Levels of monoterpene cyclase activity were determined in extracts from wounded and unwounded saplings of 10 conifer species to assess whether oleoresin biosynthesis is induced by stem wounding. Species of Abies and Picea, with low to moderate levels of constitutive monoterpene cyclase activity, exhibited a five- to 15-fold increase in cyclase activity 7 days after wounding relative to unwounded controls. In contrast, species of genera such as Pinus, with high levels of constitutive cyclase activity, did not significantly respond to wounding by alteration in the level of cyclase activity. The highest fold increase in monoterpene cyclase activity was consistently observed in Abies grandis, and the time-course of induction of activity following stem wounding in this species demonstrated a threefold increase at 2 days relative to unwounded controls, rising to a maximum increase in the response at 9 days (greater than 10-fold) followed by an apparent decline. The wound response was localized, and both bark (phloem) and wood (xylem) tissues displayed increased cyclase activity at the wound site. The magnitude of the increase in cyclase activity was dependent on the severity of the wound.  相似文献

Studies were conducted to determine whether herbivore-induced synthesis of monoterpenes occurs in the needles of ponderosa pine (Pinus ponderosa Lawson), lodgepole pine (P. contorta Douglas var. latifolia Engelmann), white fir (Abies concolor Lindl. and Gordon) and Engelmann spruce [Picea engelmanii (Parry) Engelm.]. In the needles of all species except Engelmann spruce, simulated herbivory significantly induced the activity of monoterpene cyclases 4–8 days after wounding. In ponderosa pine, real herbivory by last-instar tiger moth larvae (Halisdota ingens Hy. Edwards: Lepidoptera) induced a significantly larger response (4.5-fold increase in monoterpene cyclase activity) than did simulated herbivory (2.5-fold increase). To our knowledge, this is the first report of herbivore-induced increases in monoterpene synthesis in needle tissue. Despite this increase in monoterpene synthesis, we observed no significant increase in total monoterpene pool size in wounded needles compared to controls. Large increases in the rate of monoterpene volatilization were observed in response to wounding. We conclude that the volatile losses caused by tissue damage compensate for herbivore-induced monoterpene synthesis, resulting in no change in pool size. Tiger moth larvae consume ponderosa pine needles in a pattern that begins at the tip and proceeds downward to midway along the needle, at which point they move to an undamaged needle. Constitutive monoterpene concentrations and monoterpene cyclase activities were highest in the lower half of ponderosa pine needles. The monoterpene profile also differed between the upper and lower needle halves, the lower half possessing an additional one to four monoterpene forms. We propose that the increasing gradient in monoterpene concentrations and number of monoterpenes along the needle from tip to base deters feeding beyond the midway point and provides time for the induction of increased cyclase activity and production of new monoterpenes. The induction of new monoterpene synthesis may have a role in replacing monoterpenes lost through damage-induced volatilization and preventing extreme compromise of the constitutive defense system. A major wound response in grand fir (Abies grandis) sapling stems is the rapid increase in monoterpene production at the site of injury. Monoterpene cyclases (synthases) catalyze the formation of monoterpenes from geranyl pyrophosphate, and total cyclase activity increases markedly on wounding. At least six distinct cyclases, producing different monoterpene products, have been isolated from wounded grand fir saplings and characterized. The predominant wound-inducible cyclase produces both alpha- and beta-pinene. This pinene cyclase was purified, and polyclonal antibodies were generated in rabbits against the sodium dodecyl sulfate-denatured protein. The antibody preparation was found to cross-react by Western blotting with other grand fir monoterpene cyclases that produce different olefinic products, but not with monoterpene cyclases from related conifer species (Pinus contorta and P. ponderosa) or from angiosperms (Mentha piperita and M. spicata). The increase in monoterpene cyclase activity after wounding was closely correlated with the appearance of new cyclase protein as determined by immunoblotting. These results indicate that the wound-dependent increase in monoterpene cyclase activity is a consequence of de novo synthesis of cyclase protein.  相似文献

