Seed dormancy of a highly-dormant cultivar of celery (Apium graveolens L.) was broken by combinations of plant-derived smoke extract or N6-benzyladenine (BA) and gibberellins A4/7 (GA4/7) in the dark at temperatures between 18 and 26°C. A less dormant cultivar which responded to GA4/7 alone showed no additional response to smoke extract or BA. Neither smoke extract nor BA affected either cultivar in the dark in the absence of GA4/7. The partial dormancy-breaking effect of short exposures to red-light was also enhanced by smoke extracts in this highly-dormant cultivar. The results suggest that smoke extracts act in a similar way to cytokinins, by enhancing gibberellin activity in the celery seed system.Abbreviations BA N6-benzyladenine - GA4/7 A4 and A7 gibberellin mixture  相似文献

Summary The flowering and inflorescence development of Chrysanthemum morifolium cv. Pink Champagne under non-inductive long days was promoted by exogenous application of GA5, GA3, GA4+GA7 or GA9 in combination with the cytokinin, BA. The combination of BA and GA5 was highly effective, BA and GA3 moderately effective. Applications of the GAs alone or BA alone also resulted in some flowering, with GA3 and GA5 being most effective. In general, the effects of GA and BA were synergistic, and the concentration of both growth substances was a limiting factor with regard to the number of plants flowering under long days. Only the concentration of GA was a limiting factor for inflorescence development, however. Simultaneous application of indole-3-acetic acid reduced inflorescence development in most treatments. Kinetin at 10 mg l–1 increased the number of flowers produced on Rosa damascena plants while GA3 inhibited flowering. In the leaves of non-flowering plants GA-like activity was high while specific cytokinin activity (fraction-II) was significantly higher in flowering plants. The germination of seeds of celery (Apium graveolens L.) becomes progressively thermoinhibited on incubation in the dark at high temperatures, the inhibitory temperature being dependent on the cultivar used. In two high-dormancy cultivars of celery, the production of germination inhibitors in seeds incubated in the dark at 26°C gradually increased over a 7-day period. Inhibitor production was measured by incubating seeds of the low-dormancy cultivar Florida 683 in homogenates of the thermoinhibited seeds of the high-dormancy cultivars and recording germination either in the light or with the gibberellins A4 and A7 (GA4/7) in the dark. Most Florida 683 seeds which failed to germinate in the homogenates after 15 days were induced to germinate by addition of N6-benzyladenine (BA). The presence of BA in addition to GA4/7 throughout incubation in the dark completely overcame the inhibitory effects of homogenates. This indicates that thermoinhibition of celery seeds is associated with the accumulation of a germination inhibitor which interacts with cytokinins. Container-grownEuphorbia lathyris plants were treated with foliar sprays of various combinations of BA and GA4+7 or 0–3600 mg L?1 Promalin (1∶1 BA + GA4+7) in separate experiments. GA4+7 and Promalin stimulated plants to grow taller. BA and Promalin promoted axillary shoot growth. Multiple applications of Promalin stimulated branching more than single treatments. Dry weight accumulation was stimulated only if the growth regulators were applied to 28–33-cm and not to 56-cm tall plants. Growth Regulators and the Phytochrome-Mediated Dormancy of Celery Seeds   总被引:2,自引:0,他引:2  
Seeds of five celery (Apium graveolens L.) cultivars germinated at 15°C in the light or dark but at 22°C only in the light. This light requirement was overcome by treatment with a mixture of the gibberellins GA4 and GA7 (GA4/7) but interactions of cytokinins, daminozide, ethephon, EDTA and N-phenyl-N′-4-pyridylurea (NC5392) with GA4/7 were observed. Varietal differences in response to GA4/7 concentration and the requirement for cytokinins were related to the upper temperature limits for germination of the different cultivars. Seeds of cultivars responding to low concentrations of GA4/7 appeared to contain less natural inhibitor than those requiring either high concentrations of GA4/7 or cytokinin in addition to low GA4/7. The cytokinin requirement for germination was partially removed by leaching the seeds with water. Interaction studies with applied hormones indicated that in seeds incubated in the light inhibition by abscisic acid was partially alleviated by N6-benzyladenine but not by GA4/7 application. The implications of these results are discussed in relation to the involvement of natural plant hormones in the dormancy mechanism of celery seeds.  相似文献

