Summary. In plants, tip-growing cells represent an ideal system to investigate signal transduction mechanisms, and among those, pollen tubes are one of the favourite models. Many signalling pathways have been identified during germination and tip growth, namely, Ca2+, calmodulin, phosphoinositides, cyclic AMP, and GTPases. Not surprisingly, the apical secretory machinery, essential for tip growth, seems to be an intersection point for all these pathways. Recently, the phospholipid phosphatidic acid was also suggested to actively participate in the control of endo- and exocytosis and to interfere with the correct positioning of the actin cytoskeleton. Phosphatidic acid seems to act concertedly with the phosphoinositides phosphatidylinositol 4,5-bisphosphate and D-myo-inositol 1,4,5-trisphosphate. Here we review previous data and discuss additional evidence that these three molecules have a combined action modulating both the actin cytoskeleton and the apical secretory machinery. We further discuss how these findings can be integrated into a working model for pollen tube apical secretion that contemplates the existence of a rapid endocytosis mechanism. Summary. Signalling is an integral component in the establishment and maintenance of cellular identity. In plants, tip-growing cells represent an ideal system to investigate signal transduction mechanisms, and among these, pollen tubes (PTs) are one of the favourite models. Many signalling pathways have been identified during germination and tip growth, namely, Ca2+, calmodulin, phosphoinositides, protein kinases, cyclic AMP, and GTPases. These constitute a large and complex web of signalling networks that intersect at various levels such as the control of vesicle targeting and fusion and the physical state of the actin cytoskeleton. Here we discuss some of the most recent advances made in PT signal transduction cascades and their implications for our future research. For reasons of space, emphasis was given to signalling mechanisms that control PT reorientation, so naturally many other relevant works have not been cited. The partially-purified, thermally-stable trehalase from conidia of Humicola grisea was highly specific for trehalose and was free of potentially interfering activities. The enzyme was fully stable when stored in solution at -15°C for at least 6 months. This preparation could be used to quantify trehalose from 0.05 to 1.25 mol/ml either in carbohydrate mixtures or in complex biological materials.M.J. Neves, H.F. Terenzi and J.A. High activities of extracellular pectinase with viscosity-diminishing and reducing groups-releasing activities were produced by Penicillium frequentans after 48 h at 35°C, in agitated cultures supplemented with 0.5% citrus pectin and initial pH of 2.5. Under these conditions the fungus also produced high activity of pectinesterase. At an initial pH of 7.0 or 8.0, pectin lyase activity was also detected. Enzyme activity releasing reducing sugars was more stable at 50°C than viscosity-diminishing activity. Plathymenia foliolosa and P. reticulata are two tropical tree species economically important for their high quality wood. P. foliolosa occurs mainly in areas of Atlantic Forest (a tropical forest) whereas P. reticulata occurs mainly in the Cerrado region (a savanna vegetation), both Brazilian biomes classified as hotspots for conservation priorities. In the present study RAPD markers were used to differentiate P. foliolosa from the vicariant species P. reticulata. Using 10 random primers it was possible to obtain 84 markers, of which 22 (26.2%) were present in just one of the two species. Nine of the ten primers were successful in discriminating between the species through the detection of diagnostic markers or band frequency differences. One of the populations of P. foliolosa, located in a region where the two species co-occur, presented several markers that were characteristic of P. reticulata, suggesting the occurrence of gene flow between the two species. The aim of this research was to evaluate the biological behaviour of melonworm at different natural and artificial diets. Squash cultivar jacaré, cucumber, summer squash and an artificial diets developed by Hensley & Hammond for the sugarcane borer were tried. The research was carried out in the Laboratório de Entomologia do Centro de Ciências Agrárias of the Universidade Federal do Espírito Santo, in climatized room at 25 +/- 1 masculineC, relative humidity of 70 +/- 10% and 14h photofase. The caterpillars fed on each substrate during the whole life cycle. Differences in duration of the larval period, larvae and pupae survival, adult longevity and number of eggs were registered between the substrates. The results show that the best diet for rearing is the artificial one. Between the natural diets the cucumber showed the best results.  相似文献

