A complete protocol for the in vitro induction of Eclipta alba tetraploids has been optimized to enhance the wedelolactone content, an anti-cancerous compound. The effects of different concentrations of colchicine (0, 0.01, 0.05, 0.1, 0.2 and 0.3%; w/v) along with treatment durations (12, 24, 36 and 48 h) were investigated on shoot tip (ST) and nodal segment (NS). The treated explants were then incubated on Murashige and Skoog (MS) medium having 1.5 mg L?1 N6-benzylaminopurine and 0.5 mg L?1 α-napthalene acetic acid for shoot regeneration and afterward root was induced on 1.0 mg L?1 indole-3-acetic acid enriched ½MS medium. The tetraploids of E. alba were proficiently induced by the treatment of 0.1% colchicine for 24 h. The highest tetraploid induction efficiency was obtained from ST (30.56%) in comparison to the NS (22.22%). Analysis by spectrophotometry and flow cytometry showed that colchicine treated plants contained higher quantity of DNA than diploid plants. Cytological studies demonstrated doubled the chromosome number in tetraploids (2n?=?4x?=?44) than diploids (2n?=?2x?=?22). The ploidy level enhancement lead to alteration of other traits, like increased plant height, stem diameter, leaf size, stomatal size and chlorophyll content. As determined through high performance thin-layer chromatography, the ultimate achievement of this technique is the higher accumulation of wedelolactone in tetraploid plants (300.32 µg g?1 dry weight) in evaluation to in vitro diploid (131.31 µg g?1 dry weight) and in vivo diploid mother plants (93.26 µg g?1 dry weight), thus improving the pharmaceutical value of E. alba.  相似文献

During recent years, hazel cell suspension culture has been significantly considered as a new important source of paclitaxel. Artificial polyploidy alters different characters in plants which results in amplifying the secondary metabolites in medicinal plants. In this paper, the effects of tetraploidy induction on paclitaxel content and gene expression in hazel cell suspension culture were investigated. Various concentrations of colchicine and duration of exposure in solid and liquid media were examined and the ploidy level of cells was determined using flow cytometric analysis. The tetraploid cells were obtained from 0.2% colchicine in the exposure time of 5 and 6 days in solid medium and 0.3% colchicine in the exposure time of 3 and 4 days in liquid medium. Tetraploid cells were employed to prepare cell suspension. 3 µM of phenylalanine and 0.05 mM of vanadyl sulfate were added to both tetraploid and diploid (control) suspension to elicit paclitaxel induction. High performance liquid chromatography analysis demonstrated that the tetraploid cell suspensions produced paclitaxel of about [9.88 µg g?1 (DW)] 1.7-fold compared with diploid cells [5.74 µg g?1 (DW)]. The application of phenylalanine and vanadyl sulfate increased the concentration of paclitaxel in both diploid and tetraploid cells. Moreover, qRT-PCR analysis showed that the expression of GGPPS gene was significantly increased and PAL gene expression was altered after tetraploidization.  相似文献

A practical and reliable method for in vitro tetraploidization of Sphagneticola calendulacea (L.) Pruski [synonym Wedelia chinensis (Osbeck) Merrill] has been established to enhance the production of wedelolactone. Shoot tip and nodal explants from in vitro-grown culture (2n?=?50) were exposed to the antimitotic chemical, i.e., colchicine, at various concentrations (0, 0.025, 0.05, 0.1, 0.3, and 0.5%; w/v) for 12, 24, 36, 48, and 60 h. The treated explants were then incubated and proliferated on Murashige and Skoog (MS) medium fortified with 0.2 mg l?1 thidiazuron and 0.05 mg l?1 naphthalene acetic acid, followed by root induction in 1.0 mg l?1 indole-3 acetic acid enriched ½MS medium. Treatment of shoot tips with 0.05% colchicine for 24 h supported the maximum rate of survival (63.33%) of explants as well as tetraploid induction (42.93%). Morphological, stomatal, and cytological characteristics along with the secondary metabolite content of the in vitro tetraploids were compared to that of diploids. The recovered tetraploid plants possessed superior plant height, stem diameter, leaf size, root number, and increased length and width of stomata but decreased stomatal frequency. The tetraploid plants demonstrated twice the chromosome number (2n?=?4x?=?100) than the diploids as confirmed through cytology, spectrophotometry and flow cytometry. High-performance thin-layer chromatography showed a significant enhancement in the wedelolactone content of tetraploid plants (541.48 µg g?1 of dried sample) in comparison to diploid plants (325.43 µg g?1 of dried sample), signifying the prospective of this technique for the trade value improvement.  相似文献

