Ethanolamine kinase was purified 60-fold by fractionation with ammonium sulfate, freeze-thawing, and gel filtration from a 100,000g supernatant from spinach leaf. The 100,00g supernatant preparation was stable for weeks, but the partially purified preparation lost half of the ethanolamine kinase activity in 10–14 days at 0–4 °C or ?20 °C. A molecular weight of 110,000 was estimated by gel filtration on Sephadex G-200. The reaction required ethanolamine (Km, 42 μm), MgATP (Km, 63 μm), and free magnesium ions. The enzyme was inhibited by MgATP, with an apparent Ki of 6.7 mm. Ethanolamine kinase was inhibited by calcium (in the presence of magnesium) and o-phenanthroline. EDTA above 0.9 mm inhibited the formation of phosphorylethanolamine and EGTA stimulated at low concentrations (0.4-0.9 mm) and inhibited at 1.8 mm. Ethanolamine kinase was inhibited by monomethylethanolamine and dimethylethanolamine, but not by choline (5 mm). Larvae of Anastrepha ludens and A. serpentina that developed in mango and sapodilla fruits, respectively, were exposed to Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae) and sequentially exposed as pupae to Coptera haywardi (Oglobin) (Hymenoptera: Diapriidae). Sequential exposure to both parasitoid species contributed to a decrease in fruit fly emergence due to higher levels of parasitism, which varied according to fruit type. In creole mango, D. longicaudata represented the highest percentage of parasitism. C. haywardi parasitism was greater in pupae from Ataulfo mangos and sapodilla, where the pulp size and volume may have acted as a refuge, allowing fly larvae to escape and leaving a greater number of unparasitised pupae available to C. haywardi. Similar results were obtained under field cage conditions, but the level of parasitism by C. haywardi was lower, suggesting that its effectiveness has some limitations under natural conditions. Our results suggest that both species can exert complementary parasitism, which represents an alternative worth to investigate under open field conditions.  相似文献

The gregarious endoparasitoid Tetrastichus giffardianus Silvestri (Hymenoptera: Eulophidae) is a natural enemy of fruit flies. This parasitoid was previously used to successfully control Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) in Hawaii, USA. Despite its importance in the control of fruit fly pests, little is known about the development or characteristics of its preimaginal stages. The aim of this study was to observe the development and morphologically characterize the immature stages of Tetrastichus giffardianus. Tetrastichus giffardianus individuals were reared on C. capitata larvae/pupae under laboratory conditions at a temperature of 25 ± 2 °C, relative humidity of 60 ± 10%, and 12-h photophase. Third-instar C. capitata larvae were exposed to parasitism for 24 h. After parasitism, the pupae were dissected every 24 h to evaluate the stage of development attained by T. giffardianus, and to record their morphological characteristics. A stereomicroscope was used to observe all the immature stages of T. giffardianus. The complete development of T. giffardianus under these conditions was completed within 14 days as follows: egg (duration ? 1 day); first (? 1 day), second (? 1 day), and third (? 2 days) larval instars; pre-pupa (? 2 days); and pupa (? 7 days). The immature stages of T. giffardianus differed sufficiently in their shape, color, and size to allow morphological characterization.  相似文献

A recent shift in managing insect resistance to genetically engineered (GE) maize consists of mixing non-GE seed with GE seed known as “refuge in a bag”, which increases the likelihood of predators encountering both prey fed Bt and prey fed non-Bt maize. We therefore conducted laboratory choice-test feeding studies to determine if a predator, Harmonia axyridis, shows any preference between prey fed Bt and non-Bt maize leaves. The prey species was Spodoptera frugiperda, which were fed Bt maize (MON-810), expressing the single Cry1Ab protein, or non-Bt maize. The predators were third instar larvae and female adults of H. axyridis. Individual predators were offered Bt and non-Bt fed prey larvae that had fed for 24, 48 or 72 h. Ten and 15 larvae of each prey type were offered to third instar and adult predators, respectively. Observations of arenas were conducted at 1, 2, 3, 6, 15 and 24 h after the start of the experiment to determine the number and type of prey eaten by each individual predator. Prey larvae that fed on non-Bt leaves were significantly larger than larvae fed Bt leaves. Both predator stages had eaten nearly all the prey by the end of the experiment. However, in all combinations of predator stage and prey age, the number of each prey type consumed did not differ significantly. ELISA measurements confirmed the presence of Cry1Ab in leaf tissue (23–33 µg/g dry weight) and S. frugiperda (2.1–2.2 µg/g), while mean concentrations in H. axyridis were very low (0.01–0.2 µg/g). These results confirm the predatory status of H. axyridis on S. frugiperda and that both H. axyridis adults and larvae show no preference between prey types. The lack of preference between Bt-fed and non-Bt-fed prey should act in favor of insect resistance management strategies using mixtures of GE and non-GE maize seed.  相似文献