The turpentine fraction of conifer oleoresin is a complex mixture of monoterpene olefins and plays important roles in defense and in the mediation of chemical communication between conifer hosts and insect predators. The stereochemistry of the turpentine monoterpenes is critical in these interactions, influencing host recognition, toxicity, and potency of derived pheromones, and the stereochemical composition of these compounds lends insight into their biogenetic origin, with implications for the numbers and types of enzymes responsible and their corresponding genes. Analysis of the oleoresin from several tissues of loblolly pine (Pinus taeda) showed the derived turpentine to consist mainly of (+)-(3R:5R)-alpha-pinene and (-)-(3S:5S)-beta-pinene. Cell-free extracts from xylem tissue yielded three monoterpene synthases which together account for the monoterpene isomer and enantiomer content of the turpentine of this tissue. The major products of these enzymes, produced from the universal precursor of monoterpenes, geranyl diphosphate, were shown to be (+)-alpha-pinene, (-)-alpha-pinene, and (-)-beta-pinene, respectively. In most properties (molecular mass of approximately 60 kDa, K(m) for geranyl diphosphate of 3 microM, requirement for monovalent and divalent cations), these enzymes resemble other monoterpene synthases from conifer species.  相似文献

Two pine species (Pinus resinosa, P. banksiana) responded to inoculation with fungi carried by bark beetles by rapidly increasing monoterpene concentrations at the entry site. Changes in total monoterpenes were more pronounced than changes in proportionate compositions. The extent and rate of host response was affected by fungal species, the viability of the inoculum, and host tree species. In general, host responses were highest to fungi that are phytopathogenic and consistently associated with the major bark beetles in the study region. Simple mechanical wounding cannot account for the observed allelochemical changes, as aseptic inoculations elicited only minor reactions. Similarly, inoculation with autoclaved inviable fungi generally elicited intermediate responses, suggesting that both structural and metabolic fungal properties are important. Responses by jack pine, P. banksiana, were generally more rapid and variable than those of red pine, P. resinosa. Dose-toxicity experiments with synthetic compounds demonstrated that monoterpene concentrations present in vivo only a few days after simulated attack are lethal to most beetles. Constitutive (pre-attack) monoterpene levels can also exert some toxicity. Because bark beetles engage in pheromone-mediated mass attacks that can deplete host defenses, constitutive monoterpene levels, while a necessary early phase of successful plant defense, appear insufficient by themselves. Such interactions between constitutive and induced defense chemistry may be important considerations when evaluating general theories of plant defense.  相似文献

In response to infection by shoot infecting pathogens, Scots pine releases cortical resin into affected tissues. The resin contains a mixture of monoterpene compounds (α-pinene, β-pinene, 3-carene, β-myrcene, limonene and β-phellandrene) that retard the growth of a range of pathogens. The proportion of each monoterpene in the resin shows substantial variation among trees within a population. Thus pathogens on different trees encounter quite different monoterpene environments. To investigate the evolutionary response of pathogens to the chemically heterogeneous environment provided by Scots pine, isolates of the ascomycete canker pathogen Crumenulopsis sororia were collected from trees within a range of natural Scots pine populations. Growth rates of these isolates were measured in the presence and absence of five host monoterpenes. Substantial heritable variation for growth rate in the presence and absence of monoterpenes, and for monoterpene tolerance was recorded, suggesting the potential for the evolution of chemically specialized pathogen subpopulations on different trees within a wood. However, genetic correlations between growth rates in different monoterpene environments and between tolerance of different monoterpenes were either positive or non-significant, and there was no evidence of “tradeoffs” in performance under different monoterpene regimes. The results suggest that, on its own, the presence of monoterpene variability within Scots pine will not lead to disruptive selection on the C. sororia population. The relationship between defensive chemical diversity and pest resistance is discussed in the light of these results.  相似文献

Siberian pine (Pinus sibirica), also known as Siberian cedar pine and Siberian cedar, is an important plant that has been long used as a source of natural compounds and materials (wood, needles, soft resin, turpentine, colophony). Its chemical composition has been studied well enough; however, to our surprise, no articles that compile the phytochemical data have been published so far. Presumably, this is due to the fact that most of the studies were published in journals difficult to access and not indexed by search systems. This review, for the first time, presents a systematic compilation of available data of secondary metabolites occurring in the needles, shoots, bark, wood, seeds, and oleoresin of Pinus sibirica.  相似文献

