Beta-Lactamase Database (BLDB) is a comprehensive, manually curated public resource providing up-to-date structural and functional information focused on this superfamily of enzymes with a great impact on antibiotic resistance. All the enzymes reported and characterised in the literature are presented according to the class (A, B, C and D), family and subfamily to which they belong. All three-dimensional structures of β-lactamases present in the Protein Data Bank are also shown. The characterisation of representative mutants and hydrolytic profiles (kinetics) completes the picture and altogether these four elements constitute the essential foundation for a better understanding of the structure-function relationship within this enzymes family. BLDB can be queried using different protein- and nucleotide-based BLAST searches, which represents a key feature of particular importance in the context of the surveillance of the evolution of the antibiotic resistance. BLDB is available online at http://bldb.eu without any registration and supports all modern browsers.  相似文献

Summary The solid-state conformation of the first N-protected ornithine derivative has been established by X-ray analysis. The hydrochloride of Nα-(9-fluorenyl)methoxycarbonyl-l-ornithine crystallises as diethyl ether solvate. The backbone (ω0 ϕ, ψ χ1) torsion angles are (174.9°,–84.0°, 145.9°,–171.0°). The conformation of the urethane amide bonds istrans. The ornithine aliphatic side chain adopts preferred fully extended conformation which is stabilised by the hydrogen bonding of the-NH 3 + group to the diethyl ether molecule, carboxyl group and Cl anions.  相似文献

The complex μ-3,3′-[1,2-ethanediyl-bis(nitrilome- thylidyne)-bis(2-hydroxybenzoato)] aquadicopper(II) hydrate, C18H16N2O8Cu2, was isolated from an attempted preparation of a copper lanthanum binuclear complex. The dark purple crystals are monoclinic, space group P21/n, with 4 molecules per unit cell; dimensions a = 13.961(5), b = tl.787(3), c = 11.622(3) Å and β = 113.09(2)°. The final R was 0.046 for the 2062 reflections used in the analysis. The Cu atom in the N2O2 cavity is five coordinate with CuN distances of 1.879 and 1.880 Å and CuO distances of 1.898 and 1.900 Å. A water molecule at 2.557 Å completes the square pyramidal arrangement. The second Cu in the O4 cavity is square planar, with CuO distances to the bridging oxygens of 1.914 and 1.909 Å and to the carboxy oxygens of 1.871 and 1.882 Å. A survey of copper complexes in a square planar N2O2 arrangement has led to the equation δCu from the N2O2 plane = 0.822 – 0.275 (CuO axial distance) with a correlation coefficient of 0.98 for the 12 structures in which the Cu atom is bonded to a fifth oxygen atom. A model for the transition from square planar to square pyramidal geometry is proposed.  相似文献

(R)-(+)- and (S)-(?)-dihydrozeatins [(R)-(+)- and (S)-(?)-6-(4-hydroxy-3-methylbutylamino)purines, 1a and 1b] and their ribosides {(?)-6-[(R)-4-hydroxy-3-methylbutylamino]- and (?)-6-[(S)-4-hydroxy-3-methyl-butylamino]-9-β-D-ribofuranosylpurines, 3a and 3b} were synthesized and tested for their cytokinin activity by four bioassay systems, the growth of tobacco callus, the seed germination of lettuce, the fr. wt increase of excised radish cotyledons and the retardation of chlorophyll degradation in radish cotyledons. In tobacco callus bioassay, 1a was more active than 1b. The ribosides 3a and 3b were not less active than their corresponding aglycones 1a and 1b. In other bioassays used the activity followed the order: 1a >3a >1b >3b. In tobacco callus bioassay and lettuce seed germination, trans-zeatin [6-(4-hydroxy-3-methylbut-trans-2-enylamino)purine] showed stronger cytokinin activity than 1a.  相似文献