Sublethal effects of an azadirachtin-based botanical insecticide (commercial product NeemAzal-T/S) on behaviour, life history traits and population growth of one commercial (Dutch strain, D) and two Serbian populations (Bujanovac, B; Negotin, N) of Encarsia formosa Gahan were evaluated in laboratory bioassays. In a two-choice test, parasitoids from all tested populations preferred to parasitise the untreated whitefly nymphs over those treated with 50, 25, 12.5, 6.25 and 3.12?mg/l of azadirachtin. All concentrations caused significant deterrent effects after 24?h, and the following deterrence indices were calculated: 9.7–60.9% (B), 5.5–57.5% (N) and 12.9–63.5% (D). The longevity of adults exposed for 48?h to residues of the botanical insecticide (applied at LC50) was shorter (1.5–1.7 days) than that of control wasps. Both daily and total parasitism were significantly reduced (by 41.7, 48.3 and 60.1% for N, D and B, respectively) compared to control, as well as adult emergence in the F1 generation. The instantaneous rate of increase (ri) of surviving adults was also significantly reduced (by 13.2, 19. 3 and 20.5%, for populations N, B, and D, respectively). Adults from all tested populations, emerged from pupae treated with the botanical insecticide applied at LC50, showed reduced parasitism, adult emergence, and ri levels, but the reduction was significant only for adult emergence of B and ri of B and N. Juvenile development of the parasitoid in treated pupae was significantly extended, compared to control. The implications of these results on integrated control of the greenhouse whitefly are discussed.  相似文献

"选择-表现"假说认为,成虫应该选择有利于子代发育的高品质寄主,但在寄主选择中,除了寄主品质外,其他因素也可能影响寄主选择决策。寄主选择研究通常以成虫为对象,而对那些初龄幼虫选择寄主的寄生性昆虫很少关注。以1龄幼虫积极搜寻寄主的寄生性花绒寄甲为模式生物,采用双选试验设计,观察了花绒寄甲初孵幼虫在不同体重青杨天牛幼虫之间、在已被寄生与健康的黄粉虫蛹之间的寄生选择性;然后采用回归设计,观察了花绒寄甲寄生若干不同体重的青杨天牛幼虫后的发育表现。研究结果表明,花绒寄甲1龄幼虫对体型较大的青杨天牛幼虫的选择偏好显著大于对体型较小的寄主幼虫的选择,选择大体型幼虫的比值比是选择小体型幼虫的4.55倍;对已被寄生的寄主黄粉虫蛹的选择偏好显著大于对健康寄主蛹的选择,选择已被寄生寄主的比值比是选择健康寄主的12.57倍。寄生青杨天牛幼虫的花绒寄甲幼虫发育历期平均为11.49 d、蛹历期为26.67 d、幼虫发育至成虫的羽化率50%,这些发育表现与寄生时青杨天牛幼虫的体重没有显著关系。但刚羽化寄甲成虫体重与寄生时寄主的体重存在显著的正直线关系:寄生时的寄主体重每增大0.01 g,羽化出的寄甲成虫体重增大近0.08%;方差分析寄甲成虫体重在不同寄主体重水平之间的差异表明,从体型较大寄主中羽化的寄甲成虫体重显著大于从体型较小寄主中羽化的成虫。研究结果说明,花绒寄甲初孵幼虫在寄主选择决策时,在寄主体型大小与被寄生状态之间可能采取折衷对策,而且对体型大小不同的寄主选择与子代发育适合度表现存在一致性,从而支持"选择-表现"假说。  相似文献

