The morphological and functional changes during involution of hyperplastic goitre have been investigated in the adult male rat. Male wistar rats received an iodine-deficient diet for 6 months and during the last 2 months received propylthiouracil (PTU, 0.15%). By the end of this treatment (day 0), a hyperplastic goitre was obtained. A normal iodine supply was then given and PTU withdrawn. During the first 8 days of iodine refeeding, the plasma thyrotropin (TSH) remained at a high level (ten times the control value), whereas the thyroid iodide content was low on day 0, markedly increased on day 1 and decreased on day 4. Plasma T3 and T4 levels remained unchanged for 4 days and only increased on day 8. The total thyroid protein concentration was low on day 0 and then increased rapidly on day 8 (by 34%). The volume density of colloid remained low and unchanged until day 8, when it started to increase. However, the thyroid epithelial cell volume and the volume density of capillaries were raised on day 0, decreased rapidly in the next 8 days and more slowly later on. The total number of thyroid epithelial cells was considerably raised in the hyperplastic gland. It did not vary until day 16, when it decreased slowly, reaching a plateau on day 45 above the control value. The present data show that involution of hyperplastic goitre in the rat is due essentially to a decrease in thyroid epithelial cell volume and to a reduction of the increased number of capillary blood vessels present. The decrease in the number of epithelial cells is only 16.5%, suggesting that the death of thyroid epithelial cells contributes little. Half the process of involution, which occurs from days 0 to 8, is controlled by the thyroid iodide concentration rather than TSH, indicating the involvement of a thyroid autoregulatory mechanism. It must be emphasized, however, that the discontinuous pattern of epithelial cell number during involution may indicate that some cells with larger nuclei and more rapid turnover disappear more quickly after iodine refeeding.  相似文献

The length, total and cortical widths at midshaft, and robusticity of femora were measured in growing and mature male Holtzman rats. Analysis of covariance revealed significant differences in the regression coefficients (slopes) between young rats (1-4 months) and adult rats (5-16 months), indicating that, for a given increment in femoral length, young rats have a smaller increment in total femoral width than do adult rats. In addition, the results indicated that femoral expansion occurs in adult male Holtzman rats (5-16 months of age) without cortical thinning or loss of bone mass.  相似文献

N-Acetylcysteine derivatives of catechol estrogens have been isolated from the urine of adult male hooded rats with ligated bile ducts, following injection of [4-14C]2-hydroxyestradiol-17β and of [4-14C]estradiol-17β-By application of double isotope methods previously described, it was shown that 2-hydroxyestradiol-17β was converted into mercapturic acids in a yield of 6–8%, confirming two previous experiments with bile duct cannulated rats, and that estradiol-17β was converted into mercapturic acids to the extent of 3–6%. Since these figures are small, and since it has been shown that in two women estrogen mercapturic acids were not formed, it appears that this class of compound will not provide an answer to the problem of unidentified water-soluble metabolites of the estrogens.  相似文献

The estrogen receptor-alpha (ERalpha) knockout mouse (alphaERKO) lacks ERalpha throughout development; therefore, an adult model for the study of estrogen effects in male mice was recently developed using the antiestrogen ICI 182,780. However, differences between species have been noted during immunostaining for ERalpha in the male tract as well as in response to treatments with antiestrogens. Therefore, we developed the antiestrogen model in the adult male rat to test, in another species, the hypothesis that estrogen regulates fluid reabsorption in efferent ductules. Estrogen receptor in the rat was blocked using ICI 182,780 for 100-150 days. Male Sprague-Dawley rats were treated weekly with s.c. injections of ICI 182,780 (10 mg) or castor oil (as control). The effects of ICI included testicular atrophy and infertility, similar to terminal effects in the alphaERKO male. Additionally, ICI induced dilations of the rete testis and efferent ductules and a reduction in the height of the ductule epithelium, which are changes similar to those in both alphaERKO and ICI-treated mice. One difference between species was a large variation in effects on the rat efferent ductule epithelium, including a transient increase in the number of periodic acid-Schiff-positive, lysosomal-like granules. These data confirm that estrogen is required for normal function of the efferent ductules and is essential for long-term fertility in the male rodent.  相似文献

