Differential expression of p63 isoforms in female reproductive organs   总被引:4,自引:0,他引:4  
p63 is the identity switch for uterine/vaginal epithelial cell fate, and disruption of p63 expression by diethylstilbestrol (DES) induces cervical/vaginal adenosis in mice. In this article, we report the expression patterns of p63 isoforms (TA, DeltaN, alpha, beta and gamma) in mice, focusing on the reproductive tract. We also present the reproductive tract phenotype of female p63-/- mice. Finally, to better evaluate the potential role of p63 in human development of DES-induced cervical/vaginal adenosis, we describe the ontogeny of p63 in human female fetuses. In adult mice, the DeltaN isoforms of p63 were expressed only in squamous/basal/myoepithelial cells of epithelial tissues, while TA isoforms of p63 were highly expressed in germ cells of the ovary and testis. In fetal mice, the DeltaN and alpha forms of p63 were expressed in the cloacal and urogenital sinus epithelia. In the female p63-/- mice, the sinus vagina developed, but p63-/- sinus vaginal epithelium failed to undergo squamous differentiation confirming an essential role of p63 in squamous epithelial differentiation. Although TAp63 was highly expressed in developing primordial germ cells/oocytes, p63-/- ovaries and oocytes developed normally. The ontogeny of p63 in female reproductive organs was essentially identical in mouse and human. In the human fetus at the susceptible stage for DES-induced cervical/vaginal adenosis, most cervical/vaginal epithelial cells were columnar and negative for p63. Therefore, inhibition of p63 expression by DES should change the cell fate of human Müllerian duct epithelial cells and cause cervical/vaginal adenosis as previously demonstrated in mouse.  相似文献

This study investigated fear responses of Japanese monkeys (Macaca fuscata) to scale models. Fear responses of participants were assessed using rating scores assigned by judges. In Experiment 1, participants were presented with scale models of objects in furniture, vehicle, and mammal categories. Overall, the participants expressed stronger fear responses to mammal objects as compared to the other two kinds. In Experiment 2, participants were presented with scale models of furniture, new mammals, and animals composed of insects, birds, fish, and reptiles. As a whole, the participants showed stronger fear responses to the new mammal and animal objects than to furniture objects. Fear responses to mammal and animal objects were comparable. These results suggest that Japanese monkeys show stronger fear responses to objects possessing perceptual properties of animals, as opposed to objects that do not possess such properties.  相似文献

The anatomy and histology of the female reproductive tract of the Indonesian wild pig Babyrousa celebensis was studied by means of reproductive tracts obtained from seven animals aged between two and 22 years of age. The ovary appeared to have the ability to ovulate up to four ova at one time. However, the combined ovarian output seemed to average 1.86 ova. Ovulation can take place at any time from puberty to old age (22 years). The opening to the uterine tube was indicated by a ‘flower-like' array of tall, broad epithelial ‘petals' arising from the luminal surface of the funnel. The mucosal surfaces of these structures were covered in a mixture of prominent ciliated cells and bulbous secretory cells. The uterine tube followed a tightly convoluted path to the tip of the uterine horn. The uterus was proportionately short. The anatomical construction of the uterus was similar to those of other suids in that the columnar endometrium was heavily folded, there was a rich supply of uterine glands in the lamina propria, and the uterus was provided with a good blood supply. The cervix was thick walled and had a spiral lumen.  相似文献

A review is presented of the results of the various studies in this volume and an attempt is made to establish connections among several features of the ecology, behavior, and social structure of Japanese monkeys. Several studies in this volume suggest that intergroup direct feeding competition has been much more severe in Yakushima, in the warm-temperate region, than in Kinkazan, in the cool-temperate region of Japan. This result is consistent with the predictions that moderate abundance and clumped distribution of food incur more severe intergroup direct competition. However, the number of adult females within a group in Yakushima was smaller than that in Kinkazan even though severe intergroup direct competition should favor large groups. This contradiction can be mainly explained by the less severe intergroup indirect competition in Kinkazan than in Yakushima. By contrast, some studies in this volume also indicate that adult male to female ratio within a group has been higher in Yakushima than in Kinkazan. This result can be explained in two ways: the females in Yakushima might have recruited more males to increase the competitive ability of the group under conditions of severe intergroup direct feeding competition; alternatively, it might be profitable that the males in Yakushima defend females cooperatively as group males against the males in other groups at a moderate density of females. Some studies in this volume suggest that grooming frequency was higher in Yakushima than in Kinkazan. The higher grooming frequency in Yakushima might have been partly due to a constant increase in engaging in social behavior from a decrease in feeding time. Another reason might be that there is a stronger effect of grooming on promotion of formation of coalitions among adults under conditions of severe intergroup direct and intragroup direct competition.  相似文献

