The effect of ABA on root growth, secondary-root formation androot gravitropism in seedlings of Zea mays was investigatedby using Fluridone-treated seedlings and a viviparous mutant,both of which lack carotenoids and ABA. Primary roots of seedlingsgrown in the presence of Fluridone grew significantly slowerthan those of control (i.e. untreated) roots. Elongation ofFluridone-treated roots was inhibited significantly by the exogenousapplication of 1 mM ABA. Exogenous application of 1 µMand 1 nM ABA had either no effect or only a slight stimulatoryeffect on root elongation, depending on the method of application.The absence of ABA in Fluridone-treated plants was not an importantfactor in secondary-root formation in seedlings less than 9–10d old. However, ABA may suppress secondary-root formation inolder seedlings, since 11-d-old control seedlings had significantlyfewer secondary roots than Fluridone-treated seedlings. Rootsof Fluridone-treated and control seedlings were graviresponsive.Similar data were obtained for vp-9 mutants of Z. mays, whichare phenotypically identical to Fluridone-treated seedlings.These results indicate that ABA is necessary for neither secondary-rootformation nor for positive gravitropism by primary roots. Anatomical changes in roots of wheat seedlings (Triticum aestivumL. cv. Hatri) following oxygen deficiency in the rooting mediumwere investigated. The response of the plant to stress was testedat a very early developmental stage when the first adventitiousroots had just emerged. In order to analyze the adaptation ofdifferent roots, respiration rates of the roots 1–3 and4–n were compared with the respiration rates of the totalroot system. Oxygen deficiency was induced either by flushingnutrient solution with nitrogen or flooding of sand. In contrast to plants grown in well aerated media, both stressvariants led to a significant increase of the intercellularspace of the root cortex in seminal and first adventitious roots.Radial cell enlargement of cortical cells near the root tip,cell wall thickenings in flooded sand cultures and an increasein phloroglucinol-stainable substances were found to be furtherindicators of low oxygen supply. The roots 4–n which were promoted in growth under hypoxiashowed higher respiration rates; hence the total root respirationwas not restricted. Triticum aestivum L. cv. Starch occupies 4.2 per cent of the volume of plastids in calyptrogencells in primary roots of Zea mays L. cv. vp-7 wild type. Plastidsin calyptrogen cells are distributed randomly around large,centrally located nuclei. The differentiation of calyptrogencells into columella cells is characterized by cellular enlargementand the sedimentation of plastids to the bottom of the cells.Although sedimented plastids in columella cells do not containsignificantly more starch than those in calyptrogen cells, primaryroots are graviresponsive. The onset of root gravicurvatureis not associated with a significant change in the distributionof plastids in columella cells. These results indicate thatin this cultivar of Z. mays (1) the sedimentation of plastidsin columella cells is not based upon their increased densityresulting from increased starch content alone, (2) starch-ladenamyloplasts need not be present in columella cells for rootsto be graviresponsive, and (3) the onset of root gravicurvaturedoes not require a major redistribution of plastids in columellacells. The distribution of lateral root primordia in the root tipsof four Musa landraces (Grande Naine, Pisang Berlin, Ngok Egomeand Yangambi Km5) grown in the field has been investigated toevaluate the range of genetic variation of lateral root initiation.In banana (Musa sp.), lateral roots are initiated in the roottip, 0.6–4 mm behind the root/cap junction and arise inseveral protoxylem-based longitudinal rows or ‘ranks’.Significant differences were observed among landraces for theposition of the most distal primordium, however the longitudinalspacing between successive primordia along the ranks was similarfor all landraces. All ranks were involved in lateral root initiation.The number of ranks also showed significant variations amonglandraces and was proportional to the stelar diameter. Hencethe density of lateral roots (roots cm-1) was affected by stelardiameter variations. Finally, root elongation in the root tipwas landrace-specific and not necessarily exponential, unlikesuggested in previous studies. It is concluded that lateralroot initiation in Musa is not involved in the genetic variationsof root architecture in the field. Earlier work has established that the saturable component ofuptake of RS-[214C]ABA by bean (Phaseolus coccineus L. cv. Prizewinner)root segments can be attributed to the action of a carrier.We now show that the carrier-mediated uptake is unaffected byRS-2-trans-ABA and lunularic acid and the unnatural R-ABA alsoappears to be ineffective. The specificity for S-ABA requiresthe halving of the Km value for ABA determined previously (2.6mmol m-3 