The thymus, as part of the immune-neuroendocrine axis, is greatly influenced by factors from most endocrine glands, especially the thyroid. Antithyroid drugs (carbimazole and methimazole) were used to induce hypothyroidism in rats. Histological and ultrastructural examination of the thymus showed progressive thymic involution after 4 weeks of drug treatment to the end of observations (7 weeks). The involution was characterised by increased thymocyte apoptosis and thymocyte phagocytosis by macrophages. This resulted in thymocyte depopulation, increases in numbers of interdigitating cells, alterations to mainly subcapsular and medullary epithelial cells, an apparent increase of mast cells and collagen in the capsule and septa, and increased numbers of B cells and plasma cells. Lymphoid cells immuno-reactive with MRC OX12 (which detects B cells) were observed within blood vessel walls, suggesting that they may have been moving in and out of the thymus. The administration of drugs causing hypothyroidism, therefore, also caused marked involution of the thymus.  相似文献

We investigated in greater detail the relationship between steroid structures and their binding affinities to the microsomal progesterone-binding site of the adult male rat liver. Only six steroids of the 100 compounds tested, namely, progesterone, 3 alpha-hydroxy-5 alpha-pregnan-20-one, 3 alpha,11 beta-dihydroxy-5 alpha-pregnan-20-one, 5 alpha-pregnane-3,20-dione, pregnenolone, and pregnenolone-3-sulfate, had high binding affinities to this site. Thus, we were able to draw a conclusion on the steroid structure which should be required for the best interaction with this site. That is, regardless of the whole molecule's lipophilicity, such a steroid should possess not only a planar A-B ring configuration but also the same side chains as progesterone with C-17 beta-acetyl and non-hydroxyl groups. The exception to this are hydrophilic C-3 groups, which may somewhat increase binding affinities in some cases. We compared the steroid specificity of this binding site with those of various other progesterone-binding components. As a result, this site appears to be a novel type of progesterone binder. We, furthermore, examined the relationship between this microsomal progesterone-binding site and the microsomal progesterone-metabolizing activity. The results, although preliminary, suggest that this binding site does not participate universally in the progesterone-metabolizing processes.  相似文献

The effects of bromocriptine induced hypoprolactinemia on the testicular function were studied in adult rats. Bromocriptine treatment (1500 micrograms/day for 24 days) reduced serum and pituitary Prolactin levels, indicating a decrease in prolactin secretion and synthesis. No change in reproductive organ weights was seen in treated animals. Hypoprolactinemia had no effect on plasma testosterone or androstenedione levels and testicular androstenedione content, but decreased significantly testicular testosterone content. These findings indicated that experimental hypoprolactinemia induced a decrease in testicular testosterone content without affecting androgens levels.  相似文献

Postnatal exposure of male rats to microwaves modulated the evolution of the thyrotropin level. Indeed, this treatment increased plasma TSH without affecting significantly plasma thyroxine. Moreover, the histological study of the thyroid demonstrated a high activity of this gland in irradiated animals. The modification observed in the thyrotropin level was correlated to an increase in hypothalamic noradrenaline described previously in rats submitted to the same irradiation.  相似文献

Adult male rats were maintained on 0, 5, 10, 15, and 20% casein diets to produce a series of animals having serum alpha 2u-globulin levels varying linearly from a normal of 31 micrograms/ml to a minimum of 13 micrograms/ml. In this way, it was possible to titrate endogenously the renal reabsorption and urinary excretion of this low molecular weight protein. The average maximal reabsorption rate (Tm) was established to be 9.7 micrograms/min and was reached at a renal filtered load (F alpha 2u) of 13.6 micrograms/min. These data were expressed in terms of a Tm:F alpha 2u ratio of 0.71. Below this value, the reabsorption declined from 70% to 50% of the F alpha 2u. Above 0.71, where F alpha 2u is less than the Tm, the reabsorption increased to 80-90%. It was observed that the fractional renal uptake of the alpha 2u-globulin varied linearly with the filtered load.  相似文献

