本研究旨在用壳聚糖-聚丙烯酸纳米磁性微球纯化血红细胞超氧化物歧化酶。采用了接枝共聚法,以K2S2O8为引发剂,使壳聚糖(CTS)与聚丙烯酸(PAA)进行自由接枝共聚合成含有两性基团(-NH3,-COOH)的壳聚糖-聚丙烯酸纳米微球。化学共沉淀法制备Fe3O4磁流体,以戊二醛为交联剂,制备壳聚糖-聚丙烯酸纳米磁性微球。用傅里叶变换红外光谱仪对磁性微球结构进行检测。JEM-4000EX电镜技术对微球粒径,形貌进行表征。SOD试剂盒测定各步骤Cu-ZnSOD酶活性。结果表明,壳聚糖-聚丙烯酸纳米磁性微球有较好的粒径分布、磁响应性及蛋白吸附特性。纯化后酶比活性达6 727 U/mg,产品得率21.1%,活性回收85.7%。壳聚糖-聚丙烯酸纳米磁性微球经血液纯化血红细胞SOD具有可再生性、易操作性,其纯化效果取决于金属Cu2+的螯合程度。  相似文献

用醇类从酵母菌中释放超氧化物歧化酶   总被引:4,自引:0,他引:4  
研究了一种用醇类有机溶剂处理法从酵母菌中选择性地释放超氧化物歧化酶(SOD)的方法。当采用异丙醇浓度为90%,浸泡120min,抽提缓冲液为50mmol/L磷酸盐缓冲液(Ph7.0)时,抽提20h,SOD释放的活力为300u/ml,杂蛋白释放量最少,SOD比活达300u/mg。SOD释放率可达90%,和传统的超声波法和机械法相比,而比活提高了25倍。这种方法不需要任何复要设备,操作简单,成本低廉,在释放SOD同时,可达到初步纯化SOD的效果。  相似文献

 采用吸附、包埋、共价交联等方法固定化超氧化物歧化酶(SOD)所得固定化酶活力回收都不高,表明酶的催化反应速率受超氧阴离子自由基(O_2~-)扩散速率的控制。用海藻酸钠包埋SOD,固定化酶不表现活力,破固定化后所得的糊状物却有很高的活力。用戊二醛交联所得的固定化酶活力回收率也很低,表明ε-NH_3~+的正电荷是酶活力所必需。  相似文献

目的:研究血液中基因组DNA的简便快速提取方法。方法:取新鲜抗凝血,以红细胞裂解液除去红细胞,再破碎白细胞,除去杂蛋白,获得基因组DNA。结果:所得基因组DNA完整、无断裂,含量和纯度均较高。以所提基因组DNA作为模板能很好的扩增出p21因子启动子序列,因此该法所提取的DNA是完整可靠的。结论:该法能简便、快速、安全、廉价的提取血液中的基因组DNA,并适用于临床检测和分子生物学研究。  相似文献

玉米幼苗经过热激处理后,在随后的高温、低温、干旱和盐胁迫环境中,其存活率明显高于未热激的幼苗,表明热激能提高植物的抗热性、抗冷性、抗旱性和抗盐性,证实了玉米幼苗交叉适应现象的存在;热激还可提高谷胱甘肽还原酶(GR)和超氧化物歧化酶(SOD)的活性,且在上述4种胁迫过程中GR、SOD活性水平与玉米幼苗的存活率呈正相关,表明GR和SOD参与玉米幼苗交叉适应的形成.  相似文献

用抗坏血酸-Fe(Ⅲ)或过氧化氢为活性氧体系分别作用于牛红细胞铜锌超氧化物歧化酶(CuZn-SOD),发现酶活性降低的同时,其活性中心中Cu(Ⅱ)被还原成Cu(Ⅰ)。进一步用抗坏血酸-Fe(Ⅲ)或过氧化氢作用于马来酰亚胺标记CuZn-SOD,用ESR测定的结果表明酶分子亚基的缔合受到影响。结果提示酶的失活与活性中心中Cu(Ⅱ)的还原及酶构象的变化有关。  相似文献