Interspecific competition between Xanthopimpla stemmator and Dentichasmias busseolae was studied using pupae of the invasive crambid stemborer Chilo partellus as the host. While X. stemmator is an old association, D. busseolae formed a relatively new association with C. partellus in East Africa. Two different time intervals between parasitism (0 and 48 h) and two parasitoid sequences [i.e., X. stemmator before D. busseolae (Xs–Db) and D. busseolae before X. stemmator (Db–Xs)] were chosen. In addition, the parasitoids’ performance on pupae in maize stems and ears was assessed. For both X. stemmator and D. busseolae, there was no difference in foraging time between unparasitized pupae and pupae previously parasitized by the other species, indicating that the two species were not capable of interspecific host discrimination. In the Xs–Db sequence, the time interval between parasitism did not have an influence on the percentage of pupae producing either parasitoid species. By contrast, in the Db–Xs sequence, the percentage of pupae producing X. stemmator was almost 8 times higher in the 0-h than the 48-h interval, while for D. busseolae it was the reverse. In the 0-h interval, X. stemmator outcompeted D. busseolae irrespective of whether it parasitized first or second, while in the 48-h interval, the parasitoid parasitizing first won. While D. busseolae successfully searched for and parasitized pupae in both stems and ears, parasitism of pupae in ears by X. stemmator was negligible. It was concluded that the two species could co-exist because they partly exploit different ecological niches.  相似文献

1. Variation in parasitism risk among hosts is a key factor influencing host–parasitoid interactions; however, within-patch variation (as opposed to between-patch variation) in parasitism risk has hardly been studied. This study investigated the mechanisms of within-patch variation in parasitism risk in the oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae). 2. Bactrocera dorsalis pupates underground. Previous studies have shown that pupae at greater depths have a reduced risk of parasitism by a pupal parasitoid Dirhinus giffardii (Silvestri) (Hymenoptera: Chalcididae). When a pupa near the ground surface is parasitised, the victim may decrease the risk of parasitism risk for pupae located at deeper depths. It was hypothesised that larvae will pupate at greater depths when they perceive the presence of conspecifics because of the benefit of pupating deeper than other individuals. 3. In a series of laboratory experiments, the effects of three factors on pupation depth were examined, including: (i) the density of larvae during development; (ii) the density of concurrently burrowing larvae; and (iii) the presence of pre-existing pupae in the pupation substrate. Only the density of concurrently burrowing larvae influenced pupation depth, which suggests that when a burrowing larva perceives the presence of other burrowing larvae, it aims to burrow deeper than them. 4. This study shows not only that parasitism risk is variable among pupae within a patch, but also that it is density-dependent. A commonly made assumption (i.e. the absence of within-patch variability in parasitism risk) needs to be re-evaluated in a wide range of host–parasitoid systems.  相似文献

The earwig Euborellia annulipes (Lucas) (Dermaptera: Anisolabididae), a generalist predator, has been observed in fruits infested with fruit fly larvae, which are frequently parasitized by parasitoid wasps. Neither the capacity of earwigs to predate on fruit flies nor intraguild interactions between earwigs and fruit fly parasitoids have been investigated. Here, we studied in laboratory conditions the predation on the fruit fly Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) by the earwig E. annulipes, and whether parasitism of fruit fly larvae by the parasitoid wasp Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae) influences predation by the earwig. We evaluated the predation capacity, functional response and prey preference of E. annulipes for parasitized and non-parasitized fruit fly larvae in choice and no-choice tests. We found that earwigs prey on second- and third-instar larvae and pupae of C. capitata and consumed larger numbers of second-instar larvae, followed by third-instar larvae and pupae. Females prey on larger numbers of fruit flies than did males, regardless of the prey developmental stage, but both sexes exhibited a type II functional response. Interestingly, males killed but did not consume fruit fly larvae more than did females. In no-choice tests, earwig females consumed equal numbers of parasitized and non-parasitized fruit fly larvae. However, in choice tests, the females avoided feeding on parasitized larvae. Subsequent tests with hexane-washed parasitized and non-parasitized larvae showed that putative chemical markings left on fruit flies by parasitoids did not drive the earwig preference towards non-parasitized larvae. These findings suggest that E. annulipes is a potential biological control agent for C. capitata, and that, because the earwig avoids consuming larvae parasitized by D. longicaudata, a combination of the two natural enemies could have an additive effect on pest mortality.  相似文献

