Male and female meiosis in grasshoppers   总被引:2,自引:2,他引:0  
Male meiosis in the grasshopper Stethophyma grossum is well known as an example of proximal chiasma localisation. An investigation of female meiosis in oocytes of this species shows that both the frequency and distribution of chiasmata are quite different from the male situation. Mean chiasma frequency per cell (14.98) in considerably higher in females than in males (11.28) which agrees with the trend established in other comparative studies of male and female meiosis. More strikingly, males and females also show not only different but quite opposite patterns of chiasma distribution. In spermatocytes of males, chiasmata are strictly localised proximally in most bivalents, but in oocytes of females very few chiasmata form in proximal regions and nearly all chiasmata form either in distal or interstitial regions. The genetical significance of these findings is considered.  相似文献

Meiosis involves reciprocal exchange of genetic information between homologous chromosomes to generate new allelic combinations. In cereals, the distribution of genetic crossovers, cytologically visible as chiasmata, is skewed toward the distal regions of the chromosomes. However, many genes are known to lie within interstitial/proximal regions of low recombination, creating a limitation for breeders. We investigated the factors underlying the pattern of chiasma formation in barley (Hordeum vulgare) and show that chiasma distribution reflects polarization in the spatiotemporal initiation of recombination, chromosome pairing, and synapsis. Consequently, meiotic progression in distal chromosomal regions occurs in coordination with the chromatin cycles that are a conserved feature of the meiotic program. Recombination initiation in interstitial and proximal regions occurs later than distal events, is not coordinated with the cycles, and rarely progresses to form chiasmata. Early recombination initiation is spatially associated with early replicating, euchromatic DNA, which is predominately found in distal regions. We demonstrate that a modest temperature shift is sufficient to alter meiotic progression in relation to the chromosome cycles. The polarization of the meiotic processes is reduced and is accompanied by a shift in chiasma distribution with an increase in interstitial and proximal chiasmata, suggesting a potential route to modify recombination in cereals.  相似文献

The problem of extreme localisation of chiasmata in the grasshopper species Bryodema tuberculata has been reinvestigated, using C-banding, Q-banding and benzimidazol techniques. These techniques reveal the precise localisation of heterochromatin in different chromosomes. Single or double heterochromatic blocks are present near the centromeric regions, except in chromosomes 5 and 11, which have larger blocks. These two chromosomes possess a distal chiasma while the other autosomes have a proximal chiasma. The results with regard to the distribution of chiasmata, in relation to the localisation of heterochromatin, as well as the existence of a short arm, are compared with the earlier observations of White, and discussed briefly.  相似文献

From an analysis of metaphase I bivalent configurations in Zea mays L. it was possible to determine the effects of two supernumerary elements on chiasma formation. Both the B chromosome and abnormal chromosome 10 increased chiasma frequency. In addition to enhancing total exchanges, both elements caused a redistribution of chiasmata from distal to more proximal locations.  相似文献

An examination of the meiotic pattern of chromosome 1 isolated from a feral mouse population and containing a double insertion (Is) of homogeneously staining regions (HSRs) was carried out. The region delineated by the proximal breakpoint of Is(HSR;1C5) 1Icg and the distal breakpoint of Is(HSR;1E3)2Icg is desynapsed during the early pachytene stage and heterosynapsed at the midpachytene, as shown by electron microscopic analysis of synaptonemal complexes. The HSRs have no effect on the segregation of chromosome 1 in heterozygous mice. The lack of homosynapsis in the region under study causes chiasmata redistribution in heteromorphic bivalents. In normal males, single chiasmata are located in the medial part of the chromosome. In heterozygotes, this segment is heterosynapsed and unavailable for recombination. This leads to a significant decrease in the frequency of bivalents bearing single chiasmata. The total number of chiasmata per bivalent is much higher in heterozygous males than in normal ones. The recombination frequency between proximal markers fz and In also is higher in heterozygous animals. The increase in the total chiasma number in the heteromorphic bivalent is due to the addition of double chiasmata located mostly at precentromeric and pretelomeric regions of the chromosome.  相似文献

