The mycelium of the fungusPleurotus ostreatus was cultured on a synthetic medium with growth regulators. Best growth was observed on media with 100 ppm IAA, 200 ppm GA3 and 200 ppm of kinetin. Growth of terminal hyphae and the appearance of colonies differed characteristically. Auxin bio-assays showed that the content of substances in younger mycelium slowly increased during cultivation without conspicuous changes in the medium. After exogenous application of IAA the content of auxins in younger mycelium increased and the content of IAA decreased gradually in the culture medium. Gibberellin bio-assays showed their content in the mycelium fell during cultivation and while it increased in the medium. Application of GA3 to the medium leads to a significant increase in these substances in the young mycelium and in the medium their level remains the same. Cytokinin bio-assays showed their content to vary during cultivation. An addition of kinetin to the nutrient medium results in an increase of cytokinins in the mycelium while in the medium the level of cytokinins is maintained. Addition of one substance thus influences the levels of other endogenous regulators.  相似文献

Summary The effect of 3-indoleacetic acid (IAA), 6-furfurylaminopurine (kinetin), and gibberellic acid (GA3) on germination of the orchid Comparettia falcata was evaluated in a factorial experiment (4×4×4) with Murashige and Skoog (1962) basal medium. It was established that seeds of this orchid could be maintained under aseptic conditions as long as the necessary nutrients and appropriate concentrations of growth regulators were provided. Of the three growth regulators used, IAA significantly decreased seed germination of Comparettia falcata. There was a synergistic effect in the kinetin:GA3 combination that produced a positive response in both percentage seed germination and development of seedlings. This study describes a single medium-based protocol able to achieve more than 160000 seedlings within 21 wk, starting from a single capsule, sufficient for both large-scale propagation and in vitro conservation of this threatened orchid.  相似文献

The percent germination and seedling growth of chickpea (Cicer arietinum L. cv. PBG-1) decreased with increasing concentrations of exogenous polyethylene glycol 6000 (PEG). With 15% PEG in the growth medium germination was only 33% while with 10% PEG it was 58% as compared to 93% in control. Addition of gibberellic acid (GA3) and kinetin to medium containing 10% PEG increased the germination and seedling growth and the effect was maximum with 6 µM GA3 which was a better inducer of growth and germination under reduced water potential than kinetin. However, indole acetic acid (IAA) inhibited germination and growth of stressed seedlings. The activity of amylase in cotyledons under stress was significantly increased with GA3 while kinetin and IAA were less effective. Gibberellic acid also enhanced the mobilization of starch from cotyledons of stressed seedlings resulting in low starch levels in cotyledons compared with stressed seedlings.  相似文献

The effects of gibberellic acid (GA3) and the growth retardant AMO-1618 on ovule formation in excised pistils of Nigella sativa L. were studied by sterile culture techniques. Gibberellic acid promoted pistil growth and inhibited ovule formation. The role of endogenous gibberellins in ovule formation and pistil growth was investigated by adding AMO to the basal medium. Both pistil lengths and ovule formation were reduced significantly with increasing concentrations of AMO. The addition of low concentrations of GA3 to the medium restored pistil growth but did not reverse the inhibitory effect of AMO on ovule formation. The addition of kinetin or indoleacetic acid (IAA) to the medium containing AMO had no effect on pistil lengths. However, with the addition of 10−7 m kinetin, the number of ovules in pistils was increased but not to the levels found in pistils grown in the absence of AMO.  相似文献

The antagonistic effects of some growth regulators [i.e. indol-3-yl-acetic acid (IAA), gibberellic acid (GA3) or kinetin] on stress imposed by sea water on leaf area, pigment and photosynthetic activity in leaves of broad bean plants at different stages of development were investigated. Seed priming with GA3 alleviated either partially or completely the effects induced by the two levels of sea water (10 and 25 %) used on leaf area at all experimental stages. However, IAA, GA3 and kinetin inhibited leaf growth by themselves in almost all measurements. Seed pretreatment with kinetin alleviated the inhibition of pigment production in sea water-irrigated plants. Furthermore, GA3 or kinetin nullified the deleterious effects imposed by irrigation with sea water particularly the high level (25 %) on photosynthetic14CO2 fixation.  相似文献