for RS-; 1.3 mmol m-3 for S-ABA). The RS-1', 4'-cis-dioland RS-1'-deoxy ABA reduce the uptake of RS-[2-14C]ABA aboutas strongly as does unlabelled ABA, the K1 for 1'-deoxy ABAwas similar to the Km for ABA. The K1 for RS-1', 4'-trans-diolwas 15.7 mmol m-3. Consideration of the stereochemistry of thesecompounds suggests that the face of the ring of ABA away fromthe 1'-hydroxyl group interacts with the carrier site. Labelled material diffused out of undamaged root surfaces whichhad absorbed RS-[3H]ABA through an apical cut, suggesting thatABA is present in the apoplast. A simplified hypothesis is presented that can account for polartransport of ABA based on a gradient of a carrier in a tissuebut where the carrier is distributed uniformly on the apicaland basal ends of each cell. Fine root production and mortality in central Himalayan evergreenforests consisting of Quercus leucotrichophora (banj oak) andPinus roxburghii (chir pine) were measured. Fine root productionand mortality decreased with increasing soil depth. Annual fineroot production was higher in the broadleafed forest than inthe coniferous forest, across months and seasons (1.3 and 1.5-timesmore living and dead root biomass, respectively in banj oakthan in chir pine). Live fine root production was 2508 kg ha-1year-1inchir pine forest and 3631 kg ha-1year-1in banj oak forest. Deadfine roots accumulated at a rate of 1197 and 1525 kg ha-1year-1inchir pine and in banj oak forest, respectively. In both forests,the greatest fine root production was recorded in the rainyseason followed by summer and winter seasons. Two hydroponic experiments were conducted to determine the effectsof brief and prolonged AI3+ exposures on the hydraulic conductivity(Lp) of northern red oak (Quercus rubra L.) root systems. RootLp was determined using the pressure chamber method of Fiscus(1977). In the first experiment, 28- to 40-d-old seedlings weretreated for 4 d with complete nutrient solutions containingone of three Al concentrations (0.04, 1.85 or 3.71 mol m–3)and either 0 or 50 mmol m–3 P. Neither Lp nor daily transpirationwas affected by treatment. In Experiment II, seedlings were grown for 48–63 d incomplete solutions containing one of three Al concentrations(0, 0.75 or 2.00 mol m–3) and either 10 or 250 mmol m–3Ca. Lp and leaf area to root length ratio (LA/RL) were reducedwhen (AI3+/ Ca2+), the solution activity ratio, was 2.9 andhigher. Lp and LA/RL were also negatively correlated with Alconcentration and Al/Ca concentration ratio in the roots. Lpwas positively correlated with LA/RL in both experiments. Itis unclear whether Lp in the second experiment was reduced directlyby solution and root chemistry or whether Lp changed in responseto altered leaf/root balance. Root Growth Inhibitors from Root Cap and Root Meristem of Zea mays L.   总被引:1,自引:0,他引:1  
A micro-assay based on the growth inhibition of root segmentsof the seminal roots of Zea mays has been used to investigatethe root-growth-inhibiting substances in root caps and meristemsrespectively of the roots of Zea mays. This micro-assay is sensitiveto 50 pg of IAA or less. Paper chromatography of the acid fractionof methanolic extracts shows the presence of one main inhibitorin root caps and a different main inhibitor in root meristems.Neither is IAA, whose presence in meristems is sometimes indicatedby small inhibitions (or stimulations) at the characteristicRf of IAA. A Commelina leaf-epidermis assay shows the presenceof one stomata-closing ABA-like substance in root caps and onein meristems, one corresponding in Rf to the main root-growthinhibitor from the root cap. The implications of these findingsfor the geotropic responses of roots is briefly discussed.  相似文献

Transport of 14C-photosynthate in decapitated stems of Phaseolusvulgaris explants was dependent on the concentration of indole-3-aceticacid (IAA) applied to the cut surfaces of the stem stumps. Thephysiological age of the stem influenced the nature of the transportresponse to IAA with stems that had ceased elongation exhibitinga more pronounced response with a distinct optimum. Increasednutrient status of the explants had little influence on theshape of the IAA dose-response curve but increased, by two ordersof magnitude, the IAA concentration that elicited the optimalresponse. Applications of the inhibitor of polar auxin transport,1-(2-carboxyphenyl)-3-phenylpropane-1, 3-dione (CPD), affectedIAA-promoted transport of 14C-photosynthates. At sub-optimalIAA concentrations, CPD inhibited transport, whereas at supra-optimalIAA concentrations, 14C-photosynthate transport was marginallystimulated by CPD. Treatment with CPD resulted in a significantreduction in stem levels of [14C]IAA below the site of inhibitorapplication, while above this point, levels of [14C]1AA remainedunaltered. The divergent responses of auxin-promoted transportto CPD treatment are most consistent with a remote action ofIAA on photosynthate transport in the decapitated stems. Stirlingia latifolia, a common