Shoots of mature grafted propagules of Picea abies (L.) Karst. metabolized [3H]gibberellin A4 (GA4) to at least 14 acidic substances, two of which were tentatively identified by gas-liquid radiochromatography as GA2 (possibly an artifact) and GA34. [3H]GA9 was converted into a number of metabolites, one of which was chromatographically similar to, but not identical with, GA4. Metabolism was maximally 61 and 57% over 48 hours for GA4 and GA9, respectively, and was correlated with the rate of change (i.e. increase followed by decrease) in endogenous GA-like substances as shoot elongation progressed. Propagules covered with a clear plastic film, a treatment which promotes flowering, metabolized [3H]GA4 more slowly than did control plants in the open. Inasmuch as a GA4/7 mixture can also promote flowering in P. abies, the retarded metabolism of [3H]GA4 may reflect the manner in which trees under plastic metabolize endogenous GA-like substances. If so, then the stimulating effect of this cultural treatment on flowering may come about through an increased level of endogenous, less polar GA-like substances.  相似文献

The white-light-(WL) induced enlargement of dicotyledonous leaf cells is known to occur via an acid-growth mechanism; i.e., WL causes leaf cells to excrete protons which lead to an increase in wall extensibility and thus cell enlargement. Gibberellic acid (GA3) and N6-benzyladenine (BA) also induce leaf cell enlargement. To see if they also act via acid-induced cell wall loosening, a comparison has been made of WL-, GA3-and BA-induced growth of strips, taken from primary leaves of bean (Phaseolus vulgaris L.) plants raised in continuous red light for 10 d. White light, GA3 and BA all increased wall extensibility as measured by the Instron technique, and this change preceded the increase in growth rate. However, whereas WL induced significant proton excretion, neither GA3 nor BA caused any acidification of the apoplast. Furthermore, neutral buffers, which effectively inhibited the growth induced by WL, were without effect on growth promoted by either GA3 or BA. These results indicate that while WL, GA3 and BA all initiate growth in bean leaves by altering cell-wall properties, GA3 and BA do so through some wall loosening mechanism other than wall acidification. The effects of a plant-derived smoke extract, BA and GA3 on the thermodormancy of Grand Rapids lettuce seeds were studied. Thermodormant lettuce seeds treated either with BA, GA3 or smoke extract alone did not germinate. Combinations of BA with smoke extract and BA with GA3 were most effective in overcoming induced thermodormancy. GA3 plus smoke did not break the induced thermodormancy. The effects of the different treatments on germination were concentration dependent. Gibberellin A4/7 mixture (GA4/7), a highly effective promoter of early and enhanced flowering in the Pinaceae, caused a significant reallocation of dry matter and 14C-photosynthate within terminal buds of Pinus radiata D. Don within 8 days of hormone treatment. Treatment with GA4/7 to terminal shoots of vigorous, potentially flowering mature grafted propagules reduced the flow of photoassimilated 14C and dry matter into the terminal bud as a whole, but significantly increased the dry matter and 14C allocated within the bud to developing long-shoot primordia (potential seed-cone buds). This was accomplished at the expense of the structural tissues, the apical dome region, and the vegetative branch buds. Although GA3 caused a similar reallocation of dry matter within the terminal bud, it was significantly less effective than GA4/7 thus appears to have, in addition to any nutrient diversion abilities, a distinct morphogenic function in sexual differentiation.  相似文献

Ethephon (Eth), gibberellin A3, A4 + 7 (GA3, GA4 + 7), and 6-benzyladenine (BA) removed secondary dormancy of Amaranthus caudatus seeds. The GAs and BA potentiated the effect of ethephon or 1-aminocyclopropane-1-carboxylic acid (ACC), an ethylene biosynthesis precursor, in terms of the rate or final percent of germination. Aminoethoxyvinylglycine (AVG), an ACC synthase activity inhibitor, was observed to simultaneously inhibit the release from dormancy effected by GA3 or BA as well as the ethylene production stimulated by these regulators. Breaking of secondary dormancy by GA3, GA4 + 7 or BA was prevented by 2,5-norbornadiene (NBD), an inhibitor of ethylene binding. Ethylene completely or markedly reversed the inhibitory effect of NBD. We thus conclude that the removal of secondary dormancy in Amaranthus caudatus seeds by gibberellin or benzyladenine involves ethylene biosynthesis and action.  相似文献