During lung development, Fibroblast growth factor 10 (Fgf10), which is expressed in the distal mesenchyme and regulated by Wnt signaling, acts on the distal epithelial progenitors to maintain them and prevent them from differentiating into proximal (airway) epithelial cells. Fgf10-expressing cells in the distal mesenchyme are progenitors for parabronchial smooth muscle cells (PSMCs). After naphthalene, ozone or bleomycin-induced airway epithelial injury, surviving epithelial cells secrete Wnt7b which then activates the PSMC niche to induce Fgf10 expression. This Fgf10 secreted by the niche then acts on a subset of Clara stem cells to break quiescence, induce proliferation and initiate epithelial repair. Here we show that conditional deletion of the Wnt target gene c-Myc from the lung mesenchyme during development does not affect proper epithelial or mesenchymal differentiation. However, in the adult lung we show that after naphthalene-mediated airway epithelial injury c-Myc is important for the activation of the PSMC niche and as such induces proliferation and Fgf10 expression in PSMCs. Our data indicate that conditional deletion of c-Myc from PSMCs inhibits airway epithelial repair, whereas c-Myc ablation from Clara cells has no effect on airway epithelial regeneration. These findings may have important implications for understanding the misregulation of lung repair in asthma and COPD.  相似文献

Summary The frequency and distribution of chiasmata was investigated in two fertile carriers of reciprocal translocations, one with a 46,XY,t(9;10)(p22;q24) karyotype and one with a 46,X,-Y,+der(Y),t(Y;10)(q12;q24) karyotype. In both cases the chromosomes involved in the translocation showed an increase in chiasma frequency in comparison to karyotypically normal controls and in both cases this increase was localised, affecting only one interstitial segment of each translocation quadrivalent. In the t(9;10) case chiasmata appeared in substantial numbers in a novel location, the proximal two thirds of 9p, while in the t(Y;10) case chiasmata appeared in a conventional location, the medial region of 10q, but at an increased frequency. Furthermore there was evidence for inter-chromosomal effects in the t(9;10) case.  相似文献

A spontaneous structural heterozygote originated from a centric fusion involving two acrocentric autosomes (M5 and S8) in a male Chorthippus jucundus (2n=16+X) was tested for the effects on chiasmata characteristics and the role that these could play in metaphase-I orientation of the trivalent. Results show non-existence of inter- or intra-chromosomal effects on chiasma frequency when compared with standard individuals, and displacement of the single chiasma on the S8 to a distal position while no significant change is appreciated in the M5 chiasma distribution. The presence of two chiasmata on the M5 and one on the S8 always induces a linear orientation of the trivalent. However, when two chiasmata are present (one on each chromosome) and independently of their location on each arm (proximal, interstitial of distal) any of the possible orientations (linear or convergent) can be attained. The frequency of linear orientation is higher than that expected at random in the resulting trivalent during metaphase-I and the rate of reorientation is very low.The meitotic behaviour of such a trivalent is discussed taking into account the already mentioned aspects together with the size of the fused element.  相似文献

For many plant species, reproductive success relies on the proper timing of flowering, and photoperiod provides a key environmental input. Photoperiod-dependent flowering depends on timely expression of FLOWERING LOCUS T (FT); however, the coordination of various cis-regulatory elements in the FT promoter is not well understood. Here, we provide evidence that long-distance chromatin loops bring distal enhancer elements into close association with the proximal promoter elements bound by CONSTANS (CO). Additionally, we show that NUCLEAR FACTOR Y (NF-Y) binds a CCAAT box in the distal enhancer element and that CCAAT disruption dramatically reduces FT promoter activity. Thus, we propose the recruitment model of photoperiod-dependent flowering where NF-Y complexes, bound at the FT distal enhancer element, help recruit CO to proximal cis-regulatory elements and initiate the transition to reproductive growth.  相似文献