The distribution and relative importance of lepidopteran and coleopteran stem borers and their natural enemies on maize and sorghum were studied in cereal growing zones of the Amhara State of Ethiopia from 2003 to 2004. Sorghum is the major crop in semi-arid eastern and maize in the cool-wet western zones of the Amhara state. Four administrative zones, 10 districts and 88 localities in the semi-arid ecozone (SAE) and four zones, 19 districts and 71 localities in the cool-wet ecozone (CWE) were chosen for the study. In SAE, the species composition was 91% Chilo partellus (Swinhoe) (Lepidoptera: Crambidae), 8% Busseola fusca (Fuller) (Lepidoptera: Noctuidae) and 1% Sesamia calamistis Hampson (Lepidoptera: Noctuidae). In the CWE, maize and sorghum are grown in different ecozones and thus B. fusca was the dominant species on sorghum, whereas 61% B. fusca and 39% S. calamistis were recorded on maize. Borer density generally increased with crop growth stage. C. partellus parasitism by C. flavipes Cameron (Hymenoptera: Braconidae), which occurred only in SAE, varied among districts ranging from 5% to 39%. In the CWE, unidentified nematodes parasitized medium-sized B. fusca larvae during the wet months. Population of native parasitoids was very low. The coleopteran borer, Rhynchaenus niger (Horn) (Coleoptera: Rhynchophoridae), attacked sorghum plants in both regions. Sorghum yields were negatively related to plant damage variables and positively to larval parasitism and plant growth variables. On maize, plant damage was too low to affect yields. Taylor’s power law indicated aggregated distribution for C. partellus and B. fusca larvae and pupae combined.  相似文献

In Latin America, the diapriid Coptera haywardi (Ogloblin) attacks the pupae of tephritid fruit flies. Anastrepha spp. are among its natural hosts, but in the laboratory it also develops in the exotic Mediterranean fruit fly, Ceratitis capitata (Wiedemann). Field cage tests demonstrated that C. haywardi could locate and parasitize Mediterranean fruit fly pupae under seminatural conditions as found in a Guatemalan coffee plantation. A mean of 18.3% of the pupae buried artificially at depths of 5 mm were parasitized by C. haywardi, while those buried at 15 mm suffered 3.2% parasitism. In a laboratory experiment, larvae that buried themselves to pupate were not significantly more likely to be parasitized than artificially buried pupae, although they may have left a physical or chemical trail that betrayed their presence. Thus, the artificial burial of pupae is unlikely to grossly underestimate C. haywardi efficacy in the field. Another field cage test found that mortality levels due to unsuccessful parasitoid attacks were similar to those resulting from successful parasitism. Thus, the actual effect of a mass-release might be considerably greater than that suggested from parasitism data alone. The results are considered sufficiently positive to encourage further testing of C. haywardi as a biological control agent of the Mediterranean fruit fly.  相似文献

An overwintering population of the mushroom phorid fly Megaselia halterata parasitized by Howardula husseyi was studied in an attempt to explain the winter decline in incidence of parasitism that has been observed in flies from mushroom farms. Fly larvae from eggs hatching in November developed into pupae in December and flies emerged in May. No selective mortality of parasitized specimens of larvae, pupae, or flies was observed. Dead parasites were found in only 10% of parasitized flies. The incidence of parasitism in the emerging flies (50%) was five times that of their parental generation and although parasitism significantly delayed fly emergence the delay was only 2–3 days. There was no evidence of winter decline in parasitism; instead there was strong evidence that parasitism enhanced phorid survival through the winter.  相似文献

