Experiments were carried out to explore the involvement of the plant hormone gibberellin (GA) in the light-induced germination of lettuce seeds. Three growth retardants known to be inhibitors of GA biosynthesis were tested for their effect on red-light-induced germination. Chlormequat chloride (CCC) and AMO-1618 had no effect, but ancymidol was strongly inhibitory. Moreover, the inhibition caused by ancymidol was completely overcome by GA3. CCC and AMO-1618 inhibit the formation ofent-kaurene, while ancymidol blocks the oxidation ofent-kaurene toent-kaurenoic acid. Ancymidol also was found to inhibit GA-induced dark germination of lettuce seeds, and this inhibition was partially reversed by higher levels of GA. Therefore, the results suggest two possibilities for the relationship between phytochrome and GA in this system: first, the rate-limiting step in the germination of light-sensitive lettuce seeds, that which is regulated by phytochrome, is the oxidation ofent-kaurene toent-kaurenoic acid. Alternatively, red-light treatment may result in the release of active GAlike substances which, in turn, induce germination. In either case the results presented here support the view that phytochrome exerts its effect on lettuce seed germination by means of GA rather than via an independent pathway.  相似文献

Slender barley: A constitutive gibberellin-response mutant   总被引:13,自引:0,他引:13  
In barley (Hordeum vulgare L. cv. Herta), slender (sln1) is a single-locus recessive mutation which causes a plant to appear as if it had been grown in sturating concentrations of gibberellin (GA). We have investigated two of the GA-mediated processes in slender barley, shoot elongation and the induction of hydrolytic enzymes in aleurone layers. Shoot elongation is severely retarded in normal (wild-type) barley if the biosynthesis of GA is blocked by an inhibitor, ancymidol (-cyclopropyl--(p-methoxyphenyl)-5-pyrimidinemethanol). However, the slender mutant continues to elongate in the presence of ancymidol. In isolated normal aleurone layers, the synthesis and secretion of -amylase (EC 3.2.1.1), protease (EC 3.4) and nuclease (EC 3.1.30.2) are induced by exogenously applied GA3. However, in the aleurone layers of the slender mutant these enzymes are produced even in the absence of GA but their synthesis is still susceptible to inhibition by abscisic acid. Bioassays of half-seeds of the slender mutant and their normal siblings show no detectable differences in endogenous levels of GA-like substances. We suggest that the slender mutation allows competent tissues to express fully, or over-express, appropriate GA-induced processes independent of GA. Summary Gibberellic acid inhibition of betacyanin biosynthesis has been studied in Amaranthus caudatus L. using the pigment precursors L-tyrosine and L-dihydroxyphenylalanine. Precursors fed to gibberellic acid (GA) treated seedlings completely recovered betacyanin synthesis while the GA induced growth enhancement remained unaltered. Chlorophyll loss in the leaves of cut flowering branches of Alstroemeria pelegrina L. cv. Stajello, placed in water in darkness at 20°, was inhibited by irradiation with red light and by the inclusion of gibberellic acid (GA3) in the water. The effects of red light were abolished when it was followed by far-red light. Effects of GA3 and red light were additive over a range of GA3 concentrations (0. 01–1 μ M ). Chlorophyll breakdown was increased by the inclusion of AMO-1618, ancymidol, or tetcyclasis in the water. The effect of these inhibitors of gibberellin synthesis was fully reversed by GA3. The inhibition of chlorophyll breakdown by red light was absent when AMO-1618, ancymidol or tetcyclasis were included in the water. The results indicate that leaf yellowing is controlled by endogenous gibberellins and that the effect of phytochrome is mediated by gibberellin synthesis.  相似文献

Phytochromes are a family of related chromoproteins that regulate photomorphogenesis in plants. Ectopic overexpression of the phytochrome A in several plant species has pleiotropic effects, including substantial dwarfing, increased pigmentation, and delayed leaf senescence. We show here that the dwarf response is related to a reduction in active gibberellins (GAs) in tobacco (Nicotiana tabacum) overexpressing oat phytochrome A under the control of the cauliflower mosaic virus (CaMV) 35S promoter and can be suppressed by foliar applications of gibberellic acid. In transgenic seedlings, high concentrations of oat phytochrome A were detected in stem and petiole vascular tissue (consistent with the activity of the CaMV 35S promoter), implicating vascular tissue as a potential site of phytochrome A action. To examine the efficacy of this cellular site, oat phytochrome A was also expressed using Arabidopsis chlorophyll a/b-binding protein (CAB) and the Arabidopsis ubiquitin (UBQ1) promoters. Neither promoter was as effective as CaMV 35S in expressing phytochrome in vascular tissue or in inducing the dwarf phenotype. Collectively, these data indicate that the spatial distribution of ectopic phytochrome is important in eliciting the dwarf response and suggest that the phenotype is invoked by elevated levels of the far-red-absorbing form of phytochrome within vascular tissue repressing GA biosynthesis.  相似文献