The fern Asplenium nidus L. is in great demand as an ornamental plant. The aim of this work was to investigate the influence of phytohormones in promoting a gametophytic and sporophytic growth in homogenized sporophytes tissue. Exogenous application of 0.5 and 5 μM N 6-benzyladenine, 0.05 and 0.5 μM indole-3-acetic acid (IAA), and 0.3 and 3 μM gibberellic acid (GA3) favoured sporophyte regeneration, whereas gametophyte regeneration took place when plant material was cultured in a hormone-free liquid MS medium. The endogenous contents of the auxin IAA, the cytokinins trans-zeatin, trans-zeatin riboside, dihydrozeatin, dihydrozeatin riboside, isopentenyladenine and isopentenyladenosine, and the gibberellins GA1, GA3, GA4, GA7, GA9 and GA20 in growing gametophytes and sporophytes were evaluated. Similar levels of the auxin and cytokinins and qualitative differences in the gibberellins were found between both generations.  相似文献

Treatment of mature citrus fruit (Citrus sinensis) with ethylene induced rapid chlorophyll destruction, a rise in respiration, a release of free amino acids, an accumulation of reducing sugars, and an appearance of phenylalanine ammonia lyase activity. Gibberellin A3 (GA3) and N6-benzyladenine (BA) opposed the effects of ethylene on chlorophyll, amino acids, and to a lesser extent, reducing sugar levels. The ethylene-induced respiratory rise was only slightly modified by GA3 and BA. Phenylalanine ammonia lyase activity was not affected by GA3.  相似文献

GA12-aldehyde obtained from mevalonate via ent-kaurene, ent-kaurenol, ent-kaurenoic acid and ent-7α-hydroxykaurenoic acid in a cell-free system from immature seeds of Cucurbita maxima was converted to GA12 by the same system. When Mn2+ was omitted from the system GA12-aldehyde and GA12 were converted further to several products. Among these GA15, GA24, GA36 and GA37 were conclusively identified by GC-MS. With the exception of GA37 these GAs have not previously been found in higher plants. Another biosynthetic pathway led from ent-7α-hydroxykaurenoic acid to very polar products via what was tentatively identified as ent-6α, 7α-dihydroxykaurenoic acid. An unidentified component with an MS resembling that of a dihydroxykaurenolide was also obtained from incubations with mevalonate.  相似文献

Changes in endogenous gibberellin-like substances (GAs) and related compounds in the shoot apices of Lolium temulentum during and after flower induction by one long day was examined for plants grown in three consecutive years. The total GA level in the shoot apical tissue was high (up to 42 micrograms per gram dry weight, or 3 × 10−5 molar GA3 equivalents), increasing several-fold on the day after the long day and then declining. Of the many GA-like substances present, the putative polyhydroxylated components—with HPLC retention times between those of GA8 (three hydroxyls) and GA32 (four hydroxyls), and accounting for about a quarter of the total GA activity—were most consistent and striking in their changes. Their level in the apices increased 3- to 5-fold on the day after the long day and then subsided. When various GAs were applied to plants in noninductive short days, flower initiation was induced by several, most notably by GA32, GA5, 2,2-dimethyl GA4, GA3, and GA7. GA32 was most like one long day in eliciting a strong flowering response while having little effect on stem growth, whereas GA1 had the opposite effect. It is suggested that highly hydroxylated C-19 GAs may play a central role in the induction of flowering in this long-day plant.  相似文献

Significant male and female flowering (cone bud production) by girdled branches of 6-year-old Douglas fir (Pseudotsuga menziesii (Mirb. Franco) seedlings was promoted by applications (mid-April to June) of 1.6 or 3.2 mg per branch (in total) of certain non-polar gibberellins (GA's). Girdling alone was ineffective. When tested alone, a mixture of GA4/7 was most effective. GA9 less so, while GA5 and the more polar GA3 were essentially ineffective. For female cone buds GA4/7+ GA9 were synergistically effective, but for male cone buds GA4/7 alone was best. The auxin naphthaleneacetic acid (NAA) was not tested alone, but at low dosage (0.175 mg/branch in total) NAA enhanced the flowering efficacy of GA's for both sexes; at a high dosage (0.875 mg/branch in total) male cone bud production was further enhanced, but only at the expense of females. For female flowering the best treatment (90% frequency of flowering 6.8 cone buds/branch), was GA4/7+ GA9+ low NAA; for male flowering, it was GA4/7+ high NAA (30% frequency and 4.2 cone buds/branch. Frequency of flowering for controls was 18% and 0%, average number of cone buds/branch was 0.9 and 0, for females and males, respectively. The successful treatments did not affect promordia initiation, rather they caused the differentiation of previously initiated, but undetermined, lateral primordia into cone and latent buds at the expense of vegetative bud differentiation. The lack of success reported by earlier workers in promoting flowering in Pinaceae species by GA's appears to be the unfortunate result of selecting GA3 for initial testing. The practical implications of this early and enhanced flowering by non-polar GA's seedlings of a commercially important conifer are discussed in relation to accelerating the processes of tree improvement.  相似文献

