Summary InSmittia embryos, anterior UV irradiation causes the formation of double abdomens in which head and thorax are replaced by a mirror-image duplication of abdominal segments. The transformation is ascribed to the inactivation of cytoplasmic ribonucleoprotein particles acting as anterior determinants. During the first 5 h after egg deposition (h.a.d.), high yields of double abdomens are produced by microbeam irradiation of a small target area (diameter 20 m) behind the anterior pole. This localization coincides with a yolk-free cytoplasmic cone observed behind the anterior pole of newly depositedSmittia eggs (Zissler and Sander 1973). Irradiation of this area becomes less efficient between 5 and 7 h.a.d., while double abdomen yields increase after irradiation of adjacent target areas. A dramatic increase in the double abdomen yield is also observed after irradiation of the entire anterior surface. Under these conditions, the UV fluence required for a given yield decreases by a factor of 3.9 between 5.5. and 7 h.a.d. This can be explained quantitatively by the assumption that the effective targets move an average distance of 8 from the strongly absorbing endoplasm towards the egg surface. During the period of apparent target deshielding, the distance of nuclei from the egg surface actually decreases from 11 to 3 m. However, the timing and direction of the nuclear migration and of the apparent deshielding differ, indicating that the shift of effective targets may be triggered by, but does not exactly follow, the nuclear movement. The data suggest that a major fraction of anterior determinants shifts from a small volume behind the anterior pole to a spread-out distribution over the surface of the anterior half.  相似文献

Mediterranean beaches are very crowded during summer and, because of the high values of solar UV radiation, the potential risk for human health is relevant. In this study, all-day measurements of biologically effective global and diffuse UV radiation for skin (UVBEeryt) and eye (UVBEpker, UVBEpconj, UVBEcat) disorders were carried out on differently tilted surfaces on a summers day on a Mediterranean beach. The role played by beach umbrellas in protection from excessive sun exposure was also investigated. Erythema, photokeratitis and cataract seem to require almost the same exposure time to reach the risk threshold dose. Under full sunlight, the highest global and diffuse UV values are reached on surfaces normally oriented towards sunlight and on horizontal surfaces, respectively. Over vertical surfaces, at this northern hemisphere site, global and diffuse UV radiation reaches maxima values in the south-facing direction around noon, while maxima values are reached early in the morning and late in the afternoon over surfaces facing east and west, respectively. The quality of the beach umbrellas protection (efficiency in blocking solar UV radiation) varies with surface orientation; the highest efficiency for our specific site and geometrical conditions occurs over horizontal surfaces, with efficiency being least over vertical surfaces when incident radiation values are still relevant.  相似文献

Projected area factors for individual segments of the standing and sedentary human body were modelled for both direct and diffuse solar radiation using detailed 3D geometry and radiation models. The local projected area factors with respect to direct short-wave radiation are a function of the solar azimuth angle () between 0°<<360° and the solar altitude () angles between –90°<<+90°. In case of diffuse solar radiation from the isotropic sky the local human projected area factors were modelled as a function of the ground albedo () ranging between 0<<1. The model was validated against available experimental data and showed good general agreement with projected area factors measured for both the human body as a whole and for local quantities. Scientists can use the equations to predict the inhomogeneous irradiation and absorption of direct and diffuse solar radiation and UV-radiation at surfaces of the human body. In conjunction with detailed multi-node models of human thermoregulation the equations can be used to predict the physiological implications of solar radiation and outdoor weather conditions on humans.  相似文献

Summary The present work deals with the processes involved in the abiogenic polycondensation of nucleotides adsorbed on the clay mineral kaolinite under the action of ultraviolet (UV) radiation. The dependence of the yield of synthesis products on irradiation dose was studied. The maximum yield corresponds to a 6-h exposure. The newly synthesized substances were analyzed by ion-exchange chromatography. Some fractions were studied for the type of bonds they contained by venom phosphodiesterase and RNase T2 enzymatic hydrolysis. It was determined that some of the products synthesized by exposure of AMP adsorbed on the surface of clay particles to UV radiation may be looked upon as oligonucleotides in which some fragments have 2–5-bonded and others 3–5-bonded nucleotides  相似文献

