黑曲霉固态发酵生产单宁酶的条件优化   总被引:1,自引:0,他引:1  
研究采用响应面法优化黑曲霉固态发酵生产单宁酶的培养条件。应用Plackett—Burman试验筛选出重要影响因子:五倍子粉含量、(NH4)2SO4浓度以及接种孢子量,最陡爬坡试验逼近最大响应区域。应用Box.Behnken响应面试验对重要影响因子进一步优化。得到最佳培养条件:每250mL三角瓶中装入1.0g五倍子粉、4.4g稻壳和0.5g麸皮、液固比(mL/g)2:1且营养盐溶液组成为(NH4)2s0421g/L、MgSO4·7H2O1g/L、NaCl1g/L,培养基pH自然,接种5.7×10^7个孢子后在30℃温度下培养4d。在此条件下,单宁酶产量从40U/g提高到114U/g,3次重复验证性试验平均值为115U/g,验证了模型的可靠性。  相似文献

利用一株分离自传统发酵酸马奶中的益生干酪乳杆菌(Lactobacillus casei Zhang)进行固态发酵(Solid State Fermentation,SSF)。以发酵物中的活菌数为主要指标,采用九因素四水平(L32(4^9))的正交试验优化固态发酵培养基,并在优化的培养基基础上研究不同的初始含水量及培养时间对Lactobacillus casei Zhang活菌数的影响。实验结果表明,在固态发酵培养基组成为4g豆粕、5g麸皮、0.6g乳清粉、0.3g葡萄糖、0.3g碳酸钙、0.02g硫酸铵、0.01g硫酸镁,初始含水量为55%的优化条件下,37℃发酵60h,发酵物中Lactobacillus casei Zhang活菌数可达到4.08×10^10CFU/g。  相似文献

建立了一套两升圆柱体玻璃发酵罐的固态通风培养装置及通风系统。以球孢白僵菌Bb371为生产菌,培养基220g(82%麸皮 4.5%黄豆粉 4.5%玉米粉 9%稻壳)作为生产原料,接种30ml,23℃培养温度下,分阶段调节通风量和空气湿度,培养5d,产孢量可达到116亿/g干培养基。  相似文献

从中原油田污染土壤中通过实验室驯化培养分离到一组能以中原原油为碳源的快速生长的石油烃降解菌.用该组降解菌接种原油污染土壤,研究其原位生物联合修复实验,接种降解菌的各区分别种植大豆、施有机肥料、施有机肥料和锯末,与空白试样作对比.经过120d的联合修复,各区石油降解菌的总数(lgcfu/g)由接种时的5.25分别变为7.79、4.96、5.15、4.67.石油烃降解率分别达到89.4%、72.5%、76.7%、49.2%.表明分离的该组石油烃降解菌是一组高效降解菌且其与植物联合修复石油污染土壤能显著提高修复效果.  相似文献

两株绿脓杆菌对石油污染土壤的修复作用   总被引:2,自引:0,他引:2  
本文旨在研究环境条件下微生物对石油污染土壤的修复情况。从矿井周边土样定向筛选出两株绿脓杆菌,摇瓶降解实验发现,两菌混合培养10 d原油降解率达到95.67%,比单菌培养提高至少32%,即两菌对原油降解具有协同作用。根据降解实验结果制备了混合修复菌剂,并且人工构建石油污染场地,展开中试场地修复试验,模拟不同的操作条件下土壤中原油的降解情况。经60 d修复发现,添加了菌剂的场地,石油烃含量下降趋势明显,每克土壤中石油烃含量从初始的0.8%降至0.1%–0.3%,其中额外添加有机肥作为补充碳氮源的场地,总石油烃降解率最高,达到85.28%。而未添加菌剂的对照组石油烃含量仅减少25.85%。  相似文献

研究了根霉12号固体发酵产生纤溶酶的工艺条件。采用单因素试验、均匀设计方法对固体发酵培养基的碳源、氮源、碳氮比、初始pH、加水量、无机盐加量进行了优化;采用正交试验对发酵时间、接种量进行了研究。结果表明,实验范围内根霉12固体发酵产纤溶酶的适宜培养基组成为:麸皮:豆粕=1:2,初始pH5.0,加水量0.75ml/g物料,MnSO4-H2O 和 (NH4)2SO4加量分别为0.25%和1.42%(对物料)。适宜培养条件为接种量10^7个孢子/g 物料,培养时间 72h。优化条件下的纤溶酶产量平均达 791.81u/g物料。  相似文献

