by filter mating, it was found that the erythromycin resistance trait was conferred by a 47-kbp transposable element that transferred from the chromosome of the donor strain to different sites within the pheromone responsive plasmid pAD1 (60 kbp) of the recipient strain, suggesting that the erythromycin resistance trait was encoded on a conjugative transposon, which was named Tn950.  相似文献

A class II restriction endonuclease, PaeR7, has been isolated from a Pseudomonas aeruginosa strain containing the resistance plasmid pMG7. The activity cannot be isolated from an isogenic strain which does not contain this resistance plasmid. EndoR· PaeR7 requires Mg2+ for activity but does not require ATP or S-adenosyl-l-methionine. Specific digestion patterns are produced upon agarose gel electrophoresis of substrate DNAs which have been digested with the enzyme. The enzyme is the biochemical basis for the pMG7-mediated phage interference reported by Jacoby and Sutton (1977). DNAs isolated from restricted phages act as substrates for the enzyme while DNAs isolated from unrestricted phages and phages which have been modified by growth on strains containing pMG7 do not act as substrates for the enzyme.  相似文献

摘要:【目的】为了优化LJ1菌株的培养条件使之产生高活性的胞外褐藻胶裂解酶。【方法】通过富集培养技术从海带筛选到一株褐藻胶裂解酶产生菌LJ1, 依据表型特征、脂肪酸组成分析及16S rRNA基因序列分析对该菌株进行鉴定。通过单因子和正交试验对LJ1 菌株产胞外褐藻胶裂解酶的培养条件进行了优化。【结果】LJ1菌株属于假交替单胞菌属(Pseudoalteromonas)。该菌株产酶的最佳培养基组成为:褐藻胶3 g/L、(NH4)2SO4 3 g/L、NaCl 20 g/L、KH2PO4 0.1 g/L、CaCl2 0.1 g/L;最佳培养条件为:250 mL三角烧瓶中装液量25 mL、接种量3%、摇瓶转速150 r/min、pH7.5、培养温度为28℃、培养时间为24 h。LJ1菌株所产褐藻胶裂解酶的最适温度为40℃,最适pH7.6,最适NaCl浓度为0.3 mol/L。1 mol/L金属离子Mg2+对酶活力有明显的促进作用,而Co2+ 和Zn2+对酶活力有较强的抑制作用。【结论】LJ1菌株是Pseudoalteromonas 新的胞外褐藻胶裂解酶产生菌,在最佳培养条件下,该菌株的酶活力提高了66%。  相似文献

近年来,由于链霉菌启动子探测质粒的构建以及体外克隆技术的迅速发展,使得许多链霉菌启动子得到分离分析.已发现的链霉菌启动子大致可以分为三类:一、与原核生物典型启动子—10区和—35区类似的链霉菌启动子;二、仅与原核生物典型启动子—10区相似的启动子;三、无论在—10区还是在—35区与典型的原核生物的启动子都没有任何相似之处的启动子.链霉菌启动子的多样性还表现在—10区与—35区保守序列的间隔长短差异较大,从7bp到24bp长短不一.链霉菌中还常存在着串联的启动子.总之,链霉菌启动子比一般原核生物启动子具有更加复杂的多样性.  相似文献

A circular linkage map was developed for Streptomyces chrysomallus, a producer of actinomycin C. The map order of various marker loci was deduced from matings and to a minor extent from protoplast fusions. The map strongly resembles that of Streptomyces coelicolor A3(2). The recombination frequencies were low and highly variable (from 10(-9) to 5 X 10(-6]. Plasmid pIJ303 expressed its thiostrepton resistance gene in S. chrysomallus but did not promote chromosomal transfer or induce the Ltz+ phenotype. The data provide a background of genetics for investigations of antibiotic synthesis in this strain.  相似文献

