Summary In adult female specimens of Rana temporaria, the pars ventralis of the tuber cinereum of the hypothalamus was isolated from the brain. The isolated pars ventralis retained its normal connections with the median eminence and hypophysis. The normal blood supply of the whole isolated region (pars ventralis tuberis + median eminence + hypophysis) was preserved.The results showed (1) that, in accordance with previous experiments, as far as concerned gametogenesis, seasonal development of the gonads and of the secondary sexual characteristics, the function of the gonadotropic centre of the pars ventralis tuberis is largely independent of nervous connections between the pars ventralis of the tuber cinereum and the brain; (2) that, in animals in which the pars ventralis tuberis was isolated from the brain at the beginning of the seasonal development of the ovaries, the L.H. secretion is disturbed during the ovulation period; (3) that consequently, in such animals, the stimulating activity of the isolated gonadotropic centre on the L.H. secretion of the hypophysis is too weak to cause ovulation; (4) that nervous connections between the isolated region and the preoptic region of the brain are indispensable for ovulation.  相似文献

Summary In a large number of adult female and male specimens of Rana temporaria, the pars ventralis of the tuber cinereum of the hypothalamus was completely isolated from the brain. The pars ventralis retained its normal connections with the hypophysis. The vascular supply of the whole isolated region was left intact. Two and a half months after operation, in some animals, the interruption of the nervous pathways was still complete. In these animals it could be shown that all or nearly all A. F.-negative nerve fibres of the median eminence are axons of nerve cells localized in the area periventricularis of the pars ventralis tuberis. According to our results in Rana, the same A. F.-negative nerve fibres of the higher vertebrates are probably also axons of homologous nuclei of the tuber cinereum.In accordance with previous experiments, it could be concluded that the nerve cells of which the A. F.-negative fibres are axons, form an important centre that, via the pars distalis of the hypophysis, is responsible for the seasonal development of the ovaries, eggs and oviducts in the females and for the seasonal development of the thumb pads in the males.  相似文献

Summary In 27 adult female specimens of Rana temporaria, the total interruption of the nervous pathways to the median eminence and the hypophysis was performed, without disturbing the normal blood supply of the hypophysis. In eight cases this complete interruption persisted three months after operation. In these animals there occurred no seasonal development of the eggs, ovaries and oviducts. In the 19 other operated animals a varying degree of regeneration of the interrupted nerve fibres occurred. In these animals a varying number of ripe eggs was present, but in many of them there existed a significant difference between the weight of their ovaries compared with that of the ovaries of the control animals. In all control animals the seasonal development of the ovaries and oviducts was normal. From these and from previous experiments it could be concluded that the A. F. negative nerve fibres of the median eminence, not originating from the magnocellular neurosecretory preoptic nuclei, are responsible for the normal seasonal development of the ovaries and oviducts in Rana temporaria. It must be these nerve fibres that release gonadotropic releasing factors into the blood capillaries of the median eminence. The A. F. positive neurosecretory fibres, the portal circulation of the pars distalis and the pituicytes of the median eminence play only a secondary part during this period of control of the gonadotropic activity of the hypophysis.  相似文献

