The histology and mucus histochemistry of the pleuronectid post-gastric alimentary canal was examined using light and electron microscopy. Distinct differences in goblet cell mucus histochemistry were observed between species, with the two closest taxonomic species, the winter flounder and the yellowtail flounder showing the most diversity and the halibut showing regional variation. Numbers of goblet cells within post-gastric regions did not differ significantly between species, but were significantly different between regions within species increasing toward the rectum. The post-gastric region was divisible into two areas based upon the ultrastructural features of lipid digestion and absorption in the intestine and pyloric caeca, and of exogenous protein in the rectum. The combination of species-specific histochemical differences in mucus and general histological and ultrastructural differences within the post-gastric regions between these species suggest a correlation between lumenal environmental conditions/histology and natural prey preference.  相似文献

The ultrastructure of the female gonad of the land planarian Geoplana burmeisteri was investigated by means of electron microscopy and cytochemical techniques. It consists of two small germaria located ventral to the intestine and of two irregular, lateral rows of vitelline follicles, both enveloped by a tunica composed of an extracellular lamina and an inner sheath of accessory cells. Accessory cell projections completely surround developing oocytes and vitellocytes. The main feature of oocyte maturation is the appearance of chromatoid bodies and the development of the rough endoplasmic reticulum (RER) and Golgi complexes. These organelles appear to be correlated with the production of egg inclusions of medium electron density, about 1.5-1.8 microm in diameter, which remain scattered in the ooplasm of mature oocytes. On the basis of cytochemical tests demonstrating their glycoprotein composition, these inclusions were interpreted as residual yolk globules. Vitellocytes are typical secretory cells with well-developed RER and Golgi complexes that are mainly involved in the production of yolk globules and eggshell globules, respectively. Eggshell globules appear to arise from repeated coalescence of small Golgi-derived vesicles and, at an intermediate stage of maturation, show a multigranular pattern. Later, after vesicle fusion, they reach a diameter of 1.3-1.6 microm when completely mature and show a meandering/concentric pattern, as is typical of the situation seen in most Proseriata and Tricladida. The content of yolk globules is completely digested by pronase, while the content of eggshell globules is unaffected. Mature vitellocytes contain, in addition, a large quantity of glycogen and lipid droplets as further reserve material. On the basis of the ultrastructural characteristics of the female gonad described above and in relation to the current literature, we conclude that G. burmeisteri appears to be more closely related to the freshwater triclads, in particular to members of the Dugesiidae, than to the marine triclads.  相似文献

Preservation of Tracheal Mucus by Nonaqueous Fixative   总被引:3,自引:0,他引:3  
Two nonaqueous fixatives, composed of fluorocarbon solvents with dissolved osmium tetroxide, were used to determine the feasibility of preserving the mucous coat in bovine and rat trachea for light and electron microscopy. Aqueous fixatives, while providing excellent cytological preservation, wash away the mucous lining, precluding ultrastructural analysis. Inclusion of ruthenium red or alcian blue within aqueous fixative improved retention of mucus, but provided incomplete, patchy results. Fixation with nonaqueous fluorocarbon solvent and dissolved osmium tetroxide preserved a continuous mucous epiphase layer above a clear hypophase layer. Subcomponents of the mucus included an electron dense surface layer, interrupted patches of mucus above the surface layer and electron dense membrane-like material within the mucus. This method of fixation will preserve mucus for light, scanning and transmission electron microscopy, using either intratracheal or immersion methods of fixation. The latter would enable use of materials from large animal models, autopsy or an abattoir.  相似文献

Ultrastructural study of oogenesis in the acoel turbellarian Convoluta   总被引:2,自引:0,他引:2  
An ultrastructural investigation of oogenesis has been carried out on the acoel turbellarian Convoluta psammophyla. Developing female germ cells are not contained in well delimited ovaries but are freely distributed in the parenchyma and are surrounded by narrow cytoplasmic projections of accessory-follicle cells. Oogenesis can be divided into two periods, the previtellogenic and the vitellogenic phase. In the first period the oocyte undergoes a number of cell differentiations necessary for the intense biosynthetic activity of the second period. The ample development of nucleolus, ribosomes, endoplasmic reticulum and Golgi complexes along with the appearance of large lipid droplets and clusters of electron dense granules characterize the previtellogenic phase. The formation of yolk globules is the main feature of the second period of oogenesis. It occurs by an autosynthetic mechanism involving endoplasmic reticulum and Golgi complexes, since no endocytotic activity has been detected in the developing oocyte. The electron dense granules apparently move towards the cortical ooplasm during the late vitellogenic phase and take part in egg covering formation. Hypotheses on the role of follicle cells and on the phylogenetic significance of a comparative analysis of egg inclusions with homologous structures of other Turbellaria are suggested.  相似文献