H. grisea produced an extracellular -glucosidase (EC 3.2.1.21) at high activity in media supplemented with carboxymethyl cellulose (CMC) or cellobiose. Cellobiose-induced -glucosidase was insensitive to glucose repression whereas that of CMC-supplemented cultures was partially repressed. Molecular sieving revealed three main active components (Mr 50, 128 and 240 kDa). Glucose competitively inhibited -glucosidase activities with Ki values of 0.9mM and 3.3mM (extracellular) and 10.2mM and 22.6mM (cytosolic), induced in the presence of CMC or cellobiose respectively.The authors are with the Departamento de Biologia, Faculdade de Filosofia. The pectinases produced byPenicillium frequentans were subjected to pectin- or sodium polypectate-PAGE. The fungus secreted one endo- and one exo-polygalacturonase during the first 10 h of incubation, either in media supplemented with pectin or in the absence of carbohydrate source. After 17 h of cultivation, another two exo-and three endo-polygalacturonases were detected in cultures supplemented with pectin. The results indicate that two constitutive polygalacturonases are secreted initially, followed by the synthesis of other inducible pectinases, in a sequential production of the pectinolytic complex. The remarkable anthropological series formed by E. Tamagnini at the Universidade de Coimbra consists of the exhumed human remains from “La Conchada” cemetery, in Coimbra (Northern Portugal). From this collection, only those individuals coming from the different districts of Coimbra, where they also lived and died, were selected for study. The resulting series was made of by 483 individuals: 234 women and 249 men. The personal data sheets indicate that these people died between 1910 and 1938; their professions, age at death and its cause are also noted. They can be considered as a relatively homogeneous group of low socioeconomical level. The study of the three oraltori demonstrated the high frequencies of all of them, the absence of sexual dimorphism and only a few statistically significant differences between some of the age classes considered. The possible etiological factors (genetical and/or environmental) are also discussed.  相似文献

Summary.  Syagrus coronata is an economically important palm tree grown as an ornament, for the oil extracted from its seeds, and the wax from its leaves which has several applications in industry. Silicon biocomposites were analyzed in leaves of S. coronata. Silica bodies were found as extracellular silica masses between the hypodermal-layer cell walls and in granules present in the vacuoles of palisade cells. Scanning electron microscopy of the hypodermal layer of cells showed a collection of spherical bodies embedded in enveloping cavities that outlined the general structure of the bodies. Globular subunits with sharp edges formed the spherical bodies that ranged from 6 to 10 μm in diameter (average, 7.8 μm). X-ray microanalysis detected only silicon and oxygen homogeneously distributed throughout the bodies. Vacuoles of palisade cells contained a large number of granules ranging from 20 nm to 1.2 μm in size (average, 300 nm). Transmission electron microscopy associated with electron spectroscopic imaging and electron energy loss spectroscopy were used to determine the elemental composition of the granules. Vacuolar granules were amorphous and composed of silicon and oxygen, suggesting they consist of amorphous silica biominerals. No nitrogen, indicative of organic matter, was detected in the granules. Summary. Morphohistological analysis and histochemical studies were carried out during the induction and development of Feijoa sellowiana somatic embryos. Zygotic embryos were cultured on LPm medium containing 2,4-dichlorophenoxyacetic acid (20µM) and glutamine (8mM). Somatic embryogenesis could be induced from embryogenic cells that originated in meristematic centers or from clusters of cells. The presence of few starch grains and abundant protein bodies was observed in the globular and early torpedo stages, while in torpedo and cotyledonary-stage somatic embryos an enhanced synthesis of starch grains was associated with the accumulation of reserves to be used in the conversion of the embryos to plantlets. A phylogenetic study of selected fleshy-fruited genera of the Myrtaceae was conducted using sequences from the ITS region of nuclear DNA and the psbA-trnH region of plastid DNA. Studies to date have suggested that the fleshy-fruited state has arisen on several occasions in the Myrtaceae. The previously accepted and predominantly Neotropical tribe Myrteae has traditionally been divided into three groups, the subtribes Myrtinae, Eugeniinae and Myrciinae. This subtribal arrangement is analysed in detail here for the first time. The monophyly of the tribe and subtribes are tested and relationships of the genera within them, in particular those of the Myrciinae and anomalous genera sometimes associated with it, are discussed. Combined analyses of these two DNA regions revealed 40 shortest trees, all of which resolve Myrteae (excluding the Acmena group) as monophyletic. Myrciinae appears to be monophyletic whereas Myrtinae and Eugeniinae appear polyphyletic. The phylogenetic positions and relationships of the anomalous genera Myrceugenia, Luma and Blepharocalyx are unclear, but Myrceugenia is never included within the Myrciinae s.str. A Myrciinae s.str. clade emerges within which Myrcia, Calyptranthes and Marlierea appear polyphyletic. Summary   Croton imbricatus L. R. Lima & Pirani from Bahia, C. muellerianus L. R. Lima from Paraná and C. pygmaeus L. R. Lima from Rio Grande do Sul are described and illustrated. They are included in Croton section Lamprocroton (Müll. Arg.) Pax because they are much branched subshrubs or shrubs, bearing leaves with a lepidote indumentum (at least in part), inflorescences lacking bisexual cymules, and pistillate flowers with bifid styles.