The effect of a seaweed-derived biostimulant (Kelpak® at 1, 2.5 and 5 % dilution; v/v) on the growth, endogenous cytokinin (CK) and phytochemical content in Eucomis autumnalis (Mill.) Chitt. under hydroponic conditions was evaluated. After 4 months, the stimulatory effect of Kelpak® treatments was more noticeable in the underground organs than in the aerial organs. Total endogenous CK was also higher in plants treated with Kelpak® (c.a. 1000–1200 pmol g?1 DW) compared to control plants (860 pmol g?1 DW). Isoprenoid CKs (which mainly accumulated in the aerial organs) were more dominant than aromatic-type CKs across all the treatments. A total of 11 bioactive chemicals (8 phenolic acids and 3 flavonoids) and eucomic acid known for their diverse biological activities were quantified in the samples. The most abundant compound was p-coumaric acid (6.5 µg g?1 DW) and it was approximately sevenfold higher in 2.5 % Kelpak®-treated plants than in the control. It was also noteworthy that syringic acid only occurred in the underground organs of 5 % Kelpak®-treated plants. Eucomic acid which is a major bioactive compound in E. autumnalis was significantly enhanced in Kelpak® treatments, and the leaves accounted for more than 70 % of the overall content. Thus, Kelpak® elicited a significant influence on the growth, endogenous CK and phytochemical content in E. autumnalis. These findings provide additional evidence of the enormous potential of Kelpak® as a useful biostimulant with practical applications in various agricultural endeavours.  相似文献

The effect of rutin and gallic acid on growth, phytochemical and defense gene activation of rice (Oryza sativa L.) was investigated. The seeds of rice were primed with different concentrations of rutin and gallic acid (10–60 µg mL?1) to explicate the effect on germination on water agar plates. Further, to study the effect of most effective concentrations of gallic acid (60 µg mL?1) and rutin (50 µg mL?1), greenhouse pot experiment was set up to determine the changes in growth, antioxidant and defense parameters. The results revealed more pronounced effect of gallic acid on total chlorophyll and carotenoids as well as on total flavonoid content and free radical scavenging activities. Gene expression analysis of OsWRKY71, PAL, CHS and LOX genes involved in strengthening the plant defense further validated the results obtained from the biochemical analysis. Microscopic analysis also confirmed reduction in total reactive oxygen species, free radicals like H2O2 and O2 ? by exogenous application of gallic acid and rutin. The data obtained thus suggest that both gallic acid and rutin can affect the growth and physiology of rice plants and therefore can be used to develop effective plant growth promoters and as substitute of biofertilizers for maximizing their use in field conditions.  相似文献

Common reed (Phragmites australis (Cav.) Trin. ex Steud.) plants were harvested from four natural water ecosystems of the Bogdanka river catchment (Poznań, Poland) four times throughout the 2014 vegetative season. Over the year, average metal contents followed different decreasing trends according to the analyzed tissue: Zn > Cu ≈ Pb > Cd (rhizomes) and Zn > Pb > Cu > Cd (leaves), and mean translocation ratios (leaves/rhizomes) were found as follows: 0.93, 0.70, 0.65, 0.40 for Zn, Pb, Cd and Cu, respectively. Metal content increased gradually during the growing season, and in the case of Cu, Cd and Pb exceeded the upper limit of average concentration detected in plants from natural ecosystems. However, the content of salicylic acid did not follow the increase of metal accumulation. In rhizomes, the highest production of the metabolite was observed in May and reached 324 ng g?1 fresh weight (FW) (mean value). Afterwards, a significant drop to 50 ng g?1 FW was observed. Simultaneously, the highest values of total salicylic acid in P. australis leaves were observed in July and accompanied the intensive development of the aboveground biomass of the plant (11.3 µg g?1 FW–mean value). Subsequently, its content in leaves showed a significant decrease down to 2.1 µg g?1 FW in November. Multivariate regression analysis revealed significant interactions between analyzed metals influencing the plant response to metal-derived stress. Cu and Zn showed antagonistic properties considering their uptake and the induction of salicylic acid biosynthesis, whereas non-essential metals (Pb and Cd) acted similarly and stimulated the formation of salicylic acid glucoside.  相似文献