This study provides evidence for the Golgi‐like activity of the multilayered interlaced network (MIN) and new ultrastructural observations of the MIN in the sporoplasm of Anncaliia algerae, a microsporidium that infects both insects and humans. The MIN is attached to the end of the polar tubule upon extrusion from the germinating spore. It surrounds the sporoplasm, immediately below its plasma membrane, and most likely maintains the integrity of the sporoplasm, as it is pulled through the everting polar tube. Furthermore, the MIN appears to deposit its dense contents on the surface of the sporoplasm within minutes of spore discharge thickening the plasma membrane. This thickening is characteristic of the developmental stages of the genus Anncaliia. The current study utilizes transmission electron microscopy (TEM), enzyme histochemistry, and high voltage TEM (HVEM) with 3D tomographic reconstruction to both visualize the structure of the MIN and demonstrate that the MIN is a Golgi‐related structure. The presence of developmentally regulated Golgi in the Microsporidia has been previously documented. The current study extends our understanding of the microsporidial Golgi and is consistent with the MIN being involved in the extracellular secretion in Anncaliia algerae. This report further illustrates the unique morphology of the MIN as illustrated by HVEM using 3D tomography.  相似文献

The ultrastructure of the small granular, proteinaceous cells of 11 species of lumbricid earthworm is described and no species differences were recorded. The cells are characterized by the presence of membrane-bound, electron dense granules (0.6–0.7 urn in diameter) arising from polarized Golgi systems in close topographical relationship to the granular endoplasmic reticulum. Variations in electron density of the granules appear to be associated with maturation of the granules. The granules show a finely reticular substructure at high magnification. A less than fully mature stage of this cell type is described.
The occurrence of the small granular cell type in the annelids is discussed, as is the possible function of its secretion.
The confusions in the lumbricid literature concerning this cell type are discussed, and the much referred to figure of the 'albumen' cell type in English texts is shown not to be equivalent to the small granular, proteinaceous type referred to in this and a previous histochemical study.  相似文献

Summary In the corpora cardiaca of the insect Leucophaea the administration of serotonin elicits ultrastructural features indicative of the extrusion of neurosecretory material by exocytosis. The response to the stimulus and the process of extrusion seem to occur at considerable speed. Nearly all of the 30 test animals, fixed at various intervals starting as early as 3 min after the injection of the drug, show granules captured at the moment of leaving the axon as well as fully exteriorized secretory material. The fact that many of these granules are much smaller than the typical neurosecretory type speaks for intracellular fragmentation of the latter prior to the discharge of this cellular product. After 25 min or more the extruded electron dense structures show signs of breakdown. The apparent speed of these phenomena accounts for the dearth of omega-type configurations observed in unstimulated specimens of this species. The possible relationship between the membrane phenomena involved in exocytosis and the transient protrusions of bounding membranes of neurosecretory granules described in earlier papers remains to be clarified.Supported by N.S.F. research grant BMS 74-12456  相似文献