Control of nondisjunction in the maize B chromosome was studied using a set of B-10 translocations. The study focused on the possible effect of the proximal region of the B long arm. The experimental procedure utilized a combination of a 10B chromosome from one translocation with a B10 from another translocation. The breakpoints of the two translocations were so located that combination of the two elements created a deletion in the proximal region of the B chromosome, but no deletion in chromosome 10. Two different types of deletions were established; one involved a portion of the euchromatic region and the other the entire heterochromatic portion comprising the distal half of the B long arm, except for the small euchromatic tip. Deletion of the heterochromatic portion did not exert any effect on nondisjunction. Deletions of different portions of the euchromatic region produce different responses. Some deletions resulted in typical B nondisjunctional activity; others resulted in the disappearance of this activity. It is concluded that a region within the euchromatic portion of the chromosome is critical for the nondisjunction of B chromosomes. Among 22 translocations with breakpoints in the euchromatic regions, three were proximal to the critical region, 16 were distal and the position of three others was not determined.  相似文献

In maize microsporocytes Heterozygous for a chromosome 2 interchange chromosome (which carries a homoeologous Tripsacum chromosome segment substituted for the distal half of its short arm), normal synapsis usually occurs at pachytene throughout the bivalent, but crossing over is almost entirely restricted to the homologous proximal region of the arm, (where it apprently occurs frequently). At diakinesis chiasmata were very often found to be located immediately proximal to an easily observable terminal knob of the Tripscacum chromosome segment. It was concluded that chiasmata, initiated in the homologous proximal region, had been maintained while terminalizing across the homoeologous region. It was also noted that heterozygosity for a telomere (and distal region) foreign to maize does not seem to inhibit pairing effective for crossing over in the homologous proximal portion of the chromosome arm.  相似文献

The transport properties of potassium (K) and sodium (Na) were studied in single distal tubules of Amphiuma using free-flow micropuncture techniques and stationary microperfusion methods. The transepithelial movement of labeled potassium was measured utilizing a three-compartment system in series in which the time course of tracer disappearance from the lumen was followed. Under control conditions, in blood- and doubly-perfused kidneys, extensive active net reabsorption of sodium and potassium obtains along single distal tubules. Tubular potassium reabsorption is abolished by ouabain at a concentration of 5 x 10-6 M. Significant net secretion of K can be induced by exposing Amphiuma to a high K environment (100 mM KCl) or by adding acetazoleamide (1 x 10-4 M) to the perfusion fluid. Transepithelial movement of potassium involves mixing with only a small fraction of total distal tubular cell potassium. This transport pool of potassium increases significantly with the transition from tubular net reabsorption to net secretion. Indirect evidence is presented which indicates that increased active K uptake across the peritubular cell boundary may be of prime importance during states of net K secretion.  相似文献

In many cereal crops, meiotic crossovers predominantly occur toward the ends of chromosomes and 30 to 50% of genes rarely recombine. This limits the exploitation of genetic variation by plant breeding. Previous reports demonstrate that chiasma frequency can be manipulated in plants by depletion of the synaptonemal complex protein ZIPPER1 (ZYP1) but conflict as to the direction of change, with fewer chiasmata reported in Arabidopsis thaliana and more crossovers reported for rice (Oryza sativa). Here, we use RNA interference (RNAi) to reduce the amount of ZYP1 in barley (Hordeum vulgare) to only 2 to 17% of normal zygotene levels. In the ZYP1RNAi lines, fewer than half of the chromosome pairs formed bivalents at metaphase and many univalents were observed, leading to chromosome nondisjunction and semisterility. The number of chiasmata per cell was reduced from 14 in control plants to three to four in the ZYP1-depleted lines, although the localization of residual chiasmata was not affected. DNA double-strand break formation appeared normal, but the recombination pathway was defective at later stages. A meiotic time course revealed a 12-h delay in prophase I progression to the first labeled tetrads. Barley ZYP1 appears to function similarly to ZIP1/ZYP1 in yeast and Arabidopsis, with an opposite effect on crossover number to ZEP1 in rice, another member of the Poaceae.  相似文献