There is a growing awareness of vegetation's role as a source of potentially reactive hydrocarbons that may serve as photochemical oxidant precursors. This study assessed the influence of light and temperature, independently, on monoterpene emissions from slash pine (Pinus elliottii Engelm.). Plants were preconditioned in a growth chamber, then transferred to an environmentally controlled gas exchange chamber. Samples of the chamber atmosphere were collected; the monoterpenes were concentrated cryogenically and measured by gas chromatography. Five monoterpenes (α-pinene, β-pinene, myrcene, limonene, and β-phellandrene) were present in the vapor phase surrounding the plants in sufficient quantity for reliable measurement. Light did not directly influence monoterpene emission rates since the emissions were similar in both the dark and at various light intensities. Monoterpene emission rates increased exponentially with temperature (i. e. emissions depend on temperature in a log-linear manner). The summed emissions of the five monoterpenes ranged from 3 to 21 micrograms C per gram dry weight per hour as temperature was increased from 20 to 46 C. Initially, emission rates from heat-stressed needles were similar to healthy needles, but rates decreased 11% per day. Daily carbon loss through monoterpene emissions accounted for approximately 0.4% of the carbon fixed during photosynthesis.  相似文献

Pine-feeding bark beetles (Coleoptera: Scolytidae) interact chemically with their host pines (Coniferales: Pinaceae) via the behavioral, physiological, and biochemical effects of one class of isoprenoids, the monoterpenes and their derivatives. Pine monoterpenes occur in the oleoresin and function as behaviorally active kairomones for pine bark beetles and their predators, presenting a classic example of tri-trophic chemical communication. The monoterpenes are also essential co-attractants for pine bark beetle aggregation pheromones. Ironically, pine monoterpenes are also toxic physiologically to bark beetles at high vapor concentrations and are considered an important component of the defense of pines. Research over the last 30 years has demonstrated that some bark beetle aggregation pheromones arise through oxygenation of monoterpenes, linking pheromone biosynthesis to the host pines. Over the last 10 years, however, several frequently occurring oxygenated monoterpene pheromone components (e.g., ipsenol, ipsdienol and frontalin) have also been shown to arise through highly regulated de novo pathways in the beetles (reviewed in Seybold and Tittiger, 2003). The most interesting nexus between these insects and their plant hosts involves the late-stage reactions in the monoterpenoid biosynthetic pathway, during which isomeric dimethylallyl diphosphate and isopentenyl diphosphate are ultimately elaborated to stereospecific monoterpenes in the trees and to hydroxylated monoterpenes or bicyclic acetals in the insects. There is signal stereospecificity in both production of and response to the monoterpenoid aggregation pheromones of bark beetles and in response to␣the monoterpenes of the pines. In the California fivespined ips, Ips paraconfusus, we have discovered a number of cytochome P450 genes that have expression patterns indicating that they may be involved in detoxifying monoterpene secondary metabolites and/or biosynthesizing pheromone components. Both processes result in the production of oxygenated monoterpenes, likely with varying degrees of stereospecificity. A behavioral analysis of the stereospecific response of I. paraconfusus to its pheromone is providing new insights into the development of an efficacious bait for the detection of this polyphagous insect in areas outside the western United States. In contrast, a Eurasian species that has arrived in California, the Mediterranean pine engraver, Orthotomicus (Ips) erosus, utilizes both a monoterpenoid (ipsdienol) and a hemiterpenoid (2-methyl-3-buten-2-ol) in its pheromone blend. The stereospecificity of the response of O. erosus to the monoterpenoid appears to be the key factor to the improved potency of the attractant bait for this invasive species.Dedicated to Professor David L. Feeding by the large pine weevil (Hylobius abietis L.) causes severe damage to the bark of Scots pine (Pinus sylvestris L.) seedlings. We measured photosynthesis, the emission of volatile organic compounds from intact and weevil-damaged bark and systemic emissions from undamaged foliage. Feeding damage did not affect photosynthesis. Monoterpenes dominated the emissions from the feeding site, although some sesquiterpenes were also emitted. Weevil feeding increased bark emission of monoterpenes by nearly 4-fold and sesquiterpenes by 7-fold. The influence of weevil damage on systemic monoterpene emissions from shoots was more profound. Several compounds were substantially induced, including linalool, β-phellandrene, limonene and 1,8-cineole. Sesquiterpenes contributed only 1.2% of the total foliage emission, but comprised eight different compounds including (E,E)-α-farnesene, β-bourbonene and (E)-β-farnesene. The total emission of monoterpenes and sesquiterpenes from shoots was respectively 2.8-fold and 2.9-fold higher in the pine weevil damaged plants than the undamaged plants. As many of the induced compounds are highly reactive in the atmosphere and form organic aerosol particles, our results suggest that conifers damaged by insects could become a more important source of secondary organic aerosols than healthy trees.  相似文献