Summary In normal pars distalis of Rana temporaria, a gradient of activity of the P.A.S.-positive cells is always present. In animals, with complete and permanent interruption of all nervous pathways to the median eminence and hypophysis and without disturbance of the normal blood supply of the hypophysis, a subtotal disappearance of the gradient of activity occurred and the total activity of the P.A.S.-cells of the pars distalis was lowered. The activity of the P.A.S.-positive cells regressed in a typical way, suggesting a humoral mechanism.In animals with marked regeneration of the interrupted nerve fibres, a normal gradient of activity was present. From these and previous experiments, it is concluded that the gradient of activity of the P.A.S.-positive cells of the pars distalis of Rana temporaria is largely dependent on the A.F.-negative fibres of the median eminence, not originating from the magnocellular neurosecretory preoptic nuclei. It is suggested that these nerve fibres exert their influence on the activity of the P.A.S.-positive cells via substances released in the blood capillaries of the median eminence.The gradient of activity and its variations are mainly related to the gonadotropic function of the pars distalis.I am indebted to miss M. Summary Volumetric, karyometric and histological studies were done on the pars distalis of the hypophysis of Rana temporaria, in normal and in experimental conditions.In the normal pars distalis, a gradient of activity of the P.A.S.-positive cells is described which shows a close relation with the blood supply. This relation strongly suggests the presence of a gradient of humoral factors which would influence the activity of the pars distalis. This gradient is not dependent on the influence of A.F.-positive neurosecretory fibres. In the P.A.S.-negative cells (O.G.-cells) a similar gradient could not be found. In transplants of pars distalis the gradient of activity is absent.There exists a striking difference between autotransplants and homotransplants of pars distalis in the eye, and also between autotransplants on the iris and autotransplants in the cornea.A parallelism between autotransplants of pars distalis on the iris and the pars distalis of animals deprived of their median eminence could be established. In both groups the pars distalis shows a similar decrease in volume. The glandular cells are well differentiated, but show a low activity. This low activity is most pronounced in the P.A.S.-cells. The normal variability in staining of the P.A.S.-cells has diminished: in general, the great majority of the P.A.S.-cells are P.A.S.-purples. The gradient of activity of the pars distalis has disappeared. The seasonal development of the gonads and of the secundary sexual characteristics is absent. This absence is probably not caused by a decrease of the total number of P.A.S.-cells of the pars distalis. It is probably due to the low activity of the P.A.S.-cells, which cannot be ascribed to the absence of the neurosecretory influence of the magnocellular preoptic nuclei.A preliminary note on the gradient of activity of the pars distalis has been published (Dierickx 1963c).  相似文献

Summary Effects of Methallibure on the gonadotropic stimulation of the regressed gonads and secondary sex characters of the pars distalis-ectomized adult green frog, Rana esculenta, were studied. Regression of the gametogenetic activity and reduction of oviducts and thumb pads are the eventual consequences of the removal of pars distalis. Homoplastic pars distalis homogenates restimulate the gametogenesis and regressed secondary sex characters due to their gonadotropic hormone contents. Methallibure in this case blocks the stimulation of these organs by homoplastic pars distalis, which indicates that this compound inhibits the action of gonadotropic hormones upon their target tissues.Work supported by the Population Council (Grants M70.082 C & M71.0132 C) of New York, and National Research Council of Italy.  相似文献

Summary A light microscopic immunocytochemical study of the brain of frogs with hypothalamic lesions was performed in order to obtain evidence concerning the origin of somatostatin fibers in the median eminence and neural lobe of the hypophysis. The results indicate that the somatostatin fibers of the neural lobe originate from somatostatin perikarya located in the prechiasmatic part of the hypothalamus and possibly also in the telencephalon. The somatostatin neurons of the pars ventralis tuberis cinerei do not send axons to the neural lobe. The frog median eminence contains axon terminals of somatostatin neurons located in the pars ventralis of the tuber cinereum. Many other somatostatin fibers of the frog median eminence originate from somatostatin neurons located outside the tuber cinereum. Summary In a large number of adult female and male specimens of Rana temporaria, a total extirpation of the magnocellular neurosecretory preoptic nuclei was performed. Several months after operation, no regeneration of the neurosecretory nuclei had occurred. The results showed 1. that all A.F.-positive neurosecretory nerve fibres of the hypothalamo-hypophysial region originate from nerve cell bodies localized in the magnocellular preoptic nuclei; 2. that the magnocellular preoptic nuclei do not play an important part in the inhibitory control of the activity of the pars intermedia of Rana temporaria; 3. that the possible role of the magnocellular preoptic nuclei in the seasonal development of the gonads and of the secondary sexual characteristics of Rana, could be only of secondary importance; 4. that, in accordance with previous experiments, in Rana, ovulation is dependent on preoptic hypothalamic structures. Some results of the present and of other experiments can be interpreted as supporting the hypothesis of a possible dual higher control over the gonadotropic centre (Dierickx, 1965b, 1966, 1967) of the pars ventralis of the tuber cinereum.A part of these studies has been reported previously in abstract form (Dierickx, 1963a, 1963b, 1965a).  相似文献