Significant female flowering of 6- to 11-year-old seedlings and grafted ramets of sexually mature scions of lodgepole pine (Pinus contorta Dougl.) was promoted by both topical and spray applications of a gibberellin (GA) A4/7 mixture (1.6 to c. 5 mg per plant in total) during that period (June to September) when sexual differentiation of lateral primordia would be expected to take place. Girdling was used in most experiments to enhance the GA4/7 effect, as was the auxin, naphthaleneacetic acid (NAA). Average frequency of flowering branches on treated plants over all experiments ranged from 27 to 59% (control ranged from 0 to 36%) and average number of female strobili was increased from 2- to 6-fold by growth regulator treatment, relative to controls. Within an experiment, clonal or family frequency of flowering for treated plants ranged from 11 to 67% (controls were 0 to 28%), and number of female strobili was increased from 2- to 14-fold by growth regulator treatment, relative to controls. Movement of the flowering stimulus from the point of application was apparent in several experiments, the response in adjacent branches being correlated positively with increasing dosage of GA4/7. Significant male flowering occurred only in one experiment, girdling and GA4/7 treatment being promotive factors. The use of spray applications of GA4/7+ NAA is warranted to induce early and enhanced flowering in lodgepole pine seedlings and vegetative propagules for genetic improvement programs.  相似文献

Summary Somatic embryogenesis was induced in callus cultures derived from nucellar tissue of cashewnut (Anacardium occidentale L.). Callus was obtained from nucellar tissue after 3 wk of culture on semisolid Murashige and Skoog (MS) basal medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D, 5 μM)+gibberellic acid (GA3, 15 μM)+N6-benzyladenine (BA, 5 μM). This callus gave rise to an embryogenic mass after 9 wk on maintenance medium containing 2,4-D (10 μM)+GA3 (15 μM)+4% sucrose +0.5% activated charcoal +10% coconut water (CW) +0.05% casein hydrolysate (CH). The embryogenic mass, after transfer to medium supplemented with 2,4-D (5 μM)+GA3 (30 μM)+4% sucrose +0.5% activated charcoal +10% CW +0.05% CH, gave rise to somatic embryos. The developmental stages of somatic embryos were observed using light and stereo microscopes. Histological study of somatic embryo development was also carried out. The present study would be useful for clonal propagation, and variety improvement in cashewnut, which is essential due to its increasing demand and export potential.  相似文献

Callus was induced on the wounded immature seeds and mature zygotic embryos of Dysosma pleiantha (Hance) Woodson (Berberidaceae) on a medium based on Murashige and Skoog's (1962) formula supplemented with 1 mg/l 2,4-dichlorophenoxy-acetic acid (2,4-D). Spontaneous embryoid formation occurred on the media containing low concentrations of 2,4-D (0.1–0.5 mg/l). These embryoids germinated in either MS or B5 medium containing 1 mg/l N6-benzyladenine and 1 mg/l gibberellic acid. By GC-MS the following acidic constituents of the endosperm of Echinocystis macrocarpa were identified: abscisic acid and its trans,trans-isomer, 4′-dihydrophaseic acid, GA4, GA7, iso-GA7, GA24, GA25, two isomers of GA13, GA43, ent-6α,7α,17-trihydroxy-16αH-kauran-19-oic acid and ent-6α,7α, 16β, 17-tetrahydroxykauran- 19-oic acid. The structures of the last three new natural products were confirmed by partial synthesis. ent-Kaurene was detected in the neutral fraction.  相似文献