AnArabidopsis thaliana cDNA was isolated by complementation of theEscherichia coli mutant strain BW535 (xth, nfo, nth), which is defective in DNA base excision repair pathways. This cDNA partially complements the methyl methane sulfonate (MMS) sensitive phenotype of BW535. It also partially corrects the UV-sensitive phenothpe ofE. coli AB1886 (uvrA) and restores its ability to reactivate UV-irradiated phage. It has an insert of ca. 1.3 kb with an open reading frame of 1047 bp (predicting a protein with a molecular mass of 36 kDa). This cDNA presents a high homology to a stress related gene from two species ofFusarium (sti35) and to genes whose products participate in the thiamine biosynthesis pathway,THI4, fromSaccharomyces cerevisiae andnmt2 fromSchizosaccharomyces pombe. TheArabidopsis predicted polypeptide has homology to several protein motifs: amino-terminal chloroplast transit peptide, dinucleotide binding site, DNA binding and bacterial DNA polymerases. The auxotrophy for thiamine in the yeastthi4::URA3 disruption strain is complemented by theArabidopsis gene. Thus, the cloned gene, namedthi1, is likely to function in the biosynthesis of thiamine in plants. A canonical variate analysis was used to summarize the distribution pattern of 30 species of dung beetles (Scarabaeinae) from 162 sites in the Iberian peninsula (Spain and Portugal) in relation to 4 site variables. The best separation of beetle species, obtained using a xerothermic climatic index, was into 3 groups called “mediterranean”, “unrestricted” and “temperate”. Little further separation was achieved using altitude, dung type or soil type. These data are used to select dung beetles for introduction to south-western Australia for bush fly control.
Résumé On a fait appel à l'analyse de variables canoniques pour résumer le plan de distribution, relatif à 4 variables de localité, de 30 espèces de bousiers (Scarabaeinae) de 162 localités de la Péninsule ibérique (l'Espagne et le Portugal). La meilleure division des espèces, obtenue à l'aide d'un indice xérothermique climatique, se faisait en 3 groupes désignés respectivement comme “méditerranéen”, “ubiquiste” et “tempéré”. Peu de séparation supplémentaire a été obtenue en ayant recours à des facteurs d'altitude, de nature de la bouse ou de type de sol. Ces données ont été employées pour aider à la sélection de bousiers destinés à être introduits en Australie Occidentale pour y combattreMusca vetustissima Walker[Diptera].

The cell-cycle progression of germinating embryos of maize (Zea mays L.) was studied from 0 to 72 h after the start of imbibition using DNA flow cytometry on isolated nuclei, and analyses of thymidine kinase activity, histone biosynthesis and levels of proliferating cell nulcear antigen (PCNA). At the start of germination, 75% of the cells were in G1, but this population had decreased to 25% by 72 h. The concomitant increase of cells in S-phase did not occur continuously, but stepwise, indicating that during germination most of the cells enter S-phase as a partially synchronized population. Within the initial 60 h of embryo germination the cells passed through one S-phase; the start and duration of this period of replicative DNA synthesis was further substantiated by the analysis of S-phase-associated events, the biosynthesis of core histones and the enzyme activity of thymidine kinase, which both began to increase at about 12 h after the start of differentiation. Thymidine kinase fluctuated periodically during germination with a transient maximum at 30 h and a second peak at 72 h; histone biosynthesis was not detectable until 12 h after the start of germination. The levels of PCNA protein closely resembled the pattern of thymidine kinase during germination. The untwisting movement of the hygroscopic awn of wild oats (Avena fatua L.) causes the caryopses to rotate in anticlockwise direction and to dislodge from the spikelet. While untwisting, the awn generates more torque than is required to separate the caryopsis. The periodic shattering of caryopses caused by awn movement may be biologically significant in that the chances of having favorable moisture conditions for caryopses germination in the field at the time of dispersal are increased. Summary  A new genus, Cleistesiopsis, was segregated from Cleistes based on morphological and molecular characteristics, and two new species of Cleistes: C. batistana and C. elongata, both occurring in the Brazilian Central Plateau (Central-Western Brazil), are described and illustrated. Furthermore, a key to genera currently recognised within Pogonieae is presented.