Summary A new type of abnormal chromosome 10 has been found among maize plants grown from seeds sent by Dr. Y. C. Ting of Harvard University. This chromosome deviates in its morphology from the orthodox abnormal chromosome 10 described by Rhoades (1952) and from the one described by Ting (1958b). It produces a low degree of neo-centric activity.Cytological observations of plants heterozygous for the new abnormal chromosome 10 and either an orthodox abnormal chromosome 10 or a normal one, have suggested that the new type was derived from an orthodox abnormal 10 through spontaneous breakage and loss of an important piece of its long arm. The delection involved the distal part of the long arm of orthodox abnormal chromosome 10, proximally limited by the third most distal dissimilar and prominent chromomere. This corresponds approximately to the extra segment at the end of orthodox abnormal chromosome 10 which remains unpaired in heterozygotes with the normal 10. It bears a large heterochromatic knob. The missing piece is a part of the larger fraction of the long arm of orthodox abnormal chromosome 10 that remains unaffected by crossingover in a heteromorphic bivalent having a normal chromosome 10 (telo-segment). The telo-segment has its proximal limit at the left of the most proximal of the 3 dissimilar chromomeres, probably between the R and Sr 2 loci. It has been proposed that a factor or factors responsible for neo-centric activity are located in the portion of the telosegment between its proximal limit and the third most distal dissimilar chromomere (3 dissimilar chromomere region).Since the telo-segment of the orthodox abnormal 10 also bears a large knob in its distal half, it has been suggested that this segment has a dual role in neo-centric activity. The factor or factors located in the proximal piece of the telo-segment would stimulate over-abundance of fiber-forming substance, whereas local production of chromosomal fibers would depend ultimately on the knob's activity.If the large knob is absent, its role in neo-centric activity would be transferred to the next smaller and distally located hetero-chromatic mass, such as the knob-like body near the end of the new abnormal 10 which results from the fusion of the two most proximal prominent chromomeres of the telo-segment.This work has been partly done in the United States, under an I.C.A. — National Academy of Sciences fellowship.  相似文献

Single populations of Stethophyma gracile and S. lineatum from Eastern Canada show substantial differences in the relative amount of heterochromatic and euchromatic material in the extensively heterochromatic (megameric) M9 autosome. No differences in chromosome phenotype between these two species have been previously reported. A new polymorphism with respect to an interstitial, supernumerary H-block was also found in the S. lineatum population, and it appears to have some interchromosomal influence upon chiasma condition. These findings suggest that the North American Stethophyma forms may parallel the extensive and deme-specific heterochromatin variation found in the European populations of the Palaearctic S. grossum. Special attention was focused upon the structural organization of the proximal and distal H-segments on the M9 during the early prophase of the first meiotic division in the male sex. These conspicuous, hetregions, prior to diplotene, are clearly interrupted and contain short, euchromatic sequences. This condition can readily account for the absence of chiasma localization and the distribution of chiasmata along the H-blocks at diplotene-diakinesis in the megameric element. This is the only member of the complement showing no evidence of pronounced proximal or proximal/distal chiasma localization at diplotene. Possible explanations for this interesting phenomenon are suggested.  相似文献