Hoplosternum littorale is an Amazon fish that lives in urban areas surrounded by polluted igarapés, where elevated copper concentrations eventually occur. The central goal of this study was to evaluate the associated effects of high temperature and copper contamination on survival time and biochemical responses of the Amazonian fish species H. littorale. We exposed fish to two nominal dissolved copper concentrations (50 and 500 µg l?1) and combined temperatures of 28 and 34°C. Our findings showed that the combination of these variables affects the survival time of this species. The activity of the biotransformation enzymes ethoxyresorufin-O-deethylase and glutathione-S-transferase showed no alterations in fish within all treatments. The increase of reactive oxygen species and the decrease in potential total antioxidant capacity promoted the imbalance in the antioxidant system. An induction in superoxide dismutase activity occurred in fish exposed to copper concentrations of 50 and 500 µg l?1 at both temperatures, suggesting liver impairments. Thus, we suggest that H. littorale is sensitive to copper, and this sensitivity is increased further with exposure to high temperatures, particularly in the survival time and reactive oxygen species formation of this fish species.  相似文献

Rhododendron tomentosum Harmaja (Ledum palustre), a peat bog plant from Ericaceae family, has been used in traditional medicine as the anti-arthritis agent. Although modern researches confirm its anti-inflammatory properties, it remains threatened by habitat degradation and possibilities to collect this endangered species from its natural environment for further biological activity studies are limited. Therefore, R. tomentosum liquid in vitro cultures were established as the alternative source of that valuable plant material. Schenk–Hildebrandt medium with 24.60 μM 2-isopentenyladenine and 592.02 μM adenine provides intensive growth and proper morphology of the obtained microshoots. The R. tomentosum biomass was scaled up using the various bioreactors (immersion, temporary immersion and spraying systems) for better growth and improved volatile oil production. The largest biomass accumulation (fresh weight?=?250 g l?1, growth index?=?280, dry weight?=?20 g l?1) and essential oil content (0.5% v/m) were achieved with application of commercially available RITA® bioreactor. GC/MS analysis revealed the high content of p-cymene (6.9%), alloaromadendrene (5.5%), shyobunone (8.2%) and ledene oxide (II) (13.0%) in the volatile fraction obtained from RITA® system. The biomass growth parameters and production profile in terms of essential oil and selected terpenoid compounds were determined during the 2 month period. The influence of culture conditions and bioreactor construction on the growth and volatile oil production in R. tomentosum biomasses was discussed.  相似文献

Objectives

To evaluate the effects of 12 biotic and abiotic elicitors for increasing the production of plumbagin in Plumbago indica root cultures.

Results

Most elicitors showed minimal effects on the root dry weight, except for 250 mg chitosan l?1 and 10 mM l-alanine that markedly decreased root biomass by about 40 % compared to the untreated root cultures (5 g l?1). Treatments with 100 µM AgNO3 significantly increased intracellular plumbagin production by up to 7.6 mg g?1 DW that was 4-fold more than the untreated root cultures (1.9 mg g?1 DW). In contrast, treatments with 150 mg chitosan l?1, 5 mM l-alanine, and 50 µM 1-naphthol significantly enhanced the extracellular secretion of plumbagin by up to 10.6, 6.9, and 5.7 mg g?1 DW, respectively, and increased the overall production of plumbagin by up to 12.5, 12.5, and 9.4 mg g?1 DW, respectively.

Conclusions

Chitosan (150 mg l?1), l-alanine (5 mM), and 1-naphthol (50 µM) were the best elicitors to enhance plumbagin production in P. indica root cultures.