A method to obtain plants from embryogenic callus of Brassica nigra and protoplasts of hypocotyl expiants is described. Callus was initiated on Murashige and Skoog medium containing kinetin (kn) and 2,4-dichlorophenoxy acetic acid (2,4-D). Lowering of auxin induced embryo formation. Supplementation with gibberellic acid (GA3) enhanced embryogenic response tenfold. Passage through liquid medium devoid of growth regulators was essential for the growth of embryos. Secondary embryos were produced on transfer to solid basal medium. Embryogenic callus retained its morphogenic ability even after 12 subcultures. Both primary and secondary embryos produced fertile plants. Hypocotyl-derived protoplasts were also regenerated to plants following the same protocol. The survival of plants on transfer to soil was about 80%. Different plant explants of Persian buttercup (Ranunculus asiaticus L.) were screened for callus induction and adventitious shoot regeneration on different media to establish totipotent cultures. Murashige & Skoog (MS) medium was used, supplemented with different concentrations of the following growth regulators: kinetin, benzyladenine (BA), naphthaleneacetic acid (NAA) and indoleacetic acid (IAA). Callus was induced and adventitious buds regenerated only from cotyledonary explants after 4–5 weeks. Subculture of the regenerated buds on the same basal medium in presence of gibberellic acid (GA3) and BA produced well-organized shoots. Rooting was obtained by transferring shoots to growth regulator-free MS medium. A high rate of shoot multiplication has been achieved on medium with high concentration of kinetin and long-day photoperiod. Finally the plants were successfully transferred to soil and grown in a greenhouse.  相似文献

Fourteen d after pollination they were deprived of achenes and the receptacles were treated for 3 d with a mixture of indoleacetic acid (IAA), gibberellic acid (GA3) and kinetin at a concentration of 3·10-4 M. Then [14C]sucrose and 32PO 4 3- were applied to the surface of the leaves or to the root medium. Young excised floral buds of Aquilegia were grown on defined medium containing kinetin, indoleacetic acid (IAA), or gibberellic acid (GA3). Only when 10−6 or 10−7 m kinetin was added to the basal medium was there a significant increase in the number of initiated whorls of primordia. Buds on the basal medium or on medium with IAA or GA3 failed to initiate carpels. On medium with 10−6 or 10−7 m kinetin, buds successfully initiated a normal whorl of five carpels. A high level of inorganic nitrogen was also required for the initiation of carpels. With 10−5 m kinetin, individual buds initiated from 6–18 carpels. Staminodial primordia of these buds were replaced with carpels, or the floral apex enlarged to accommodate a single whorl of many carpels. Kinetin did not support the further differentiation of the floral organs. Sepals, petals, and carpels did differentiate on medium with GA3, but stamens aborted. However, on medium with GA3 and kinetin, stamen primordia differentiated into short filaments and anthers. Further unknown growth factors appear to be required for the complete differentiation of floral primordia into mature organs.  相似文献

