A cell-free protein synthesizing system was used to study the mechanism of resistance to streptomycin (Str) and spectinomycin (Spc) in laboratory mutants and clinical isolates of Neisseria gonorrhoeae. The 70S ribosomes from sensitive strains were sensitive to the effects of Str and Spc on synthesis directed by several synthetic polynucleotide messengers, whereas 70S ribosomes from resistant strains were resistant to these same effects. In each case, the alteration was localized to the 30S ribosomal subunit by studying antibiotic sensitivities of hybrid 70S ribosomes formed by combining subunits from sensitive and resistant strains. No evidence was found for streptomycin- or spectinomycin-inactivating enzymes.  相似文献

Suppression of streptomycin dependence in Escherichia coli strain K-114, a spectinomycin-sensitive strain, is correlated with modification of 30S ribosomal protein P4, the component modified in spectinomycin-resistant mutants. The mutant is unusual in that reversion from dependence has previously been correlated only with modification in 30S protein P4a. Introduction into K-114 of another mutation conferring spectinomycin resistance results in a further alteration in protein P4.  相似文献

A dominant mutation at a new locus affecting resistance to cycloheximide has been isolated by exploiting a synergistic relationship with a previously known mutation for cycloheximide resistance in Tetrahymena. The new mutation (ChxB) was induced in a line homozygous for ChxA and was recovered from that background by a new technique termed interrupted genomic exclusion. Segregation data from the interrupted genomic exclusion suggest that ChxA and ChxB are separate, linked loci showing 30% recombination. Minimal lethal doses of cycloheximide for the four possible combinations of the wild-type and mutant alleles of these two genes are: wild type 6 µg/ml, ChxA 125 µg/ml, ChxB 10 µg/ml, ChxA-ChxB 175 µg/ml.  相似文献

SYNOPSIS. The responses of a strain of Amoeba proteus to conditions which could give rise to induced increases in resistance to streptomycin have been investigated. An indirect selection method, based on survival time in high concentrations of streptomycin, was devised. All attempts to obtain substrains with enhanced sensitivity or enhanced resistance by indirect methods were unsuccessful. It was therefore concluded that increases in resistance observed as a result of other treatments were likely to be the result of adaptation of a majority of individuals, rather than selection of pre-existing resistant variants. Direct treatment with regularly increased concentrations of streptomycin led to a 10-fold increase in resistance of large cultures. Nuclear transfers were made by the de Fonbrune technique to produce resistant nucleus/normal cytoplasm, and normal cytoplasm/resistant nucleus combinations. Examination of the descendants of these 'crosses' showed that the resistance was nuclear dependent and not influenced by the cytoplasm. The resistance proved transitory, disappearing slowly during continued culture without further contact with streptomycin.
These results are discussed with reference to possible mechanisms responsible for the increase in streptomycin resistance and its mode of origin.  相似文献

Two phenotypically distinct sets of nystatin-resistant mutants were investigated. One set is resistant, respiratory competent, and requires no lipid for growth. The other set is more resistant, respiratory deficient, and lipid requiring (unsaturated fatty acid or sterol). Both sets show altered sterol composition as demonstrated by the Liebermann-Burchard colorimetric reaction, ultraviolet spectrophotometry, and gas-liquid chromatography. Genetic analysis indicates that all nystatin-resistant mutants can be placed into one of six distinct genetic groups. The phenotype's nystatin resistance, lipid requirement, and respiratory deficiency are recessive. There was one case of allelism for mutants from different sets. Revertants of mutants which have the tripartite phenotype retain a residual level of nystatin resistance, but they are no longer lipid requiring or respiratory deficient. Growth studies in mutants which have the tripartite phenotype reveal that the addition of ergosterol to the growth medium results in decreased resistance to nystatin.  相似文献

We have isolated 15 spontaneous mutants resistant to one or several antibiotics like chloramphenicol, erythromycin and spiramycin. We have shown by several criteria that all of them result from mutations localized in the mitochondrial DNA. The mutations have been mapped by allelism tests and by two- and three-factor crosses involving various configurations of resistant and sensitive alleles associated in cis or in trans with the mitochondrial locus omega which governs the polarity of genetic recombination. A general mapping procedure based on results of heterosexual (omega(+)x omega(-)) crosses and applicable to mutations localized in the polar segment is described and shown to be more resolving than that based on results of homosexual crosses. Mutations fall into three loci which are all linked and map in the following order: omega-R(I)-R(II)-R(III). The first locus is very tightly linked with omega while the second is less linked to the first. Mutations of similar resistance phenotype can belong to different loci and different phenotypes to the same locus. Mutations confer antibiotic resistance on isolated mitochondrial ribosomes and delineate a ribosomal segment of the mitochondrial DNA. Homo- and hetero-sexual crosses between mutants of the ribosomal segment and those belonging to the genetically unlinked ATPase locus, O(I), have been performed in various allele configurations. The polarity of recombination between R(I), R(II), R(III) and O(I) decreases as a function of the distance of the R locus from the omega locus rather than as a function of the distance of the R locus from the O(I) locus.  相似文献

A genetic analysis of resistance to tetracycline (TC), streptomycin (SM), sulfanilamide (SA), and penicillin G was carried out through transduction with phage lysates obtained from a multiply resistant strain of Staphylococcus aureus by ultraviolet irradiation. All transductants acquired resistance to both TC and SA, even when singly selected for either SA or TC resistance. The locus responsible for TC resistance could not be separated genetically from that for SA resistance. On the other hand, in transduction of SM resistance, about 30% of the transductants jointly acquired resistance to both TC and SA. These observations suggest that the loci governing resistance to TC, SA, and SM exist close together on a single genetic unit, this probably being the chromosome.  相似文献

红霉素是一类广谱大环内酯类抗生素 ,在临床上具有广泛的应用。近 10年来用基因工程方法对红霉素结构改造已经取得了很大的进展。通过基因工程不仅可以改造红霉素内酯环环的大小、环的骨架和环的侧链 ,而且可以对后修饰的羟基、糖基和甲基进行改造。迄今用基因工程方法合成的新的大环内酯环结构已超过 10 0种 ,所合成的各种红霉素类似物也有数十种 ,且经过基因改造的红霉素类似物都具有生物活性。但基因工程产物产量都普遍降低 ,抑菌活性也不理想 ,因此未来红霉素基因工程研究的重点应加强产量和高活性结构筛选的研究。  相似文献

Resistance to tobacco mosaic virus (TMV) was activated by various forms of induction in Samsun NN tobacco leaves, and the intensity of the different forms was compared. Induced resistance was highest in leaf tissue between TMV inoculated stripes parallel to the mid-vein and after injection of ethylene maleic anhydride copolymer (EMA), followed by that induced in distal half leaves after inoculating the basal halves with TMV. Resistance in upper leaves following inoculation of the lower leaves with TMV was relatively low, while induction due to lesions caused by ethrel gave an intermediate degree of resistance. Estimation of resistance by size and number of local lesions was correlated with the amount of extractable virus as measured by enzyme-linked immunosorbent assay (ELISA), thus indicating that in the resistant tissue virus replication, and not only the development of necrotic local lesions, is suppressed. An increase in a specific ribosomal fraction (R2), recovered by a two-step procedure, was observed in tissues where resistance was most intense, i.e., between TMV stripes and after EMA injection. It may be that this specific ribosomal fraction participates in maintaining the resistant state.  相似文献