To study the modes of actinomycin biosynthesis and the mechanism responsible for resistance to the antibiotic producing S. chrysomallus No. 2, the authors undertook an examination and studies into the cloning system for gene(s) of resistance to actinomycin from a S. chrysomallus No. 2 actinomycin C producer and the cloning of a S. chrysomallus No. DNA fragment to the actinomycin-sensitive Streptomyces Sp. 26-115 H-I on the vector plasmid pIJ702. The cloning gave rise to actinomycin-resistant strains. The character of actinomycin resistance is inheritable in a steady fashion.  相似文献

The fifth phage resistance factor from the prototype phage-insensitive strain Lactococcus lactis subsp. lactis ME2 has been characterized and sequenced. The genetic determinant for Prf (phage resistance five) was subcloned from the conjugative plasmid pTN20, which also encodes a restriction and modification system. Typical of other abortive resistance mechanisms, Prf reduces the efficiency of plaquing to 10(-2) to 10(-3) and decreases the plaque size and burst size of the small isometric-headed phage p2 in L. lactis subsp. lactis LM0230. However, normal-size plaques occurred at a frequency of 10(-4) and contained mutant phages that were resistant to Prf, even after repeated propagation through a sensitive host. Prf does not prevent phage adsorption or promote restriction and modification activities, but 90% of Prf+ cells infected with phage p2 die. Thus, phage infections in Prf+ cells are aborted. Prf is effective in both L. lactis subsp. lactis and L. lactis subsp. cremoris strains against several small isometric-headed phages but not against prolate-headed phages. The Prf determinant was localized by Tn5 mutagenesis and subcloning. DNA sequencing identified a 1,056-nucleotide structural gene designated abiC. Prf+ expression was obtained when abiC was subcloned into the lactococcal expression vector pMG36e. abiC is distinct from two other lactococcal abortive phage resistance genes, abiA (Hsp+, from L. lactis subsp. lactis ME2) and abi416 (Abi+, from L. lactis subsp. lactis IL416). Unlike abiA, the action of abiC does not appear to affect DNA replication. Thus, abiC represents a second abortive system found in ME2 that acts at a different point of the phage lytic cycle.  相似文献

将欧芹(Petroselinumcrispum)苯丙氨酸脱氨酶(PAL)cDNA亚克隆到组成型表达载体pMG36e启动子P32下游,电穿孔法转化乳酸乳球菌,获得有PAL表达活性的乳酸乳球菌工程菌(pMG36ePAL/L.lactisMG1363)。通过递归PCR合成了一段120bp的调控片段,用以将pMG36e改造为分泌型表达载体pXHS,以翻译偶联的方式表达PAL,可使PAL的N末端带上usp45信号肽,结果亦检测到PAL酶活性。自行分离克隆了乳酸乳球菌热休克蛋白基因dnaJ的启动子区域,构建了热诱导表达载体pXHJ,获得PAL热诱导表达工程菌(pXHJPAL/L.lactisIL1403),经30℃至37℃热诱导,可使PAL表达活性提高至2倍。本文还就乳酸乳球菌PAL工程菌在经典型苯丙酮尿症防治中的应用进行了分析和讨论  相似文献

Plasmid pMG1 (65.1 kb) was isolated from a gentamicin-resistant Enterococcus faecium clinical isolate and was found to encode gentamicin resistance. EcoRI restriction of pMG1 produced five fragments, A through E, with molecular sizes of 50.2, 11.5, 2.0, 0.7, and 0.7 kb, respectively. The clockwise order of the fragments was ACDEB. pMG1 transferred at high frequency to Enterococcus strains in broth mating. pMG1 transferred between Enterococcus faecalis strains, between E. faecium strains, and between E. faecium and E. faecalis strains at a frequency of approximately 10−4 per donor cell after 3 h of mating. The pMG1 transfers were not induced by the exposure of the donor cell to culture filtrates of plasmid-free E. faecalis FA2-2 or an E. faecium strain. Mating aggregates were not observed by the naked eye during broth mating. Small mating aggregates of several cells in the broth matings were observed by microscopy, while no aggregates of donor cells which had been exposed to a culture filtrate of E. faecalis FA2-2 or an E. faecium strain were observed, even by microscopy. pMG1 DNA did not show any homology in Southern hybridization with that of the pheromone-responsive plasmids and broad-host-range plasmids pAMβ1 and pIP501. These results indicate that there is another efficient transfer system in the conjugative plasmids of Enterococcus and that this system is different from the pheromone-induced transfer system of E. faecalis plasmids.  相似文献