We report synthesis of silver nanoparticles (AgNPs) from Streptomyces xinghaiensis OF1 strain, which were characterised by UV–Vis and Fourier transform infrared spectroscopy, Zeta sizer, Nano tracking analyser, and Transmission electron microscopy. The antimicrobial activity of AgNPs alone, and in combination with antibiotics was evaluated against bacteria, namely Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis, and yeasts viz., Candida albicans and Malassezia furfur by using micro-dilution method. The minimum inhibitory concentration (MIC) and minimum biocidal concentration of AgNPs against bacterial and yeast strains were determined. Synergistic effect of AgNPs in combination with antibacterial and antifungal antibiotics was determined by FIC index. In addition, MTT assay was performed to study cytotoxicity of AgNPs alone and in combination with antibiotics against mouse fibroblasts and HeLa cell line. Biogenic AgNPs were stable, spherical, small, polydispersed and capped with organic compounds. The variable antimicrobial activity of AgNPs was observed against tested bacteria and yeasts. The lowest MIC (16 µg ml?1) of AgNPs was found against P. aeruginosa, followed by C. albicans and M. furfur (both 32 µg ml?1), B. subtilis and E. coli (both 64 µg ml?1), and then S. aureus and Klebsiella pneumoniae (256 µg ml?1). The high synergistic effect of antibiotics in combination with AgNPs against tested strains was found. The in vitro cytotoxicity of AgNPs against mouse fibroblasts and cancer HeLa cell lines revealed a dose dependent potential. The IC50 value of AgNPs was found in concentrations of 4 and 3.8 µg ml?1, respectively. Combination of AgNPs and antibiotics significantly decreased concentrations of both antimicrobials used and retained their high antibacterial and antifungal activity. The synthesis of AgNPs using S. xinghaiensis OF1 strain is an eco-friendly, cheap and nontoxic method. The antimicrobial activity of AgNPs could result from their small size. Remarkable synergistic effect of antibiotics and AgNPs offer their valuable potential in nanomedicine for clinical application as a combined therapy in the future.  相似文献

The antioxidative phytochemicals in globe artichoke (Cynara scolymus L.) have received increasing attention for their health-promoting properties related to the high levels of caffeoylquinic acids and flavones in capitula and leaves. Since phytochemicals in plants vary in relation to both biotic and abiotic factors, we explored the possibility to use in vitro-derived materials as a source of antioxidant compounds. Two suspension cultures, an anthocyanin-producing and not-producing cultures, and the sourced callus were evaluated in terms of their total polyphenol (TP) content and qualitative profile, total anthocyanin (TA) content and antioxidant activity (AA). TP and TA content were quantified by spectrophotometric assays, while the polyphenol profile was estimated by HPLC analysis. AA was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. Growth kinetics and polyphenol accumulation were investigated for 25 days in red suspension cultures. The latter accumulated a higher TP and TA content (25.7 and 2.61 g kg?1 of DM, respectively) than calluses and green suspension cultures. During cell growth, the TA content in red suspension cultures ranged from 1.43 to 2.41 g kg?1 of DM. Optimum production of polyphenols was achieved on day 25 of culture; a positive correlation existed between TP and both DPPH (r?=?0.84) and FRAP (r?=?0.85). The 1,5–O-dicaffeoylquinic acid and cyanidin malonylglucoside (21.18 and 1.24 g kg?1 of DM, respectively) were the primary compounds. The results of this investigation indicate that cell suspension of globe artichoke could represent a potential source of bioactive compounds with high antioxidant properties for industrial applications.  相似文献

This study evaluates the production of biomass and mycosporine-like amino acids (MAAs) throughout the year in Gracilaria vermiculophylla (Rhodophyta) collected in Ria de Aveiro (Portugal). The algae were grown in outdoor tanks in seawater with the addition of fishpond effluents under two different water flows (100 and 200 L h?1) in an integrated multi-trophic aquaculture (IMTA) system (tanks 1200 L; 1.5 m2) and different algal densities (3, 5, and 7 kg m?2). MAA content in IMTA seaweeds was significantly affected by the interaction of time and stocking density, but not by the water flow. The highest MAA content was observed in April (about 3.13 mg g?1 DW) followed by May (1.79 mg g?1 DW). Seaweed biomass productivity was higher in May (372.06 g DW m?2 week?1) than in April (353.40 g DW m?2 week?1). Four MAAs were identified by HPLC and electrospray ionization mass spectrometry (ESI-MS) in G. vermiculophylla: Porphyra-334, Shinorine, Palythine and Asterina-330. The highest levels of Porphyra-334 and Shinorine were reached from November to January and the Palythine + Asterina-330 from April to August. Taking into account the average biomass and MAA production of G. vermiculophylla growing in this IMTA system (8.56 g of MAA in 18 m2 culture along 8 months; 35.5% produced in April), a total amount of 71.33 g MAA year?1 could be produced in this system by scaling up to 100 m2. The sharptooth catfish Clarias gariepinus is an emerging global invader for which control strategies might include the use of piscicides such as rotenone. Experimental exposure demonstrated that C. gariepinus was less susceptible to rotenone than most other fish species, with unexpected survival observed at rotenone concentrations of 87.5 and 100 µg L?1. C. gariepinus were also observed exhibiting avoidance behaviour to rotenone treated water and were found to be capable of recovering from rotenone exposure. As such, effective eradication might not be attainable even at a dose exceeding 100 µg L?1 with exposures of longer than 24 h. This exposure scenario may pose an unacceptable risk to non-target fauna and highlights the difficulty associated with managing current and future invasions.  相似文献