Summary Post-irradiation DNA degradation in P. mirabilis rec + strains after UV irradiation is found to be more extensive in starvation buffer than in growth medium. In growth medium restriction of protein synthesis, but not DNA synthesis, largely prevents the expression of breakdown limitation. By the addition of chloramphenicol during post-irradiation incubation in growth medium the expression of break-down limitation was followed and found to occur 20 to 40 min after UV irradiation. Pre-irradiation by a low dose of UV leads after a corresponding time of post-irradiation incubation to breakdown limitation even in starvation buffer after a second UV exposure.Post-irradiation DNA degradation is presumed to be initiated at the sites of DNA lesions which arise at replication points damaged by UV. While pre-starvation restricts the efficiency of postirradiation DNA degradation by the reduction of the number of replication points active at the time of irradiation, caffeine as well as 2,4-dinitrophenol inhibit DNA degradation even in rec - cells probably by the interference with nicking or exonucleoltytic events initiated at those sites in the absence of breakdown limitation.Breakdown limitation is postulated to be due to inducible derepression of REC-functions which lead to the protection and, probably, repair of DNA lesions arising at the replication points following UV exposure.  相似文献

Effects of various physical and chemical treatments of Colorado potato beetle [Leptinotarsa decemlineata (Coleoptera: Chrysomelidae)] eggs on parasitization and development of the egg parasitoid Edovum puttleri (Hymenoptera: Eulophidae) were investigated. UV irradiation did not affect host acceptance but reduced host suitability for UV exposure times 90 min. Susceptibility of host eggs to UV irradiation varied with host age; eggs were most vulnerable to damage from irradiation at 12, 18, and 24 h post-oviposition. The rate of parasitization also was influenced by host age. Percent parasitization was greatest in freshly laid eggs and 24–30 h old eggs. Seventy-seven percent of host eggs frozen at –20 °C (5 min) were parasitized by E. puttleri, but extended exposure of eggs to –20 °C reduced both acceptance and suitability. Host eggs that had been washed with hexane (removal of kairomone and sticky layer) also were parasitized. After 5 min of washing, application of kairomone significantly increased the rate of parasitism (from 74.7% to 88.2%), but with longer periods of washing, kairomone application had no significant effect on percent parasitism. Thus, the sticky material(s) coating the egg did not appear to be essential for parasitization to occur. Our results provide effective methods and times for treating Colorado potato beetle eggs to maximize parasitization and development of E. puttleri.  相似文献

Effects of Pb2+, Ni2+, Hg2+ and Se4+ on cultured human glioma U-343MG cells were investigated considering uptake, toxicity and, in combination with radiation, clonogenic cell survival. The cells were exposed to 0-100 m of the metals for a week before the evaluation. The tests showed a tendency to toxicity with 10 m nickel although not significant (P > 0.05). Selenium, lead and mercury exerted a significant toxicity (P < 0.05) at 2.5 m, 10 m and 1 m, respectively. To challenge the clonogenic cell survival capacity, the cells were irradiated with60Co photons after being exposed to the highest nontoxic concentration of the different metals. The clonogenic cell survival tests, after irradiation, showed no significant change if the cells were exposed to 5 m nickel, 0.5 m selenium or 5 m lead compared with those not exposed. Mercury, 0.1 m, gave a relative reduction in survival compared with only irradiated cells of 58 ± 17%. Thus, only mercury affected the radiation-induced damage and/or repair. When exposed to the highest nontoxic concentrations of the different metals, the cultures did not display a significant uptake ratio (metal concentration ratio of exposed cells to control cells) of nickel (3.1 ± 3.3), only a small uptake ratio of selenium (4.0 ± 0.4), while there was a large uptake ratio of both lead (2.6 ± 1.7) x 102 and mercury (1.5 ± 0.2) x 101. The results indicated that nickel was neither especially toxic nor influenced the clonogenic cell survival after irradiation. Mercury was more toxic and also influenced the radiation sensitivity. Lead was taken up strongly but did not influence the radiation sensitivity. Selenium accumulated but gave no detectable effect on the radiation sensitivity.  相似文献