AIMS: The aim of the study is to evaluate whether xylanase can be used as a potential reporter gene for cloning and expression studies in Lactococcus. METHODS AND RESULTS: The 750 bp xylanase gene was amplified and subcloned into the unique NheI restriction enzyme site of pMG36e and subsequently transformed into competent Escherichia coli XLI-blue MRF cells and Lactococcus lactis cells. Bacterial culture containing pMG36e-Xy has an enzyme activity of 390 microg xylose ml(-1) culture 30 min(-1), respectively, when compared with 40 microg xylose ml(-1) culture 30 min(-1) for the negative control (plasmidless strain). CONCLUSIONS: The thermostable xylanase gene was successfully expressed in both E. coli and L. lactis. The activity of xylanase can be easily detected by the formation of visible clearing zones around the transformed colonies on Remazol Brilliant Blue-Xylan (RBB-Xylan) agar media. However, there were some significant differences in the optimum growth temperature and plasmid stability in the new clones. SIGNIFICANCE AND IMPACT OF THE STUDY: The constructed reporter vector has the potential to be used as a reporter system for Lactococcus as well as E. coli, and it is an addition to the pool of lactococcal vector systems.  相似文献

The work was aimed at studying the resistance of three streptomycetes (Streptomyces chrysomallus, S. azureus and S. roseoflavus var. roseofungini) and their spontaneous Nocardia-like variants lacking aerial mycelium and spores against nine polyphages isolated mainly from soil. Some Nocardia-like variants were found to differ from their parent cultures in the resistance against certain actinophages. S. chrysomallus VKM Ac-590 and Ac-628 variants lost resistance against the phages. S. azureus VKM Ac-719 and S. roseoflavus var. roseofungini VKM Ac-770 variants became resistant to the phages. The changed phage resistance of the streptomycetes and their Nocardia-like variants was attributed to the disorganised process of adsorption (8 and 7%, respectively, against 70 and 90% for the parent strains).  相似文献

Abstract Two recombinant DNA clones, pMG286.2 and pMG301.1, were isolated from the partial genomic library of Mycoplasma gallisepticum strain S6. Recombinant M. gallisepticum specific fragments were used as probes in Southern hybridisation with 10 M. gallisepticum strains whose DNA was digested by Eco RI, Hin dIII, Bgl II, Rsa I and Bam HI. The 1.5 kb fragment pMG301.1 did not show polymorphism in hybridisation patterns with M. gallisepticum strains, while the 3.5 kb fragment pMG286.2 enabled differentiation of M. gallisepticum strains into clusters. The DNA sequence of pMG301.1 was used to design a pair of 27-mer oligonucleotides flanking a 1.3 kb genomic region. These two primers directed specific in vitro amplification of all M. gallisepticum strains assayed giving an expected 1.3 kb product. Digestion of polymerase chain reaction products by Dde I enabled simple differentiation between clusters of M. gallisepticum strains and may be useful for improved epizootiological studies of M. gallisepticum infections in poultry.  相似文献