目的 综合利用鲎试剂生产的废料血浆提取超氧化物歧化酶 (Superoxide dismutase,SOD),探讨鲎血SOD对耐甲氧西林金色葡萄球菌(MRSA)、对甲氧西林敏感金色葡萄球菌(MSSA)、野生型绿脓杆菌(PAO1)、大肠杆菌(E.coli)的抑菌作用.方法 采用离心分离、丙酮沉淀、热变性去杂蛋白的方法从生产鲎试剂废料血浆中提取SOD,然后把SOD加入菌液培养,24 h后观察结果.结果 SOD对耐甲氧西林金色葡萄球菌、对甲氧西林敏感金色葡萄球菌、野生型绿脓杆菌、大肠杆菌的最低抑菌浓度分别为20.02,40.05,20.02,20.02 μg/ml.结论 本方法可成功地从生产鲎试剂废料血浆中提取得到SOD粗品,实验表明鲎血SOD具有抗菌作用.  相似文献

以二硫苏糖醇(DTT)或L-抗坏血酸(Vit C)为终止剂改进的经典邻苯三酚法测定超氧化物歧化酶(SOD)活性.反应体系:50mmol/L邻苯三酚,pH8.20,总体积9ml,25℃,加一滴DTT(100mmol/L)或Vit C(5%),约50μl,终止自氧化反应.终止后,反应体系的420nm光吸收在lh内保持恒定.终止剂法的邻苯三酚自氧化率及酶活测定灵敏度与经典法相近,一个酶活单位相当于纯SOD 100μg/L.  相似文献

为考察来源于极端高温酸性环境中的超氧化物歧化酶,建立了一个由嗜酸热原体茵的sod基因编码的蛋白质的可靠三维分子结构,并对这个蛋白质进行进化踪迹分析识别出了11个踪迹残基?其中,残基Asn39,Gly105和Glu162的位置随机遍布整个结构,其余的残基却全部都明显地聚于一个靠近铁原子的子类当中?由此从在三维结构确认sod基因编码含铁的超氧化物歧化酶.此外,那些被识别的靠近铁原子的踪迹残基可能跟铁的绑定和催化功能直接相关?  相似文献

目的:研究大黄蛰虫胶囊联合维生素C钠治疗黄褐斑的临床疗效及对血清和血液流变学的影响。方法:选取2012年8月到2013年8月我院收治的黄褐斑患者140例,按照随机数字表法平均分成研究组和对照组,每组70例,研究组给予大黄蛰虫胶囊联合维生素C钠治疗,对照组给予大黄蛰虫胶囊治疗,两组均治疗1个月。3个月后比较两组临床疗效,并检测两组患者的血清睾酮(T)、雌二醇(E2)、孕酮(P)、过氧化脂质分解产物丙二醛(MDA)、超氧化物歧化酶(SOD)以及血液流变学水平的变化。结果:研究组总有效率85.7%(60/70)显著高于对照组的57.1%(40/70),两组比较差异具有统计学意义(P0.05);与治疗前和对照组相比,治疗后研究组SOD、E2显著升高,MDA、T显著降低,差异均具有统计学意义(P0.05);两组P水平均无明显变化;治疗后研究组血液流变学各指标较治疗前和对照组显著下降,差异均有统计学意义(均P0.05)。结论:大黄蛰虫胶囊联合维生素C钠治疗黄褐斑具有较好的临床疗效,可以调节患者的性激素水平,改善患者的血流变学。  相似文献

A novel metal-free low molecular weight superoxide dismutase mimic   总被引:8,自引:0,他引:8  
2-Ethyl-1-hydroxy-2,5,5-trimethyl-3-oxazolidine (OXANOH), the one-electron reduction product of the stable nitroxide radical, 2-ethyl-2,5,5-trimethyl-3-oxazolidinoxyl (OXANO), is reportedly oxidized by superoxide, and its oxidation has been proposed as a method for assaying superoxide. We find that superoxide can both reduce OXANO and oxidize OXANOH. The respective rate constants, k1 and k2, were determined using two superoxide-generating systems (xanthine oxidase/xanthine as well as ionizing radiation). OXANOH oxidation and OXANO reduction are both inhibitable by superoxide dismutase, pH-dependent (4.5-9.3), and result in a steady state distribution of [OXANO] and [OXANOH], independent of their initial concentrations, i.e. the OXANO/OXANOH couple exhibits a metal-independent superoxide dismutase-like function. Thus it provides a prototype for future development of improved low molecular weight superoxide dismutase mimics which will also function in cellular hydrophobic (aprotic) compartments such as membranes.  相似文献