Five laboratory-raised Japanese monkeys (Macaca fuscata) were presented various types of photographs of Japanese and rhesus monkeys (Macaca mulatta) in upright, horizontal, and inverted orientations in a sensory-reinforcement experiment. The ratio of the duration of potential viewing time for the photographs which the subjects controlled to the interval between subject-controlled presentations of them (the D/I score) was used as a measure of preference for the photographs. When inverted photographs were presented, the D/I scores were lower than for upright photographs. The difference in D/I scores between photographs of the two species, which indicated discriminability between them, also diminished when the photographs were inverted. The results obtained suggest an inversion effect in face perception in macaque monkeys.  相似文献

Daily intake of gross energy (DGEI) and protein (DGPI) of Japanese monkeys in a cool temperature forest were investigated in different four seasons. As a result, dramatic seasonal differences in DGEI and DGPI were found: DGEI and DGPI in winter were significantly lower than those in spring and autumn, being only about 17–42% of those in the two seasons. DGEI and DGPI in summer were also lower than those in autumn and spring, being only about 29–52% of those in the two latter seasons. Seasonal comparisons of some variables on food qualities revealed that much lower values of DGEI and DGPI in winter and summer were influenced srongly not so much by the lower calorie and protein content of food as by the lower speed of dry weight intake. Multiple regression analyses for each food item revealed that the unit weight was a more important factor in determining the speed of dry weight intake than was the speed of unit intake. In addition, the speed of dry weight intake contributed much more to the speed of calorie and protein intake than did the content of calorie and protein. Multiple regression analyses, employing DGEI and DGPI (as dependent variables) and daily mean values of some variables related to food quality (as independent variables), revealed that the mean values of speed of calorie and protein intake on food items eaten in a day could explain 79.0% and 85.9% of the variance in DGEI and DGPI, respectively. In addition, the mean value of the speed of dry weight intake of food items eaten in a day can be used to explain 74.3% and 52.8% of the variance in DGEI and DGPI, respectively. These findings suggested that not only nutritional content of food but also the speed of dry weight intake strongly determined the nutritional condition of nonhuman primates. Am. J. Thirteen adult female Japanese monkeys (Macaca fuscata) were sacrificed in the early and the late follicular phase, and in the mid-luteal phase of the menstrual cycle, and also after castration. Pituitary LH content correlated significantly with plasma estradiol levels but not progesterone levels in the cycling monkeys. Pituitary LH species were fractionated by isoelectrofocusing (1EF). Pituitary LH species were separated into six fractions by IEF. High alkaline LH and alkaline LH were the most abundant species in the late follicular phase and the mid-luteal phase, respectively. Neutral LH and acidic LH were the most abundant in the early follicular phase and in the castrates. Pituitary LH thus changed qualitatively and quantitatively during the menstrual cycle and after castration.  相似文献

We employed techniques of behavioral entropy to carry out a quantitative analysis of sequences of behavioral patterns evident in the interaction between infants and other members of a group of Japanese monkeys (Macaca fuscata fuscata). The group concerned included examples both of monkeys in captivity and in the wild state. The results were examined as a function of the animal age and environmental differences (cage-field). An example is given to illustrate the use of information theory. Findings partially confirmed that the variability of social behavior decreases as the age of the animals increases.  相似文献

