Tonic activity of neurons of the superior cervical sympathetic ganglion was recorded by the "sucrose gap" method and in the 4th and 5th lumbar sympathetic ganglia with the aid of focal nonpolarizing electrodes in acute experiments on anesthetized cats and rabbits. The preganglionic fibers of the ganglia were left intact. Stimulation of the depressor nerve not only sharply inhibited the tonic activity of the ganglia but also led to the appearance of electropositive potentials of 0.7 ± 0.2 mV in the superior cervical ganglion and 20–250 µV in the lumbar ganglia. The amplitude of this potential was unchanged by atropine (1 · 10–6M). A similar effect occured without stimulation of the depressor nerve, after division of the preganglionic fibers or blocking of their conduction; it is attributed to the cessation of preganglionic tonic impulses which induce not only spikes, but also many EPSPs in neurons of the ganglion. Their frequency in the lumbar ganglia was 4/sec. Summation of these EPSPs leads to constant electronegativity of the ganglion surface relative to the postganglionic fibers, and its disappearance is recorded as a positive potential. Stimulation of the depressor nerve thus does not induce IPSPs in the ganglion; consequently, the inhibition of synaptic activity observed under these circumstances is located in the CNS and not in the ganglion.A. Spontaneous and evoked unit activity in response to repeated application of clicks at a frequency of 0.3–2.0 Hz in the caudate nucleus was studied by an extracellular recording technique in chronic experiments on cats. Four types of spontaneous unit activity in the caudate nucleus were distinguished. Altogether 44% of neurons tested responded by changes in spontaneous activity to clicks. Five types of responses of caudate neurons to clicks were discovered: phasic excitation, phasic inhibition, tonic activation, tonic inhibition, and mixed tonic responses; the commonest type was tonic activation. During prolonged stimulation by clicks extinction of the phasic responses was not observed. Complete or partial extinction of tonic responses in the course of frequent repetition of stimulation was observed in 33% of responding neurons. A study of the tonic electrical activity of nerves containing preganglionic and postganglionic fibers in the superior cervical and stellate sympathetic ganglia of cats and rabbits has shown that this activity consists of groups of spikes synchronous with the pulse or respiration, and occurs on a background of irregular low-amplitude impulses. The frequency of spikes is higher (250/sec) in nerves containing preganglionic fibers than in those containing postganglionic fibers (100/sec). Groups of spikes in a nerve containing preganglionic fibers correspond in some preparations to groups of spikes of lower frequency in a nerve containing postganglionic fibers of the same ganglion; in other preparations, this correspondence was lacking, apparently due to the absence of synaptic contacts between those groups of pre- and postganglionic neurons whose activity was recorded. Neurons send axons to different nerves (cardiac and vertebral) of the stellate ganglion discharged synchronously in some preparations, and asynchronously in others. Stimulation of the head of the caudate nucleus in cats anesthetized with chloralose and pentobarbital evoked spike responses of the Purkinje cells and other cerebellar cortical neurons in the paramedian lobes, lobulus simplex, and the tuber of the vermis. Phasic responses in the form of simple discharges (on account of activation of the neurons through mossy fibers) appeared mainly after a latent period of 5–12 and 14–20 msec; the latent period of responses consisting of complex discharges (on account of activation of Purkinje cells through climbing fibers) was 5–6, 9–22 msec, or more. Depending on the latent period, the spike responses differed in their rhythm of generation. In response to stimulation of the caudate nucleus with a frequency of 4–6/sec recruiting responses were found. An inhibitory pause was an invariable component of the tonic responses. During stimulation of the globus pallidus responses of the same types (phasic and tonic) appeared as during stimulation of the caudate nucleus, but they differed in the distribution of the neurons by latent period of spike responses. The minimal latent period was 4 msec. Recruiting also was observed during repetitive stimulation of the globus pallidus. During stimulation of the substantia nigra Pukinje cells activated by climbing fibers responded. Evoked complex discharges appeared after a stable latent period of 8.5±0.3 msec. Arguments are put forward regarding the role of the substantia nigra, the globus pallidus, nuclei of the inferior olive, and also the thalamic nuclei in the mechanism of caudato-cerebellar oligosynaptic and polysynaptic connections.N. When responses in some nerves of the pterygopalatine ganglion of the cat in situ to stimulation of its other nerves were recorded it was found that most fibers passing through the ganglion are continuous sympathetic postganglionic fibers (at least three groups). Most of the parasympathetic preganglionic fibers forming synapses on neurons of the ganglion constitute a group of fibers with the same threshold of excitation. Intracellular recording from single neurons of the pterygopalatine ganglion showed that stimulation of the Vidian nerve evokes orthodromic spike potentials in some neurons of the ganglion with a short latent period, and in others with a long latent period (2.5–6.0 and 10–44 msec, respectively). Evidently only fast-conducting fibers