The mechanism by which mutated copper-zinc superoxide dismutase (SOD1) causes familial amyotrophic lateral sclerosis is believed to involve an adverse gain of function, independent of the physiological antioxidant enzymatic properties of SOD1. In this study, we have observed that mutant SOD1 (G41S, G85A, and G93A) but not the wild type significantly reduced the stability of the low molecular weight neurofilament mRNA in a dosage-dependent manner. We have also demonstrated that mutant SOD1 but not the wild type bound directly to the neurofilament mRNA 3'-untranslated region and that the binding was necessary to induce mRNA destabilization. These observations provide an explanation for a novel gain of function in which mutant SOD1 expression in motor neurons alters an intermediate filament protein expression.  相似文献

Paraquat exerts a cytotoxic effect of Chinese hamster ovary cells in culture via the superoxide radical (O2. We have described a superoxide dismutase (SOD) mimic based on manganese (DF-Mn) which consists of a one-to-one complex between desferrioxamine B (Desferal) and MnO2. It is a small molecular weight molecule, easy to prepare and possesses considerable stability. It is now shown to protect mammalian cells from paraquat toxicity. Thus, 20 μM DF-Mn affords up to complete protection against the cytotoxicity of 200 μM paraquat in Chinese hamster ovary cells. Desferrioxamine B or MnO2 alone gave no protection. MnCl2 or catalase provided little or no protection against the paraquat, respectively. Equivalent amounts of human Cu-Zn SOD in terms of activity, also provided no protection. Copper diisopropylsalicylate (CuDIPS) provided limited, yet significant, protection, but this is explained in terms other than SOD activity. Finally, at higher concentrations, purified human SOD, exerts a limited toxicity as well as a protective ability against paraquat (similar to DF-Mn) both of which are eliminated upon heat denaturation of the enzyme. It appears that the SOD mimic, DF-Mn, can enter mammalian cells and can protect against the cytotoxic effects of O2.  相似文献

A simple and rapid procedure for the purification of superoxide dismutase (SOD) from bovine erythrocytes by means of affinity-purified anti-SOD antibodies coupled to Sepharose 4B is reported. The method allowed 80% recovery of electrophoretically pure enzyme directly from hemolyzed red blood cells. The mild elution conditions (3.5 M MgCl2) and the stability of the immunoadsorbent permit the column to be reused several times (greater than 50). The physicochemical properties of the enzyme obtained by this method are identical to those of the native protein.  相似文献

Soluble extracts of Plectonema boryanum have been shown to contain a single, electrophoretically distinct, superoxide dismutase. The enzyme has been isolated and has been found to be an iron-containing enzyme similar to that described from the periplasm of Escherichia coli. It contains 1 Fe3+/mole of enzyme. The molecular weight was approximately 36 500, and the enzyme appeared to be composed of two subunits of equal size joined by non-covalent interactions. ESR data are presented, as are the results of amino acid analysis.  相似文献

1. Several apparent molecular weights (mol. wt) are reported for plasma or serum extracellular superoxide dismutase (EC SOD) activity. This study found species-dependent heterogeneity for apparent mol. wt using gel filtration with Sephadex G-150. 2. EC SOD activity in rabbit and guinea-pig serum, measured by a modified pyrogallol assay, eluted just before ceruloplasmin activity, but rat and bovine serum activity eluted after ceruloplasmin (apparent mol. wt of 142,000 and 73,000, respectively). 3. The heterogeneity between rat and rabbit serum was not eliminated by substituting a cytochrome-c-based SOD assay for the pyrogallol method, by substituting lung extracts for serum, by analysing a mixture of rat and rabbit serums, nor by analysing hemolysed serum. The apparent mol. wt of bovine serum EC SOD activity was not duplicated by gel filtration analysis of a mixture of bovine cytosolic SOD and albumin. 4. In conclusion, species-specific variation in apparent mol. wt for serum EC SOD activity was demonstrated under several circumstances.  相似文献

为探索简便实用纯化SOD的工艺路线,以人或猪血红细胞溶血上清液,经铜胺中空纤维透析器(分子量截留值为15kD)透析和超滤,收集分子量大于15.0kD的物质,再加热60℃10min,离心取上清即得。Cu、ZnSOD和MnSOD分子量分别为32.0kD和80.0kD。人血和猪血纯化的SOD总收率分别为88.2%和89.2%,比活性分别为17429U/mg和18228U/mg。工艺简便实用,适于工业纯化生产。  相似文献