A genetic system selecting for deletion events (DEL recombination) due to intrachromosomal recombination has previously been constructed in the yeastSaccharomyces cerevisiae. Intrachromosomal recombination is inducible by chemical and physical carcinogens. We wanted to understand better the mechanism of induced DEL recombination and to attempt to determine in which phase of the cell cycle DEL recombination is inducible. Yeast cells were arrested at specific phases of the cell cycle, irradiated with UV or -rays, and assayed for DEL recombination and interchromosomal recombination. In addition, the contribution of intrachromatid crossing-over to the number of radiation induced DEL recombination events was directly investigated at different phases of the cell cycle. UV irradiation induced DEL recombination preferentially in S phase, while -rays induced DEL recombination in every phase of the cell cycle including G1. UV and -radiation induced intrachromatid crossing over preferentially in G1, but it accounted at the most for only 14% of the induced DEL recombination events. The possibility is discussed that single-strand annealing or one-sided invasion events, which can occur in G1 and may be induced by a double-strand break intermediate, may be responsible for a large proportion of the induced DEL recombination events.  相似文献

-Crystallin, a major protein of the eye lens, is known to have chaperone activity in preventing heat-induced aggregation of enzymes and other crystallins. In this study, we investigate the ability of -crystallin to inhibit UV-light-induced aggregation of other lens proteins and the effect of exposure of -crystallin to UV irradiation on its chaperone activity. The chaperone activities of -crystallin preincubated at different temperatures were found to be different and could be correlated with its change in quaternary structure as determined by the fluorescence probe ANS (8-anilo-1-naphthalene sulfonate). -Crystallin can inhibit the aggregation of -crystallin from UV irradiation at room temperature, and the preheated -crystallins provide more protection than the native one. Upon irradiation by UV light, -crystallin gradually lost its ability to protect -crystallin against thermal aggregation. The loss of the chaperone efficacy of -crystallin to protect other lens proteins may shed light on human cataract formation induced by long-term exposure to UV irradiation.  相似文献

We used a device called a Phototron to measure the effects of UV radiation on the cosmopolitan algae, Cryptomonas erosa, grown in continuous cultures. In the Phototron, we investigated changes in photosynthetic parameters (Pmax – specific production rate at optimal light intensity; – initial slope of the linear portion of the Photosynthesis-Irradiance curve; and – the convexity or rate of bending) and carbon allocation as a function of irradiance at three different environmentally-realistic doses of UV radiation in unconditioned (no prior UV exposure) and conditioned algae (15 d previous UV exposure). For unconditioned control algae, Pmax-Total was lower, although not significantly, than the two highest UV treatments. For conditioned control algae, Pmax-Total was higher, although not significantly, than all UV treatments. Our data suggest that short term (4 h) exposure to low levels of UV (8.09 W m–2 unweighted) does not affect Pmax-Total in C. erosa, but does change the proportion of carbon allocated to lipids and proteins. Also, comparisons of lipids, polysaccharides and proteins as a percent of total carbon uptake between unconditioned and conditioned algae indicate that exposure history to UV radiation can have a negative impact on carbon allocation to lipids and proteins, in a wetland alga species that is crucial to the efficient transfer of energy through freshwater food webs.  相似文献

1. 1. Intensities of ultraviolet rays (UV) from every direction and the amount of UV absorbed by each body part was measured in four situations (track, beach, golf course and skiing slope).

2. 2. The intensity of UV was measured by a portable UV meter and the amount of UV absorbed by every body part was measured with a UV irradiation energy measurement label.

3. 3. The intensity of UV was strong from S45° and SE45° in all cases. On snow, the intensity of UV from every direction was stronger than in other situations, it was especially strong in reflection from below and from the north. The UV reflection from the sand was slightly larger than that from mud and grass.

4. 4. The experimental day's weather was fine and occasionally cloudy during golf, beach activity and gate-ball, and clear during skiing.

5. 5. The UV deal of a horizontal plane was 135, 98, 156 and 120 kJ/m2/h at golf, beach activity, gate-ball and skiing, respectively.

6. 6. This study suggests that players should be concerned with the prevention of UV radiation during sporting activity.