The female gonad of the land planarians Microplana scharffi and Microplana terrestris consists of two small germaria located ventrally in the anterior third of the body and of two ventro‐lateral rows of oblong vitelline follicles distributed between the intestinal pouches. Both these structures are enveloped by a tunica composed of an outer extracellular lamina and an inner sheath of accessory cells. Oocyte maturation is characterized by the appearance of chromatoid bodies and the development of endoplasmic reticulum and Golgi complexes. These organelles appear to be correlated with the production of egg granules with a fenestrated/granular content of medium electron density, about 4–5 μm in diameter, which remain dispersed in the ooplasm of mature oocytes. On the basis of cytochemical tests showing their glycoprotein composition, and their localization in mature oocytes, these egg granules have been interpreted as yolk. In the vitelline follicles, vitellocytes show the typical features of secretory cells with well‐developed rough endoplasmic reticulum and Golgi complexes involved in the production of eggshell globules and yolk. The eggshell globules, which appear to arise from repeated coalescences of two types of Golgi‐derived vesicles, contain polyphenols and, when completely mature, they measure about 1–1,2 μm in diameter and show a meandering/concentric content pattern as is typical of the situation observed in most Proseriata and Tricladida. Mature vitellocytes also contain a large amount of glycogen and lipids as further reserve material. On the basis of the ultrastructural features of the female gonad and in relation to the current literature the two species of rhynchodemids investigated appear to be closely related to the freshwater planarians belonging to the family Dugesiidae. Transmission electron microscope investigations of the serous (poison) skin glands in the New World tree frogs Osteopilus septentrionalis and Phrynohyas venulosa revealed that they produce granules with closely similar substructures, namely, a dense cortex and pale medulla. In both species these features, that contrast the complex, sometimes repeating patterns described in other hylid frogs, derive from similar secretory and maturational processes starting from the Golgi phase of poison biosynthesis. Observations on secretory discharge showed that the two species share common release mechanisms, based on bulk discharge (holocrine) processes. Our data provide novel evidence of the extensive ultrastructural polymorphism of serous skin products in Hylidae and agree with phylogenies that regard this family as polyphyletic in origin. Assuming that ultrastructural features of cutaneous poison biosynthesis and maturation are adequate clues for tracking anuran phylogeny, the present findings also support a close relationship between Osteopilus and Phrynohyas taxa as previously suggested by osteological evidence.  相似文献

Electron microscopic studies of unstimulated and stimulated spider fibroin glands show that the fibroin synthesis stimulus evokes visible changes in both the endoplasmic reticulum and Golgi apparatus of their secretory epithelium. Gradual increase in distension of the reticulum accompanies the increase of evoked fibroin synthetic activity. The flattened translucent Golgi vesicles, seen in inactive cells, display a gradual increase in size and number, also with time. The stimulation also elicits a gradual transition in the gland's luminal membrane, during which the microvilli on the lining gradually disappear acquiring an electron dense appearance. Correlations of the observed transitions to the gland's increase in rate of elicited synthetic activity are discussed. The parallelisms between the ultrastructural modifications observed in the spider secretory cells with those described in the silkworm glands during their progression through the fifth instar have been stressed.  相似文献

The pollen morphology of 49 species and one variety, representing 18 genera of the family Annonaceae from Thailand, is described and illustrated based on observations using scanning electron microscopy (SEM). The pollen grains of Anaxagorea show double-spheres as a result of the intine extrusion. The palynological evidence confirms the close relationship of Dasymaschalon, Desmos and Asian Friesodielsia. Decussate tetrads in Goniothalamus and coexisting tetragonal, rhomboidal, T-shaped, tetrahedral and decussate tetrads in Mitrephora are reported. Pollen morphology is consistent within Huberantha, Marsypopetalum and Monoon, but is more diverse in Polyalthia sensu stricto Pollen unit, shape, size, ornamentation and aperture number reaffirm the great diversity among and within genera in Annonaceae.  相似文献

The female Afghan pika is known as a species of copulatory ovulators exhibiting persistent estrus unless copulatory stimuli are employed. The plasma LH concentration rose quickly after copulation, peaked at 6 h and descended thereafter until 12 h. Before copulation (estrus), immunoreactive LH-containing cells of the pituitary in this species were filled with numerous large electron dense secretory granules, but the Golgi apparatus and the granulated endoplasmic reticulum were not well developed. One hour later the stainability of the cells was faint, their secretory granules were malls in size and in number, and were of low electron density. These alterations were the most predominant at 12 h after copulation. No exocytotic extrusion of secretory granules was observed throughout the experiment.  相似文献