terminate synaptically on most neurons of the ganglion and only slow-conducting fibers on some of them. Recording from intact nerves of the pterygopalatine ganglion revealed no tonic activity in them. Microelectrode recording from single neurons of the ganglion showed that either the frequency of generation of spike potentials is relatively low (1–3/sec) or such potentials are absent altogether.A. Single unit responses in the superior olive of the greater horseshoe bat to ultrasonic stimuli with a filling frequency within the echolocation range were investigated. Some neurons were found to have three completely unconnected response regions with characteristic frequencies of 1/2 and 1/3 of the basic frequency, which was within the 80–86 kHz band. An increase in strength of the stimulus with filling frequency equal to the characteristic frequency of the neuron changed the tonic regime of activity into phasic. Presentation of two stimuli, overlapping in time, replaced the phasic regime by tonic. The frequency of the tonic response corresponded exactly to the beating frequency up to 1200 Hz (synchronization of unit discharges with each beating cycle). The opercularis muscle of Rana catesbeiana originates on the suprascapular cartilage of the shoulder girdle and inserts on the otic opercular element. It is part of the levator scapulae musculature and lies dorsomedial to the levator scapulae superior and inferior muscles. Bipolar electrode recordings from all three muscles show electrical activity linked to cyclical firing of the posterior intermandibularis muscle, an important ventilatory muscle. The opercularis muscle shows low amplitude, erratic signals when animals are sumerged. Upon emergence of the snout region, the opercularis muscle shows rhythmic low amplitude activity at twice the rate of buccal pumping. Lung ventilation is synchronized with this rhythm and at ventilation the opercularis muscle shows higher amplitude activity. Upon submergence, opercularis activity again shows low level activity with no rhythmic pattern. Opercularis muscle activity has a major low frequency component (about 30 Hz) that probably corresponds to activity of tonic muscle fibers. Higher frequency signals (about 200–250 Hz) comparable to those of the levator scapulae muscles are also present and probably represent activity of phasic muscle fibers. Activity of the opercularis muscle is correlated with conditions in which aerial respiration is possible, and this pattern of activity supports an opercularis role in aerial hearing and/or detection of substrate vibrations. As far as we know, this is the first report of electromyographic analysis of a vertebrate tonic muscle.  相似文献

This paper summarizes information obtained in the experimental study of the dynamics of polarization of central primary afferent endings and modifications of segmental responses to afferent stimuli during fictitious locomotion and fictitious scratching in immobilized, decorticated, decerebrate, and spinal cats. Fictitious locomotion was accompanied by tonic hyperpolarization, fictitious scratching by tonic depolarization of central primary afferent endings. Against the background of these long-lasting changes in primary afferent depolarization, it exhibited periodic changes in the rhythm of efferent activity. Periodic changes of depolarization were virtually in phase in different ipsilateral segments of the lumbosacral enlargement. Data on groups of afferent fibers in whose central endings tonic and phasic changes of polarization took place. The appearance of fictitious locomotion was accompanied by a tonic increase, and of fictitious scratching by tonic inhibition of several evoked segmental responses. These tonic changes were a background against which segmental responses were modulated in step with the working rhythm of the locomotion and scratching generators. Many of the changes in evoked segmental responses were shown to be based on modulation of polarization of central endings of primary afferents by locomotion and scratching generators. It is concluded that active tonic and phase-dependent selection of incoming afferent information is effected through modulation of presynaptic inhibition of the generator. The role of this selection in peripheral collection of activity of locomotion and scratching generators is discussed.A. A. The kinetics of primary afferent depolarization (PAD) in the lumbar spinal cord was studied in immobilized decerebrate and decapitated cats during fictitious scratching. Fictitious scratching was accompanied by tonic and periodic PAD. Periodic PAD was cophased in different segments of the lumbosacral enlargement. Tonic depolarization was observed in terminals of all groups of afferent fibers studied (low-threshold cutaneous and of groups Ia and Ib). Periodic changes were observed only in endings of low-threshold cutaneous fibers and group Ib fibers. The physiological role of modulation of PAD by the spinal scratch generator is discussed.A. A. Focal extracellular recordings were made of postjunctional currents produced at synapses of the inferior rectus eye muscle fibers by the spontaneous release of quanta of transmitter. These consisted of miniature endplate currents, or MEPC, in phasic fibers and miniature postjunctional currents, or MPJC, in tonic fibers. Open time of ionic channels (chan) was also registered. In tonic fibers, MPJC lasted considerably longer than MEPC did in phasic fibers: rising time, decay time, and chan in the former measured respectively 2.5, 4–5, and 2.2 times higher than in the latter. Acetylcholinesterace (AChE) inhibition produced a