In autotetraploids, chromosome pairing may be in the form of quadrivalents or bivalent pairs. Whether or not the quadrivalents are maintained until first meiotic metaphase depends on the formation of chiasmata. The relative frequencies of M I configurations thus contain information both on pairing and on chiasma formation. With distal chiasma localisation six configurations can be recognised and their relative frequencies determined: ring quadrivalents, chain quadrivalents, trivalents (with univalent), ring bivalents, open (rod) bivalents, univalent pairs. These represent five degrees of freedom permitting five parameters to be estimated: the frequency (f) of quadrivalent pairing; the frequencies of chiasmate association of the two ends (arms in metacentrics), a′, b′, after quadrivalent pairing, and a, b after bivalent pairing. — The appropriate formulae have been derived and applied to observations on Tradescantia virginiana (4n=24) which has pronounced distal chiasma localisation. Slight modifications make the model applicable to autotetraploids with interstitial in addition to distal chiasmata. In T. virginiana, chromosome pairing appeared to be random between homologues (65.8% quadrivalent pairing; 55.4% observed at M I). After quadrivalent pairing chiasmate association is frequent in the “average long” arm (95.0%) and much less so in the other arm (60.5%). This is attributed to partner exchange. After bivalent pairing chiasma frequencies are still different for the two arms (93.8% and 83.5% association respectively) but much less pronounced. Various complications are discussed.  相似文献

Trivalents resulting from polymorphic Robertsonian rearrangements must have a regular orientation in metaphase I if the polymorphisms are to be maintained. It has been argued that redistribution of proximal and interstitial chiasmata to more distal positions is necessary for a convergent orientation, the only one that produces viable gametes. Cornops aquaticum is a South-American grasshopper that lives and feeds on water-hyacinths, and has three polymorphic Robertsonian rearrangements in its southernmost distribution area in Central Argentina and Uruguay. The orientation of trivalents in metaphase I, the formation of abnormal spermatids and the frequency and position of chiasmata in the trivalents, was analysed in a polymorphic population of C. aquaticus. In this study we observed a correlation between the number of trivalents with the frequency of abnormal spermatids; additionally, the number of chiasmata, especially proximal and interstitial ones, was strongly correlated with the frequency of the linear orientation. Therefore we confirmed our previous assumption, based on other evidence, that the chiasmata redistribution in fusion carriers is essential to the maintenance of the polymorphisms.  相似文献

The endemic Australian grasshopper Heteropternis obscurella shows considerable variation in respect of both chromosome structure and chromosome behaviour. The structural differences depend upon different patterns of heterochromatin distribution as revealed by C-banding. These involve differences between populations in respect of polytypic variation in the size of paracentromeric C-bands and differences within populations in respect of polymorphisms both for terminal blocks of heterochromatin in autosomes 3 to 8 and a large proximal block of heterochromatin in autosome 7. The behavioural differences stem in part from genotypically determined variation in the chiasma distribution pattern which is markedly localised in a majority of populations but more randomly distributed in populations from the south of Western Australia. Behavioural differences also arise as secondary consequences of the presence of those heterochromatic blocks which occur as polymorphisms. The distal blocks on autosomes 5, 6, 7 and 8 lead to a redistribution of chiasmata to more proximal sites while the proximal block on 7 leads to the virtual abolition of chiasma formation in that bivalent and its replacement by a non-chiasmate mechanism of segregation. This depends upon a persistent proximal heterochromatic association between the pairing partners. The presence of distal C-blocks on bivalents 3 to 8 gives rise to persistent pseudomultiples, formed as a result of heterochromatic associations between these blocks. Such pseudomultiples involve any two or three of these six bivalents, provided they carry distal blocks, and their frequency rises dramatically in the presence of the proximal heterochromatic block on chromosome 7.  相似文献

Concentrations of macrominerals; Na, K, Ca, Mg, P, and Cl were measured in different sections of the alimentary tract of five roe deer, Capreolus capreolus, kept in captivity and fed a diet of grass pellets and oats. By means of the non-absorbed marker-slaughter technique (using 51CrEDTA as marker), sites of secretion and absorption of minerals in the alimentary tract were determined. Large amounts of P, Na and K were secreted into the rumen, whereas Cl was secreted into the abomasum. The larger amounts of these minerals were absorbed from the distal small intestine and caecum/proximal colon. In the coiled colon, small quantities of Na, K and Cl were absorbed which is essential for the maintenance of mineral balance. Emphasis is put on the role of the large hindgut in concentrate selectors both with respect to fementation and conservation of minerals and other nutrients.  相似文献