Three copper-resistant, gram-negative bacteria were isolated and characterized. Of the three strains, Alcaligenes denitrificans AH tolerated the highest copper concentration (MIC = 4 mM CuSO(4)). All three strains showed various levels of resistance to other metal ions. A. denitrificans AH contains sequences which cross-hybridized with the mer (mercury resistance) determinant of Tn21 and the czc (cobalt, zinc, and cadmium resistance), cnr (cobalt and nickel resistance), and chr (chromate resistance) determinants of A. eutrophus CH34. DNA-DNA hybridization with probes prepared from A. eutrophus CH34 and Tn21 revealed the presence of chr-, cnr-, and mer-like sequences on the 200-kb plasmid pHG27 and of czc, cnr, and mer homologs located on the chromosome. The second strain, classified as Alcaligenes sp. strain PW, carries czc, cnr, and mer homologs on the 240-kb plasmid pHG29-c and a chr determinant on the 290-kb plasmid pHG29-a; a third plasmid, the 260-kb large plasmid pHG29-b, is cryptic. In contrast to the Alcaligenes strains, which were isolated from metal-contaminated water, Pseudomonas paucimobilis CD was isolated from the air. This strain harbors two cryptic plasmids: the 210-kb large plasmid pHG28-a and the 40-kb plasmid pHG28-b. Southern analysis revealed no homology between the metal ion resistance determinants of A. eutrophus CH34 and P. paucimobilis CD.  相似文献

When Streptomyces chrysomallus 2703 grows on solid media, lytic zones appear in the form of negative colonies which are not caused by the virulent phage. The material from these colonies and the cultural broth of S. chrysomallus 2703 were examined by electron microscopy. Four different morphological types of particles were revealed, three of which were defective phage particles (tails). Particles of the fourth type had a regular hexagonal shape and a diameter of 200A. The particles prevailed in all of the preparations. Their origin is discussed. S. chrysomallus in considered as a poly- and defective lysogenic culture.  相似文献

本文报告以CD2cDNA5’端的片段作探针,从人T淋巴细胞基因组文库筛选阳性重组克隆,经限制性内切酶降解和Southern杂交分析,证明其中一个阳性克隆的插入片段中含CD2基因5’侧翼顺序。经插入片段的亚克隆、限制性内切酶图谱及DNA序列分析,鉴定出一含转录起始点及其上游序列的4.0kb片段。将此片段中含转录起始点和两个DN(ase)Ⅰ高敏感位点的2.5kb片段定向克隆到以虫萤光素酶为报告基因的表达载体pMG3中,并用限制性内切酶对此2.5kb片段作不同程度缺失,构成一系列突变子。这些重组的表达质粒转染人JurkatT细胞后,以瞬时表达实验分析各突变子驱动虫萤光素酶基因的表达,结果发现在CD2基因5’上游具有很弱的启动子活性,初步测定该启动子位于-1.2kb~-98bp域。CD2基因具有弱启动子、强增强子的特点与T细胞表面其它抗原分子基因是相似的。  相似文献

The donor properties of K. pneumoniae PI 220 with multiple drug resistance were studied. It was shown that the above strain carried 2 plasmids, i.e. R-plasmid pPI 220 controling resistance to ampicillin, streptomycin, tetracycline, chloramphenicol, kanamycin and sulphanylamides and plasmid pPI 221 controlling lactose fermentation. Both plasmids can be transfered on conjugation to strain E. coli P678 at a temperture of 28 degrees C at a rate of 10(-5) for pPI 220 and 10(-4) for pPI 221. The drug resistance controlled by pPI 221 was transfered mainly in a "blocks" simultaneously to 6 drugs. Deletion of plasmid pPI 220 was observed rarely. The donor properties of the strain were defined by the conjugative plasmid pPI 220 controlling the self-transfer and mobilization of plasmid pPI 221 incapable of the self-transfer. E. coli P678 (pPI 220) (PPI 221) acquired the donor properties and transfered both plasmids to E. coli J62 on crossing simultaneously at a rate of 10(-2), as well as to S. typhimurium LT2 and P. rettgeri at a rate of 10(-5). In all the recipient strains studied the transfered plasmids were unstable and segregated also simultaneously at a rate being the highest for P. retgari PI 230. The clones with stable preservation of the plasmids could be obtained by selection.  相似文献