Understanding the factors influencing host-selection behavior of parasitoids is essential in studies on host-parasitoid ecology and evolution, and in combining sustainable strategies of pest management, such as host-plant resistance and biological control. The effects of host-plant resistance on the olfactory response and parasitism success by Cotesia vestalis, a parasitoid of diamondback moth (Plutella xylostella) larvae were examined. Here, it was demonstrated that host-plant resistance can strongly influence foraging behavior and parasitism success of the parasitoid. In olfactometer experiments, C. vestalis did not differentiate between crucifer plant types with similar levels of susceptibility or resistance to P. xylostella but showed a strong preference for susceptible compared with partially-resistant host plants. The influence of previous oviposition activity varied with the host-plant type experienced by the parasitoid. In cage experiments, C. vestalis preferred to parasitize P. xylostella larvae on a susceptible plant compared with larvae on a partially resistant host plant when exposed to hosts for 24 h. However, this preference appeared to be transitory, and was not found after 96 h exposure. The present study suggests that combining partial host-plant resistance with biological control by C. vestalis for the control of P. xylostella may in some circumstances be antagonistic and negatively affect parasitism success.  相似文献

Wetlands are potential sites for mosquito breeding and are thus important in the context of public health. The use of chemical and microbial controls is constrained in wetlands in view of their potential impact on the diverse biota. Biological control using generalist aquatic insects can be effective, provided a preference for mosquito larvae is exhibited. The mosquito prey preferences of water bugs and larvae of odonate species were evaluated using chironomid larvae, fish fingerlings and tadpoles as alternative prey. Manly's selectivity (αi) values with 95% confidence intervals (CIs) were estimated to judge prey preference patterns. Multivariate analysis of variance (manova) and standardized canonical coefficients were used to test the effects of density on prey selectivity. The αi values indicated a significant preference (P < 0.05) in all of the insect predators tested for mosquito larvae over the alternative prey as a density‐dependent function. On a comparative scale, chironomid larvae had the highest impact as alternative prey. In a multiple‐prey experiment, predators showed a similar pattern of preference for mosquito larvae over alternative prey, reflecting a significant (P < 0.05) niche overlap. The results suggest that, in a laboratory setting, these insect predators can effectively reduce mosquito density in the presence of multiple alternative prey.  相似文献

Many caterpillars construct shelters by folding leaves and feeding from within. Many shelter-constructing species suffer high rates of parasitism as larvae or pupae. In spite of the likely significance, the effects of these shelters on the survival of pupae and the trade-off between feeding and constructing shelters have attracted little experimental attention. In both field and laboratory experiments, fern-feeding caterpillars [Herpetogramma theseusalis (Walker) (Lepidoptera: Crambidae)] invested heavily in shelters, losing weight and significantly delaying pupation rather than feeding or pupating in exposed locations. Experimentally thinning the walls of shelters in the field doubled the parasitism rate. Parasitism of pupae exceeded predation by an order of magnitude or more in both seasons of this study. Caterpillars constructed similarly sized shelters regardless of availability of fern fronds, resulting in incomplete shelters on especially small fronds, putting pupae at increased risk of parasitism. We suggest that similar interactions are common and merit more attention.  相似文献

Pyruvate decarboxylase (PDC) was purified from mature, dry maize kernels and from roots of anaerobically treated maize seedlings and partially characterized. PDC was purified to a specific activity of 96 units per milligram protein from kernels and to 41 units per milligram protein from root. The subunit molecular masses were estimated to be 61,000 and 60,000 for kernel PDC and 59,000 and 58,000 for root PDC. The pH optimum for each enzyme was 5.8. Since the pH optimum is nearly one pH unit below the value reported for the cytoplasm of anaerobically metabolizing maize roots (pH 6.7 ± 0.2), we investigated the effects of pH 5.8 and 6.6 on the cooperative kinetics observed for PDC from each source. The maximum Hill coefficients (nH) were much greater at each pH for the kernel PDC (pH 5.8, nH = 2.5 and pH 6.6, nH = 3.2) than for the root PDC (pH 5.8, nH = 1.4 and pH 6.6, nH = 1.8). The cooperative kinetics observed with respect to pyruvate were asymmetric. Potassium inhibited maize PDC and was competitive with pyruvate (root PDC Ki = 16 millimolar and kernel PDC Ki = 10 millimolar).  相似文献