shrub of Banksia woodlands ofSW Australia, is a highly successful resprouter species recoveringfrom fire by multiple sprouting of new shoots from its upperroot stock. in comparison with the congeneric fire-sensitive(obligate seeder) species Stirlingia tenuifolia it exhibitsa low shoot:root dry weight ratio and high concentrations ofstored starch in the cortical tissue of its roots. The relationshipbetween root reserves of starch and development of newly sproutingshoot material following fire is examined in S. latifolia afterspring and summer burns. During the initial 2-5 month periodafter fire, levels of stored starch in the roots fall by 50-75%,followed by a slow increase as plants reproduce and the attainmentof pre-fire starch levels by 1·5-2 years after the fire.Starch reserves of roots can be further reduced by shading theregenerating shoots to limit their input of photosynthates andalmost totally eliminated by monthly removal of successive flushesof new shoots over a 10-12 month period. New shoots continueto sprout until all the starch is eliminated. Species of the genus Veronica differ in habitat preferences,growth form and in adventitious root production. The annualspecies rarely or never produce adventitious roots in intactplants in the field but some, for example V. persica and V.arvensis will root vigorously from single node stem segmentsin culture. Others, such as V. agrestis require the presenceof IAA for substantial levels of root formation to occur incultured stem segments. Veronica hederifolia cuttings rarelyproduce roots. Stem cuttings of the perennial species, in general,rooted more vigorously than those of annual plants. Both V.fihiformis and V. serpyllifolia root very strongly. The position of root production from the stem cuttings differedfrom species to species. Roots arose either from the node, theregion of the base or at some intermediate point. Veronica arvensis,V. chamaedrys and V. persica rooted mainly from the basal regionwhereas V. filiformis rooted mainly from the node. Veronicaserpyllifolia cuttings rooted at both of these locations. Veronica filiformis, a perennial species that is infertile inBritain, produces root primordia in intact plants at nodes whichare close to the shoot apex. Thus, even very young stem segmentshave ‘preformed’ root primordia. For this reason,detached stem segments of V. filiformis root very rapidly andthis probably has been of great significance in its successfulinvasion and spread in lawns and short turf areas. The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 10–7m s–1 MPa–1 for F. acanthodes and 5.2 ? 10–7m s–1 MPa–1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (1–3 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of –0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from–0.016 to –1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Nuclear volumes and cell areas were determined for seven regionsof the meristem of roots of Zea mays. Roots were fixed in 10per cent neutral buffered formalin, in 3 per cent glutaraldehydeor in acetic acid/alcohol; they were prepared as sections oralls were teased apart. Mean volumes of interphase nuclei weresimilar in all regions of the root except the vascular tissueof the stele. Mean nuclear volumes and the overall range ofvolumes were similar in sub-populations of cells with differentproportions of G1, S and G2 cells, e.g. in row I of root capinitials, whose cells lack a G1 phase, and in quiescent centrecells, which are mainly in G1. Nuclear volume does not appearto be closely correlated with DNA content. Nuclear volumes covereda 6 to 12-fold range within a meristem and even within specificregions, in which cells are part of the same cell lineages,there was a 4- to 9-fold range. Nuclear volumes were comparedin sister cells in rows I and II of the root cap initials. In10 per cent of the pairs, sister nuclei had identical volumes;the other pain had different volumes and mean difference was68 µm3. Mechanisms by which this variability could begenerated are discussed, particularly asymmetry, at mitoses,of factors that regulate nuclear growth. Polar auxin transport inhibitors, including N-1-naphthylphthalamicacid (NPA) and 2,3,5-triiodobenzoic acid (TIBA), have variouseffects on physiological and developmental events, such as theelongation and tropism of roots and stems, in higher plants.We isolated NPA-resistant mutants of Arabidopsis thaliana, withmutations designated pir1 and pir2, that were also resistantto TIBA. The mutations specifically affected the root-elongationprocess, and they were shown ultimately to be allelic to aux1and ein2, respectively, which are known as mutations that affectresponses to phytohormones. The mechanism of action of auxintransport inhibitors was investigated with these mutants, inrelation to the effects of ethylene, auxin, and the polar transportof auxin. With respect to the inhibition of root elongationin A. thaliana, we demonstrated that (1) the background levelof ethylene intensifies the