Carotenoids are essential components of the photosynthetic apparatus involved in plant photoprotection. To investigate the protective role of zeaxanthin under high light and UV stress we have increased the capacity for its biosynthesis in tobacco plants (Nicotiana tabacum L. cv. Samsun) by transformation with a heterologous carotenoid gene encoding -carotene hydroxylase (crtZ) from Erwinia uredovora under constitutive promoter control. This enzyme is responsible for the conversion of -carotene into zeaxanthin. Although the total pigment content of the transgenics was similar to control plants, the transformants synthesized zeaxanthin more rapidly and in larger quantities than controls upon transfer to high-intensity white light. Low-light-adapted tobacco plants were shown to be susceptible to UV exposure and therefore chosen for comparative analysis of wild-type and transgenics. Overall effects of UV irradiation were studied by measuring bioproductivity and pigment content. The UV exposed transformed plants maintained a higher biomass and a greater amount of photosynthetic pigments than controls. For revelation of direct effects, photosynthesis, pigment composition and chlorophyll fluorescence were examined immediately after UV treatment. Low-light-adapted plants of the crtZ transgenics showed less reduction in photosynthetic oxygen evolution and had higher chlorophyll fluorescence levels in comparison to control plants. After 1 h of high-light pre-illumination and subsequent UV exposure a greater amount of xanthophyll cycle pigments was retained in the transformants. In addition, the transgenic plants suffered less lipid peroxidation than the wild-type after treatment with the singlet-oxygen generator rose bengal. Our results indicate that an enhancement of zeaxanthin formation in the presence of a functional xanthophyll cycle contributes to UV stress protection and prevention of UV damage.  相似文献

Oral administration of immune milk, that had been obtained from cows immunized with a variety of human gut bacteria containingE. coli, S. typhimurium, S. dysenteriae and 23 others, protected AKR/J mice from the lethal effect of radiation, when immune milk was orally given to mice at 150 g kg–1 day–1 for 7 days prior to-irradiation of 8 Gy. Mean survival times were 24.8 days for the group given immune milk but only 16.8 days for the group given control milk from unimmunized cows. Enterobac teriaceae were detected in various organs such as liver, lung and kidney on day 13 after irradiation, whereas the numbers were significantly fewer in the study group as compared with the control group. And fewer number of intestinal Enterobacteriaceae were detected in the study group compared with the control group prior to irradiation. Immune milk also enhanced the mitogenic response of mesentric lymph node cells, the redirected cytolytic activity of intestinal intraepithelial lymphocytes to P815 tumor cells with anti-CD3 mAb, andin vitro killing activities of the phagocytes in mesenteric lymph nodes toE. coli as compared with control milk. These results suggest that immune milk may reduce the number of bacteria translocating from the intestinal-tract and augment the activities of the gut-associated lymphoid tissues against the invasion of intestinal bacteria, causing protection against the lethal effect of radiation.  相似文献

Summary Attempts to measure the nesting density or territory size of bald eagles led to a fundamental difficulty, inherent in all such measurements on organisms which are distributed along an irregular boundary, such as a coastline. The length of such a boundary is not a meaningful measure, and neither can a meaningful area be associated with each nest. Mandelbrot's (1983) fractal geometry applies to the problem, but has not previously supplied practical units of measurement for fractals such as coastlines or rugged surfaces. A practical method is given for measuring the extent of such fractals, introducing a unit of variable dimension, the metron, which includes the existing SI units of length, area and volume as special cases. A linear measure, the spacing allows densities on fractals of different dimensions to be compared directly. The method is applied to the distribution of bald eagle nests along the coastlines of two islands in the Aleutians, and an extension of the method to handle distributions on mountainsides and island surfaces is indicated.  相似文献

Flash-induced absorbance measurements at 830 nm on both nanosecond and microsecond timescales have been used to characterise the effect of ultraviolet light on Photosystem II core particles. A combination of UV-A and UV-B, closely simulating the spectrum of sunlight below 350 nm, was found to have a primary effect on the donor side of P680. Repetitive measurements indicated reductions in the nanosecond components of the absorbance decay with a concomitant appearance and increase in the amplitude of a component with a 10 s time constant attributed to slow reduction of P680+ by Tyrz when the function of the oxygen evolving complex is inhibited. Single-flash measurements show that the nanosecond components have amplitudes which vary with S-state. Increasing UV irradiation inhibited the amplitude of these components without changing their S-state dependence. Salmon from a 3 male×3 female sockeye salmon cross were raised under three different light regimes (UV: 300–850nm for 8 hoursday-1, visible: 400–850nm for 8 hoursday-1, and a control in which eggs were kept in darkness until hatching, followed by increasing amounts of light as the embryos approached swim-up) up to the parr interval (105 days from fertilization). The dose rates (Wm-2), and total intensities per day (Jm-2), for each treatment were less than 0.46 and 0.28 times of those measured in the shaded part of a stream, respectively. Egg mortality was higher for the UV and visible treatments compared to the control, and the number of alevins alive after day 83 was significantly different between treatments: highest for the control and lowest for the UV treatment. Eggs and hatched fish from female F2 suffered higher mortality than those from the other two females raising the possibility that sensitivity to UV-B in sockeye salmon may be either a maternally inherited trait or due to physiological or physical differences between egg groups. Alevins and parr from the control treatment transferred to the UV treatment suffered little mortality compared to animals raised in the UV and visible treatments from fertilization. These results, in conjunction with light measurements from streams, suggest that light and UV-B radiation could be a source of young salmon mortality in perturbed environments such as those resulting after forestry intervention.  相似文献