The range of inducible antibacterial and other associated haemolymph proteins in Manduca sexta larvae and pupae was examined by high resolution two-dimensional (2D) isoelectric focusing-polyacrylamide gel electrophoresis. Twenty-two major inducible proteins were consistently resolved on gels of haemolymph from bacteria-injected larvae. Haemolymph from bacteria-injected pupae showed a different pattern of induced proteins. The proteins of the two stages include those which (i) are induced in both stages, (ii) those which are exclusively induced in either larvae or pupae, (iii) those which are inducible in larvae, but consititutively present in pupae, and, (iv) those which are induced in larvae, and which are present at intermediate levels but may be induced to higher levels in pupae.The antibacterial activity of the haemolymph from larvae and pupae was compared on acidpolyacrylamide gels, and the apparent Mr and pI of the inducible proteins determined. Certain of the inducible proteins appear to resemble the cecropin and attacin proteins of Hylophora cecropia.  相似文献

Acetohydroxyacid synthase has been purified from maize (Zea mays, var Black Mexican Sweet) suspension culture cells 49-fold by a combination of ion exchange chromatography, gel filtration, and hydroxyapatite chromatography. Use of the nondenaturing, zwitterionic detergent 3-([3-cholamidopropyl]dimethyl-ammonio)-1-propanesulfonate was necessary to dissociate the enzyme from the heterogeneous, high molecular weight aggregates in which it appears to reside in vitro. The solubilized maize acetohydroxyacid synthase had a relative molecular mass of 440,000. The purified enzyme was highly unstable. Acetohydroxyacid synthase activities in crude extracts of excised maize leaves and suspension cultured cells were reduced 85 and 58%, respectively, by incubation of the tissue with 100 micromolar (excised leaves) and 5 micromolar (suspension cultures) of the imidazolinone imazapyr prior to enzyme extraction, suggesting that the inhibitor binds tightly to the enzyme in vivo. Binding of imazapyr to maize acetohydroxyacid synthase could also be demonstrated in vitro. Evidence is presented which suggests that the interaction between imazapyr and the enzyme is reversible. Imazapyr also exhibited slow-binding properties when incubated with maize cell acetohydroxyacid synthase in extended time course experiments. Initial and final Ki values for the inhibition were 15 and 0.9 micromolar, respectively. The results suggest that imazapyr is a slow, tight-binding inhibitor of acetohydroxyacid synthase.  相似文献

Recent research in Ghana has demonstrated the effectiveness of an isolate of B. bassiana, sensu lato (IMI 389521) from the U.K. against the larger grain borer Prostephanus truncatus (Horn) (Coleoptera: Bostrichidae), a major pest of stored maize. To determine whether this isolate is effective on immature stages of P. truncatus, a laboratory study of the response of immature stages (egg, larvae and pupae) of P. truncatus in maize grains to two concentrations (1?×?109 and 3.16?×?109?cfu/kg maize) of B. bassiana, IMI 389521 formulated with Entostat? and kaolinite was undertaken. Adult emergence, per cent survival of adults that emerged and the number of larvae in each immature stage were assessed after 45 days. Apart from the egg experiment, higher numbers of adults emerged in grains containing larvae and pupae treated with B. bassiana product compared to the untreated maize control. This notwithstanding, survival of emerged adults of P. truncatus was greatly reduced in B. bassiana treatments (<10%) compared with 75–95% in the controls. Surface treatment with B. bassiana on pre-infested maize showed a significant effect on the developmental biology of P. truncatus in Ghana.  相似文献

The detection of the lime leafminer Phyllonorycter issikii in Novosibirsk in 2008–2009 is reported. Mines of this pest have been found on the small-leaved lime Tilia cordata and the Siberian lime T. sibirica, but not on the Amur lime T. amurensis (common host plant of moth). Leaves of T. cordata carried 2–3 times more mines than T. sibirica. On both host plants, the parasitism of mature larvae and pupae did not exceed 7%. Lack of control by local parasitoids can give the pest an opportunity to increase population density and spread further in Western Siberia.  相似文献