Geranyl diphosphate synthase (GPS) is generally considered to be responsible for the biosynthesis of monoterpene precursors only. However, reduction of LeGPS expression in tomato (Lycopersicon esculentum) by virus-induced gene silencing resulted in severely dwarfed plants. Further analysis of these dwarfed plants revealed a decreased gibberellin content, whereas carotenoid and chlorophyll levels were unaltered. Accordingly, the phenotype could be rescued by application of gibberellic acid. The dwarfed phenotype was also obtained in Arabidopsis thaliana plants transformed with RNAi constructs of AtGPS. These results link geranyl diphosphate (GPP) to the gibberellin biosynthesis pathway. They also demand a re-evaluation of the role of GPS in precursor synthesis for other di-, tri-, tetra- and/or polyterpenes and their derivatives.  相似文献

Mutations (Rht genes) blocking sensitivity to gibberellic acid (GA) were used to examine phytohormone mediated cell wall expansion in wheat (Triticum aestivum L.). Irreversible extensibility of immature leaf segments, as determined by stress/strain (instron) measurements, declined with Rht gene dose. Exogenous GA3 significantly increased wall extensibility in the nonmutant controls but had no effect on the near-isogenic GA-insensitive genotypes. Furthermore, ancymidol, an inhibitor of gibberellin biosynthesis, diminished wall extensibility in the nonmutant control. Extensibility of immature segments was highly correlated with mature leaf sheath length (R = +0.95). The results indicate that wall yielding properties of expanding wheat leaves are associated with leaf cell expansion potential and that GA is involved in the determination of those properties.  相似文献

The plant growth retardant alpha-cyclopropyl-alpha-(4-methyoxyphenyl)-5-pyrimidine methyl alcohol (ancymidol) and a series of analogs of this substance in which one or more of the substituents were varied were tested for their comparative biological activity. The compounds were tested as inhibitors of internode elongation in peas and as inhibitors of the oxidation of ent-kaurene catalyzed by microsomal preparations from the liquid endosperm of Marah macrocarpus seeds. The relative effectiveness of a substance was generally the same as an inhibitor of the two processes. Ancymidol was the most effective. Substitution of the alcohol group of ancymidol by either methoxy or hydrogen groups reduced the activity only slightly. Substitution of the cyclopropyl group by an isopropyl moiety also had little effect on the activity. However, substitution of the cyclopropyl group with a phenyl or other aryl substituent greatly reduced the effectiveness of the analog as an inhibitor. Replacement of the 4-methoxyphenyl substituent with a similar substituent such as 4-chlorophenyl had little effect on activity, but replacement with a 2-methoxyphenyl group greatly reduced activity. Analogs in which the pyrimidyl moiety of ancymidol was modified were inactive in whole plants, but moderately active in the cell-free ent-kaurene oxidation system. The application of gibberellic acid can overcome the growth inhibitions due to treatment of the test plants with 10(-5)m or lower concentrations of the inhibitors. However, the inhibitory effects of 10(-4)m or higher concentrations of inhibitors on test plants were not overcome by the applications of exogenous gibberellic acid. These results support the idea that the effects of low concentrations of these substances on plant growth are primarily a consequence of their ability to inhibit ent-kaurene oxidation and gibberellin biosynthesis. Other modes of inhibition may operate at higher inhibitor concentrations.  相似文献

We studied how mitochondria affect ethylene response via modulation of CTR1 expression in cytoplasmic male-sterile (CMS) Brassica juncea. The expression of CTR1 gene was reduced in CMS compared with male-fertile (MF) lines. We observed that hypocotyl and root lengths were shorter than in the MF line during germination in the dark. An enhanced ethylene response was observed in CMS plants as shown by the CMS and maintainer line phenotypes treated with 1-aminocyclopropane-1-carboxylic acid. The phenotype in CMS plants could be recovered to the maintainer line when treated with Ag(+) . One ethylene response gene, plant defensin gene, was detected to be induced in CMS. The behavior of this phenotype could be mimicked by treating the maintainer line with antimycin A that disturbs mitochondrial function, which showed reduced length of hypocotyl and roots, and resulted in similar expression patterns of ethylene-related genes as in CMS. The reduced length of hypocotyl and roots could be recovered to the maintainer line by treatment with gibberellic acid (GA(3) ). In addition, the GA(3) content was reduced in CMS plants and in the MF line treated with antimycin A. Ethylene treatment markedly affects GA(3) content; however, GA(3) did not significantly affect ethylene-related gene expression in regards to regulation of hypocotyl and root length, which suggests that ethylene acts upstream via gibberellin to regulate hypocotyls and root development. Taken together, our results suggest a link between mitochondrial modulation of the ethylene and gibberellin pathway that regulates the development of hypocotyl and roots.  相似文献