The ultrastructural study of the supramedullary neurons in Crenilabrus quinquemaculatus shows that these cells are engaged in intense synthetic activity as is testified by the nuclear morphology, the plentiful rough endoplasmic reticulum and polysomes, and finally the remarkably developed Golgi complexes, many of them active. Moreover the cytoplasm of the supramedullary neurons shows numerous membrane-bounded bodies (1,800-4,000 A) containing electron dense material, more or less finely granular. Oogenesis in fetal pig ovaries comprises the successive changes from the primordial germ cells to the dictyotene oocytes in primordial ovarian follicles. In this study the observations were carried out with an electron microscope and stereological analysis was performed. At the ultrastructural level there are no differences between the primordial germ cells and oogonia, but oogonia are connected with the intercellular bridges. The onset of the dictyotene phase was accompanied by the changes in the cytoplasm of oocytes. Near the nucleus, the yolk nucleus is formed containing numerous Golgi bodies, endoplasmic reticulum (ER), mitochondria and granules. ER proliferates in contact with the external leaflet of the nuclear envelope forming the narrow ER cisterns. Between the nuclear envelope and ER cisterns, the vesicles with grey content are visible. The proliferating ER forms numerous concentric cisterns around the nucleus. Next, the most external cisterns fragment, detach, and then form the cup-like structures. These structures separate the distinct areas of cytoplasm-compartments, which contain mitochondria, ribosomes and lipid droplets. The cells of cortical sex cords of the ovary, which encloses the oocyte, form the follicles. The volume of oocytes in forming follicle increases due to the increase in the number of the cell inclusions: lipid droplets, vacuoles and yolk globules. In the oocytes of primordial ovarian follicles, the compartments are transformed into the yolk globules, which are encountered by a sheath of ER cisterns and the grey vesicles; they contain the mitochondria, lipid droplets and light vacuoles. The role of the compartments and yolk globules as metabolic units is discussed in comparison with similar structures of the mature eggs of pigs and other mammal species.  相似文献

SYNOPSIS. The taxonomy of the genera Mallomonas, Mallomonopsis and Synura is based on their silica scales. Examination of water samples from Southern Scotland in the electron microscope revealed that they were plentiful in the area and permitted detailed observations on new species. Comparative studies are made on the flagella of three known species of Synura ; the "flimmergeissel" of these biflagellates appear to be more or less similar, but the whip-lash flagella show specific differences suggesting a possible secondary taxonomic criterion. The distribution of the three genera is discussed with particular reference to species found in Iceland, Scotland and Japan. A stage in the conjugation of a species of Mallomonas is described.  相似文献

The ultrastructural features, characterizing the different types of spermatogonia and spermatocytes in the blue fox, have been studied within and near the reproductive season, and also in the summer and autumn. Two distinct types of spermatogonia — A and Β — are described. The A-spermatogonia often have a prominent nucleolus and numerous cytoplasmic organelles including characteristic whorls of AER. Large vacuoles containing electron dense particles are sometimes observed. In the B-spermatogonia the chromatin forms condensed areas of varying size, and the nucleolus is usually absent. The number of cytoplasmic organelles is generally small. Ultrastructural characteristics are further used to distinguish between the different stages in the prophase of the primary spermatocytes. In leptotene the nucleus contains a thread-like chromatin with electron dense peripheral areas. Towards the end of the stage the mitochondria display dilated cristae, and aggregations of a granular material can be observed in the intermitochondrial matrix. Zytogene is characterized by the appearance of syniaptinemal complexes in the nucleus, and of the chromatoid body and piles of annulate lamellae in the juxtanuclear cytoplasm. In pachytene the chromosomes become apparent as aggregations of condensed chromatin associated with the synaptinemal complexes. The Golgi complex is more prominent than in the previous stages, and the number of the other cytoplasmic organelles is increasing. In the last stages of the prophase (diplotene and diakenesis) the chromosomes become still more electron dense, the nucleolus appears as a very prominent structure, and there is a marked vesiculation of the cytoplasm. The secondary spermatocytes have a characteristic nucleus with a somewhat irregular outline and larger peripheral areas of condensed chromatin. In the cytoplasm a double Golgi complex is frequently observed. In the summer and autumn spermatocytes in zygotene seem to represent the most advanced form of spermatogenic cells.  相似文献