A series of 1-acyl-3,5-bis(benzylidene)-4-piperidones 37 were designed and synthesized as novel cytotoxic agents. These compounds displayed potent cytotoxic properties towards human Molt4/C8, CEM, HSC-2, HSC-3 and HSC-4 neoplasms and also to murine L1210 cells. The majority of the compounds have sub-micromolar or very low micromolar IC50 and CC50 values and are significantly more potent than the reference alkylating drug melphalan. Evaluation of these compounds against non-malignant HGF and HPLF cells revealed the tumour-specific toxicity. In particular, 3e emerged as a promising lead cytotoxic agent which caused apoptosis and PARP1 cleavage in HSC-2 cells.  相似文献

The crystal structure of a regenerated form of (1→3)-α-d-glucan, obtained by solid state deacetylation of the triacetate derivative, has been determined by combined X-ray diffraction analysis and stereochemical model refinement. The structure crystallizes in an orthorhombic unit cell with parameters a = 16.46 A?, b = 9.55 A? and c (fibre repeat)=8.44 Å, and space group P212121. The chain conformation is nearly completely extended and is very close to a 2/1 helix, even though the dimer residue is the crystallographic repeat unit. An intramolecular O(2)  O(4)′ hydrogen bond stabilizes the conformation and extensive intermolecular hydrogen-bonding abilizes the packing. The resulting structure is sheet-like, with an alternating polarity of chain directions within the sheet. In its sheet-like character, extensive hydrogen-bonding, and insolubility in water, this polymorph of (1→3)-α-d-glucan resembles regenerated cellulose. The reliability of the structure analysis is indicated by the X-ray residual R=0.206.  相似文献

(±)-Licarin A (1) was obtained by oxidative coupling, and its enantiomers, (?)-licarin A (2) and (+)-licarin A (3), were resolved by chiral HPLC. Schistosomicidal and trypanocidal activities of these compounds were evaluated in vitro against Schistosoma mansoni adult worms and trypomastigote forms of Trypanosoma cruzi. The racemic mixture (1) displayed significant schistosomicidal activity with an LC50 value of 53.57 μM and moderate trypanocidal activity with an IC50 value of 127.17 μM. On the other hand, the (?)-enantiomer (2), displaying a LC50 value of 91.71 μM, was more active against S. mansoni than the (+)-enantiomer (3), which did not show activity. For the trypanocidal assay, enantiomer 2 showed more significant activity (IC50 of 23.46 μM) than enantiomer 3, which showed an IC50 value of 87.73 μM. Therefore, these results suggest that (±)-licarin A (1) and (?)-licarin A (2) are promising compounds that could be used for the development of schistosomicidal and trypanocidal agents.  相似文献

Early studies led to the identification of 3β-(4-methoxyphenyl)tropane-2β-carboxylic acid methyl ester (5) with high affinity at the DAT (IC50 = 6.5 nM) and 5-HTT (Ki = 4.3 nM), while having much less affinity at the NET (Ki = 1110 nM). In the present study, we replaced the 4′-methoxy group of the 3β-phenyl ring with a bioisosteric 4′-methylthio group to give 7a. We also synthesized a number of 3β-(4-alkylthiophenyl)tropanes 7be, 3β-(4-methylsulfinylphenyl) and 3β-(4-methylsulfonylphenyl)tropane analogues 7fh as well as the 3β-(4-alkylthiophenyl)nortropane derivatives 811 to further characterize the structure–activity relationship of this type of compound for binding at monoamine transporters. With exception of the 4′-methylsulfonyl analogue 7h, all the tested compounds possessed high binding affinities at the 5-HTT. The Ki values ranged from 0.19 nM to 49 nM. The 3β-(4-methylthiophenyl)tropane 7a and its N-(3-fluoropropyl) analogue 9a and N-allyl analogue 10a are the most selective compounds for the 5-HTT over the NET (NET/5-HTT = 314–364) in the series. However, none of the compounds showed selectivity similar to 5 for both the DAT and 5-HTT relative to the NET. This study provided useful SAR information for rational design of potent and selective monoamine transporter inhibitors.  相似文献