【目的】研究内生真菌菌株YCEF005、YCEF053固态发酵培养基。【方法】通过单因素试验和正交试验设计优化固态发酵培养基基质组成;通过单因素试验和均匀试验设计及神经网络结合遗传算法优化基质外加组分和接种量。【结果】优化的YCEF005培养基基质组成(质量比)为麸皮50%、豆饼粉10%、米糠20%、玉米碎粒20%;YCEF053培养基基质组成为麸皮60%、豆饼粉10%、米糠10%、玉米碎粒20%。YCEF005基质外加组分和接种量(/kg基质)为蔗糖12.96 g、蛋白胨12.70 g、NH4NO3 4.00 g、KH2PO4 1.50 g、Ca SO4 10.00 g、Mg SO4 0.48 g、含水量500 g、接种量24 m L。YCEF053基质外加组分和接种量(/kg基质)为蔗糖13.37 g、蛋白胨14.02 g、Na NO33.85 g、KH2PO4 1.23 g、Ca SO4 10.89 g、Mg SO4 0.52 g、含水量480 g、接种量20 m L。在优化培养基上发酵7-9 d获得最大发酵生物产量。【结论】通过对固态发酵培养基的优化提高了发酵的生物产量。  相似文献

高效石油降解菌的筛选及其降解性能研究   总被引:4,自引:0,他引:4  
从长期被石油污染的土壤中驯化筛选、分离出2株高效石油降解菌Y-7和Y-9,通过形态学特征的观察和生理生化试验对其进行初步鉴定,鉴定结果分别为假单胞菌属(Pseudomonas sp.)和芽孢杆菌属(Bacillus sp.)。同时,研究并分析了不同pH、温度、初始石油浓度、接种量、吐温80等条件对菌体生长和石油降解率的影响。结果表明,在试验条件下,2株优势菌在初始pH为7左右,对石油的降解率可分别高达68.7%,74.5%,偏酸或偏碱的环境均不利于菌体的生长;培养温度对2株菌体生长和石油降解率的影响较大,最佳温度为35℃,降解率达到最大,分别为73.1%和69.6%;石油初始浓度大于0.4g/L时,Y-7降油率从69%降到49%,Y-9基本变化不大,控制石油物质浓度在0.4g/L,有利于对石油的生物处理;最佳接种量为2mL/L;吐温80对石油的降解促进作用有待进一步分析与研究。  相似文献

利用Bello Cell这种新型的生物反应器,来培养COS7细胞和昆虫Sf9细胞。COS7细胞和昆虫sf9细胞分别用T75培养瓶及spinnerflask培养之后,经细胞计数接入Bello Cell之中,同时检测并分析培养基中葡萄糖、谷氨酰胺、乳酸和氨浓度变化情况。COS7细胞初始接种量为4.208×10^7cells,最终在培养156h后细胞数量达到了4.68×10^8cells,是初始细胞量的11倍。sf9细胞初始接种细胞量为1×10^8cells,在培养192h时,细胞总量达到了最高为4.01×10^9cells,是最初细胞量的40倍。培养基的代谢物进行有规律的变化。Bello Cell适合COS7细胞和昆虫Sf9细胞高密度大规模培养,为动物细胞高效大规模表达药物蛋白,奠定重要的基础。  相似文献

Among about 200 Rhizopus strains isolated in Thailand, Rhizopus sp. MB46 was selected as a producer of raw cassava starch-digestive glucoamylase. Rice bran was effective for the enzyme production in a solid culture as well as wheat bran. Addition of turpentine oil into the rice bran solid culture increased the productivity. Rhizopus sp. MB46 was found to produce glucoamylase in a liquid culture containing 1% rice bran but not in one consisting of 10% raw cassava starch of 2% glucose. The productivity per 1 g solids in the medium in liquid culture was finally improved 6-times by utilization of n-hexane-treated rice bran, supplement of 0.1% meat extract and addition of gauze as a support. The activity was superior to that in turpentine oil-supplemented solid culture.  相似文献

In this study, Bacillus sphaericus NRC 69 was grown in culture media, in which 12 agricultural wastes were tested as the main carbon, nitrogen and energy sources under solid state fermentation. Of the 12 tested agricultural by-products, wheat bran was the most efficient substrate for the production of B. sphaericus mosquitocidal toxins against larvae of Culex pipiens (LC50 1.2 ppm). Mixtures of tested agricultural wastes separately with wheat bran enhanced the produced toxicity several folds and decreased LC50 between 3.7- and 50-fold in comparison with that of agricultural wastes without mixing. The toxicity of B. sphaericus grown in wheat bran/rice hull at 8/2 (g/g) and wheat bran/barley straw at 1/4 (g/g) showed the same toxicity as that in wheat bran medium (LC50 decreased 17- and 16-fold, in comparison with that in rice hull or barely straw media, respectively). In wheat bran medium, the maximum toxicity of the tested organism obtained at 50% moisture content, inoculum size 84 × 106 CFU/g wheat bran and incubation for 6 days at 30°C. Addition of cheese whey permeate at 10% to wheat bran medium enhanced the toxicity of B. sphaericus NRC 69 about 46%.  相似文献