Thymus daenensis Celak. is an aromatic herb used as a popular medicine and its natural products in the form of extracts and essential oil have significant economic values in Iran. We hypothesized that spraying plants grown under deficit irrigation system with chitosan can be considered as an applicable method to enhance essential oil and antioxidant activity in thyme. Response of thyme to three irrigation regimes including well-watered, moderate stress, and severe stress along with three levels of chitosan application rates 0, 200, and 400 μL L?1 was evaluated in a 2-year study in 2014 and 2015. Drought stress condition significantly shortened phenologic stages, more specifically in the first (establishment) year. All growth parameters were reduced dramatically as drought stress intensified. Imposing even moderate stress reduced leaf area as much as 59 and 44% in the first year and the second year, respectively. Biomass yield of plants grown under severe drought stress decreased substantially, whereas essential oil content and the share of thymol in thyme oil which possesses the greatest degree of biological activity improved. Maximum oil yield (1.50 g plant?1) was obtained from plants under mild drought stress when sprayed with 400 μL L?1 chitosan in the second year when plants were well-established. Foliar applications of chitosan reduced the adverse effect of water deficit on oil yield and improved thymol content of the essential oil. Chitosan also increased secondary metabolites including α-terpinene, p-cymene, γ-terpinene, thymol, carvacrol and β-caryophyllene. Leaf flavonoid reduced under deficit irrigation while more phenol was found in plants grown under deficit irrigation. The essential oil of thyme exhibited antioxidant property when the plants were sprayed with 400 μL L?1 chitosan. The results of this study indicated that thyme can be grown successfully under moderate stress and that application of chitosan elicitor can to some degree compensate the negative impact of deficit irrigation on its biomass and essential oil yield.  相似文献

Wild tubers of Dioscorea bulbifera (Db) and Dioscorea hirtiflora (Dh) mainly used as sources of famine food and in herbal preparations are often indiscriminately collected in Africa and Asia. Therefore, there is the need to complement wild sourcing of the tubers to promote their conservation. The present study reports in vitro tuberous induction (80%) for the first time from Dh cultured on MS?+?NAA (2.5 mg/L) with IC50 of 472.5?±?1.77 µg/mL using DPPH, whereas tuberous root (60%) from Db on MS?+?Kn (2.5 mg/L)?+?NAA (0.25 mg/L) had IC50 of 26.97?±?1.00 µg/mL. Genetic fidelity assessment of in vitro plants compared to the wild plants revealed similar amplicon size of amplified DNA using trnH–psbA and rbcL. Similarly, micromorphological diagnostic features like oil gland, crystals (raphides), trichome and stomata type were obtained from the epidermal peels of the wild and in vitro plants. The ethyl acetate (EtOAc) extract of the flesh of Dh (wild) had the highest catechin content (108.3?±?0.69 µg/g DW). Protocatechuic acid was highest in the methanol (MeOH) extract of the flesh of Dh (0.42?±?0.02 µg/g DW), while it was detected in trace amount in the in vitro tuberous roots of MeOH extracts of Dh treated with NAA. The in vitro protocol developed in this study could be employed to multiply Dioscorea bulbifera L. and Dioscorea hirtiflora Benth. to offer genetically stable clones for the optimization of bioactive compounds and germplasms conservation.  相似文献