Summary By means of single and double immuno-enzyme cytochemical staining techniques, it was shown that the external region of the amphibian median eminence contains separate vasotocinergic and mesotocinergic nerve fibres. Moreover, it was demonstrated that the vasotocinergic fibres also contain neurophysin. In animals in which the hypothalamic magnocellular neurosecretory preoptic nuclei had been completely removed, the immuno-reactive vasotocinergic and mesotocinergic fibres of the median eminence had disappeared. From this result, it is concluded that, at least the great majority of the vasotocinergic and mesotocinergic fibres of the external region of the amphibian median eminence are processes of neurosecretory perikarya located in the hypothalamic magnocellular preoptic nuclei. On the other hand, our results do not exclude the possibility that a minority of these neurosecretory fibres originate from small immuno-reactive perikarya which were found in the tuber cinereum. The observation that both kinds of processes accumulate around blood capillaries of the hypophysial portal system strongly suggests that they play a role in the control of the activity of the pars distalis of the hypophysis.This investigation was supported by a grant from the Belgian Nationaal Fonds voor Geneeskundig Wetenschappelijk Onderzoek  相似文献

Summary The pars tuberalis of the hypophysis of Rana temporaria presents the general structural and the cytological characteristics of an endocrine gland. It is composed of elongated cells with long, branching processes ending on the external basement membrane of the pericapillary space. The pars tuberalis cells produce secretory granules which are accumulated in the pericapillary endings of the processes.Corresponding to its separate localization, the pars tuberalis of Rana temporaria has a separate vascularization of which the efferent capillaries anastomose with the capillary plexus of the median eminence. The general direction of the blood flow of the pars tuberalis is towards the capillaries of the median eminence. Also, the secretory products of the pars tuberalis pass into the blood stream of the hypophysial portal system.Several characteristics of the pars tuberalis show that its function must be different from that of the pars distalis of the hypophysis. Moreover, in contrast with the pars distalis, the activity of the pars tuberalis is not regulated by neurohumoral factors.The results show that a role of the pars tuberalis in the regulation of the activity of the pars distalis of the hypophysis is not excluded.  相似文献

The pars distalis of the avian adenohypophysis consists of well-defined cephalic and caudal lobes which are distinct in their cellular constituents. Immunocytochemical investigations on the pituitary hormones of the pars distalis of the Japanese quail reveal five types of secretory cells, adenocorticotropin (ACTH) cells, prolactin (PRL) cells, thyroid-stimulating hormone (TSH) cells, growth hormone GH (STH) cells, and FSH/LH (gonadotropic) cells. The ACTH cells, TSH cells, and PRL cells are restricted to the cephalic lobe, and GH (STH) cells are confined to the caudal lobe, while FSH/LH cells are distributed throughout the cephalic and caudal lobes. The median eminence of birds has distinct anterior and posterior divisions, each with different neuronal components. The avian hypophysial portal vessels also consists of two groups, anterior and posterior. The peculiar arrangement and distribution of the avian hypophysial portal vessels are possibly related to the distribution of neuropeptides in the two divisions of the median eminence and to the cytological and functional differentiation of two lobes of the pars distalis. The localization of perikarya and fibers containing luteinizing hormone releasing hormone (LHRH), somatostatin, vasotocin, mesotocin, corticotropin-releasing factor (CRF), vasoactive intestinal polypeptide (VIP), glucagon, metenkephalin, and substance P in the hypothalamus and median eminence of the Japanese quail has been investigated by means of immunohistochemistry using antisera against the respective neuropeptides. LHRH-, somatostatin-, VIP-, met-enkephalin-, and substance P-immunoreactive fibers are localized in the external layer of the anterior and posterior divisions of the median eminence, while CRF- and vasotocin-reactive fibers are demonstrated only in the external layer of the anterior division of the median eminence. The metenkephalin fibers are thicker in the anterior median eminence but the substance P fibers are more abundant in the posterior division. Mesotocin fibers occur only in the internal layer of the median eminence and neural lobe.  相似文献