Five microorganisms, three bacteria and two yeasts, capable of degrading Tapis light crude oil were isolated from oil-contaminated soil in Bangkok, Thailand. Soil enrichment culture was done by inoculating the soil in mineral salt medium with 0.5% v/v Tapis crude oil as the sole carbon source. Crude oil biodegradation was measured by gas chromatography method. Five strains of pure microorganisms with petroleum degrading ability were isolated: three were bacteria and the other two were yeasts. Candida tropicalis strains 7Y and 15Y were identified as efficient oil degraders. Strain 15Y was more efficient, it was able to reduce 87.3% of the total petroleum or 99.6% of n-alkanes within the 7-day incubation period at room temperature of 25 ± 2 °C.  相似文献

A search for oxidases that catalyze the oxidation of oligosaccharides has resulted in the isolation of several soil-derived fungus strains which produced novel oligosaccharide oxidases with different substrate specificity on wheat bran solid culture. One of these oxidases produced by Acremonium strictum T1 strain has been characterized. This enzyme showed high reactivity toward maltose, lactose, cellobiose and maltooligosaccharides composed of up to seven glucose units, and was named as glucooligosaccharide oxidase based on its substrate specificity. Strain T1 was subjected to a strain improvement program, and an enzyme hyper-producing mutant strain T1-38 was selected. This mutant strain produced glucooligosaccharide oxidase 75 times higher than the wild type strain T1. When cultivated in a solid medium comprised of 1 part of wheat bran and 1 part of water (w/w), enzyme activity reached a maximum level of 6 units per g of culture medium after 4 days cultivation. Characteristics of the enzyme including the substrate specificity were compared with two other novel oligosaccharide oxidases isolated in this laboratory. Batch type conversion of lactose to lactobionic acid using crude enzyme was also discussed.  相似文献

Mutant strains of Aspergillus sojae exhibited coordinate increases of acid proteinase, α-amylase, and cellulase and a decrease of pectin trans-eliminase accompanied with the hyperproduction of alkaline proteinase in wheat bran koji culture. The production of these enzymes in the wheat bran solid medium, liquid wheat bran-defatted soybean medium, and liquid glucose-peptone medium were surveyed. The analyses on the production patterns of these enzymes under the different cultural conditions suggest that mutation in these mutants producing elevated levels of the above enzymes is due to a more complex alteration than a single gene mutation.  相似文献

为了获得生产用廉价型灵芝发酵培养基,采用中心组合旋转设计法和响应曲面法对低成本培养基组分进行了优化。优化的四个组分为玉米粉(x1)、麸皮粉(x2)、豆饼粉(x3)和蔗糖(x4)。结果表明,灵芝菌体发酵和多糖发酵的培养基预测模型分别为:Y1=15.1–0.31x1–0.34x2+0.36x3–0.44x4–1.26x12–1.98x22–0.85x32–1.15x42–0.59x2x3和Y2=2.0–0.08x1–0.08x2+0.04x3–0.09x4–1.13x12–0.33x22–0.08x32–0.16x42–0.16x2x3–0.10x1x4。从中获得菌体发酵的最优配方为:玉米粉19.7g/L,麸皮粉11.3g/L,豆饼粉6.3g/L,蔗糖19.5g/L;多糖发酵的最优配方为:玉米粉19.6g/L,麸皮粉11.0g/L,豆饼粉6.7g/L,蔗糖19.1g/L。150L发酵罐中试放大结果表明,灵芝菌体的产量为16.92g/L,多糖产量为1.86g/L。所得培养基为灵芝产品的高效低成本生产提供了基础。  相似文献

In a locally isolated Rhizopus oryzae strain highest-production of protease (388.54/g wheat bran) was observed in presence of Tween-80 and dioctyl sodium sulfosuccinate individually at 40mg/g wheat bran concentration. Under solid state fermentation biotin (0.0025mg/g wheat bran); Ca2+ (0.05mg/g wheat bran) and 1-Naphthyl acetic acid (0.01mg/g wheat bran) also showed some inducing effect on the synthesis of the enzyme protease by solid state fermentation.  相似文献

Low cost fermentation media using agricultural by-products (wheat bran extract, rice bran extract and soybean meal extract) as a major nutrient source, were evaluated for the production of tyrosinase from the fungus Auricularia auricula in submerged culture. In single-factor experiments, three components (wheat bran extract, casein and CuSO4) were chosen to further optimize medium composition using response surface methodology (RSM). The central composite experimental results showed the following optimum medium composition: wheat bran extract 36.0 %, casein 1.1 g/l and CuSO4 0.13 g/l. Under these conditions, the highest tyrosinase activity was 17.22 U/ml, which was 2.1 fold higher than that obtained using the non-optimized medium. The present study is the first to report the statistical optimization of medium composition for production of tyrosinase by A. auricula using cheaper wheat bran extract as a major nutrient source. These results might provide a reference for the development of a cost-effective medium for commercial production of tyrosinase.  相似文献