The interrelationship between ethylene and growth regulators in the senescence of romaine lettuce (Lactuca sativa L.) leaves was studied. Gibberellic acid (GA3), kinetin, and 3-indoleacetic acid (IAA) retarded chlorophyll loss from leaf discs which were floated on hormone solutions. Abscisic acid (ABA) and ethephon enhanced chlorophyll loss and antagonized the senescence-retarding effect of GA3 and kinetin. A high concentration of IAA (10–4 M) caused accelerated chlorophyll loss, whereas a similar concentration of kinetin neither retarded nor promoted chlorophyll loss. The ineffectiveness of IAA and kinetin at their supraoptimal concentrations in retarding leaf senescence was related to increased production of ethylene induced in the treated leaf discs. GA3 was the most effective in retarding chlorophyll loss and did not stimulate ethylene production at all. The senescence-enhancing effect of ABA was not mediated by ethylene. However, the moderately increased production of ethylene, induced by relatively high concentrations of ABA, could act synergistically with the latter to accelerate chlorophyll loss. The effects of indole-3-acetic acid (IAA), abscisic acid (ABA), gibberellic acid (GA3) and kinetin on the hydrolytic activity of proton pumps (adenosine triphosphatase, H+-ATPase, pyrophosphatase, H+-PPase) of tonoplasts isolated from stored red beet (Beta vulgaris L. cv. Bordo) roots were studied. Results suggest that the phytohormones can regulate the hydrolytic activities of H+-ATPase and H+-PPase of the vacuolar membrane. Each of the proton pumps of the tonoplast has its own regulators in spite of similar localization and functions. IAA and kinetin seem to be regulators of the hydrolytic activity for H+-PPase whereas for H+-ATPase it may be GA3. Abscisic acid (ABA) inhibits the production of ethylene induced by water stress in excised wheat leaves and counteracts the stimulatory effect of 6-benzyladenine (BA) on this process. The stimulatory effect of BA and the inhibitory effect of ABA were equally pronounced whether external or endogenous ethylene levels were determined. When leaves were sprayed or floated on solutions of BA, indole-3-acetic acid (IAA), gibberellic acid (GA3), or ABA, the relative activities of these growth regulators on stress-induced ethylene at 10-4 mol l-1 were BA>IAA >GA3>controls>ABA. In non-stressed leaves, however, where the levels of ethylene produced were 2–20 times smaller, the relative activities were IAA >BA>GA3>controls>ABA. The effects of BA and ABA spray treatment on water stress induced ethylene were closely similar whether the solutions were applied 2 or 18 h prior to the initiation of water stress. In Nigella damascena, where “doubleness” behaves as a single gene character, recessive to the normal wild “single” form, we detected a number of differential responses of cultured floral apices of the two forms to plant growth regulators. “Single” buds initiated all organ types on a defined medium lacking any plant growth regulators. “Double” buds, however, required either kinetin or GA3 for the development of a normal organ complement. Kinetin enhanced the growth and development of buds of both genotypes, whereas GA3, at all concentrations, specifically inhibited the initiation and growth of stamens and nectaries in “single” buds. No combination of IAA or kinetin overcame this inhibition of “single” buds by GA3. In the presence of both IAA and GA3, “single” buds, following the initiation of normal carpels, initiated a few abnormal carpelloid structures from regions which had failed to produce stamens and nectaries. These observations, coupled with other studies including those on the endogenous levels of gibberellin in the two genotypes, suggest that “doubleness” correlates strongly with a modification of gibberellin metabolism. The causal connection between the two phenomena is as yet to be discovered.  相似文献

All the concentrations (25-150 mgl-1) of the phytohormones kinetin, IAA (indol-3-ylacetic acid) and GA3 (gibberellic acid) increased the activity of DCPIP (2,6 dichlorophenolindophenol)-Hill reaction, chlorophyll and protein contents over the control data in leaves ofSechium edule Sw. on Darjeeling hill of the Eastern Himalayas; while ethrel (2-chloroethylphosphonic acid) treatments decreased these parameters in the hilly species. The most effective concentrations in increasing these parameters were 50 mg 1-1 of kinetin, 50 mg 1-1 of IAA and 100 mg 1-1 of GA3; whereas 50 mg 1-1 of ethrel was most effective in decreasing these parameters during the induction of senescence in the hilly vegetable crop. The increase in these parameters was greatest with kinetin, followed by IAA and least with GA3 in the hilly plant species studied.  相似文献

Adventitious root formation in stem cuttings of mung bean was enhanced by ethrel, which had an additive effect when employed simultaneously with indolebutyric acid (IBA). Abscisic acid (ABA) did not influence the number of roots per cutting whereas gibberellic acid (GA3) and kinetin were without effect on rooting at lower concentrations but were inhibitory at higher concentrations. Nevertheless, all three of these chemicals showed synergistic interactions with IBA and/or indol-3-ylacetic acid (IAA) and thereby significantly promoted root formation. A localised application of morphactin to the epicotyl of cuttings totally inhibited root production irrespective of which of the foregoing growth regulators were suppliedvia the hypocotyl. Morphactin application also prevented root formation in cuttings treated with vitamin D2. The various growth regulators employed had differing effects on growth of roots but there was no simple relationship between their effects on root formation and subsequent root growth.  相似文献