much greater (4.4-fold extension of current decay in phasic than in tonic fibers, where a 1.8-fold increase was seen, thereby reducing the gap between the decay time of currents in these fibers to a difference of 1.6 times. The more protracted decay of MPJC in tonic fibers compared with MEPC in phasic fibers is determined by the lower functional activity of AChE as well as the higher value of chan. Duration of MEPC and magnitude of chan in the "slow" phasic fibers of rat skeletal muscles fell well below the same parameters measured in the tonic fibers of the ocular muscle.I. M. Changes in electrical polarization of primary afferent terminals in the lumbosacral portion of the spinal cord were investigated during fictitious locomotion in immobilized decorticated and spinal cats. Fictitious locomotion was accompanied by stable hyperpolarization of the afferent terminals, against the background of which they were periodically depolarized in rhythm with efferent activity. These tonic and phasic changes were observed in terminals of all groups of afferent fibers tested: cutaneous and muscular (Ia and Ib). Periodic in-phase depolarization was carried out in different ipsilateral segments of the lumbosacral enlargement. During ficitious galloping changes in depolarization of the primary efferents were in phase on different sides; during fictitious walking, these periodic changes were out of phase. On the basis of these results the physiological importance of changes in electrical polarization of primary afferent terminals of the spinal locomotor generator is discussed.A. A. Some characteristics of spinal reflex reaction inhibition were studied in cat fetuses during the last three weeks of antenatal development. The experiments were conducted on fetuses with intact placental circulation. Restoration of the excitability of the spinal reflex arcs was very slow after stimulation of the dorsal root by a single stimulus. In embryos studied 20 days before birth the full inhibition of reflex responses lasted about 500 msec. Even 2–3 sec after a single stimulation of the afferent fibers the amplitude of the reflex response to the second stimulus was only 30–40% of the control value. It was determined that such long postactivation depression is unrelated to refractoriness or antidromic inhibition. The presence of a prolonged intense depolarization of afferent terminal fibers at these stages suggests a presynaptic inhibition as one of the most probable reasons for the prolonged postactivation depression. Another important factor in the appearance of postactivation depression is probably the morphologic and functional immaturity of synaptic structures. A reciprocal inhibition was observed in cat fetuses on the 10–12th antenatal day. Intracellular recording techniques were used to record electrical response from neurons of the rabbit (isolated) superior cervical ganglion to single stimuli applied to bundles of preganglionic fibers as well as tonic electrical neuronal activity in this ganglion during acute experiments in situ. A review of the findings obtained confirms that neurons of the ganglion receive preganglionic synaptic inputs of two types, the first of a single pattern, formed by a preganglionic fiber; excitatory action of the latter on ganglion units suffices to induce postsynaptic action potentials (AP I) and the second of a multiple pattern, formed by several preganglionic fibers with relatively faint excitatory action, capable of evoking postsynaptic action potentials (AP II) only when excited in unison. In experiments on rabbits anesthetized with urethane, we recorded spontaneous sympathetic activity of single nerve fibers. The activity was recorded from fine filaments dissected from the cervical sympathetic nerve trunk (CSNT). Conduction velocities in CSNT fibers and action-potential amplitude of single B-fibers were measured during recording of whole nerve trunk. Spectral analysis of the activity recorded was carried out. It was shown that unmyelinated CSNT fibers are under stronger baroreceptor control than myelinated fibers. Also, a periodic component of 2–3 Hz, noted by a number of authors after denervation of baroreceptors, was detected in intact animals along with a rhythm at the pulse rate. This component was found equally often in the activity of B- and C-fibers. Cross-correlation analysis of the simultaneously recorded activities of 24 pairs of CSNT single fibers was carried out. A correlation due to synchronization of the activities of pairs of fibers with a physiological rhythm (respiration, pulse) was found in five pairs. The correlation in one pair suggests the presence of a common excitatory synaptic input into two distinct preganglionic neurons.A. A. The total electrical response and action potentials of separate neurons in the cochlear nuclei in Vespertilionidae and Rhinolophidae were investigated. Maximum sensitivity to ultrasound was recorded in Vespertilionidae in the frequency ranges 10–30 and 70–80 kc/sec, and in Rhinolophidae in the frequency ranges 10–30 and 84–86 kc/sec. Mininum off-response thresholds were observed in Vespertilionidae in the range 50–60 kc/sec, and in Rhinolophidae in the range 78–80 kc/sec. The areas of responses by neurons in the cochlear nuclei in both species of bats were similar in shape to those recorded in the same structure in other animals. An exception was provided by Rhinolophidae, in which three peculiar types of neurons were observed: 1) neurons whose response area lay in the frequency ranges up to 78 kc/sec or from 80 to 90 kc/sec; 2) neurons responding in the range 40–90 kc/sec, but not sensitive