Artibeus lituratus is one of the most recognized bat species in the neotropics, probably due to its high abundance in urban areas. Despite its wide distribution, the morphology and physiology of its reproductive organs remain unclear. Thus, the aim of the present study was to describe the morphology and histology of the female reproductive organs of this species, and evaluate morphological variations during its different reproductive phases (nonreproductive, pregnant, lactating, etc.). Thirty adult females were collected, divided into six (6) sample groups, according to the reproductive status, and submitted to anatomical and histological analyses. We show that the ovaries of A. lituratus are polarized, present a low number of interstitial glands, have large and well-developed Graafian follicles and a persistent corpus luteum, and may present a functional everted corpus luteum. Ovulation is simple, unilateral and nonpreferential, possibly alternated in successive ovulations, and may be sequential, with the occurrence of postpartum estrus. The uterus is simplex, the implantation is fundic and interstitial and the placenta is chorioallantoic, discoidal, and hemochorial. The female reproductive organs of A. lituratus share similarities with the pattern observed in human, which indicates that A. lituratus may be an interesting animal model in reproductive studies.  相似文献

Summary The effects of short-term (8 weeks) treatment with different doses of cyproterone acetate (CPA), levonorgestrel (LN) and progesterone (PRO) on the adrenal gland, ovary, uterus and vagina were studied in cycle-synchronised beagle bitches (first anoestrus). The same organs from non-treated primiparous beagle bitches at the 6th and 9th weeks of pregnancy were also included. In the animals treated with the highest doses of CPA (4.0 mg/kg/day orally) and PRO (42.5mg/kg/day subcutaneously), as well as in pregnant bitches (9th week of pregnancy), a decrease in adrenal weight and cortex width and also an apparent loss of cells in the zona fasciculata and zona reticularis were observed. A marked increase in ovarian weight was recorded only in pregnant bitches (6th week of pregnancy). This was reflected by the presence of multiple well-developed corpora lutea. The ovaries of virgin control and progestagen-treated bitches revealed ovarian atrophy. Progestagen treatment caused marked stimulation of the uterus, resulting in dose-related oedematous and hyperplastic changes. Comparable findings were also observed during pregnancy. The vaginal epithelium of the progestagen-treated and pregnant bitches showed marked mucification as compared with control bitches. Advances in our understanding of ovarian cyclicity, pathogenesis of subfertility and/or infertility and reproductive pathology in food animals have frequently entailed examination of abattoir material. Despite the fact that most lesions in ewes are likely to be of relatively minor significance to fertility, results of previous studies suggest that lesions of the female reproductive system may represent a significant source of loss to sheep husbandry. The objective of this paper is to review the pathophysiology, the effects on reproductive efficiency and the key gross and histological diagnostic features of congenital and acquired pathology of ovary and tubular genital organs in ewes.  相似文献

Grooming among adult and older juvenile females in a wild group of blue monkeys (Cercopithecus mitis stuhlmanni) is analyzed and related to agonistic behavior. Both grooming and agonistic behavior were less frequent than would be expected in baboon and macaque groups. Fair shares of grooming were given and taken, both within and between interactions, in amost dyads, and all females were seen to groom with at least half the others. The reciprovity of grooming was its most outstanding feature. There was no evidence that grooming was preferentially directed at kin. Frequent grooming partnerships fell into two clusters, one of which ranked higher than the other. We suggest that grooming throughout the group helps to establish a coherent team of females which effectively, defends its territory in daily encounters at the boundaries with neighboring groups.  相似文献

The present study investigated the influence of dominance rank in combination with kinship on age-related differences in social grooming among adult females in a free-ranging group of Japanese monkeys (Macaca fuscata). Eighty-three adult females were divided into six sub-groups according to age-class (younger: 5–9 years old; middle: 10–14 years old; older: 15–22 years old) and dominance rank (high and low rank). The ratio of the number of unrelated females that each female groomed to the total number of available unrelated females and grooming bouts which she gave to unrelated females decreased with increasing age for both high- and low-ranking females, whereas age did not appear to affect corresponding values for related females. On the other hand, compared with low-ranking females, high-ranking females of all age-classes received grooming more often from a larger number of unrelated females. Moreover, older females of low rank received grooming less often from a smaller number of unrelated females than younger females of low rank. These results indicate that with increasing age females are more likely to concentrate on related females when they have grooming interactions with other females. This tendency seems to be more apparent for low-ranking females. Moreover, the present findings also indicate that older high-ranking females could maintain their social attractiveness as high as younger high-ranking females.  相似文献