Summary The vascularization of the pars distalis of the hypophysis of the toad, Bufo bufo (L.), was studied by the traditional method of injecting a mixture of India-ink and gelatine into the circulatory system of the head via the arteria carotis communis. Further, methyl-methacrylate corrosion casts of the brains were made; the hypothalamo-adenohypophysial region of these corrosion casts was studied with the scanning electron microscope. The results showed that the portal vessels which arise from the median eminence do not supply distinct areas in the pars distalis as is supposed by the point-to-point-hypothesis. The portal vessels enter the ventro-median region of the pars distalis and branch off into a three-dimensional network of the secondary capillary plexus of the pars distalis. The plexus is made up mostly by four- to six-sided meshes. This angioarchitecture guarantees an optimal supply of the glandular cells of the pars distalis with nutritional factors and releasing hormones, on the one hand, and facilitates the removal of the hormones which are released by these cells, on the other hand. Summary By means of electron microscopy, in the median eminence of Rana temporaria, the terminal arborizations of axons of six different types of neurosecretory cells, located in the pars ventralis of the tuber cinereum, were identified. In addition, phenomena connected with the release of neurosecretory material from the axon terminals of these neurosecretory cells into the blood capillaries of the median eminence are described.Preliminary results suggested the existence, in the median eminence, of additional different neurosecretory axon types which could also belong to corresponding neurosecretory cell types probably located in the apical part of the pars ventralis of the tuber cinereum. Moreover, in the external region of the normal median eminence, separate monoaminergic nerve fibres were tentatively identified. Arguments are adduced which plead (1) against the assumption that the ependyma or the pituicytes of the median eminence could produce adenohypophysiotropic hormones; (2) against the inference that the ependymal cells of the median eminence might be involved in the transport of adenohypophysiotropic hormones from the cerebrospinal fluid into the blood capillaries of the median eminence.  相似文献

Summary Nerve fibres containing granular vesicles first appear in the median eminence of the rat on the 16th foetal day while secretory granules in the cells of the adenohypophysis are not present till the 17th foetal day. These observations suggest that the differentiation and early activity of pars distalis cells may depend on substances elaborated at nerve terminals in the median eminence. Although the loops of the primary plexus of portal vessels do not develop until the 4th postnatal day, substances released by nerve fibres in the neurohypophysis could reach the pars distalis through vessels already present at the 15th foetal day in the mesenchyme between the diencephalon and the adenohypophysis. This view is supported by the fact that the earliest cells to exhibit ultrastructural evidence of secretory activity are in the rostral pole of the pars distalis, the first region of the gland to become vascularized. The earliest granules to appear in the cells of the pars distalis correspond to those which are considered to contain mucoprotein hormones; somatotrophin type granules were seen only in postnatal tissues.The finding that, in the median eminence, the development of granular vesicles precedes that of agranular vesicles is discussed with reference to the times at which neurosecretory materials and monoamines become detectable in the region.We should like to thank Miss Ann Pearson, Mr. D. The immunohistochemical avidin-biotin complex method was used to study hormone-producing cells in the adenohypophysis of the skink Chalcides chalcides during embryonic development. In Chalcides, the formation of Rathke's pouch was evident between stages 28 and 30 of embryonic development. The adenohypophysial cells begin to differentiate before the morphological development of the gland was complete. At stage 29, few corticotropic cells were present only in the dorsal face of Rathke's pouch. No other immunoreactive cell type was revealed at this stage. At stage 32, the hypophysis had developed to a great extent though it was not yet elongated in a cephalic-caudal direction. At this stage, the corticotropic cells appeared more numerous and well differentiated in the rostral pars distalis and in the pars intermedia. Melanotropic, somatotropic and gonadotropic cells appeared simultaneously, with the same distributions as in the adult skink. At stage 34, the first thyrotropic cells appeared in the pars distalis but also in the pars intermedia, whereas rare prolactin cells were observed only at stage 35 in the medial pars distalis. Between stages 36 and 38, the gland was developed in the cephalic-caudal direction and all the cell types were completely differentiated with an evident increase in the number of prolactin cells. In embryos close to birth (stages 39-40), the hypophysis and the adenohypophysial cells were already similar to those of the adult animal.  相似文献