A group of novel l-serinamides, substituted (S)-2-(benzylideneamino)-3-hydroxy-N-tetradecylpropanamides (3ao) and substituted (S)-2-(benzylamino)-3-hydroxy-N-tetradecyl propanamides (4c, 4i, 4l, and 4o), were synthesized as potential anti-tumor lead compounds. In vitro cell viability assay results indicate treatment with 3ao compounds resulted in significant inhibition of cell viability in the chemoresistant breast cancer cell line, MCF-7TN-R. Compounds 3i and 3l, both ortho-substituted analogs, show the greatest efficacy with IC50 values of 10.3 μM and 12.5 μM, respectively. The SAR analysis indicate that the imine functional group of 3ao is critical for the cellular anti-viability effect, and the partial atomic charge (PAC) value of imine C atom is a valuable structural parameter for predicting the activity of these ceramide analogs.  相似文献

Structure–activity relationships of 6-(benzoylamino)benzoxaborole analogs were investigated for the inhibition of TNF-α, IL-1β, and IL-6 from lipopolysaccharide stimulated peripheral blood mononuclear cells. Compound 1q showed potent activity against all three cytokines with IC50 values between 0.19 and 0.50 μM, inhibited LPS-induced TNF-α and IL-6 elevation in mice and improved collagen-induced arthritis in mice. Compound 1q (AN4161) is considered to be a promising lead for novel anti-inflammatory agent with an excellent pharmacokinetic profile.  相似文献

This work analyzes the spatial heterogeneity of consanguinity in the Basque province of Guipúzcoa (Spain), using data provided by Catholic dispensations (1862-1980). Secular trends in consanguinity rates (%M(C)) and mean inbreeding coefficient (F) were similar in the seven Guipúzcoan regions considered, with peaks between 1881-1920 and subsequently a gradual decline. Substantial differences in consanguinity characteristics emerged when the regions were classified according to level of urbanization. Principal component analysis (accounting for more than 85% of the total variance in consanguinity variables) clearly discriminated between urbanized and less urbanized regions. The latter stand out for their high consanguinity rates (3.57-6.73%), mean inbreeding coefficient (0.00112-0.00240), and M22/M33 ratio (M22, first cousins; M33, second cousins), which ranged between 0.89-1.48. Moreover, in less urbanized regions, marital consanguinity was eminently local, and mainly conditioned by regional endogamy (71.4-85.0%). By contrast, urban subpopulations showed the lowest consanguinity rates (1.60-1.96%) and mean inbreeding coefficient (around 0.0007). In these regions, the M22/M33 ratio also exhibited high values (1.07-1.56), but this time at the expense of the contribution of the immigrant group. Discussion of the factors that could have modeled this spatial variation in consanguinity centers on: 1) demographic aspects related to the chronology and intensity of industrialization, 2) the geography of the territory and the geography of peopling, and 3) linguistic differences, expressed in the uneven distribution of Basque-speakers among the different territories considered.  相似文献

The preparations and properties of tris(dipivaloylmethanato)technetium(III), tris(trifluoroacetylacetonato)technetium(III), and tris(hexafluoroacetonato)technetium(III) are described. The oxidation of the dipivaloyl derivative to tris(dipivaloyl)technetium(IV) hexafluorophosphate was shown to take place readily. Voltammetric studies and magnetic resonance results on the new complexes are reported. The large shifts observed for the complexes seem to be due to a contact interaction.  相似文献

Summary The conformation of chymotryptic fragment C2 of bacteriorhodopsin (residues 1–71) was studied by 2D1H NMR. The fragment was solubilized in a mixture of chloroform/methanol (11), 0.1 M LiClO4. Most of the resonances in1H NMR spectra of fragment C2 were assigned using phase-sensitive DQF-COSY, TOCSY, and NOESY techniques. To simplify the assignment procedure for overlapping regions of NMR spectra, an analog of fragment C2 with leucines deuterated in -positions was used. Deuterium exchange rates for amide protons were measured in a series of TOCSY spectra. Two right-handed -helical regions Pro8-Lys30 and Lys41-Leu62 were identified on the basis of NOE connectivities and deuterium exchange rates. The N-terminal part of the fragment (Ala.2-Gly6) adopts the helical conformation stabilized by 3 hydrogen bonds.  相似文献