Juvenile trees of temperate and boreal regions cease growth and set buds in autumn in response to short day-lengths (SD) detected by phytochrome. Growth cessation and bud set are prerequisites for the development of winter dormancy and full cold hardiness. In this study we show that the SD-requirement for bud set and cold hardening can be overcome in hybrid aspen (Populus tremula L. × tremuloides Michx.) by low night temperature and inhibition of gibberellin (GA) biosynthesis. Bud set and increased cold hardiness were observed under normally non-inductive long day-length (LD) in wild-type plants, when exposed to low night temperature and paclobutrazol. In addition, the effect of PHYA overexpression could be overcome in transgenic plants, producing bud set and cold acclimation by treatment with: SD, low night temperature and paclobutrazol. After cold acclimation, the degree of bud dormancy was lower for wild-type plants prior treated with LD and transgenic plants (overexpressing PHYA), than SD-treated, wild-type plants. Thus, low night temperature in combination with reduced GA content induced bud set and promoted cold hardiness under normally non-inductive photoperiods in hybrid aspen, but was unable to affect development of dormancy. This might suggest separate signalling pathways from phytochrome regulating the induction of cold/cold hardiness and bud dormancy in hybrid aspen or alternatively, there might be one pathway that fails to complete its action in the transgenic and paclobutrazol treated plants.  相似文献

The relatively rapid and extensive corolla expansion in Ipomoea nil (L.) Roth cv. ‘Scarlett O'Hara’ appears to be initiated by a shift in the balance between a growth promoter (gibberellin, GA) and a growth inhibitor (ethylene) two days before anthesis. The effects of applied growth regulators in vitro, were measured as a change in area of segments from whole young corollas 16–17 mm long. Applied gibberellic acid (GA3) strongly promoted growth, while a GA action inhibitor (ancymidol) reduced growth. Inhibitors of GA biosynthesis (AMO-1618, paclobutrazol, tetcyclasis, and chlorocholine chloride) had little effect, implying that the GAs were not being synthesized within the corolla segments, at least at this stage of corolla growth. Both the level of endogenous GAs and response of the segments to sucrose increased as the corolla size also increased from 15 to 20 mm in length. Applied 1-aminocyclopropane-l-carboxylic acid (ACC), an ethylene precursor, inhibited the response of corollas to applied GA3. Applications of ethylene biosynthesis inhibitors (AVG and cobalt ions) promoted growth of the corolla segments. Rapid ethylene production by corollas 15–17 mm long (48 hr before flower opening) appeared to inhibit the growth that may have been induced by low levels of endogenous GAs. Older corollas (longer than 18 mm) had very low levels of ethylene production and much higher levels of GA-like substances; these changes apparently allow the corollas to begin to expand in vivo and to strongly respond to sucrose applications alone in vitro. Acid-induced growth does not seem to be an important component of corolla expansion; applied fusicoccin (a proton efflux promoter) and sodium orthovanadate (a proton efflux inhibitor) had no significant effect on growth of the 16–17 mm long corollas. Buffers (Good's and phosphate) over a wide pH range had no effect on corolla expansion. Taken together, our results indicate that the regulation of corolla expansion depends (at least in part) on compensatory shifts in the levels of two plant growth regulators [PGRs], ethylene and GA.  相似文献