Summary In the pars distalis of the hypophysis of adult Rana temporaria, three types of nerve-fiber profiles were found at two distinct sites, in both lateral parts of the bordering regions of the anterior lobe with the intermediate lobe of the hypophysis. The first type of nerve-fiber profile consists of bundles of very fine axonal elements (diameter: <0.7 m). The second type is formed by larger nerve fibers (diameter up to 4 m) containing a few neurosecretory granules of approximately 100 nm. The third type of nervefiber profile resembles the second type but these nerve fibers make synaptoid contacts on at least two different types of glandular cells. The possible functional significance of these nerve fibers in the pars distalis is discussed.No nerve fibers were found (1) in the central part of the bordering region of the pars distalis with the intermediate lobe, (2) at the bordering region with the median eminence and (3) with the neurohypophysial stalk, and (4) in all other parts of the pars distalis.  相似文献

An immunohistochemical, light- and electron-microscopial study was made of the pars distalis in adult tammar wallabies (Macropus eugenii). The pars distalis of this marsupial mammal was divided into three regions, based on the distribution of cell types within the gland. Somatotropic, mammotropic, luteotropic, folliculotropic, corticotropic and thyrotropic cells were identified on the basis of their immunohistochemistry, cytology and ultrastructure. Non-granulated (folliculo-stellate) cells, identified in electron micrographs, were found throughout the pars distalis. Somatotropic cells were predominant in the posterior pars distalis in all animals examined. In the single male specimen and in the non-lactating females examined, small numbers of apparently inactive mammotropic cells were scattered throughout the pars distalis; the same cell type was apparently active and present in considerable numbers in lactating females. Only one morphological type of gonadotropic cell was evident; these cells were scattered throughout the pars distalis, but in largest numbers in the median region. Small numbers of thyrotropic cells were found, most commonly in the anterior pars distalis. Corticotrops were also observed in moderate numbers, predominantly in the anterior regions of the pars distalis.  相似文献

The pituitaries of the exotic carp (Carassius carassius) are studied at the light microscopic level, for the characterization of the adenohypophysial cell-types with particular emphasis to the gonadotropic potency of the pituitary in relation to the annual reproductive patterns. The gland in the fish is of the cranioleptobasic type. Based on the classical staining methods and localization of the differential cell-types, the adenohypophysis of the fish can be subdivided into the rostral (RPD) and proximal (PPD) pars distalis and the pars intermedia (PI). The pars nervosa (PN) of the fish consists of the neurosecretory (NS) fibres which show varied accumulations of the NS material in concert with the seasonal reproductive cycle. The NS tract is constituted of AF--ve nucleus praeopticus and AF + ve nucleus lateralis tuberis. 2 tinctorially different cell-types, corresponding to corticotrops and lactotrops of other teleosts, are identifiable in the RPD of the present species. In the PPD, 3 cell-types can be distinguished with various granule-staining procedures: they represent somatotrops, thyrotrops, and gonadotrops. Further, 2 gonadotropic cells with apparently different staining properties are discernible. In the PI are dispersed besides a few agranular cells, islets of 2 kinds of cells separable with combined PAS-PbH technique. Seasonal changes in the gonadosomatic index and diameter of the testicular lobules and egg diameter as indices of gonadal maturation are found to be closely interwoven with the morphometric alterations in the basophilic cell area (BCA) chosen as criterion of gonadotropic potency of the PPD. The maximum BCA values are preceded with the time of greatest atresia of the eggs indicating the influence of the pituitary gonadotropins in the follicular resorption in the test fish.  相似文献

Indirect immunofluorescence technique with anti-beta FSH and anti-beta oLH antisera has allowed us to detect "gonadotropic cells" in the pars distalis and in the pars tuberalis of the adenohypophysis of the monkey Macacus irus. In the pars distalis, 85-90 % of the "gonadotropic cells" react simultaneously with these two antisera ; 10-15 % of these cells react only either with anti-beta hFSH or anti-beta oLH antisera. The gonadotropic cells are dispersed in the whole pars distalis, amid the other cellular types ; indeed, in the female, there is a "gonadotropic zone" in the median zone of the lateral lobes of the gland. In the pars tuberalis, we have observed "gonadotropic cells" which react only with anti-beta oLH antiserum. These results are compared with observations of some authors.  相似文献