The aim of this work is to compare the composition and seasonality of the phytoplankton population in a natural oligotrophic lake (Lake Sanabria) and a mesotrophic reservoir (Valparaíso). Both ecosystems are located on the Tera river course (NW Spain), which runs along an area of ancient metamorphic and plutonic rocks. Some physical and chemical parameters, chlorophyll a and phytoplankton biovolume were studied from monthly samples collected at different depths during the periods 1987–1989 (Lake Sanabria) and 1991–1992 (Valparaíso). Phytoplankton biovolume and chlorophyll a concentration were about five times higher in Valparaíso than in Lake Sanabria. Species composition (and main phytoplankton groups) were different. Valparaíso was highly dominated by diatoms and Lake Sanabria by cryptophytes and small chlorophytes. In spite of the fact that both sites were nitrogen limited, heterocystous cyanophytes (Anabaena sp.) were detected only in Valparaíso. The relationships between phytoplankton structure and trophic level, hydrological conditions and nitrate content are discussed.  相似文献

1-(2-(2,5-Dimethoxyphenylthio)benzylidene)semicarbazide (2a) was discovered as a potent apoptosis inducer through our cell based HTS assay. SAR study led to the discovery of a more aqueous soluble analog (2-(2,5-dimethoxyphenylthio)-6-methoxybenzylideneamino)guanidine (5e) with EC50 value of 60 nM in the caspase activation assay and GI50 value of 62 nM in the growth inhibition assay in T47D cells. Compound 5e was found to be an inhibitor of tubulin polymerization and efficacious in a MX-1 breast tumor model.  相似文献

Mixed-metal thiocyanate complexes [MnHg(SCN)4(NOP)] (1) and [MnHg(SCN)4(DMSO)] (2) (NOP = 3-methyl-4-nitropyridine-N-oxide, DMSO = dimethylsulfoxide) have been synthesized and structurally characterized by single crystal X-ray analysis. Complex 1 and 2 both contain a [Mn22-O)2] lozenge, which is bridged to Hg(II) ions by end-to-end thiocyanate groups to form a 2-D and 3-D polymeric network, respectively. Magnetic studies indicate that both complexes are anti-ferromagnets, with 1 showing anti-ferrimagnetic ordering below 8.0 K.  相似文献

The one pot aqueous reaction of M(ClO4)2 (M = Cu2+ or Ni2+) with N-methylbis[2-(2-pyridylethyl)]amine (MeDEPA) and N,N′-dimethyl-N,N′-bis(2-pyridylmethyl)ethylenediamine (bpmen) and 1,4,7,10-tetraazacyclododecane (cyclen) in presence of sodium dicyanamide (Nadca) yielded dicyanamido-bridged polynuclear complex {[Cu(MeDEPA)(μ-1,5-dca)]ClO4}n (1), and two dinuclear complexes [Cu2(bpmen)2(μ-1,5-dca)]2(ClO4)5dca (2) and [Ni(cyclen)(μ-1,5-dca)]2(ClO4)2 (3). These complexes were characterized by IR and UV-Vis spectroscopy. Room temperature single-crystal X-ray studies have confirmed that the Cu(II) centers in 1 and 2 adopt geometries that are more close to trigonal bipyramidal (TBP) in 1 and close to square pyramidal (SP) in 2, whereas in 3, the Ni(II) centers are located in octahedral environment with doubly bridged μ-1,5-dca bonding mode. The intermolecular M···M distances in these complexes are in the range of 7.3-8.6 Å. Variable temperature magnetic susceptibility studies have confirmed that the dca-bridges mediate very weak antiferromagnetic interaction between the M(II) centers with J values of −0.35, −0.18 and −0.43 cm−1 for 1, 2 and 3, respectively. The results are compared and discussed in the light of other related bridged μ-1,5-dca Cu(II) and Ni(II) complexes.  相似文献

The 2:1 reaction of [Ru(H2O)2(NH3)5]2+ with 1,2,4,5-tetrazine (tz) gives rise to the formation of the dinuclear complex ion [{Ru(NH3)5}2(μ-tz-N1:N4)]4+. Its tetraphenylborate and hexafluoro-phosphate salts have been fully characterized; the X-ray structure of the former has also been determined.  相似文献