Monoterpene cyclase activity is greatly increased in grand fir (Abies grandis) sapling stems in response to wounding and the composition of the cyclic olefin mixture generated differs from that produced constitutively as determined by radio gas-liquid chromatography. Cell-free extracts from wounded stems and from non-wounded controls were systematically compared for monoterpene cyclase activities following partial purification and separation of these enzymes by anion-exchange chromatography (Mono Q FPLC) and native PAGE. The increase in monoterpene cyclase activity following wounding represents both the apparent enhancement of constitutive cyclase activities and the appearance of novel cyclization enzymes that are absent in nonwounded controls. A pinene cyclase was shown to be the major wound-inducible enzyme directly responsible for oleoresin monoterpene formation and was tentatively identified as a 62-kDa protein by SDS-PAGE.  相似文献

Callus cultures of Pinus radiata that synthesized monoterpenes de novo and which were stable for at least 1 year have been established. The products differed from those of parent plants in that α-pinene (87–100%) rather than β-pinene was the main component. The best lines accumulated monoterpenes (ca 2 × 10?3% wt/wet wt)in yields 40–20% of that in the parent stem and needles. The composition of the extractable oil depended on the light regime. After culture in total darkness toluene and acetone accumulated. These compounds also occurred (at low levels) in dark-grown seedlings and in seeds of P. radiata and a route for their formation from α-pinene is suggested. Cell-free extracts of the culture lines converted [14C] IPP into geraniol, nerol and α- and β-pinenes in up to 46% total yield. These are the most active crude extracts for monoterpene biosynthesis that have been reported from either tissue cultures or higher plants.  相似文献

Wood from 92 trees of Pinus culminicola, Pinus nelsonii, Pinus pinceana and Pinus maximartinezii from nine localities and oleoresin from several trees of Pinus nelsonii from one Iocality were analyzed for monoterpenes by gas liquid chromatography. Monoterpenes of Pinus nelsonii were composed mainly of α- and β-pinenes, while the main constituent of the monoterpenes of Pinus pinceana and Pinus maximartinezii was limonene, present in amounts of about 90 percent. The similarity in composition of the monoterpenes of Pinus pinceana and Pinus maximartinezii strongly supports the contention of Rzedowski [12] of the close evolutionary relation of the two pines. Pinus culminicola wood produced monoterpenes composed mainly of α-pinene, sabinene, terpinolene and limonene, with other monoterpenes occurring sporadically in sizeabie amounts. The variability between individual populations was relatively high, probably due to their long-time isolation by the intervening low-elevation barriers. Appreciable differences were found in compositions of monoterpenes from stem and from twigs of Pinus culrninicola.  相似文献