The procera mutant of tomato (Lycopersicon esculentum L.) has a phenotype which is remarkably similar to that of normal tomatoes treated with exogenous gibberellin (GA), indicating that it might be a GA over-producer. However, analysis of endogenous GAs by gas chromatography-mass spectrometry showed that Procera actually has lower levels of GA20 and GA1 than normal. The reason for these anomalously low GA levels is not clear, as there was no difference between procera and normal plants in their ability to metabolize [3H]GA20. The procera mutant responded to exogenous gibberellic acid with increased extension growth, but the proportional response for a given dose of GA was the same in procera and normal plants. Bulbing was studied in shallot plants cultured in vitro. Bulbing occurred under a 16 h photoperiod with fluorescent + incandescent light and 30-50 g 1(-1) sucrose in the culture medium. Exogenous gibberellin (10 microM GA3) inhibited leaf and root growth and bulbing. When added to the medium at a concentration of 10 microM, three inhibitors of gibberellin biosynthesis (ancymidol, flurprimidol and paclobutrazol) promoted bulb formation and the percentage of bulbing. When ancymidol was used in combination with GA3, it did not reverse the effect of GA3 applied alone. Under treatments with 30-70 g l(-1) sucrose, bulbing ratios greater than those found in control plants were achieved by addition of ancymidol, and bulb fresh weight was increased in the same way. Ancymidol caused a 66% decrease in sucrose content in leaf bases but greatly increased the glucose, fructose and fructan contents. The increase in fructan content by ancymidol could result from the three-fold rise in total [14C]sucrose uptake per plant from the culture medium associated with a marked increase in leaf base labelling at the expense of root labelling. The possible role of ancymidol is discussed and evidence supports a major regulatory role for gibberellins in bulbing.  相似文献

In wild-type (WT) Columbia and Landsberg erecta ecotypes of Arabidopsis thaliana (L.) Heynh., trichomes are present on the adaxial surfaces of all rosette leaves but are absent from the abaxial surfaces of the first-formed leaves. We have determined that both long-day (LD) photoperiod and gibberellin (GA) stimulate trichome formation. WT plants grown in LD conditions produce the first abaxial trichome on earlier leaves than plants grown in short-day (SD) conditions. Photoperiod sensitivity of abaxial trichome formation on WT plants develops gradually over time, reaching the maximum sensitivity about 24 d after germination. Application of gibberellic acid to WT plants growing in SD conditions accelerates the onset of abaxial trichomes. Conversely, application of 20 to 80 mg L-1 paclobutrazol, a GA biosynthesis inhibitor, to wild-type plants suppresses trichome initiation on the abaxial epidermis. The GA-deficient mutants ga1-5 and ga4-1 and the GA-insensitive mutant gai-1 exhibit delayed onset of abaxial trichomes when grown in LD conditions. The null mutant ga1-3 produces completely glabrous leaves when grown in SD conditions. Application of gibberellic acid to glabrous ga1-3 plants consistently induces earlier formation of trichomes on the adaxial epidermis than on the abaxial epidermis, demonstrating a difference between the adaxial and abaxial surfaces in their response to GA with regard to trichome formation.  相似文献

The germination of Amaranthus paniculatus seeds was inhibited by applying paclobutrazol, a specific inhibitor of gibberellin biosynthesis. This inhibition was markedly counteracted by gibberellin A3 (GA3), suggesting that endogenous gibberellins are required for germination in this species. The inhibitory effect of paclobutrazol was also overcome by ethephon (2-chloroethylphosphonic acid) or the precursor of ethylene biosynthesis, ACC (1-aminocyclopropane-l-carboxylic acid). Thus the physiological effect of gibberellin can be mimicked by ethylene released from ethephon or synthesised from exogenous ACC. The biosynthesis of heme, a plant tetrapyrrole, was studied in the leaves of a chlorophyll-deficient plastome mutant of the sunflower (Helianthus annuus L, line 2-24, albina form). In the light, the content of 5-aminolevulinic acid (ALA) in white mutant leaves was, on the average, ten times less than in that of the wild-type form (line 3629). Chlorophyll content in mutant leaves comprised only 0.3% of that of control plants. The activities of Fe-chelatase and ALA dehydratase in the heme synthesis were either comparable to or even higher than those in the wild-type leaves. A normal respiration rate in white mutant leaves, the equal content of phytochrome apoproteins in plants of both types, and the lack of noticeable morphogenetic differences realized through the phytochrome system can indicate that mutant and wild-type leaves are similar in their levels of phytochrome and the cytochromes of mitochondrial respiration. Nevertheless, in the mutant, the content of heme noncovalently bound by apoproteins amounted to only one third of its content in the wild-type plants. It seems that a dramatic decrease in the capability of white leaves for chlorophyll biosynthesis and for the formation of the photosynthetic apparatus is responsible for a low demand for chloroplast cytochromes, which is the major cause of a reduced heme content in the mutant.  相似文献

Summary In henbit (Lamium amplexicaule L.) seeds, micromolar levels of gibberellic acid (GA3) introduced at the onset of imbibition markedly stimulated germination. Sensitivity to GA3 stimulation is rapidly lost if the seeds are pretreated in darkness at 15° for 1 day or more on a water substrate. Loss in responsiveness to GA3 is hastened by pretreatment in far-red or by dark pretreatment at higher (25°) temperatures and is reduced by pretreatments at 5°.