effects of auxin transport inhibitors,(2) auxin transport inhibitors might act also via an inhibitorypathway that does not involve ethylene, auxin, or the polartransport of auxin, (3) the hypothesis that the inhibitory effectof NPA on root elongation is due to high-level accumulationof auxin as a result of blockage of auxin transport is not applicableto A. thaliana, and (4) in contrast to NPA, TIBA itself hasa weak auxin-like inhibitory effect. Waterlogging tolerance, root porosity and root anatomy wereevaluated for 20 Trifolium accessions (species and sub-species,all annuals) selected from the eight Sections of the genus.Nine accessions were sensitive [relative growth rate (RGR) reducedby up to 80%] to waterlogging, nine accessions were tolerant(RGR not reduced), and in two accessions RGR increased (up to1.9-fold), when compared to drained controls. Growth of themain (i.e. tap) root axis was severely reduced in all accessionswhen waterlogged. Lateral roots formed the bulk of the rootsystem of tolerant accessions when grown in waterlogged soil.Lengths of the longest lateral roots were up to three-timeslonger than the main root axis. Root porosity varied from 0.7–12%among accessions when grown in aerated solution and from 1.1–15.5%in plants grown in hypoxic (0.031–0.045 mol O2m-3) solution.In some accessions aerenchyma formed by cell lysigeny; in othersit formed by schizogenous cell separation, or a combinationof both processes. O2consumption rates of expanded lateral roottissues varied by up to 1.7-fold (on a mass basis) among thesix accessions tested and was reduced by an average of 24% forroots of plants grown in hypoxic solution prior to measurements.Accessions with the highest root porosity tended to have longerroots when grown in waterlogged soil. Three accessions formed‘aerotropic roots’ and the lateral root lengthsof these plants exceeded those of all other accessions, suggestingenhanced O2movement to the submerged lateral root axis via theaerotropic roots. Waterlogging-tolerant accessions were identifiedin seven of the eight Sections in Trifolium, and the tolerantaccessions tended to be those with extensive lateral root systemsof relatively high porosity. Transformed roots of Tagetes erecta were obtained followinginfection of stems of sterile plantlets with Agrobacterium rhizogenesstrain TR105. The thiophenes detected were 5-(4-hydroxy-l-butenyl)-2,2'-bithienyl,5-(4-acetoxy-l-butenyl)-2,2'-bithienyl, 5-(3-buten-l-enyl)-2,2',-bithienyl and 2.2': 5', 2'-terthienyl. The thiophene patternwas the same in normal root cultures and roots of the intactplant. Transformed roots showed a higher growth rate and a higherbiomass yield than normal root cultures on a hormone-free media. Cytokinins, cis-zeatin and cis- and (trans-ribosylzeatin, wereidentified in the root exudate of the rice plant (Oryza sativa,indica cultivar IR-24) after several chromatographic separationsand combined gas-liquid chromatography-selected ion monitoring(GC-SIM) analysis. The presence of trans-zeatin ribotide wassuggested by enzyme hydrolysis, subsequent chromatographic separationand GC-SIM. The comparatively high content of the ribotide inthe root exudate suggests the form of cytokinins to be transportedfrom roots to other parts in the rice plant. Root morphogenesis and cell cycle kinetics of intact and aseptically-grownexcised roots of Helianthus annuus L. were studied. Intact rootsshow predominant cell arrest in G1 with an absence of polyploidcells coincident with secondary vascularization. Exposure ofthe cut ends of aseptically grown excised roots to known concentrationsof indol-3-yl acetic acid, benzyladenine, and myo-inositol for8 weeks initiated the production of secondary vascular tissuesand predominant cell arrest in G2 concommitant with poiyploidization.Excised roots grown in the absence of these substances producedroots with only primary vascularization and predominant cellarrest in G1 coincident with an absence of polyploidization.These results indicate that (a) root cells of H. annuus havethe ability to undergo polyploidization that may be inducedby exogeneously applied chemicals, (b) a general relationshipbetween predominant cell arrest in G1 coincident with the absenceof secondary vascularization does not hold true and (c) althoughsecondary vascularization occurs in cultured roots exposed toall three additives similar to secondary vascularization inintact roots, the two roots should not be considered identicalin all respects. Bot. 38: 42–52 Oilseed rape (Brassica napus L. cv. Bien venu) was grown for49 d in flowing nutrient solution at pH 6?0 with root temperaturedecrementally reduced from 20?C to 5?C; and then exposed todifferent root temperatures (3, 5, 7, 9, 11, 13,17 or 25?C)held constant for 14 d. The air temperature was 20/15?C day/nightand nitrogen was supplied automatically to maintain 10 mmolm–3 NH4NO3 in solution. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g–1 d–1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration.