Invasive populations of small spruce bark beetle Ips amitinus were first registered in 2019 in the southeast of Western Siberia. In natural stands of Siberian pine (Pinus sibirica Du Tour), several hundred hectares of outbreak foci of the alien bark beetle were identified. In 2020, a local focus of the bark beetle was found in the conifer collection in the arboretum “Kedr” of the Institute of Monitoring Climatic and Ecological Systems SB RAS, 30 km from Tomsk. The bark beetle caused the main damage to the collection of pines. I. amitinus colonized both host plants Scotch pine (Pinus sylvestris L.) and mountain pine (Pinus mugo Turra), which were previously known to it in the native range in Europe, and the local Siberian species Siberian pine (Pinus sibirica Du Tour), Siberian spruce (Picea obovata Ledeb.) and introduced Far Eastern Korean pine (Pinus koraiensis Sieb. et Zucc.). Demographic characteristics of I. amitinus studied on damaged trees indicate its high reproduction potential in Siberia. The bark beetle outbreak focus was suppressed; however, this plantation requires further annual monitoring of pest abundance and distribution, both to preserve the scientific dendroecological field station and to study the implementation of sential plant conception in relation to the invasion of I. amitinus.  相似文献

The mountain pine beetle (Dendroctonus ponderosae Hopkins) is the most serious pest of pines (Pinus) in western North America. Host pines protect themselves from attack by producing a complex mixture of terpenes in their resin. We sampled lodgepole pine (Pinus contorta variety latifolia) phloem resin at four widely separated locations in the interior of British Columbia, Canada, both just before (beginning of July) and substantially after (end of August) the mountain pine beetle dispersal period. The sampled trees then were observed the next spring for evidence of survival, and the levels of seven resin monoterpenes were compared between July and August samples. Trees that did not survive consistently had significantly higher phloem resin monoterpene levels at the end of August compared with levels in July. Trees that did survive mainly did not exhibit a significant difference between the two sample dates. The accumulation of copious defense-related secondary metabolites in the resin of mountain pine beetle-killed lodgepole pine has important implications for describing the environmental niche that the beetle offspring survive in as well as that of parasitoids, predators, and other associates.  相似文献

Cortex resin collected from 7-year-old trees of Pinus brutia from different seed sources growing in experimental plots was analyzed by GLC and GC-MS. The monoterpene composition of the cortex resin Pinus brutia subsp, brutia differs markedly from that of Pinus brutia subsp; eldarica, and of Pinus halepensis Mill. trees of the same age. Within a provenance, monoterpene composition varies considerably, yet the provenances differ significantly when analyzed by multiple analysis of variance and Duncan's Multiple Range Test. Five chemotypes were identified using the Fastclus procedure; one of which has a monoterpene composition which corresponds only to the mean monoterpene composition of the subsp. elderica. Most of the subsp. brutia provenances include a range of chemotypes, thereby implying the occurrence of genotypic variation. Cluster analysis discriminates conspicuously between subsp. eldarica and subsp. brutia. Subsp. brutia provenances are arranged into two groups, the dividing line between them being approximately the 35°E meridian.  相似文献

Monoterpene composition of cortical oleoresin was determined in a number of 9-yr-old grafts of Scots pine (Pinus sylvestris), growing in a clone trial at two different levels of mineral nutrient supply. Variation between clones was considerable for some of the monoterpene constituents, indicating genetic control. In one of the clones, δ3-carene, α-terpinene and terpinolene were totally absent. The monoterpene composition was not significantly affected by the annual application of macro-nutrients.  相似文献