to stimuli with a fill frequency of 78–80 kc/sec; and 3) neurons whose response area lay in the range 78–80 kc/sec, but in which the character of the response changed from tonic to phasic when there was a change in the fill frequency of the stimulus. Temporal and amplitude characteristics of evoked potentials of the sensomotor cortex in waking cats were studied during variation in the intensity of electrodermal stimulation. The results obtained in experiments on intact animals and on the same animals for several months after division of the spinocervical tracts at the cervical level were compared. After blocking of the inflow of afferent impulses along these tracts of the spinal cord, statistically significant changes in evoked potentials were observed, mainly in response to medium and strong stimulation. These changes were more clear in the motor and second somatosensory areas of the cortex. A decrease in sensitivity to pain also was found. During recovery of the motor functions, cutaneous sensation remained impaired and the amplitude characteristics of the evoked somatosensory activity were not restored. The motor unit (MU) potentials of the human m. rectus femoris were recorded during voluntary isometric contraction by means of a bipolar needle electrode. The frequency of impulse discharge of individual motoneurons was defined as a quantity inverse to the average interval between impulses during 0.5 or 1.0 sec. The force of contraction varied from 0 to 4–14 kg (17–47% of the maximum). The investigations showed that in addition to switching on and off of motoneurons during a change of contraction force, the frequency of their impulse discharges also changes. Motoneurons recruited at a low force (low-threshold) reached the highest frequency (up to 18–21 impulses/sec). As a rule, the higher the threshold, the lower the frequency in the entire range of changes. In the case of prolonged contraction with a constant force the frequency of discharges dropped during the first 1–2 min. The established frequency level did not exceed 10–13 impulses/sec. A voluntary increase of contraction force at this period was related with a new increase of frequency. Recruitment of new motoneurons was observed during prolonged contraction. The data obtained show that the mechanism of change of the firing frequency of motoneurons actively participates in contraction gradation, mainly its dynamic component. It is regarded as a mechanism of smooth and precise control. Global and single unit responses of the superior olivary complex were investigated during ultrasonic stimulation at different frequencies in two species of bats from the Vespertilionidae, which emit frequency-modulated signals and the Rhinolophidae, which utilize almost monochromatic (80 ± 1 kHz) echolocation cries. Maximal sensitivity to ultrasound in the Vespertilionidae was found at frequencies of 10–40 kHz, and in the Rhinolophidae also within the range 10–40 kHz but with a second increase in sensitivity in the region 82–86 kHz. Sharply tuned neurons were more numerous in the Rhinolophidae than in the Vespertilionidae. Neurons whose response in the echolocation frequency band changed in character depending on the fill frequency of the stimulus were found in Rhinolophidae: a phasic discharge occurs over a wide range of frequencies and a tonic discharge at the characteristic frequency; the latter was also observed over a limited range of intensities.A. It was shown that the rabbit sensorimotor cortex received afferent fibers from neurons located in the specific, nonspecific, and association thalamic nuclei using the retrograde axonal transport technique. The distribution, dimensions, and shape of the somata of relay neurons spread through the thalamic nuclei were analyzed. The total number of neurons sending out thalamo-sensorimotor-cortical fibers was calculated and the coordinates of loci with the highest density of these cells in each thalamic nucleus were identified. Multipolar and stellate cells with somata measuring 12–20 µm and 10–15 µm in diameter, respectively, prevailed amongst relay neurons. Amongst the specific nuclei, the majority of afferent fibers are sent out by the ventrolateral, ventral anterior, and anterior ventral nuclei. A comparable number of afferent fibers are sent out by the mediodorsal and paracentral nuclei; these split up among the association nuclei and paracentral nuclei, respectively. Efferent fibers of the hind limbs were divided and electrical activity of a filament from the ventral root of S1 was recorded in experiments on mesencephalic cats capable of locomotion in response to stimulation of the "locomotor region" of the midbrain. In response to weak stimulation of the locomotor region, when the forelimbs were not performing stepping movements, regular waves of activity appeared in the filament with a period close to the duration of the step during walking (0.5–1.0 sec). This periodic process was largely dependent on the tonic afferent inflow: various extero- and interoceptive stimuli applied to the hind limb could change the period of generation or abolish it. Active stepping movements of the forelimbs as well as passive movements of the hind limbs led to synchronization of activity in the filament with these movements. After division of the afferent fibers to the hind limbs the animals performed one or two steps in response to stimulation of the dorsal root of S1 by a short series of pulses. They could also perform independent stepping movements of the hind limb if 15–30% of the fibers in the dorsal root of L7 remained intact.M. V. Lomonosov Moscow State University.