Electron microscope studies of young oocytes have demonstrated that the plate-like, hexagonally shaped yolk bodies previously observed in living cells are wholly within the substance of oocyte mitochondria and that they remain within these mitochondria while increasing in size. These bodies possess a crystalline structure consisting of what appear to be lines, with a spacing of 70 to 85 A, and appear very dense in the electron microscope. After formalin fixation such bodies give an intense positive test for protein, and when viewed in the electron microscope are only slightly less dense than after OsO4 fixation. Evidence is presented for the origin of these crystals within a single crista. The clusters of yolk globules previously studied in living cells are seen to consist of several types of bodies, but an irregular dense droplet predominates. This dense material is apparently secreted by small spherical bodies which, the evidence suggests, originate from the breaking up of filamentous mitochondria and which possess an outer double membrane and sometimes internal cristalike membranes. When thin sections of young oocytes are immersed in xylol the dense globules of the clusters are dissolved, but the hexagonal bodies are unaffected, indicating that the globules are of a predominantly fatty nature, while the hexagonal bodies are of a predominantly protein nature. Examination of mature or almost mature oocytes has revealed that the main body of the yolk platelets is crystalline in nature and is surrounded by a thick matrix which, in light microscope study, masks the fact that the face view of the main body of the platelets is often hexagonal. The spacing within the main body is found to be 70 to 85 A. The crystal laminae of this material can be resolved quite clearly into rows of particles. Dense globules of varying sizes are found in the cytoplasm between the platelets. When thin sections of these OsO4-fixed oocytes are immersed in xylol, the material of the globules is extracted and the crystalline material of the platelets remains unaffected, indicating the fatty nature of the globules and the protein nature of the platelets. The platelets of the mature egg resemble the hexagon bodies, previously described in young oocytes, in their protein nature, their crystalline spacing, and their hexagonal outline. This is given as strong evidence for the origin of the mature platelets by the growth of the intramitochondrial hexagon bodies. The biochemical implications of this study are discussed.  相似文献

The ovary of the freshwater planarian Schmidtea mediterranea has been studied for the first time using both light and electron microscopy methods. The ultrastructure of the ovary revealed two types of cells: accessory cells and germinal cells at various stages of differentiation, distributed along a maturation axis. Initially, oogonia underwent cytoplasm growth due to the development of organelles, such as endoplasmic reticulum, Golgi complex, and mitochondria, which are all involved in the production of cytoplasmic inclusions or yolk globules. It is shown that the chromatoid body and fibrogranular aggregates may participate in the synthesis of vitelline inclusions. When completely mature, the oocytes have become larger, due to the accumulation of nutritive inclusions, which are round in shape and have a paracrystalline structure. These inclusions are interpreted as being yolk globules and may represent a kind of nutritive material for the developing embryo. These ultrastructural features of the ovary agree with the available phylogenetic tree, based on morphological and karyological characters that considers Schmidtea group as a genus and not a subgenus. The presence of sperm between the oocytes suggests that fertilization may occur within the ovary, representing an uncommon condition within the Triclads, in which fertilization usually takes places outside of the ovaries.  相似文献

Summary The gastrin-producing cells in the cat have been studied under various experimental conditions indicating the secretory cycle of these cells. Normal gastrin cells in animals fed ad libitum show secretory granules of different electron density. After fasting for 24 hrs the cells are granulated with electron dense secretory granules and after refeeding the cells are degranulated, showing clear secretory granules. The implication of the endoplasmic reticulum and the Golgi apparatus in the secretory cycle is discussed on the basis of the ultrastructural findings.Supported by a grant from the Fonds national suisse de la Recherche scientifique, Berne, Switzerland.  相似文献

Gametogenesis of Tyzzeria chalcides Probert, Roberts & Wilson, 1988, from the ocellated skink, Chalcides ocellatus , occurs within the epithelium of the gali bladder. Transmission electron microscopy reveals that macrogamonts contain 2 types of wall-forming bodies. Type I bodies are large densely stained structures associated with rough endoplasmic reticulum and the Golgi apparatus. They appear to be formed within the Golgi itself. Type II bodies are less densely stained, smaller and appear to form directly from the rough endoplasmic reticulum. Canaliculi are associated with Type I wall-forming bodies and probably function to transport the wall-forming bodies to the pellicle. Micropores occur in the pellicie and large amylopectin granules, lipid globules and dense bodies are found within the cytoplasm of the macrogamont. Mature microgamonts contain in excess of 20 microgametes, each of which has 2 flagella and an associated mitochondrion. Both types of gamont are found within a parasitophorous vacuole, in the host cell, which is filled with vesicular material on which the gamonts probably feed.  相似文献