Rhodotorula glutinis CCT 2182, Rhodosporidium toruloides CCT 0783, Rhodotorula minuta CCT 1751 and Lipomyces starkeyi DSM 70296 were evaluated for the conversion of sugars from Brazilian molasses into single-cell oil (SCO) feedstock for biodiesel. Pulsed fed-batch fermentations were performed in 1.65 l working volume bioreactors. The maximum specific growth rate (µmax), lipid productivity (Pr) and cellular lipid content were, respectively, 0.23 h?1, 0.41 g l?1 h?1, and 41 % for Rsp. toruloides; 0.20 h?1, 0.27 g l?1 h?1, and 36 % for Rta. glutinis; 0.115 h?1, 0.135 g l?1 h?1, and 27 % for Rta. minuta; and 0.11 h?1, 0.13 g l?1 h?1, and 32 % for L. starkeyi. Based on their microbial lipid productivity, content, and profile, Rsp. toruloides and Rta. glutinis are promising candidates for biodiesel production from Brazilian molasses. All the oils from the yeasts were similar to the composition of plant oils (rapeseed and soybean) and could be used as raw material for biofuels, as well as in food and nutraceutical products.  相似文献

The aim of this work was to evaluate the effects of co-inoculation with phosphate-solubilizing and nitrogen-fixing rhizobacteria on growth promotion, yield, and nutrient uptake by wheat. Out of twenty-five bacteria isolated from the rhizosphere soils of cereal, vegetable, and agro-forestry plants in eastern Uttar Pradesh, three superior most plant growth-promoting (PGP) isolates were characterized as Serratia marcescens, Microbacterium arborescens, and Enterobacter sp. based on their biochemical and 16S rDNA gene sequencing data and selected them for evaluating their PGP effects on growth and yield of wheat. Among them, Enterobacter sp. and M. arborescens fixed significantly higher amounts (9.32?±?0.57 and 8.89?±?0.58 mg Ng?1 carbon oxidized, respectively) of atmospheric nitrogen and produced higher amounts (27.06?±?1.70 and 26.82?±?1.63 TP 100 µg mL?1, respectively) of IAA in vitro compared to S. marcescens (8.32?±?0.39 mg Ng?1 carbon oxidized and 21.29?±?0.99 TP 100 µg mL?1). Although both M. arborescens and S. marcescens solubilized remarkable amounts of phosphate from tricalcium phosphate likely through production of organic acids, however, Enterobacter sp. was inactive. The effects of these three rhizobacteria were evaluated on wheat in alluvial soils of the Indo-Gangetic Plain by inoculation of plants with bacterial isolates either alone or in combinations in both pot and field conditions for two successive years. Rhizobacterial inoculation either alone or in consortium of varying combinations significantly (P?≤?0.05) increased growth and yield of wheat compared to mock inoculated controls. A consortium of two or three rhizobacterial isolates also significantly increased plant height, straw yield, grain yield, and test weight of wheat in both pot and field trials compared to single application of any of these isolates. Among the rhizobacterial treatment, co-inoculation of three rhizobacteria (Enterobacter, M. arborescens and S. marcescens) performed best in promotion of growth, yield, and nutrient (N, P, Cu, Zn, Mn, and Fe) uptake by wheat. Taken together, our results suggest that co-inoculation of Enterobacter with S. marcescens and M. arborescens could be used for preparation of an effective formulation of PGP consortium for eco-friendly and sustainable production of wheat.  相似文献

The land crab Cardisoma guanhumi is one of the most common species in mangroves of the American Atlantic coast and Caribbean islands however, studies of its effects on the physical and chemical soil properties are scarce. This study compares specific physicochemical properties of soil between C. guanhumi burrows (B) and adjacent zones (AZ), and provides the first insights on their role as an ecosystem engineer in mangroves. The study was conducted in an estuarine system dominated by Rhizophora mangle, located at the Río Chico estuary, Miranda state of Venezuela. Random soil samples were taken digging each burrow until reaching the bottom and at the same depth for AZ. Data analysis was carried out using Bayesian inference. Credible mean differences between B and AZ, were found for sand (B?=?26.53?±?10.76, AZ?=?17.25?±?5.7%), silt (B?=?73.16?±?10.77, AZ?=?82.42?±?5.69%), pH (B?=?8.71?±?0.36, AZ?=?9.12?±?0.30), soil organic matter (SOM, B?=?0.43?±?0.21, AZ?=?0.17?±?0.06%), total N (TN, B?=?786?±?232, AZ?=?529?±?107 µg g?1), Mg (B?=?4.42?±?0.60, AZ?=?3.48?±?0.71 cmolc kg?1) and K (B?=?0.12?±?0.05 AZ?=?0.06?±?0.02 cmolc kg?1). Chemical variables as SOM, K, Mg and TN showed the highest values of effect size (>?1.4). With the exception of the pH, all chemicals variables—which were different between B and AZ—showed strong and decisive evidences of correlations with SOM. When SOM variable was controlled, the relationships between pH–TN, TN–K and Mg–K decreased, even though the correlation evidence between each pair remained. Differences in chemical contents found in B respect to AZ suggest that the activities of C. guanhumi (feeding, moulting, excretion and defecation) within their burrows promote the spatial heterogeneity of mangrove soils.  相似文献