A field experiment was conducted to investigate the effects of presoaking the wheat grains (Triticum aestivum L.) in different levels of salinity (33 or 66 mM) and in growth regulators (indolyl-3-acetic acid, IAA at SO g m-3, gibberellic acid, GA3 at 100 g m-3, or kinetin at 100 g m-3) on the shoot growth and pigment content of the developing wheat flag leaf. Salinity at 33 or 66 mM led to an insignificant increase in the fresh and dry masses as well as in the shoot diameter and shoot length, but it attenuated the flag leaf area. In the majority of cases, salinity increased the chlorophyll (Chla, Chlb) and carotenoid contents as well as the number of chloroplasts per a mesophyll cell. The growth in the wheat shoot of the saline-treated plants was, in general, stimulated in response to presoaking the grains in kinetin or GA3. On the other hand, IAA + salinity led to a negligible effect on the growth in the wheat plants particularly at the early stages of growth. The presoaking of grains in NaCl at 33 mM + IAA or 66 mM + kinetin induced a marked increase in the pigment content of the wheat flag leaf particularly at the early stages of growth. The interaction between salinity and phytohormones increased the number of chloroplasts; kinetin was the most effective.  相似文献

The effects of three growth regulators, namely gibberellic acid (GA3), kinetin (6-furfuryl-aminopurine) and α-naphthyl-acetic acid (NAA), each at five concentrations (0, 25, 50,102 and 103 mg/litre) on the growth of coconut seedlings (Cocos nucifera) were examined. GA3 at 25 mg/litre increased growth in height; higher concentrations had no significant effect. Kinetin at 102 and 103 mg/litre and NAA at all concentrations reduced height growth compared with the control. No treatment caused a significant increase in total dry weight.  相似文献

Summary On a simple nutrient medium in explants from roots of Cichorium intybus form shoots visible after about 14 days. Gibberellic acid (GA3) does not influence the spontaneous development of the chicory explants. GA3 in combination with kinetin inhibits shoot formation whereas kinetin alone promotes the process. On the other hand high concentrations of IAA inhibit the regeneration of shoots.The soluble proteins of chicory cultures treated with growth regulators were examined by disc-electrophoresis. It was shown that the proteins detected by staining with Amido black, phosphatases, esterases and glutamate dehydrogenase (GDH) present in the original root tissue remained constant under the different culture conditions during a period of 12 days. The quantitative changes of some of the proteins, phosphatases and esterases observed during the culture period were identical for all the different cultures in spite of the different morphogenetic behaviour. Only the activities of GDH and peroxidase changed after treatment with different growth regulators; however, in these cases, there was also no direct connection with the morphogenetic responses of the cultures.The specific activity of the GDH-band was promoted by IAA and at the same time the formation of peroxidases was inhibited. Kinetin delayed the formation of peroxidases during the first days of the culture period but promoted it later on. There was a repression by IAA of a specific kationic peroxidase. In the tissues treated with GA3 the activity of peroxidases was always higher than in the control tissue. This effect of GA3 can be partly explained by the fact that GA3 inhibits the release of peroxidases of the explants into the nutrient medium.  相似文献

Hypocotyl sections and isolated meristem-tips from 7- to 10-day-old seedlings of flax were cultured on a Murashige and Skoog (1962) medium. The optimum concentrations of naphthaleneacetic acid (NAA) and benzyladenine (BA) were determined for shoot and root initiation from hypocotyls. Meristem-tip cultures differentiated shoots and roots in response to these concentrations. The frequency and yield of shoots, promoted by BA, decreased if NAA or gibberellic acid (GA3) was included in the medium. At high NAA concentration GA3 and BA inhibited root initiation. Morphological development of cultures differed depending on the growth regulators in the medium.  相似文献