Summary Stationary phase cells of strain phr/MC 2 ofE. coli are not photoreactivable but the frequency of UV-induced mutations to low Streptomycine-resistance (S 3, 3/ml) is decreased strongly by illumination with light of fluorescence tubes (310 to 500 nm) after UV-irradiation. Also dark-reversion (DRM) of these mutations due to keeping UV-irradiated cells in saline is observed. Illumination before UV-irradiation decreases the frequency of the mutations (photoprotection against mutation=PPM) to the same extent as the combined action of photoreversion (PRM) and DRM. The lag-phase of cell division is prolonged strongly by illumination from 80 min without light to 150 min by the light-dose of highest activity. The additional lag is nearly the same if the illumination is done before, after or without UV-irradiation; this lag is about additive to the small lag caused by UV. Pre-illumination of the stationary-phase cells does not cause photoprotection against killing (PP), it even decreases the survival after high UV-doses. The observations support the hypothesis that PRM in this strain may be indirect, i.e. caused by the light-induced additional division lag which enhances the dark repair of UV-premutations. Also spontaneous premutations which are apparently present in the stationary-phase cells seem to be influenced by the light in this way.  相似文献

A variety of chemotherapeutic agents induce cell death via apoptosis. We had shown previously that gemcitabine (2,2-difluorodeoxycytidine) induced an atypical apoptosis in BG-1 human ovarian cancer cells; therefore, further studies were conducted to characterize more precisely gemcitabine-induced apoptosis in BG-1 cells compared to a general inducer of apoptosis, staurosporine. BG-1 cells exposed to 0.5, 1.0 and 10 M gemcitabine for 8 h, or staurosporine (1.0 M) for 6 h, exhibited high molecular weight DNA fragmentation (50 kbp); however, only staurosporine treatment produced internucleosomal DNA fragments (200 bp) in a laddered pattern on the agarose gel. Staurosporine (1.0 M) rapidly induced phosphatidylserine plasma membrane translocation that increased linearly with time as measured by annexin V-FITC binding, and similar kinetics were observed for caspase activation by staurosporine in BG-1 cells. In contrast, 10 M gemcitabine increased phosphatidylserine expression in a small fraction of cells (5–10%) vs. untreated controls over the course of 48 h and significant caspase activity was detected within 12 h of drug exposure. Time-lapse video microscopy of BG-1 cells exposed to 1.0 M staurosporine or 10 M gemcitabine for up to 72 h showed that the morphologic changes and kinetics of cell death induced by these agents differed significantly. We also evaluated the apoptosis induced by paclitaxel (a mitotic poison) and cisplatin (an agent not dependent on cell cycle functions) in BG-1 cells by these methods because these drugs are used clinically to treat ovarian cancer. Our findings demonstrate that the kinetics of apoptotic cell death induced by gemcitabine and other chemotherapeutic agents should be taken into account when designing treatment strategies for ovarian cancer.  相似文献

Summary The carotenoids ofLophocolea bidentata (L.)Dum. have been investigated by adsorption and partition TLC and through their visible and UV spectra. The following carotenes and xanthophylls have been isolated and identified: -carotene, -carotene, neo--carotene U, zeaxanthin, mono-cis-neoxanthin, trans-neoxanthin, poly-cis-neoxanthin, Violaxanthin neo V, violaxanthin, antheraxanthin, lutein, and lutein-5,6-epoxide.