Grand fir (Abies grandis) is a useful model system for studying the biochemistry, molecular genetics, and regulation of defensive oleoresin formation in conifers, a process involving both the constitutive accumulation of resin (pitch) in specialized secretory structures and the induced biosynthesis of monoterpenes and sesquiterpenes (turpentine) and diterpene resin acids (rosin) by nonspecialized cells at the site of injury. A similarity-based cloning strategy, employing primers designed to conserved regions of existing monoterpene synthases and anticipated to amplify a 1000-bp fragment, unexpectedly yielded a 300-bp fragment with sequence reminiscent of a terpenoid synthase. Utilization of this amplicon as a hybridization probe afforded four new, full-length cDNA species from a wounded fir stem cDNA library that appeared to encode four distinct monoterpene synthases. Expression in Escherichia coli, followed by enzyme assay with geranyl diphosphate (C(10)), farnesyl diphosphate (C(15)) and geranylgeranyl diphosphate (C(20)), and analysis of the terpene products by chiral phase gas chromatography and mass spectrometry confirmed that these sequences encoded four new monoterpene synthases, including (-)-camphene synthase, (-)-beta-phellandrene synthase, terpinolene synthase, and an enzyme that produces both (-)-limonene and (-)-alpha-pinene. The deduced amino acid sequences indicated these enzymes to be 618 to 637 residues in length (71 to 73 kDa) and to be translated as preproteins bearing an amino-terminal plastid targeting sequence of 50-60 residues. cDNA truncation to delete the transit peptide allowed functional expression of the "pseudomature" forms of these enzymes, which exhibited no change in product outcome as a result of truncation. Sequence comparison revealed that these new monoterpene synthases from grand fir are members of the Tpsd gene subfamily and resemble sesquiterpene (C(15)) synthases and diterpene (C(20)) synthases from conifers more closely than mechanistically related monoterpene synthases from angiosperm species. The availability of a nearly complete set of constitutive and inducible monoterpene synthases from grand fir (now numbering seven) will allow molecular dissection of the resin-based defense response in this conifer species, and detailed study of structure-function relationships among this large and diverse family of catalysts, all of which exploit the same stereochemistry in the coupled isomerization-cyclization reaction.  相似文献

The elicitor activity of compounds extracted from the mycelia of six species of phytopathogenic fungi was assessed from the sizes of necrotic lesions on the external surface of the living trunk phloem of five coniferous species inhabiting Siberia: Siberian larch (Larix sibirica L.), Scotch pine (Pinus sylvestris L.), Siberian spruce (Picea obovata Ledeb.), Siberian fir (Abies sibirica L.), and cedar pine (Pinus sibirica (Rupr.) Mayr.). The compounds for inoculation were extracted from the mycelium of ascomycetes imperfect, and basidium fungi; the living mycelia of these fungi were also used. The fungal extract or mycelium was placed into the hollows 7 mm in diameter in the trunk bark. Infection triggered the formation of hypersensitivity necrotic lesions in the inner bark exceeding in size those appeared after control wounding of four tree species (larch, pine, spruce, and cedar); fir was an exclusion. In experiments with tree trunks and conifer calluses, a dependence of immune response parameters (the sizes of necrotic lesions and the content of lignin and bound proanthocyanidins) on the quantity of the fungal preparation was elucidated. The largest necrotic lesions appeared after injection of 500μg of the fungal preparation into the hollow in the trunk, and its higher quantities did not increase the indices measured. The size of the necrotic lesion on the trunk bark is supposed to be used as a promising index characterizing the level of tree immunity and tolerance under various ecological conditions.  相似文献

Conifers respond to herbivore attack with defensive chemicals, which are toxic to both insects and their associated microorganisms. Microorganisms associated with insects have been widely reported to metabolize toxic chemicals, which may help both microorganisms and host insects overcome host conifer defense. Dendroctonus valens LeConte, an introduced exotic pest from North America to China, has killed millions of healthy pines. Alpha‐pinene is the most abundant defensive monoterpene in Chinese Pinus tabuliformis. Although microorganisms associated with D. valens have already been investigated, little is known about their bioactivities when encountering host defensive monoterpenes. In this study, we evaluated the influences of different concentrations of α‐pinene to D. valens and the three most frequently isolated yeasts and bacteria of D. valens, and further assayed microorganisms’ capabilities to degrade α‐pinene. Results showed that the gallery lengths and body weight changes of bark beetles were significantly affected by 6 mg/mL and 12 mg/mL of α‐pinene applied in media compared to controls. The tolerance of experimental microorganisms to α‐pinene varied depending on the microbial species. Two out of three yeast strains and all three bacterial strains degraded 20%–50% of α‐pinene compared to controls in 24 h in vitro. The microorganisms capable of α‐pinene degradation in vitro and their tolerance to high levels of α‐pinene suggested that D. valens‐associated microorganisms may help both microorganisms and the bark beetle overcome host α‐pinene defense.  相似文献