Cubilin is a peripheral membrane protein that cooperates with the endocytic receptor megalin to mediate endocytosis of ligands in various polarized epithelia. Megalin is expressed in the male reproductive tract where it has been implicated in the process of sperm membrane remodeling. A potential role for cubilin in the male reproductive tract has not been explored. Using RT-PCR, we found that cubilin and megalin mRNAs are expressed in the efferent ducts, corpus and cauda epididymis, and proximal and distal vas deferens. Immunohistological analysis revealed that cubilin was expressed in nonciliated cells of the efferent ducts, principal cells of the corpus and cauda epididymis and vas deferens. Immunogold EM showed cubilin in endocytic pits, endocytic vesicles, and endosomes of these cells. The expression profile of cubilin in the male reproductive tract was coincident with that of megalin except in principal cells of the caput epididymis. Double immunogold labeling showed that cubilin and megalin co-localized within the endocytic apparatus and recycling vesicles of efferent duct cells. Neither protein was found in lysosomes. Injection of RAP, an antagonist of megalin interaction with cubilin, reduced the level of intracellular cubilin in cells of the efferent ducts and vas deferens. In conclusion, cubilin and megalin are co-expressed in cells of the epididymis and vas deferens and the endocytosis of cubilin in these tissues is dependent on megalin. Together, these findings highlight the potential for a joint endocytic role for cubilin and megalin in the male reproductive tract.  相似文献

Neonatal male rats were castrated either at 0, 6 or 24 hrs. after birth. As adults, testosterone was delivered by subcutaneous implantation of a Silastic capsule containing this hormone. The probability to display mounting behavior in presence of an estrous female was lower when the animals were castrated at 0 hr. than at 6 or 24 hrs. or when they received a subcutaneous injection of 1 microgram of testosterone propionate, at the time of castration at 0 hr. These results suggest that in the rat, during the 6 hrs. following birth, neonatal testes influence the sensitivity of the adult central nervous system to testosterone.  相似文献

Equol (EQ), a metabolite of the soy isoflavone daidzein, has well known estrogenic properties. Data from animal studies suggested that EQ may act also as an anti-androgen. However, data regarding how EQ may affect brain functions like the regulation of neuroendocrine activity and reproductive outcomes in adult male rats are still lacking. We therefore investigated the effects of EQ on sex-steroid regulated gene expression in the brain [medial preoptic area/anterior hypothalamus (MPOA/AH) and medial basal hypothalamus/median eminence (MBH/ME)], pituitary, and prostate as a reference androgen-dependent organ. Furthermore reproductive outcomes were evaluated. The anti-androgen flutamide (FLUT) served as reference compound. Male rats (n = 12 per group) were treated by gavage for 5 days with either EQ (100 or 250 mg/kg BW/day), or FLUT 100 mg/kg BW/day. All vehicle- and EQ-treated males showed successful reproductive outcomes, whereas FLUT-exposed males had severe reproductive impairments resulted in infertility. FLUT decreased relative weights of prostate, seminal vesicles and epididymides, and increased serum levels of luteinizing hormone, follicle-stimulating hormone, testosterone and 5α-dihydrotestosterone without altering prolactin levels, whereas EQ exerted opposite effects. Both EQ and FLUT decreased gonadotropin releasing hormone (GnRH) expression in the MPOA/AH. Only FLUT upregulated levels of GnRH receptor expression both in the MBH/ME and pituitary. While EQ downregulated the hypothalamic ERα and ERβ expressions, but FLUT did not. In the prostate, only FLUT upregulated both ERα and AR mRNA expression levels. Taken together, our findings are the first data that EQ did not induce anti-androgenic effects on brain, prostate and male reproductive parameters, however, estrogenic neuroendocrine and reproductive effects of EQ were observed.  相似文献