Blood vessel homeostasis and endothelial cell survival depend on proper signalling through angiopoietin receptors such as the receptor tyrosine kinases Tie-1 and Tie-2. We have studied the presence and subcellular localization of these receptors in murine female reproductive organs using confocal microscopy analysis of antibody stained tissue sections of ovary and oviduct. We show that Tie-2 principally localizes to primary cilia of the surface epithelium of the ovary, bursa and extra-ovarian rete ducts as well as to plasma membranes of ovarian theca and endothelial cells. Primary cilia of follicular granulosa cells were negative. Further, Tie-1 and Tie-2 localized to motile cilia of the oviduct. Western blotting detection and immunolocalization of anti-Tie-2 in ovary and oviduct were abolished by administration of an anti-Tie-2 blocking peptide, confirming antibody specificity. In a series of immunohistochemical analysis on human ovarian tissues we also observed a unique localization of Tie-2 to the primary cilia of ovarian surface epithelium. These observations are the first to show ciliary localization of angiopoietin receptors. Our results support the hypothesis that cilia of the female reproductive organs play a novel and important sensory role in relaying physiochemical changes from the extracellular environment to epithelial cells of the oviduct, the ovary and extra-ovarian tissues.  相似文献

Hexachlorobenzene (HCB) is a persistent environmental contaminant which has been measured in human serum, fat, semen, and follicular fluid. In animal testing HCB has been shown to be a reproductive toxin. Discrepant results were obtained from prior studies concerning the effect of HCB treatment on ovarian steroidogenesis. The current study was designed to assess the impact of HCB on the ovary and gonadal steroid levels in the superovulated rat. Female Sprague-Dawley rats (n = 24) were dosed with HCB (0.0, 1.0, 10.0, or 100.0 mg/kg BW/day) for 21 days. All rats received 10 IU pregnant mare serum gonadotropin (PMSG) s.c. on day 18 of treatment and 15 IU of human chorionic gonadotropin (hCG) on day 20. A terminal blood sample was collected and circulating levels of estradiol (Emr2) and progesterone. (P4) were determined. Serum concentrations of P4 were significantly (p < 0.0034) elevated by HCB treatment at all dose levels. Ovarian weights were significantly increased (p < 0.05) in the lowest dose group only compared to the control group. Serum concentrations of Ev uterine weight, weight gain, and general animal health were not affected by HCB treatment. We conclude that during HCB treatment the rat ovary remains responsive to gonadotropin stimulation. Moreover, it is suggested that HCB effects on ovarian steroidogenesis are indirect.  相似文献

The menstrual cycles of a captive group of patas monkeys were followed for 15 months by taking vaginal smears and lavages three times a week. Without an adult male in the group, menstrual cycles still showed the expected qualitative changes previously associated with the onset and with the end of a mating period. The addition of an adult male to the female group, once mating season cycles were evident, did not result in further changes in erythrocytes or sediment levels in vaginal samples or cycle regularity. Menstrual-cycle onsets for related females were significantly more synchronized than onsets for unrelated females. Preliminary observations on adult male patas housed separately from the females indicate that they too undergo seasonal changes in physiology and behavior.  相似文献

Two ovariectomized female rhesus monkeys treated year-round with estradiol-filled capsules were used in hour-long behavioral tests with male rhesus monkeys both in and out of the normal breeding season. The study was designed primarily to test male hormonal responses to copulatory behavior and it was expected that behavior of the females would be essentially the same under both seasonal conditions. Several behaviors of the female, however, were found to fluctuate on a seasonal basis, namely (1) proximity to the male, (2) grooming of the male, (3) sexual presentations, and (4) threatening away (a form of sexual invitation). All of these behaviors, except proximity, were found to be positively correlated with the male partner's testosterone levels before and after the behavior test in the only test condition in which most males ejaculated. The preliminary suggestion is made that these females responded to some cue, either from the environment or from the males, that caused a change in their behavior between breeding and nonbreeding seasons despite the constant hormonal environment provided by the estradiol capsules. Since the same behaviors which were sensitive to seasonal effects were positively correlated with male testosterone levels, it is possible that the male's hormonal status affects affects female behavior.  相似文献