The effects of plant growth regulators (PGRs) and organic elicitors (OEs) on in vitro propagation of Eucomis autumnalis was established. Three-year-old ex vitro grown plants from organogenesis of E. autumnalis and somatic embryogenesis (previously reported protocol) of Drimia robusta were investigated for antibacterial activity. In vitro propagation from leaf explants of E. autumnalis was established using different PGRs and OE treatments for mass propagation, biomass production and bioactivity analysis to supplement the use of wild plant material. Prolific shoots (16.0?±?0.94 shoots per explant) were obtained with MS (Murashige and Skoog in Physiol Plant 15:473–497, 1962) medium containing 100 mg l?1 haemoglobin (HB), 10 µM benzyladenine (BA) and 2 µM naphthaleneacetic acid (NAA). The shoots were rooted effectively with a combination of 2.5 µM indole-3-acetic acid and 5.0 µM indole-3-butyric acid. The plantlets were successfully acclimatized in a vermiculite-soil mixture (1:1 v/v) in the greenhouse. Three-year-old ex vitro-grown E. autumnalis and D. robusta plants derived via organogenesis and somatic embryogenesis respectively exhibited antibacterial activity and varied with PGR and OE treatments, plant parts and bacteria. The leaves of E. autumnalis ex vitro-derived from a combination of HB, BA and NAA followed by the individual treatments of BA and HB gave the best antibacterial activities (<?1 mg ml?1: minimum inhibitory concentration from 0.098 to 0.78 mg ml?1) against all tested pathogenic bacteria (Bacillus subtilis, Enterococcus faecalis, Micrococcus luteus, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa). The bulbs of D. robusta ex vitro-derived from solid culture with 10 µM picloram, 1 µM thidiazuron and 20 µM glutamine exhibited good antibacterial activity against E. faecalis, M. luteus and S. aureus when compared with other treatments and mother plants. The ex vitro-grown E. autumnalis and D. robusta biomass produced with PGRs along with OE treatments confirmed a good potent bioresource and can be used as antibacterial agents. The in vitro plant regeneration of E. autumnalis and D. robusta protocols and ex vitro plants could be used for conservation strategies, bioactivity and traditional medicinal use.  相似文献

The aim of the study was to evaluate the biosynthesis and exudation of 10 low-molecular weight organic acids (LMWOAs) into the rhizosphere with a simultaneous analysis of the acid contents in the roots and leaves of 9 Salix taxa growing on two experimental areas, differing in their concentrations of copper (Cu), lead (Pb) and zinc (Zn) in the soil (Area 1—low, Area 2—high concentration). The obtained results reveal a significant difference in the phytoextraction of the tested Salix taxa for the analysed metals in both areas. The highest contents of Cu, Pb and Zn were observed for all Salix collected from Area 2, especially in S. × smithiana roots (116 ± 8.76, 87.84 ± 7.30 and 203.42 ± 14.62 mg kg?1 DW, respectively). The results obtained in Area 2 also revealed acidification of the rhizosphere and a higher concentration of acids, mainly oxalic, malic, malonic, acetic and citric acids. Contents of oxalic, malic, acetic and citric acids increased in the roots of Salix taxa from Area 2, while in the leaves formic and succinic acids were also present. S. × smithiana was the taxon with the highest concentration of acids in the rhizosphere and roots (73.48 ± 6.77 and 49.79 ± 2.65 μM 100 g?1 DW, respectively), while in leaves a higher content was observed for S. alba and S. viminalis ‘PR’ taxa (78.12 ± 3.95 and 71.12 ± 3.75 μM 100 g?1 DW, respectively).  相似文献
